ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. This study assessed the effects of volume concentration (0.3%, 0.6%, 1.2%, and 12% v/v) and type of polymer flocculant (acrylamide, chitosan, and purified and non-purified acrylamide grafted chitosan) on the flocculation mechanism of kaolinite-rich mining tailings samples. We measured the flocculated samples’ transmittance using UV-VIS spectrometry to check for particles in the supernatant medium in order to verify the flocculation efficiency. It was observed that flocculation directly depends on the pH of the medium, the isoelectric point of the kaolinite, and the polymer chains’ molecular mass. The concentration of H+ and OH- ions is responsible for producing charges of different electrical nature on the particles present in the suspension, thereby affecting the flocculation process. In the acrylamide samples, the aggregation of particles occurs by electrostatic interaction; and in the other samples by bridging, suggesting that the chains’ molecular mass interferes in the coagulation and flocculation mechanisms. Moreover, the results of absorbance measurements and flocs’ apparent density highlight that the grafting of acrylamide on the chitosan chain may improve the kaolin waste flocculation. For flocculant concentrations of up to 1.2% (v/v), the absorbances were mostly below 1.0, indicating removal of suspended materials.

ABSTRACT Background Triatoma infestans (Hemiptera: Reduviidae) is a hematophagous insect and the main vector of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae). In the present study, the authors investigated whether a serine protease activity from the saliva of T. infestans has a role in vasomotor modulation, and in the insect-blood feeding by cleaving and activating protease-activated receptors (PARs). Methods T. infestans saliva was chromatographed as previously reported for purification of triapsin, a serine protease. The cleavage activity of triapsin on PAR peptides was investigated based on FRET technology. Mass spectrometry was used to analyze the sites of PAR-2 peptide cleaved by triapsin. NO measurements were performed using the DAN assay (2,3-diaminonapthalene). The vasorelaxant activity of triapsin was measured in vessels with or without functional endothelium pre-contracted with phenylephrine (3 µM). Intravital microscopy was used to assess the effect of triapsin on mouse skin microcirculation. Results Triapsin was able to induce hydrolysis of PAR peptides and showed a higher preference for cleavage of the PAR-2 peptide. Analysis by mass spectrometry confirmed a single cleavage site, which corresponds to the activation site of the PAR-2 receptor. Triapsin induced dose-dependent NO release in cultured human umbilical vein endothelial cells (HUVECs), reaching a maximum effect at 17.58 nM. Triapsin purified by gel-filtration chromatography (10-16 to 10-9 M) was applied cumulatively to mouse mesenteric artery rings and showed a potent endothelium-dependent vasodilator effect (EC30 = 10-12 M). Nitric oxide seems to be partially responsible for this vasodilator effect because L-NAME (L-NG-nitroarginine methyl ester 300 µM), a nitric oxide synthetase inhibitor, did not abrogate the vasodilation activated by triapsin. Anti-PAR-2 antibody completely inhibited vasodilation observed in the presence of triapsin activity. Triapsin activity also induced an increase in the mouse ear venular diameter. Conclusion Data from this study suggest a plausible association between triapsin activity mediated PAR-2 activation and vasodilation caused by T. infestans saliva.

ABSTRACT Matrix Assisted Laser Desorption/Ionization and Time of Flight mass spectrometry (MALDI-TOF MS) is a powerful tool for the identification of bacteria through the detection and analysis of their proteins or fragments derived from ribosomes. Slight sequence variations in conserved ribosomal proteins distinguish microorganisms at the subspecies and strain levels. Characterization of Leptospira spp. by 16S RNA sequencing is costly and time-consuming, and recent studies have shown that closely related species (e.g., Leptospira interrogans and Leptospira kirschneri) may not be discriminated using this technology. Herein, we report an in-house Leptospira reference spectra database using Leptospira reference strains that were validated with a collection of well-identified Brazilian isolates kept in the Bacterial Zoonosis Laboratory at the Veterinary Preventive Medicine and Animal Health Department at Sao Paulo University. In addition, L. interrogans and L. kirschneri were differentiated using an in-depth mass spectrometry analysis with ClinProTools™ software. In conclusion, our in-house reference spectra database has the necessary accuracy to differentiate pathogenic and non-pathogenic species and to distinguish L. interrogans and L. kirschneri.

Abstract Introduction: Donor shortage and organ allocation is the main problem in pediatric heart transplant. Mechanical circulatory support is known to increase waiting list survival, but it is not routinely used in pediatric programs in Latin America. Methods: All patients listed for heart transplant and supported by a mechanical circulatory support between January 2012 and March 2016 were included in this retrospective single-center study. The endpoints were mechanical circulatory support time, complications, heart transplant survival and discharge from the hospital. Results: Twenty-nine patients from our waiting list were assessed. Twelve (45%) patients were initially supported by extracorporeal membrane oxygenation (ECMO) and a centrifugal pump was implanted in 17 (55%) patients. Five patients initially supported by ECMO were bridged to another device. One was bridged to a centrifugal pump and four were bridged to Berlin Heart Excor®. Among the 29 supported patients, 18 (62%) managed to have a heart transplant. Thirty-day survival period after heart transplant was 56% (10 patients). Median support duration was 12 days (interquartile range [IQR] 4 - 26 days) per run and the waiting time for heart transplant was 9.5 days (IQR 2.5-25 days). Acute kidney injury was identified as a mortality predictor (OR=22.6 [CI=1.04-494.6]; P=0.04). Conclusion: Mechanical circulatory support was able to bridge most INTERMACS 1 and 2 pediatric patients to transplant with an acceptable complication rate. Acute renal failure increased mortality after mechanical circulatory support in our experience.

Abstract This study aimed to assess the feasibility of comet and cytogenetic tests as tools for evaluating genomic instability in seeds of Oryza sativa L. (rice) and Phaseolus vulgaris (beans) L. from gene banks. Rice and beans were exposed to methyl methanesulfonate (MMS) as a reference DNA damaging agent. Seeds of two accessions of rice and beans were obtained from Embrapa Rice and Beans - Brazil. Seed groups were imbibed in three concentrations of MMS for three periods of time to carry out cytogenetic tests, and for one period for the comet test. At concentrations of 10 and 15 mg/L, MMS induced cytotoxic and/or mutagenic effects in the meristematic cells of roots from all the accessions of both species. In the comet test, MMS induced genotoxic effects at all the concentrations in the evaluated accessions of rice and beans, except in one accession of beans at the lowest concentration (5 mg/L). Both species showed sensitivity to MMS. The comet test can be proposed for the measurement of genomic instability in accessions of rice and beans in gene banks, as being more sensitive than the cytogenetic tests used.

Composites based on nanosized particles of magnetite and natural microsized zeolite were prepared and their efficiency to adsorb arsenate from aqueous systems was evaluated through kinetic data and adsorption isotherms, considering the proportion of magnetic iron oxide, composite:solution ratio and zeolite particle size. A well-crystallized nanomagnetite with an averaged particle size of 50 nm when supported on zeolite was obtained; the nanomagnetite enhances the arsenate adsorption of zeolite. The composite prepared with a nanomagnetite:zeolite mass ratio of 0.43:1 and 0.30:1 presented on zeolite milled 20 and 120 min, respectively, showed a similar adsorption capacity as that observed for pure magnetite. Supported nanomagnetite prevents risks of the nanoparticles contamination. The smaller the mean particle size of zeolite the lower the mass proportion of nanomagnetite in the composite required to reach the maximum adsorptive efficiency. Acidic conditions improve arsenate remotion, in agreement with the development of positive surface charge in the composite.

RACIONAL: Isquemia gástrica representa importante desafio médico e geralmente é decorrente de processos vasculares obstrutivos ou não-obstrutivos. Apesar da rica arcada vascular do estômago, lesões de isquemia gástrica têm sido observadas. Quando a isquemia progride, pode culminar em necrose do órgão. OBJETIVO: Avaliar os efeitos dos diferentes tempos de isquemia gástrica sobre os tecidos da parede do estômago de coelhos mediante desvascularização total de ambas as curvaturas gástricas. MÉTODO: Foram utilizados 15 coelhos machos da raça Nova Zelândia. Os animais foram distribuídos nos seguintes grupos: grupo 1, observados por 3 h; grupo 2, por 6 h; grupo 3, por 12 h. A técnica cirúrgica foi a mesma em todos os animais e consistiu na ligadura e secção de todas as veias e artérias da parede gástrica. Após o período pré-determinado de observação, o estômago foi removido por completo para estudo macro e microscópico. RESULTADOS: As alterações macroscópicas e histológicas tornaram-se mais intensas com o aumento do tempo de isquemia e foram mais evidentes nas regiões do fundo e do corpo. Por outro lado, o antro manteve-se preservado na maioria dos animais operados. Necrose de mucosa do corpo e do fundo foi observada em todos os animais estudados, e foi acompanhada por hemorragia em 60% dos coelhos dos grupos 2 e 3. CONCLUSÃO: O modelo experimental de isquemia gástrica foi eficaz para produzir necrose hemorrágica de fundo e corpo do estômago de coelhos mesmo em curto tempo. Por outro lado, a região do antro pilórico desses animais mostrou-se significativamente mais resistente à isquemia do que as demais regiões.
BACKGROUND: Despite the rich vascular arcade of the stomach, gastric ischemia represents an important medical challenge and can be the consequence of obstructive or non-obstructive vascular processes of pathological or iatrogenic origin. AIM: To assess the effects of acute gastric ischaemia on the different regions of the stomach. METHOD: Fifteen New Zeland rabbits were divided into three groups: group 1, animals were observed during 3 h; group 2, during 6 h; group 3, during 12 h. Rabbit stomachs were subjected to devascularization of the greater and lesser curvatures. After predetermined time, the stomachs were removed for macro and microscopic studies. RESULTS: Haemorrhagic necrosis was more marked in the gastric fundus and body. In contrast, the antropylorus remained preserved in 80% of the animals. Necrosis of the gastric body and fundus mucosa were observed in all animals after 6 h and 12 h of ischaemia. CONCLUSION: Acute gastric ischaemia in rabbits produces haemorrhagic necrosis of the gastric fundus and body even in a short period of time. Beside this, the antropyloric region was significantly more resistant to ischaemia.

Recentemente nosso grupo demostrou experimentalmente que a hipótese de produção de radical acetil em meio tamponado no sistema diacetil/peroxinitrito era possível. Diacetil é um flavorizante em alimentos, cigarros e bebidas. O peroxinitrito é encontrado na mitocôndria, e em certas condições como um processo de infeção em humanos, a concentração aumenta significativamente. Em sistemas biológicos, radicais podem facilmente modificar a estrutura e atividade de ácidos nucleicos, proteínas e outras biomoléculas, causando significativo estres oxidativo. Baseado em dados de ressonância magnética e espectrometria de massas, este artigo apresenta os produtos que provam a produção do radical acetil e a formação de compostos estáveis devido à ligação covalente entre o acetil e os aminoácidos (produtos acetilados), e os adutos de peptídeos e proteínas. Estes materiais foram separados por eletroforese capilar e identificados por espectrometria de massas. O meio reacional consistiu da mistura de diacetil e peroxynitrito em uma relação de 1:2 e em 20 mmol L-1 de fosfato de sódio, no valor de pH 7,2. Estes experimentos também revelam a dupla acetilação da lisina, demonstrando a alta reatividade do composto com biomoléculas contendo grupos de nitrogênio, abundantemente encontradas em sistemas biológicos. A mudança estrutural de uma molécula acetilada é uma fonte de modificações pós-tradução com inúmeras consequências biológicas.
Recently, our group proposed a process that generated acetyl radicals in a reaction medium buffered with a diacetyl/peroxynitrite system. Diacetyl is a flavoring agent in food, cigarettes and drinks. Peroxynitrite is found in mitochondria, and in certain conditions, such as an infection in humans, its concentration is augmented significantly. In biological systems, radical compounds can easily modify the structure and activity of nucleic acids, proteins and other biomolecules, causing significant oxidative stress. Based on paramagnetic resonance and mass spectrometry data, this work discusses products that prove acetyl radicals are produced and are able to form stable covalent bonds with amino acid (acetylated products), peptide and protein adducts. These materials were separated and detected by capillary electrophoresis coupled with tandem mass spectrometry or offline mass spectrometry. The reaction medium contained a 1:2 mixture of diacetyl and peroxynitrite dissolved in 200 mmol L-1 of pH 7.2 sodium phosphate buffer. These experiments also reveal the double acetylation of lysine, demonstrating the high reactivity of these compounds when in contact with nitrogen-containing biomolecules readily found in biological systems. These structural changes might be an epigenetic source of post-translational protein modification.

INTRODUÇÃO: Isquemia gástrica representa um importante desafio médico nas áreas de Patologia e Cirurgia. O objetivo do presente trabalho foi avaliar os efeitos da isquemia gástrica aguda nas diferentes regiões do estômago. MÉTODO: Estômagos de coelhos foram submetidos à completa desvascularização das curvaturas maior e menor durante 3, 6 e 12 horas. Após esses períodos de tempo, os órgãos foram removidos para análise macro e microscópica. RESULTADOS: Necrose hemorrágica foi mais evidente nas regiões de corpo e fundo. Por outro lado, a região antropilórica manteve-se preservada em 100% dos coelhos após 3 horas de isquemia (grupo I), e em 80% dos coelhos após 6 e 12 horas de isquemia (grupos II e III). Necrose da camada mucosa do corpo e do fundo gástricos foi observada em todos os animais após 6 e 12 horas de isquemia. CONCLUSÃO: Conclui-se que esse modelo de isquemia gástrica aguda foi eficaz para produzir necrose hemorrágica do corpo e fundo de coelhos mesmo por um curto período de tempo. Além disso, a região antropilórica manteve-se preservada na maioria dos animais.
INTRODUCTION: Gastric ischemia represents an important medical challenge in pathology and surgical practice. OBJECTIVE: The purpose of this study was to assess the effects of acute gastric ischemia on different regions of the stomach. METHOD: Rabbit stomachs were subjected to devascularization of the greater and lesser curvatures for 3, 6 and 12 hours. After these periods, the stomachs were removed for macro and microscopic analysis. RESULTS: Hemorrhagic necrosis was more marked in the gastric fundus and body. In contrast, the antropylorus remained preserved in 100% of the rabbits after 3 hours of ischemia (group I), and in 80% of the rabbits after 6 and 12 hours of ischemia (groups II and III). Necrosis of the gastric body and fundus mucosa were observed in all animals after 6 and 12 hours of ischemia. CONCLUSION: We concluded that this experimental model of acute gastric ischemia was effective in producing hemorrhagic necrosis of the gastric fundus and body in rabbits even within a short period of time. Furthermore, the antropyloric region was preserved in most animals.

Solanum lycocarpum A.St.-Hil (Solanaceae) é um arbusto típico da região central do Brasil (Cerrado). A atividade alelopática do extrato aquoso de folhas e frutos dessa espécie já foi verificada em estudos anteriores. O objetivo desse trabalho foi avaliar a atividade alelopática de diferentes extratos de S. lycocarpum na germinação e crescimento de quatro espécies-alvo. As folhas foram coletadas, secas e trituradas e submetidas a dois métodos distintos de extração: 1- líquido-líquido (acetato de etila e diclorometano) do extrato aquoso das folhas e 2- com solventes em polaridade crescente (hexano, diclorometano, acetato de etila, acetona, metanol e água) diretamente das folhas. Cada extração foi realizada com equipamento de ultrassom durante uma hora, filtrado e evaporado. Desses extratos, soluções de 800, 400 e 200 ppm foram preparadas, e água e Logran® foram usados como controle positivo e negativo, respectivamente. Cada solução, bem como os controles, foi dissolvida em DMSO para os bioensaios. As espécies alvo usadas foram: alface, agrião, tomate e cebola. Cada placa era composta de 20 sementes e foi adicionado 1 mL de solução teste com 4 repetições. As placas foram incubadas a 25 ºC no escuro. Posteriormente, as plântulas tiveram suas partes aéreas e raízes medidas e a porcentagem de germinação e inibição calculada para cada extrato. Tomate foi a espécie que mostrou maior sensibilidade para todos os extratos, seguido de agrião, cebola e alface. Os extratos que tiveram maior atividade foram o acetato de etila, acetona e as extrações líquido-líquido, indicando as frações que devem conter os princípios ativos da folha dessa espécie.
Solanum lycocarpum A.St.-Hil (Solanaceae) is a typical shrub in the Cerrado of central Brazil. The allelopathic activity of aqueous extracts of the leaves and fruits of this species has already been proven in previous studies. The goal of this work was to verify the allelopathic activity of different leaf extracts of S. lycocarpum on the germination and growth of four target species. The leaves were collected, dried, triturated and submitted to two distinct methods of extraction: 1- liquid-liquid (ethyl acetate and dichloromethane) from the aqueous extract and 2- with solvents of increasing polarities (hexane, dichloromethane, ethyl acetate, acetone, methanol and water) directly from the leaves. Each extraction was made with ultrasound equipment for one hour, filtered and evaporated. From these extracts, solutions of 800, 400 and 200 ppm were prepared, and water and Logran® were used as positive and negative controls, respectively. Each solution, as well as the controls, was dissolved in DMSO for the bioassays. The target species used were lettuce, watercress, tomato and onion. To each plate, 20 seeds were added and 1 mL of the tested solutions (with 4 repetitions). The plates were incubated at 25 ºC without light, and the shoots and roots of the seedlings were then measured and the percentage of germination and the inhibition of each extract were calculated. Tomato was the most sensitive to the extracts, followed by watercress, onion and lettuce. The extracts with stronger activity were AcOEt, acetone and the liquid-liquid extraction, indicating the fractions that may contain the active principles of the leaves in this species.

Cloning and endogenous expression of a Eucalyptus grandis UDP-glucose dehydrogenase cDNA

UDP-glucose dehydrogenase (UGDH) catalyzes the oxidation of UDP-glucose (UDP-Glc) to UDP-glucuronate (UDP-GlcA), a key sugar nucleotide involved in the biosynthesis of plant cell wall polysaccharides. A full-length cDNA fragment coding for UGDH was cloned from the cambial region of 6-month-old E. grandis saplings by RT-PCR. The 1443-bp-ORF encodes a protein of 480 amino acids with a predicted molecular weight of 53 kDa. The recombinant protein expressed in Escherichia coli catalyzed the conversion of UDP-Glc to UDP-GlcA, confirming that the cloned cDNA encodes UGDH. The deduced amino acid sequence of the cDNA showed a high degree of identity with UGDH from several plant species. The Southern blot assay indicated that more than one copy of UGDH is present in Eucalyptus. These results were also confirmed by the proteomic analysis of the cambial region of 3- and 22-year-old E. grandis trees by 2-DE and LC-MS/MS, showing that at least two isoforms are present. The cloned gene is mainly expressed in roots, stem and bark of 6-month-old saplings, with a lower expression in leaves. High expression levels were also observed in the cambial region of 3- and 22-year-old trees. The results described in this paper provide a further view of the hemicellulose biosynthesis during wood formation in E. grandis.
