Biostimulant potential of Azospirillum brasilense and nicotinamide for hydroponic pumpkin cultivation

RESUMO A aplicação de bioestimulantes na agricultura tem sido estudada para aumentar a produção, utilizando menores espaços físicos e menos insumos agrícolas, visando aumentar a sustentabilidade nos sistemas de produção. Apesar dos resultados promissores, há necessidade de estudar os efeitos de aplicações conjuntas destes produtos, identificando possíveis interações durante o desenvolvimento da cultura. Assim, este estudo teve como objetivo verificar o potencial bioestimulante do uso isolado e combinado de nicotinamida e Azospirillum brasilense durante o crescimento inicial de plantas de abóbora em cultivo hidropônico. Os tratamentos foram compostos pela combinação da aplicação de nicotinamida (foliar) e A. brasilense (foliar ou aplicada em solução hidropônica). Foi realizado um delineamento inteiramente casualizado com seis tratamentos e quatro repetições. Os tratamentos aumentaram as trocas gasosas, principalmente a taxa de assimilação e a eficiência de carboxilação, sendo superiores ao tratamento controle. Em complemento, para as características de crescimento, a aplicação isolada de nicotinamida favoreceu o desenvolvimento da parte aérea e, quando combinada com A. brasilense via foliar, aumentou o crescimento radicular. Assim, a aplicação de nicotinamida e A. brasilense isoladamente ou em conjunto potencializa o crescimento e as trocas gasosas de plantas de abóbora cultivadas em sistema hidropônico, principalmente quando se utiliza aplicação foliar de A. brasilense. ABSTRACT The application of biostimulants in agriculture has been studied to increase production while using smaller physical spaces and agricultural inputs to increase sustainability in production systems. Despite the promising results, there is a need to study the effects of joint applications of these products, identifying possible interactions during crop development. Thus, this study aimed to verify the biostimulant potential of the isolated and combined use of nicotinamide and Azospirillum brasilense during the initial growth of pumpkin plants in hydroponic cultivation. A completely randomized design with six treatments and four replicates was performed. The treatments were composed by the combination of the application of nicotinamide (foliar) and A. brasilense (foliar or applied in the hydroponic solution). Treatments increased the gas exchanges, particularly the assimilation rate and carboxylation efficiency, being superior to the control treatment. However, for growth characteristics, the isolated application of nicotinamide favored the development of the shoot and, when combined with A. brasilense via foliar, increased root growth. Thus, the application of nicotinamide and A. brasilense alone or together enhances the growth and gas exchanges of pumpkin plants grown in a hydroponic system, especially when using foliar application of A. brasilense. ABSTRACT Purpose: This study aimed to evaluate the therapeutic effect of an ethanol extract of Ocimum basilicum L. (EEOb) aerial parts against Ehrlich’s experimental tumor (EET) in mice. Methods: Swiss mice were divided into two groups (control and treated; n = 6). On day 21, all mice were inoculated subcutaneously with 2 × 106 (0.05 mL) EET cells in the left paw for solid tumor development. This study lasted 28 days. Treatment began 24 hours after inoculation with EET. Measurements of dorsoplantar thickness were used to assess tumor growth. The paw pad was collected for histopathological analysis and stained using the argyrophilic nucleolar organizing regions (AgNOR) technique and immunohistochemistry for proliferating cell nuclear antigen, Bcl-2 and Bax. Results: The treatment of animals with EEOb at 100 mg/kg intraperitoneally was able to reduce the growth (Control = 3.7 ± 0.1 mm vs. EEOb = 5.7 ± 0.2 mm) and the number of AgNORs of solid Ehrlich tumor. The antitumor effect of EEOb was associated with the induction of apoptosis of tumoral cell, as suggested by the reduction of the content of Bcl-2 induced by extract. Conclusions: The study demonstrated that daily administration of EEOb is able to reduce the growth of EET by induce apoptosis of tumoral cells. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO Tecnologias outrora aplicadas exclusivamente em sistemas convencionais, têm maximizado os ganhos em sistemas hidropônicos, porém, necessitando ainda de informações para uma adequada utilização. Assim, o estudo teve como objetivo avaliar o efeito da aplicação foliar de tiamina e niacina sobre o crescimento e atividades de troca gasosa em duas cultivares de alface em sistema hidropônico de fluxo e refluxo. O delineamento experimental utilizado foi o inteiramente casualizado (DIC), com quatro tratamentos e oito repetições. Os tratamentos foram: T1 - controle; T2 - tiamina em concentração de 200 mg L-1; T3 - niacina em concentração de 200 mg L-1; T4 - tiamina e niacina combinadas, em concentração de 100 mg L-1 de cada vitamina. Cada parcela experimental foi constituída por um vaso, contendo uma muda de alface. A aplicação conjunta de tiamina e niacina elevou as trocas gasosas das plantas de alface, aumentando em cerca de 30% a massa fresca, em relação ao controle, para ambas as cultivares, seguida pela aplicação isolada de niacina. Para a tiamina, a aplicação de forma isolada não resultou em alterações significativas. ABSTRACT Technologies once applied exclusively in conventional systems have maximized gains in hydroponic systems; however, they still need information for proper use. Thus, the study aimed to evaluate the effect of foliar application of thiamine and niacin on growth and gas exchange activities in two lettuce cultivars in an ebb and flow hydroponic system. The completely randomized design (CRD) was used with four treatments and eight replications. The treatments were as follows: T1 - control, T2 - thiamine at a concentration of 200 mg L-1, T3 - niacin at 200 mg L-1, and T4 - thiamine and niacin combined at 100 mg L-1 of each vitamin. Each experimental plot consisted of a lettuce plant. The combined application of thiamine and niacin stood out, increasing the activity of gas exchange of lettuce plants and increasing by about 30% the fresh mass, concerning the control, for both cultivars, followed by the isolated application of niacin. For thiamine, the application alone did not result in significant changes. The vitamins affect the morphophysiology of lettuce, where the use of niacin has the potential for use in hydroponic production systems. BACKGROUND Leishmania RNA virus 1 (LRV1) is commonly found in South American Leishmania parasites belonging to the subgenus Viannia, whereas Leishmania RNA virus 2 (LRV2) was previously thought to be restricted to the Old-World pathogens of the subgenus Leishmania. OBJECTIVES In this study, we investigated the presence of LRV2 in strains of Leishmania (L.) infantum, the causative agent of visceral leishmaniasis (VL), originating from different hosts, clinical forms, and geographical regions. METHODS A total of seventy-one isolates were screened for LRV2 using semi-nested reverse transcription-polymerase chain reaction (RT-PCR) targeting the RNA-dependent RNA polymerase (RdRp) gene. FINDINGS We detected LRV2 in two L. infantum isolates (CUR268 and HP-EMO) from canine and human cases, respectively. Resumo Fundamento O bloqueio atrioventricular (BAV) descreve um comprometimento na condução dos átrios para os ventrículos. Embora o curso clínico do BAV tenha sido avaliado, os achados são de países de alta renda e, portanto, não podem ser extrapolados para a população latina. Objetivo Avaliar a associação entre BAV e mortalidade. Métodos Foram incluídos pacientes do estudo CODE (Clinical Outcomes in Digital Electrocardiology), maiores de 16 anos que realizaram eletrocardiograma (ECG) digital de 2010 a 2017. Os ECGs foram relatados por cardiologistas e por software automatizado. Para avaliar a relação entre BAV e mortalidade, foram utilizados o modelo log-normal e as curvas de Kaplan-Meier com valores de p bicaudais < 0,05 considerados estatisticamente significativos. Resultados O estudo incluiu 1.557.901 pacientes; 40,23% eram homens e a média de idade foi de 51,7 (DP ± 17,6) anos. Durante um seguimento médio de 3,7 anos, a mortalidade foi de 3,35%. A prevalência de BAV foi de 1,38% (21.538). Os pacientes com BAV de primeiro, segundo e terceiro graus foram associados a uma taxa de sobrevida 24% (taxa de sobrevida relativa [RS] = 0,76; intervalo de confiança [IC] de 95%: 0,71 a 0,81; p < 0,001), 55% (RS = 0,45; IC de 95%: 0,27 a 0,77; p = 0,01) e 64% (RS = 0,36; IC de 95%: 0,26 a 0,49; p < 0,001) menor quando comparados ao grupo controle, respectivamente. Os pacientes com BAV 2:1 tiveram 79% (RS = 0,21; IC de 95%: 0,08 a 0,52; p = 0,005) menor taxa de sobrevida do que o grupo controle. Apenas Mobitz tipo I não foi associado a maior mortalidade (p = 0,27). Conclusão BAV foi um fator de risco independente para mortalidade geral, com exceção do BAV Mobitz tipo I.
Abstract Background Atrioventricular block (AVB) describes an impairment of conduction from the atria to the ventricles. Although the clinical course of AVB has been evaluated, the findings are from high-income countries and, therefore, cannot be extrapolated to the Latinx population. Objective Evaluate the association between AVB and mortality. Methods Patients from the CODE (Clinical Outcomes in Digital Electrocardiology) study, older than 16 years who underwent digital electrocardiogram (ECG) from 2010 to 2017 were included. ECGs were reported by cardiologists and by automated software. To assess the relationship between AVB and mortality, the log-normal model and the Kaplan-Meier curves were used with two-tailed p-values < 0.05 considered statistically significant. Results The study included 1,557,901 patients; 40.2% were men, and mean age was 51.7 (standard deviation ± 17.6) years. In a mean follow-up of 3.7 years, the mortality rate was 3.35%. The AVB prevalence was 1.38% (21,538). Patients with first-, second-, and third-degree AVB were associated with 24% (relative survival rate [RS] = 0.76; 95% confidence interval [CI]: 0.71-0.81; p < 0.001), 55% (RS = 0.45; 95% CI: 0.27-0.77; p = 0.01), and 64% (RS = 0.36; 95% CI: 0.26-0.49; p < 0.001) lower survival rate when compared to the control group, respectively. Patients with 2:1 AVB had 79% (RS = 0.21; 95% CI: 0.08-0.52; p = 0.005) lower survival rate than the control group. Only Mobitz type I was not associated with higher mortality (p = 0.27). Conclusion AVB was an independent risk factor for overall mortality, with the exception of Mobitz type I.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Laurus nobilis L. is a large shrub belonging to the Lauracea family. Its leaves are widely used for food seasoning as well as in folk medicine. Various studies have demonstrated the antiproliferative, antifungal and antibacterial effects of Laurus nobilis, but no studies have investigated the genotoxic effect of the aqueous extract of the plant. The objective of this study was to analyze the genotoxic potential of an aqueous extract of leaves, using the Allium cepa assay and mouse peripheral blood cell micronucleus test. The results showed that the extract did not have any genotoxic activity, but cytotoxic activity was observed in the two experimental models used. The extract had an antiproliferative effect, detected through the reduction of the mitotic index and the polychromatic/normochromatic erythrocyte (PCE/NCE) ratio. The tests also demonstrated a large number of cells undergoing apoptosis and with nuclear abnormalities related to cell death processes. These results can be explained by the presence of phenolic compounds, saponins, flavonoids and alkaloids, detected in the phytochemical analysis of the extract. Therefore, the extract from L. nobilis in the form generally used by the population does not pose risks related to its genotoxic potential, and also contains components with apoptotic and antigenotoxic potential.

Abstract Essential oils (EOs) are a promising alternative for food preservation and can be used instead of synthetic antimicrobial compounds such as sanitizers, as long as they are efficient and safe. In view of this possibility of use, Lippia alba (Mill) (LAEO) and Cymbopogon citratus D.C. Stapf. (CCEO) essential oils were evaluated as natural antimicrobials and applied into coriander for its biocontrol. The EOs were extracted and their physical properties, total phenolics, cytotoxicity to Artemia salina L. and antimicrobial potential in vitro and on natural coriander microbiota were determined. The EOs had a LD50 (median lethal dose - checked), are considered biologically active. MIC (Minimum Inhibitory Concentration) varied from 1.25 to 5.00 μL/ mL and from 0.62 to 5.00 μL/ mL for LAEO and CCEO, respectively, on standard strains of Staphylococcus aureus, Salmonella Thyphimurium, Salmonella Enteritidis, Escherichia coli and Listeria monocytogenes. The lowest MIC values for LAEO and CCEO were selected for application in coriander. EOs reduced the pathogenic microbial load in the coriander revealing its potential as a natural sanitizer, and thus demonstrating superior or equivalent efficacy to commercial sanitizer. By correlating the cytotoxic and antimicrobial results obtained, we have shown that these EOs present potential for application in foods.

Resumo Objetivou-se com o presente estudo determinar o teor de compostos bioativos e atividade antioxidante presentes nos frutos de três espécies vegetais do Cerrado Brasileiro: carnaúba (Copernicia prunifera (Mill.) H.E. Moore), murici (Byrsonima crassifolia L. Rich) e oiti (Licania tomentosa (Benth.) Fritsch). Entre os frutos analisados, o fruto oitiapresentou maior conteúdo de fenólicos totais (1236,42 ± 34,06 mg EAG 100 g-1), seguido do murici (468,90 ± 27,30 mg EAG 100 g-1) e do côco da carnaúba (314,44 ± 9,50 mg EAG 100 g-1). Quanto à atividade antioxidante, o murici apresentou 4350,31 ± 1,85 µmol TEAC.100 g-1 e o oiti que apresentou 14721,69 ± 0,85 µmol TEAC.100 g-1. Foi verificado também um elevado conteúdo de antocianinas no côco da carnaúba (9,35 ± 0,00 mg-cy-3-glu.100 g-1) e elevado conteúdo de carotenoides (20,0 ± 1,23 mg-ß-carot.100 g-1) e vitamina C (58,60 ± 1,32 mg.100 g-1) no murici. Os dados obtidos somam informações ao conhecimento atual sobre as características nutricionais e funcionais de frutos do Cerrado Brasileiro ainda pouco explorados, como os analisados, que apresentaram elevado conteúdo de compostos bioativos e atividade antioxidante.
Abstract The aim of this study was to determine the content of bioactive compounds and antioxidant activity present in three fruit species from the Brazilian Cerrado: carnauba (Copernicia prunifera (Mill.) In conclusion, data obtained add valuable information to the current knowledge on the nutritional and functional features of fruits from the Brazilian Cerrado that have been scarcely explored, such as those analyzed, which showed high content of bioactive compounds and antioxidant activity.

During biodiesel storage, chemical reactions may occur, producing sludges. The aim of this study was to characterize the chemical and microbial composition of the sludge found in a biodiesel storage tank. The material was collected in a biodiesel production plant. The sludge chemical characterization was performed by Fourier transform infrared (FTIR) spectroscopy, flame atomic absorption spectroscopy (FAAS), gas chromatography (GC), and the microbial investigation used culture-dependent techniques. The deteriogenic potential of the native microbial community was evaluated using the sludge as a microbial inoculum in a 60 day experiment. The microbial growth, biodiesel degradation, pH alterations, and the detection of esters in the aqueous phase were evaluated. The chemical analysis indicated the sludge composition as fatty acids esters and metallic ions; sterols glycosides were not detected. Seven bacteria and five fungi species were obtained from the sludge. The microbial growth analysis indicated that the native community does not have high biodiesel deteriogenic capacity.

Estimation, morphometry and ultrastructure of ovarian preantral follicle population in agouti (Dasyprocta leporina)

RESUMO: O objetivo deste trabalho foi caracterizar a população folicular ovariana pré-antral em cutias (D. leporina) estimando o número de folículos em cada categoria de desenvolvimento, e também descrever a morfometria e as características específicas do folículo e oócito usando microscopia de luz e eletrônica de transmissão. O comprimento de cada ovário foi medido utilizando um paquímetro, seccionados longitudinalmente em duas metades e ambos foram imediatamente fixados para realizar a estimativa da população folicular e análise ultraestrutural. A média (±S.E.M.) da população folicular por par de ovário foi estimada em 4419,8±532,26 e 5397,52±574,91 nos ovários direito e esquerdo, respectivamente, mas não foram observadas diferenças entre eles. Os diâmetros dos folículos, oócito e núcleos, respectivamente, foram: 18,62±3,40μm, 12,28±2.37μm e 6,10±0,93μm para primordial, 23,75±5,70μm, 14,22±3,00μm e 6,70±1,24μm para primário e 88,55±17,61μm, 52,85±17,56μm e 22,33±17,61μm de folículos secundários. A maioria dos folículos encontrados pertencia à categoria primordial (86,63%), seguido pelo primário (13,01%) e um secundário (0,35%). Adicionalmente, os folículos poliovulares foram observados em todos os animais e representavam 7,51% do total de folículos contados. A análise ultra-estrutural mostrou que o oócito apresentou núcleos centrais e regulares, exibindo uma massa homogênea. Dentre as organelas, as mitocôndrias foram as mais abundantes e o aparelho de Golgi do oócito foi raramente observado. Em conclusão, este trabalho mostra pela primeira vez a caracterização da população de folículos pré-antrais do ovário da Dasyprocta leporina. Essas informações serão úteis para o desenvolvimento e adaptação de biotécnicas, como a criopreservação de germoplasma e manipulação de gametas in vitro.
ABSTRACT: The aim of this study was to characterize the preantral ovarian follicular population in agoutis (D. leporina) by estimating the number of follicles at each developmental category, and also describe the morphometry and the specific features of the follicle and the oocyte by using light and transmission electron microscopy. The length of each ovary was measured using a caliper rule, longitudinally sectioned into two halves and both were immediately fixed to perform the estimation of follicular population and ultrastructural analysis. The mean (±S.E.M.) population of follicular per pair of ovary was estimated at 4419.8±532.26 and 5397.52±574.91 for right and left ovaries, respectively, but no differences were observed between them. The diameters for follicles, oocyte and nuclei were: 18.62±3.40μm, 12.28±2.37μm and 6.10±0.93μm for primordial, 23.75±5.70μm, 14.22±3.00μm and 6.70±1.24μm for primary and 88.55±17.61μm, 52.85±17.56μm and 22.33±17.61μm for secondary follicles, respectively. The most of the follicles found belonged to the primordial category (86.63%), followed by primary (13.01%) and secondary (0.35%) one. Additionally, polyovular follicles were observed in all the animals and they represented 7.51% of the total follicles counted. The ultrastructural analysis showed that the oocyte presented a central and regular nuclei, displaying a homogenous mass. Among the organelles, the mitochondria were the most abundant and the oocyte Golgi apparatus was rarely observed. In conclusion, this work shows for the first time the characterization of the population of preantral follicles in the ovary of Dasyprocta leporina. Those information will be useful for further development and adaptation of biotechniques such as germplasm cryopreservation and in vitro gametes manipulation.

Abstract Cashew, Anacardium occidentale L. (Anacardiaceae), is one of the most important sources of agricultural income in northeastern Brazil, but many of the arthropods associated with the crop have yet to be identified. We describe here for the first time the damage caused by Holopothrips fulvus (Morgan) (Thysanoptera: Phlaeothripidae) to dwarf-cashew trees cultivated in the municipality of Pacajús, Ceará, Brazil. Leaf tissue injuries were caused by the sucking mouthparts of the insect and were characterized by dark necrotic spots on the epidermis that resulted in yellowing, wilting and, ultimately, abscission of the leaves. H. fulvus also fed on developing kernels and pseudofruits producing injuries that manifested in the form of chlorotic specks. Additional information is given on the pest status and important aspects of the morphology of the insect, including sexual dimorphism, redescription of the adults and description of the second instar larvae.

Resumo: O objetivo deste estudo foi criopreservar o sêmen de tatus-peba (Euphractus sexcinctus) em diluente Tris-gema e glicerol, e determinar os danos causados pelo processo de congelação-descongelação, utilizando marcadores fluorescentes e análise ultraestrutural. As amostras de sêmen (n=11) coletadas de 4 tatus-peba adultos por eletroejaculação foram criopreservadas em diluente Tris acrescido de 20% de gema de ovo e 3% de glicerol, em curva rápida de congelação. A análise clássica das amostras foi realizada após a diluição, refrigeração e descongelação, seguida por análise de fluorescência, utilizando uma combinação de sondas fluorescentes para avaliar a integridade da membrana (Iodeto de Propídio - PI e Hoechst - H342), e a atividade mitocondrial (CMXRos - Mito Tracker RED®). Foi também utilizada a análise ultraestrutural para verificar possíveis alterações morfológicas causadas pela crioinjúria. Quando comparadas com as amostras a fresco, verificou-se uma queda significativa em todos os parâmetros seminais dos tatus após a descongelação, em que apenas 6,1% de espermatozoides móveis foram encontrados. No entanto, o percentual de espermatozoides que permaneceu com membrana viável (13%) e funcional (24,7%) após a descongelação sugere que algumas células podem estar vivas, mas imóveis. Análises utilizando marcadores fluorescentes revelaram que as mitocôndrias dos espermatozoides de tatus são altamente sensíveis ao protocolo de congelação e os achados através da análise ultraestrutural comprovaram esta afirmação. Além disso, as imagens obtidas por microscopia eletrônica de transmissão revelaram que espermatozoides congelados-descongelados apresentaram membranas plasmáticas danificadas, modificações nucleares como alterações na cromatina, e alterações acrossomais relativas à capacitação espermática. Em conclusão, este estudo é a primeira tentativa de criopreservação de sêmen em uma espécie de tatu, e nos auxiliou a identificar pontos críticos no processo de congelação-descongelação, a fim de melhorar o protocolo.
Abstract: The aim of the study was to cryopreserve the semen of six-banded armadillos (Euphractus sexcinctus) in Tris-yolk and glycerol diluent, and to determine the damage caused by the freezing-thawing process, using fluorescent markers and ultrastructural analysis. Semen samples (n=11) collected from 4 adult six-banded armadillos by electroejaculation were cryopreserved in Tris diluent plus 20% egg yolk and 3% glycerol, in a fast freezing curve. Classical analysis of samples was performed after dilution, refrigeration and thawing, followed by fluorescence analysis, using a combination of fluorescent probes to assess membrane integrity (propidium iodide - PI and Hoechst - H342), and mitochondrial activity (CMXRos - Mito Tracker Red®). We also used the ultrastructural analysis to verify possible morphological alterations caused by cryoinjuries. When compared with fresh samples, we verified a significant decline in all the armadillos' semen parameters after thawing, in which only 6.1% motile sperm were found. However, the percentage of sperm which remained with viable (13%) and functional (24.7%) membranes after thawing suggests that some cells could be live but immotile. Analysis using fluorescent markers revealed that the mitochondria of armadillos' sperm is highly sensible to the freezing protocol and the findings through ultrastructure analysis proved this statement. Additionally, the images obtained by transmission electron microscopy revealed that frozen-thawed sperm presented damaged plasma membrane, nuclear modifications as changes in chromatin and acrossomal changes relative to sperm capacitation. In conclusion, this study is the first attempt to cryopreserve the semen of an armadillo species, and to help us to identify critical points on the freezing-thawing procedure in order to improve the protocol.

Spatialization of soil quality index in the Sub-Basin of Posses, Extrema, Minas Gerais

RESUMO Objetivou-se, com este estudo, determinar o índice de qualidade do solo (IQS), em relação a atributos químicos e físicos, e espacializá-lo, além de avaliar a utilização deste índice no pagamento por serviços ambientais na Sub-Bacia Hidrográfica das Posses, Extrema, Minas Gerais, representativa do Bioma Mata Atlântica. Os valores do IQS foram influenciados tanto pela substituição da mata nativa por povoamento de eucalipto, quanto por pastagens e culturas anuais, refletindo na redução da qualidade do solo na profundidade amostrada nos sistemas avaliados. A espacialização do IQS apresentou valores variando de 0,40 a 0,80, ocorrendo algumas áreas pontuais com elevados índices e algumas com índices superiores a 1,00 (mata nativa). O reflorestamento com eucalipto condicionou solos, em sua maioria, com baixas deteriorações físicas e químicas devido ao acúmulo de serrapilheira. Já os menores valores do IQS estão associados às pastagens. O modelo pelo qual o IQS se ajustou foi o exponencial, possibilitando a krigagem ordinária. O IQS apresenta grande potencial para uso no pagamento de agricultores que prestam serviços de conservação do solo e água.
ABSTRACT This study aimed to determine and spatialize the soil quality index (SQI), in relation to chemical and physical attributes, and evaluate its use in the payment for environmental services in the Sub-Basin of Posses, Extrema-MG, Brazil, which represents the Atlantic Forest Biome. SQI values were influenced by both the replacement of native forests by stands of eucalyptus and by pastures and annual crops, reflecting in the reduction of soil quality in the sampled layer in the evaluated systems. The spatialization of SQI showed values ranging from 0.40 to 0.80, with some specific areas with high values and others with values above 1.00 (native forest). The reforestation with eucalyptus conditioned most of the soils with low chemical and physical deterioration, due to accumulation of litter. The lowest SQI values are associated with pastures. SQI adjusted to the exponential model, which allowed the use of ordinary Kriging. The SQI has a great potential of use in the payment to farmers who provide services of soil and water conservation.
