Resumen En esta nota técnica, presentamos uno de los principales legados empíricos de la investigación sobre la publicidad televisiva de las candidatas/de los candidatos a diputado federal en el Estado de Río de Janeiro en los Horarios Gratuitos de Publicidad Electoral (HGPE) en el periodo de 1998 a 2018. El objetivo es presentar y describir el proceso de recopilación de esta base de datos, los procedimientos utilizados a lo largo del tiempo para sistematizar la información y los posibles usos de dos bases de datos vinculadas a la investigación: la primera tiene como unidad de análisis los programas electorales registrados durante el periodo; y la segunda tiene como unidad de análisis las candidatas/los candidatos, contabilizando el tiempo (en segundos) que aparecieron en el HGPE; el rendimiento electoral de cada candidato/candidata (votos y resultado final); la financiación de la campaña y otras características políticas, sociales y demográficas. Abstract In this technical note, we present one of the main empirical legacies of the research on the electoral campaign of candidates for Federal Deputy in the state of Rio de Janeiro during the free time granted by the government for electoral propaganda (Horário Gratuito de Propaganda Eleitoral − HGPE) from 1998 to 2018. The aim is to introduce and describe the process of data collection for this database, the procedures used over time for the systematization of information, and the potential uses of two databases linked to the research: the first analyzes the electoral programs recorded during the period and the second the unit of analysis is the individual candidates, coding the time (in seconds) of appearance in the HGPE, the electoral performance of the candidates (votes and final result), campaign financing, and other political, social, and demographic characteristics. Resumo Nesta nota técnica apresentamos um dos principais legados empíricos da pesquisa sobre propaganda das candidatas/dos candidatos a Deputado Federal do Estado do Rio de Janeiro no Horário Gratuito de Propaganda Eleitoral (HGPE) no período de 1998 a 2018. Trata-se de introduzir e descrever o processo de coleta dessa base de dados, os procedimentos utilizados ao longo do tempo para a sistematização das informações e os possíveis usos de dois bancos de dados vinculados à pesquisa: o primeiro possui como unidade de análise os programas eleitorais gravados no período; e, o segundo, possui como unidade de análise os/as candidatos/candidatas, contabilizando o tempo (em segundos) de aparição no HGPE; desempenho eleitoral de cada candidata/candidato (votos e resultado final); financiamento de campanha e outras características políticas, sociais e demográficas. Résumé Dans cette note technique, nous présentons l’un des principaux legs empiriques de la recherche sur la publicité télévisée des candidats à la députation fédérale de l’État de Rio de Janeiro dans le cadre de la période de 1998 à 2018. Resumen La literatura sobre las reformas electorales, en general, y las reformas de la financiación de las campañas, en particular, se ha beneficiado poco de las aportaciones del campo del análisis de las políticas públicas. Ya sea en términos teóricos y/o metodológicos. Este artículo propone un modelo para analizar las reformas de la financiación de las campañas electorales basado en el concepto de ciclo de políticas (policy cycle), considerando cómo se forma la agenda sobre reformas (1); la toma de decisiones (2); la aplicación (3); y la evaluación (4). Abstract The literature regarding electoral reforms, in general, and campaign finance reforms, in particular, has benefited little from the contributions of the public policy field, whether in theoretical or methodological terms. ABSTRACT The aim of the study was to assess the effectiveness of a pelvic floor muscle (PFM) rehabilitation programme in overcoming faecal incontinence (FI). Methodology: a case study of a 64-year-old, self-employed teacher diagnosed with FI because of rectal prolapse surgery. A Pelvic Floor Muscle Training (PFMT) programme was carried out, one session/week for 16 weeks, supervised by anorectal biofeedback. Each session took 45 minutes, of which 20 minutes was for PFMT and the rest for physical examination, manual therapy, massage and patient teaching/recording: symptom diary, supervision of diet management, bowel training, defecation techniques/postures and emotional support. In the first session (T0) and the last (T1), an anamnesis, Wexner Index, Bristol Stool Form Scale (BSFS) and Fecal Incontinence Quality of Life (FIQL) were also taken. The results showed gains in PFM strength, which went from grade 2 to 4 on the modified Oxford scale; in quality of life (QoL), absence of faecal losses, stools went from consistency type 2 to 4 on the BSFS. Conclusion: the uropelvic rehabilitation programme proved to be effective in re-educating the PFM and significantly improved the participant's QoL.
RESUMO O objetivo do estudo foi avaliar a eficácia de um programa de reabilitação dos Músculos do Pavimento Pélvico (MPP) para superar a Incontinência Fecal (IF). Metodologia: estudo de caso com uma participante de 64 anos, professora, autónoma, com diagnóstico de IF devido a sequela de cirurgia a prolapso retal. Realizou-se um programa de Treino dos Músculos do Pavimento Pélvico (TMPP), de uma sessão/semana, durante 16 semanas, supervisionado por Biofeedback anorretal. Cada sessão demorou 45 minutos, dos quais 20 minutos para TMPP e o restante para o exame físico, terapia manual, massagem e ensinos/registos da doente: diário de sintomas, supervisão da gestão da alimentação, treino intestinal, técnicas/posturas de defecação e suporte emocional. Na primeira sessão (T0) e na última (T1), foi ainda efetuado a anamnese, Índice de Wexner, Bristol Stool Form Scale (BSFS) e Fecal Incontinence Quality of Life (FIQL). Os resultados evidenciaram ganhos na força dos MPP, que evoluíram de grau 2 para 4 na escala de Oxford modificada; na qualidade de vida (QdV), ausência de perdas fecais, as fezes passaram de consistência tipo 2 para 4 na BSFS. Conclusão: o programa de reabilitação uropélvica, mostrou-se eficaz na reeducação dos MPP e melhorou significativamente a QdV da participante.

Lipases have been used in industrial processes as biocatalysts for transesterification reactions. The synergism between enzymes and magnetic properties may be reached by using magnetic nanoparticles (MNPs) as support to immobilize them in aggregate structures, denominated by magnetic crosslinked enzyme aggregates (MCLEA). One of the advantages of such supports is the possibility of using magnetic separation for enzyme recovery, reducing costs and allowing reuse in continuous systems. Here, porcine pancreatic lipase (PPL) was immobilized onto functionalized magnetite support (Fe3O4-APTS) with a protein binding efficiency of 78.84%. Physical and chemical properties of the nanoparticles and immobilized lipase were characterized by X-ray diffraction (XDR), transmission electron microscopy (TEM), infrared spectroscopy (FTIR), dynamic light scattering (DLS), zeta potential, vibrating sample magnetometer measurements (VSM), and 57Fe Mössbauer spectroscopy. The immobilized lipase additionally exhibited improved stability across wide pH and temperature ranges compared with free lipase. The immobilized derivate also attained good reusability, maintaining 61.37% of its initial activity after 6 reaction cycles. Abstract We investigate the occurrence of enterobacteria and antimicrobial resistance in passerines of the genus Sporophila seized from illegal trafficking. A total of 35 birds, apparently healthy and from illegal breeding, were sent to the Laboratory of Ornithological Studies at the State University of Ceará (Fortaleza, Ceará, Brazil) by environmental agencies. Cloacal swabs were collected from each bird and subjected to traditional microbiological processing, using standard cultivation and bacterialidentification techniques. We conducted antimicrobial susceptibility testing using the disc-diffusion method. Twenty-three birds (65.7%) exhibited enterobacteria, with Escherichia coli (28.6%) and Serratia liquefaciens (25.7%) being the most common. Antimicrobial resistance in total isolates was observed most frequently in relation to ciprofloxacin (28.1%), followed by tetracycline (25.0%) and enrofloxacin (18.8%). The rate of resistance to Serratia liquefaciens was the highest (i.e., 66.6% of isolates). The birds’ cloacal samples revealed several members of the Enterobacterales order, with E. coli and S. liquefaciens being the most prevalent. We also observed antimicrobial resistance to several drugs, as well as cases of multi-resistance. Antimicrobial resistance is clearly present in wild birds resulting from illegal keeping. Resumo O objetivo desta pesquisa foi investigar a ocorrência de enterobactérias em passeriformes do gênero Sporophila, provenientes do tráfico ilegal de animais, e analisar sua resistência antimicrobiana. Um total de 35 aves, aparentemente saudáveis e provenientes de criações ilegais, foram encaminhadas ao Laboratório de Estudos Ornitológicos da Universidade Estadual do Ceará (Fortaleza, Ceará, Brasil) por órgãos ambientais. Amostras obtidas a partir de suabes cloacais foram coletadas de cada ave e submetidas a processamento microbiológico tradicional, utilizando técnicas padrão de cultivo e identificação bacteriana. O teste de sensibilidade aos antimicrobianos foi avaliado pelo método de disco de difusão. Das amostras examinadas, 23 (65,7%) apresentaram a presença de enterobactérias, sendo Escherichia coli (28,6%) e Serratia liquefaciens (25,7%) as mais frequentes. A resistência antimicrobiana referente aos isolados totais foi observada com mais frequência em relação à ciprofloxacina (28,1%), seguida por tetraciclina (25,0%) e enrofloxacina (18,8%). Especificamente em relação à Serratia liquefaciens, a taxa de resistência foi a mais altas, atingindo 66,6% dos isolados. Com base nos resultados obtidos, podemos concluir que as amostras cloacais das aves apresentaram isolamento de diversos membros da ordem Enterobacteriales, sendo E. coli e S. liquefaciens as mais frequentes. Também foi observada a ocorrência de resistência antimicrobiana a diversos fármacos utilizados, assim como casos de multiresistência, o que mostra que esse problema está presente em pássaros silvestres oriundo de manutenções ilegais. Isso ressalta a necessidade de considerar a presença de bactérias resistentes a antimicrobianos em programas de soltura, visando evitar a dispersão desses microrganismos no meio ambiente. ABSTRACT Wheel slipping and fuel consumption are variables influenced by tire pressure, speed, ballasting, and soil surface. Once the tractor is present in all agricultural operations, knowledge of these variables is essential to achieving the most operational efficiency. This study aimed to develop an embedded system to collect data and show tractor slipping and fuel consumption in real-time via smartphone. The system was developed using prototyping platforms and validated through randomized field tests under four speeds and two terrain conditions. Three methods for determining slippage under different conditions were compared. The developed embedded system determined an agricultural tractor's slipping and fuel consumption conditions. The slip and fuel consumption rates varied depending on the type of rolling surface (ground preparations) and the tractor's operating speeds. Conventional tillage surface increased fuel demand by 6% and slippage by 17% compared to no-till rates. The proposed embedded system found similar results and could present the slipping values in the developed Android to real-time operation compared to a conventional method. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract We aimed to find new therapeutic targets related to Cancer Stem Cell alterations in recurrent patients from two TCGA cohorts: Testicular Germ Cell Tumor (TGCT) and Uterine Corpus Endometrial Carcinoma (UCEC). Raw sequencing data were downloaded from the TCGA database. Datasets containing RNA expression and Methylation files were directly downloaded from cBioportal. Variant Call Format files (VCFs) were downloaded from the GDC portal. Gene enrichment analysis was performed using GSEA (Gene Set Enrichment Analysis) software. Transcriptome profiling, coexpression co-occurrence, networks, and survival analyses were performed using cBioportal tools, while mutational analysis of patients was processed using UNIX scripts. We found that cancer stem cell transcription factors were highly expressed in Testicular Germ Cell Tumor (TGCT) and Uterine Corpus Endometrial Carcinoma (UCEC) cohorts, compared to the other 29 cancer cohorts in TCGA. Patients presented a poorer diagnosis when the genes (POU5F1, NANOG, SOX2, SALL4, ABCB1, ABCC1, and ABCG2) were altered. In UCEC cohorts, recurrent patients showed the ABCG2 potentially phosphorylated by the PIM1 kinase. In the TGCT cohort, genes ABCB1 and ABCG2 only appeared in the phosphonetwork in recurrent patients potentially phosphorylated by the same kinase, PIM1, but also by PRKACA. Our data indicate that PRKACA and PIM1 may modulate POU5F1 phosphorylation. Abstract We estimated genetic parameters of two Pinus caribaea var. caribaea progeny tests based on growth traits aiming at the identification of the best selection strategy. This study was conducted in two progeny tests located in Selvíria (MS), Brazil. Tests were set up in block design, one with 76 progenies (area 1) and the other with 99 progenies (area 2). Genetic parameters were estimated using the SELEGEN software through REML/BLUP method. To estimate the genetic gain through selection, four selection methods and ten selection intensities were tested. The intensity of selection that maximizes genetic gain and genetic diversity was calculated by the optimization point. Significant variation was observed between and with progeny for all traits in area 2 and only within plots for height in area 1. Higher values of heritability were observed in area 1. ABSTRACT Breath passion fruit (Passiflora nitida) has been used as rootstock for sour passion fruit (P. edulis) in order to control wilting and collar rot caused by Fusarium spp. However, it is supposedly susceptible to root-knot nematode (Meloidogyne incognita), which is widespread in tropical crop fields. Two trials were carried out to assess the effect of M. incognita on the growth of a P. nitida accession, and a third one was performed to assess the effect on the growth of P. edulis grafted onto P. nitida and the anatomical changes induced by this nematode in P. nitida. In view of the high multiplication rate of this nematode in P. nitida, causing intense root galling and reducing plant growth of both P. nitida and P. edulis, P. nitida should not be used as rootstock, or even main crop, in M. incognita-infested fields. RESUMO O maracujá-suspiro (Passiflora nitida) tem sido utilizado como porta-enxerto para o maracujá-azedo (P. edulis), com o objetivo de controlar a murcha e a podridão do colo causadas por Fusarium spp. No entanto, acredita-se que ele seja suscetível ao nematoide-das-galhas (Meloidogyne incognita), que está amplamente distribuído em campos de cultivo tropical. Foram realizados dois ensaios para avaliar o efeito de M. incognita no crescimento de um acesso de P. nitida, e um terceiro ensaio foi conduzido para avaliar o efeito no crescimento de P. edulis enxertado em P. nitida e as alterações anatômicas induzidas por esse nematoide em P. nitida. Periodontal dental ligament mesenchymal stem cells (PDLMSCs) play a major role in periodontal tissue regeneration by the neoformation of root cementum and alveolar bone. These cells are highly heterogeneous, and many present low potential to renovate the hard tissue damaged by periodontal disease. A previous study found that the low osteoblast/cementoblast (O/C) differentiation potential of PDLMSCs is related to high asporin (ASPN) expression, which was identified as a negative regulator of PDL cells differentiation and mineralization, suppressing BMP-2-induced O/C differentiation. Objective This study aimed to investigate whether 1,25(OH)2D3 treatment could stimulate the O/C differentiation of periodontal ligament mesenchymal progenitor cells characterized as low osteoblast potential (LOP), by asporin and bone morphogenetic protein-2 alteration. Methodology Three LOP cell populations were cultured in standard medium (CONTROL), osteogenic medium (OM), and osteogenic medium associated with 1 nM of 1,25(OH)2D3 (OM + VD). The following assays were performed: 1) MTT to evaluate metabolic activity; 2) gene expression for asporin (ASPN), bone morphogenetic protein-2 (BMP-2), runt-related transcription factor 2 (RUNX2), alkaline phosphatase (ALP), osteocalcin (OCN), and vitamin D receptor (VDR) using qRT-PCR; 3) BMP-2 extracellular expression; and 4) quantification of mineralized nodule deposition by Alizarin Red Staining. Data were subjected to two-way ANOVA and Tukey’s test (P<0.05). Results The results showed that the 1,25(OH)2D3 treatment did not affect the cell viability, as demonstrated by metabolic activity increase over the 10 days in culture. After 14 days of 1,25(OH)2D3 treatment, the mRNA levels for ASPN and VDR decreased (P<0.05), while BMP-2 transcripts and extracellular expression increased (P<0.05). In parallel, RUNX2, ALP, and OCN gene expression was upregulated by 1,25(OH)2D3 treatment, resulting in an increase of mineral nodule deposition in vitro (P<0.05). Conclusions These data show that 1,25(OH)2D3 improves osteoblast/cementoblast differentiation of low osteoblast potential accompanied by alterations in ASPN and BMP-2 expression. ABSTRACT The largest proportion of greenhouse gas (GHG) emissions in the Agriculture sector of the Brazilian national GHG inventory is derived from the large (>200 million head) herd of cattle. The greatest contribution to these emissions comes from the enteric methane from cattle, but the direct and indirect emissions of nitrous oxide (N2O) from cattle excreta and N fertilizer are responsible for approximately 9 % of all national anthropogenic GHG emissions. Ammonia (NH3) can be volatilized from N fertilizer and cattle excreta and deposited in sites remote from the source, constituting an indirect source of N2O. This study aimed to determine whether direct N2O emissions and NH3 volatilization from N-fertilized pastures were greater than those derived from a mixed grass-legume pasture without N fertilizer addition. Emissions of N2O and NH3 from excreta and N fertilizer from a Palisade grass (Urochloa brizantha cv. Marandu) monoculture fertilized with 2 × 60 kg N ha-1 yr-1 urea were compared to those from a mixed Palisade grass-forage peanut (Arachis pintoi) pasture. Dung and urine were collected from these cattle, and NH3 losses and N2O emissions from the excreta and from N fertilizer were monitored using static chamber techniques. Volatilization of NH3 and N2O emissions were found to be greater from urine than from dung. Ammonia losses from excreta and urea fertilizer were low, not exceeding 6.8, 1.1, and 4.7 % of the N applied as urine, dung, and fertilizer, respectively. The N2O emissions showed a tendency to be greater for the urine from the N-fertilized compared to the mixed grass-legume pasture, and the N2O emissions from the urine of the N-fertilized pasture ranged from 0.08 to 0.94 % of applied urine N. The N2O emission from the N fertilizer was at maximum 0.46 % of the applied N. The direct N2O emissions and the loss of NH3 by volatilization (indirect N2O emission) from the excreta of cattle grazing the mixed grass-legume pasture were similar to, or lower than, the grazed grass monoculture fertilized with 120 kg N ha-1 yr-1. As the mixed pasture received no N fertilizer and hence no GHG emission from its manufacture or application, introducing forage peanut to the Urochloa brizantha pastures shows potential to be responsible for lower GHG emissions than the N fertilized grass pasture. ABSTRACT Background: In the past decades, endoscopic ultrasound has developed from a diagnostic tool to a platform for many therapeutic interventions. Various technological advancements have emerged since the last Brazilian Consensus, demanding a review and update of the recommendations based on the best scientific evidence. Methods: A group of 32 renowned echoendoscopists selected eight relevant topics to be discussed to generate clinical questions. After that, a literature review was conducted to answer these questions based on the most updated evidence. Results: Thirty-three statements were formulated and voted on by the experts to reach a consensus. The Oxford System was used to grade the level of evidence. Conclusion: There is moderate evidence to support that the needle shape, gauge, or aspiration technique does not influence the yield of endoscopic ultrasound (EUS)-guided tissue sampling of pancreatic solid lesions. There is moderate evidence to support using EUS-TTNB of the cyst wall to differentiate between mucinous and non-mucinous cystic neoplasms. There is little evidence to support the EUS-guided treatment of gastric varices. There is a high level of evidence to support that EUS-guided biliary drainage and ERCP present similar outcomes in patients with distal malignant biliary obstruction. There is a high level of evidence for using EUS to diagnose neoplastic pancreatic cysts and detect necrosis before indicating drainage. There is moderate evidence to support EUS-GE over duodenal stent for malignant gastric outlet obstruction in patients with a life expectancy higher than 2 months. RESUMO Contexto: Nas últimas décadas, a ecoendoscopia evoluiu de uma ferramenta diagnóstica para uma plataforma para diversas intervenções terapêuticas. Vários avanços tecnológicos surgiram desde o último Consenso Brasileiro, demandando uma revisão e atualização das recomendações baseadas nas melhores evidências científicas. Métodos: Um grupo de 32 ecoendoscopistas renomados selecionou oito tópicos relevantes para serem discutidos a fim de gerar questões clínicas. Em seguida, foi realizada uma revisão da literatura para responder a essas perguntas com base nas evidências mais atualizadas. Resultados: Trinta e três tópicos foram formulados e votados pelos especialistas para alcançar um consenso. O Sistema de Oxford foi utilizado para classificar o nível de evidência. Conclusão: Há evidências moderadas para sustentar que a forma da agulha, calibre ou técnica de aspiração não influenciam no rendimento da amostragem tecidual guiada por ultrassom endoscópico (USE) de lesões sólidas pancreáticas.
