Prevalence of myopic eyes in private practice and public care services in Brazil: 11 years of retrospective analysis Brazil 1

ABSTRACT Purpose: This study aimed to examine the prevalence of myopic eyes over 11 years (2008-2018) in a private clinic and a public assistance service. Methods: We retrospectively evaluated 6332 individuals (12,664 eyes) between 5 and 25 years old, seen at a private clinic-CEMO (2,663 individuals) and a public service-HOIP (3,669 individuals) from 2008 to 2018. We evaluated the prevalence of myopic eyes (EE ≤-0.50) and high myopic eyes (EE ≤-6.00). Results: Sex and services did not show statistical differences. The variation in the prevalence of myopic and high myopic eyes showed a random pattern during the study period (this prevalence could not be increased). Prevalences ranged from 20.7% (in 2017) to 32.4% (in 2015) for myopic eyes and from 1.6% (in 2009 and 2016) to 3.3% (in 2015) for eyes with high myopia. The prevalence of myopia showed a statistically significant increase based on the age group. Conclusion: The prevalence of myopic eyes did not increase in our study. Resumo O aumento da modificação dos habitats e da perda de espécies demanda esforços consistentes para descrever e compreender os padrões de biodiversidade. O programa BIOTA/FAPESP foi criado nesse contexto e é uma iniciativa de sucesso para promover estudos em biodiversidade e conservação no Brasil. O BIOTA/Araçá é um projeto interdisciplinar que promoveu uma avaliação detalhada da biodiversidade da Baía do Araçá, um ecossistema costeiro localizado ao Norte do estado de São Paulo, Sudeste do Brasil. A baía engloba múltiplos habitats, tais como praias, manguezais, costões rochosos, e uma planície de maré, e também fornece importantes serviços ecossistêmicos. Infelizmente, a baía está sujeita à conflitos sócio-ambientais complexos que contrastam demandas econômicas, sociais e ambientais (i.e. a expansão das atividades do porto vizinho vs. a pesca artesanal de pequena escala e a proteção da biodiversidade). O presente estudo apresenta um levantamento das espécies bentônicas que ocorrem nos diferentes habitats da Baía do Araçá, incluindo dados obtidos durante o projeto BIOTA/Araçá e de investigações realizadas anteriormente na área. As espécies bentônicas desempenham um papel importante no ambiente marinho, e estudar a diversidade desses organismos que vivem associados ao fundo é indispensável para compreender o funcionamento do meio ambiente. A macrofauna, meiofauna, e microorganismos associados aos fundos consolidado e inconsolidado foram listados, e informações adicionais foram fornecidas para cada espécie, tais como a distribuição geográfica e nos habitats. O checklist inclui 826 espécies, quase 70% registradas durante o projeto BIOTA/Araçá. Os taxa mais especiosos foram os anelídeos (225 spp.), moluscos (194 spp.), e crustáceos (177 spp.). Entre as espécies bentônicas listadas, sete são endêmicas da Baía do Araçá, 14 são consideradas ameaçadas de extinção, e sete são exploradas economicamente. Abstract Increasing habitat modification and species loss demand consistent efforts to describe and understand biodiversity patterns. The BIOTA/FAPESP Program was created in this context and it has been a successful initiative to promote studies on biodiversity and conservation in Brazil. The BIOTA/Araçá is an interdisciplinary project that provided a detailed evaluation of the biodiversity of Araçá Bay, a coastal seascape located on the North coast of the state of São Paulo, Southeast Brazil. The bay encompasses multiple habitats, such as beaches, mangroves, rocky shores, and a tidal flat, and provides important ecosystem services. Unfortunately, the bay is the subject of complex social-environmental conflicts that oppose economic, social, and environmental demands (i.e., the expansion of neighboring harbor activities vs. small-scale artisanal fisheries and protection of biodiversity). The present study presents a survey of the benthic species occurring in the different habitats of Araçá Bay, including data obtained during the BIOTA/Araçá project and previous assessments of the area. The benthic species play an important role in marine environments and studying the diversity of these organisms that live associated with the bottom is indispensable for comprehending the environment’s functioning. The macrofauna, meiofauna, and microorganisms associated with soft and hard bottom were listed, and additional information, such as the habitat and geographical distribution, were provided for each species. The checklist includes 826 species, almost 70% recorded during the BIOTA/Araçá project. The most speciose taxa were the annelids (225 spp.), mollusks (194 spp.), and crustaceans (177 spp.). Seven benthic species are endemic to Araçá Bay, 14 are considered threatened, and seven are economically exploited. Furthermore, the bay is the type locality of many taxa, and 11 new benthic species were described based on specimens sampled during the project. ABSTRACT In 2000, Cleide Costa published a paper presenting the state of knowledge of the Neotropical Coleopte ra, with a focus on the Brazilian fauna. Twenty-four years later, thanks to the development of the Coleoptera section of the Taxonomic Catalog of the Brazilian Fauna (CTFB - Catálogo Taxonômico da Fauna do Brasil) through the collaboration of 100 coleopterists from all over the globe, we can build on Costa’s work and present an updated overview of the state of knowledge of the beetles from Brazil. There are currently 35,699 species in 4,958 genera and 116 families known to occur in the country, including representatives of all extant suborders and superfamilies. Our data show that the Brazilian beetle fauna is the richest on the planet, concentrating 9% of the world species diversity, with some estimates accounting to up to 15% of the global total. The most diverse family in numbers of genera is Cerambycidae (1,056 genera), while in number of species it is Chrysomelidae (6,079 species). Conotrachelus Dejean, 1835 (Curculionidae) is the most species-rich genus, with 570 species. The French entomologist Maurice Pic is the author who has contributed the most to the naming of species recorded from Brazil, with 1,794 valid names in 36 families, whereas the Brazilians Ubirajara R. Martins and Maria Helena M. Galileo are the only ones among the top-ten authors to have named species in the 21st century. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT Objective: Obesity is characterized by a state of chronic, low-intensity systemic inflammation frequently associated with insulin resistance and dyslipidemia. Materials and methods: Given that chronic inflammation has been implicated in the pathogenesis of mood disorders, we investigated if chronic obesity that was initiated early in life – lasting through adulthood – could be more harmful to memory impairment and mood fluctuations such as depression. Results: Here we show that pre-pubertal male rats (30 days old) treated with a high-fat diet (40%) for 8-months gained ~50% more weight when compared to controls, exhibited depression and anxiety-like behaviors but no memory impairment. The prefrontal cortex of the obese rats exhibited an increase in the expression of genes related to inflammatory response, such as NFKb, MMP9, CCl2, PPARb, and PPARg. There were no alterations in genes known to be related to depression. Conclusion: Long-lasting obesity with onset in prepuberal age led to depression and neuroinflammation but not to memory impairment.

ABSTRACT The plasmid-mediated colistin-resistance gene named mcr-1 has been recently described in different countries and it became a public health challenge. Of note, few studies have addressed the spread of Escherichia coli harboring the mcr-1 gene in both, community and hospital settings. A total of seven colistin-resistant E. coli carrying mcr-1, collected from 2016 to 2018, from community (n=4), healthcare-acquired infections (n=2) and colonization (n=1) were identified in three high complexity hospitals in Sao Paulo, Brazil. These colistin-resistant isolates were screened for mcr genes by PCR and all strains were submitted to Whole Genome Sequencing and the conjugation experiment. The seven strains belonged to seven distinct sequence types (ST744, ST131, ST69, ST48, ST354, ST57, ST10), and they differ regarding the resistance profiles. Transference of mcr-1 by conjugation to E. coli strain C600 was possible in five of the seven isolates. The mcr-1 gene was found in plasmid types IncX4 or IncI2. Three of the isolates have ESBL-encoding genes (blaCTX-M-2, n=2; blaCTX-M-8, n=1). We hereby report genetically distinct E. coli isolates, belonging to seven STs, harboring the mcr-1 gene, associated to community and healthcare-acquired infections, and colonization in patients from three hospitals in Sao Paulo. These findings point out for the potential spread of plasmid-mediated colistin-resistance mechanism in E. coli strains in Brazil.

SUMMARY: Mesenchymal stem cells are characterized by in vitro high proliferation and multilineage potential maintenance. This study aimed to isolate and characterize equine YS mesenchymal stem cells and compare these with amniotic membranes. The yolk sac (YS) and amniotic membranes (AM) were obtained from 20 pregnant mares with gestational age around 30 days. Cells were cultured in α-MEM supplemented with 15 % FBS, 1 % antibiotic solution, 1 % L-glutamine and 1 % nonessential amino acids. To cell characterization we used cytogenetic analysis, fibroblast colony-forming unit assays, cell growth curves, immunophenotyping, flow cytometry, differentiation assays and teratoma formation. Results: Both cell sources presented fibroblastoid and epithelioid-like format. The YS cells have lower colony formation potential then AM ones, 3 versus 8 colonies per 103 plated cells. However, YS cells grew progressively while AM cells showed steady. Both, the YS and amnion cells immunolabeled for Oct-4, Nanog, SSEA-3, cytokeratin 18, PCNA, and vimentin. In addition, presented mesenchymal, hematopoietic, endothelial and pluripotency markers in flow cytometry. Discussion: Both cell sources presented high plasticity and differed into osteogenic, adipogenic, and chondrogenic lineages, and no tumor formation in nude mice was observed. The results suggest that horse YS may be useful for cell therapy such as amnion-derived cells.
RESUMEN: Las células madre mesenquimales se caracterizan por una alta proliferación in vitro y un mantenimiento potencial de múltiples líneas. Este estudio tuvo como objetivo aislar y caracterizar las células madre mesenquimales del saco vitelino equinas y compararlas con las membranas amnióticas. Se obtuvo el saco vitelino (SV) y las membranas amnióticas (MA) de 20 yeguas preñadas con edad gestacional de aproximadamente 30 días. Las células se cultivaron en α -MEM suplementado con 15 % de FBS, 1 % de solución antibiótica, 1 % de L-glutamina y 1 % de aminoácidos no esenciales. Para la caracterización celular utilizamos análisis citogenéticos, ensayos de unidades de colonias de fibroblastos, curvas de crecimiento celular, inmunofenotipaje, citometría de flujo, ensayos de diferenciación y formación de teratomas. Ambas fuentes celulares presentaron formato fibroblastoideo y epitelioide. Las células SV tienen un potencial de formación de colonias más bajo que las de MA, 3 versus 8 colonias por 103 células en placa. Sin embargo, las células SV crecieron progresivamente mientras que las células MA se mostraron estables. Tanto las células YS como las células amnios están inmunomarcadas para Oct-4, Nanog, SSEA-3, citoqueratina 18, PCNA y vimentina. Además, presentó marcadores mesenquimales, hematopoyéticos, endoteliales y pluripotenciales en citometría de flujo. Ambas fuentes celulares presentaron alta plasticidad y diferían en linajes osteogénicos, adipogénicos y condrogénicos, y no se observó formación de tumores en ratones. Los resultados sugieren que el SV de caballo puede ser útil para la terapia celular, como las células derivadas de amnios.

Abstract The identification of families at-risk for hereditary cancer is extremely important due to the prevention potential in those families. However, the number of Brazilian genetic services providing oncogenetic care is extremely low for the continental dimension of the country and its population. Therefore, at-risk patients do not receive appropriate assistance. This report describes the creation, structure and management of a cancer genetics service in a reference center for cancer prevention and treatment, the Barretos Cancer Hospital (BCH). The Oncogenetics Department (OD) of BCH offers, free of charge, to all patients/relatives with clinical criteria, the possibility to perform i) genetic counseling, ii) preventive examinations and iii) genetic testing with the best quality standards. The OD has a multidisciplinary team and is integrated with all specialties. The genetic counseling process consists (mostly) of two visits. In 2014, 614 individuals (371 families) were seen by the OD. To date, over 800 families were referred by the OD for genetic testing. The support provided by the Oncogenetics team is crucial to identify at-risk individuals and to develop preventive and personalized behaviors for each situation, not only to the upper-middle class population, but also to the people whose only possibility is the public health system.

RESUMO A fertilização é uma das maneiras para aumentar a produtividade e a utilização de fertilizantes de liberação lenta pode ser vantajosa, permitindo a assimilação dos nutrientes à medida que as plantas crescem. O objetivo deste trabalho foi avaliar o efeito da utilização de fertilizante de liberação lenta no crescimento inicial de um plantio de Eucalyptus urophylla x E.grandis, visando reduzir o número de fertilizações de cobertura. O delineamento experimental foi o de blocos ao acaso, com quatro tratamentos fornecendo as mesmas quantidades de nutrientes, porém variando o parcelamento e a fonte do fertilizante: T1-Fertilizante convencional com aplicação parcelada; T2-Fertilizante convencional com aplicação em dose única aos três meses; T3-Fertilizante de liberação lenta no plantio; e T4-Fertilizante de liberação lenta aos três meses. Foram avaliadas as seguintes características: mortalidade, altura, DAP, volume de madeira, teores foliares dos macronutrientes e de N e K na solução do solo. A mortalidade média foi de 4%, altura de 8,5 m, DAP de 7,5 cm e volume de 24 m3.ha-1, aos 18 meses de idade, não diferindo entre os tratamentos. Apenas o teor foliar de K diferiu entre os tratamentos, sendo a menor concentração com a aplicação parcelada do fertilizante. A aplicação do fertilizante de liberação lenta aos três meses proporcionou os menores teores na solução do solo. A aplicação parcelada dos nutrientes não resultou em ganho de produtividade ou benefícios nutricionais. O uso de fertilizantes de liberação lenta possibilita a redução do número de fertilizações de cobertura, sem o risco de lixiviação dos nutrientes.
ABSTRACT Fertilization is one of the most effective ways to increase crop productivity, and the use of slow release fertilizers could be advantageous, allowing the assimilation of nutrients as the plants grow. The objective was evaluating the effect of slow release fertilizers in a Eucalyptus urophylla x E. grandis stand in order to reduce the number of fertilization applications. The experimental design was randomized blocks with four treatments. Treatments were applied with the same amounts of nutrients: T1-Convetional fertilizer with split application; T2-Conventional fertilizer in a single dose at 3 months; T3-Controlled-release fertilizer applied at planting; and T4-Controlled-release fertilizer applied 3 months after planting. We evaluated the mortality, initial growth, leaf nutrition and N and K in the soil solution. The experiment showed an average mortality of 4%, height of 8.5 m, DBH of 7.5 cm and volume of 24 m3.ha-1 at 18 months of age, with no difference among treatments for these characteristics. There were no differences in foliar concentrations of N, P, Ca, Mg and S, only the K differed among treatments, with the lowest concentration at conventional fertilizer split application treatment. The application of slow release fertilizer at 3 months showed the lowest concentrations of N and K in the soil solution. The split application of nutrients showed no improvement in eucalypt growth or nutritional benefits. The use of slow-release fertilizer is possible to reduce the number of fertilization application with no risk of nutrients leaching.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Efeito de extratos de plantas do Cerrado em Dipetalogaster maxima (Uhler) (Hemiptera, Reduviidae)

A transmissão da doença de Chagas ocorre, principalmente, por meio de fezes de hemípteros hematófagos (Triatominae), os quais ingerem o Trypanosoma cruzi ao se alimentarem do sangue de pessoas ou outros mamíferos infectados. Para o controle dos triatomíneos, os piretróides são os principais inseticidas utilizados. Entretanto, algumas populações de insetos demonstraram resistência a determinados piretróides, indicando a necessidade do desenvolvimento de novos inseticidas eficazes no controle desses vetores. Assim, foi avaliada a atividade inseticida de 83 extratos vegetais, pertencentes a 35 espécies diferentes, em ninfas do primeiro estádio de Dipetalogaster maxima (Uhler, 1894) (Hemiptera: Reduviidae), triatomíneo encontrado no México. Para o teste tópico, foram aplicados 50 ìg de cada extrato nos tergitos abdominais de dez ninfas, em duplicata. Como controles, foram utilizados insetos tratados com etanol, acetona ou sem nenhum tipo de tratamento. Os triatomíneos foram observados durante 28 dias. Nenhum extrato apresentou atividade inseticida significativa, entretanto, o extrato hexânico do fruto e o etanólico da casca do caule de Simarouba versicolor (Simaroubaceae) inibiram a taxa de ecdise em D. maxima (40% e 25%, respectivamente). Sugere-se que estes extratos sejam quimicamente investigados e monitorados por ensaios biológicos a fim de determinar os componentes, para que estes possam ser utilizados como modelos moleculares ou como compostos biorracionais nos programas de controle de insetos.
Chagas disease, also known as American Trypanosomiasis, is chiefly transmitted by faeces of haematophagous bugs (Triatominae) that ingested Trypanosoma cruzi from blood of infected people or other mammals. Pyrethroids have been the main insecticides used against these insects. However, some populations of insects have shown significant levels of resistance to several pyrethroids, indicating the need of new insecticides for the control of triatomines. Thus, the insecticidal activity of 83 Cerrado plant extracts belonging to 35 species were assayed on first instar nymphs of Dipetalogaster maxima (Uhler, 1894) (Hemiptera, Reduviidae), species found in Mexico. For the extract application on triatomines, 50 μg of the extract were topically applied in duplicate on dorsal tergites of ten insects. Insects topically treated with acetone, ethanol, as well as insects with no treatment were used as controls. Triatomines were observed over a 28-day period. The extracts showed no insect mortality, however, fruit hexane and stem bark ethanolic extracts of Simarouba versicolor (Simaroubaceae) inhibited the rate of ecdysis in D. maxima (40% and 25%, respectively). These preliminary data suggest that this extracts should be chemically investigated and monitored through biological assays in order to determine the components, so that it may be used as a molecular model or as biorational compounds for use in insect control programmes.

Assim, foi avaliada a atividade inseticida de 24 extratos vegetais em ninfas do quarto estádio de Rhodnius milesi Carcavallo, Rocha, Galvão & Jurberg, em condições de laboratório. Para o teste tópico, foram aplicados 50 µg de cada extrato nos tergitos abdominais de dez ninfas, em duplicata. Como controles, foram utilizados insetos tratados com etanol, acetona ou sem nenhum tipo de tratamento. Os triatomíneos foram observados durante 28 dias. Extratos hexânicos e etanólicos de Simarouba versicolor, Guarea kunthiana, Guarea guidonia e Talauma ovata causaram mortalidade entre 20% e 95% de R. milesi em comparação com os controles, onde não houve mortalidade dos insetos. Estes dados preliminares sugerem que o extrato etanólico da casca da raiz de S. versicolor e o extrato hexânico da raiz de G. guidonia, os quais foram responsáveis pela mortalidade de 95% e 75%, respectivamente, devem ser quimicamente investigados e monitorados por ensaios biológicos a fim de determinar seus componentes inseticidas, a serem utilisados como modelos moleculares ou como compostos biorracionais nos programas de controle de insetos.
Chagas' disease is chiefly transmitted by feces of haematophagous bugs (Triatominae) that ingested Trypanosoma cruzi from blood of infected people or animals. Pyrethroids have been the main insecticides used against these insects. However, some populations of insects have shown significant levels of resistance to several pyrethroids, indicating the need of new insecticides for the control of triatomines. Insecticidal activity of 24 Cerrado plant extracts belonging to five species of four families were assayed on fourth instar nymphs of Rhodnius milesi Carcavallo, Rocha, Galvão & Jurberg (Hemiptera: Reduviidae), under laboratory conditions. For the extract application on triatomines, 50 µg of the extract were topically applied in duplicate on dorsal tergites of ten insects. Insects topically treated with acetone, ethanol, as well as insects with no treatment were used as controls. Triatomines were observed over a 28-day period. Hexanic and ethanolic extracts of Simarouba versicolor, Guarea kunthiana, Guarea guidonia and Talauma ovata caused mortality between 20% and 95% of R. milesi in comparison with the controls, which showed no insect mortality. These preliminary data suggest that the ethanolic extract of the root bark of S. versicolor and the hexanic extract of the root of G. guidonia, responsible for a 95% and 75% insect mortality, respectively, should be chemically investigated and monitored through biological assays in order to determine their insecticidal components, that could be used as a molecular model or as biorational compounds for use in insect control programmes.

The most important features of the CE-ICP hyphenation, as well as its advantages and drawbacks as a tool for speciation are discussed. The fundamental principles of capillary electrophoresis and inductively coupled plasma mass spectrometry are also presented. Some applications involving different designs proposed in the literature to couple CE and ICP system for elemental speciation are reviewed.

No presente trabalho nós examinamos o uso potencial de sondas de oligonucleotídeos para caracterizar sorotipos de Neisseria meningitidis sem o uso de anticorpos mono-clonais (MAbs). A diversidade antigênica da proteína PorB forma a base do método de sorotipagem, todavia, o atual painel de MAbs utilizados, sub-estima em no mínimo 50% a diversidade desta proteína devido a falta de reagentes para as várias regiões variáveis (VRs) da proteína PorB ou porque várias variantes das VRs não são reagentes com os MAbs disponíveis. Nós analisamos o uso de sondas de oligonucleotídeos para caracterizar os sorotipos 10 e 19 de N. meningitidis. O gene porB da cepa protótipo do sorotipo 10 foi sequenciado e alinhado com outras 7 sequências de diferentes sorotipos, e as individuais VRs foram então analisadas. Os resultados com as sondas 21U (VR1-A) e 615U (VR3-B) contra 72 cepas de N. meningitidis confirmaram que VR1-A e VR3-B codificam epítopos para os MAbs 19 e 10 respectivamente. É possível o uso de sondas para a caracterização dos sorotipos e podemos tipar 100% da diversidade da VR do gene porB. Trata-se de um método simples, rápido, e especialmente útil para a análise de um grande número de amostras.
In the present study we examine the potential use of oligonucleotide probes to characterize Neisseria meningitidis serotypes without the use of monoclonal antibodies (MAbs). Antigenic diversity on PorB protein forms the bases of serotyping method. However, the current panel of MAbs underestimated, by at least 50% the PorB variability, presumably because reagents for several PorB variable regions (VRs) are lacking, or because a number of VR variants are not recognized by serotype-defining MAbs12. We analyzed the use of oligonucleotide probes to characterize serotype 10 and serotype 19 of N. meningitidis. The porB gene sequence for the prototype strain of serotype 10 was determined, aligned with 7 other porB sequences from different serotypes, and analysis of individual VRs were performed. The results of DNA probes 21U (VR1-A) and 615U (VR3-B) used against 72 N. meningitidis strains confirm that VR1 type A and VR3 type B encode epitopes for serotype-defined MAbs 19 and 10, respectively. The use of probes for characterizing serotypes possible can type 100% of the PorB VR diversity. It is a simple and rapid method specially useful for analysis of large number of samples.
