ABSTRACT In Experiment one, a total of 60 blue peacocks were randomly assigned into the control group (12.00 MJ/kg) and treatment group (12.30 MJ/kg). Each group consisted of three repeats and each replicate contained 10 peacocks (2 males and 8 females). In Experiment two, based on better metabolic energy (ME) (12.30 MJ/kg), 90 blue peacocks were randomly assigned to three dietary treatments with 18.00%, 19.00% and 20.00% crude protein (CP) levels. The results of Experiment one revealed that the different dietary ME levels had minor effect on productive performance of peacocks (p>0.05). Decreasing dietary ME levels decreased the concentration of estradiol (E2) and increased the concentrations of follicle stimulating hormone (FSH) and luteinizing hormone (LH) (p<0.01). However, 12.00 MJ/kg ME stimulated creatinine levels of blue peacock, which exceeded normal values. Based on this result, blue peacocks fed with 12.30 MJ/kg ME had better performance. Results from Experiment two indicated that egg production of blue peacocks was significantly improved by the 20.00% CP diet. The concentrations of FSH, LH, and E2 remained at high levels fed with the 20.00% CP diet. Changes in serum biochemical indices such as total cholesterol, creatinine and globulin of peacocks supplied with 18.00% and 19.00% CP diets were abnormal (p<0.05). ABSTRACT To improve the flow field structure of the air distribution chamber of the microwave–hot-air coupling dryer for a straw-based nutrient seeding-growing bowl tray and enhance the uniformity of the outlet gas velocity distribution, computational fluid dynamics software was used to simulate the flow field of the air distribution chamber based on the gas motion differential equation and the (RNG) k–ε turbulence model. Using the front panel height, upper apex angle, and side wall inclination angle as factors and the speed nonuniformity coefficient as the index, the results showed that when the air distribution chamber front panel height was 33 mm, the upper apex angle was 108°, the side wall inclination angle was 78°, the height of the two spoilers was 40 mm, the outlet airflow speed varied from 5.36 to 5.71 m/s, and the speed unevenness coefficient was reduced from 13.95% in the original model to 6.05%. The maximum deviation between the numerical simulation results and test data was less than 4%, which met the uniformity requirement of the outlet airflow speed of the dryer. The results provide theoretical support for the design and industrial production of microwave–hot-air coupling dryers.

Abstract Objective: To investigate the impact of recombinant human interferon α1b (rhIFNα1b) treatment in infants hospitalized with lower respiratory tract infections on subsequent wheezing. Methods: The clinical data of infants (n = 540) with viral pneumonia, wheezy bronchitis, or bronchiolitis hospitalized in 19 Chinese hospitals from June 2009 to June 2015 were retrospectively analyzed. The parameters relevant to wheezing episodes within the last year were collected by telephone and questionnaires. The rhIFNα1b treatment group (n = 253) and control group (n = 287) were compared in terms of wheezing episodes within the last year. Moreover, the wheezing group (95 cases) and non-wheezing group (445 cases) were compared. Results: Out of 540 cases, 95 (17.6%) experienced wheezing episodes, 13.8% (35/253) cases treated with rhIFNα1b, and 20.9% (60/287) cases without rhIFNα1b experienced wheezing episodes within the last year. The rhIFNα1b treatment significantly improved wheezing episodes within the last year, compared with the control peers (p = 0.031). Single-factor regression showed statistically significant differences between the wheezing and non-wheezing groups in terms of age, rhIFNα1b use, childhood and family history of allergy, housing situation, and feeding history (p < 0.05). Binary logistic regression showed a childhood history of allergy (OR = 2.14, p = 0.004), no rhIFNα1b use (OR = 1.70, p = 0.028), and living in a crowded house (OR = 1.92, p = 0.012) might be risk factors of subsequent wheezing. Accordingly, breastfeeding (OR = 0.44, p = 0.008) and hospitalization age of 1-year-old (OR = 0.58, p = 0.024) were protective factors. Conclusions: Early use of rhIFNα1b in infants hospitalized with lower respiratory tract infections and breastfeeding could prevent subsequent wheezing. Living in a crowded house could promote subsequent wheezing.

Resumo A autofagia desempenha um papel importante na manutenção da homeostase celular através da degradação de proteínas desnaturadas e outras macromoléculas biológicas. Nos últimos anos, muitos pesquisadores se concentraram no mecanismo da autofagia em parasitas apicomplexos, mas pouco se sabe sobre esse processo na coccidia aviária. No presente estudo, a clonagem, sequenciamento e caracterização de gene relacionado à autofagia Etatg8 foram investigados pela PCR quantitativa em tempo real (RT-qPCR), mancha ocidental (WB), ensaios indiretos de imunofluorescência (IFAs) e microscopia eletrônica de transmissão (TEM), respectivamente. Os resultados mostraram que o gene Etatg8 de E. tenella possui uma ORF de 375 bp, codificando uma proteína de 124 aminoácidos com estrutura e propriedades semelhantes à de outros apicomplexos. RT-qPCR revelou que Etatg8 é expresso durante os quatro estágios de desenvolvimento de E. tenella. Entretanto, seus níveis transcricionais foram significativamente mais elevados na fase de oocisto não esporulados. Os ensaios de manchas ocidental (WB) e de imunofluorescência (IFA) mostraram que a proteína EtATG8 foi lipidada para ligar-se à membrana do autofagossomo sob condições de deficiência nutritiva (em presença de rapamicina) e se agregar no citoplasma de esporozoítas e merozoítas. No entanto, o processo de produção de membrana do autofagossomo pode ser inibido por um inibidor de autofagia (3-meetiladeninatiladenina, 3-MA). Em conclusão, foi demonstrado que E. tenella tem um mecanismo de autofagia conservado, semelhante ao de outros parasitas apicomplexos, e que EtATG8 pode ser usado como um marcador para futuras pesquisas sobre autofagia direcionada à coccidiose aviária.

Abstract Autophagy plays an important role in maintaining cell homeostasis through degradation of denatured proteins and other biological macromolecules. In recent years, many researchers focus on mechanism of autophagy in apicomplexan parasites, but little was known about this process in avian coccidia. In our present study. The cloning, sequencing and characterization of autophagy-related gene (Etatg8) were investigated by quantitative real-time PCR (RT-qPCR), western blotting (WB), indirect immunofluorescence assays (IFAs) and transmission electron microscopy (TEM), respectively. The results have shown 375-bp ORF of Etatg8, encoding a protein of 124 amino acids in E. tenella, the protein structure and properties are similar to other apicomplexan parasites. RT-qPCR revealed Etatg8 gene expression during four developmental stages in E. tenella, but their transcriptional levels were significantly higher at the unsporulated oocysts stage. WB and IFA showed that EtATG8 was lipidated to bind the autophagosome membrane under starvation or rapamycin conditions, and aggregated in the cytoplasm of sporozoites and merozoites, however, the process of autophagosome membrane production can be inhibited by 3-methyladenine. In conclusion, we found that E. tenella has a conserved autophagy mechanism like other apicomplexan parasites, and EtATG8 can be used as a marker for future research on autophagy targeting avian coccidia.

ABSTRACT Objective To evaluate the effects of Arf6 downregulation on human prostate cancer cells. Materials and Methods The effects of Arf6 downregulation on cell proliferation, migration, invasion and apoptosis were assessed by MTT, BrdU, scratch, Transwell assays and flow cytometry respectively. AKT, p-AKT, ERK1/2, p-ERK1/2 and Rac1 protein expressions were detected by Western blot. Results Downregulating Arf6 by siRNA interference suppressed the mRNA and protein expressions of Arf6. The proliferation capacities of siRNA group at 48h, 72h, and 96h were significantly lower than those of control group (P <0.05). The migration distance of siRNA group at 18h was significantly shorter than that of control group (P <0.01). The number of cells penetrating Transwell chamber membrane significantly decreased in siRNA group compared with that of control group (P <0.01). After 24h, negative control and normal control groups had similar apoptotic rates (P >0.05) which were both significantly lower than that of siRNA group (P <0.01). After Arf6 expression was downregulated, p-ERK1/2 and Rac1 protein expressions were significantly lower than those of control group (P <0.05). Conclusion Downregulating Arf6 expression can inhibit the proliferation, migration and invasion of prostate cancer cells in vitro, which may be related to ERK1/2 phosphorylation and Rac1 downregulation.

ABSTRACT Recently the use of antibiotic growth promoter (AGPs) in poultry feed is banned in China, leading to the increasing demand for AGPs substitutes. Probiotics have been considered as a potential substitute. The purpose of this study was to evaluate the effects of probiotic on serum biochemistry, and growth promotion of body weight and visceral organs and in Lohmann Brown chicks aged 0 to 9 weeks. Five hundred and forty 1-day-old male chicks were randomly divided into three groups, with six replicates and 30 chicks in each replicate. The experiment was carried out over 70 days. Dietary treatments were: (1) basal diet alone (control group); (2) basal diet containing 0.1% probiotics (probiotic group); and (3) basal diet containing 0.01% zinc bacitracin in the feed (antibiotic group). The body weight (BW) of probiotic group was significantly higher than that of the control group during the whole trial period (p<0.05). ADG (0~70 d) in the probiotic group and the antibiotic group was higher than that in the control group (p<0.05). The Oliver weight and serum total protein (TP) in the probiotic group was higher than that in the control group on the 63rd day (p<0.05). The length of the duodenum, the weight of the spleen, duodenum and Jejunum in both the probiotic and antibiotic groups were higher than those in the control group (p<0.05). The ileum weight in the probiotic group was significantly higher than those in the control group (p<0.05). The ileum length in the probiotic group was significantly longer than that in both the antibiotic and control groups (p<0.05). Probiotic mix with Bacillus subtilis and Pediococcus acidilactici play a similar role in promoting growth of body and visceral organs, and it is a promising growth promoter for Lohmann Brown chicks (0 to 9 weeks).

ABSTRACT Soil microbial biomass plays a significant role in soils, and it is often used as an early indicator of change in soil quality. Soil microbial biomass is affected by different fertilization management practices. Therefore, the impact of different long-term fertilization management practices on the soil organic carbon (SOC) content, soil microbial biomass carbon (SMBC), and soil microbial biomass nitrogen (SMBN), as well as the soil microbial quotient (SMQ) in the tilled layer (0.00-0.20 m) were studied in the present paper, together with grain yield, in a double-cropping rice (Oryza sativa L.) system. The experiment in NingXiang county of Hunan Province, China, begin in 1986, and the experiment included five fertilization treatments: without fertilizer input (CK), mineral fertilizer alone (MF), rice straw residues and mineral fertilizer (RF), 30 % organic matter and 70% mineral fertilizer (LOM), and 60% organic matter and 40 % mineral fertilizer (HOM). The results showed that there is no significant difference in effect on SOC, SMBC, and SMBN contents and on the SMQ in the paddy field with MF treatment compared with the CK treatment at the main growth stages of early and late rice. The SOC, SMBC, SMBN contents, and the SMQ in the paddy field were highest in the LOM and HOM treatments, followed by the RF treatment, at the main growth stages of early and late rice. The results indicated that grain yields of early and late rice with the LOM, HOM, and RF treatments were higher than the yields under the MF and CK treatments. As a result, combined application of organic matter or rice straw residues with mineral fertilizer is a practice available for increasing SOC and microbial biomass contents in double-cropping rice paddy soils.

ABSTRACT Residue management in cropping systems may improve soil quality. However, there are few studies on the effects of residue management on soil aggregation and carbon content in the humin (C-HUM), humic acid (C-HAF) and fulvic acid (C-FAF) fractions in South China. Therefore, the effects on soil aggregation and on the C-HUM, C-HAF, C-FAF from incorporating winter cover crop residues in a double-cropped rice (Oryza sativa L.) system in South China fields were studied. The experiment has been conducted since winter 2004. Five winter cropping systems were used: rice-rice-ryegrass (Ry-R-R), rice-rice-Chinese milk vetch (Mv-R-R), rice-rice-potato (Po-R-R), rice-rice-rape (Ra-R-R) and rice-rice with winter fallow (Fa-R-R). The results indicated that the organic C content in the paddy soil under the Ry-R-R, Mv-R-R, Po-R-R, and Ra-R-R systems was significantly higher than the content in the Fa-R-R system at the early rice and late rice maturity stages. The different sizes of aggregates under the five treatments showed similar trends. The Po-R-R systems had the highest percentage of soil aggregates in each size class and the Fa-R-R systems had the lowest percentage of soil aggregates in each size class in the 0.00-0.05, 0.05-0.10, and 0.10-0.20 m soil depth at the early rice and late rice maturity stages. The C-HUM, C-HAF, and C-FAF increased through long-term application of winter cover crop residues. Statistical analysis showed that the C-HAF under the Ra-R-R systems was significantly higher than that in the Fa-R-R systems at the early rice and late rice maturity stages. The C-FAF and C-HUM under the Mv-R-R systems was significantly higher than the C-FAF and C-HUM in the Fa-R-R systems at the early rice and late rice maturity stages. As a result, the soil organic C content, the soil aggregates in each size class, and the C-HUM, C-HAF, and C-FAF increased from application of winter cover crop residues in double-cropped rice systems.

Y chromosomal STRs show sufficient variability among individduals in a population and a high degree of geographical differentiation, such that their polymorphic character makes them especially suited for population genetic studies. To investígate the polymorphism of a set of 17 Y-STR loci in northern China, we genotyped the 17 Y chromosomal STR loci in a population sample of 377 unrelated males from eight ethnic populations in northern China. We calculated the haplotype frequencies, Rst value and carried out the analysis of molecular variance (AMOVA). We then drew the multidimensional scaling analysis (MDS) plot and phylogenetic tree based on the Rst value. All populations showed a high level of haplotype diversity, with low inter-population variance as measured by an analysis of molecular variance. However, the genetic distances were significant when the eight populations were compared to other populations. By MDS and the phylogenetic tree, we found that the eight populations had a close relationship and Xibo had a northeast origination.

Todas as nucleobases são regiosseletivamente alquiladas e os produtos desejados são obtidos com rendimentos moderados a altos, sob irradiação de microondas por 8 minutos em água como solvente e na presença de Et3N como base.

A facile and eco-friendly approach has been developed for the preparation of N-(ethyoxycarbonylmethyl) nucleobases and N-(iso-propoxycarbonylmethyl) nucleobases, which are important building blocks for Peptide Nucleic Acids (PNA). All the nucleobases are regioselectively alkylated and the desired products are obtained in moderate to high yields under microwave irradiation for 8 min in water as the solvent and in the presence of Et3N as the base.

OBJECTIVE: To determine the validity of a recently developed rapid test - a colloidal dye immunofiltration assay (CDIFA) - used by health workers in field settings to identify villagers infected with Schistosoma japonicum. METHODS: Health workers in the field used CDIFA to test samples from 1553 villagers in two areas of low endemicity and an area where S. japonicum was not endemic in Anhui, China. All the samples were then tested in the laboratory by laboratory staff using a standard parasitological method (Kato-Katz), an indirect haemagglutination assay (IHA), and CDIFA. The results of CDIFA performed by health workers were compared with those obtained by Kato-Katz and IHA. FINDINGS: Concordance between the results of CDIFA performed in field settings and in the laboratory was high (kappa index, 0.95; 95% confidence interval, 0.93-0.97). When Kato-Katz was used as the reference test, the overall sensitivity and specificity of CDIFA were 98.5% and 83.6%, respectively in the two villages in areas of low endemicity, while the specificity was 99.8% in the non-endemic village. Compared with IHA, the overall specificity and sensitivity of CDIFA were greater than 99% and 96%, respectively. With the combination of Kato-Katz and IHA as the reference standard, CDIFA had a sensitivity of 95.8% and a specificity of 99.5%, and an accuracy of 98.6% in the two areas of low endemicity. CONCLUSION: CDIFA is a specific, sensitive, and reliable test that can be used for rapid screening for schistosomiasis by health workers in field settings.

OBJETIVO: Determinar la validez de una prueba rápida desarrollada recientemente -prueba de inmunofiltración con colorante coloidal (PICC)- y empleada por agentes de salud sobre el terreno para identificar a los lugareños infectados por Schistosoma japonicum. MÉTODOS: Agentes de salud sobre el terreno utilizaron la PICC para analizar muestras de 1553 lugareños en dos zonas de baja endemicidad y una zona sin S. japonicum endémica de Anhui, China. Todas las muestras fueron analizadas en el laboratorio por personal técnico utilizando un método parasitológico estándar (Kato-Katz), una prueba de hemaglutinación indirecta (IHA) y la PICC. Los resultados de la PICC llevada a cabo por los agentes de salud fueron comparados con los obtenidos mediante las técnicas de Kato-Katz e IHA. RESULTADOS: Se observó una elevada concordancia entre los resultados de la PICC aplicada sobre el terreno y la practicada en el laboratorio (índice kappa = 0,95; intervalo de confianza del 95% = 0,93-0,97). Cuando se utilizó el método Kato-Katz como prueba de referencia, la sensibilidad y la especificidad generales de la PICC fueron de un 98,5% y un 83,6%, respectivamente, en las dos aldeas de las zonas de endemicidad baja, mientras que la especificidad fue del 99,8% en la aldea sin endemicidad. En comparación con la IHA, la especificidad y la sensibilidad generales de la PICC superaron el 99% y el 96%, respectivamente. Utilizando la combinación de Kato-Katz e IHA como referencia, la PICC presentó una sensibilidad del 95,8% y una especificidad del 99,5%, con una precisión del 98,6%, en las dos zonas de endemicidad baja. OBJECTIF: Déterminer la validité d'un test diagnostique rapide (CDIFA) récemment mis au point, faisant appel à un colorant colloïdal et à la technique d'immunofiltration et destiné au dépistage sur le terrain par des agents de santé de Schistosoma japonicum chez les villageois. MÉTHODES: Des agents de santé présents sur le terrain ont appliqué le test CDIFA à des prélèvements effectués sur 1553 villageois provenant de deux zones de faible endémicité de la schistosomiase sino-japonaise et d'une zone où cette maladie n'est pas endémique, appartenant à la province d'Anhui, en Chine. Tous ces prélèvements ont ensuite été analysés en laboratoire par du personnel spécialisé utilisant une technique de diagnostic parasitologique classique (technique de Kato), la technique d'hémagglutination indirecte (IHA) et le test CDIFA. Les résultats des tests CDIFA effectués par les agents de santé ont été comparés à ceux obtenus par la technique de Kato et par hémagglutination indirecte. RÉSULTATS: Le taux de concordance entre les résultats des tests CDIFA effectués sur le terrain et en laboratoire est élevé (indice kappa : 0,95, intervalle de confiance à 95 % : 0,93- 0,97). Si l'on utilise la technique Kato comme référence, on parvient à une sensibilité et à une spécificité globales pour le test CDIFA de 98,5 % et de 83,6 % respectivement dans les deux villages situés en zones de faible endémicité, ainsi qu'à une spécificité de 99,8 % dans le village exempt de schistosomiase sino-japonaise à l'état endémique. Par comparaison avec la technique d'hémagglutination indirecte (IHA), la spécificité et la sensibilité globales du test CDIFA sont respectivement supérieures à 99 et 96 %. Si l'on se réfère à une combinaison de la technique KATO et de la technique d'hémagglutination indirecte, on obtient pour le test CDIFA une sensibilité de 95,8 % et une spécificité de 99,5 %, ainsi qu'une précision de 98,6 % dans les deux zones d'endémicité. CONCLUSION: Le test CDIFA offre une méthode spécifique, sensible et fiable permettant le dépistage rapide sur le terrain de la schistosomiase par les agents de santé.
