Twelve microsatellite loci previously developed in the tropical tree Cariniana estrellensis were genetically characterized in Cariniana legalis. Polymorphisms were assessed in 28 C. legalis individuals found between the Pardo and Mogi-Guaçu River basins in the state of São Paulo, Brazil. Of the 12 loci, 10 were polymorphic and exhibited Mendelian inheritance. The allelic richness at each locus ranged from 2-11, with an average of 7 alleles per locus, and the expected heterozygosity ranged from 0.07-0.88. These loci showed a high probability of paternity exclusion. The characteristics of these heterologous microsatellite markers indicate that they are suitable tools for investigating questions concerning population genetics in C. legalis.

Este trabalho teve como objetivo otimizar um protocolo econômico, rápido e eficaz de minipreparação de DNA genômico, para as espécies florestais Copaifera langsdorffii (Óleo de Copaíba), Hymenaea courbaril (Jatobá), Eugenia uniflora (Pitanga), Tabebuia roseo alba (Ipê Branco) e Cariniana estrellensis (Jequitibá Branco). Este método é uma adaptação da técnica de extração CTAB de Doyle e Doyle (1990), o qual consiste principalmente na adição de PVP para eliminar polifenoles, somente uma etapa de extração com clorofórmio-álcool isoamílico e a adição da RNase A imediatamente após a extração com clorofórmio. O método também dispensa o uso de nitrogênio líquido, o uso do fenol e a adição de proteinase K. Os DNAs das espécies florestais extraídos apresentaram alto rendimento e boa qualidade, com rendimento de 25.7 a 42.1 µg de DNA a partir de 100 mg de tecido foliar congelado. Com este protocolo, em apenas 1 dia de trabalho, uma pessoa pode completar o isolamento do DNA de aproximadamente 50 amostras de folhas (dependendo da capacidade da centrífuga). O DNA obtido pode ser usado para métodos de análise baseados em PCR (SSR e RAPD).

An efficient and rapid DNA minipreparation modified method for frozen samples was developed for five tropical tree species: Copaifera langsdorffii, Hymenaea courbaril, Eugenia uniflora, Tabebuia roseo alba and Cariniana estrellensis. This procedure that dispenses the use of liquid nitrogen, phenol and the addition of proteinase K, is an adaptation of the CTAB-based DNA extraction method. The modifications included the use of PVP to eliminate the polyphenols, only one chloroform-isoamyl alcohol step and the addition of RNase immediately after extraction with chloroform. The yields of the DNA samples ranged from 25.7 to 42.1 µg from 100 mg leaf tissue. The DNA samples extracted by this method were successfully used for PCR (SSR and RAPD) analyses in these five and other twelve tropical tree species.

Microsatellite markers were transferred and characterized for two Neotropical fig tree species, Ficus citrifolia and Ficus eximia. Our study demonstrated that microsatellite markers developed from different subgenera of Ficus can be transferred to related species. In the present case, 12 of the 15 primer pairs tested (80%) were successfully transferred to both of the above species. Eleven loci were polymorphic when tested across 60 F. citrifolia and 60 F. eximia individuals. For F. citrifolia, there were 4 to 15 alleles per locus, whereas expected heterozygosities ranged from 0.31 to 0.91. In the case of F. eximia, this was 2 to 12 alleles per locus and expected heterozygosities from 0.42 to 0.87.

Um total de 119 índios Waiãpi e 348 habitantes de Manaus, Amazonas, foram testados usando nove sistemas genéticos enzimáticos na primeira população e dois na segunda. Os Waiãpi mostraram algumas diferenças marcantes quanto às médias dos indígenas sul-americanos em ACP*A (1% vs. 14 ± 9%), GLO1*1 (9% vs. 30 ± 14%) e ESD*1 (99,6% vs. 70 ± 17%), enquanto as prevalências de ESD*1 e CA2*1 na população de Manaus mostraram-se dentro do intervalo encontrado previamente na região norte do Brasil (ESD*1: 85% vs. 80-94%; CA2*1: 98% vs. 89-99.8%). As freqüências quase idênticas obtidas para esses dois marcadores nesta população, quando a amostra foi subdividida de acordo com a cor da pele, sugerem que tais classificações morfológicas têm pouco valor nesta região. Distâncias genéticas entre os Waiãpi e outras tribos, obtidas usando esses e outros marcadores genéticos, mostraram bom paralelismo com as distâncias geográficas que separam esses índios dos outros grupos considerados.

One hundred nineteen Waiãpi Indians and 348 inhabitants of Manaus, Amazonas, were tested using nine enzyme genetic systems in the first population and two in the second. The Waiãpi showed some marked differences compared to South American Indian averages in ACP*A (1% vs. 14 ± 9%), GLO1*1 (9% vs. 30 ± 14%) and ESD*1 (99.6% vs. 70 ± 17%), while ESD*1 and CA2*1 prevalences in the Manaus population were within the range previously found in the northern region of Brazil (ESD*1: 85% vs. 80-94%; CA2*1: 98% vs. 89-99.8%). The almost identical frequencies obtained for these two markers in this population, when the sample was subdivided according to skin color, suggest that such morphological classifications have little value in this region. Genetic distances between the Waiãpi and other tribes, obtained using these and other genetic markers, showed good parallelism with the geographical distances which separate these Indians from the other groups considered.

Este estudo foi conduzido no sentido de otimizar as técnicas de preparo de extratos de folíolos e identificar tampões e condições de corrida que proporcionassem uma boa definição dos perfis eletroforéticos de clones de seringueira. Foram testados 25 diferentes sistemas enzimáticos utilizando-se onze sistemas tamponantes. Não foram encontradas diferenças na qualidade dos zimogramas obtidos nos diferentes estádios fenológicos analisados, em amostras submetidas a diferentes condições de centrifugação e nas diferentes plantas de um mesmo clone. Extratos de folíolos liofilizados apresentaram resolução semelhante para Adh, Pgi, 6Pgdh, Lap-1, Skdh, Acp, Mdh, ßGlu, Pgm e Idh que homogenados de folíolos não liofilizados preparados até 2 dias pós-coleta. Os padrões de Got e Est apresentaram boa nitidez somente em extratos de folíolos frescos. As demais enzimas não apresentaram resolução satisfatória. Com o uso de diferentes substratos, foi possível detectar nove regiões de atividade esterásica nas condições empregadas, embora variação genética tenha sido caracterizada para apenas três locos nos clones analisados. Os fenótipos observados para esterase-7 utilizando-se ésteres derivados de naftol foram semelhantes aos padrões encontrados utilizando L-benzoil-ßnaftilamida como substrato, indicando que esta esterase seja, provavelmente, uma en-dopeptidase. A utilização de substratos fluorogênicos permitiu ainda a detecção de uma variante eletroforética de Acp em Hevea nítida e uma variante de ßGlu em Hevea rigidifolia.

This study aimed at optimizing techniques for preparing clonal leaflet extracts and identifying buffers and migration conditions so as to achieve better resolution of electrophoretic patterns in rubber tree clones. Twenty-five enzymatic systems in eleven gel and electrode buffers were tested. Similar electrophoretic profiles were obtained from extracts of clones in different phenological stages, in samples submitted to different centrifugation conditions and in plants from the same clone. Extracts from liophylized leaflets showed the same electrophoretic resolution as nonliophylized leaflets after two days for Adh, Pgi, 6Pgdh, Lap-1, Skd, Acp, Mdh-1, ß-Glu, Pgm and Idh. Got and Est electrophoretic patterns were well resolved only in fresh leaflets extracts. The other enzymes studied did not show a good electrophoretic resolution. Employing different types of substrates, nine regions of esterase activity were observed under the conditions used, although genetic variation has been characterized for only three loci in the clones analysed. Est-7 phenotypes observed using naphtyl-esters were similar to patterns found using L-benzoyl-β-naphtylamide assubstrate, suggesting that this esterase is an endopeptidase. The use of fluorogenic substrates allows the detection of a new electrophoretic Acp variant in Hevea nitida and another βGlu variant in Hevea rigidifolia leaflet extracts.
