This study details the preparation, antileishmanial assay, and in silico analysis of twenty 2-hydroxy-3-(1,2,3-triazolyl)propyl vanillin derivatives. These compounds were synthesized in three steps and evaluated against Leishmania infantum, Leishmania amazonensis, and Leishmania braziliensis promastigotes. Compounds 3s and 3t were the most effective, showing good activity against all Leishmania species tested. Molecular docking indicated that all compounds bind favorably to the sterol 14α-demethylase (CYP51) enzyme from L. infantum. ADMET (absorption, distribution, metabolism, excretion and toxicity) analysis indicated good oral bioavailability, non blood-brain barrier penetration, and high gastrointestinal absorption. Posaconazole and compounds 3e, 3s, and 3t remained stable in the CYP51 binding region during 100 ns molecular dynamics (MD) simulations. RESUMO A aplicação de cinza vegetal na agricultura surge como técnica promissora para potencializar a produtividade agrícola. Sua composição química tem aptidão para aprimorar os atributos do solo, atuando como fertilizante, corretivo e auxiliando na retenção de água. Entretanto, estudos recomendam adoção controlada, para evitar impactos adversos. O objetivo da pesquisa foi avaliar o potencial da cinza vegetal como estratégia de adubação na cultura do amendoim, com ênfase na otimização da produtividade das plantas e na redução do déficit hídrico. O experimento foi conduzido em casa de vegetação em vasos de 5 dm3; o delineamento experimental aplicado foi em blocos casualizados, formando um esquema fatorial 5x5, correspondente a cinco doses de cinza vegetal (0; 8; 16; 24 e 32 g dm3) e cinco disponibilidades hídricas (25; 50; 75; 100 e 125% da capacidade de campo), com quatro repetições. RESUMO Compostos derivados de petróleo presentes no solo podem comprometer o crescimento das plantas e inibir o desenvolvimento da população microbiana. Esses contaminantes diminuem a porosidade do solo, dificultam a absorção de água e podem interromper processos metabólicos das plantas. Assim, nesta pesquisa buscou-se avaliar os efeitos da presença no solo de três hidrocarbonetos no solo sobre o crescimento e desenvolvimento de espécies forrageiras e nos indicadores microbiológicos do solo. O experimento foi realizado em casa de vegetação no delineamento experimental de blocos casualizados, com quatro repetições. Os tratamentos foram dispostos em fatorial de 5×4, sendo o primeiro fator constituído pelas espécies avaliadas (Pennisetum glaucum, Zea mays, Brachiaria ruziziensis, Panicum maximum e Sorghum bicolor), e o segundo fator correspondendo a presença dos contaminantes: benzeno, tolueno ou xileno e solo sem contaminante. ABSTRACT Oil-derived compounds present in the soil can compromise plant growth and inhibit the development of the microbial population. These contaminants reduce soil porosity, hinder water absorption, and can disrupt plant metabolic processes. The aim of this research was to evaluate the effects of the presence of three hydrocarbons in the soil on the growth and development of forage species and on soil microbiological indicators. The experiment was carried out in a greenhouse using a randomized block design with four replications. Treatments were arranged in a 5×4 factorial, with the first factor consisting of the species evaluated (Pennisetum glaucum, Zea mays, Brachiaria ruziziensis, Panicum maximum and Sorghum bicolor) and the second factor corresponding to the presence of contaminants: benzene, toluene or xylene and soil without contaminants. The species were grown for 42 days. The tolerance of the species depended on the type of contaminant present in the soil. Resumo Introdução: O exercício físico está sendo incorporado ao tratamento de pacientes em hemodiálise, porém pouco se sabe sobre as principais características dessas intervenções. Objetivo: Descrever os protocolos de exercício físico prescritos para pacientes em hemodiálise no Brasil. Métodos: Uma revisão de escopo foi conduzida de acordo com as diretrizes JBI e Prisma-ScR. Foram realizadas pesquisas na Medline, Embase e em outras três bases de dados até maio de 2024. Outras fontes (sites, livros e diretrizes) também foram pesquisadas. Foram incluídas evidências de pacientes em hemodiálise, descrevendo protocolos de exercício físico em todos os ambientes e desenhos no Brasil. Resultados: Encontradas 45 evidências, resultando em 54 protocolos de exercício físico de 16 estados brasileiros. O exercício de força (33,3%), seguido do exercício aeróbico (22,2%), foi o mais prescrito para ser realizado durante a diálise (85,2%). Os profissionais mais prevalentes na supervisão dos programas foram fisioterapeutas e profissionais de educação física (37,0% e 18,5%, respectivamente). Todos os protocolos adotaram os princípios de treinamento tipo e frequência, enquanto a progressão foi adotada em apenas 53,7%. A frequência mais prescrita foi três vezes por semana (88,9%). A intensidade do exercício foi determinada predominantemente por métodos subjetivos (33,3%). Abstract Introduction: Exercise is being incorporated into the treatment of patients on hemodialysis; however, little is known about the major characteristics of these interventions. Objective: To describe the exercise protocols prescribed for hemodialysis patients in Brazil. Methods: A scoping review was conducted following JBI and Prisma-ScR guidelines. Searches were carried out in Medline, Embase and three other databases until May 2024. Other sources (websites, books and guidelines) were also investigated. Evidence from patients on hemodialysis describing exercise protocols in all settings and designs in Brazil was included. Results: Forty-five pieces of evidence were found, resulting in 54 exercise protocols from 16 Brazilian states. Strength exercises (33.3%), followed by aerobic exercises (22.2%), were the most prescribed, mainly to be performed during dialysis (85.2%). The most prevalent professionals supervising the programs were physiotherapists and exercise physiologists (37.0% and 18.5%, respectively). All protocols implemented the principles of type and frequency training, while progression was adopted in only 53.7%. The main prescription was three times per week (88.9%). Exercise intensity was predominantly determined by subjective methods (33.3%). Conclusion: Aerobic and strength exercises during dialysis were the most commonly prescribed modalities in Brazil, with the majority of programs being properly supervised by qualified professionals. However, existing protocols have not employed systematic progression throughout the intervention, which would be appropriate for providing better physiological responses and adaptations. RESUMO A contaminação do solo com metais tóxicos traz consigo graves problemas ambientais. Dentre esses metais, o cobre (Cu) é prejudicial ao desenvolvimento das plantas quando atinge elevados níveis de contaminação no solo. Assim, identificar espécies capazes de resistir a esse tipo de contaminação auxilia nos processos de revegetação e descontaminação desses locais. Portanto, o presente estudo teve como objetivo investigar a tolerância de plantas de Cedrela fissilis ao excesso de Cu através de variáveis morfofisiológicas e bioquímicas. Mudas de C. fissilis foram submetidas a cinco concentrações de Cu (0, 2, 4, 6 e 8 mg L-1). O delineamento experimental foi inteiramente casualizado, com cinco tratamentos e quatro repetições. ABSTRACT Soil contamination with toxic metals brings along severe environmental issues. Among these metals, copper (Cu) is harmful to plant development when it reaches high contamination levels in the soil. Thus, identifying species capable of resisting this contamination type helps these sites’ revegetation and decontamination processes. Therefore, the present study aimed to investigate the tolerance of Cedrela fissilis plants to excess Cu through morphophysiological and biochemical variables. C. fissilis seedlings were subjected to five concentrations of Cu (0, 2, 4, 6, and 8 mg L-1). The experimental design was completely randomized, with five treatments and four repetitions. Morphophysiological (number of leaves, shoot height, root length, dry weight, morphological variables of the root system, and leaf area) and biochemical (antioxidant enzymes, lipid peroxidation, hydrogen peroxide concentration, and photosynthetic pigments) traits and Cu accumulated in roots and shoot were evaluated. High concentrations of Cu had a negative effect on the shoot and root dry weight, photosynthetic rate, stomatal conductance, and transpiration rate. Abstract The objective of this study was to examine the impact of a high-fat finishing diet on the in vivo performance, anatomical and carcass characteristics, and meat quality of adult culled goats. Over a period of 28 days leading up to slaughter, eighteen adult culled goats were subjected to a finishing diet provided in quantities sufficient to meet 3.0 times the nutritional requirements of adult non-dairy maintenance goats. The female goats were divided into two groups: one fed with the baseline diet without fat supplementation (n = 9) and the other fed a diet concentrated supplemented with whole full-fat linseed (n = 9). The diets varied in fat content (2.8% vs. 8.4% DM). The high-fat group exhibited a higher dry matter intake compared to the control group (+37%), along with elevated plasma cholesterol and triglyceride levels. The control group experienced a significant decrease in daily weight gain between the second and final week of the finishing period (-70%), in contrast to the increase observed in the high-fat group (+59%). At slaughter, no differences were noted between the groups in anatomical and carcass characteristics. Additionally, no differences were found between the diets regarding proximate composition, pH, yellow color component, and lightness of loin. The high-fat group exhibited a higher redness of loin. The inclusion of high-fat levels in finishing diets for culled adult goats allows for increased feed intake and performance but does not appear to affect carcass or meat quality.

Resumo Fundamento: A Cardiomiopatia Chagásica Crônica (CCC) é causada por um processo inflamatório induzido pelo Trypanosoma cruzi, que leva à miocardite com fibrose reativa e reparativa. A CCC progride com alterações de perfusão miocárdica e eventos histopatológicos que afetam a Aptidão Cardiorrespiratória (ACR). Objetivos: Avaliamos os efeitos do Treinamento Físico Aeróbico (TFA) na perfusão miocárdica e nos comprometimentos morfológicos e funcionais relacionados à inflamação e fibrose em hamsters sírios com CCC. Como objetivo secundário, analisamos as áreas de secção transversa do músculo esquelético. Métodos: Hamsters com CCC e seus respectivos controles foram divididos em quatro grupos: CCC sedentário, CCC-TFA, controle sedentário e controle TFA. Sete meses após a infecção, os animais foram submetidos à ecocardiografia, à cintilografia de perfusão miocárdica e ao teste de esforço cardiopulmonar. TFA de intensidade moderada foi realizado durante cinquenta minutos, cinco vezes por semana, por oito semanas. Posteriormente, os animais foram reavaliados. A análise histopatológica foi realizada após os procedimentos acima mencionados. O nível de significância foi estabelecido em 5% em todas as análises (p<0,05). Resultados: Animais com CCC sedentários apresentaram piores Defeitos de Perfusão Miocárdica (DPM) ao longo do tempo, Fração de Ejeção do Ventrículo Esquerdo (FEVE) reduzida, e apresentaram mais inflamação e fibrose quando comparados aos demais grupos (análise ANOVA mista). Por outro lado, o TFA foi capaz de mitigar a progressão do DPM, atenuar a inflamação e a fibrose e melhorar a eficiência da ACR em animais CCC-TFA. Conclusão: Nosso estudo demonstrou que o TFA melhorou a disfunção cardíaca, DPM e reduziu a inflamação e a fibrose em modelos de hamster com CCC. Abstract Background: Chronic Chagas cardiomyopathy (CCC) is caused by an inflammatory process induced by Trypanosoma cruzi, which leads to myocarditis with reactive and reparative fibrosis. CCC progresses with myocardial perfusion abnormalities and histopathological events that affect cardiorespiratory fitness (CRF). Objectives: We evaluated the effects of aerobic physical training (APT) on myocardial perfusion and on morphological and functional impairments related with inflammation and fibrosis in Syrian hamsters with CCC. As a secondary objective, we analyzed the cross-sectional areas of the skeletal muscle. Methods: Hamsters with CCC and their respective controls were divided into four groups: CCC sedentary, CCC-APT, sedentary control and APT control. Seven months after infection, the animals underwent echocardiography, myocardial perfusion scintigraphy and cardiopulmonary exercise testing. Moderate-intensity APT was performed for fifty minutes, five times a week, for eight weeks. Subsequently, the animals were reassessed. Histopathological analysis was conducted after the above-mentioned procedures. The level of significance was set at 5% in all analyses (p<0.05). Results: CCC sedentary animals presented worse myocardial perfusion defects (MPD) over time, reduced left ventricle ejection fraction (LVEF) and showed more inflammation and fibrosis when compared to other groups (mixed ANOVA analysis). Conversely, APT was able to mitigate the progression of MPD, ameliorate inflammation and fibrosis and improve CRF efficiency in CCC-APT animals. Conclusions: Our study demonstrated that APT ameliorated cardiac dysfunction, MPD, and reduced inflammation and fibrosis in CCC hamster models. Additionally, CCC-SED animals presented skeletal muscle atrophy while CCC-APT animals showed preserved skeletal muscle CSA. ABSTRACT In Brazil, the lima bean is the second most economically significant legume within the genus Phaseolus. Climate change, particularly water scarcity, threatens the production of this species. The application of salicylic acid has mitigated the adverse effects of stress. This study aimed to assess the impact of salicylic acid on acclimatisation to water restriction in three genotypes of Phaseolus lunatus (‘Cara Larga’, ‘Cearense’, and ‘Orelha de Vó’). A completely randomised design with a triple factorial included three broad bean genotypes, two pre-conditionings with 1.0 mM salicylic acid and without this elicitor (0.0 mM), and three levels of water availability (75, 50, and 25%), totalling 18 treatments with eight replicates. Physiological and biochemical responses were evaluated after 60 days of treatment. The responses varied among the genotypes. ‘Cara Larga’ stood out regarding osmoregulatory and antioxidant parameters compared to the other genotypes. RESUMO A alta concentração de sais no solo provoca impacto na disponibilidade de água e nutrientes essenciais para o crescimento e fisiologia das plantas. Assim, são necessárias estratégias de manejo que possam mitigar os efeitos negativos do estresse salino, como por exemplo o uso de hormônios vegetais. Assim, este estudo teve como objetivo avaliar o efeito do estresse salino e aplicação de melatonina sobre crescimento e aspectos produtivos de plantas de rabanete. O experimento foi realizado em delineamento em blocos casualizados, em esquema fatorial 3 × 3, com quatro repetições, sendo considerada uma planta por parcela experimental. RESUMO O manejo dos níveis de condutividade elétrica é um dos fundamentos necessários para se obter maior produção e melhor qualidade de hortaliças. O objetivo deste trabalho foi avaliar o efeito da condutividade elétrica da solução nutritiva na cultura da couve-rábano cultivada em sistema semi-hidropônico. O delineamento experimental foi em blocos casualizados com cinco tratamentos associados à condutividade elétrica da solução nutritiva (1,31; 1,71; 2,37; 2,98 e 3,75 dS m-1) e quatro repetições. As plantas foram colhidas 78 dias após o transplante e avaliadas quanto ao crescimento, produção e qualidade pós-colheita. ABSTRACT Managing electrical conductivity levels is one of the fundamentals required to obtain greater production and better quality of vegetables. The aim of this study was to evaluate the effect of electrical conductivity of the nutrient solution on the kohlrabi crop cultivated in semi-hydroponic system. A randomized block design was used with five treatments associated with the electrical conductivity of the nutrient solution (1.31, 1.71, 2.37, 2.98, and 3.75 dS m-1) and four replications. The plants were harvested 78 days after transplanting and evaluated for growth, production, and postharvest quality. Nutrient solution with electrical conductivity (EC) of 1.31 dS m-1 promotes greater production of purple kohlrabi. The increase in EC reduced linearly most of the analyzed variables (leaf area, fresh mass of leaves, bulb and shoot, dry mass of leaves and total, bulb volume and firmness), with more significant losses for the bulb fresh mass (50.54%) and bulb volume (57.37%) variables. Resumo Nos últimos anos, o desenvolvimento de tecnologias de alto rendimento para obtenção de dados sequenciais potencializou a possibilidade de análise de dados proteicos in silico. No entanto, quando se trata de estudos de interação de poliproteínas virais, existe uma lacuna na representação dessas proteínas, devido ao seu tamanho e comprimento. Para estudos utilizando técnicas de ponta como o Aprendizado de Máquina, uma boa representação dessas proteínas é imprescindível. Apresentamos uma alternativa para este problema, implementando um protocolo de fragmentação e modelagem para preparar essas poliproteínas na forma de fragmentos de peptídeos. Tal procedimento é feito por diversos scripts, implementados em conjunto no workflow que chamamos de PolyPRep, uma ferramenta escrita em script Python e disponível no GitHub. Este software está disponível gratuitamente apenas para usuários não comerciais.

Abstract In recent years, the development of high-throughput technologies for obtaining sequence data leveraged the possibility of analysis of protein data in silico. However, when it comes to viral polyprotein interaction studies, there is a gap in the representation of those proteins, given their size and length. The prepare for studies using state-of-the-art techniques such as Machine Learning, a good representation of such proteins is a must. We present an alternative to this problem, implementing a fragmentation and modeling protocol to prepare those polyproteins in the form of peptide fragments. Such procedure is made by several scripts, implemented together on the workflow we call PolyPRep, a tool written in Python script and available in GitHub. This software is freely available only for noncommercial users.

Resumo A verificação de mudanças morfológicas e fisiológicas sob diferentes condições luminosas em espécies frutíferas nativas em estágio juvenil é importante, uma vez que indicam o ambiente adequado para a formação de mudas com alto vigor. Objetivou-se verificar o crescimento e as alterações morfofisiológicas sob gradiente de sombreamento em mudas de feijoa (Acca sellowiana (O. Berg) Burret) para obter mudas de boa qualidade, adequadas para fomentar os plantios da espécie em pomares. As mudas foram cultivadas por vinte e um meses sob quatro tratamentos de sombreamento (0%, 30%, 50% e 80%). Foram avaliados parâmetros de crescimento, pigmentos fotossintéticos, trocas gasosas, fluorescência da clorofila e anatomia foliar. Mudas a pleno sol e 30% de sombra apresentaram maior crescimento em altura, diâmetro e acúmulo de massa seca, devido à maior taxa de fotossíntese. Como principais mecanismos de aclimatação sob 80% de sombra, as mudas desenvolveram maior área foliar, redução da espessura do limbo foliar e aumento do rendimento quântico do fotossistema II. Mesmo assim, a assimilação líquida de CO2 e a taxa de transporte de elétrons foram menores e, consequentemente, houve restrição ao crescimento e acúmulo de massa seca das mudas no maior nível de sombreamento. Portanto, para a obtenção de mudas de feijoa de maior qualidade, recomendamos que seja realizada a pleno sol ou até 30% de sombra, para maximizar o vigor das mudas em viveiros e, posteriormente, este ambiente de luz também pode ser utilizado em pomares para favorecer o crescimento e a produção de frutos.

Abstract Understanding morphological and physiological changes under different light conditions in native fruit species in juveniles’ stage is important, as it indicate the appropriate environment to achieve vigorous saplings. We aimed to verify growth and morphophysiological changes under shade gradient in feijoa (Acca sellowiana (O. Berg) Burret) to achieve good quality saplings adequate to improve cultivation in orchards. The saplings were grown for twenty-one-month under four shading treatments (0%, 30%, 50%, and 80%). Growth, photosynthetic pigments, gas exchanges, chlorophyll fluorescence, and leaf anatomy parameters were evaluated. Saplings under full sun and 30% shade had higher height and diameter growth and dry mass accumulation due to higher photosynthesis rate. As main acclimatization mechanisms in feijoa saplings under 80% shade were developed larger leaf area, reduced leaf blade thickness, and enhanced quantum yield of photosystem II. Even so, the net CO2 assimilation and the electron transport rate was lower and, consequently, there was a restriction on the growth and dry mass in saplings under deep shade. Therefore, to obtain higher quality feijoa saplings, we recommend that it be carried out in full sun or up to 30% shade, to maximize the sapling vigor in nurseries and, later, this light environment can also be used in orchards for favor growth and fruit production.
