Abstract Aiming to compare and update the sand fly fauna of Portão de Pedra site, Sumidouro District, Rio de Janeiro State, Brazil, and considering the environmental changes occurred, the biology and ecology of the local sandfly species were examined five years later as a complementary study carried. Captures were made in the cave, surroundings of cave and forest of the region, from 6 p.m. to 6 a.m. Among the 2323 sandflies of eigth species of the Lutzomyia were captured: L. gasparviannai, L. edwardsi, L. tupynambai, L. hirsuta, L. whitmani, L. migonei, L. intermedia, Lutzomyia. sp and one species of the Brumptomyia Kind: B. brumpti. In 2009 and 2010 were collected 1756 samples from ten species of the former genus and two of the second. L. gasparviannai was predominant, in the three collection sites, in both periods. Five species implicated as vectors of Leishmania: L. intermedia, L. whitmani, L. migonei, L. hirsuta and L. davisi have been collected in the area. Poisson regression and ANOVA were used to perform statistical analysis of species most relevant. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract Obesity, associated with the intake of a high-fat diet (HFD), and anxiety are common among those living in modern urban societies. Recent studies suggest a role of microbiome-gut-brain axis signaling, including a role for brain serotonergic systems in the relationship between HFD and anxiety. Evidence suggests the gut microbiome and the serotonergic brain system together may play an important role in this response. Here we conducted a nine-week HFD protocol in male rats, followed by an analysis of the gut microbiome diversity and community composition, brainstem serotonergic gene expression (tph2, htr1a, and slc6a4), and anxiety-related defensive behavioral responses. We show that HFD intake decreased alpha diversity and altered the community composition of the gut microbiome in association with obesity, increased brainstem tph2, htr1a and slc6a4 mRNA expression, including in the caudal part of the dorsomedial dorsal raphe nucleus (cDRD), a subregion previously associated with stress- and anxiety-related behavioral responses, and, finally, increased anxiety-related defensive behavioral responses. The HFD increased the Firmicutes/Bacteroidetes ratio relative to control diet, as well as higher relative abundances of Blautia, and decreases in Prevotella. We found that tph2, htr1a and slc6a4 mRNA expression were increased in subregions of the dorsal raphe nucleus in the HFD, relative to control diet. Specific bacterial taxa were associated with increased serotonergic gene expression in the cDRD. Thus, we propose that HFD-induced obesity is associated with altered microbiome-gut-serotonergic brain axis signaling, leading to increased anxiety-related defensive behavioral responses in rats.

Chagas disease, caused by the parasite Trypanosoma cruzi, occurs most commonly in Latin America. As the treatment is highly toxic and ineffective in the chronic phase of the disease, alternative treatments are needed. Through quantitative structure-activity relationship modeling (QSAR) analysis using ligand-based and structure-based virtual screening methods, we predicted the trypanocidal potential of 47 neolignans against three targets, the enzymes cruzain, trypanothione reductase, and sterol 14-alpha demethylase. A combined analysis allowed for the selection of potent inhibitors against Trypanosoma cruzi. Of these compounds, two were isolated and shown to inhibit the growth of epimastigotes at concentrations of 9.64 and 8.72 µM, and trypomastigote forms at 4.88 and 2.73 µM. ABSTRACT Purpose: To evaluate the modulatory properties of Calendula officinalis L. (Asteraceae) (C. officinalis) extract on cafeteria diet-fed rats. Methods: A cafeteria diet was administered ad libitum for 45 days to induce dyslipidemia. Then, the rats were treated with the formulations containing C. officinalis in the doses of 50, 100, and 150 mg/kg or only with the vehicle formulation; the control group received a commercial ration. Results: The cafeteria diet decreased glutathione S-transferase activity and high-density lipoprotein plasmatic levels and damaged the hepatic architecture. The C. officinalis extract was able to reduce lipid infiltration in liver tissue and to modulate oxidative stress and lipid profile markers. Conclusions: The correlations between the variables suggest a pathological connection between oxidative stress markers and serum lipid profile. Abstract Since the first reported case of COVID-19 in Brazil, the public and private educational system started to close. Up to November 2020, scientific discussions about the return of schooling activities have been rarely performed by the national scientific community and police-makers. The great delay of school returning in Brazil contrasts with successful international strategies of school reopening worldwide and seems counterintuitive with the reopening of non-essential activities. Here, important issues to be considered before and during school reopening are reviewed and discussed. COVID-19 testing is essential to avoid disease spreading, but high cost of individual RT-qPCRs impairs an extensive testing strategy for school returning. To reduce costs and increase the speed of diagnosis, we tested the efficiency of a pooled-sample PCR strategy in a cohort of the educational staff in the city of Macaé/RJ, finding five asymptomatic individuals (0,66%) among the 754 people tested. Thus, a polled-sample PCR testing strategy of the educational staff might prevent infection spreading in schools at a reasonable cost. We discuss how our test strategy could be coupled with internationally recognized safety rules to allow for a safe school return and how countries from different world regions are dealing with educational activities during COVID-19 pandemic.

Abstract This study investigated the renal function of soccer players after an entire game-season. Thirty-five athletes recruited to play for the Macae Futebol Clube were invited for this study, of which 18 athletes completed the entire game season. Blood and 24-hour urine were collected at the beginning (Pre-Season) and the end of the game season (Post-Season). Kidney functions were assessed by calculating the urinary excretion, clearance, and fractional excretion of the selected solutes. Plasma creatinine, sodium, total protein, and osmolality were lower in the Post-Season . In contrast, plasma urea was higher in the Post-Season period. Urinary excretion of urea was reduced while albumin excretion was higher in comparison to Pre-Season. The clearances of creatinine, total proteins, and albumin were higher in the Post-Season period. In accordance, the fractional excretion of albumin increased. On the other hand, the clearance and fractional excretion of urea was lower in the Post-Season period. These results show that soccer-associated exercise throughout the entire game-season induces kidney functions adaptations that may prevent dehydration in these athletes through increased urea reabsorption to conserve water. In addition, this data corroborates to increased glomerular permeability to plasma proteins, such as albumin, that soccer players may experience.

Resumo O BRAMS-SFIRE é um novo sistema de modelação atmosférica com componente de propagação de fogo desenvolvido no Centro de Previsão de Tempo e Estudos Climáticos (CPTEC / INPE) no Brasil em colaboração com o Instituto Mediterrâneo de Agricultura, Ambiente e Desenvolvimento (MED) em Portugal. O presente artigo descreve a incorporação do modelo de propagação de fogo no Brazilian developments on the Regional Atmospheric Modeling System (BRAMS). Os principais objetivos foram desenvolver o acoplamento entre um modelo atmosférico e um modelo de propagação de fogo que simulasse os efeitos da circulação atmosférica sobre um incêndio florestal e vice-versa. Esta ferramenta tem como objetivo ajudar a entender as relações entre o fogo e a paisagem nas florestas de carvalho mediterrâneas, e avaliar os resultados das simulações deste acoplamento na escala fina no ecossistema do Montado na região do Alentejo. Para isso, três grades de alta resolução espacial ao longo de três incêndios foram configuradas com dados de caracterização de superfície realistas e propriedades dos modelos de combustível. Uma grade foi colocada sobre fogo em uma planície e as outras nas montanhas, para avaliar os diferentes tipos de propagação do fogo. Este trabalho demonstra que este sistema simula de forma consistente a interação entre o fogo, os modelos de combustível e a atmosfera, mostrando que o fogo altera a circulação local ao nível da superfície, intensifica as correntes de vento ascendentes e descendentes, alterando a estrutura da atmosfera.
Abstract BRAMS-SFIRE is a new atmospheric modeling system with a fire spreading component developed at the Centro de Previsão de Tempo e Estudos Climáticos (CPTEC/INPE) in Brasil, in collaboration with the Instituto Mediterrâneo para a Agricultura, Ambiente e Desenvolvimento (MED) in Portugal. The present paper describes the incorporation of the fire model into the Brazilian development of the Regional Atmospheric Modelling System (BRAMS). The main objectives were to develop the coupling between an atmospheric and a spreading fire models which simulate the effects of atmospheric circulation over a wildfire and vice-versa. This tool is intended to help understand fire-landscape relationships in Mediterranean oak woodlands and evaluate the simulation results on a fine-scale in the Alentejo region's Montado ecosystem. For this purpose, three grids of very high spatial resolution over three fires were configured with realistic surface characterization data and fuel model properties. One grid was placed at a fire in the plains and the others in the mountains to evaluate fire propagation types. This work demonstrates that this system consistently simulated the interaction between the fire, the fuel models, and the atmosphere, showing the fire changes the local circulation at the surface level, intensify wind currents, and changes the atmosphere structure.

Abstract In canine visceral leishmaniasis, coinfections can aggravate the disease. Our aim was to investigate Brucella canis in dogs infected with Leishmania infantum. One hundred and six L. infantum-seropositive dogs were submitted to serology for B. canis, PCR for B. canis and L. infantum, and histopathological analysis of the genital tract. Anti-B. canis antibodies were detected in seven dogs whose clinical signs, L. infantum load and histological alterations were similar to those of seronegative animals. The circulation of anti-B. canis antibodies was low but demonstrates the exposure of dogs to this bacterium in a visceral leishmaniasis-endemic area.

This work presents the application of a simple and sensitive method developed for the simultaneous analysis of the levels of twenty two underivatizated free L-amino acids in plasma samples of preeclampsia patients by high performance liquid chromatography coupled to high-resolution mass spectrometer (LC-HRMS) using hydrophilic interaction liquid chromatography (HILIC) silica column. The linearity was evaluated in the range of 0.009 to 8.018 µmol L-1 showing coefficients of determination (R2) in the range of 0.999 to 0.911 (p < 0.05). The intra- and inter-day precisions were above of 9.84 and 15.31, respectively. The limits of detection (LOD) and quantification (LOQ) were in the range of 0.003 to 0.683 µmol L-1 and 0.009 to 2.276 µmol L-1, respectively. The results of the investigation of free L-amino acids showed that the levels of GABA, Arg, Ser, Leu, Ile, Met, Val, Ala, Gly, Bet, His and Lys in plasma of patients with preeclampsia were significantly different from healthy patients.

ABSTRACT Resident and circulating immune cells have been extensively studied due to their almost ubiquitous role in cell biology. Despite their classification under the “immune cell department”, it is becoming increasingly clear that these cells are involved in many different non-immune related phenomena, including fetus development, vascular formation, memory, social behavior and many other phenotypes. There is a huge potential in combining high-throughput assays - including flow cytometry and gene analysis - with in vivo imaging. This can improve our knowledge in both basic and clinical cell biology, and accessing the expression of markers that are relevant in the context of both homeostasis and disease conditions might be instrumental. Here we describe how we generated a novel mouse strain that spontaneously express three different fluorescence markers under control of well-studied receptors (CX3CR1, CCR2 and CD11c) that are involved in a plethora of stages of cell ontogenesis, maturation, migration and behavior. Also, we assess the percentage of the expression and co-expression of each marker under homeostasis conditions, and how these cells behave when a local inflammation is induced in the liver applying a cutting-edge technology to image cells by confocal intravital microscopy.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Lesões pulmonares associadas ao parasitismo por Sebekia oxycephala (Pentastomida) em jacarés-açu (Melanosuchus niger Spix, 1825) oriundos de vida livre na Amazônia brasileira

O presente estudo objetivou descrever as lesões pulmonares macro e microscópicas associadas ao parasitismo por Sebekia oxycephala em 100 espécimes de jacarés-açu (Melanosuchusniger), abatidos na Reserva de Desenvolvimento Sustentável Mamirauá, situada no Estado do Amazonas, Brasil. Durante a avaliação macroscópica, exemplares dos parasitos encontrados no tecido pulmonar foram coletados individualmente em AFA (Álcool etílico - Formalina - Ácido acético glacial) e formol a 5% para avaliação parasitológica e classificação taxonômica. Amostras de tecido pulmonar de todos os animais, independentemente da ocorrência de lesões macroscópicas, foram fixadas em formol 10% e incluídas em parafina. Secções histológicas coradas por Hematoxilina-Eosina destas amostras foram avaliadas por meio de microscopia óptica. Macroscopicamente, 4 dos 100 animais (4%) apresentaram espécimes de pentastomídeos no parênquima pulmonar. Os parasitos foram classificados taxonomicamente como pertencentes à espécie Sebekia oxycephala. Nenhuma alteração macroscópicas foi observada, porém, microscopicamente, lesões pulmonares foram encontradas em 37% dos casos, sendo que, lesões inflamatórias associadas ao parasitismo corresponderam a 75,6% dos mesmos (28/37). Nestes, segmentos degenerados e ovos de S. oxycephala encontravam-se envolvidos por cápsula de tecido conjuntivo fibroso e infiltrado inflamatório predominantemente composto por células gigantes do tipo corpo estranho. Três espécimes apresentaram espessamento de septos alveolares e sete exemplares continham infiltrado inflamatório granulocítico multifocal no parênquima pulmonar. As lesões associadas ao parasitismo, de modo geral, apresentaram intensidade discreta e parecem não representar uma causa importante de doença pulmonar entre a população estudada. Esta é a primeira descrição de lesões pulmonares em M. niger associadas ao parasitismo por S. oxycephala na Amazônia brasileira.
The goal of this study was to describe gross and microscopic lung lesions associated with the parasitism by Sebekia oxycephala between 100 specimens of black caiman (Melanosuchus niger) slaughtered in the Mamirauá Sustainable Development Reserve, situated in the Amazonas State, Brazil. During the macroscopic evaluation, specimens of parasites found in the lung tissue were collected individually in AFA (ethyl alcohol - formalin - glacial acetic acid) and 5% formalin for parasitological evaluation and taxonomic classification. Lung tissue samples of all the animals, regardless of the occurrence of gross lesions, were fixed in 10% formalin and embedded in paraffin. Histological sections obtained from the samples stained by Hematoxylin-Eosin were evaluated by light microscopy. Grossly, 4 of the 100 animals (4%) presented pentastomid specimens in the lung parenchyma. These parasites were taxonomically classified as Sebekia oxycephala. No macroscopic alterations were observed, however microscopically pulmonary lesions were found in 37% of the cases, and inflammatory lesions associated with the parasitism corresponded to 75.6% of them (28/37). In these cases, degenerate segments and eggs of S. oxycephala were surrounded by a fibrous capsule and inflammatory infiltrate composed predominantly by foreign body giant cells. Three specimens exhibited thickening of the alveolar septa and seven specimens presented multifocal inflammatory granulocytic infiltration in the lung parenchyma. Generally, the associated parasitic lesions had mild intensity and did not appear to represent an important cause of lung disease in the studied population. This is the first description of lung lesions in M. niger associated with parasitism by S. oxycephala in the Brazilian Amazon.

Carros de emergencia: disponibilidad de los artículos esenciales en un hospital de urgencia norteriograndense

It is a descriptive, exploratory and quantitative study that evaluates, by the standards of the Brazilian Society of Cardiology (SBC), the availability of the items of the Level I Emergency Cars (EC) of the largest emergency hospital in Rio Grande do Norte. Data were collected during the month of November 2010, the sample consists of 12 EC. All EC analyzed showed gaps in availability of essential items, being the greatest deficiencies related to materials intended for vascular access, circulation control and drugs aimed at treating the causes of cardiac arrest. It was felt institution, as well as a protocol, difficult routine check of the EC, the nurse assigned task.
Trata-se de uma pesquisa descritiva, exploratória e quantitativa com o objetivo de avaliar, sob os padrões da Sociedade Brasileira de Cardiologia (SBC), a disponibilidade dos itens de nível I dos Carros de Emergência (CE) do maior hospital de urgência do Rio Grande do Norte. Os dados foram coletados durante o mês de novembro de 2010, sendo a amostra composta por 12 CE. Todos os CE analisados apresentaram falhas na disponibilidade de itens essenciais, estando as maiores deficiências relacionadas aos materiais destinados ao acesso vascular, controle circulatório e medicamentos voltados ao tratamento das causas de PCR. Percebeu-se que a falta de recursos humanos e materiais na instituição, bem como de um protocolo, dificulta a rotina de checagem do CE, tarefa atribuída ao enfermeiro.
Se trata de una investigación descriptiva, exploratoria y cuantitativa con el objetivo de evaluar, bajo los patrones de la Sociedad Brasileña de Cardiología (SBC), la disponibilidad de los artículos de nivel I de los Carros de Emergencia (CE) del mayor hospital de urgencia de Rio Grande do Norte. Los datos fueron colectados durante el mes de noviembre de 2010, estando compuesta la muestra por 12 CE. Todos los CE analizados presentaron fallos en la disponibilidad de artículos esenciales, estando las mayores deficiencias relacionadas con los materiales destinados al acceso vascular, control circulatorio y medicamentos dirigidos al tratamiento de las causas de PCR. Se percibe que la falta de recursos humanos y materiales en la institución, así como de un protocolo, dificulta la rutina de comprobación del CE, tarea atribuída al enfermero.
