RESUMO: O presente estudo objetivou avaliar a eficácia de um protocolo one-step de congelamento do sêmen suíno utilizando dimetilacetamida (DMA) e etilenoglicol como crioprotetores. Durante 10 semanas, a fração rica dos ejaculados de dois machos suínos foram coletados, uma vez por semana. Após a coleta, os ejaculados foram diluídos (1:1; v/v) no diluidor de resfriamento. Após a avaliação da concentração espermática, os ejaculados foram agrupados em um pool com o mesmo número de espermatozoides de cada macho e estabilizados a 20 °C por 120 min. Os criopropetores foram adicionados ao diluidor de congelamento a 20 °C, em diferentes concentrações, compondo seis tratamentos: glicerol (controle), 1,25% e 2,5%; etilenoglicol, 1,5% e 2,5%; e DMA, 2,5% e 5,0%. As amostras foram armazendadas em palhetas de 0,25 mL contendo 35 x 106 espermatozoides. Após 90 min a 20 °C as palhetas foram submetidas a uma curva de resfriamento até 5 °C (0,3 a 0,5 °C/mim) e mantidas a 5 °C por 60 min. O congelamento foi realizado a partir da colocação das palhetas horizontalmente a 5 cm acima do nitrogênio líquido por 10 min, com sua posterior imersão no nitrogênio líquido. Após o descongelamento a 37 °C por 30 segundos a qualidade espermática foi avaliada através de um sistema computadorizado e por citometria de fluxo. A motilidade espermática foi maior (P < 0,05) nos tratamentos com 5,0% e 2,5% DMA (22,2 ± 2,6% e 20,0 ± 2,8%, respectivamente) do que no tratamento com 2,5% etilenoglicol (8,2 ± 1,0%). A integridade da membrana plasmática (P = 0,08) e potencial de membrana mitocondrial (P = 0,27) foi similar entre os tratamentos. O tratamento com 2,5% de etilenoglicol foi menos eficiente em manter membrana acrossomal intacta (P< 0,01). Alguns parâmetros de cinética espermática (DAP, DCL, DSL, VAP, VCL, VSL e ALH) foram afetados positivamente pelo uso de DMA a 5.0%. ABSTRACT: The present study evaluated the cryoprotectant efficacy of dimethylacetamide (DMA) and ethylene glycol in a one-step protocol to freeze boar sperm. The sperm-rich portion of the ejaculates from two boars were collected once a week, for 10 weeks. After collection, the ejaculates were diluted (1:1; v/v) in the cooling extender. After determining their spermatozoa concentration, the ejaculates were pooled with the same number of spermatozoa from each boar and stabilized at 20°C for 120 min. Distinct cryoprotectants were added to the cooling extender at 20 °C, at different concentrations, composing six treatments: 1.25% and 2.5% glycerol (control); 1.25% and 2.5% ethylene glycol; 2.5% and 5.0% DMA. The samples were stored in 0.25 mL straws, containing 35 × 106 spermatozoa. After 90 min at 20 °C, the straws were submitted to a cooling curve until 5 °C (0.3 to 0.5 °C/min) and kept at 5°C for 60 min. Freezing was conducted by placing the straws horizontally 5 cm above the liquid nitrogen for 10 min, followed by immersion on liquid nitrogen. After thawing at 37 °C for 30 seconds, sperm quality was evaluated through a computer-assisted semen analysis system and flow cytometry. Sperm motility was greater (P< 0.05) in treatments with 5.0% and 2.5% DMA (22.2 ± 2.6% and 20.0 ± 2.8%, respectively) than in treatment with 2.5% ethylene glycol (8.2 ± 1.0%). The integrity of the plasma membrane (P = 0.08) and mitochondrial membrane potential (P = 0.27) was similar among the treatments. The treatment with 2.5% ethylene glycol was the least efficient to maintain intact acrosome membrane (P< 0.01). Some kinetics parameters (DAP, DCL, DSL, VAP, VCL, VSL e ALH) were positively affected by 5.0% DMA. ABSTRACT Image phenotyping presents great potential for field experiment evaluations in tomato crop. Validating this method for tomato plants is important to reduce costs and time. Thus, the objective of the present study is to investigate the agronomic performance of tomato hybrids and inbred lines using vegetation index. A randomized block design with three replications was used. A total of36 experimental hybrids and 9 inbred lines were evaluated, in addition to the recurrent genitor UFU-57 and a commercial hybrid, culminating in 47 treatments. During the experiment, average weight, transverse and longitudinal diameter, pulp thickness, number of lobules, soluble solids, β-carotene and lycopene concentrations were evaluated. RESUMO O uso da fenotipagem por imagens apresenta alto potencial para avaliação de experimentos na cultura do tomateiro. Validar a aplicação desse método é importante par reduzir custos e tempo. Assim, a pesquisa teve como objetivo investigar o desempenho agronômico de híbridos e linhagens de tomateiro a partir de índices de vegetação. Foi utilizado o delineamento em blocos casualizados com três repetições. Foram avaliados 36 híbridos experimentais e 9 linhagens, além do genitor recorrente UFU-57 e um híbrido comercial, totalizando 47 tratamentos. Foram avaliados o peso médio, diâmetro transversal e longitudinal, espessura da polpa, número de lóculos, teores de sólidos solúveis, β-caroteno e licopeno. Além disso, analisou-se o desenvolvimento vegetativo da planta por meio de índices de vegetação obtidos com o suporte de uma aeronave e remotamente pilotada. Abstract The ocean is a unique system connecting ecological, economic, social, and cultural components, through which goods and services regulate the planetary condition and support the development of mankind. However, its increasing use has followed the exponential growth of the global economic system, outpacing humanity’s ability to develop the knowledge necessary to establish a basis for its proper use. Hence, there is an added perception that our necessary knowledge about the functioning of the ocean for its appropriate planning and management, advances at a slow pace, with which the ocean would be losing quality and sustainability. Systemic views of the ocean tend to highlight dominant components and processes instead of structural details, establishing a quality shortcut to the knowledge where society can understand current and future ocean conditions. To achieve the desired ocean health and sustainability, we propose the formation of a base of knowledge of the marine and coastal environments, capable of supporting best practices and policies for planning and management. We drew from the interdisciplinary research developed by the Brazilian research group “Ecosystem-Based Marine and Coastal Management (Eco-MCM), ” which has been developing projects based on three fundamental steps: (1) systemic analysis of the marine and coastal environments, highlighting their ecosystems, ecosystem services, social and economic benefits produced by the services and the stakeholders benefited; (2) modeling of the studied systems, and (3) propositional phase to incorporate models to support the practices and policies for their planning, management, and governance. As such, they are aligned with the United Nations Decade of Ocean Science for Sustainable Development (2021-2030) challenges and outcomes.

ABSTRACT Objective To describe the sociodemographic and clinical characterization of patients with chronic hepatitis C followed at the outpatient clinic of a reference hospital in infectology. Method A cross-sectional, descriptive, quantitative study with chronic hepatitis C patients attended at a referral hospital during November 2015 to April 2016 with a sample of 47 users. Results The participants were male (76.6%), 57 years old (57.5%), brown (38.3%), married (55.3%), (61.7%), with a discovery time of up to 6 years (68.1%), not knowing the form of contamination (57.5%), immunized against hepatitis B (65.9%), undergoing drug therapy (85.1%) with Ribavirin (55.6%); And 70.2% had adverse effects. Conclusion Sociodemographic and clinical characterization assist the clinical practice of the multiprofessional team with patients with chronic hepatitis C.

Abstract INTRODUCTION: Chronic hepatitis C is a leading cause of liver disease. Infection triggers an immediate immune response in the host that is mediated by humoral/cellular mechanisms. T cells respond to infection via secretion of cytokines, which inhibit or stimulate one another, leading to cytokine imbalance and ultimately affecting treatment. Studies using interferon (IFN) and ribavirin (RBV) showed that TCD8+ cells and cytokine levels are associated with sustainable virological response (SVR). However, studies that investigated the effects of triple therapy (TT) are limited. METHODS: The study included hepatitis C virus (HCV)+ RNA, naives, genotype 1, ≥18 years, and advanced fibrosis (F≥3) patients. Samples were collected at baseline and after 12 weeks (W12) of TT. Six cytokines were analyzed by flow cytometry. RESULTS: Of 31 patients, four were excluded (two deaths, one interrupted TT, and one F2 patient). Of the 27 remaining patients, 21 (78%) were cirrhotic. SVR was achieved in 63% of the patients. The patients had a mean age of 55.11 ± 10.03 years. Analyses at baseline showed that the chemokine CCL5/Regulated on Activation, Normal T Cell Expressed and Secreted (RANTES) (p=0.04) and interleukin (IL)-6 (p=0.02), which was associated with SVR. RANTES (p=0.04) and IL-8 (p=0.01) levels were associated with SVR at W12. CONCLUSIONS Similar to patterns observed during double therapy, IL-6, IL-8, and RANTES levels were associated with SVR in TT, indicating the potential role of interferon in immune response to hepatitis C virus.

Abstract Faced with the need for greater knowledge of the different physalis species, the aim of this study was to characterize different Native American physalis species (Physalis peruviana L., Physalis pubescens L., Physalis angulata L., Physalis mínimos L. and Physalis ixocarpa Brot) as to their physicochemical characteristics, bioactive compounds and antioxidant activity. Besides that, in order to increase their use and add even more value to this fruit, we also evaluate the influence of these different species on the physicochemical, rheological and sensory characteristics of physalis jelly. In addition, this study evaluated the sensory acceptance of the combination of physalis jellies obtained from different species with brie-type cheese. The Peruviana, Pubences and Angulata, are highlighted for being the nutritionally richest species, with the highest levels of phenolic compounds, vitamin C and antioxidant. Moreover, they stand out for originating the most widely sensory accepted jellies, either in pure form or in combination with brie-type cheese.

ABSTRACT Objective: This study aims to estimate the prevalence of insulin resistance (IR) among chronic hepatitis C (CHC) patients and their related laboratory and demographic data. Subjects and methods: In this study, non-diabetic CHC patients referred to Viral Hepatitis Ambulatories from Rio de Janeiro (Brazil) donated blood samples. Insulin was measured using a chemiluminescence immunoassay. IR was determined by HOMA-IR, where HOMA-IR > 2 was defined as IR. Results: A total of 214 CHC patients were recruited (123 females aged 53.6 years ± 10.9 years). IR was present in 133 patients (62.1%) and was associated in bivariate analysis to higher mean values of age (p = 0.040), triglycerides (p = 0.032), glucose (p = 0.000), insulin (p = 0.000), waist circumference (p = 0.001), and body mass index (p = 0.007); however, none of these variables were significant in the multivariate analysis. Conclusions: The high prevalence of IR was observed among CHC patients, and there was no difference in clinical or laboratory parameters when both groups were compared in the multivariate analysis. This high IR prevalence could lead to a high risk for development of cardiovascular disease and metabolic disorders.
