Adaptability and stability of sweet potato genotypes in Western São Paulo

RESUMO A batata-doce (Ipomoea batatas L.) possui grande variabilidade genética que contribui para maior produção e qualidades das raízes. Os agricultores de batata-doce do Oeste Paulista, Brasil, plantam durante as quatro estações e utilizam há duas décadas os mesmos genótipos. Desse modo, são necessárias pesquisas que visem avaliar novos genótipos e indicar os mais promissores. Portanto, objetiva-se identificar a adaptabilidade e estabilidade de genótipos de batata-doce em seis épocas de plantio: Setembro de 2019, Janeiro de 2020, Abril de 2020, Setembro de 2020, Outubro de 2020 e Dezembro de 2020 no Oeste Paulista, por meio da metodologia da regressão linear proposta por Eberhart & Russell (1966) e pelo método Centróide. Avaliou-se 17 genótipos, dois genótipos experimentais do Banco de Germoplasma da Universidade do Oeste Paulista e dois genótipos que são cultivados há mais de duas décadas pelos agricultores (Canadense e Ligeirinha Paulista). A partir dos resultados obtidos, os genótipos Canadense, IAPAR 69 e SCS 272 Marina, podem ser considerados como de ampla adaptação na região Oeste Paulista (ambientes favoráveis e desfavoráveis). Por sua vez, os genótipos UBD 01 e UBD 02 são indicados exclusivamente para ambientes desfavoráveis, enquanto que Ligeirinha Paulista, INIA Arapey, SCS 369 Águas Negras, BRS Rubissol, Brazlândia Branca, Brazlândia Roxa e BRS Amélia são indicados para ambientes favoráveis. batatadoce batata doce Ipomoea L. ABSTRACT Sweet potato (Ipomoea batatas L.) have great genetic variability that contribute to higher production and root quality. Sweet potato farmers in the Western region of São Paulo state, Brazil, plant during the four seasons and have used the same genotypes for two decades. Thus, research is needed to evaluate new genotypes and indicate the most promising ones. Therefore, the objective of this study was to identify the adaptability and stability of sweet potato genotypes in six planting seasons: September 2019, January 2020, April 2020, September 2020, October 2020, and December 2020 in Western São Paulo using the linear regression methodology proposed by Eberhart & Russell (1966) and the centroid method. We evaluated 17 genotypes, two experimental genotypes from the germplasm bank of the University of Western São Paulo and two genotypes that have been cultivated for more than two decades by farmers (Canadense and Ligeirinha Paulista). The results show that Canadense, IAPAR 69, and SCS 272 Marina can be considered widely adapted in the Western region of São Paulo (favorable and unfavorable environments). ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT Chickpea has recently gained prominence in agriculture, being an excellent option for crop rotation in areas cultivated with soybean or corn. Seeds must have good physiological quality, mainly during storage, to ensure crop success. Seeds undergo metabolic reactions that impair their performance when stored improperly. In this context, a study was carried out to evaluate the physiological variables of the chickpea cultivar BRS Aleppo in different types of packaging and storage environments. Abstract Dinizia Excelsa is an Amazonian tree with a wide range of applications as a raw material in the industry. The objective of this study was to extract, characterize, and evaluate the biological activities of xylan extracted from Dinizia excelsa wood. The xylan was obtained in five stages, including delignification, precipitation, purification, and freeze-drying. The physicochemical analysis of xylan included the determination of monosaccharides, elemental composition, FTIR analysis, 2D nuclear magnetic resonance spectroscopy, and the determination of molecular weight. Xylan had an extraction yield of 28.44% and an elemental composition of 35.03% carbon, 5.65% hydrogen, and 59.32% oxygen. FTIR analysis revealed similarities between Dinizia excelsa xylan and commercial xylan. 2D NMR analysis confirmed the presence of characteristic xylan groups. Furthermore, xylan has a low molecular weight. In vitro cytotoxicity tests demonstrated low toxicity, indicating its potential for biological applications. Immunomodulatory activity assays revealed that xylan stimulated cell proliferation and the production of anti-inflammatory cytokines. The anticoagulant activity of xylan was low compared to heparin. The antioxidant activity of xylan was weaker compared to ascorbic acid and butylated hydroxytoluene (BHT). The São Luís Island, Maranhão State, is an ecotonal region, a mix of humid forest, cocais forest, dune vegetation, restingas and mangroves. Unlike the interior of the State, their coastal areas have a gap in knowledge of the Passifloraceae. Here, through field expeditions and herbaria visit, we provide taxonomic treatment, identification key and comments on the Passifloraceae of the São Luís Island. Ten species were recorded: Passiflora auriculata Kunth, P. edulis Sims, P. foetida L., P. glandulosa Cav., P. laurifolia L., P. misera Kunth, P. nitida Kunth, P. picturata Ker Gawl., P. subrotunda Mast., and P. vespertilio L. Passiflora picturata and P. misera were recorded for the first time on São Luís Island.
A Ilha de São Luís, Estado do Maranhão, configura-se como uma região ecotonal, um misto de floresta úmida, mata-de-cocais, vegetação de dunas, restingas e manguezais. Diferentemente do interior do Estado do Maranhão, suas zonas costeiras possuem uma lacuna sobre o conhecimento da família Passifloraceae. Aqui, por meio de expedições de campo e consulta a herbários, nós fornecemos o tratamento taxonômico, chave de identificação e comentários sobre as Passifloraceae da Ilha de São Luís. Foram registradas dez espécies: Passiflora auriculata Kunth, P. edulis Sims, P. foetida L., P. glandulosa Cav., P. laurifolia L., P. misera Kunth, P. nitida Kunth, P. picturata Ker Gawl., P. subrotunda Mast. e P. vespertilio L. ABSTRACT Purpose: To evaluate the long-chain fatty acid and major compounds levels in the feces after prophylactic oral use of Lacticaseibacillus casei in an experimental model of intestinal mucositis. Methods: Fifteen Swiss mice were randomly divided into three groups (n=5/group): The negative or positive control groups (n = 5) received saline orally for 18 days and an the intraperitoneal (i.p.) of saline or 5 Fluorouracil (450 mg/kg) in 15th day, respectability. L. casei group received oral concentration of L. casei (1x109 CFU/mL) for 18 days, the i.p. injection of 5-fluorouracil (450 mg/kg) in 15th days. Tissue samples from colon and each small intestine segment were collected for histopathological analysis. Stool samples were collected. Fecal composition of long-chain fatty acids and sterols were analysed by gas chromatography-mass spectrometry on the 15th and the 18th day. Results: The mucosa layer of all small intestine segments of animals from L. casei showed well preserved epithelium and glands, without necrosis signs, but Goblet cells number decreased. Several long-chain fatty acids and sterols have been identified before and after in the groups. L. casei administration after 5-FU treatment reduced concentrations of linoleic acid (18:2) (p < 0.001) and oleic acid (18:1) (p < 0.001) in feces. Conclusions: L. casei prevented the mucosal damage associated with 5-FU-induced intestinal mucositis reduced long-chain fatty acid levels in the feces. Abstract Based on the genetic, morphological, and morphometric data of blood gamonts, we identified Hepatozoon parasites in colubrid snakes sampled in the Eastern Amazon region. Hepatozoon trigeminum was detected in the mussurana snake Clelia clelia and exhibited wide and elongated gamonts (mean dimensions: 14.25±0.65 × 4.31±0.43 μm) with an evident parasitophorous vacuole. Hepatozoon odwyerae sp. nov. was described in the indigo snake Drymarchon corais, whose gamonts have elongated and thin bodies (mean dimensions: 13.41±0.79 × 3.72±0.35 μm) with one end more tapered than the other. Phylogenetic analyses, based on the amplification of a 441 bp fragment of the 18S rRNA gene, revealed that the novel sequences of Hepatozoon spp. from our study were closely related to hemogregarine lineages found in lizards and snakes from Brazil, forming a well-supported monophyletic clade with them. The present study provides the first species description of Hepatoozon in D. corais and a new record of a host species for C. clelia using the integrated taxonomic data. RESUMO O manejo de nitrogênio (N) na cultura do milho é fator chave para o sucesso de seu cultivo, de modo que se objetivou com esse estudo avaliar a resposta da cultura, em segunda safra, à doses de N aplicadas exclusivamente em cobertura ou na adubação de base (semeadura) complementada com a adubação de cobertura. Para tal, em 2019 e 2020 foram realizados cinco experimentos em áreas da região Sudoeste do Estado de Goiás, testando 0 e 30 kg ha-1 de N aplicados na base combinados com cinco doses totais de N (0, 75, 150, 225 e 300 kg ha-1) completadas em cobertura, quando as plantas estavam em estádio V3. Avaliou-se a produção de massa seca na fase vegetativa V6 ou V9, e, por ocasião da colheita, os parâmetros produtivos. Verificou que a aplicação de N não interfere no número de fileiras de grãos, mas aumenta o número de grãos por fileira e, principalmente, o peso de grãos. Em 4 dos 5 ensaios, a cultura respondeu positivamente ao incremento da dose total de N; em 1 desses, a adubação de base contribuiu para aumento da eficiência da adubação de cobertura, de modo que a produtividade do milho foi maior e atingida com uma dose total de N menor. Nos outros 3, respostas positivas foram observadas quando da aplicação total do N em cobertura e em dose única. A maior produtividade média de grãos de milho (8.233 kg ha-1), no entanto, ocorreu justamente no ensaio com ausência de resposta à adubação nitrogenada - provavelmente devido às melhores condições de fertilidade química da área aliada à adequada distribuição das chuvas, considerando assim múltiplos fatores para N recomendações de aplicação. ABSTRACT Nitrogen (N) fertilizer management is a key factor in the success of maize crops. The objective of this work was to evaluate the response of second-crop maize to N applied exclusively as topdressing or at planting + topdressing. Five experiments were conducted in areas in southwestern Goiás in 2019 and 2020 to evaluate N application rates of 0 and 30 kg ha-1 at planting, combined with five N rates applied as topdressing at the maize V3 stage over a total of 0, 75, 150, 225, and 300 kg ha-1. Dry matter production in the V6 or V9 stage and yield parameters at harvest were evaluated. N application did not affect the number of grain rows per ear but increased the number of grains per row and, particularly, grain weight. The crop responded positively to increases in the total N rate in 4 of the 5 experiments: in 1 of these experiments, N fertilizer application at planting contributed to an increase in the efficiency of topdressing fertilization, resulting in a higher maize yield with a lower total N rate; in the other 3, a positive maize response to N occurred when total N was applied as topdressing at V3. Abstract Although widely used in medicine, separation technology, and other fields, the effects of cyclodextrins on the activities of phosphoryl transfer enzymes have not been previously evaluated. In vivo studies evaluated the function of cyclodextrins as active compounds. Despite the use of cyclodextrins as active compounds, the effects of cyclodextrins on hepatic and renal tissues remain to be fully elucidated. The primary objective of this study was to evaluate the effects of β- cyclodextrins, methyl-β-cyclodextrin (M-β- cyclodextrins), and (2-hydroxypropyl)-β-cyclodextrin (HP-β-cyclodextrins) on enzyme activities regulating the maintenance of energy homeostasis in the kidney and liver tissues in relation to toxicity. Serum levels of liver and kidney markers were measured, and oxidative stress parameters were assessed. After 60-day treatments, we observed that the administration of β-cyclodextrins and M-β-cyclodextrins inhibited the hepatic activity of pyruvate kinase, an irreversible enzyme within the glycolytic pathway. Additionally, administration of HP-β-cyclodextrins inhibited creatine kinase activity and increased the total sulfhydryl content in kidneys. Here, we demonstrated for the first time that β-cyclodextrins, M-β-cyclodextrins, and HP-β-cyclodextrins cause bioenergetic dysfunction in renal and hepatic tissues. Abstract This study presents the first complete mitogenome of the Brazilian Atlantic bushmaster Lachesis with insights into snake evolution. The total length was 17,177 bp, consisting of 13 PCGs, 22 tRNAs, two rRNAs and a duplicate control region (CRs). Almost all genes were encoded by the heavy-strand, except for the ND6 gene and eight tRNAs (tRNA-Gln, Ala, Asn, Cys, Tyr, Ser[TGA anticodon], Glu, Pro). Only ATG, ATA, and ATC were starting codons for protein-coding sequences. Stop codons mainly were TAA, AGA, AGG, and TAG; whereas ND1, ND3, and CYTB terminated with incomplete stop codons. Phylogeny retrieved Lachesis within the Crotalinae as the sister group of Agkistrodon; and the Lachesis+Agkistrodon clade as the sister group of (Sistrurus+Crotalus)+Bothrops. The tree supports Crotalinae, Viperinae, and Azemiopinae in the Viperidae family, being sister taxa of Colubridae+(Elapidae+Psammophiidae). The mean genetic distance across 15 snake families and 57 nucleotide sequences was 0.37. The overall mean value of genetic distance across the Crotalinae was 0.23, with Lachesis muta exhibiting the shortest distance of 0.2 with Agkistrodon piscivorus, Protobothrops dabieshanensis and P. flavoviridis and the greatest 0.25 with Gloydius blomhoffii, Trimeresurus albolabris, S. miliarius, and Deinagkistrodon acutus. Do native plant species functionally similar to invasive species suffer more impacts from the invasion in seasonally dry tropical forests?

Resumo Testamos a hipótese que das espécies estudadas do Semiárido brasileiro, as funcionalmente mais semelhantes com a arbórea exótica Azadirachta indicaseriam mais afetadas na germinação e no desenvolvimento inicial. Inicialmente foi produzido extrato bruto das folhas frescas da exótica em seis diluições. Para cada diluição foram colocadas para germinar 25 sementes, de quatro espécies nativas ( Cenostigma pyramidale, Libidibia ferrea, Mimosa caesalpiniifoliae Amburana cearensis), em placas de Petri com quatro repetições para cada espécie, onde foram avaliados: comprimento da radícula, porcentagem de emergência, índice de velocidade de emergência e tempo médio de emergência. Para avaliar a influência da exótica sobre o desenvolvimento dos indivíduos das espécies estudadas foi realizado um experimento em casa de vegetação. As sementes foram plantadas em 10 baldes de 20 l., sendo cinco baldes para cada espécie nativa isolada e cinco para ela em interação com a espécie exótica. O experimento durou quatro meses, e os atributos funcionais dos indivíduos foram coletados. Das nativas analisadas, de acordo com a análise de agrupamento usando os atributos funcionais das espécies plantadas isoladamente, M. caesalpiniifolia, C. pyramidalee L. ferreaforam funcionalmente mais próximas da exótica. No experimento de germinação notou-se que o extrato afetou apenas o comprimento da raiz, independente da proximidade funcional. Notamos que apenas M. caesalpiniifolia, plantada em interação, teve seu desenvolvimento inicial fortemente inibido. A semelhança funcional entre a exótica e as nativas não influenciou na germinação, mas afetou negativamente o desenvolvimento.
Abstract We tested the hypothesis that of the species studied in the Brazilian Semiarid, the ones most functionally similar to tree Azadirachta indicawould be more affected by this exotic species during germination and early development. At first, we produced a crude extract of A. indica fresh leaves in six different dilutions. We placed 25 seeds of four native species ( Cenostigma pyramidale, Libidibia ferrea, Mimosa caesalpiniifolia, and Amburana cearensis) to germinate in Petri dishes for each dilution, with four replicates for each species. We assessed: radicle length, emergence percentage, emergence speed index, and mean emergence time. We conducted an experiment in a greenhouse to assess how the exotic plant affected the development of individual plants of the studied species. We planted five seeds of each native species alone and five in interaction with the exotic species using ten 20-l buckets. The experiment lasted four months, and the functional attributes of the individuals were collected. Among the native species analyzed, according to the cluster analysis using the functional attributes of the species planted alone, M. caesalpiniifolia, C. pyramidaleand L. ferreawere functionally more similar to the exotic species. In the germination experiment, the extract affected only root length, regardless of the functional similarity. We noticed that only M. caesalpiniifoliahad its early development strongly inhibited when planted in interaction with the exotic species. Although the functional similarity between the exotic and the native species did not influence germination, it negatively affected the development.

ABSTRACT We assessed the impacts of the biological invasion of Azadirachta indica on the quality of plant species native to a Seasonally Dry Tropical Forest. We planted Amburana cearensis, Libidibia ferrea, Mimosa caesalpiniifolia, and Cenostigma pyramidale individuals alone and interacting with the exotic species. We measured their diameters at ground level and heights for four months, calculating their monthly and general average. After this period, seedlings were destroyed to obtain their number of leaves, leaf area, specific leaf area, length and primary root area, wood density, wood dry matter, leaf dry matter, shoot dry matter, primary root dry matter, secondary root dry matter, root system dry matter, and total dry matter. The presence of the exotic species affected the functional characteristics of native species in different ways. We observed negative effects in M. caesalpiniifolia and A. cearensis, positive effects in C. pyramidale, and mixed effects in L. ferrea. In addition, we identified conflicting demands between pairs of characteristics of the interacting species A. cearensis and M. caesalpiniifolia. Based on the results, it is possible to conclude that A. indica decreases the quality of individuals of most native species studied, which may have consequences for their establishment and the local biodiversity.

ABSTRACT Owing to numerous health benefits, the cultivation of medicinal plants has significantly increased. Achillea millefolium L. (common names: milefoil, yarrow) is attributed important medicinal properties. Consumers of herbal medicinal products demand for production systems based on ecological farming principles, thus an interesting technology would be application biostimulant based on humic acids. The objective of this study was to evaluate the rooting and initial performance of Achillea millefolium in response to the use of humic acids (HA) isolated from composted cattle manure and poultry litter and applied in different concentrations (0, 10, 20, 30, and 40 mmolL-1 of HA) at the propagation phase. Rooting and plant biomass accumulation were analyzed. Humic acids derived from poultry litter promoted root development and increase of fresh biomass and total dry mass, proving their biostimulant effect. The optimal concentration of humic acids estimated for the initial development of A. millefolium was 22.25 mmolL-1.

RESUMO O Plano Global de Eliminação da Filariose Linfática, lançado pela Organização Mundial da Saúde em 2000, propõe o uso de testes de detecção de antígeno circulante filarial como ferramenta diagnóstica para avaliação e monitoramento das ações de controle da parasitose. Entretanto, esses testes, apesar de apresentarem alta sensibilidade, não conseguem detectar com eficiência a infecção em seu estágio inicial, quando ainda não existe a presença de helmintos adultos. Considerando essa limitação, a pesquisa de anticorpos antifilariais tem sido apontada como uma alternativa, uma vez que os anticorpos produzidos contra as larvas infectantes do parasito são detectados antes da presença de antígeno circulante filarial. O objetivo deste estudo foi definir o ponto de corte e avaliar a acurácia do kit Filaria Detect™ IgG4 produzido com o antígeno recombinante Wb123 para diagnóstico da filariose linfática no Brasil. Para isso, foi realizado um estudo de avaliação de teste diagnóstico, no qual foram utilizadas 256 amostras de soro: 79 (30,9%) obtidas de indivíduos microfilarêmicos e 177 (60,1%), de indivíduos amicrofilarêmicos e que testaram negativo para os testes imunológicos Bm14 CELISA e Og4C3 ELISA. A definição do ponto de corte ideal, bem como da acurácia do kit Filaria Detect™ IgG4, foi obtida através da construção de curvas ROC, sendo a densidade óptica de 0,239 aquela na qual o teste obteve melhor desempenho, com sensibilidade de 81,0% e especificidade de 96,6%. Os resultados obtidos demonstraram que o kit Filaria Detect™ IgG4 é uma ferramenta promissora para investigação e monitoramento de áreas submetidas ao tratamento em massa para filariose linfática.
ABSTRACT The Global Programme to Eliminate Lymphatic Filariasis, launched by the World Health Organization in the year 2000, proposes the use of circulating filarial antigen tests as a diagnostic tool to assess and monitor initiatives to control filarial infection. However, despite a high sensitivity, these tests are not efficient to detect infection at early stages, before worms have reached the adult stage. Considering this limitation, anti-filarial antibody testing has been suggested as an alternative, given that the antibodies produced against the larvae are detectable before the presence of circulating filarial antigen. The objective of the present study was to determine the diagnostic cut-off and the accuracy of the Filaria Detect™ IgG4 kit employing recombinant Wb123 antigen for diagnosis of lymphatic filariasis in Brazil. For that, we performed a diagnostic evaluation study in which 256 serum samples were analyzed: 79 (30.9%) obtained from microfilaremic individuals and 177 (60.1%) from amicrofilaremic individuals who tested negative with the Bm14 CELISA and Og4C3 ELISA immunologic tests. The ideal cutoff as well as the Filaria Detect™ IgG4 kit accuracy were determined based on ROC curve analyses, with an optical density of 0.239 identified as the cutoff with the best performance, with 81.0% sensitivity and 96.6% specificity. The results show that the Filaria Detect™ IgG4 kit is a promising tool for investigation and monitoring of areas undergoing mass drug administration for lymphatic filariasis.
RESUMEN En el programa mundial de eliminación de la filariasis linfática, puesto en marcha por la Organización Mundial de la Salud en el año 2000, se propone el uso de pruebas de detección del antígeno filárico circulante como instrumento de diagnóstico para la evaluación y el seguimiento de las medidas de control de la parasitosis. Sin embargo, esas pruebas, a pesar de tener un alto grado de sensibilidad, no permiten detectar con eficiencia la infección en su fase inicial, cuando todavía no existen helmintos adultos. En vista de esa limitación, se ha señalado como una opción el estudio de anticuerpos antifiláricos, puesto que los anticuerpos producidos contra las larvas infectantes del parásito se detectan antes de la existencia de antígeno filárico circulante. El objetivo de este estudio fue definir el punto de corte y evaluar la exactitud del estuche Detect™ para pruebas de anticuerpos antifiláricos IgG4, fabricado con el antígeno recombinante Wb123, para el diagnóstico de la filariasis linfática en Brasil. Para ello, se realizó un estudio de evaluación de la prueba diagnóstica, en el cual se utilizaron 256 muestras de suero, a saber, 79 (30,9%) obtenidas de personas microfilarémicas y 177 (60,1%) de personas amicrofilarémicas, que arrojaron resultados seronegativos en las pruebas inmunológicas CELISA Bm14 y ELISA Og4C3. La definición del punto de corte ideal y de la exactitud del estuche Detect™ se obtuvo con la construcción de curvas de la característica operativa del receptor (ROC); una densidad óptica de 0,239 marcó el mejor nivel de desempeño de la prueba, con una sensibilidad de 81,0% y una especificidad de 96,6%. Los resultados obtenidos demostraron que el estuche Detect™ es un instrumento prometedor para la investigación y el seguimiento de las regiones donde se realiza un tratamiento masivo de la filariasis linfática.
