RESUMO: No Brasil, a Mata Atlântica vem sofrendo com o desmatamento, que tem impactado sua flora, fauna e microbiota. No entanto, a diversidade fúngica presente nesses ambientes é pouco conhecida e estudada. Neste trabalho, um total de 90 amostras de 45 aves silvestres (45 penas e 45 fezes) foram coletadas na Ilha da Marambaia, Sudeste do Brasil. Fungos filamentosos isolados dessas amostras foram identificados por meio de características macroscópicas e microscópicas. Alguns isolados foram identificados por biologia molecular usando a técnica de PCR. Foram identificados Acremonium, Alternaria, Aspergillus, Cunninghamella, Curvularia, Eurotium, Fusarium, Geotrichum, Neosartorya, Pestalotia, Paecilomyces, Penicillium, Rhizopus, Mucor e Syncephalastrum. Esses resultados indicam a presença de espécies de fungos saprofíticos nas penas e fezes de aves silvestres do local de captura. Novos estudos devem ser realizados a fim de elucidar se o perfil da micobiota se modifica com a antropização e se interfere na saúde das aves e na recuperação ambiental. ABSTRACT: In Brazil, the Atlantic Forest has been suffering from deforestation, which has had impacts on its flora, fauna, and microbiota. However, the fungal diversity present in these environments is little known and studied. In this study, a total of 90 samples of 45 wild birds (45 feathers and 45 feces) were collected in Ilha da Marambaia, southeastern Brazil. Filamentous fungi isolated from these samples were identified through macroscopic and microscopic characteristics. Some isolates were identified by molecular biology using the PCR technique. Acremonium, Alternaria, Aspergillus, Cunninghamella, Curvularia, Eurotium, Fusarium, Geotrichum, Neosartorya, Pestalotia, Paecilomyces, Penicillium, Rhizopus, Mucor and Syncephalastrum were identified. These results indicate the presence of saprophytic fungi species in the feathers and feces of wild birds of the capture site. Further studies should be conducted to elucidate if the mycobiota profile modifies with anthropization and if it interferes with bird health and environmental recovery. Apurinic/apyrimidinic endonuclease 1/redox factor-1 (APE1/REF-1) is a multifunctional protein acting on cellular signaling pathways, including DNA repair and redox activities. APE1/REF-1 has emerged as a target for cancer therapy, and its role in breast cancer models would reveal new strategies for cancer therapy. APX2009 is a specific APE1/REF-1 redox inhibitor whose anticancer properties have not been described in breast cancer cells. Here, we investigated the effect of the APX2009 treatment in the breast cancer cell lines MDA-MB-231 and MCF-7. Breast cancer cell lines were cultured, and WST1 and colony formation assays were performed to evaluate cell proliferation. Annexin V-FITC/7-AAD and LDH-Glo™ assays were performed to evaluate cell death. The wound healing assay and Matrigel transwell assay were performed after APX2009 treatment to evaluate the cellular migration and invasion processes, respectively. Our findings demonstrated that APX2009 treatment decreased breast cancer cell proliferative, migratory, and invasive properties. Furthermore, it induced apoptosis in both cell lines. Our study is the first to show the effects of APX2009 treatment on apoptosis in a breast cancer cell. Therefore, this study suggested that APX2009 treatment is a promising anticancer molecule for breast cancer. Screener, a board game supplemented with online resources, was introduced and distributed by the Brazilian Society of Pharmacology and Experimental Therapeutics to postgraduate programs as an instructional tool for the process of drug discovery and development (DDD). In this study, we provided a comprehensive analysis of five critical aspects for evaluating the quality of educational games, namely: 1) description of the intervention; 2) underlying pedagogical theory; 3) identification of local educational gaps; 4) impact on diverse stakeholders; and 5) elucidation of iterative quality enhancement processes. We also present qualitative and quantitative assessments of the effectiveness of this game in 11 postgraduate courses. We employed the MEEGA+ online survey, comprising thirty-three close-ended unipolar items with 5-point Likert-type response scales, to assess student perceptions of the quality and utility of Screener. Based on 115 responses, the results indicated a highly positive outlook among students. In addition, we performed a preliminary evaluation of learning outcomes in two courses involving 28 students. Pre- and post-quizzes were applied, each consisting of 20 True/False questions directly aligned with the game's content. The analysis revealed significant improvement in students' performance following engagement with the game, with scores rising from 8.4 to 13.3 (P<0.0001, paired t-test) and 9.7 to 12.7 (P<0.0001, paired t-test). These findings underscore the utility of Screener as an enjoyable and effective tool for facilitating a positive learning experience in the DDD process. Anthelmintic intoxication in small ruminants diagnosed in Northeastern Brazil

RESUMO: Descrevem-se os achados clínicos, epidemiológicos e anatomopatológicos das intoxicações por anti-helmínticos em pequenos ruminantes diagnosticadas no Hospital Veterinário da Universidade Federal de Campina Grande, Patos, Paraíba, durante o período de janeiro de 2000 a dezembro de 2022. Foram realizadas 1.928 necropsias de pequenos ruminantes, sendo 1.032 caprinos e 896 ovinos. Destes, foram identificados quatro surtos de intoxicação por anti-helmínticos nessas espécies: um surto de intoxicação por ivermectina em ovino, um surto de disofenol em caprinos e ovinos e dois surtos por nitroxinil em caprinos. Essas intoxicações representaram 0,78% e 0,23% das doenças de caprinos e ovinos diagnosticadas na rotina, respectivamente. Na intoxicação por ivermectina o animal apresentava sinais clínicos neurológicos e não foram observadas alterações macroscópicas e histopatológicas. Nos surtos de disofenol e nitroxinil, os animais demonstravam anorexia, hipertermia, taquicardia, tremores, dispneia, incoordenação e rigidez de membros. Macroscopicamente observou-se lesões hepáticas e renais associadas a alterações circulatórias como hemorragias e edemas. Microscopicamente observou-se necrose e degeneração de hepatócitos e das células epiteliais tubulares associadas a hemorragia e congestão. A prevalência dessas intoxicações em pequenos ruminantes é considerada baixa no Semiárido nordestino, entretanto, os surtos podem ocorrer com alta letalidade nos rebanhos. O principal fator de risco para a ocorrência das intoxicações foi a sobredosagem aliada aos demais fatores como idade dos animais, temperatura ambiental alta, exercício constante e escore corporal baixo, que provocaram o aumento da toxicidade dos anti-helmínticos. ABSTRACT: The clinical, epidemiological and anatomopathological findings of anthelmintic poisoning in small ruminants diagnosed at the Veterinary Hospital of the Federal University of Campina Grande, Patos, Paraíba, from January 2000 to December 2022 are described. A total of 1,928 necropsies were carried out on small ruminants, of which 1,032 were goats and 896 sheep. Four outbreaks of anthelmintic poisoning were identified in these species: one outbreak of ivermectin in sheep, one outbreak of disophenol in goats and sheep and two outbreaks of nitroxinil in goats. These intoxications accounted for 0.78% and 0.23% of goat and sheep diseases diagnosed routinely. In the case of ivermectin poisoning, the animal had neurological clinical signs, and no macroscopic or histopathological alterations were observed. In the outbreaks of disophenol and nitroxinil, the animals showed anorexia, hyperthermia, tachycardia, tremors, dyspnea, incoordination and limb rigidity. Macroscopically, there were liver and kidney lesions associated with circulatory alterations, such as hemorrhages and edema. Microscopically, necrosis and degeneration of hepatocytes and tubular epithelial cells associated with hemorrhage and congestion were observed. The prevalence of these intoxications in small ruminants is considered low in the Semi-arid Northeast, but outbreaks can occur with high lethality in herds. The main risk factor for the occurrence of intoxications was overdose, combined with other factors such as the age of the animals, high environmental temperature, constant exercise and low body score, which increased the toxicity of anthelmintics. RESUMO: O trinca-ferro Saltator similis (d’Orbigny & Lafresnaye, 1837) é um pássaro muito valorizado por criadores de aves e pelo tráfico ilegal de animais silvestres. Os cromistas coccídios são agentes etiológicos da coccidiose, a qual pode ser assintomática e enzoótica em populações de aves quando as condições ambientais são favoráveis, mas surtos epizoóticos com doença grave podem ocorrer como resultado de mudanças ambientais. Nesse contexto, este artigo relata um caso de coccidiose em um espécime juvenil de vida livre de S. similis capturado no Parque Nacional de Itatiaia, no sudeste do Brasil. Foram observados sinais clínicos típicos de coccidiose, associados a diarreia mucóide esverdeada contendo alta densidade de oocistos. Três Isospora spp. foram especificamente identificadas em 14 defecações coletadas durante um período de duas horas. Isospora saltatori (Berto, Balthazar, Flausino & Lopes, 2008) foi a espécie com maior densidade e positividade durante a maior parte do período de coleta. Por fim, este artigo discute a importância da avaliação contínua da saúde das aves como bioindicadores, e de seus biomarcadores ecológicos coccídios, como uma das estratégias para avaliação do estado de conservação do Parque Nacional de Itatiaia. ABSTRACT: The green-winged saltator Saltator similis (d’Orbigny & Lafresnaye, 1837) is a passerine bird highly valued by bird breeders and a target of illegal wildlife trafficking. Coccidian chromists are etiological agents of coccidiosis, which may be asymptomatic and enzootic in bird populations when environmental conditions are favorable. However, epizootic outbreaks with severe disease can occur due to environmental changes. In this context, this study reports a case of coccidiosis in a free-living juvenile specimen of S. similis captured at the Itatiaia National Park in southeastern Brazil. Typical clinical signs of coccidiosis were observed, and it was associated with greenish mucoid diarrhea containing a high density of coccidian oocysts. Three Isospora spp. were specifically identified in 14 fecal samples (fecal droplets) collected over two hours. Isospora saltatori (Berto, Balthazar, Flausino & Lopes, 2008) had the highest density and positivity throughout most of the collection period. Finally, this paper discusses the importance of the continuous evaluation of the health of birds as bioindicators and their coccidian ecological biomarkers as one of the strategies for evaluating the conservation status of Itatiaia National Park. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract The objective of this study was to evaluate the effects and economic viability of diets containing different levels of antibiotic and buriti oil (BO) on performance, carcass and cut yields, and relative weight of organs of broilers. A total of 432 one- to 42-day-old male chicks were distributed in a completely randomized experimental design with six treatments, each consisting of six replicates of 12 birds. The treatments consisted of one diet with antibiotic without BO, one diet without antibiotic (DWA) without BO, and four DWA containing increasing levels of BO (0.2, 0.4, 0.6, and 0.8%). Average weight and weight gain (WG) of broilers fed with DWA + BO were similar to those of birds fed control diet. Feed intake and feed conversion (FC) were not different among treatments. Relative weight of pancreas linearly increased in the birds fed diets containing BO. The inclusion of 0.45 and 0.40% of BO in the diets promoted the improvement of WG and FC, respectively. Cost of feed management, ratio, gross margin, and gross income did not differ among treatments. It was concluded that the inclusion of 0.45% of BO in diets without antibiotics is economically feasible and allows recovering the performance of broilers. RESUMO: A raiva é uma doença viral fatal que afeta animais e o homem, estando disseminada entre os sistemas ecológicos nacionais. Descrevem-se os achados anatomopatológicos e imuno-histoquímicos de um caso de raiva em um jaguarundi (Herpailurus yagouaroundi), encontrado em área peridomiciliar rural, após apresentar agressividade e ataque a dois animais. Microscopicamente, observou-se meningoencefalite linfoplasmocítica associada a corpúsculos de inclusão intracitoplasmáticos eosinofílicos em neurônios, gliose e neuronofagia. O diagnóstico de raiva foi estabelecido pelos achados patológicos e confirmado por imuno-histoquímica e qPCR. A ocorrência de raiva em felinos silvestres do gênero Puma só foi descrita por meio de exames sorológicos. Os achados patológicos são essenciais para o diagnóstico e elucidação da participação desta espécie no ciclo silvestre da doença, bem como na manutenção e conservação do equilíbrio da fauna silvestre. ABSTRACT: Rabies is a fatal viral disease that affects animals and humans and is widespread among national ecological systems. We describe the anatomopathological and immunohistochemical findings of a case of rabies in a jaguarundi (Herpailurus yagouaroundi) found in a rural peri-domestic area after showing aggression and attacking two animals. Histopathology of brain tissue showed lymphoplasmacytic meningoencephalitis associated with intracytoplasmic eosinophilic inclusion bodies in neurons, gliosis and neuronophagia. The diagnosis of rabies was based on the lesions and confirmed by immunohistochemistry and qPCR. In Brazil, rabies in wild felines of the genus Puma has only been described in animals through serological examinations. The pathological findings are essential for the diagnosis and elucidation of the participation of this species in the wild cycle of the disease, as well as in the maintenance and conservation of the balance of wild fauna. High-intensity interval training (HIIT) has stood out as a treatment for obesity, leading to adaptations of the cardiovascular system and reducing body adiposity. In addition, the search for alternative therapies for weight loss has intensified. The administration of Hibiscus sabdariffa (Hs) has been described as an efficient supplement in weight loss and in the treatment of metabolic changes associated with obesity. In this context, the objective was to investigate the effects of the association of Hs and HIIT on metabolic adaptations and lipid metabolism in obese rats. Wistars rats were subjected to obesity and subsequently randomized into 4 groups: obese (Ob), obese + HS (ObHs), obese + HIIT (ObHIIT), and obese + HS + HIIT (ObHsHIIT). For 8 weeks, ObHs and ObHsHIIT rats received Hs extract daily (150 mg/kg of body weight) and trained groups (ObHIIT and ObHsHIIT) were subjected to a HIIT program on a treadmill. Nutritional profile, glycemic curve, biochemical profile, and liver glycogen were determined. HIIT decreased caloric intake, feed efficiency, body adiposity, total body fat, and body weight gain, associated with improvements in physical performance parameters and a smaller glycemic curve and area. Hs had a hepatoprotective effect, reducing alkaline phosphatase values, but its effects were more pronounced when associated with HIIT. Therefore, the combination of treatments promoted a reduction in food consumption and body adiposity, as well as an improvement in physical performance and glycemic profile, but without changes in lipid metabolism. RESUMO A criptococose é uma importante micose sistêmica de distribuição mundial, que acomete homens, animais domésticos e silvestres. A infecção é adquirida pela inalação do fungo ou de basidiósporos presentes no ambiente e pode manifestar-se por lesão nasal ou ocular localizada, por comprometimento pulmonar, ou, menos comumente, como meningoencefalomielite. O diagnóstico definitivo nos casos neurológicos é desafiador, assim como seu tratamento. Este trabalho tem como objetivo relatar um caso de meningoencefalomielite e pneumonia por criptococose diagnosticado por meio de histopatologia, em um felino doméstico. ABSTRACT Cryptococcosis is an important systemic mycosis with worldwide distribution that affects men and also domestic and wild animals. The infection is acquired by inhalation of the fungus or basidiospores present in the environment and may manifest as localized nasal or ocular injury, pulmonary illness or, less commonly, as meningoencephalomyelitis. Long-term treatment is necessary, and the prognosis is reserved. This paper aims to report a case of invasive cryptococcosis in a domestic cat that was presented with neurological signs. Abstract In Compost-Bedded Pack Barn (CBP) systems, air velocity is linked with the thermal comfort of housed dairy cattle and bedding quality and, therefore, assessing ventilation efficiency is essential. In this context, the objective of this study was to evaluate and characterize dependence and spatial distribution of air velocity at the 1.5 m height (vair,M) and at bedding level (vair,B) in an open CBP system with positive pressure ventilation. The study was conducted in 2021, in a facility located in the Zona da Mata region, Minas Gerais, Brazil. The facility area was divided into a mesh composed of 55 equidistant points, where vair,M and vair,B data were collected in the morning (09:00 a.m.) and afternoon (03:00 p.m.) periods, during three weeks in Brazilian winter. Geostatistics techniques were used to assess dependence and spatial distribution. In both periods evaluated, there were a strong occurrence of spatial dependence and non-uniform vair,M and vair,B distributions. The vair,M and vair,B values were lower than recommended (1.8 m∙s–1) in more than 65.0% of the area. Adequate ventilation levels were observed only in the first 20.0 m of the facility, from Southeast to Northwest, because of the fan lines present. DiGeorge syndrome is a disorder caused by a microdeletion on the long arm of chromosome 22. Approximately 1% of patients diagnosed with DiGeorge syndrome may have an absence of a functional thymus, which characterizes the complete form of the syndrome. These patients require urgent treatment to reconstitute T cell immunity. Thymus transplantation is a promising investigational procedure for reconstitution of thymic function in infants with congenital athymia. Here, we demonstrate a possible optimization of the preparation of thymus slices for transplantation through prior depletion of thymocytes and leukocyte cell lineages followed by cryopreservation with cryoprotective media (5% dextran FP 40, 5% Me2SO, and 5% FBS) while preserving tissue architecture. Thymus fragments were stored in liquid nitrogen at -196°C for 30 days or one year. The tissue architecture of the fragments was preserved, including the distinction between medullary thymic epithelial cells (TECs), cortical TECs, and Hassall bodies. Moreover, depleted thymus fragments cryopreserved for one year were recolonized by intrathymic injections of 3×106 thymocytes per mL, demonstrating the capability of these fragments to support T cell development. Lolium multiflorum grass is the major pollen allergen source in the southern region of Brazil, but most of its allergens remain poorly characterized. The aim of this study was to investigate antibody reactivity to L. multiflorum crude and carboxymethyl-ligand extracts in allergic patients and healthy individuals. Ion exchange carboxymethyl (CM) chromatography (CM-Sepharose) was used to isolate proteins (S2) from L. multiflorum crude extract (S1), which were assessed by SDS-PAGE. S1- and S2-specific IgE and IgG4 levels were measured by ELISA using sera from 55 atopic and 16 non-atopic subjects. Reactive polypeptide bands in S1 and S2 were detected by immunoblotting, and the most prominent bands in S2 were analyzed by mass spectrometry (MS-MS). Similar IgE and IgG4 levels were observed to both S1 (IgE median absorbance: 1.22; IgG4 median absorbance: 0.68) and S2 (IgE median absorbance: 1.26; IgG4 median absorbance: 0.85) in atopic subjects. S1 and S2 had positive correlations for IgE and IgG4 (IgE: r=0.9567; IgG4: r=0.9229; P<0.0001) levels. Homology between S1 and S2 was confirmed by IgE (84%) and IgG4 (83%) inhibition. Immunoblotting revealed that the 29-32 kDa band was recognized by 100% of atopic subjects in both S1 and S2. MS-MS analysis identified similarity profile to groups 1 and 5 grass allergens. This study revealed that carboxymethyl-ligand fraction played an important role for pollen allergy diagnosis by containing clinically relevant allergens and constituted a promising candidate for allergen-specific immunotherapy. In this work the effect of the number of cyclic loads on the mechanical behaviour of a chemically stabilised soft soil, reinforced with synthetic polypropylene fibres, and not reinforced, are analysed. This experimental study was carried out on a Portuguese soft soil collected in the Baixo Mondego area in the centre of the country. The laboratory work is based on the following tests: i) monotonic unconfined compression test (UCS), ii) cyclic unconfined compression test performed with different numbers of cycles (2500, 5000, 10000) and several frequencies (0.25, 0.5, 1.0, 2.0 Hz) for a load corresponding to 50% of the strength evaluated in the monotonic tests (UCS) and iii) monotonic UCS tests performed after the cyclic loading stag (UCSpc). The analysis is complemented with the study of the accumulated permanent axial deformation developed during the cyclic stage. The results show that the accumulated permanent axial strain increases with the number of cycles, also the strength and stiffness after the cyclic loading stage increase their values.
En este trabajo se analiza el efecto de parámetros claves de la solicitud cíclica en el comportamiento mecánico de un suelo blando químicamente estabilizado, no reforzado y reforzado con fibras sintéticas de polipropileno. Este estudio experimental se realizó en un suelo blando portugués recogido en la zona del Baixo Mondego en el centro del país. El trabajo de laboratorio se llevó a cabo desarrollando las siguientes pruebas: i) monotónico de compresión no confinada (UCS), ii) prueba cíclica de compresión no confinada realizada con diferentes números de ciclos (2500, 5000, 10000) y diferentes frecuencias (0.25, 0.5, 1.0, 2.0 Hz) para una carga correspondiente al 50% de la resistencia evaluada en las pruebas monotónicas (UCS) y iii) pruebas de UCS monotónicas realizadas luego de la etapa cíclica (UCSpc. El análisis se complementa con el estudio de la deformación axial permanente acumulada desarrollada durante la etapa cíclica. Los resultados muestran que la deformación axial permanente acumulada aumenta con el número de ciclos, también se observa que aumenta la resistencia y la rigidez después de la etapa de carga cíclica.

Glioblastoma is the most prevalent and malignant brain tumor identified in adults. Surgical resection followed by radiotherapy and chemotherapy, mainly with temozolomide (TMZ), is the chosen treatment for this type of tumor. However, the average survival of patients is around 15 months. Novel approaches to glioblastoma treatment are greatly needed. Here, we aimed to investigate the anti-glioblastoma effect of the combination of matteucinol (Mat) (dihydroxyflavanone derived from Miconia chamissois Naudin) with the chemotherapeutic TMZ in vitro using tumor (U-251MG) and normal astrocyte (NHA) cell lines and in vivo using the chick embryo chorioallantoic membrane (CAM) assay. The combination was cytotoxic and selective for tumor cells (28 μg/mL Mat and 9.71 μg/mL TMZ). Additionally, the combination did not alter cell adhesion but caused morphological changes characteristic of apoptosis in vitro. Notably, the combination was also able to reduce tumor growth in the chick embryo model (CAM assay). The docking results showed that Mat was the best ligand to the cell death membrane receptor TNFR1 and to TNFR1/TMZ complex, suggesting that these two molecules may be working together increasing their potential. In conclusion, Mat-TMZ can be a good candidate for pharmacokinetic studies in view of clinical use for the treatment of glioblastoma.
