Resumo O aumento da modificação dos habitats e da perda de espécies demanda esforços consistentes para descrever e compreender os padrões de biodiversidade. O programa BIOTA/FAPESP foi criado nesse contexto e é uma iniciativa de sucesso para promover estudos em biodiversidade e conservação no Brasil. O BIOTA/Araçá é um projeto interdisciplinar que promoveu uma avaliação detalhada da biodiversidade da Baía do Araçá, um ecossistema costeiro localizado ao Norte do estado de São Paulo, Sudeste do Brasil. A baía engloba múltiplos habitats, tais como praias, manguezais, costões rochosos, e uma planície de maré, e também fornece importantes serviços ecossistêmicos. Infelizmente, a baía está sujeita à conflitos sócio-ambientais complexos que contrastam demandas econômicas, sociais e ambientais (i.e. a expansão das atividades do porto vizinho vs. a pesca artesanal de pequena escala e a proteção da biodiversidade). O presente estudo apresenta um levantamento das espécies bentônicas que ocorrem nos diferentes habitats da Baía do Araçá, incluindo dados obtidos durante o projeto BIOTA/Araçá e de investigações realizadas anteriormente na área. As espécies bentônicas desempenham um papel importante no ambiente marinho, e estudar a diversidade desses organismos que vivem associados ao fundo é indispensável para compreender o funcionamento do meio ambiente. A macrofauna, meiofauna, e microorganismos associados aos fundos consolidado e inconsolidado foram listados, e informações adicionais foram fornecidas para cada espécie, tais como a distribuição geográfica e nos habitats. O checklist inclui 826 espécies, quase 70% registradas durante o projeto BIOTA/Araçá. Os taxa mais especiosos foram os anelídeos (225 spp.), moluscos (194 spp.), e crustáceos (177 spp.). Entre as espécies bentônicas listadas, sete são endêmicas da Baía do Araçá, 14 são consideradas ameaçadas de extinção, e sete são exploradas economicamente. A baía é a localidade tipo de vários taxa, e 11 novas espécies bentônicas foram descritas com base em espécimes amostrados durante o projeto. Abstract Increasing habitat modification and species loss demand consistent efforts to describe and understand biodiversity patterns. The BIOTA/FAPESP Program was created in this context and it has been a successful initiative to promote studies on biodiversity and conservation in Brazil. The BIOTA/Araçá is an interdisciplinary project that provided a detailed evaluation of the biodiversity of Araçá Bay, a coastal seascape located on the North coast of the state of São Paulo, Southeast Brazil. The bay encompasses multiple habitats, such as beaches, mangroves, rocky shores, and a tidal flat, and provides important ecosystem services. Unfortunately, the bay is the subject of complex social-environmental conflicts that oppose economic, social, and environmental demands (i.e., the expansion of neighboring harbor activities vs. small-scale artisanal fisheries and protection of biodiversity). The present study presents a survey of the benthic species occurring in the different habitats of Araçá Bay, including data obtained during the BIOTA/Araçá project and previous assessments of the area. The benthic species play an important role in marine environments and studying the diversity of these organisms that live associated with the bottom is indispensable for comprehending the environment’s functioning. The macrofauna, meiofauna, and microorganisms associated with soft and hard bottom were listed, and additional information, such as the habitat and geographical distribution, were provided for each species. The checklist includes 826 species, almost 70% recorded during the BIOTA/Araçá project. The most speciose taxa were the annelids (225 spp.), mollusks (194 spp.), and crustaceans (177 spp.). Seven benthic species are endemic to Araçá Bay, 14 are considered threatened, and seven are economically exploited. Furthermore, the bay is the type locality of many taxa, and 11 new benthic species were described based on specimens sampled during the project. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract This study aimed to characterize the spatial distribution and composition of living Benthic Foraminifera (BF) and to comprehend how environmental conditions (e.g., organic matter) can affect communities of these protozoa in the northern and southern sectors of the Santos Basin (SB), in the continental slope and São Paulo Plateau. In this context, 23 stations (65 samples including replicates at each station) were collected between 400 and 2,400 m water depth. Multivariate analyses revealed that the ecological structure of the community changes mainly along the bathymetric gradients. Stations located between 400 and 700 m, both in northern and southern sectors, are characterized by the presence of indicator species of high intensity of currents, such as Globocassidulina subglobosa and Trifarina bradyi. These stations are also mainly marked by the occurrence of Epistominella exigua, a phytodetritivore species. The stations at 1,000 and 1,300 m depth, in both sectors, are characterized by high accumulation of organic matter in the sediments, which favors the development of agglutinated foraminifera species, such as those of the genus Reophax. Finally, the lower slope and the São Paulo Plateau, in both sectors, are oligotrophic regions, with pulses of labile organic carbon, probably low current velocities and the presence of Alabaminella weddellensis. The quantity and quality of food, which are closely related to hydro-sedimentary dynamics and bentho-pelagic coupling in the slope and São Paulo Plateau, are the main factors that influence the distribution of living BF assemblages in the SB. Resumo A utilização de resíduos da produção de café para a obtenção de biocarvão é uma abordagem sustentável, que visa minimizar o impacto ambiental desses materiais. Assim, o objetivo deste trabalho foi estudar o efeito da adição de diferentes concentrações de biocarvão de café na composição do substrato sobre a qualidade fisiológica de sementes de alface (Lactuca sativa) e sorgo (Sorghum bicolor). As variáveis analisadas foram: teor de água das sementes, porcentagem de germinação, índice de velocidade de germinação, comprimento da parte aérea, comprimento da raiz e massa seca das plântulas. O delineamento experimental utilizado foi inteiramente casualizado, com cinco concentrações de biocarvão (0; 7,5; 15; 30 e 60%), conduzido com quatro repetições de 25 sementes. A utilização do biocarvão nas concentrações estudadas não proporciona aumento na porcentagem de germinação e no vigor das sementes de alface e sorgo. O aumento da concentração de biocarvão causou menor vigor das sementes, sugerindo efeito tóxico. Para germinação de sementes, não houve diferença significativa entre as espécies de alface e sorgo, independente do tratamento. Para o índice de velocidade de germinação, as sementes de sorgo apresentam maiores médias, excetuando-se para o tratamento com adição de 15% de biocarvão de palha de café. As sementes de alface apresentam maiores médias de comprimento de parte aérea, excetuando-se para o tratamento com 100% de substrato comercial. Abstract The use of residues from coffee production to obtain biochar is a sustainable approach, which aims to minimize the environmental impact of these materials. In this study, the effect of adding coffee straw biochar on the physiological quality of lettuce and sorghum seeds was investigated. Thus, the objective of this work was to study the effect of adding different concentrations of coffee biochar in the substrate composition on the physiological quality of lettuce (Lactuca sativa) and sorghum (Sorghum bicolor) seeds. The experimental design used was completely randomized, with five concentrations of biochar (0; 7.5; 15; 30 and 60%), conducted with four replications of 25 seeds. The use of biochar in the concentrations studied does not provide an increase in the average germination percentage and vigor of lettuce and sorghum seeds. The increase in the concentration of biochar caused less seed vigor, suggesting a toxic effect. For seed germination, there was no significant difference between lettuce and sorghum species, regardless of treatment. For the germination speed index, sorghum seeds have higher means, except for the treatment with the addition of 15% coffee straw biochar. Lettuce seeds have higher shoot length averages, except for treatment with 100% commercial substrate. ABSTRACT The expression ‘you need to know to conserve’ is a well-known cliche among biologists. Documenting the richness of a group of organisms is the first step towards understanding biodiversity and preparing efficient conservation plans. In this context, many efforts have been made to quantify the number of species on Earth and estimate the number of species still unknown to science. A few countries have complete and integrated databases estimating the approximate number of species recorded for their territory, particularly in the Global South. In Brazil, a country of continental dimensions, revealing the richness of the second most diverse clade of invertebrates (=Mollusca) has been a goal of taxonomists. Recently, in an unprecedented, collective, and integrated effort among Brazilian malacologists, it was possible to estimate how many valid species of molluscs are there in Brazil. In this effort, more than 30 mollusc experts joined together to update the Taxonomic Catalogue of the Brazilian Fauna (TCBF), a governmental website that allows a quick and real-time updating of all Metazoan. So far, more than 5,000 updates have been made in TCBF, indicating the presence of 3,552 valid species of molluscs in Brazil, distributed among the main clades as follows: Caudofoveata (10 spp.), Solenogastres (6 spp.), Polyplacophora (35 spp.), Scaphopoda (43 spp.), Cephalopoda (92 spp.), Bivalvia (629 spp.) and Gastropoda (2,737 spp.). Abstract The continental margin off the southeastern Brazilian coast is punctuated by a series of geological-geomorphological features, such as subsurface saline diapirs and pockmarks at the seafloor interface, which evidence the abundant presence of oil and gas in the region. In several of these sites, hydrocarbons can be naturally released into the water column, areas are cold seep areas. These are marked by the presence of oil- and gas-dependent ecosystems, where specific organisms are able to fix carbon from hydrocarbon chemosynthesis. In addition, light hydrocarbon fluid flow through the sediment may build up authigenic carbonates that can be further colonized by cold-water corals, generating large carbonate mounds over geological time, normally positioned at the border of these pockmark features. The present work reports on a multidisciplinary oceanographic cruise carried out in the Santos Basin, SW Atlantic, to seek, map, and collect geological, chemical, and biological data from different deep-sea habitats. The cruise occurred in November 2019 on the R/V Alpha Crucis of the Oceanographic Institute of the University of São Paulo (IOUSP). We intended to discover and detail different geomorphological features, characterize free-living and symbiotic microorganisms, determine the chemosynthetic rates in relation to heterotrophic microbial production, and characterize the fauna and study their ecological and evolutionary links within and across ocean basins. All discoveries made during the cruise and their respective results will be presented separately in several papers that comprise this special volume.

Resumo O potencial biotecnológico das microalgas tem sido alvo de uma série de pesquisas que visam utilizar seu potencial para produzir macromoléculas de alto valor agregado. Uma especial atenção tem sido dada à produção de biocombustíveis, como biohidrogênio, biodiesel e bioetanol, a partir de lipídios e carboidratos de microalgas. A bioprospecção e a identificação precisa de microalgas são importantes na busca de linhagens com melhor desempenho em termos de produção de biomassa e lipídios. Metodologias que combinam morfologia e técnicas moleculares permitem uma identificação mais precisa das espécies. Assim, este trabalho teve como objetivo identificar a nova cepa LGMM0013 coletada na Represa do Iraí, localizado no estado do Paraná, Brasil e avaliar a produção de biomassa, carboidratos e lipídios. Com base na morfologia e na árvore filogenética obtida com sequências ITS (“Internal Transcribed Spacer”), a cepa LGMM0013 foi identificada como Desmodesmus abundans. D. abundans acumulou 1500 mg L-1 de biomassa seca após 22 dias de crescimento em condições autotróficas, 50% superior a Tetradesmus obliquus (LGMM0001) (Scenedesmaceae-Chlorophyceae), uma cepa cultivada frequentemente em fotobiorreatores para produção de biomassa no NPDEAS, na Universidade Federal do Paraná (UFPR) para a produção de biomassa. Na análise da biomassa de D. abundans LGMM0013 constatou-se acúmulo de 673,39 mg L-1 de carboidratos, 130% maior que T. obliquus (LGMM0001). A produção de lipídios foi de 259,7 mg L-1, equivalente à de T. obliquus. A privação de nitrogênio aumentou a produção de biomassa e carboidratos em D. abundans LGMM0013, indicando uma maior capacidade de produção de biomassa desta cepa.
Abstract The biotechnological potential of microalgae has been the target of a range of research aimed at using its potential to produce macromolecules with high added value. Particular focus has been given to biofuels' production, such as biohydrogen, biodiesel, and bioethanol from lipids and carbohydrates extracted from microalgal biomass. Bioprospecting and accurate identification of microalgae from the environment are important in the search for strains with better performance. Methodologies that combine morphology and molecular techniques allow more precise knowledge of species. Thereby, this work aimed to identify the new strain LGMM0013 collected at Iraí Reservoir, located in Paraná state, Brazil, and to evaluate the production of biomass, carbohydrates, and lipids from this new microalgal strain. Based on morphology and phylogenetic tree from internal transcribed spacer (ITS), strain LGMM0013 was identified as Desmodesmus abundans. D. abundans accumulated 1500 mg L-1 of dried biomass after 22 days of cultivation in autotrophic conditions, 50% higher than Tetradesmus obliquus (LGMM0001) (Scenedesmaceae-Chlorophyceae), usually grown in photobioreactors located at NPDEAS at the Federal University of Paraná (UFPR) to produce biomass. Analysis of the D. abundans biomass from showed an accumulation of 673.39 mg L-1 of carbohydrates, 130% higher than T. obliquus (LGMM0001). Lipid production was 259.7 mg L-1, equivalent to that of T. obliquus. Nitrogen deprivation increased the production of biomass and carbohydrates in D. abundans LGMM0013, indicating this new strain greater biomass production capacity.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Analysis of structural, morphological and magnetic properties of diluted magnetic semiconductor ZnO:Eu obtained by combustion reaction

Resumo Matriz semicondutora de ZnO, dopada com Eu nas concentrações de 0,05 e 0,10 mols, foi sintetizada por reação de combustão utilizando nitrato de zinco, nitrato de európio e ureia como combustível. No intuito de analisar o efeito da concentração de európio e da sinterização na estrutura, band gap, propriedades magnéticas e morfológicas do ZnO, as amostras foram sinterizadas a 1100 °C por 30 min e analisadas antes e após sinterização por difração de raios X, espectroscopia na região do ultravioleta e visível, magnetometria de amostra vibrante e microscopia eletrônica de varredura. A partir dos resultados obtidos, constatou-se que houve formação da fase semicondutora ZnO e também de uma segunda fase (Eu2O3). Observou-se que as amostras antes e após a sinterização apresentaram valores de band gap dentro da faixa dos semicondutores e ferromagnetismo à temperatura ambiente.
Abstract Eu-doped semiconductor matrix of ZnO at concentrations of 0.05 and 0.10 mols was synthesized by combustion reaction, using zinc nitrate, europium nitrate, and urea as fuel. In order to analyze the effect of europium concentration and sintering on the structure, band gap, magnetic and morphological properties of ZnO, the samples were sintered at 1100 °C for 30 min and analyzed before and after sintering via X-ray diffraction, ultraviolet and visible spectroscopy, vibrant sample magnetometry, and scanning electron microscopy. From the results obtained, it was found that there was the formation of the semiconductor phase ZnO and also a second-phase (Eu2O3). It was observed that the samples before and after sintering presented band gap values within the semiconductor range and ferromagnetism at room temperature.

Effects of resistance exercise training and stretching on chronic insomnia

Objective: The aim of this study was to assess the effects of resistance exercise and stretching on sleep, mood, and quality of life in chronic insomnia patients. Methods: Three 4-month treatments included: resistance exercise (n=10), stretching (n=10), and control (n=8). Sleep was evaluated with polysomnography, actigraphy, and questionnaires. Mood and quality of life were assessed with the Profile of Mood States (POMS) and the Medical Outcomes Study Short-Form 36-Item Health Survey (SF-36), respectively. Results: There were no significant treatment differences between resistance exercise and stretching. However, compared with the control treatment, resistance exercise and stretching led to significantly greater improvements in Insomnia Severity Index scores (-10.5±2.3, -8.1±2.0 vs. 2.3±1.8, respectively), and actigraphic measures of sleep latency (-7.1±4.6, -5.2±1.9 vs. 2.2±2.1 min), wake after sleep onset (-9.3±2.8, -7.1±3.0 vs. 3.6±4.2 min), and sleep efficiency (4.4±1.8, 5.0±0.8 vs. -2.3±2%). Pittsburgh Sleep Quality Index (PSQI) global scores (-5.3±0.8, -3.9±1.5 vs. -0.1±0.8) and sleep duration (1.2±0.3, 1.6±0.6 vs. -0.1±0.2 h) also improved following both experimental treatments compared with control. PSQI-Sleep efficiency increased after resistance exercise compared with control (19.5±3.9 vs. 2.1±4.3%). No significant differences were observed in polysomnography or quality of life measures. Tension-anxiety was lower in the stretching group than the control group. Conclusion: Moderate-intensity resistance exercise and stretching led to similar improvements in objective and subjective sleep in patients with chronic insomnia. A presente pesquisa visou elucidar quais aspectos da resposta imune em Diatraea flavipennella são suprimidos pelo parasitoide Cotesia flavipes, para garantir o sucesso do parasitismo. Investigou-se a presença de células em apoptose no corpo gorduroso pela técnica de TUNEL. De acordo com os resultados verificou-se redução nos níveis de oxido nítrico e na atividade da fenoloxidase nas lagartas parasitadas, aos três dias, e redução do número total de hemócitos aos três e sete dias. Na contagem diferencial, observou-se aumento no número de plasmatócitos e redução de esferulócitos, no terceiro dia de parasitismo. O número de micro-esferas melanizadas, nas lagartas parasitadas, foi inferior mostrando uma melanização menos intensa. A análise ultra-estrutural comfirmou o efeito imunossupressor de C. flavipes para suprimir a resposta de encapsulação em D. flavipennella, pois, somente foi evidenciado à formação de cápsula de hemócitos, aderidas à superfície das micro-esferas, nas lagartas não parasitadas. Como efeito do parasitismo, foi registrado ainda no terceiro dia presença de hemócitos e células do corpo gorduroso em apoptose, além deste último apresentar aspecto de degeneração. Conclui-se que C. flavipes suprimi a resposta imunológica celular e humoral de D. flavipennella e afeta drasticamente o tecido gorduroso.
The present research aimed to elucidate which aspects of immune responses in Diatraea flavipennella are suppressed by the parasitoid Cotesia flavipes, thus, ensuring parasitism success. We investigated the presence of apoptosis in fat body cells through the TUNEL technique. According to the results, reduced levels of nitric oxide and phenoloxidase activity were observed in larvae parasitized for three days, and reduced total number of hemocytes, after three and seven days. An increase in plasmatocytes and decrease in spherulocytes numbers were observed in the differential count on the third day of parasitism. The number of melanized microspheres in parasitized larvae was low and indicated less intense melanization. The ultrastructural analysis confirmed the immunosuppressive effect of C. flavipes on the encapsulation response of D. flavipennella because only the formation of hemocytes capsules, adhered to the microspheres' surface, was evidenced in non-parasitized caterpillars. The effect of parasitism was also recorded on the third day with the presence of hemocytes and apoptosis in fat body cells, including aspects of degeneration in the latter. We concluded that C. flavipes suppresses cellular and humoral immunological responses in D. flavipennella and drastically affects the host's fat tissue.

Amongst other botanical sources, Annona muricata L., Annonaceae, seeds and Piper nigrum L., Piperaceae, fruits are particularly enriched with acetogenins and piperine-related amides, respectively. These crude ethanolic extracts are potent Aedes aegypti bioactives that can kill Aedes aegypti larvae (dengue fever mosquito). A. muricata displayed a 93.48 µg/mL LC50 and P. nigrum an 1.84 µg/mL LC50. An uncommon pharmacognostical/toxicological approach was used, namely different combinations of both extracts to achieve an improved lethal effect on the larvae. The independence test (χ²) was utilized to evaluate the combination of the two crude extracts. All of the tested combinations behaved synergistically and these novel results were attributed to the completely different biochemical mechanisms of the differentiated chemical substances that were present in the two botanical sources. Besides the two above selected plants, Melia azedarach L., Meliaceae, Origanum vulgare L., Lamiaceae, and Ilex paraguariensis A. St.-Hil., Aquifoliaceae, in order of decreasing toxicity, may also be sought as potential extracts for the sake of synergic combinations.

scrapie is a transmissible neurodegenerative disorder of sheeps and goats. It is characterized by the deposition in the central nervous system of an abnormal isoform of a normal cellular protein (PrPc), coded by the PrP gene. Mutations at 136, 154 and 171 codons of exon 2, are described. Combinations of these codons show different levels of association with the susceptibility to this disease; from highly resistants (ARR/ARR) to highly susceptibles (VRQ/VRQ). Codons 171 and 136 are the main determinant of scrapie susceptibility while codon 154 plays a minor role. In the present communication, a sample of Uruguayan Creole sheeps (N=28) was analysed for 136 and 171 codons. Samples of genomic DNA were processed by PCR-RFLP. The results were obtained by electrophoresis in polyacrylamide gel (10.5%) stained with AgNO3. Genotypic frequencies for the 136 and 171 codons were analysed. The results revealed 5 different genotypes. The most frequent genotypes were those associated with more susceptibility to the disease: VQ/AQ= 0.39 y AQ/AQ= 0.29. The analysis of genetic risk revealed a 72% of highly susceptible individuals, in case of proper environmental conditions.
El scrapie es una enfermedad neurodegenerativa transmisible de ovinos y caprinos. Se caracteriza por la acumulación en el sistema nervioso central de una isoforma anómala de una proteína celular normal (PrPc) codificada por el gen PrP. En el exón 2 del gen se han identificado mutaciones en los codones 136, 154 y 171. La combinación de estos codones muestran diferentes niveles de asociación con la susceptibilidad a la enfermedad, desde muy resistentes (ARR/ ARR) a muy susceptibles (VRQ/VRQ). Los codones 171 y 136 son los mayores determinantes de la susceptibilidad al scrapie, mientras que el codón 154 juega un rol menor. En el presente trabajo se estudió una muestra de ovinos Criollos Uruguayos (N=28) para los codones 136 y 171. Las muestras de ADN genómico se procesaron por PCR-RFLP. Los resultados se obtuvieron por electroforesis en geles de poliacrilamida (10,5%) teñidos con AgNO3. Se analizaron las frecuencias genotípicas para los codones 136 y 171. Cinco genotipos diferentes fueron obtenidos, encontrándose en mayor frecuencia los genotipos de alta susceptibilidad: VQ/AQ= 0,39 y AQ/AQ= 0,29. El análisis de riesgo genético de esta majada según los genotipos obtenidos, reveló un 72% de animales muy susceptibles a la enfermedad en caso de manifestarse las condiciones ambientales propicias.
