Cytotoxicity of dilutions of bioceramic materials in stem cells of human exfoliated deciduous teeth

Abstract Several materials have been developed to preserve pulp vitality. They should have ideal cytocompatibility characteristics to promote the activity of stem cells of human exfoliated deciduous teeth (SHED) and thus heal pulp tissue. Objective To evaluate the cytotoxicity of different dilutions of bioceramic material extracts in SHED. Methodology SHED were immersed in αMEM + the material extract according to the following experimental groups: Group 1 (G1) –BBio membrane, Group 2 (G2) - Bio-C Repair, Group 3 (G3) - MTA Repair HP, Group 4 (G4) – TheraCal LC, and Group 5 (G5) - Biodentine. Positive and negative control groups were maintained respectively in αMEM + 10% FBS and Milli-Q Water. The methods to analyze cell viability and proliferation involved MTT and Alamar Blue assays at 24, 48, and 72H after the contact of the SHED with bioceramic extracts at 1:1 and 1:2 dilutions. Data were analyzed by the three-way ANOVA, followed by Tukey’s test (p<0.05). Results At 1:1 dilution, SHED in contact with the MTA HP Repair extract showed statistically higher cell viability than the other experimental groups and the negative control (p<0.05), except for TheraCal LC (p> 0.05). At 1:2 dilution, BBio Membrane and Bio-C showed statistically higher values in intra- and intergroup comparisons (p<0.05). BBio Membrane, Bio-C Repair, and Biodentine extracts at 1:1 dilution showed greater cytotoxicity than 1:2 dilution in all periods (p<0.05). Conclusion MTA HP Repair showed the lowest cytotoxicity even at a 1:1 dilution. At a 1:2 dilution, the SHED in contact with the BBio membrane extract showed high cell viability. Thus, the BBio membrane would be a new non-cytotoxic biomaterial for SHED. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Resumo O estudo visa avaliar a promoção dos subgrupos de AIN/MP anunciados em encartes de supermercados, considerando as características das redes e a tipologia dos encartes. Foram analisados encartes de cinco redes de supermercados da região metropolitana do Rio de Janeiro (junho/2019-maio/2020). Foram apresentados dados de abrangência e perfil socioeconômico das redes e tipo de encarte. Dos 68.110 alimentos, 30,6% são AIN/MP. Carnes/ovos foram mais promovidos (42,3%), seguido de frutas/hortaliças (20,2%) anunciados mais em encartes de hortifruti. Laticínios; cereais/farinhas/massas e bebidas/infusões representaram cerca de 10,0% dos anúncios. Leguminosas (3,8%), especiarias e cogumelos (0,2%) e oleaginosas (0,1%) foram pouco promovidos. Foram encontradas diferenças estatisticamente significativas na promoção de subgrupos segundo abrangência e perfil socioeconômico das redes. Frutas/hortaliças foram mais anunciadas em redes nacionais e voltadas para o público de alta renda em comparação às locais e voltadas para um público de média/baixa renda. Constatou-se desequilíbrio na promoção dos subgrupos de AIN/MP, com diferenças entre as redes e os tipos de encartes. Diante disto, deve-se dar luz à discussão sobre a regulação da promoção de alimentos no varejo alimentar. ABSTRACT This study evaluated how nutritional insults applied at different stages of intrauterine development affect the growth and development of total tract organs and the mRNA expression of genes that encode growth factors, tight junction proteins, digestive enzymes, and glucose transporters in the small intestine of newborn goats. Fourteen nulliparous dairy goats were assigned to one of two nutritional plans over gestation: maintenance-restriction (M-R, n = 8) – 100% of maintenance requirement from d 8-84 of gestation followed by feeding at 50% maintenance requirement from d 85 of gestation to parturition; or restriction-maintenance (R-M, n = 6) – 50% of maintenance requirement from d 8-84 of gestation followed by feeding at 100% maintenance requirement from d 85 of gestation to parturition. Fisher's least significant difference test was performed using SAS 9.4, considering a full fixed-effect model. When expressed per kg of body weight, the weights of the small intestine and total intestine were 18.7 and 18.1% lower, respectively, for R-M offspring. The lengths of the small intestine and the total intestine of the R-M newborns were 19.1 and 9.5% shorter, respectively. This group also had a lower villus height:intestinal crypt depth ratio. This study aimed to investigate a miniaturized extraction methodology with analysis by GC-MS for simultaneous detection and quantification of cocaine (COC), benzoylecgonine (BZE) and cocaethylene (CE) in hair to monitor drug addicts. The process was done following the international guide of the Scientific Task Force on Forensic Toxicology. After sample extraction, derivatizing solution was added, clean-up was done by the Hollow Fiber Liquid Phase Microextraction (HF-LPME) and adapted to use the ultrasonic bath, a simple and easy-to-handle device. Isobutylchloroformate was first used as derivatization reagent to convert benzoylecgonine to isobutylbenzoylecgonine. Analytes quantification occurred within a linear range of 0.1 to 20 ng mg-1 for COC and 0.05 to 5 ng mg-1 for BZE and CE, with a correlation coefficient of r > 0.99. Limits of detection were 0.05; 0.03 and 0.03 ng mg-1, whereas limits of quantification were 0.1; 0.05 and 0.05 ng mg-1 for COC; BZE and CE, respectively. There was no matrix effect interference. Intra and inter-day precision and accuracy were acceptable according to international guidelines. The analytical method HF-LPME-GC-MS was successfully applied to 14 hair samples from patients admitted in drugs detoxification programs. All samples resulted positive for cocaine (0.80-> 20 ng mg-1) and benzoylecgonine (0.20-> 5 ng mg-1), 11 samples were positive for cocaethylene (0.10-> 0.60 ng mg-1).

Resumo Objetivos: este estudo revisou sistematicamente a literatura para melhor compreender a associação entre COVID-19, gravidez e neonatos. Métodos: MEDLINE, EMBASE, Web of Science, BVS e SCOPUS foram acessadas, considerando os termos: (covid 19 OR covid-19 OR novel coronavirus OR 2019 novel coronavirus OR 2019-nCoV OR sarscov 2 OR sars-cov-2 OR sarscov2 OR sars cov-2) AND (pregnancy OR pregnant OR pregnant women OR gestation OR gestational) AND (infant OR fetal OR neonatal). Trinta textos completos foram incluídos (408 gestantes, 11 mulheres nãogestantes e 279 recém-nascidos). Resultados: febre (45,83%) e tosse (31,61%) foram os principais sintomas da COVID-19 durante a gestação. Baixos níveis de linfócitos (32,10%), elevados níveis de proteínas Creativa (32,35%); leucocitose (29,41%); neutrófilo (5,88%); e alterações radiográficas sob tomografia computadorizada de tórax, radiografia ou ultrasom (45,84%) foram os principais achados laboratoriais. Parto por cesárea e prematuridade foram registrados em 239 e 49 casos, respectivamente. Dez recém-nascidos testaram positivo para o virus SARS-CoV-2. Conclusão: quando a pneumonia COVID-19 afeta mulheres durante a gravidez, os sintomas são semelhantes aos experimentados por mulheres não grávidas. Além disso, ainda não há evidências plausíveis que sugiram a transmissão vertical do vírus SARS-CoV-2 de mãe para filho.

Low levels of lymphocytes (32.10%), elevated levels of C-reactive protein (32.35%); leukocytosis (29.41%); neutrophil (5.88%); and radiographic alterations on chest CT, x-ray or ultrasound (45.84%) were the main laboratorial findings. Cesarean delivery and preterm were registered in 239 and 49 cases, respectively. Ten neonates tested positive for SARS-CoV-2. Conclusion: when COVID-19 pneumonia affects women during pregnancy, the symptoms are similar to those experienced by non-pregnant women. In addition, there is still no plausible evidence suggesting vertical transmission of SARS-CoV-2 virus from mother to child.

ABSTRACT Introduction: syphilis is a sexually transmitted disease with an annual incidence of 12 million people worldwide. In Brazil, there are increasing rates of infant mortality and incidence due to this disease. Objective: to report a case of primary syphilis with manifestation in the mouth. Case report: patient 50-years-old, male, homosexual with a complaint of an asymptomatic lesion on the lower left lip for about 2 months. In the intraoral physical examination, it was observed ulcerated lesion, with white-yellowish pseudomembrane, non-suppurative with hardened, elevated and badly-delimited edges measuring approximately 3 cm on the lower left lip. An incisional biopsy of the lesion was carried out and it was prescribed high doses of prednisolone, but without success. During follow-up, erythematous lesions target-shaped appeared in the palmar region and the resulting histopathological report showed ulcerated lichenoid cheilitis. Hematological exams confirmed positive for syphilis in the VDRL test. The treatment was penicillin G benzathine for 3 weeks. Conclusions: in the present case, the syphilitic lesion has clinical characteristics common to other diseases. Thus, dentists should know their clinical manifestations, that would aid in early diagnosis and treatment.
RESUMEN Introducción: La sífilis es una enfermedad de transmisión sexual con una incidencia anual de 12 millones de personas en el mundo. En Brasil, hay crecientes tasas de incidencia y mortalidad infantil por esa enfermedad. Objetivo: Describir un caso de sífilis primaria con manifestación en cavidad oral. Presentación del caso: Paciente masculino de 50 años de edad, homosexual, con queja de lesión sin dolor en el labio inferior izquierdo que había comenzado aproximadamente dos meses antes de asistir a la consulta. En la inspección clínica intraoral se observó lesión ulcerada, que medía aproximadamente 3 cm en el labio inferior izquierdo, con seudomembrana blanco-amarillenta, no supurativa, con bordes indurados, elevados y mal delimitados. Se tomó una biopsia de la lesión y se prescribieron altas dosis de prednisolona, pero sin éxito. Durante el seguimiento, lesiones eritematosas en “tiro al blanco” aparecieron en la región palmar y el estudio histopatológico reveló queilitis liquenoide ulcerada. Los exámenes hematológicos confirmaron positividad para sífilis en la prueba VDRL. El tratamiento fue penicilina G benzatínica por tres semanas. Conclusiones: En el presente caso la lesión sifilítica tenía características clínicas comunes a otras enfermedades. Por lo tanto, los dentistas deben conocer sus manifestaciones clínicas, que ayudarían en el diagnóstico y tratamiento tempranos.

RESUMO. A amputação é um procedimento irreversível que gera consequências sociais, psicológicas e funcionais. A dor do membro fantasma ocorre entre 50 e 80% dos amputados, apresentando implicações na qualidade de vida. Objetivo: Avaliar a sensação, dor fantasma e qualidade de vida entre pacientes amputados. Métodos: Trata-se de um estudo transversal retrospectivo realizado na região de Juiz de Fora, Minas Gerais, Brasil. Os critérios de inclusão foram possuir pelo menos uma amputação de membro inferior e ser paciente de um dos hospitais referência para região. Vinte participantes foram incluídos nas análises. O questionário da pesquisa consistiu em itens, adaptados do Groningen Questionnaire Problems After Leg Amputation, que descrevem frequência e incômodo da sensação fantasma, dor fantasma, causas e nível da amputação, e questões sobre os tratamentos além do questionário qualidade de vida WHOQOL-BREF. Resultados: A maioria dos participantes era do sexo feminino (55%) com idade média de 55,6 (DP=14,8) anos. A maior frequência de amputação foi transfemural (65%), e o principal motivo da amputação foi o diabetes mellitus (40%). Com relação ao incômodo da sensação fantasma, 29% a classificou como moderado ou severa, e 15% alegou frequência diária desse fenômeno. Quanto {a dor fantasma, apenas 6% afirmou frequência diária. A qualidade de vida média dos participantes foi de 4,1 (DP=1,1) em 5 (cinco significa muito satisfeito), de acordo com a primeira questão do WHOQOL-BREF; o domínio físico apresentou a menor média 3,4 (DP=0,7). Conclusão: A sensação e dor fantasmas foram prevalentes nos entrevistados que apresentaram menor satisfação com o domínio físico da qualidade de vida.
ABSTRACT. An amputation is an irreversible event that causes social, psychological, and functional consequences that reduces the quality of life of the amputee. Phantom pain generally is reported by 50 to 80% of amputees. Objective: To describe the pain and phantom sensation and quality of life among lower-limb amputees. Methods: This was a cross-sectional study carried out in the region of Juiz de Fora, state of Minas Gerais, Brazil. Inclusion criteria were being a patient in one of two hospitals in the region at the time of the interview and having at least one lower-limb amputation. A total of 20 amputees were included in the analysis. The interview questionnaire had items adapted from the Groningen Questionnaire Problems After Leg Amputation - describing the frequency and discomfort of phantom pain and sensation, causes and the level of the amputation, as well as the WHOQOL-BREF, for assessing quality of life. Results: Most participants were women (55%) and had a mean age of 55.6 years (SD=14.8). Femoral amputation was the most prevalent (65%), and diabetes (40%) was the main reason for amputation. 29% of amputees classified the phantom pain as moderate or severe, and 15% claimed daily frequency of this phenomenon. As for phantom pain, only 6% stated daily frequency. The mean quality of life was 4.1 (SD=1.1, five score means very satisfied), the physical domain of quality of life had the lowest mean (3.4, SD=0.7). Conclusions: Phantom sensation and pain were prevalent among lower-limb amputees who were, in general, less satisfied with their physical domain of quality of life.

Resumen Este artículo presenta una reflexión y discusión sistematizada en torno a dos ejes orientadores: en el primero, se habla sobre el envejecimiento y las vulnerabilidades a las pérdidas biológicas, físicas, cognitivas, sociales y afectivas que requieren atención específica, y, así mismo, sobre las vulnerabilidades a COVID-19 a las que están expuestos los ancianos institucionalizados; en el segundo eje, se reflexiona sobre la adopción de medidas restrictivas y de protección para prevenir la propagación del virus con el objetivo de mantener la salud del anciano y mitigar los efectos de la pandemia. La conclusión es que esta ha incrementado las numerosas vulnerabilidades a las que ya estaban expuestas las personas mayores institucionalizadas, agregando, además, la vulnerabilidad a una enfermedad nueva como es el COVID-19, debido a la alta letalidad y comorbilidad que representa, la cual, es agravada por la precariedad de las instituciones brasileñas de larga estancia debido a la negligencia de las autoridades públicas, la sociedad civil, la gerencia de la institución y las familias de los pacientes. El escenario post-pandemia requerirá esfuerzos colectivos para proteger y garantizar la supervivencia de los ancianos que viven en estas residencias.
Abstract This article presents a systematized reflection and discussion around two guiding axes: the first discusses aging and vulnerabilities to biological, physical, cognitive, social and affective losses that require specific attention, as well as vulnerabilities to COVID-19 to which institutionalized elderly people are exposed; the second, we reflect on the adoption of restrictive and protective measures to prevent the spread of the virus, aiming to keep the elder health and mitigate the effects of the pandemic. The conclusion is that the pandemic has increased the many vulnerabilities to which institutionalized older people were already exposed, adding vulnerability to a new disease, such as COVID-19, due to its high lethality and comorbidity, aggravated by precariousness of long-term Brazilian institutions due to the negligence of public authorities, civil society, the management of the institution and the families of the patients. The post-pandemic scenario will require collective efforts to protect and ensure the survival of the elderly living in those residences.
Resumo Este artigo apresenta uma reflexão sistematizada e discussão em torno de dois eixos orientadores: o primeiro discute o envelhecimento e vulnerabilidades a perdas biológicas, físicas, cognitivas, sociais e afetivas que requerem atenção específica, bem como vulnerabilidades à COVID-19 a que os idosos institucionalizados são expostos; e na segunda, refletimos sobre a adoção de medidas restritivas e protetivas para prevenir a propagação do vírus, com o objetivo de manter a saúde dos idosos e mitigar os efeitos da pandemia. A conclusão é que a pandemia aumentou as muitas vulnerabilidades às quais os idosos institucionalizados já estavam expostos, agregando vulnerabilidade a uma nova doença, como a COVID-19, devido à alta letalidade e comorbidade que representa, agravada pela precariedade das instituições brasileiras de longo prazo devido à negligência do poder público, da sociedade civil, da gestão da instituição e das famílias dos pacientes. O cenário pós-pandemia exigirá esforços coletivos para proteger e garantir a sobrevivência dos idosos que vivem nessas residências.

Resumo Objetivo: Construir e validar quanto ao conteúdo uma terminologia especializada de Enfermagem, no cuidado a adultos vivendo com aids, fundamentado no Modelo de Sete Eixos da Classificação Internacional para a Prática de Enfermagem. Métodos: Estudo metodológico, realizado em um Hospital Escola no Nordeste do Brasil. Seguiram-se as seguintes etapas: extração dos termos de prontuários de pessoas vivendo com aids; normalização; mapeamento cruzado entre os extraídos e os constantes na Classificação Internacional para a Prática de Enfermagem; distribuição destes nos sete eixos e validação de conteúdo por meio de índice de concordância entre enfermeiros peritos. Resultados: Extraíram-se 2.000 termos. A normalização resultou em 557 termos pertinentes, estando 319 constantes e 238 não constantes na Classificação Internacional para a Prática de Enfermagem. Foram validados pelos peritos 522 termos, sendo 319 constantes e 203 não constantes, os quais atingiram um índice de concordância ≥ 0,80. Conclusão: O estudo permitiu identificar e validar os termos utilizados por enfermeiros na assistência às pessoas vivendo com aids, o qual subsidiará as etapas subsequentes à construção de um subconjunto terminológico para informação e comunicação à prática de Enfermagem.
Abstract Objective: To build and validate, in terms of content, a specialized nursing terminology in care of adults infected with AIDS, based on the Seven-Axis Model of the International Classification for Nursing Practice. Method: A methodological study carried out at a teaching hospital in northeastern Brazil. The following steps followed extraction of terms from the medical records of people infected with AIDS; normalization; cross-mapping between extracted and constant in the International Classification for Nursing Practice; distribution in seven axes and content validation through a concordance index among expert nurses. Results: Two thousand terms have been extracted. Normalization resulted in 557 pertinent terms, 319 of which were constant and 238 not included in the International Classification for Nursing Practice. Five hundred and twenty-two terms were validated by experts, of which 319 were constant and 203 were not constant, which reached a concordance index ≥ 0.80. Conclusion: This study allowed to identify and validate the terms used by nurses in assisting people infected with AIDS, which will subsidize the steps subsequent to the construction of a terminological subset for information and communication to nursing practice.
Resumen Objetivo: Elaborar y validar, en cuanto al contenido, una terminología especializada de enfermería en el cuidado de adultos que viven con sida, fundamentada en el Modelo de Siete Ejes de la Clasificación Internacional de la Práctica de Enfermería. Métodos: Estudio metodológico, realizado en un hospital universitario en la región nordeste de Brasil. Se llevaron a cabo las siguientes etapas: extracción de los términos de historias clínicas de personas que viven con sida; normalización; mapeo cruzado entre los términos extraídos y los que constan en la Clasificación Internacional de la Práctica de Enfermería; distribución de estos en los siete ejes y validación de contenido mediante el índice de concordancia por enfermeros peritos. Resultados: Se extrajeron 2.000 términos. La normalización dio como resultado 557 términos pertinentes, de los cuales 319 constaban en la Clasificación Internacional de la Práctica de Enfermería y 238 no. Los peritos validaron 522 términos, de los cuales 319 constaban y 203 no constaban, que alcanzaron un índice de concordancia ≥ 0,80. Mediação de conflito: soluções propostas em atendimento a casos de violência contra a pessoa idosa

Resumo Objetivo Descrever os motivos da violência contra pessoas idosas e as soluções propostas na mediação de conflito em um ambulatório especializado em geriatria e gerontologia do Distrito Federal, Brasil, entre os anos de 2008 a 2018. Método Estudo retrospectivo, documental, descritivo, com abordagem quantitativa, desenvolvido através da análise de informações obtidas em livros-ata da unidade, com registro de reuniões de mediação de conflito de casos de violência contra a pessoa idosa. A coleta abrangeu os motivos de violência contra a pessoa idosa e as soluções propostas na mediação de conflitos. Resultado: Foram analisados 111 casos. Os principais motivos da violência foram: sobrecarga do cuidador principal (77,4%); filhos acharem que os pais idosos eram capazes de se cuidarem sozinhos (27%); ressentimento dos filhos para com o idoso (24,3%); e desconhecimento da doença do idoso (14,4%). As principais soluções propostas foram: acompanhamento regular com médico (82,8%), assistente social e/ou psicólogo (58,5%); comprometimento de todos os filhos com a divisão dos cuidados e despesas dos pais idosos (52,2%); introdução da pessoa idosa em atividades sociais da comunidade (27%); e contratação de um cuidador formal (24,3%). Conclusão A sobrecarga do cuidador foi o principal motivo de conflito encontrado e as propostas apontadas se relacionavam a maior necessidade de cuidados em saúde com a pessoa idosa e seu cuidador, acerca disto, percebeu se a importância da disponibilidade de uma equipe multidisciplinar frente as situações de violência. A mediação de conflito permitiu o estabelecimento de estratégias reais e direcionadas para alcance de resultados frente aos casos de violência.
Abstract Objective To describe the reasons for violence against older people and the solutions proposed for conflict mediation in an outpatient clinic specialized in geriatrics and gerontology in the Federal District, Brazil, between 2008 and 2018. Method A retrospective, documentary, descriptive study with a quantitative approach developed with the analysis of information obtained in the unit’s minutes books via the records of conflict mediation meetings in cases of violence against older people. The collection covered the reasons for violence against older people and the solutions proposed for conflict mediation. Result We analyzed 111 cases. The main reasons for the violence were main caregiver burden (77.4%), children thinking that their older parents were able to take care of themselves (27%), resentment of children towards their older parents (24.3%), and being unaware of the older person’s disease (14.4%). The main solutions proposed were regular follow-up with a doctor (82.8%), social worker and/or psychologist (58.5%), the commitment of all children in sharing care and expenses of their older parents (52.2%), introducing the older person to social activities in the community (27%), and hiring a formal caregiver (24.3%). Conclusion The caregiver burden was the main cause for conflict found, and the proposals identified were related to the greater need for health care for the older person and their caregiver. In this regard, the importance of a multidisciplinary team available in situations of violence was perceived. Conflict mediation made it possible to establish real and targeted strategies to achieve results in cases of violence.

Abstract The objective of this study was to assess and compare two methodologies to predict the mortality of individual trees in a seasonal semi-deciduous forest located in the municipality of Viçosa, Minas Gerais, Brazil. The data came from 10 permanent plots and were randomly divided into two groups: fitting (6 plots) and validation (4 plots). When assessing Methodology 1, the mortality was considered a dichotomous variable in a logistic model, assuming a value of 1 for a dead tree and 0 for a living tree. In Methodology 2, the mortality probability was estimated by the group of trees for each plot and for each measurement interval. After the analysis, it was observed that Methodology 2 provided more consistent mortality estimates when compared to the mortality estimated by Methodology 1.
