Versatile Applications of Cyanoacetic Acid in Organic Chemistry: Active Methylene Compound for the Knoevenagel Condensation and Organocatalyst for the Biginelli Reaction Chemistry

The application of cyanoacetic acid as a catalyst for the Biginelli reaction and as an active methylene compound for the Knoevenagel condensation reaction was evaluated. Using cyanoacetic acid as a Bronsted acid catalyst, after a synthetic optimization process, it was possible to synthesize eight dihydropyrimidinones with good yields (80-99%) using ethanol as solvent. It is the first time, to our knowledge, that the use of cyanoacetic acid is reported in the synthesis of this class of compounds, which have a wide bioactive potential. Also, cyanoacetic acid was used as a reagent in the Knoevenagel condensation, through which polyfunctionalized olefins were obtained and can be used as building blocks for structurally complex molecules. By using KOH as catalyst, eleven Knoevenagel adducts were synthesized with good yields (65-97%), under microwave irradiation as heating source, in water. Moreover, Knoevenagel adducts containing halogenated substituents (F, Cl) showed potential larvicidal activity with lethal concentrations (LC50) of 19.63, 33.84 µg mL 1 and LC90 of 27.46 and 48.16 µg mL1. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Progenesis QI (PQI) is a multiplatform bioinformatics tool that facilitates the identification workflow for metabolomics experiments. PQI uses fragmentation data provided by MassBank of North America (MoNA) libraries, among others, for metabolite annotation. However, PQI does not officially support MoNA libraries and other libraries based on structure-data files (.sdf). This paper describes the development and application of a software named MoNA to Progenesis QI Library Converter, allowing PQI and MoNA by correcting the fragmentation data of the library for Progenesis readability. We evaluated several public experimental datasets, including human plasma, plant extracts, cultured cells, bacteria, rat serum, and rat hippocampus. The results showed that it is mandatory to proceed with file conversion of each library to allow PQI to access fragmentation information from .msp (main spectra profile) files. Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development. Assessing the Spatial Distribution of Key Flavonoids in Mentha × piperita Leaves: An Application of Desorption Electrospray Ionization Mass Spectrometry Imaging (DESI-MSI)

Desorption electrospray ionization mass spectrometry imaging (DESI-MSI) was used to assess the spatial distribution of some key flavonoids in peppermint (Mentha × piperita L.) leaves. The chemical images were generated by applying DESI-MSI on the peppermint leaves imprinting while acquiring the spectra in the negative ion mode. The following key flavonoids were detected and grouped within well-known biosynthetic routes in plants: naringenin route [naringenin, sakuranetin, hesperetin, hesperidin, luteolin, and apigenin]; luteolin route [luteolin, chrysoeriol, luteolin-7-O-D-glucuronide, luteolin-7-O-D-glucoside, luteolin-7-O-neohesperidoside]; apigenin route [apigenin, apigenin-7-O-gentiobioside, apigenin-7-O-neohesperidoside, thymusin/pilosin, pebrellin, acacetin/genkwanin, ladanein, xanthomicrol/pedunculin/nevadensin]. Maps of the spatial distribution of these flavonoids throughout the peppermint leaves within each route were then displayed. The results described herein comprise an important (although still underexplored) subject that certainly will experience a remarkable growth in the next years.

Amino acids/ureides and carbohydrates are the main nutrients for the plants growth. These metabolites are continuously translocated through the plant organism via well-known transportation mechanisms. The well-established source-to-sink relationship involves the transfer of these nutrients from mature (source) to young (sink) leaves. Desorption electrospray ionization mass spectrometry imaging (DESI-MSI) is used herein to determine the spatial distribution of amino acids/ureides and carbohydrates in the imprintings of Mentha × piperita L. leaves at three distinct maturation stages: young, expanding, and mature. Hence, chemical images of some typical carbohydrates (glucose, sucrose, sorbitol/mannitol) and free amino acids/ureides (phenylalanine, leucine, histidine, aspartate, asparagine, tyrosine, glutamate, allantoic acid) clearly point to a source-to-sink relationship, as the young leaves are the final destination for some of these key metabolites. These results therefore reveal the high potential of DESI-MSI to investigate biochemical processes in plants, a still underexplored area.

First record of Rhipidocotyle santanaensis (Digenea) parasitizing Acestrorhynchus lacustris from Batalha River, Brazil

Resumo O presente trabalho descreve a ocorrência do digenético Rhipidocotyle santanaensis Lunaschi, 2004 no Brasil. Estes parasitos foram encontrados no fígado, coração, gônadas, intestino, cecos pilóricos, estômago, bexiga natatória e cavidade do peixe Acestrorhynchus lacustris (Lütken, 1875) provenientes do rio Batalha, Estado de São Paulo, com maior abundância nas gônadas e trato digestório. Os espécimes dos parasitos encontrados neste trabalho apresentaram características morfológicas muito semelhantes aos espécimes provenientes da Argentina, com diferenças apenas em relação ao ducto ejaculatório e vesícula seminal. Porém, os exemplares brasileiros apresentaram grandes diferenças morfométricas entre os espécimes descritos na Argentina, com medidas muito superiores. Este é o primeiro registro desta espécie de parasito no Brasil e neste hospedeiro.
Abstract This paper describes the occurrence of digenetic Rhipidocotyle santanaensis Lunaschi, 2004 in Brazil. This parasite was found in the liver, heart, gonads, intestine, pyloric caeca, stomach, swim bladder and cavity of Acestrorhynchus lacustris (Lütken, 1875) from the Batalha River, São Paulo State, with higher incidence in gonads and digestive tract. The parasite specimens found in this study showed morphological characteristics very similar to specimens from Argentina, with differences only in relation to the ejaculatory duct and seminal vesicle. However, Brazilian specimens presented great morphometrical differences between specimens described in Argentina, with much higher measures. This is the first record of this metazoan parasite species in Brazil and in this host fish.

Fifteen naphthoquinones, sixteen phenazines and fifteen aryl triazoles were synthesized and evaluated against Mycobacterium tuberculosis. Twenty five substances are reported here for the first time and, among all of the compounds evaluated, six presented MIC (minimal inhibitory concentration) values < 6.25 µg mL-1. These substances are promising antimycobacterial prototypes.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A listeriose é uma infecção não incomum, geralmente associada com recém-nascidos e pacientes imunodeprimidos que tem sido poucas vezes encontrada em pacientes com AIDS. Esta escassez de relatos despertou o interesse de diversos investigadores. Neste artigo, os autores relatam um caso de septicemia e meningite por Listeria monocytogenes em paciente com AIDS e fazem uma revisão da literatura. Novos conhecimentos sobre a interação agente - hospedeiro nas infecções por listeria, como o papel do fator de necrose tumoral e da integralina, estão permitindo novas interpretações da aparente escassez de infecções por listeria em pacientes com AIDS. O número de relatos vem crescendo em período recente, sugerindo que a associação pode ser mais freqüente do que aparenta ser, em parte explicada por inadequação no diagnóstico.
Listeriosis is a not uncommon infection in humans, usually associated with immunodeficient states and with newborns. However, relatively few cases have been reported in HIV-infected patients. This scarcity of reported cases has aroused interest in the association of listerosis and AIDS. In this paper we present a case of meningitis and septicemia caused by Listeria monocytogenes in a female patient with AIDS. A review of recent medical literature indicates that association of listeriosis and AIDS may be more common than it seems. Recent research in host-parasite interaction in listerial infection suggests an important role for tumor necrosis factor (TNF) and for integralin, a bacterial protein, in modulating listerial disease in AIDS patients. Inadequate diagnosis may be in part responsible for the scarcity of reports.
