In this study, we aimed to investigate the anticancer activity of the extract, fractions, and isolated compounds of the flowers of Fridericia platyphylla, and to characterize the bioactive compounds. The chemical diversity of the extracts and fractions was investigated using liquid chromatography coupled with high resolution mass spectrometry (LC-HRMS). We were able to annotate 26 compounds from the classes of flavones, flavonols, flavanones, isoflavones, and cinnamic acid and its derivatives. The dichloromethane fraction showed greater cytotoxicity at a concentration of 100 µg mL−1. In addition, the inhibitory concentrations of the dichloromethane fraction were 22.14 and 30.9 µg mL−1 for MCF-7 and MDA-MB-231 cell lines, respectively, and were capable of inhibiting tumor cell migration. RESUMO Os modelos de balanço hídrico do solo podem ser uma boa opção para estudar o crescimento das culturas e as respostas de rendimento sob diferentes níveis de fornecimento de água. O objetivo deste estudo foi avaliar os efeitos de diferentes estratégias de irrigação na produtividade do girassol e na produtividade da água usando experimentos de campo e simulações de balanço hídrico do solo. Dois experimentos de campo foram realizados. Em 2018/19, foram utilizadas quatro estratégias de irrigação (mantida a água disponível no solo em 80, 70, 60 e 50% da água total disponível). Em 2020/21 foi utilizado um tratamento irrigado e de sequeiro. O modelo SIMDualKc foi calibrado e validado para simular o balanço hídrico do solo e derivar os coeficientes duais usando variáveis da planta a partir de observações de campo. ABSTRACT Soil water balance models can be a good option for studying crop growth and yield responses under different water supply levels. The aim of this study was to evaluate the effect of different irrigation strategies on sunflower yield and water productivity using field experiments and soil water balance simulations. Two field experiments were performed. In 2018/19, four irrigation strategies (aimed at maintaining soil available water at 80, 70, 60, and 50% of total available water) were evaluated. In 2020/21 an irrigated and a rain-fed treatment were evaluated. The SIMDualKc model was calibrated and validated to simulate soil water balance and derive the dual coefficients using plant variables from field observations. RESUMO Objetivou-se com esta pesquisa determinar e avaliar o comportamento da forma e do tamanho do feijão mungo durante a secagem. O feijão mungo tem sido destaque no meio científico e agronômico devido suas propriedades nutricionais, medicinais e produtivas, e o conhecimento de suas propriedades físicas em diferentes teores de água é fundamental para otimização dos processos de plantio, colheita e pós-colheita. Os grãos foram colhidos com o teor de água de aproximadamente 27% (base úmida, bu), e posteriormente submetidos à secagem artificial com ventilação natural, até atingirem o teor de água de aproximadamente 9% (bu). Ao longo do processo de secagem a forma e o tamanho dos grãos foram avaliados mediante determinação dos eixos ortogonais, comprimento, largura e espessura em 30 grãos por repetição. De posse desses dados foram calculados a circularidade, esfericidade, volume, área superficial, área projetada, relação superfície-volume e diâmetro geométrico. ABSTRACT This study investigates the impact of drying on the shape and size of mung beans (Vigna radiata L.), which are well-regarded for their nutritional, medicinal, and agronomic benefits. Understanding their physical properties at varying moisture levels is crucial for improving agricultural practices from planting through post-harvest. Initially harvested at about 27% moisture content (wet basis, wb), the beans were air-dried naturally to approximately 9% moisture content (wb). The analysis involved measuring the orthogonal axes, length, width, and thickness of 30 representative beans. Resumo A melhoria da germinação de plantas é fundamental para garantia de mudas de melhor qualidade. Assim, objetivou-se avaliar se o condicionamento de sementes com qualidade de luz (light quality - LIQ) e extrato aquoso de tubérculos de Cyperus rotundus (AEC) modula a germinação e o crescimento inicial de plântulas de Moringa oleifera. Utilizou-se delineamento inteiramente casualizado, em esquema fatorial 4x4, sendo quatro condições de LIQ (luz branca, azul, vermelha e vermelho distante) e quatro concentrações de AEC (0, 25, 50 e 100%). O condicionamento de sementes com luz vermelha reduziu o tempo médio de emergência, enquanto que as luzes azul, vermelha e vermelho extremo associadas a 50% de extrato aquoso de C. rotundus aumentaram o comprimento inicial da parte aérea e o acúmulo de pigmentos fotossintéticos. Condicionamento de sementes com luz azul induziu a formação de plântulas com menor comprimento final da parte aérea, no entanto, a aplicação de 100% de extrato aquoso de C. rotundus reverteu o menor crescimento. Abstract Improving plant germination is essential to guarantee better quality seedlings. Thus, this research aimed to evaluate whether the seed priming with light quality (LIQ) and the aqueous extract of Cyperus rotundus (AEC) tuber could modulate the germination and initial growth of Moringa oleifera L. seedlings. The experimental design was a completely randomized in the 4x4 factorial scheme, composed of four LIQ conditions (white, blue, red, and distant red light) and four AEC concentrations (0, 25, 50 and 100%). Seed priming with red light reduced the average emergence time, while blue, red, and extreme red lights associated with 50% of aqueous extract of C. rotundus increased shoot initial length and photosynthetic pigment accumulation. Seed priming with blue light resulted in seedlings with a shorter final shoot length. However, application of 100% of aqueous extract of C. rotundus reversed this. The white light in combination with concentrations of 50 and 100% of AEC promoted a higher relative shoot growth rate of seedlings. The research revealed that seed priming with light quality and aqueous extracts of C. rotundus tubers modulates the germination and initial growth of M. oleifera seedlings. More work needs to be done to determine the responsible compounds in AEC that is responsible for priming growth as phytohormones.

Resumo Os microRNAs (miRNAs) são reconhecidos como biomarcadores do diabetes mellitus tipo 2 (DM2), úteis para a compreensão do metabolismo da doença, e possuem grande potencial como alvos terapêuticos. O aumento da expressão de BDNF e IGF1 está altamente envolvido nos benefícios as vias de insulina e glicose, porém, são regulados negativamente em condições de resistência à insulina, enquanto seu aumento de expressão está correlacionado com a melhora do metabolismo da glicose e da insulina. Estudos sugerem a regulação desses genes por microRNA em vários contextos diferentes, proporcionando uma nova abordagem de investigação para compreender o metabolismo do DM2 e revelar potenciais alvos terapêuticos. No presente estudo, investigamos em diferentes modelos animais (humanos, ratos e camundongos) miRNAs que têm como alvo BDNF e IGF1 em tecido muscular esquelético com condições fisiológicas de DM2. As análises foram realizadas utilizando ferramentas de bioinformática e bancos de dados para predição de miRNA, homologia molecular, validação experimental de interações, expressão na condição fisiológica estudada e interação em rede. Os resultados mostraram três candidatos a miRNAs para IGF1 (miR-29a, miR-29b e miR-29c) e um para BDNF (miR-206). As avaliações experimentais e a busca pela expressão no músculo esquelético de indivíduos com DM2 confirmaram a interação prevista entre miRNA-mRNA para miR-29b e miR-206 através de modelos humanos, ratos e camundongos. Essa interação foi reafirmada em múltiplas análises de rede. Em conclusão, nossos resultados mostram a relação de regulação entre miR-29b e miR-206 com os genes investigados, em diversos tecidos, sugerindo um padrão de inibição. Contudo, esses dados mostram um grande número de possíveis processos fisiológicos de interação para perspectivas biotecnológicas.

Abstract microRNAs (miRNAs) are recognized as diabetes mellitus type 2 (T2DM) biomarkers useful for disease metabolism comprehension and have great potential as therapeutics targets. BDNF and IGF1 increased expression are highly involved in the benefits of insulin and glucose paths, however, they are down-regulated in insulin resistance conditions, while their expression increase is correlated to the improvement of glucose and insulin metabolism. Studies suggest the microRNA regulation of these genes in several different contexts, providing a novel investigation approach for comprehending T2DM metabolism and revealing potential therapeutic targets. In the present study, we investigate in different animal models (human, rat, and mouse) miRNAs that target BDNF and IGF1 in skeletal muscle tissue with T2DM physiological conditions. Bioinformatics tools and databases were used to miRNA prediction, molecular homology, experimental validation of interactions, expression in the studied physiological condition, and network interaction. The findings showed three miRNAs candidates for IGF1(miR-29a, miR-29b, and miR-29c) and one for BDNF (miR-206). The experimental evaluations and the search for the expression in skeletal muscle from T2DM subjects confirmed the predicted interaction between miRNA-mRNA for miR-29b and miR-206 through human, rat, and mouse models. This interaction was reaffirmed in multiple network analyses. In conclusion, our results show the regulation relationship between miR-29b and miR-206 with the investigated genes, in several tissues, suggesting an inhibition pattern. Nevertheless, these data show a large number of possible interaction physiological processes, for future biotechnological prospects.

Resumo A hibernação é uma condição natural dos animais que vivem na zona temperada, embora alguns lagartos tropicais também experenciem hibernação anualmente, como é o caso do lagarto nativo da América do Sul, Salvator merianae ou “teiú”. Embora as características fisiológicas e metabólicas associadas à hibernação tenham sido amplamente estudadas, possíveis alterações na integridade das hemácias durante esse período ainda permanecem obscuras. A desidratação e o jejum são consequências naturais da hibernação por vários meses e podem estar relacionadas a algumas modificações celulares. Neste estudo, investigamos se a tolerância osmótica de hemácias do lagarto teiú sob hibernação são diferentes das células obtidas de animais em atividade normal. Além disso, investigamos indiretamente por meio da quantificação de biomoléculas oxidadas e da atividade de enzimas antioxidantes se a membrana das hemácias dos teiús em hibernação poderia estar associada à oxidação. Nossos resultados sugerem que as hemácias possuem maior fragilidade durante o período de hibernação, embora não tenhamos encontrado evidências de um cenário de estresse oxidativo associado à essa fragilidade acentuada. Embora não tenhamos excluído a possibilidade de dano oxidativo durante a hibernação, sugerimos que um aumento no volume das hemácias como consequência de sangue hipoosmótico durante a hibernação também poderia afetar a integridade de hemácias, tal como foi observado.

Abstract Hibernation is a natural condition of animals that lives in the temperate zone, although some tropical lizards also experience hibernation annually, such as the lizard native from South America, Salvator merianae, or “tegu” lizard. Even though physiological and metabolic characteristic associated with hibernation have been extensively studied, possible alterations in the red blood cells (RBC) integrity during this period remains unclear. Dehydration and fasting are natural consequences of hibernating for several months and it could be related to some cellular modifications. In this study, we investigated if the osmotic tolerance of RBCs of tegu lizard under hibernation is different from the cells obtained from animals while normal activity. Additionally, we indirectly investigated if the RBCs membrane of hibernating tegus could be associated with oxidation by quantifying oxidized biomolecules and the activity of antioxidant enzymes. Our findings suggest that RBCs are more fragile during the hibernation period, although we did not find evidence of an oxidative stress scenario associated with the accentuated fragility. Even though we did not exclude the possibility of oxidative damage during hibernation, we suggested that an increased RBCs volume as a consequence of hypoosmotic blood during hibernation could also affect RBCs integrity as noted.

Reports on freeze-cast ceramic materials frequently focus on the study of the organized macroporosity and the properties of the materials. This study aims to describe the microstructure evolution of freeze-cast alumina during the sintering process, analyzing grain growth, densification, pore elimination and crystal structure at different sintering temperatures (1300-1500 °C). Aqueous suspensions with 20 vol% alumina were freeze-cast in liquid N2 and sintered. The microstructure was analyzed by stereological analysis, N2 adsorption and X-ray diffraction. Grain sizes varied within 237-500 nm, and the intergranular porosity decreased from 8.8% at 1300 °C to 1.4% at 1500 °C. N2 isotherm analysis revealed pore shrinking from the region of macro and mesopores (20-80 nm), to smaller residual mesopores (3.7-15 nm) at temperatures above 1400 °C. Rietveld refinement of the XRD diffractograms confirmed increased crystallite size and decreased lattice strain at higher sintering temperatures. ABSTRACT Purpose: To assess Meibomian gland dysfunction using meibography in patients with xeroderma pigmentosum and correlate with ocular surface changes. Methods: This cross-sectional study evaluated patients with xeroderma pigmentosum. All patients underwent a comprehensive and standardized interview. The best-corrected visual acuity of each eye was determined. Detailed ophthalmic examination was conducted, including biomicroscopy examination of the ocular surface, Schirmer test type I, and meibography, and fundus examination was also performed when possible. Meibomian gland dysfunction was assessed by non-contact meibography using Oculus Keratograph® 5M (OCULUS Inc., Arlington, WA, USA). Saliva samples were collected using the Oragene DNA Self-collection kit (DNA Genotek Inc., Ottawa, Canada), and DNA was extracted as recommended by the manufacturer. Factors associated with abnormal meiboscores were assessed using generalized estimating equation models. Results: A total of 42 participants were enrolled, and 27 patients underwent meibography. The meiboscore was abnormal in the upper eyelid in 8 (29.6%) patients and in the lower eyelid in 17 (62.9%). The likelihood of having abnormal meiboscores in the lower eyelid was 16.3 times greater than that in the upper eyelid. In the final multivariate model, age (p=0.001), mutation profile (p=0.006), and presence of ocular surface malignant tumor (OSMT) (p=0.014) remained significant for abnormal meiboscores. For a 1-year increase in age, the likelihood of abnormal meiboscores increased by 12%. Eyes with OSMT were 58.8 times more likely to have abnormal meiboscores than eyes without ocular surface malignant tumor. Conclusion: In the final model, age, xeroderma pigmentosum profile, previous cancer, and clinical alterations on the eyelid correlated with a meiboscore of ≥2. ABSTRACT This article discusses some aspects of the process of political emigration resulting from the dynamics between revolution and counterrevolution in Portugal in the 1820s and 1830s. Although the direction of the exiles varied according to the outcome of the successive disputes that took place in the country between royalists and liberals, it was when D. Resumo Foi avaliada a eficácia larvicida do lotilaner (Credeli®) no tratamento de cães naturalmente infestados com larvas de Dermatobia hominis. Foram medicados 12 cães, apresentando pelo menos três larvas vivas de D. hominis. Os animais foram medicados por via oral com dose única não inferior a 20 mg/kg de lotilaner. Após a administração desse medicamento, os animais permaneceram em seus domicílios, e foram feitas observações para verificar o efeito larvicida 6 horas após o tratamento. Larvas vivas foram consideradas quaisquer parasitas que exibissem motilidade após a remoção. Para cada animal foi utilizada a fórmula: 100 x [(total de larvas vivas antes do tratamento - total de larvas vivas após o tratamento) /total de larvas vivas antes do tratamento] como critério para avaliar a eficácia do lotilaner. Foram contadas 98 larvas em 12 cães, com média de 8,1 larvas por animal. A eficácia do lotilaner foi de 80,6%. Dezenove larvas foram encontradas vivas, porém apresentando hipomobilidade e comportamento letárgico. Abstract An evaluation was made of the larvicidal efficacy of lotilaner (Credeli®) in the treatment of dogs naturally infested with Dermatobia hominis larvae. A total of 12 dogs presenting at least three live D. hominis larvae were medicated. The animals were medicated orally with a single dose of no less than 20 mg/kg lotilaner. After drug administration, the animals remained at their homes, and observations were made to verify the larvicidal effect 6 hours after treatment. Live larvae were considered any parasite that exhibited motility after removal. RESUMO O objetivo deste estudo foi avaliar o efeito de diferentes níveis de ácidos graxos poli-insaturados ômega-3 (PUFA n-3) na dieta de Sardinella brasiliensis sobre seus parâmetros hematológicos. ABSTRACT This study aimed to evaluate the effect of different levels of omega-3 polyunsaturated fatty acids (n-3 PUFA) in the diet of Sardinella brasiliensis on their hematological parameters. A total of 375 fish were distributed in 15 experimental units to evaluate the effects of the diet on their blood for 45 days. The levels of n-3 PUFA added in the diet were 0, 0.3, 0.6, 0.9, and 1.2% of the lipid fraction. These values represent the total percentage of n-3 PUFA in the lipid fraction of the diet, and 0% n-3 PUFA was used as the negative control. Five fish from each experimental unit were sampled at the end of the experiment for hematological analysis, and the parameters measured include the total number of erythrocytes, hematocrit percentage, hemoglobin and glucose concentration and differential leukocyte count. Resumo O feijão-caupi é uma planta leguminosa pertencente à família das Fabaceae cultivada nas regiões Norte e Nordeste do Brasil, com potencial produtivo. Entre os fatores abióticos, a deficiência hídrica é uma das principais limitações ambientais que influenciam a produção agrícola no mundo. O objetivo do trabalho foi estudar o conteúdo relativo de água e os osmorreguladores de plantas de feijão-caupi submetidas ao estresse hídrico. O experimento foi conduzido em casa de vegetação da Universidade Federal Rural da Amazônia (UFRA, Belém, PA), foram utilizadas plantas de feijão-caupi BR-17 Gurguéia Vigna unguiculata (L.) Walp. O delineamento experimental foi inteiramente casualizado (DIC) em esquema fatorial 2 × 2, duas condições hídricas (controle e déficit hídrico) e dois tempos de estresse (quatro e seis dias de suspensão hídrica), com 7 repetições, totalizando 28 unidades experimentais. Abstract Cowpea is a leguminous plant belonging to the fabaceae family cultivated in the North and Northeast regions of Brazil, with productive potential. Among the abiotic factors, water deficiency is one of the main environmental limitations that influence agricultural production in the world. The objective of this work was to study the relative water content and osmoregulators of cowpea plants subjected to water stress. The experiment was carried out in a greenhouse at the Universidade Federal Rural da Amazônia (UFRA, Belém, PA), cowpea plants BR-17 Gurguéia Vigna unguiculata (L.) Walp were used. The experimental design was completely randomized (DIC) in a 2 × 2 factorial scheme, two water conditions (control and water deficit) and two times of stress (four and six days of water suspension), with 7 replications, totaling 28 experimental units. The water deficit affected plants, causing a reduction in relative water content (69.98%), starch (12.84% in leaves and 23.48% in roots) and carbohydrates (84.34%), and an increase in glycine-betaine, sucrose (114.11% in leaves and 18.71% in roots) and proline (358.86%) at time 2. The relative water content was negatively affected by water conditions, with a decrease in relation to the interaction of the aerial part and the root system. Resumo Fundamento Não existem estudos randomizados comparando a manutenção do ritmo sinusal após ablação por cateter (AC) em relação ao tratamento com fármacos antiarrítmicos (AA) em pacientes idosos portadores fibrilação atrial (FA) paroxística. Objetivos Comparar os resultados clínicos do isolamento das veias pulmonares (VPs) com o cateter PVAC Gold de segunda geração com o uso de AA em idosos com FA paroxística sintomática, recorrente, apesar do uso de fármacos AA. Métodos Sessenta pacientes com FA paroxística ≥ 65 anos e sem cardiopatias estruturais foram randomizados para duas formas de tratamento: grupo 1: AC e grupo 2: AA. O desfecho primário foi a taxa livre de recorrência de FA após pelo menos um ano de seguimento. Os desfechos secundários foram: progressão para formas persistentes de FA, impacto na qualidade de vida (QVFA) e complicações. O nível de significância adotado na análise estatística foi de 5% (p<0,05). Resultados A taxa livre de recorrência de FA foi de 80% (10% com amiodarona) no grupo AC, após 1,3 procedimentos por paciente e de 65% no grupo AA (60% com amiodarona), (p = 0,119) num seguimento médio de 719 dias (Q1: 566; Q3: 730). A taxa livre de FA persistente foi de 83,4% no grupo AC e de 67,7% no grupo AA (p = 0,073). Ambas as estratégias apresentaram melhora no escore de QVFA durante o seguimento (p < 0,001), sem diferença entre os grupos. Abstract Background There are no randomized studies comparing the maintenance of sinus rhythm after catheter ablation (CA) concerning treatment with antiarrhythmic drugs (AA) in elderly patients with paroxysmal atrial fibrillation (AF). Objectives To compare the clinical results of pulmonary vein (PV) isolation with the second-generation PVAC Gold catheter against AA treatment in elderly people with recurrent symptomatic paroxysmal AF, refractory to at least one AA, and without structural heart disease. Methods Sixty patients with paroxysmal AF ≥ 65 years old were randomized to two forms of treatment: group 1: CA and group 2: AA drugs. The primary outcome was the AF recurrence-free rate after at least one year of follow-up. Secondary outcomes were: progression to persistent forms of AF, impact on quality of life (QOLF), and complications. The significance level adopted in the statistical analysis was 5% (p<0.05). Results The AF recurrence-free rate was 80% (10% with amiodarone) in the CA group, after 1.3 procedures per patient and 65% in the AA group (60% with amiodarone), (p = 0.119) in an average follow-up of 719 days (Q1: 566; Q3: 730).
