ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The exploration of bioproducts from these industrial rejects can reduce environmental impact besides of producing high value-added materials. In this scenario, carboxymethyl starch nanoparticles were produced from mango (Mangifera indica L.) kernel starch. These nanoparticles were then decorated with thermoresponsive chains of the amino terminated poly(N-isopropylacrylamide) (PNIPAM-NH2), with the intention of evaluating their applicability in the biomedical area. Elemental analysis, Fourier transform infrared (FTIR) and 1H nuclear magnetic resonance (NMR) spectroscopy confirmed successful grafting of PNIPAM-NH2 onto the carboxymethyl starch backbone. Scanning electron microscopy (SEM) images and dynamic light scattering (DLS) data showed sizes of 100 and 112 nm in the dry state and of 744 and 598 nm in the hydrated state, when the grafting degree (GD) was of 6 and 14.3%, respectively. The degree of swelling was of 41,100 and 15,100% for GD of 6 and 14.3% respectively, suggesting that the nanogels are suitable for drug incorporation. The toxicity of the nanogels to human adipose-derived stem cells (ADSCs) and red blood cells (RBCs) was evaluated by lactate dehydrogenase (LDH), alamarBlue and hemolysis assays. Both nanogels were non-cytotoxic and non-hemolytic, suggesting the suitability of these biomaterials for cell-and blood-contacting applications.

ABSTRACT Objectives: Candida spp. has been reported as one of the common agents of nosocomial bloodstream infections and is associated with a high mortality. Therefore, this study evaluated the clinical findings, local epidemiology, and microbiological aspects of candidemia in eight tertiary medical centers in the state of Parana, South of Brazil. Methods: In this study, we reported 100 episodes of candidemia in patients admitted to eight different hospitals in five cities of the state of Parana, Brazil, using data collected locally (2016 and 2017) and tabulated online. Results: The incidence was found to be 2.7 / 1000 patients / day and 1.2 / 1000 admissions. C. albicans was responsible for 49% of all candidemia episodes. Cancer and surgery were the two most common underlying conditions associated with candidemia. The mortality rate within 30 days was 48%, and removal of the central venous catheter (p = 0.029) as well as empirical or prophylactic exposure to antifungals were both related to improved survival (p = 0.033). Conclusions: This study highlights the high burden and mortality rates of candidemia in hospitals from Parana as well as the need to enhance antifungal stewardship program in the enrolled medical centers.

O Consórcio inclui três coortes de nascimento de Ribeirão Preto, Estado de São Paulo (1978/1979, 1994 e 2010), quatro de Pelotas, Estado do Rio Grande do Sul (1982, 1993, 2004 e 2015) e duas de São Luís, Estado do Maranhão (1997 e 2010). As coortes provêm de três regiões do Brasil, de três estados diferentes, com importantes diferenças socioeconômicas, culturais e de infraestrutura. As coortes foram iniciadas ao nascer dos participantes, exceto a mais recente em cada município, onde as mães foram recrutadas durante a gestação. Os instrumentos para a coleta de dados foram refinados para aproximar diferentes exposições na primeira infância e a influência, a longo prazo, no processo saúde-doença. Os investigadores das nove coortes realizaram estudos perinatais e depois examinaram o capital humano, saúde mental, nutrição e sinais percursores de doenças crônicas. Um total de 17.636 nascidos vivos foram recrutados em Ribeirão Preto, 19.669 em Pelotas e 7.659 em São Luís. Nas coortes que foram iniciadas durante a gestação, foram entrevistadas 1.400 gestantes em Ribeirão Preto, 3.199 em Pelotas e 1.447 em São Luís. Foram utilizadas diferentes estratégias para reduzir as perdas de seguimento. A rede de pesquisa do Consórcio permite analisar a incidência de doenças e identificar possíveis relações causais que podem explicar os desfechos de saúde nessas populações e contribuir para o desenvolvimento de medidas públicas e políticas de saúde que estejam mais de acordo com as respectivas realidades.

Different strategies were employed to reduce losses to follow-up. This research network allows the analysis of the incidence of diseases and the establishment of possible causal relations that might explain the health outcomes of these populations in order to contribute to the development of governmental actions and health policies more consistent with reality.

A phytochemical investigation from aerial parts of Casearia arborea (Rich.) Urb. (Salicaceae) led to isolation and identification of: sitosterol, 4-en-stigmast-3-one, 13-hydroxy-trans-ent-cleroda-3,14-diene,3-hydroxy-2-oxo-trans-ent-cleroda-3,14-diene (kolavelone), a mixture of 13-hydroxy-trans-ent-cleroda-3,14-diene and an ester ethyl hexadecanoate, kaempferol-3-O-a-L-arabinofuranoside and 4',5,7-trihydroxy-3',5'-dimethoxyflavone (tricin). The compounds have been described for the first time in this species. Five trihydroxy-flavone-hexoside derivatives have been identified by LC-ESI-HR-MS. The antimicrobial activity of kolavelone was evaluated against strains of Staphylococcus, Bacillus, Pseudomonas, Shigella and Candida spores, and its minimum inhibitory concentration (MIC) was determined. The results showed the antimicrobial activity of kolavelone against several bacteria and Candida tropicalis indicating its potential use as antimicrobial agent.

Abstract Schistosomiasis, a chronic disease that affects million people worldwide, is caused by trematode flukes of the genus Schistosoma. The lack of an anti-schistosomiasis vaccine and massive monotherapy with praziquantel reinforces the need for search and development of new therapeutic drugs. Recently, we demonstrated that the essential oil of Piper cubeba L., Piperaceae, and their derivative dibenzylbutyrolactolic (-)-6,6'-dinitrohinokinin, presents in vitro and in vivo activities against Schistosoma mansoni. Here, we identified changes in the protein expression after exposure to dibenzylbutyrolactolic (-)-6,6'-dinitrohinokinin. We applied two-dimensional gel electrophoresis (2-DE) to S. mansoni soluble protein extracts and observed at least 38 spots to be affected by dibenzylbutyrolactolic (-)-6,6'-dinitrohinokinin. We further identified 25 differentially expressed proteins by mass spectrometry. Enrichment for biological processes and predictive analyses of protein-protein interactions suggest that dibenzylbutyrolactolic (-)-6,6'-dinitrohinokinin targets proteins involved mainly in metabolic processes, especially carbohydrate metabolism. In summary, this study provides an interesting approach to understand the anti-parasitic activity of semi-synthetic (-)-6,6'-dinitrohinokinin a derivative compound from lignan and for the development of new therapy strategies.

Association among measures of mobility-related disability and self-perceived fatigue among older people: a population-based study

Objective: To investigate the relationship between self-perceived fatigue with different physical functioning tests and functional performance scales used for evaluating mobility-related disability among community-dwelling older persons. Method: This is a cross-sectional, population-based study. The sample was composed of older persons with 65 years of age or more living in Cuiabá, MT, and Barueri, SP, Brazil. The data for this study is from the FIBRA Network Study. The presence of self-perceived fatigue was assessed using self-reports based on the Center for Epidemiologic Studies-Depression Scale. The Lawton instrumental activities of daily living scale (IADL) and the advanced activities of daily living scale (AADL) were used to assess performance and participation restriction. The following physical functioning tests were used: five-step test (FST), the Short Physical Performance Battery (SPPB), and usual gait speed (UGS). Three models of logistic regression analysis were conducted, and a significance level of α<0.05 was adopted. Results: The sample was composed of 776 older adults with a mean age (SD) of 71.9 (5.9) years, of whom the majority were women (74%). The prevalence of self-perceived fatigue within the participants was 20%. After adjusting for covariates, SPPB, UGS, IADL, and AADL remained associated with self-perceived fatigue in the final multivariate regression model. Conclusion: Our results suggest that there is an association between self-perceived fatigue and lower extremity function, usual gait speed and activity limitation and participation restriction in older adults. Further cohort studies are needed to investigate which physical performance measure may be able to predict the negative impact of fatigue in older adults.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Synthesis and evaluation of octocrylene-inspired compounds for UV-filter activity

Octocrylene (2-ethylhexyl 2-cyano-3,3-diphenyl-2-propenoate) is present in several sunscreens and is known to work synergistically with UV filters. We prepared eight octocrylene-related compounds to test their photoprotective activities by measuring diffuse transmittance. The compounds had varied photoprotection profiles, with Sun Protection Factors (SPF) ranging from 1 to 5 and UVA Protection Factors (UVAPF) ranging from 1 to 8. Compounds 4, 5, and 7 showed the best protection against UVB sunrays, while compounds 5, 6, and 7 presented the best results for protection from UVA, so compound 7 had the most balanced protection overall. Results for compounds 4, 8, and 9 are reported for the first time in the literature.

No semiárido nordestino o aumento da área desflorestada e não recuperada da Caatinga degradada fragiliza seus solos e seu equilíbrio hidrológico. Este trabalho teve como objetivo mapear a alteração da cobertura vegetal na Sub-Bacia do Rio Espinharas no período 2000-2010. Os dados das imagens do sensor Thematic Mapper do satélite Landsat-5 foram processados no ambiente Spring. Para avaliação da cobertura vegetal utilizou-se o índice de vegetação por diferença normalizada. 65% da área apresentam alto grau de antropismo, vegetação rala e solos expostos. A imagem diferença foi analisada considerando-se os valores da média de distribuição dos níveis de cinza na imagem (µ) e do desvio padrão (σ), segundo os limiares [0, μ - σ], [μ - σ, μ + σ] e [μ + σ, 254] relacionando-os às classes: desmatamento/redução, não mudança e áreas de regeneração/incremento da vegetação. Os resultados mostram que 80% da área se mantêm inalterados; em 12,6% da área ocorreu redução da cobertura vegetal pelas práticas de corte raso para obtenção de lenha para uso doméstico ou comercial e preparo da área para a agricultura e em 7,4% da área registrou-se incremento possivelmente favorecido pelo cultivo de pastagens irrigadas e não irrigadas em solos de aluvião.
In the semiarid northeast, the increase in the area of deforestation and non-reclaimed 'Caatinga' sites already degraded weaken its soils and hydrological balance. This study aimed to map the alteration in the vegetation cover along the Espinharas River sub-basin area during the period 2000-2010. The image data collected from the thematic mapper sensor of the satellite Landsat-5 were processed with the aid of computational systems Spring. In order to evaluate the vegetation cover, the normalized difference vegetation index was used. Sixty five percent of the area presented high level of anthropogenic disturbance, sparse vegetation and exposed soils. The difference in image, whose values were analysed taking into account the values of the average distribution of gray levels in the image (µ) standard deviation (σ) according to the thresholds [0, μ - σ], [μ - σ, μ + σ] e [μ + σ, 254], related to the following classifications: deforestation/reduction of vegetation cover, no change and areas of regeneration/vegetation growth. The results showed that 80% of the area remains unchanged, reduction of vegetation cover occurred in 12.6% of the area due to the practice of clear cutting of vegetation in order to obtain firewood for domestic or commercial use and also area preparation for agriculture purposes; some increment was observed in 7.4% of the area possibly due to cultivation of irrigated and non-irrigated pastures in alluvial soils.

O objetivo deste estudo foi avaliar a potencialidade da espectroscopia no infravermelho próximo (NIRS) como um método rápido e não destrutivo para determinação do teor de sólidos solúveis (SSC) e pH em ameixas intactas. Amostras de ameixa com teores de sólidos solúveis variando de 5,7 a 15% e pH entre 2,72 e 3,84 foram adquiridas em supermercados em Natal (Rio Grande do Norte, Brasil) e espectros NIR foram coletados no intervalo de 714-2500 nm. Uma comparação de algumas técnicas de calibração multivariada com relação ao preprocessamento dos dados e algoritmos de seleção de variáveis, tais como mínimos quadrados parciais por intervalos (iPLS), algoritmo genético (GA), algoritmo das projeções sucessivas (APS) e seleção de preditores ordenados (OPS), foi realizada. Modelos de validação para SSC e pH tiveram um coeficiente de correlação (R²) de 0,95 e 0,90, com erro quadrático médio de previsão (RMSEP) de 0,45 e 0,07, respectivamente. A partir desses resultados, conclui-se que a espectroscopia NIR pode ser usada como uma técnica não destrutiva para determinação de SSC e pH em ameixas.
The aim of this study was to evaluate the potential of near-infrared reflectance spectroscopy (NIRS) as a rapid and nondestructive method to determine the soluble solid content (SSC) and pH of intact plums. Samples of plum with total solid contents ranging from 5.7 to 15% and pH from 2.72 to 3.84 were collected from supermarkets in Natal (Rio Grande do Norte, Brazil) and NIR spectra were acquired in the 714-2500 nm range. A comparison of some multivariate calibration techniques with respect to several pre-processing data and variable selection algorithms, such as interval partial least squares (iPLS), genetic algorithm (GA), successive projection algorithm (SPA) and ordered predictor selection (OPS), was performed. Validation models for SSC and pH had a coefficient of correlation (R²) of 0.95 and 0.90, as well as a root mean square error of prediction (RMSEP) of 0.45 and 0.07, respectively. From these results, it can be concluded that NIR spectroscopy can be used as a nondestructive technique for determining SSC and pH in plums.

Microsatellite markers for population genetic studies of the blowfly Chrysomya putoria (Diptera: Calliphoridae)

The investigation of the genetic variation and population structure of Chrysomya species is of great interest for both basic and applied research. However, very limited genetic information is available for this genus across its geographical distribution. Here, we describe 12 polymorphic microsatellite loci isolated from Chrysomya putoria with expected heterozygosities ranging from 0.1402-0.8312. These markers are of potential applied interest for forensic entomologists and for the characterisation of the genetic structure of C. putoria from recently colonised regions, with great promise for understanding the colonisation dynamics and spread of the genus Chrysomya in the New World.

A infecção pela Chlamydophila abortus (C. abortus) em ruminantes domésticos está relacionada com distúrbios reprodutivos. Apesar de ainda pouco estudada em todo o mundo, a infecção já foi identificada em alguns países europeus e também nos EUA. No Brasil, estudos preliminares demonstraram evidências sorológicas da infecção em alguns rebanhos com baixa prevalência de anticorpos. Até o momento ainda não foi possível a identificação de C. abortus a partir de material biológico proveniente de fêmeas com problemas reprodutivos em rebanhos brasileiros. O objetivo deste trabalho foi avaliar a presença da C. abortus em uma coleção de abortos bovinos (n=85), ovinos (n=12) e caprinos (n=8), em amostras de muco vaginal bovino (n=13), ovino (n=90) e caprino (n=20), e em sêmen ovino (n=10) e caprino (n=5). As amostras biológicas (n=243) foram avaliadas por meio de técnica de PCR desenvolvida para a amplificação do espaço intergênico 16S-23S RNAr da C. abortus. Um controle interno da reação, que amplifica um fragmento do gene ND5 do DNA mitocondrial de bovino, foi utilizado para a avaliação da eficiência da extração e da amplificação do DNA nas amostras provenientes de abortamento bovino. Todas as amostras biológicas (n=243) incluídas nesse estudo resultaram negativas para a C. abortus na PCR. O controle interno da reação possibilitou a amplificação de um produto do gene ND5 mitocondrial bovino em todas as amostras de aborto bovino (n=85). Apesar de evidências,,sorológicas indicarem a presença da infecção por C. abortus em rebanhos de ruminantes no Brasil, e considerando o número de amostras biológicas avaliadas, a não identificação de C. abortus nesse estudo sugere que a frequência de sinais clínicos nos animais infectados pode ser baixa ou mesmo ausente.
Chlamydophila abortus (C. abortus) infection is related to reproductive failure in domestic ruminants. Although it has not been well characterized worldwide, this pathogen has already been identified in some European countries and in the USA. In Brazil, preliminary studies have shown serological evidence of C. abortus infection in herds with low antibody prevalence. Until now, the identification of C. abortus in biological samples from females presenting reproductive failures has not been described in Brazilian herds of domestic ruminants. The aim of this study was to evaluate the presence of the C. abortus in a collection of abortions from cattle (n=85), sheep (n=12), and goats (n=8), in samples of vaginal mucus from cows (n=13), sheep (n=90), and goats (n=20), and in semen from sheep (n=10) and goats (n=5). The specimens (n=243) were evaluated using a PCR assay developed to amplify the 16S-23S rRNA intergenic space of C. abortus. A PCR assay with an internal control, which amplifies a fragment from the ND5 gene of bovine mitochondrial DNA, was used in order to evaluate the efficiency of the DNA extraction and of the PCR reaction. All biological samples (n=243) included in this study were negative for C. abortus in the PCR assay. The internal control enabled the amplification of a product from the bovine mitochondrial ND5 gene in all cattle abortion samples (n=85). Given the serological evidence indicating the presence of C. abortus infection in Brazilian herds of domestic ruminants, and considering the wide sampling evaluated, the failure to identify C. abortus in this survey suggests that the frequency of clinical signs in infected animals may be low or even absent.

An increasing interest has been growing during the past years for the search of natural origin antioxidants, particularly those from marine algae. In this context, the main objective of current research was to evaluate the chemical composition and some antioxidant properties of the aqueous extract of the seaweed Bryothamnion triquetrum. The extracts contains: Proteins (9.5%), Lipids (1.3%), Carbohydrates (5.9%), Fibers (10.2%) and Ashes (43%). In current approach, the following results were obtained for the different procedures assessed: DPPH• radicals scavenging (38% for 4 mg of lyophilized); beta-Carotene-Linoleic assay (12% for 4 mg of lyophilized); O2•- radicals scavenging (IC50 0.36 mg/mL); OH• radicals scavenging (IC50 2.11 mg/mL) and iron quelation ability (IC50 0.37 mg/mL). Thus, antioxidant properties of this natural product seem to be related to its ability to scavenge free species. In previous reports of our group, cinnamic and phenolic acids were proposed as at least partially responsible for the antioxidant properties of the extract, but the necessity for the presence of other components was also shown. Then, the antioxidant properties of the extract could be envisioned as the result of the additive and/or synergic effect between phenolic constituents and the other antioxidant components, such as minerals, carotenes and ascorbic acid.
En la actualidad existe un marcado interés por la búsqueda de antioxidantes de fuentes naturales, incluidas las algas marinas. El objetivo de este trabajo fue evaluar la composición química y propiedades antioxidantes del alga Bryothamnion triquetrum. Se estudió la composición centesimal y de minerales, identificación de ácidos grasos y sustancias antioxidantes. La composición centesimal es la siguiente: Proteínas (9,5%), Lípidos (1,3%), Carbohidratos (5,9%), Fibras (10,2%) y Cenizas (43%). Los resultados de la actividad antioxidante para las diferentes metodologías empleadas fueron: atrapamiento de radicales DPPH• (38%, 4 mg de liofilizado), beta-Caroteno-Linoleico (12%, 4 mg de liofilizado), actividad atrapadora de radicales O2•- (CI50 0,36 mg/mL), de radicales OH• (CI50 2,11 mg/mL) y unión al Fe (CI50 0,37 mg/mL). Las propiedades antioxidantes de esta alga parecen explicarse por la capacidad atrapadora de radicales libres, particularmente relacionada con mecanismos de dismutación de radicales O2•-, inactivación de radicales OH• y quelación de Fe. En trabajos previos se identificaron ácidos cinámicos y fenólicos como moléculas que pudieran explicar la actividad antioxidante, sin embargo adicionalmente se debe considerar un efecto sumatorio y/o sinérgico de otros componentes antioxidantes del extracto, como los descritos en este trabajo, incluidos minerales, carotenoides y vitamina C.
