Phytochemical study of the roots of Euphorbia phosphorea Mart. (Euphorbiaceae) was carried out through chromatographic techniques, resulting in the isolation of a new ent-abietane diterpene named 11β,12β-dihydroxy-ent-abieta-8(14),13(15)-dien-16,12α-olide (1), and of nine known ent-abietane diterpenes jolkinolide A (2), jolkinolide E (3), euphorin H (4), euphopilolide (5) jolkinolide F (6), ent-12-hydroxy-12[R]-abieta-8(14),13(15)-dien-16,12-olide (7), ent-11α-hydroxyabieta-8(14),13(15)-dien-16,12α-olide (8), 17-hydroxyjolkinolide B (9) and caudicifolin (10). The structures of all compounds were established using spectroscopic techniques such as 1D and 2D NMR, and the structure of the compound 1 was established also with MS, IR and ECD. All compounds were submitted to an in silico study through of a predictive model and then submitted to in vitro tests against Mycobacterium tuberculosis and M. smegmatis for evaluation of their antimycobacterial activity. Compounds 5 and 9 showed mycobacterial growth inhibition with MIC values of 62.5 μM against M. tuberculosis and M. smegmatis, respectively.

OBJECTIVES: This prospective, randomized, open-label study aimed to compare the effects of antihypertensive treatment based on amlodipine or hydrochlorothiazide on the circulating microparticles and central blood pressure values of hypertensive patients. METHODS: The effects of treatments on circulating microparticles were assessed during monotherapy and after the consecutive addition of valsartan and rosuvastatin followed by the withdrawal of rosuvastatin. Each treatment period lasted for 30 days. Central blood pressure and pulse wave velocity were measured at the end of each period. Endothelial, monocyte, and platelet circulating microparticles were determined by flow cytometry. Central blood pressure values and pulse wave velocity were recorded at the end of each treatment period. RESULTS: No differences in brachial blood pressure were observed between the treatment groups throughout the study. Although similar central blood pressure values were observed during monotherapy, lower systolic and diastolic central blood pressure values and early and late blood pressure peaks were observed in the amlodipine arm after the addition of valsartan alone or combined with rosuvastatin. Hydrochlorothiazide-based therapy was associated with a lower number of endothelial microparticles throughout the study, whereas a higher number of platelet microparticles was observed after rosuvastatin withdrawal in the amlodipine arm. CONCLUSIONS: Despite similar brachial blood pressure values between groups throughout the study, exposure to amlodipine was associated with lower central blood pressure values after combination with valsartan, indicating a beneficial interaction. Differences between circulating microparticles were modest and were mainly influenced by rosuvastatin withdrawal in the amlodipine arm.

O perfil metabólito baseado na aplicação de análises multivariadas (análise de componentes principais, PCA) dos dados de espectrometria de massas com ionização electrospray (ESIMS) no modo positivo e de ressonância magnética nuclear (RMN) do ¹H de extratos brutos de espécies de Piper destacou algumas espécies caracterizadas pela produção de lignanas (P. solmsianum, P. truncatum e P. cernuum), neolignanas (P. regnellii) e cromenos (P. gaudichaudianum). Análises específicas em conjunto de espécies caracterizadas morfologicamente por apresentarem inflorescências pêndulas e globosas (P. caldense, P. carniconnectivum, P. bowiei e P. permucronatum) ou em espécies que produzem amidas indicaram o potencial mais significativo para tais análises como critério para estudos fitoquímicos posteriores. Análises intraespecíficas de plântulas das espécies P. solmsianum, P. regnellii e P. gaudichaudianum indicaram uma composição química nas folhas baseada na presença dos fenilpropanóides dilapiol e apiol, diferentemente do que produzem as plantas adultas. No caso das espécies que produzem amidas, a composição apresentou-se relativamente constante independentemente do estágio de desenvolvimento.

The metabolomic profiling based on the application of multivariate analysis (principal component analysis, PCA) of positive mode electrospray ionization mass spectrometric (ESIMS) and ¹H nuclear magnetic resonance (NMR) data of crude extracts highlighted some species characterized by lignans (P. solmsianum, P. truncatum and P. cernuum), neolignans (P. regnellii) and chromenes (P. gaudichaudianum). A specific analysis focusing on species having pendant and globular inflorescences (P. caldense, P. carniconnectivum, P. bowiei and P. permucronatum) or amides-producing species indicated higher potential of the methodology in determining similarities and establishing priorities for further phytochemical investigation. Such intraspecific analysis applied to analyzed seedling leaves of the P. solmsianum, P. regnellii and P. gaudichaudianum species revealed the production of dillapiole and apiole instead of lignans, neolignans or prenylated benzoic acid, produced by the adult leaves, respectively. In case of amides-producing species, a similar profile was observed regardless the developmental stage.

Intrahepatic cholestasis of pregnancy is associated with preterm labor and delivery. We hypothesize that in the course of this disease, bile acids activate the oxytocin receptor pathway leading to the premature onset of labor. We evaluated in vitro and in vivo myometrial sensitivity to oxytocin and the oxytocin-receptor expression in patients with cholestasis and control. Furthermore, we evaluated the ability of cholic acid to mediate the activation of the oxytocin-receptor pathway. Patients with cholestasis of pregnancy, compared to controls, required lower doses of oxytocin to elicit 3 uterine contractions in 10 minutes (1.3 ± 0.6 vs 3.6 ± 0.8 Units; p < 0.05) and had lower in vitro ED50 (1.6 x 10_10 M vs 1.0 x 10_8 M; p < 0.05), reflecting higher oxytocin sensitivity. The long-term (24 h) incubation of myometrial strips from control patients with cholic acid (20 µM) markedly increased the sensitivity to oxytocin, but not to endothelin-1 or prostaglandin F2a. The 24 h incubation of cultured myometrial cells with cholic acid increased the expression of oxytocin-receptor at mRNA (northern blot) and protein levels (western blot and immunohistochemistry). Our results demonstrate that in the setting of intrahepatic cholestasis of pregnancy, an activation of the oxytocin-receptor pathway occurs. The activation of such pathways seems to be the result of a cholic acid mediated increase in receptor expression, which in turn could explain the increase in the preterm labor and delivery rate observed in this disease

Se ha reportado una asociación entre CIE y distrés fetal, muerte fetal in utero y síntomas de parto prematuro/parto prematuro. Con la hipótesis que en la CIE el aumento en los ácidos biliares, en particular ácido cólico, activa la vía ocitocina-receptor de ocitocina (OT-ROT) en el miometrio, generando un inicio prematuro del trabajo de parto, evaluamos la sensibilidad miometrial a OT in vitro e in vivo, y la expresión de mRNA en pacientes CIE y controles y la capacidad de ácido cólico para regular la activación de la vía OT-ROT. Las pacientes con CIE mostraron una alta sensibilidad a OT in vivo e in vitro comparado con controles. Al incubar segmentos/células en cultivo de miometrio con ácido se observó una marcado aumento de la sensibilidad contráctil para la OT y un aumento de la expresión celular del mRNA y los niveles de la proteína del ROT. La activación de la vía OT-ROT parece ser el resultado de un aumento de la síntesis/función de RsOT mediada por el ácido cólico. Esto resultados entregan las bases celulares/moleculares que podrían explicar la asociación entre CIE y parto prematuro

A biologia de Hippodamia convergens Guérin-Meneville foi estudada utilizando-se ovos de Anagasta kuehniella (Zeller) como alimento das larvas, em comparação com dieta comumente utilizada, constituída de adultos dos pulgões Schizaphis graminum (Rondani) e Brachycaudus (Appelia) schwartzi Börner. Para os adultos do predador, utilizaram-se adultos do pulgão S. graminum. Os experimentos foram conduzidos a 25 ± 1°C, UR de 70 ± 10% e fotofase de 12 h. O período embrionário, quando as larvas foram alimentadas com as três dietas foi de 3,1; 3,2 e 3,2 dias respectivamente. A duração do período de larva a adulto foi maior na dieta constituída de ovos de A. kuehniella (19,5 dias), entretanto a viabilidade para esse período não diferiu entre as três dietas testadas. Para a fase adulta do predador oriundo de larvas criadas sob os três regimes alimentares, todos os parâmetros reprodutivos e a longevidade foram estatisticamente iguais. Assim, verificou-se que ovos de A. kuehniella podem ser considerados uma dieta em potencial, em substituição aos pulgões S. graminum e B. schwartzi na criação de larvas de H. convergens.

The biology of Hippodamia convergens Guérin-Meneville was studied on three diets, at temperature 25 ± 1°C; 70 ± 10% RH and 12 hours photofase. Eggs of Anagasta kuehniella (Zeller) and two species of aphids, Schizaphis graminum ( Rondani) and Brachycaudus (Appelia) schwartzi Börner, were fed to the larvae. S. graminum was fed to the adults. The lenght of the larval-adult period of H. convergens was higher when the larvae fed on eggs of A. kuehniella (19.5 days), than when they fed on the aphids S. graminum (17.0 days) or B. schwartzi (17.9 days). However, the viability of larva-adult period showed no significant difference among all three tested diets. Longevity and all the reproductive parameters at the adult phase of H. convergens showed no significant difference when the larvae were reared on the three diets. Eggs of A. kuehniella were shown as a suitable alternative food, that be used as a substitute of the aphids, the most commom prey to larvae rearing of H. convergens.

We report the occurence of the bamboo aphid, Takecallis taiwanus (Takahashi) (Homoptera: Aphididae) in Contagem, Minas Gerais State. This aphid was observed causing injuries on leaves and shoots of bamboo Phyllostachys cf. aurea A & C. Reveerii. This is the first record of this aphid on bamboo in Brazil.

Estudamos 11 pacientes (9 do sexo masculino) com diagnóstico citogenético de síndrome do X frágil (SXF) com o objetivo de se investigar o circuito neuronal afetado nesta entidade. As idades variaram de 8 a 19 anos. Todos os indivíduos apresentaram pavilhões auriculares grandes, faces alongadas e traços autistas. Dez deles tinham retardo mental grave. Detectou-se a presença de distúrbio da atenção em 10 pacientes. O eletrencefalograma revelou-se anormal em 6 de 10 indivíduos examinados, mostrando descargas epileptiformes focais predominantemente em áreas frontais e parietais. Todos foram submetidos a ressonância magnética craniana que se revelou anormal em 8. As anormalidades mais importantes foram redução do verme cerebelar e aumento do IV ventrículo. Realizou-se tomografia por emissão de photon único (SPECT) em 7 pacientes. Todos os exames estavam alterados sendo o principal achado a hipoperfusão das porções inferiores dos lobos frontais. Com base no quadro clínico, achados neuropsicológicos e resultados dos exames funcionais e de imagem, sugerimos que a SXF apresenta disfunção de ampla rede neuronal: cerebelo regiões fronto basais lobos parietais. A literatura aponta para a presença de distúrbio envolvendo o mesmo circuito neuronal em pacientes autistas.

We studied 11 patients (9 males) with cytogenetic diagnosis of fragile X syndrome (FXS) with the purpose of investigating the neural circuitry involved in this condition. The ages ranged from 8 to 19. All the individuals presented large ears, elongated faces and autistic features. Ten patients had severe mental retardation. Attention disorder was found in 10 individuals. Electroencephalographic recordings were abnormal in 6 of 10 patients examined, showing focal epileptiform discharges predominantly in frontal and parietal areas. All patients underwent magnetic resonance imaging studies which were abnormal in 8 of them. The most important abnormalities were reduction of the cerebellar vermis and enlargement of the IV ventricle. Single photon emission computerized tomography (SPECT) was performed in 7 patients and was abnormal in all of them, the most frequent finding being a hypoperfusion of the inferior portions of the frontal lobes. Based on the clinical picture, neuropsychological findings and functional and structural imaging studies we suggest that FXS presents with a dysfunction involving a large area of the central nervous system: cerebellum - basal frontal regions - parietal lobes. The literature points to a disturbance involving the same neural circuitry in patients with autism.

Estudou-se a ocorrência e a flutuação populacional dos inimigos naturais (predadores e parasitóides), associados ao pulgão Brachycaudus schwartzi (Börner), em pessegueiro (Prunus persica). Foram amostrados quinzenalmente de agosto de 1994 a julho de 1995, dois ramos de aproximadamente 25 cm por planta em 10 plantas. No laborátório, foi realizada a triagem e contagem dos pulgões e de seus inimigos naturais. Constatou-se a presença de predadores das famílias Coccinellidae (seis espécies) e Syrphidae (sete espécies) e também do parasitóide Aphidius colemani Viereck (Aphidiidae). Os predadores foram mais abundantes de agosto a dezembro de 1994, coincidindo com a maior ocorrência do pulgão B. schwartzi. Dentre as espécies de coccinelídeos amostradas, a espécie Scymnus (Pullus) argentinicus (Weise) destacou-se, correspondendo a 43,9% do total de coccinelídeos amostrados. Os coccinelídeos apresentaram o pico populacional no mês de outubro. A espécie Allograpta neotropica Curran (Syrphidae) foi a de maior ocorrência, com maior pico em setembro, correpondendo a 86,7% dos sirfídeos amostrados. O pico populacional de A. colemani significativo também ocorreu no mês de setembro.

The occurrence and population fluctuation of natural enemies (predators and parasitods) associated with Brachycaudus schwartzi.(Börner) were studied on peach (Prunus persica). Two twigs 25 cm long were sampled every 15 days in 10 plants from August 1994 to July 1995 and taken to the laboratory for assessing the incidence of aphids and their natural enemies. Over the period of greatest aphid occurrence, predators of the families Coccinellidae (six species) and Syrphidae (seven species) and the parasitoid Aphidius colemani Viereck (Aphidiidae) were observed. Predators occurred at higher incidence from August to December, 1994, where occurrence of B. schwartzi was greatest. Among the species of Coccinellidae surveyed, the occurrence of Scymnus (Pullus) argentinicus (Weise) was higher and represented 43,9% of the total coccinellids surveyed. The coccinellid population peaked in October. The species Allograpta neotropica Curran (Syrphidae) showed highest occurrence (86,7%) among Syrphidae and its population peaked in September as did A. colemani.
