Novel isatinspirooxazine derivatives were designed and synthesized as potential anti-proliferative agents. The new compounds were obtained from aldol condensation reactions between isatin and 3-(hydroxyimino)butan-2-one in the presence of an organic base in order to generate an aldol adduct, followed by cyclization in trifluoroacetic acid, providing the desired isatinspirooxazines in 30 to 80% yield. All the synthesized compounds, including the starting material and the synthetic intermediates, were tested for in vitro anti-proliferative activity against cell lines MCF-7 and MDA-MB231 (breast cancer) and A549 (lung cancer), highlighting the compound 4-methyl,5'-methyl-spiro[(5-aza-4-eno-3-one-cyclohexane)-1,3'-(1H-indol-one)] with an IC50 (half maximal inhibitory concentration) = 0.34 µM, more potent than the reference drug, doxorubicin (IC50 = 1.88 µM), in breast cancer line MDA-MB231.

O estudo fitoquímico das flores e frutos verdes de Senna spectabilis forneceu um novo alcaloide 2,6-dialquilpiperidin-3-ol chamado de (-)-7-hidróxi-cassina, juntamente com cinco alcaloides conhecidos: (-)-cassina, (-)-espectalina, (-)-3-O-acetil-espectalina, (-)-7-hidróxi-espectalina e (-)-6-iso-espectalina. Além dos alcaloides, também foram isolados constituintes químicos de outras classes como o esteroide β-sitosterol, os flavonoides luteolina e 3-metóxi-luteolina, o triterpeno ácido betulínico e o ácido trans-cinâmico. A partir do levantamento bibliográfico foi possível observar que os compostos estão sendo relatados pela primeira vez nesta espécie.

The phytochemical study of flowers and green fruits of Senna spectabilis furnished a new substituted 2,6-dialkylpiperidin-3-ol alkaloid, named (-)-7-hydroxycassine, along with five known piperidine alkaloids: (-)-cassine, (-)-spectaline, (-)-3-O-acetylspectaline, (-)-7-hydroxyspectaline and (-)-iso-6-spectaline. In addition to non-alkaloidal, chemical constituents from other chemical classes were also identified, including the steroid β-sitosterol, the flavonoids luteolin and 3-methoxyluteolin, the triterpene betulinic acid and trans-cinnamic acid. To our knowledge, compounds are being reported for the first time in this species.

Ethanol extracts from six selected species from the Cerrado of the Central-Western region of Brazil, which are used in traditional medicine for the treatment of infectious diseases and other medical conditions, namely Erythroxylum suberosum St. Hil. (Erythroxylaceae), Hyptis crenata Pohl. ex Benth. (Lamiaceae), Roupala brasiliensis Klotz. (Proteaceae), Simarouba versicolor St. Hil. (Simaroubaceae), Guazuma ulmifolia Lam. (Sterculiaceae) and Protium heptaphyllum (Aubl.) March. (Burseraceae), as well as fractions resulting from partition of these crude extracts, were screened in vitro for their antifungal and antibacterial properties. The antimicrobial activities were assessed by the broth microdilution assay against six control fungal strains, Candida albicans, C. glabrata, C. krusei, C. parapsilosis, C. tropicalis and Cryptococcus neoformans, and five control Gram-positive and negative bacterial strains, Escherichia coli, Enterococcus faecalis, Klebsiella pneumoniae, Pseudomonas aeruginosa and Staphylococcus aureus. Toxicity of the extracts and fractions against Artemia salina was also evaluated in this work. All plants investigated showed antimicrobial properties against at least one microorganism and two species were also significantly toxic to brine shrimp larvae. The results tend to support the traditional use of these plants for the treatment of respiratory and gastrointestinal disorders and/or skin diseases, opening the possibility of finding new antimicrobial agents from these natural sources.Among the species investigated, Hyptis crenata, Erythroxylum suberosum and Roupala brasiliensis were considered the most promising candidates for developing of future bioactivity-guided phytochemical investigations.

The resin obtained from species of the genus Protium (Burseraceae) is formed mostly of pentacyclic triterpenes: α-amyrin and β-amyrin. The separation and purification of these triterpenes were optimized in three steps.

Four eudesmane-type sesquiterpenes, costic acid (1), 12-carboxyeudesman-3,11(13)-diene (2), viscic acid (3), 3-oxo-γ-costic acid (4) and two rearranged eudesmane derivatives, 3α-hydroxyisoiphion-11(13)-en-12-oic acid (5) and 5β-hydroxy-4-oxo-11(13)-dehydroiphionan-12-oic acid (6), in addition to (-)-epicatechin, have been isolated from the trunk bark of Nectandra cissiflora. This is the first reported occurrence in the Lauraceae of 3-6. The structures of the isolated compounds have been established on the basis of 1D and 2D NMR spectroscopic techniques. The 13C NMR assignments of 3, 5 and 6 are given here for the first time, as well as some corrections to the previously reported chemical shift assignments of 4.

Dois novos e incomuns derivados de epicatequina e epiafzelequina contendo unidades feniloctanóides glicosiladas e denominados mascagninas A e B foram isolados, juntamente com quercetina-3-O-±-L-ramnopiranosil-(1→6)-²-D-glucopiranosídeo, das partes aéreas de Mascagnia pubiflora(Malpighiaceae), uma planta tóxica para o gado. Suas estruturas foram determinadas pela combinação de espectroscopia de ressonância magnética nuclear (RMN) uni e bidimensionais e dados de espectroscopia de massas (MS).

Mascagnins A and B, two novel and unusual epicatechin and epiafzelechin derivatives bearing glucosylated phenyloctanoid units, along with quercetin-3-O-±-L-rhamnopyranosyl-(1→6)-²-Dglucopyranoside were isolated from the aerial parts of Mascagnia pubiflora(Malpighiaceae), a plant toxic to cattle. Their structures were established by a combination of 1D- and 2D-nuclear magnetic resonance spectroscopic techniques and mass spectrometry data.

From the trunk bark of Nectandra megapotamica (Lauraceae) four phenylpropanoids, elemicin, isoelemicin, (±)-erythro-1-(3,4,5-trimethoxyphenyl)-1,2-propanediol and (±)-threo-1-(3,4,5-trimetoxyphenyl)-1,2-propanediol have been isolated, in addition to 3,4,5-trimethoxybenzoic acid, (-)-epicatechin and trans-1(10)-epoxy-4(15)-caryophyllene. The diastereoisomeric erythro- and threo- phenylpropanoids are being reported for the first time in a plant taxon as well as the occurrence of the other compounds in Nectandra. The structures of the isolated compounds have been established on the basis of 1D and 2D NMR spectroscopic techniques. Their in vitro antifungal activities against standard strains of Candida albicans, C. krusei, C. tropicalis and Cryptococcus neoformans and antioxidant properties were also evaluated in this work.

Chromatographic fractionation of the organic extract from leaves of Ouratea multiflora afforded the flavone dimers heveaflavone, amentoflavone-7<FONT FACE=Symbol>¢¢</FONT>,4<FONT FACE=Symbol>¢¢¢</FONT>-dimethyl eter, podocarpusflavone-A and amentoflavone. Their structures were elucidated from spectral data, including 2D-NMR experiments of the natural substances. Biological activities of all isolates were evaluated, using antimicrobial assay against Gram-positive bacteria Staphylococcus aureus and Bacillus subtilis, cytotoxicity assay against mouse lymphoma (L5178) and KB cell lines, TLC screening for acetylcholinesterase inhibitors and antioxidant activity measured by DPPH test.

O fracionamento cromatográfico do extrato orgânico das folhas de Ouratea multiflora forneceu os flavonóides diméricos, heveaflavona, 7<FONT FACE=Symbol>¢¢</FONT>,4<FONT FACE=Symbol>¢¢¢</FONT>-dimetilamentoflavona, podocarpusflavona-A e amentoflavona. Suas estruturas foram elucidadas com base nos dados espectrais, incluindo experimentos bidimensionais de RMN, das substâncias naturais. A atividade antibiótica de todos os isolados foi avaliada, usando-se as bacterias Gram-positivas Staphylococcus aureus and Bacillus subtilis. Teste de citotoxicidade nas linhagens de linfoma de ratos (L5178) e KB também foram conduzidos para avaliar os extratos e os flavonóides isolados. a triagem biológica para a avaliação de atividade antioxidante e inibidora de acetil colinesterase foram conduzidas pela técnica da bioautografia com DPPH e teste pelo teste de Ellman respectivamente.

The present communication reports the isolation and identification of four triterpenoid saponins from the chloroform extract of the leaves of Tocoyena brasiliensis: 3-O-beta-D-quinovopyranosyl quinovic acid, 3-O-beta-D-quinovopyranosyl cincholic acid, 3-O-beta-D-glucopyranosyl quinovic acid and the 28-O-beta-D-glucopyranosyl ester derivative of quinovic acid as binary mixtures, respectively. From the ethanol extract a flavonoid identified as ramnazin-3-O-rutinoside was obtained. The structures of these compounds were assigned by data analysis of 1D and 2D NMR spectrometry and comparison with data recorded in the literature for these compounds.

Phytochemical studies with leaves of Uncaria guianensis resulted in the isolation of the oxindole alkaloids isomitraphylline (1), 3-isoajmalicine (2) mitraphylline (3), and isomitraphylinic acid (4). Structural assignments of these alkaloids, including relative configurations and conformations, were performed through spectral data and physical properties. 1D and 2D homonuclear and heteronuclear NMR spectroscopy was a valuable tool for the establishment of the relative stereochemistry of those compounds.
