RESUMO A pandemia de covid-19 gerou um numeroso volume de produções científicas com diferentes níveis de qualidade. A velocidade com que o conhecimento era produzido e compartilhado a nível mundial impôs à gestão em saúde o desafio de buscar meios de identificar as melhores evidências disponíveis para subsidiar suas decisões. Em resposta a este desafio, o Departamento de Ciência e Tecnologia do Ministério da Saúde do Brasil estabeleceu um serviço para elaborar e disponibilizar conhecimento científico abordando questões prioritárias de saúde pública no cenário da pandemia. Entre os temas abordados estão tratamentos medicamentosos, medidas não farmacológicas, testagem, reinfecção e resposta imunológica, imunização, fisiopatologia, síndrome pós-covid e eventos adversos. Neste artigo, discute-se os pontos fortes e lições aprendidas, bem como os desafios e perspectivas que fornecem um exemplo real sobre como disponibilizar as melhores evidências científicas, em tempo hábil e de forma oportuna, para auxiliar o processo decisório durante uma emergência em saúde pública.
ABSTRACT The COVID-19 pandemic generated a large volume of scientific productions with different quality levels. The speed with which knowledge was produced and shared worldwide imposed on health management the challenge of seeking ways to identify the best available evidence to support its decisions. In response to this challenge, the Department of Science and Technology of the Brazilian Ministry of Health started offering a service to produce and provide scientific knowledge addressing priority public health issues in the pandemic scenario. Drug treatments, non-pharmacological measures, testing, reinfection and immunological response, immunization, pathophysiology, post-COVID syndrome and adverse events are among the topics covered. In this article, we discuss the strengths and lessons learned, as well as the challenges and perspectives that present a real example of how to offer the best scientific evidence in a timely manner in order to assist the decision-making process during a public health emergency.

Resumo Introdução O smartphone, dispositivo móvel e popular, tornou-se atrativo pela facilidade de utilização e multifuncionalidade. Seu uso prolongado, com flexão anterior do pescoço e movimentos repetitivos dos membros superiores, contribui para sintomas musculoesqueléticos. Objetivo Avaliar as alterações da postura cervical pelo uso do smartphone em adultos, bem como verificar a associação da postura com fatores relacionados à saúde. Métodos Estudo transversal, em duas universidades em Fortaleza, Ceará, Brasil, entre 2018 e 2019. Participaram 769 adultos (>18 anos) que usavam rotineiramente o smartphone. Foram coletadas variáveis socioeconômicas, condições de saúde e uso do smartphone. Realizou-se a avaliação do alinhamento postural da cervical, sendo mensurado o alinhamento vertical da cabeça (AVC) pela fotogrametria, na posição anatômica (baseline) e digitando no smartphone. Resultados A idade média foi de 23 anos (± 6,7), com maior proporção do sexo feminino (72,1 %; n = 559) e média de 7,9h (± 4,4) utilizando o dispositivo. O uso do smartphone gerou anteriorização de cabeça relacionada ao sexo masculino (p < 0,05 pelo ΔAVC), tempo de uso do dispositivo (p < 0,05 pela ΔAVC), incapacidade funcional na região cervical (p < 0,05 pela ΔAVC) e não dormir bem (p = 0,019 pela ΔAVC lado D). Abstract Introduction The smartphone, a popular mobile device, has become attractive because it is easy to use and due to it multifunctionality. Its prolonged use, with anterior flexion of the neck and repetitive movements of the upper limbs, contributes to musculoskeletal symptoms. Objective To evaluate changes in cervical posture due to smartphone use in adults, as well as verify the association of posture with health-related factors. Methods Cross-sectional study, carried out at two universities in Fortaleza, Ceará, Brasil, between 2018 and 2019. A total of 769 adults (>18 years old) who routinely used smartphones participated. Data on socioeconomic variables, health conditions and smartphone use were collected. The cervical postural alignment was assessed, with the vertical head alignment (VHA) being measured using photogrammetry, in the anatomical position (baseline) and while typing on the smartphone. Results The mean age was 23 years (± 6.7), with a higher proportion of females (72.1%; n = 559) and an average of 7.9h (± 4.4) using the device. Smartphone use led to the forward head movement related to males (p < 0.05 by ΔVHA), time spent using the device (p < 0.05 by ΔVHA), functional disability in the cervical region (p < 0.05 by ΔVHA) and not sleeping well (p = 0.019 by ΔVHA on the R side). The corrosion resistance of chromium-plated steel sheets produced on an industrial scale was investigated. Two parameters of the production process were considered: pickling and chemical treatment. The specimens were produced at the industrial plant considering two different pickling conditions and two different chemical treatments: non-electrolytic and electrolytic pickling and non-electrolytic and electrolytic passivation respectively. Accelerated corrosion tests in the humid chamber, microscopy analyses, analyses of roughness and content of trivalent chromium in the passivation film and analyses of the content of metallic chromium in the coating by coulometry technique were performed. The results showed a low corrosion resistance of the materials produced with the non-electrolytic passivation, regardless of the pickling condition. The highest degree of protection of the base steel was achieved for the specimen produced by using the electrolytic route for both pickling and chemical treatment steps. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract Areas of high concentration of seal carcasses have been observed in localized areas of James Ross Island, Antarctica. Such carcasses show an unusual vegetation development, in a semi-arid area with bare soils under intense winds, high salinity and sandy texture. We investigated carcasses of seals around a lake in James Ross Island, with four different stages of decomposition, with three replicates: Seal (S01), with recently mummified carcasses; S02, with partially degraded carcasses; S03, with broken carcasses with partially degraded exposed bones, and S04, with completely broken, scattered skeletons. The vegetation showed a maximum degree of development in carcasses at stages S02 and S03, with the environment between the skin and the skeleton as the preferred place for vegetation establishment. The chemical alteration was greater with increasing carcass decomposition but reduced with the spreading and final decomposition of the bones, with anomalous values observed only in the vicinity of the carcasses. It is concluded that the presence of carcasses of seals, concentrated in wet places, even in a semi-desert climate, represent important oases of nutrients, with a combination of physical and chemical effects throughout the decomposition process that favor plant establishment and succession. RESUMO Objetivo: Este artigo analisa o cenário brasileiro do trabalho mediado por plataformas digitais (digitrab). Originalidade/valor: O digitrab representa o arranjo laboral alternativo ao modelo de emprego tradicional de longo termo que mais tem crescido nos últimos anos. Entretanto, não conhecemos com clareza esse cenário, nem em termos de quantidade nem de diversidade de produtos e serviços ofertados, muito menos quanto ao volume de indivíduos envolvidos. O presente artigo apresenta informações e reflexões que colaboram para a compreensão desses dados no Brasil. Design/metodologia/abordagem: Trata-se de um estudo analítico-exploratório. Artigos científicos foram utilizados para discutir as influências tecnológicas, políticas e socioeconômicas que favoreceram a emergência do digitrab e seus efeitos no mundo do trabalho e nos trabalhadores. Para sustentar tal discussão, foi feita uma análise documental de sites e aplicativos móveis (apps) que permitiu um mapeamento quali-quantitativo acerca das plataformas digitais de intermediação ativas no Brasil atualmente. Resultados: Foram identificadas mais de 100 empresas atuantes no país que oferecem 11 tipos de serviços nessa modalidade de negócio, com indicação de tendência de crescimento. Observou-se também um movimento de diversificação, absorvendo profissionais de diferentes tipos de formação e níveis educacionais. Tal crescimento se alinha à tendência político-econômica de incentivo ao afrouxamento das relações de trabalho, que individualiza e informaliza os vínculos, enfatizando a neces-sidade de debate sobre a regulamentação desse arranjo de trabalho e de mais pesquisas que se debrucem a investigar suas repercussões sobre a saúde do trabalhador, considerando, ainda, a heterogeneidade presente nessa nova classe. ABSTRACT Purpose: This article analyzes the Brazilian scenario of work mediated by digital platforms (digiwork). Originality/value: Digiwork represents an alternative work arrangement to the traditional long-term employment model, with the highest growth rate in recent years. Details of this scenario are still unclear regarding the quantity and diversity of the offered products and services and the number of individuals involved. This article presents information and reflections that contribute to understanding these data in Brazil. Design/methodology/approach: This is an analytical-exploratory study. Scientific articles were used to discuss technological, political, and socioeconomic influences that supported the emergence of digiwork and its effects on the labor market and the workforce. In order to support this discussion, a documental analysis of websites and mobile application software was carried out, allowing for a qualitative-quantitative mapping of Brazil’s current digital intermediation platforms. Findings: More than 100 companies operating in the country were identified as offering 11 types of services in this business, indicating a growing trend. A diversification movement was also observed, absorbing professionals with different kinds of training and schooling levels. Abstract Although widely used in medicine, separation technology, and other fields, the effects of cyclodextrins on the activities of phosphoryl transfer enzymes have not been previously evaluated. In vivo studies evaluated the function of cyclodextrins as active compounds. Despite the use of cyclodextrins as active compounds, the effects of cyclodextrins on hepatic and renal tissues remain to be fully elucidated. The primary objective of this study was to evaluate the effects of β- cyclodextrins, methyl-β-cyclodextrin (M-β- cyclodextrins), and (2-hydroxypropyl)-β-cyclodextrin (HP-β-cyclodextrins) on enzyme activities regulating the maintenance of energy homeostasis in the kidney and liver tissues in relation to toxicity. Serum levels of liver and kidney markers were measured, and oxidative stress parameters were assessed. After 60-day treatments, we observed that the administration of β-cyclodextrins and M-β-cyclodextrins inhibited the hepatic activity of pyruvate kinase, an irreversible enzyme within the glycolytic pathway. Additionally, administration of HP-β-cyclodextrins inhibited creatine kinase activity and increased the total sulfhydryl content in kidneys. Here, we demonstrated for the first time that β-cyclodextrins, M-β-cyclodextrins, and HP-β-cyclodextrins cause bioenergetic dysfunction in renal and hepatic tissues. Resumo Fundamento Estudos anteriores revelaram alto risco de eventos tromboembólicos arteriais e venosos como consequência de danos virais diretos do SARS-CoV-2 em células endoteliais e um meio procoagulante devido ao aumento de biomarcadores como o D-dímero, fibrinogênio, fator VIII. Foram realizados ensaios controlados randomizados de terapias antitrombóticas em pacientes internados, no entanto, poucos estudos avaliaram o papel da tromboprofilaxia no ambiente ambulatorial. Objetivo Avaliar se a profilaxia antitrombótica com rivaroxabana reduz o risco de eventos trombóticos venosos ou arteriais, suporte ventilatório invasivo e morte em pacientes ambulatoriais com COVID-19. Métodos O estudo CARE é um ensaio randomizado, aberto, multicêntrico e controlado por rivaroxabana 10 mg uma vez por dia durante 14 dias ou tratamento local padrão isolado, para a prevenção de resultados adversos, registrado no Clinicaltrials.gov (NCT04757857). Os critérios de inclusão são adultos com infecção confirmada ou suspeita do SARS-CoV-2, com sintomas leves ou moderados, sem indicação de hospitalização, no prazo de 7 dias após o início dos sintomas e um fator de risco de complicação da COVID-19 (>65 anos, hipertensão, diabetes, asma, doença pulmonar obstrutiva crônica ou outras doenças pulmonares crônicas, tabagismo, imunossupressão ou obesidade). O desfecho primário composto inclui tromboembolismo venoso, necessidade de ventilação mecânica invasiva, eventos cardiovasculares agudos maiores e mortalidade no prazo de 30 dias após a randomização, sendo avaliado segundo o princípio da intenção de tratar. Todos os pacientes assinaram termo de consentimento. Foi estabelecido um nível de significância de 5% para todos os testes estatísticos. Resultados Os principais desfechos trombóticos e hemorrágicos, hospitalizações e mortes serão avaliados centralmente por um comitê de eventos clínicos independente, sob a condição cega para a alocação dos grupos de tratamento. Abstract Background Previous studies have demonstrated a high risk of arterial and venous thromboembolic events as a consequence of direct viral damage to endothelial cells by SARS-CoV-2 and a procoagulant milieu due to increased biomarkers, such as D-dimer, fibrinogen, and factor VIII. Although randomized controlled trials of antithrombotic therapies have been conducted in hospitalized patients, few have evaluated the role of thromboprophylaxis in an outpatient setting. Methods The COVID Antithrombotic Rivaroxaban Evaluation (CARE) study, a multicenter, randomized, open-label, controlled trial of rivaroxaban 10 mg once daily for 14 days or local standard treatment alone to prevent adverse outcomes, is registered in clinicaltrials.gov (NCT04757857). The inclusion criteria are adults with confirmed or suspected SARS-CoV-2 infection and mild or moderate symptoms without indication for hospitalization, within 7 days of symptom onset, and 1 risk factor for COVID-19 complication (> 65 years, hypertension, diabetes mellitus, asthma, chronic obstructive pulmonary disease or other chronic lung diseases, smoking, immunosuppression, or obesity). The primary composite endpoint, which includes venous thromboembolism, invasive mechanical ventilation, major acute cardiovascular events, and mortality within 30 days of randomization, will be assessed according to the intention-to-treat principle. All patients will provide informed consent. A significance level of 5% will be used for all statistical tests. Results Major thrombotic and bleeding outcomes, hospitalizations, and deaths will be centrally adjudicated by an independent clinical events committee blinded to the assigned treatment groups. Conclusion The CARE study will provide relevant and contemporary information about the potential role of thromboprophylaxis in outpatients with COVID-19. Abstract Natural resources are depleted in fragmented landscapes that have their vegetation also altered. As a result, the microhabitat diversity and the composition and distribution of local species are affected. In this study, we evaluated the small mammals’ community diversity, composition and microhabitat selection in two Atlantic Forest fragments, in an ecotone area with the Pampa biome, southern Brazil. We recorded five rodents (Akodon montensis, Oligoryzomys nigripes, Sooretamys angouya, Juliomys pictipes and the exotic Rattus rattus) and one marsupial (Didelphis albiventris). Both fragments were dominated by the generalist rodent A. montensis. Akodon montensis and O. nigripes showed similar habitat preferences: ground covered by rocks and higher values of vegetation obstruction. Sooretamys angouya preferred places with higher abundance of trees. Fruit availability was important for A. montensis and D. albiventris, highlighting the importance of this food resource for local wildlife, and the potential role of these species as seed predators and dispersers. Small species richness, the presence of an exotic species and high dominance level suggest that the study area is highly degraded.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

ABSTRACT The thermal and luminous conditions in aviaries affect the production and quality of eggs. Thus, the aim of the present study was to evaluate the thermal and luminous environment of commercial laying hen aviaries equipped with compact fluorescent (CF) and light-emitting diode (LED) lamps and their influence on egg production and quality. The study was carried out in two commercial laying hen facilities with similar construction features and management. The first facility was illuminated with CF lamps and the second with LED lamps. The thermal environment was assessed via air temperature, air relative humidity, and enthalpy. The evaluated lamps provided a similar thermal environment in both facilities. The aviary equipped with LED lamps showed higher illuminance than the one equipped with a CF lamp. Production performance was evaluated through feed intake (FI), feed conversion (FC), mortality (M), and laying percentage (LP). Specific gravity (SG) was used to evaluate eggshell quality. The variables related to production performance and eggshell quality of laying hens raised in aviaries equipped with LED lamps were statistically equal (F-test, p > 0.05) to those raised in aviaries equipped with CF lamps. These results indicate that the replacement of CF lamps with LED lamps does not adversely affect the yield and quality of eggs.

Association between SNP rs527616 in lncRNA AQP4-AS1 and susceptibility to breast cancer in a southern Brazilian population

Abstract Breast cancer (BC) is the leading cause of death by this disease in women worldwide. Among the factors involved in tumorigenesis, long non-coding RNAs (lncRNAs) and their differential expression have been associated. Differences in gene expression may be triggered by variations in DNA sequence, including single nucleotide polymorphisms (SNPs). In the present study, we analyzed the rs527616 (C>G), located in the lncRNA AQP4-AS1, using PCR-SSP in 306 BC patients and 312 controls, from a Brazilian population. In the BC group, the frequency found for CG heterozygotes was above the expected and the overdominant model is the best one to explain our results (OR: 1.70, IC 95%: 1.23-2.34, P<0.001). Furthermore, the SNP were associated with age at BC diagnosis and the risk genotype more frequent in the older age group. According to TCGA data, AQP4-AS1 is down-regulated in BC tissue, and the overexpression is associated with better prognoses, including Luminal A, HER2-, stage 1 of disease and smaller tumor. In conclusion, the CG genotype is associated with increased susceptibility in the southern Brazilian population. This SNP is mapped in the lncRNA AQP4-AS1, showing differential expression in BC samples. Based on these results, we emphasize the potential of the role of AQP4-AS1 in cancer.

Meliaceae representatives are economically important in several aspects including the production of highly prized woods (mahogany, cedar, etc.), constituents for cosmetics, and insecticides. The present study aimed to verify the chemical composition as well as leishmanicidal and cytotoxic potential of essential oils from leaves of two different populations of Guarea macrophylla collected at cities of São Paulo (population I) and Cubatão (population II), São Paulo State, Brazil. Chemically, the oils showed the predominance of sesquiterpenes: cis-β-guaiene, bicyclogermacrene, viridiflorol, and isolongifolan-7α-ol from population I and α-copaene, E-caryophyllene, cis-β-guaiene, and γ-amorphene from population II. In vitro antileishmanial activity against promastigote forms of Leishmania (L.) amazonensis of essential oils was evaluated and displayed 50% effective concentration (EC50) values ranging from 11.8 to 20.5 µg mL-1. Furthermore, toxicity against peritoneal macrophages of BALB/c mice was observed, with 50% cytotoxic concentration (CC50) ranging from 17.7 to > 100 µg mL-1. Multivariate statistical analysis revealed the influence of each constituent of the oils against L. amazonensis being 1,10-di-epi-cubenol, α-amorphene, E-caryophyllene, isopimara-7,15-diene, and β-elemene associated with the antileishmanial potential.

Abstract: Electrochemotherapy (ECT) is a technique that combines chemotherapy with local application of specific electric pulses with the aim of increasing the permeability of the plasma membrane in a reversible way, improving the influx of chemotherapeutic drugs into the cytoplasm and potentiating their cytotoxic effects. This technique has broadened the range of possible treatments for oncological patients, either on its own or as adjuvant to surgical procedures. It is especially useful in tumors located in regions with only a small surgical safety margin, such as the limb extremities, skull, oral cavity, neck and perianal region, among others. ECT makes it feasible to perform procedures more conservatively, or even to perform otherwise infeasible procedures, by expanding the margins without removing healthy tissues. The objective of this paper is to provide a brief bibliographic review of the principles, applications and future possibilities of electrochemotherapy, helping to disseminate pertinent information about this relatively new technique for the treatment of cancer.
