Abstract This study compares the effects of virus-cell interactions among SARS-CoV-2 variants of concern (VOCs) isolated in Brazil in 2021, hypothesizing a correlation between cellular alterations and mortality and between viral load and transmissibility. For this purpose, reference isolates of Alpha, Gamma, Zeta, and Delta variants were inoculated into monolayers of Vero-E6 cells. Viral RNA was quantified in cell supernatants by RT‒PCR, and infected cells were analyzed by Transmission Electron Microscopy (TEM) for qualitative and quantitative evaluation of cellular changes 24, 48, and 72 hours postinfection (hpi). Ultrastructural analyses showed that all variants of SARS-CoV-2 altered the structure and function of mitochondria, nucleus, and rough endoplasmic reticulum of cells. Monolayers infected with the Delta variant showed the highest number of modified cells and the greatest statistically significant differences compared to those of other variants. Viral particles were observed in the cytosol and the cell membrane in 100 % of the cells at 48 hpi. Alpha showed the highest mean particle diameter (79 nm), and Gamma and Delta were the smallest (75 nm). Alpha and Gamma had the highest particle frequency per field at 48 hpi, while the same was observed for Zeta and Delta at 72 hpi and 24 hpi, respectively. The cycle threshold of viral RNA varied among the target protein, VOC, and time of infection. Abstract Background Patients with immune-mediated rheumatic diseases (IMRDs) have been prioritized for COVID-19 vaccination to mitigate the infection severity risks. Patients with rheumatoid arthritis (RA) are at a high risk of severe COVID-19 outcomes, especially those under immunosuppression or with associated comorbidities. However, few studies have assessed the safety of the COVID-19 vaccine in patients with RA. Objective To evaluate the safety of vaccines against SARS-CoV-2 in patients with RA. Methods This data are from the study “Safety and Efficacy on COVID-19 Vaccine in Rheumatic Diseases,” a Brazilian multicentric prospective phase IV study to evaluate COVID-19 vaccine in IMRDs in Brazil. Adverse events (AEs) in patients with RA of all centers were assessed after two doses of ChAdOx1 (Oxford/AstraZeneca) or CoronaVac (Sinovac/Butantan). Stratification of postvaccination AEs was performed using a diary, filled out daily and returned at the end of 28 days for each dose. Results A total of 188 patients with RA were include, 90% female. CoronaVac was used in 109 patients and ChAdOx1 in 79. Only mild AEs were observed, mainly after the first dose. The most common AEs after the first dose were pain at the injection (46,7%), headache (39,4%), arthralgia (39,4%), myalgia (30,5%) and fatigue (26,6%), and ChAdOx1 had a higher frequency of pain at the injection (66% vs 32 %, p < 0.001) arthralgia (62% vs 22%, p < 0.001) and myalgia (45% vs 20%, p < 0.001) compared to CoronaVac. The more common AEs after the second dose were pain at the injection (37%), arthralgia (31%), myalgia (23%), headache (21%) and fatigue (18%). Arthralgia (41,4% vs 25%, p = 0.02) and pain at injection (51,4% vs 27%, p = 0.001) were more common with ChAdOx1. No serious AEs were related. With Regard to RA activity level, no significant difference was observed between the three time periods for both COVID-19 vaccines. Conclusion In the comparison between the two immunizers in patients with RA, local reactions and musculoskeletal symptoms were more frequent with ChAdOx1 than with CoronaVac, especially after the first dose. In summary, the AE occurred mainly after the first dose, and were mild, like previous data from others immunizing agents in patients with rheumatoid arthritis. RESUMO Objetivo: Descrever o IMPACTO-MR, um estudo brasileiro de plataforma nacional em unidades de terapia intensiva focado no impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Métodos: Descrevemos a plataforma IMPACTO-MR, seu desenvolvimento, critérios para seleção das unidades de terapia intensiva, caracterização da coleta de dados, objetivos e projetos de pesquisa futuros a serem realizados na plataforma. Resultados: Os dados principais foram coletados por meio do Epimed Monitor System® e consistiram em dados demográficos, dados de comorbidades, estado funcional, escores clínicos, diagnóstico de internação e diagnósticos secundários, dados laboratoriais, clínicos e microbiológicos e suporte de órgãos durante a internação na unidade de terapia intensiva, entre outros. De outubro de 2019 a dezembro de 2020, 33.983 pacientes de 51 unidades de terapia intensiva foram incluídos no banco de dados principal. Conclusão: A plataforma IMPACTO-MR é um banco de dados clínico brasileiro de unidades de terapia intensiva focado na pesquisa do impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Essa plataforma fornece dados para o desenvolvimento e pesquisa de unidades de terapia intensiva individuais e ensaios clínicos observacionais e prospectivos multicêntricos. ABSTRACT Objective: To describe the IMPACTO-MR, a Brazilian nationwide intensive care unit platform study focused on the impact of health care-associated infections due to multidrug-resistant bacteria. Methods: We described the IMPACTO-MR platform, its development, criteria for intensive care unit selection, characterization of core data collection, objectives, and future research projects to be held within the platform. Results: The core data were collected using the Epimed Monitor System® and consisted of demographic data, comorbidity data, functional status, clinical scores, admission diagnosis and secondary diagnoses, laboratory, clinical, and microbiological data, and organ support during intensive care unit stay, among others. From October 2019 to December 2020, 33,983 patients from 51 intensive care units were included in the core database. Conclusion: The IMPACTO-MR platform is a nationwide Brazilian intensive care unit clinical database focused on researching the impact of health care-associated infections due to multidrug-resistant bacteria. This platform provides data for individual intensive care unit development and research and multicenter observational and prospective trials. Resumo Fundamento A hipertrofia e a dilatação do ventrículo direito observadas na hipertensão arterial pulmonar (HAP) prejudicam a dinâmica do ventrículo esquerdo (VE) achatando o septo interventricular. Objetivo Investigar se o treinamento físico resistido (TFR) de intensidade baixa a moderada é benéfico para funções contráteis do VE e de cardiomiócitos em ratos durante o desenvolvimento de HAP induzida por monocrotalina (MCT). Métodos Foram usados ratos Wistar machos (Peso corporal: ~ 200 g). Para avaliar o tempo até o possível surgimento de insuficiência cardíaca (ou seja, ponto de desfecho), os ratos foram divididos em dois grupos, hipertensão com sedentarismo até a insuficiência (HSI, n=6) e hipertensão com treinamento até a insuficiência (HTI, n=6). Para testar os efeitos do TFR, os ratos foram divididos entre grupos de controle sedentários (CS, n=7), hipertensão com sedentarismo (HS, n=7) e hipertensão com treinamento (HT, n=7). A HAP foi induzida por duas injeções de MCT (20 mg/kg, com um intervalo de 7 dias). Os grupos com treinamento foram submetidos a um protocolo de TFR (subir escadas; 55-65% da máxima carga carregada), 5 dias por semana. A significância estatística foi definida em p <0,05. Resultados O TFR prolongou o ponto de desfecho (~25%), melhorou a tolerância ao esforço físico (~55%) e atenuou as disfunções de contratilidade de VE e de cardiomiócitos promovidas pela MCT preservando a fração de ejeção e o encurtamento fracional, a amplitude do encurtamento, e as velocidades de contração e relaxamento nos cardiomiócitos. O TFR também preveniu os aumentos de fibrose e colágeno tipo I no ventrículo esquerdo causados pela MCT, além de manter as dimensões de miócitos e colágeno tipo III reduzidas por MCT. Conclusão O TFR de intensidade baixa a moderada é benéfico para funções contráteis de VE e cardiomiócitos em ratos durante o desenvolvimento de HAP induzida por MCT.
Abstract Background The right ventricular hypertrophy and dilation observed in pulmonary artery hypertension (PAH) damages the left ventricle (LV) dynamics by flattening the interventricular septum. Objective To investigate whether low- to moderate-intensity resistance exercise training (RT) is beneficial to LV and cardiomyocyte contractile functions in rats during the development of monocrotaline (MCT)-induced PAH. Methods Male Wistar rats (Body weight: ~ 200 g) were used. To assess the time to potential heart failure onset (i.e., end point), rats were divided into sedentary hypertension until failure (SHF, n=6) and exercise hypertension until failure (EHF, n=6) groups. To test RT effects, rats were divided into sedentary control (SC, n = 7), sedentary hypertension (SH, n=7), and exercise hypertension (EH, n=7) groups. PAH was induced by two MCT injections (20 mg/kg, with 7 days interval). Exercise groups were submitted to an RT protocol (Ladder climbing; 55-65% of carrying maximal load), 5 times/week. Statistical significance was assumed at P < 0.05. Results RT prolonged the end point (~25 %), enhanced the physical effort tolerance (~ 55%), and mitigated the LV and cardiomyocyte contractility dysfunctions promoted by MCT by preserving the ejection fraction and fractional shortening, the amplitude of shortening, and the velocities of contraction and relaxation in cardiomyocytes. RT also prevented increases in left ventricle fibrosis and type I collagen caused by MCT, and maintained the type III collagen and myocyte dimensions reduced by MCT. Conclusion Low- to moderate-intensity RT benefits LV and cardiomyocyte contractile functions in rats during the development of MCT-induced PAH.

RESUMO Objetivos: Há diversas terapias sendo utilizadas ou propostas para a COVID-19, muitas carecendo de apropriada avaliação de efetividade e segurança. O propósito deste documento é elaborar recomendações para subsidiar decisões sobre o tratamento farmacológico de pacientes hospitalizados com COVID-19 no Brasil. Métodos: Um grupo de 27 membros, formado por especialistas, representantes do Ministério da Saúde e metodologistas, integra essa diretriz. Foi utilizado o método de elaboração de diretrizes rápidas, tomando por base a adoção e/ou a adaptação de recomendações a partir de diretrizes internacionais existentes (GRADE ADOLOPMENT), apoiadas pela plataforma e-COVID-19 RecMap. A qualidade das evidências e a elaboração das recomendações seguiram o método GRADE. Resultados: Foram geradas 16 recomendações. Entre elas, estão recomendações fortes para o uso de corticosteroides em pacientes em uso de oxigênio suplementar, para o uso de anticoagulantes em doses de profilaxia para tromboembolismo e para não uso de antibacterianos nos pacientes sem suspeita de infecção bacteriana. Não foi possível fazer uma recomendação quanto à utilização do tocilizumabe em pacientes hospitalizados com COVID-19 em uso de oxigênio, pelas incertezas na disponibilidade e de acesso ao medicamento. Foi feita recomendação para não usar azitromicina, casirivimabe + imdevimabe, cloroquina, colchicina, hidroxicloroquina, ivermectina, lopinavir/ ritonavir, plasma convalescente e rendesivir. Conclusão: Até o momento, poucas terapias se provaram efetivas no tratamento do paciente hospitalizado com COVID-19, sendo recomendados apenas corticosteroides e profilaxia para tromboembolismo. Diversos medicamentos foram considerados ineficazes, devendo ser descartados, de forma a oferecer o melhor tratamento pelos princípios da medicina baseada em evidências e promover economia de recursos não eficazes.
ABSTRACT Objective: Several therapies are being used or proposed for COVID-19, and many lack appropriate evaluations of their effectiveness and safety. The purpose of this document is to develop recommendations to support decisions regarding the pharmacological treatment of patients hospitalized with COVID-19 in Brazil. Methods: A group of 27 experts, including representatives of the Ministry of Health and methodologists, created this guideline. The method used for the rapid development of guidelines was based on the adoption and/or adaptation of existing international guidelines (GRADE ADOLOPMENT) and supported by the e-COVID-19 RecMap platform. The quality of the evidence and the preparation of the recommendations followed the GRADE method. Results: Sixteen recommendations were generated. They include strong recommendations for the use of corticosteroids in patients using supplemental oxygen, the use of anticoagulants at prophylactic doses to prevent thromboembolism and the nonuse of antibiotics in patients without suspected bacterial infection. It was not possible to make a recommendation regarding the use of tocilizumab in patients hospitalized with COVID-19 using oxygen due to uncertainties regarding the availability of and access to the drug. Strong recommendations against the use of hydroxychloroquine, convalescent plasma, colchicine, lopinavir + ritonavir and antibiotics in patients without suspected bacterial infection and also conditional recommendations against the use of casirivimab + imdevimab, ivermectin and rendesivir were made. Conclusion: To date, few therapies have proven effective in the treatment of hospitalized patients with COVID-19, and only corticosteroids and prophylaxis for thromboembolism are recommended. Several drugs were considered ineffective and should not be used to provide the best treatment according to the principles of evidence-based medicine and promote economical resource use.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Resumo Anaplasma marginale é um patógeno transmitido por vetores que causam uma doença conhecida como anaplasmose. Até a presente data, não há genomas sequenciados de cepas brasileiras. O objetivo deste estudo foi comparar o genoma completo das cepas brasileiras de A. marginale (Palmeira e Jaboticabal) com os genomas de cepas de outras regiões (cepas dos EUA e Austrália). As sequências dos genomas das cepas brasileiras foram obtidas mediante sequenciamento de nova geração. As “reads” foram mapeadas usando-se como referência o genoma de A. marginale da cepa Florida. Foram identificados polimorfismos de nucleotídeo único (SNPs) e analisadas inserções/deleções (INDELs). As duas linhagens brasileiras se agruparam em um clado particular que, por sua vez, agrupou-se em um grupo maior junto com as linhagens norte-americanas. Além disso, foram identificadas diferenças significativas nas proteínas de superfície entre os dois isolados brasileiros. Esses resultados lançam luz sobre a história evolutiva de A. marginale e fornecem as primeiras informações de genomas de isolados sul-americanos. Avaliar as sequências de genomas de cepas de diferentes regiões é essencial para aumentar o conhecimento do pan-genoma dessa bactéria.
Abstract Anaplasma marginale is a vector-borne pathogen that causes a disease known as anaplasmosis. No sequenced genomes of Brazilian strains are yet available. The aim of this work was to compare whole genomes of Brazilian strains of A. marginale (Palmeira and Jaboticabal) with genomes of strains from other regions (USA and Australia strains). Genome sequencing of Brazilian strains was performed by means of next-generation sequencing. Reads were mapped using the genome of the Florida strain of A. marginale as a reference sequence. Single nucleotide polymorphisms (SNPs) and insertions/deletions (INDELs) were identified. The data showed that two Brazilian strains grouped together in one particular clade, which grouped in a larger American group together with North American strains. Moreover, some important differences in surface proteins between the two Brazilian isolates can be discerned. These results shed light on the evolutionary history of A. marginale and provide the first genome information on South American isolates. Assessing the genome sequences of strains from different regions is essential for increasing knowledge of the pan-genome of this bacteria.

Resumo: O objetivo deste trabalho foi avaliar a variabilidade genética de Araucaria cunninghamii, em teste de procedência, e estimar ganhos genéticos para a produção de sua madeira, com base em diferentes intensidades de seleção. O experimento foi estabelecido em Luís Antônio, no estado de São Paulo, Brasil, com sementes de polinização aberta coletadas em Queensland, Austrália. O delineamento experimental utilizado foi o de blocos ao acaso, com seis repetições e quatro plantas por parcela. O diâmetro à altura do peito (DBH), a forma do fuste e a sobrevivência foram medidos 29 anos após o plantio. Diferenças significativas foram detectadas entre as procedências. O coeficiente de variação genotípica foi alto (60%) para DBH. Os valores médios de DBH e forma do fuste foram de 23,21 cm e 4,50, respectivamente. As procedências Langkelly Creek, Brooweena L.A., Yarraman e St. Agnes apresentaram melhor desempenho em DBH, e Imbil Seed Orchard, Brooweena L.A., St. Agnes, St. John e Yarraman, em forma do fuste. Os ganhos estimados com a seleção variaram de 0,5 a 24,8%, para DBH, e de 0,9 a 3,5%, para forma do fuste.
Abstract: The objective of this work was to assess the genetic variability of Araucaria cunninghamii in a provenance test and to estimate the genetic gains in its wood production, based on different selection intensities. The trial was established in Luís Antônio, in the state of São Paulo, Brazil, using open-pollinated seed from Queensland, Australia. The experimental design used was a randomized complete block with six replicates, and four plants per plot. Diameter at breast height (DBH), stem form, and survival were measured at 29 years after planting. Significant differences were detected among provenances. The coefficient of genotypic variation between provenances was high (60%) for DBH and stem form. The mean values for DBH and stem form were 23.21 cm and 4.50, respectively. The provenances Langkelly Creek, Brooweena L.A., Yarraman, and St. Agnes showed the best performance for DBH, and Imbil Seed Orchard, Brooweena L.A, St. Agnes, St. John, and Yarraman, for stem form. Genetic gains ranged from 0.5 to 24.8% for DBH, and from 0.9 to 3.5% for stem form.

Abstract The striking and complex phenotype of Cockayne syndrome (CS) patients combines progeria-like features with developmental deficits. Since the establishment of the in vitro culture of skin fibroblasts derived from patients with CS in the 1970s, significant progress has been made in the understanding of the genetic alterations associated with the disease and their impact on molecular, cellular, and organismal functions. In this review, we provide a historic perspective on the research into CS by revisiting seminal papers in this field. We highlighted the great contributions of several researchers in the last decades, ranging from the cloning and characterization of CS genes to the molecular dissection of their roles in DNA repair, transcription, redox processes and metabolism control. We also provide a detailed description of all pathological mutations in genes ERCC6 and ERCC8 reported to date and their impact on CS-related proteins. Finally, we review the contributions (and limitations) of many genetic animal models to the study of CS and how cutting-edge technologies, such as cell reprogramming and state-of-the-art genome editing, are helping us to address unanswered questions.

RESUMO Foram avaliados os parâmetros genéticos e fenotípicos da bifurcação (BIF), forma do fuste (RET) e diâmetro à altura do peito (DAP) para as procedências e progênies de Dipteryx alata. O experimento foi instalado em delineamento em blocos ao acaso, com 43 tratamentos (26 e 17 progênies dos municípios de Três Lagoas e Brasília no Estado de Mato Grosso do Sul e Distrito Federal, respectivamente) e cinco plantas por parcela. O espaçamento entre plantas foi de 3 x 3 metros. A análise dos dados foi realizada pelo método REML / BLUP (máxima verossimilhança restrita melhor predição linear não viesada). Foram detectadas diferenças significativas entre progênies apenas para DAP. Os valores genéticos individuais variaram de 3,7 a 21,7% para BIF e DAP; A variação genética entre as progênies variou de 9 a 10,5% para BIF e DAP entre as procedências. A herdabilidade individual no sentido restrito foi alta para o DAP (mínimo de 0,31), revelando o potencial de melhoramento genético. As procedências e progênies demonstraram ter variabilidade genética, que pode ser explorada em programas de melhoramento florestal, com ampla base genética.

The individual genetic values ranged from 3.7 to 21.7 % for BIF and DBH; genetic variation among progenies ranged from 9 to 10.5 % for BIF and DBH among provenances. Individual heritability in the strict sense was high for DBH (minimum from 0.31), revealing the potential for genetic improvement. Provenance and progenies trial showed genetic variability which can be exploited in forest improvement programs with a broad genetic base.

RESUMO O presente estudo foi conduzido com o objetivo de analisar estrutura de covariância e estimativas de repetibilidade das variáveis relacionadas à produtividade da goiabeira como massa dos frutos (MF), número de frutos (NF) e produção de frutos (PROD) de três safras, em 95 genótipos de uma população segregante. E por meio dos critérios de AIC (Akaike’s Information Criterion) e SBC (Schwarz’s Bayesian Criterion) escolher a estrutura de covariância mais adequada das observações dentro de um mesmo indivíduo. Foi possível incorporar ao modelo estatístico, uma estrutura de covariância entre medidas repetidas, sendo as formas autorregressiva e simetria composta as mais adequadas. Os valores dos coeficientes de repetibilidade obtidos para PF (0,25), NF (0,14) e PROD (0,29) foram considerados baixos, indicando que para a população em estudo as três safras realizadas não foram suficientes para selecionar os melhores indivíduos com maior acurácia. Para as variáveis PF e PROD, a partir de cinco medições seria possível a obtenção de estimativas de acurácias em torno de 0,50. Enquanto que para a variável NF, observou-se que mais safras seriam necessárias. Estes valores indicam que em populações segregantes de goiabeira, avaliações nas primeiras safras, não são suficientes para seleção de genótipos mais estáveis para as variáveis consideradas neste estudo.
ABSTRACT The present study was conducted with the objective of analyzing the covariance structure and repeatability estimates of the variables related to guava productivity, such as fruit weight (FW), fruit number (FN) and fruit production (FP) of three harvests, in 95 genotypes of a segregating population. The study also aims to choose the most appropriate covariance structure of the observations within the same individual by means of AIC (Akaike's Information Criterion) and SBC (Schwarz's Bayesian Criterion) criteria. A covariance structure between repeated measures could be incorporated into the statistical model, with the self-regression and compound symmetry forms being the most adequate. The values of repeatability coefficients obtained for FW (0.25), FN (0.14), and FP (0.29) were considered low, indicating that the three harvests were not sufficient to select the best individuals with greater accuracy for the study population. For the variables PF and FP, estimates of accuracy around 0.50 could be obtained from five measurements, while for the variable FN more harvests would be necessary. These values indicate that in guava-segregating populations, evaluations in the first harvests are not enough to select more stable genotypes for the variables considered in this study.

Microcirculation improvement after short-term infusion of vasopressin in septic shock is dependent on noradrenaline

OBJECTIVES: To assess the impact of vasopressin on the microcirculation and to develop a predictive model to estimate the probability of microcirculatory recruitment in patients with septic shock. METHODS: This prospective interventional study included patients with septic shock receiving noradrenaline for less than 48 hours. We infused vasopressin at 0.04 U/min for one hour. Hemodynamic measurements, including sidestream dark-field imaging, were obtained immediately before vasopressin infusion, 1 hour after vasopressin infusion and 1 hour after vasopressin withdrawal. We defined patients with more than a 10% increase in total vascular density and perfused vascular density as responders. ClinicalTrials.gov: NCT02053675. RESULTS: Eighteen patients were included, and nine (50%) showed improved microcirculation after infusion of vasopressin. The noradrenaline dose was significantly reduced after vasopressin (p=0.001) and was higher both at baseline and during vasopressin infusion in the responders than in the non-responders. The strongest predictor for a favorable microcirculatory response was the dose of noradrenaline at baseline (OR=4.5; 95% CI: 1.2-17.0; p=0.027). For patients using a noradrenaline dose higher than 0.38 mcg/kg/min, the probability that microcirculatory perfusion would be improved with vasopressin was 53% (sensitivity 78%, specificity 77%). CONCLUSIONS: In patients with septic shock for no longer than 48 h, administration of vasopressin is likely to result in an improvement in microcirculation when the baseline noradrenaline dose is higher than 0.38 mcg/kg/min.

RESUMO Visando à obtenção de novas variedades de goiaba com atributos superiores, este trabalho foi conduzido e analisado pelo procedimento REML/BLUP, em nível de indivíduos.Foram avaliadas 17 famílias segregantes de goiabeira, seguindo o delineamento experimental de blocos casualizados, com 2 repetições, com 12 plantas por parcela. As famílias foram obtidas após polinizações biparentais controladas. Os indivíduos estudados apresentaram elevada variância genotípica para peso de fruto (PF), produtividade (PRD) e teor de ácido ascórbico (TAA). As magnitudes dos coeficientes de herdabilidade da média das progênies conduziram a um alto valor de acurácia para seleção de progênies para rendimento de polpa (RP), teor de sólidos solúveis (TSS), além de PF, PRD e TAA; moderado para acidez do fruto (AF) e para a relação TSS/AF, e baixo em espessura do mesocarpo (EM) e pH. Averiguou-se que a seleção entre famílias (h2mp) apresentou os maiores valores para PF, RP, PRD, TSS e TAA, mostrando-se que, para o presente estudo, a seleção entre família seria efetiva, já que estas características foram as que permitiram maiores valores de acurácia seletiva entre famílias. Quanto ao ordenamento dos indivíduos, observa-se um destaque para as famílias provenientes do cruzamento entre UENF 1835 x UENF 1834; UENF 1831 x UENF 1832; UENF 1831 x UENF 3739,que ocupam as primeiras posições no rankeamento para a maioria das características.
ABSTRACT Aiming at the generation of new guava varieties with superior attributes, we conducted this study adopting the REML/BLUP procedure at individual level. Seventeen segregating guava families were evaluated in a randomized-block design with two replicates and 12 plants per plot. Families were obtained after controlled biparental pollination. The studied individuals showed high genotypic variance for fruit weight (FW), total yield (YLD), and ascorbic acid content (AAC). The heritability coefficients of the mean of progenies led to high progeny-selection accuracy for pulp yield (PY), soluble solids content (SSC), in addition to FW, YLD, and AAC; moderate accuracy for fruit acidity (FA) and SSC/FA ratio; and low accuracy for mesocarp thickness (MT) and pH. Selection among families (h2mp) indicated the highest values for FW, PY, YLD, SSC, and AAC, revealing that, for the present study, this practice would be effective, since these traits allowed for the highest selection accuracy values among families. As for the ranking of individuals, families originating from crosses UENF 1835 × UENF 1834, UENF 1831 × UENF 1832, and UENF 1831 × UENF 3739 stood out, occupying the first positions for most traits.

Rotary-vacuum-filter mud (RVFM) is waste generated during the manufacturing process of titanium dioxide. In this work, RVFM and ceramic bricks containing different ratios of this waste are investigated. The mud samples were characterized using thermal analysis (TG/DTG). The aim of the present work was to determine the effect of adding RVFM on the ceramic properties of clay, such as apparent porosity, water absorption, linear shrinkage and flexural strength, used to produce red ceramics (bricks and roofing tiles). Samples were dried out at 110°C and fired at 800°C, 950°C and 1100°C. The addition of RVFM tends to increase the apparent porosity and water absorption and to decrease the flexural strength of the ceramic specimens. Based on the results, ceramic specimens with 20% RVFM content that are burned at 800ºC can not be used as bricks, and ceramic specimens with 20% RVFM content that are fired at 800ºC and 950°C can not used as roofing tiles, according to Brazilian standards.

Electric-arc furnace dust (EAFD) is a by-product of steel production and recycling. This fine-grained material contains high amounts of zinc and iron as well as significant amounts of potentially toxic elements such as lead, cadmium and chromium. Therefore, the treatment and stabilization of this industrial residue is necessary. Concrete is a well-known suitable environment for stabilization/solidification of materials which have leachable elements in need of fixation. The effect of the EAFD content on the mechanical and chemical performance of Portland cement concrete is investigated in this paper. The effect of the EAFD content on the setting time of cement slurry was also analyzed. The axial compressive strength of the concrete samples increases with the EAFD addition in the range of 10 to 20 wt. (%) EAFD; also the tensile strength increases with the EAFD addition. An increase in EAFD content significantly increases the setting time of the concrete. The acetic acid leaching and water solubilization tests indicate low mobility of the potentially toxic elements from the EAFD concrete composite. The results of the immersion tests show that the addition of EAFD to the concrete seems to reduce chloride penetration, which may help prevent pitting corrosion in reinforced concrete.
