RESUMO Leptospirose e toxoplasmose são zoonoses de caráter ocupacional disseminadas mundialmente, principalmente em áreas tropicais e socioeconomicamente vulneráveis. Ambas as doenças têm hospedeiros vertebrados, que espalham o agente infeccioso para o meio ambiente e são potencialmente infecciosos para o homem, as quais podem evoluir para casos fatais. O presente estudo reforça a importância da saúde única para o controle e a profilaxia da leptospirose e da toxoplasmose, mediante a utilização de animais sentinela em áreas de vulnerabilidade socioeconômica, com base em estudo sorológico. Um total de 336 amostras de sangue foi obtido da população canina de um distrito urbano que apresenta altos índices de pobreza e ausência de saneamento básico. Os resultados foram analisados com base em animais sororreagentes e georreferenciados por mapeamento espacial de Kernel. Um total de 43,3% (52/120) dos animais foi sororreagente para anti-Leptospira spp., com predominância de Canicola (35/52 = 67,3%) e Pirogenes (5/52 = 9,6%), enquanto 52,97% (178/336) dos cães foram sororreagentes para anti-Toxoplasma gondii. ABSTRACT Leptospirosis and toxoplasmosis are zoonoses of occupational character that are spread worldwide, mainly in tropical and socioeconomically vulnerable areas. Both diseases have vertebrate hosts, which spread the infectious agent to the environment and are potentially infectious to humans and can progress to fatal cases. The present study reinforces the importance of One Health for the control and prophylaxis of leptospirosis and toxoplasmosis using sentinel animals in areas of socioeconomic vulnerability based on a serological study. RESUMO Utilizaram-se quatro garanhões nos meses de janeiro, abril, julho e outubro de 2016, em dois protocolos, em que: GI (n=4; 5mL de solução salina, i.v.) e GII (n=4; 5000UI de hCG, i.v.) e subdividiram-se esses protocolos em ciclos (C1 e C2), seguindo o esquema crossover, sendo: CI=animais 1 (GI) e 2 (GII), avaliados nos dias D1, D3 e D5, e animais 3 (GI) e 4 (GII), em D2, D4 e D6; CII= animais 1 (GII) e 2 (GI), em D1, D3, D5, e animais 3 (GII) e 4 (GI), em D2, D4 e D6. Realizou-se o tratamento em D1 e D2 de cada ciclo e a ultrassonografia testicular no modo color Doppler e Doppler espectral, uma hora antes de cada coleta de sêmen e imediatamente após. Avaliou-se: número de reflexo de Flehmen, de montas sem ereção, início da monta, tempo de reação à ereção e total da monta e análises de qualidade seminal. Estatisticamente foram utilizados os testes de qui-quadrado e ANOVA. Não houve diferenças estatísticas (P>0,05) entre os parâmetros analisados. Concluiu-se que uma única dose de 5000UI de hCG em garanhões não causou alterações significativas nos parâmetros avaliados em diferentes estações do ano.

ABSTRACT Four stallions were used in January, April, July and October 2016 in two protocols: GI (n=4; 5ml saline, iv) and GII (n=4; 5000 hCG, iv), and these protocols were subdivided into cycles (C1 and C2) following the cross over scheme, as follows: CI=animal 1 (GI) and 2 (GII) evaluated on days D1, D3 and D5 and animal 3 (GI) and 4 (GII) at D 2, D 4 and D 6; CII=animal 1 (GII) and 2 (GI) at D1, D3, D5 and animal 3 (GII) and 4 (GI) at D2, D4 and D6. Treatment was performed on D1 and D2 of each cycle and testicular ultrasound in color Doppler and spectral Doppler mode, one hour before each semen collection and immediately after. We evaluated: Flehmen's reflex number, mounts without erection, start of the mount, reaction time to erection and total mount and seminal quality analyzes. Statistically, the Chi-square and ANOVA tests were used. There were no statistical differences (P>0.05) between the analyzed parameters. It was concluded that a single dose of 5000IU hCG in stallions did not cause significant changes in the parameters evaluated in different seasons of the year.

The influence of aging on memory has been extensively studied, but the importance of short-term memory and recall sequence has not. The objective of the current study was to examine the recall order of words presented on lists and to determine if age affects recall sequence. Physically and psychologically healthy male subjects were divided into two groups according to age, i.e., 23 young subjects (20 to 30 years) and 50 elderly subjects (60 to 70 years) submitted to the Wechsler Adult Intelligence Scale-Revised and the free word recall test. The order of word presentation significantly affected the 3rd and 4th words recalled (P < 0.01; F = 14.6). In addition, there was interaction between the presentation order and the type of list presented (P < 0.05; F = 9.7). Also, both groups recalled the last words presented from each list (words 13-15) significantly more times 3rd and 4th than words presented in all remaining positions (P < 0.01). The order of word presentation also significantly affected the 5th and 6th words recalled (P = 0.05; F = 7.5) and there was a significant interaction between the order of presentation and the type of list presented (P < 0.01; F = 20.8). The more developed the cognitive functions, resulting mainly from formal education, the greater the cognitive reserve, helping to minimize the effects of aging on the long-term memory (episodic declarative).

Soros de 358 javalis (Sus scrofa), criados em sistema de semiconfinamento em propriedades do estado de São Paulo, foram coletados entre 1998 e 2000 e testados para anticorpos contra o vírus da doença de Aujeszky (VDA), pela técnica de soroneutralização (SN) e ensaio imunoenzimático (ELISA). Foram detectados animais soropositivos em três das sete propriedades analisadas. Do total de javalis testados, 30,7% apresentaram anticorpos neutralizantes contra o VDA, com variação de 25,2% a 100% nas propriedades com animais sororreagentes. O ELISA detectou menor número de sororeagentes (19,3%), sendo a sensibilidade e a especificidade 57,3% e 97,6%, respectivamente, e a correlação observada de 85,2% (P<0,0001). Os resultados mostram que a infecção pelo vírus da doença de Aujeszky ocorre em criações de javalis no estado de São Paulo, e compromete o sucesso de um futuro programa de erradicação da doença na região.

Serum samples collected from 358 wild boars (Sus scrofa) in breeding farms in São Paulo, southeast Brazil, from 1998 to 2001, were tested for antibodies against pseudorabies virus (PRV) by means of serum neutralization (SN) and enzyme-linked immunobsorbent assay (ELISA). Seropositive animals were detected in three of seven herds analyzed. Overall seroprevalence as assessed by SN was 30.7%, ranging from 25.2% to 100% for the herds that presented seropositive animals. Indirect ELISA detected lower seroprevalence (19.3%). Sensitivity and specificity of ELISA were equal to 57.3% and 97.6%, respectively. Agreement was equal to 85.2% (P<0.0001). These results showed that PRV infections occurred in farmed feral swine in southeast Brazil, and affect pseudorabies eradication program.

RESUMO Estudou-se o efeito da vacinação sobre o perfil sorológico de um rebanho Nelore fechado, que apresentava leptospirose endêmica. Amostras de soro de 100% das fêmeas (=12meses a 8 anos) foram examinadas (out/98) pela à Soroaglutinação Microscópica (SAM) frente aos sorovares Icterohaemorrhagiae, Canicola, Pomona, Grippotyphosa, Hardjo e Wolffi. Baseado nesses resultados os animais foram vacinados e subdivididos em 4 grupos: I) SAM +, vacinados; II) SAM +, não vacinados; III) SAM -, vacinados e IV) SAM -, não vacinados. Tres mL de uma vacina comercial contendo os mesmos 6 sorovares foram aplicados por via SC, em dez./99, com reforço em jan/00 e revacinação em out/00. Os soros foram colhidos em mar/00 e nov/00 e reexaminados pela SAM. A proporção de reagentes nas 3 colheitas foi de 62,5% (120/ 192), 77,4% (106/137) e 74% (91/123) com médias geométricas dos títulos de 2,59, 2,34 e 2,15 respectivamente. O perfil sorológico, considerando-se a freqüência de reagentes e títulos de aglutininas antes da vacinação, mostrou distribuição bimodal, com 39,6% negativos. Após vacinação, houve aumento na freqüência de reagentes, provavelmente, devido a soroconversão de novilhas e vacas negativas, entretanto não promoveu aumento significante na média geométrica de títulos para Hardjo e Wolffi, ao contrário, houve tendência decrescente. O monitoramento sorológico de um rebanho vacinado pode ser utilizado como ferramenta na avaliação do "status" protetor conferido pela vacina, onde o decréscimo progressivo dos títulos pós vacinação significaria eficiência vacinal. ABSTRACT The effect of vaccination on the serological profile of a closed Nelore beef cattle herd, endemically infected by leptospirosis was studied. Serum samples of 100% of the females (= 12 months to 8 years) were collected in Oct/1998 and examined by Microscopic Agglutination test (MAT) using 6 serovars of Leptospira spp.: icterohaemorrhagiae, canicola, pomona, grippotyphosa, hardjo and wolffi. After MAT, the animals were vaccinated and divided in 4 groups: I) hardjo/ wolffi MAT positive and vaccinated; II) hardjo/wolffi MAT positive and non-vaccinated; III) MAT negative and vaccinated and IV) MAT negative and non-vaccinated. The first vaccination was in Dec/1999 with reinforcement in Jan/2000; and revaccination in Out/2000. The 3 mL dose of a commercial vaccine containing the same 6 serovars was given subcutaneously. Serum samples were taken in Mar/2000 and Nov/2000 and examined the same way by MAT. The proportion of reagents in the 3 collections was: 62.5% (120/192), 77.4% (106/137) and 74% (91/123). Geometrical means titers were equal to 2.59, 2.34 and 2.15, respectively. Serological profile considering reagents frequency and pre-vaccination titers showed a bimodal distribution with 39.6% of negative results. After vaccination, there was an increase in the frequency of reactors, probably due to the seroconversion of negative heifers and cows, however, the vaccination did not elicit significant increase in the magnitude of the Hardjo or Wolffi titers. Instead, the geometric means in the whole herd, vaccinated and unvaccinated groups, showed a decreasing tendency. RESUMO A sensibilidade e especificidade de uma nested reação em cadeia da polimerase (nested PCR) para detecção do vírus da leucose bovina (VLB) foi avaliada em amostras de sangue bovino, como uma metodologia confiável para diagnóstico preciso de animais naturalmente infectados. O protocolo de nested PCR padronizado, baseia-se na identificação de fragmentos de 340 pb e 444 pb do DNA de VLB, produzidos pela utilização de 2 diferentes conjuntos de primers do gene env proviral. Este protocolo foi validado através de testes em 40 amostras de sangue bovino de um rebanho leiteiro VLB positivo e pela comparação dos resultados com o diagnóstico sorológico por imunodifusão em gel de ágar (IDGA) e por ensaio imunoenzimático (ELISA). O protocolo de nested PCR foi capaz de discriminar animais infectados dentre aqueles que foram diagnosticados sorologicamente como negativos. Esse ensaio mostrou ter aplicabilidade, também, a outros fluídos corporais ou amostras de tecidos. Investigações feitas em 40 amostras de sangue bovino mostraram que 37 animais foram positivos para VLB com ELISA, 36 com nested PCR e somente 25 com IDGA. O nested PCR demonstrou uma concordância nos resultados com o ELISA em até 95%. Esses resultados comprovaram que o nested PCR pode ser indicado para uma detecção precoce, sensível e direta do VLB em bovinos naturalmente infectados. Comparado com os testes sorológicos, esse método mostra-se mais adequado para a identificação de bovinos infectados possuindo títulos de anticorpo anti-VLB baixos, transientes ou ausentes. Além disso, esse ensaio pode ser usado para confirmar os resultados sorológicos duvidosos ou a infecção em bovinos assintomáticos. ABSTRACT A sensitive and specific nested polymerase chain reaction (nested PCR) for detection of bovine leukemia virus (BLV) in blood samples was evaluated as a reliable assay for accurate diagnosis of naturally infected animals under field conditions. The standardized nested PCR relies on the identification of BLV DNA fragments of 340 or 444 bp produced by applying 2 different pairs of primers sets to the proviral env gene. This protocol was validated by testing 40 bovine blood samples from a BLV-positive dairy herd, and by comparing results with serological diagnosis provided by agar-gel immunodiffusion (AGID) and enzyme-linked immunosorbent assay (ELISA). The nested PCR protocol was able to discriminate infected animals from that ones serodiagnosed as negative. This method also showed applicability for other body fluids or tissue samples. Investigation of 40 blood samples from a dairy herd showed that 37 animals were positive in ELISA, 36 in nested PCR, and only 25 in AGID. The nested PCR has demonstrated a good agreement in the results with ELISA up to 95%. These results demonstrated that nested PCR could be indicated for an early, sensitive and direct detection of BLV-infection in naturally infected cattle. Compared to the serological tests, this method proved to be the most suitable for identification of BLV-infected cattle with low, transient or without BLV-antibody titers. RESUMO A Artrite-encefalite dos caprinos (AEC) é uma doença infecciosa, multissistêmica, causada por um lentivírus, que acomete caprinos de todas as idades. As principais manifestações clínicas da doença são a leucoencefalomielite que acomete cabritos e a artrite mais freqüente nos animais adultos. Os pulmões e a glândula mamária também podem ser afetados pelo vírus causando, respectivamente, uma pneumonia crônica intersticial e uma mamite endurativa. Esta pesquisa teve como objetivo comparar as técnicas de imunodifusão em gel de ágar, ensaio imunoenzimático e imunofluorescência indireta, utilizadas como técnicas auxiliares no diagnóstico da Artriteencefalite dos caprinos, assim como avaliar a sensibilidade e a especificidade de cada uma das técnicas. ABSTRACT Caprine arthritis encephalitis (CAE) is an infectious multisystemic disease caused by a lentivirus, and affects goats at all ages. Main clinical signs of the disease are leukoencephalomyelitis, which affects kids and arthritis, more frequent in adult animals. Lungs and the mammary gland may also be affected by the virus, causing, respectively, chronic interstitial pneumonia and indurative mastitis. The objective of the present trial was to compare the techniques used to aid CAE diagnosis - agar gel immunodiffusion, immunoenzymatic assay and indirect immunofluorescence - as well as to evaluate their sensitivity and specificity. Sensitivity and specificity obtained in this trial for ELISA were 100% and 83.78%, respectively. Immunofluorescence technique presented sensitivity equal to 98.25% and specificity equal to 79.25%.
