Resumo O artigo objetiva analisar a percepção de participantes sobre o efeito de intervenções desenvolvidas no âmbito de um projeto que integra o Programa de Apoio ao Desenvolvimento Institucional do Sistema Único de Saúde (PROADI-SUS). Adotou-se o desenho metodológico de avaliação normativa, na perspectiva da abordagem de pesquisa qualitativa. Na pesquisa, foram envolvidos 21 profissionais de saúde de cinco projetos de intervenção, representando as cinco regiões brasileiras. As mudanças percebidas e tangibilizadas foram constatadas como resultado das intervenções. Dentre as conclusões, destaca-se que houve convergência entre percepções e intencionalidades do projeto avaliado.

Abstract The article aims to analyze the participants’ perception of the effect of interventions developed within a project nested in the Support Program for Institutional Development of the Unified Health System (PROADI-SUS). We adopted normative assessment from the perspective of a qualitative research approach. Twenty-one health professionals from five intervention projects representing the five Brazilian regions were involved in the research. The changes perceived and made tangible were identified as a result of the interventions. Among the conclusions, we underscore a convergence between perceptions and intentions of the evaluated project.

Abstract The number of antral follicles is considered an important fertility trait because animals with a high follicle count (HFC) produce more oocytes and embryos per cycle. Identification of these animals by genetic markers such as single nucleotide polymorphisms (SNPs) can accelerate selection of future generations. The aim of this study was to perform a genome wide association study (GWAS) on Nelore and Angus heifers with HFC and low (LFC) antral follicle counts. The groups HFC and LFC for genotyping were formed based on the average of total follicles (≥ 3 mm) counted in each breed consistently ± standard deviation. A total of 72 Nelore heifers (32 HFC and 40 LFC) and 48 Angus heifers (21 HFC and 27 LFC) were selected and the DNA was extracted from blood and hair bulb. Genotyping was done using the Illumina Bovine HD 770K BeadChip. The GWAS analysis showed 181 and 201 SNPs with genotype/phenotype association (P ≤ 0.01) in Nelore and Angus heifers, respectively. Functional enrichment analysis was performed on candidate genes that were associated with SNPs. A total of 97 genes were associated to the 181 SNPs in the Nelore heifers and the functional analysis identified genes (ROBO1 and SLIT3) in the ROBO-SLIT pathway that can be involved in the control of germ cell migration in the ovary as it is involved in lutheal cell migration and fetal ovary development. In the Angus heifers, 57 genes were associated with the 201 SNPs, highlighting Fribilin 1 (FBN1) gene, involved in regulation of growth factors directly involved in follicle activation and development. In summary, GWAS for Nelore and Angus heifers showed SNPs associated with higher follicle count phenotype. Furthermore, these findings offer valuable insights for the further investigation of potential mechanism involved in follicle formation and development, important for breeding programs for both breeds. Abstract This study aimed to evaluate the impact of Meloidogyne javanica, Meloidogyne incognita, and Meloidogyne arenaria on different aspects of the development of sugarcane plants under greenhouse conditions. For this purpose, seedlings of the RB867515 genotype were individually inoculated with 5,000 eggs + second-stage juveniles of their respective nematodes/plant, and non-inoculated plants were used as control. After 330 days of inoculation, the plants were removed from the pots, and the following characteristics were evaluated: fresh mass of the aerial part and root system; leaf area; leaf chlorophyll index; culm diameter; fresh mass of culms; broth volume; contents of neutral and acid detergent fiber, cellulose, hemicellulose, lignin, apparent sucrose in broth, and reducing sugars in broth; total soluble solids concentration. Subsequently, the final population of nematodes in the root system of inoculated plants was determined to calculate the reproduction factor of nematodes. The results showed that all tested Meloidogyne species negatively affected plant development and the composition of some analyzed fractions, in comparison to the non-inoculated control. Resumo: Este estudo investiga a dominância bacteriana em uma das mais estudadas lagoas costeiras eutróficas contaminadas por esgoto, a Lagoa Imboassica em Macaé (RJ), Brasil, utilizando sequenciamento massivo do gene codificador do RNAr 16S. Foram coletadas amostras de água de três locais na lagoa. O DNA microbiano total foi extraído e a região V3-V4 do gene RNAr 16S foi amplificada e sequenciada na plataforma Illumina MiSeq. Um total de 744.879 sequências parciais do gene RNAr 16S foram agrupadas, revelando a ausência de uma dominância bacteriana única na lagoa costeira eutrófica influenciada por esgoto. Os filos predominantes detectados na lagoa foram Cyanobacteria (27,8%), Proteobacteria (23,7%) e Actinobacteria (14,6%). Proteobacteria emergiu como o filo mais abundante no local da lagoa impactado pelo esgoto, enquanto Cyanobacteria dominou os outros dois locais de amostragem. A família de maior prevalência encontrada foi a Synechococcaceae, sendo representada pelo gênero Synechococcus. Famílias do filo Cyanobacteria consideradas potencialmente tóxicas representaram menos de 1% do total de famílias. A região da lagoa impactada pelo esgoto exibiu maior diversidade e riqueza bacteriana. A dominância de comunidades bacterianas associadas ao esgoto bruto, como membros da família Enterobacteriaceae, não foi confirmada, a qual representou somente 0,75% das famílias no local mais afetado. Este estudo apresenta a análise inicial da comunidade bacteriana na Lagoa Imboassica e sugere que a dominância responde a eutrofização e descarga de esgoto. Abstract: This study investigates the presence of bacterial dominance in one of the most studied sewage-driven eutrophic coastal lagoons, the Imboassica Lagoon in Macaé (RJ), Brazil, utilizing high-throughput sequencing of 16S rDNA. Water samples were collected from three sites within the lagoon. Total microbial DNA was extracted, and the V3-V4 region of the 16S rRNA gene was amplified and sequenced on the Illumina MiSeq platform. A total of 744,879 partial 16S rRNA sequences were clustered, revealing the absence of a single bacterial dominance in the sewage-driven eutrophic coastal lagoon. The prominent phyla detected in the lagoon were Cyanobacteria (27.8%), Proteobacteria (23.7%), and Actinobacteria (14.6%). Proteobacteria emerged as the most abundant phylum in the sewage-impacted lagoon site, whereas Cyanobacteria dominated the other two sampling sites. Among families, Synechococcaceae predominated with genus Synechococcus exhibited the highest prevalence. Families of potentially toxic Cyanobacteria represented less than 1% of the total families. The sewage-impacted lagoon section displayed greater bacterial diversity and richness. The dominance of bacterial communities associated with raw sewage, such as members of the Enterobacteriaceae family, was not confirmed, constituting only 0.75% of the families in the most affected site. This study presents the initial analysis of the bacterial community in the Imboassica Lagoon and suggests that dominance in the lagoon responds to the eutrophication and sewage discharge. Resumo Neste trabalho, foi testado o uso de um polímero superabsorvente (hidrogel) em sistema silvipastoril com jacarandá-da-bahia (Dalbergia nigra (Vell.) Allemão ex Benth.), em área de pastagem degradada. A área experimental faz parte da fazenda experimental Bananal do Norte, Cachoeiro de Itapemirim, Espírito Santo, Brasil. Mais de 480 mudas foram plantadas e submetidas aos tratamentos Hidrogel seco (DRY); Hidrogel hidratado ao redor do torrão (HAR); Hidrogel hidratado misturado com solo (HMS); e apenas água (WAT). O delineamento foi em blocos casualizado, com três repetições. As variáveis testadas foram sobrevivência da planta, altura e diâmetro do coleto, até 672 dias após o plantio, umidade do solo aos 93 dias após o plantio e biomassa total e análise química foliar aos 184 dias após o plantio. A sobrevivência foi positivamente influenciada pela aplicação do hidrogel, mas pouca ou nenhuma diferença foi observada para os métodos de aplicação. O hidrogel e suas técnicas de aplicação não afetaram o crescimento em altura e diâmetro do coleto. O polímero não influenciou o desenvolvimento em biomassa e conteúdo nutricional das folhas, mas influenciou na distribuição de biomassa. Abstract This work tested the use of a superabsorbent polymer (hydrogel) in a silvopastoral system with Brazilian-rosewood (Dalbergia nigra (Vell.) Allemão ex Benth.) in a degraded pasture area. The experimental site is part of the Bananal do Norte experimental farm, Cachoeiro de Itapemirim, Espírito Santo, Brazil. Over 480 seedlings were planted and submitted to the treatments: Dry hydrogel (DRY); Hydrated hydrogel around the root ball (HAR); Hydrated hydrogel mixed with soil (HMS); and water only (WAT). The design was in randomized blocks, with three repetitions. The tested variables were plant survival, height and root collar diameter until 672 days after planting, soil moisture at 93 days after planting and total biomass and leaf chemical analysis at 184 days after planting. Survival was positively influenced by the application of the hydrogel, but there was little or no effect regarding the application method. The hydrogel and its application techniques did not alter growth in height and root collar diameter. The polymer did not influence the development in biomass and nutritional content of the leaves, but did influence the biomass distribution. The use of the superabsorbent polymer was therefore not effective in accelerating plant growth and anticipating the stages in the implantation of the silvopastoral system. Resumo O mero do Atlântico Epinephelus itajara é a maior espécie de garoupa no Oceano Atlântico. Apesar do seu status de conservação Vulnerável (VU) globalmente e Criticamente em Perigo (CR) no Brasil, a espécie continua enfrentando ameaças da sobrepesca em toda a sua área de distribuição. Utilizamos o sequenciamento de DNA por código de barras, empregando o gene mitocondrial da Citocromo c oxidase subunidade I (COI), para identificar a comercialização ilegal de E. itajara em mercados de peixe nas costas norte (CN) e sul (CS) do Brasil. A coleta de amostras foi realizada em mercados de peixe e teve como objetivo confirmar a identificação de peixes descaracterizados e vendidos como E. itajara na CN, bem como identificar filés de peixe vendidos como garoupa verdadeira na CS. O sequenciamento de DNA por código de barras permitiu a identificação inequívoca de 22 (84,6%) das 26 amostras de mercado de peixe analisadas. Ambas as áreas amostradas tiveram confirmação da comercialização ilegal de E. itajara, e das 22 amostras analisadas, 17 (77,3%) foram confirmadas como E. itajara. Nós relatamos aqui dois crimes, a venda ilegal de E. itajara e a fraude comercial por substituição de espécies. Este estudo ressaltou que a legislação existente que protege E. itajara em águas brasileiras requer a adoção de melhores políticas públicas para a conservação da espécie. Abstract The Atlantic goliath grouper Epinephelus itajara is the largest grouper species in the Atlantic Ocean. Despite the conservation status being Vulnerable (VU) globally and Critically Endangered (CR) in Brazil, the species continues to face threats from overfishing throughout its range. We used DNA barcoding employing the mitochondrial Cytochrome c oxidase subunit I (COI) gene to identify the illegal commercialization of E. itajara in fish markets from the northern (NC) and southern (SC) Brazilian coasts. Sampling was conducted in fish markets and aimed to confirm the identification of mischaracterized fish sold as E. itajara in the NC, as well as identifying fish fillets sold as the Dusky grouper in the SC. DNA barcoding allowed the unambiguous identification of 22 (84.6%) of the 26 analyzed fish market samples. Both sampled areas had confirmation of E. itajara illegal commercialization and from the 22 analyzed samples, 17 (77.3%) were confirmed to be E. itajara. Here we report two crimes, the illegal sale of E. itajara and commercial fraud by species substitution. This study has highlighted that the existing legislation that protects E. itajara in Brazilian waters requires the adoption of better public policies for the conservation of the species. BACKGROUND In Brazil, transmission of visceral and cutaneous leishmaniasis has expanded geographically over the last decades, with both clinical forms occurring simultaneously in the same area. OBJECTIVES This study characterised the clinical, spatial, and temporal distribution, and performed entomological surveillance and natural infection analysis of a leishmaniasis-endemic area. METHODS In order to characterise the risk of leishmaniasis transmission in Altos, Piauí, we described the clinical and socio-demographic variables and the spatial and temporal distribution of cases of American visceral leishmaniasis (AVL) and American cutaneous leishmaniasis (ACL) cases and identified potential phlebotomine vectors. FINDINGS The urban area concentrated almost 54% of ACL and 86.8% of AVL cases. The temporal and spatial distribution of AVL and ACL cases in Altos show a reduction in the number of risk areas, but the presence of permanent disease transmission foci is observed especially in the urban area. 3,808 phlebotomine specimens were captured, with Lutzomyia longipalpis as the most frequent species (98.45%). Of the 35 females assessed for natural infection, one specimen of Lu. longipalpis tested positive for the presence of Leishmania infantum and Leishmania braziliensis DNA. MAIN CONCLUSION Our results indicate the presence of risk areas for ACL and AVL in the municipality of Altos and highlight the importance of entomological surveillance to further understand a possible role of Lu. longipalpis in ACL transmission. Abstract The hematological parameters of fish contribute to the understanding of the health status in response to diseases, treatments, handling, and environmental factors. This study aimed to verify the hematological parameters of mullet Mugil liza after one-hour immersion baths in five concentrations of hydrogen peroxide (H2O2), an effective chemical treatment for several diseases in fish. A total of 108 fish (35.25 ± 6.09g) was distributed into six treatments with three replicates each: untreated fish (control), and fish treated with five concentrations of H2O2 (150, 200, 250, 300 and 350 mg L-1). Blood collection was performed immediately after bathing and 30 days after. Erythrogram, leukogram, thrombocytogram, hematocrit, plasma glucose, hemoglobin concentration and hematimetric indexes were analyzed. There was no significant difference in the erythrocyte and total leukocyte counts. Regarding plasma glucose and hematimetric indexes, the values were increased (p<0.05), except for mean corpuscular volume (MCV), that was decreased (p<0.05) after bath, including control. These responses may be associated to stress from handling and bathing procedure. In fish treated with H2O2, the number of thrombocytes and lymphocytes was lower (p<0.05) immediately after bath, while neutrophils and monocytes showed increased (p<0.05) values immediately after bath, which indicates adverse effects due to H2O2; however, these effects seem to be reversible after 30 days of recovery. Resumo Fundamento Dados robustos sobre a curva de aprendizagem (LC) da substituição da válvula aórtica transcateter (TAVR) são escassos nos países em desenvolvimento. Objetivo Avaliar a LC da TAVR no Brasil ao longo do tempo. Métodos Analisamos dados do registro brasileiro de TAVR de 2008 a 2023. Pacientes de cada centro foram numerados cronologicamente em número sequencial de caso (NSC). A LC foi realizada usando um spline cúbico restrito ajustado para o EuroSCORE-II e o uso de próteses de nova geração. Ainda, os desfechos hospitalares foram comparados entre grupos definidos de acordo com o nível de experiência, com base no NSC: 1º ao 40º caso (experiência inicial), 41º ao 80º caso (experiência básica), 81º ao 120º caso (experiência intermediária) e 121º caso em diante (experiência alta). Análises adicionais foram conduzidas de acordo com o número de casos tratados antes de 2014 (>40 e ≤40 procedimentos). O nível de significância adotado foi p <0,05. Resultados Foram incluídos 3194 pacientes de 25 centros. A idade média foi 80,7±8,1 anos e o EuroSCORE II médio foi 7±7,1. A análise da LC demonstrou uma queda na mortalidade hospitalar ajustada após o tratamento de 40 pacientes. Um patamar de nivelamento na curva foi observado após o caso 118. A mortalidade hospitalar entre os grupos foi 8,6%, 7,7%, 5,9%, e 3,7% para experiência inicial, básica, intermediária e alta, respectivamente (p<0,001). A experiência alta foi preditora independente de mortalidade mais baixa (OR 0,57, p=0,013 vs. experiência inicial). Centros com baixo volume de casos antes de 2014 não mostraram uma redução significativa na probabilidade de morte com o ganho de experiência, enquanto centros com alto volume de casos antes de 2014 apresentaram uma melhora contínua após o caso de número 10. Conclusão Observou-se um fenômeno de LC para a mortalidade hospitalar do TAVR no Brasil. Abstract Background Robust data on the learning curve (LC) of transcatheter aortic valve replacement (TAVR) are lacking in developing countries. Objective To assess TAVR’s LC in Brazil over time. Methods We analyzed data from the Brazilian TAVR registry from 2008 to 2023. Patients from each center were numbered chronologically in case sequence numbers (CSNs). LC was performed using restricted cubic splines adjusted for EuroSCORE-II and the use of new-generation prostheses. Also, in-hospital outcomes were compared between groups defined according to the level of experience based on the CSN: 1st to 40th (initial-experience), 41st to 80th (early-experience), 81st to 120th (intermediate-experience), and over 121st (high-experience). Additional analysis was performed grouping hospitals according to the number of cases treated before 2014 (>40 and ≤40 procedures). The level of significance adopted was <0.05. Results A total of 3,194 patients from 25 centers were included. Mean age and EuroSCORE II were 80.7±8.1 years and 7±7.1, respectively. LC analysis demonstrated a drop in adjusted in-hospital mortality after treating 40 patients. A leveling off of the curve was observed after case #118. In-hospital mortality across the groups was 8.6%, 7.7%, 5.9%, and 3.7% for initial-, early-, intermediate-, and high-experience, respectively (p<0.001). High experience independently predicted lower mortality (OR 0.57, p=0.013 vs. initial experience). Low-volume centers before 2014 showed no significant decrease in the likelihood of death with gained experience, whereas high-volume centers had a continuous improvement after case #10. Conclusion A TAVR LC phenomenon was observed for in-hospital mortality in Brazil. Resumo Contaminação por microplásticos (MPs) é um impacto bem estabelecido nos oceanos, embora sejam escassos enfoques combinando análises simultâneas dos componentes bióticos e abióticos. Este estudo foi direcionado para esta lacuna, demonstrando a ingestão de MPs por Atherinella brasiliensis (espécie de peixe) e comparando com a presença do contaminante na água e sedimento. Três praias de Ubatuba (exposta, calma e estuário abrigado) foram amostradas para os componentes peixe, água e sedimento durante o verão e o inverno. Dados ambientais evidenciaram diferenças espaciais e sazonais (ACP/ANOVA). A presença de partículas sintéticas (PSs) em peixes foi alta (~38%). Concentrações máximas ocorreram no estuário, para água (490 PSs/m³), e na praia exposta, para sedimento (62 PSs/50g). O formato fibra predominou em todos os componentes. Preferência dos peixes pela cor azul parece ocorrer. Relações estatísticas significativas foram determinadas para comprimento dos peixes e tamanho das PSs e entre concentrações de PSs na água e nos peixes. As identidades químicas (μ-FTIR spectra), polipropileno, polietileno, poliamida, poliéster e celulose/papelão predominaram. Influências da hidrodinâmica local (e.g., tamanhos das PSs) e da redução do turismo durante a epidemia de Covid-19 (e.g., menos PSs no verão) são discutidas. Abstract Microplastics (MPs) contamination is a well-established impact in oceans, but integrated approaches combining simultaneous analyzes of biotic and abiotic components are scarce. This study addresses this gap, demonstrating Atherinella brasiliensis (fish species) ingestion of MPs and comparing with the contaminant presence in water and sediment. Three Ubatuba beaches (exposed, calm and sheltered estuary) were surveyed for fish, water, and sediment components in summer and winter. Environmental data evidenced spatial and seasonal differences (PCA/ANOVA). Presence of synthetic particles (SPs) in fish was high (~38%). Maximum concentrations occurred in the estuary, for water (490 SPs/m³), and in the exposed beach, for sediment (62 SPs/50g). Fibers format predominated in all components. Fish preference for blue color seems to occur. Significant statistical relationships were determined for fish length and SPs size and between SPs concentrations in water and fish. The chemical identities (μ-FTIR spectra) polypropylene, polyethylene, polyamide, polyester, and cardboard/cellulose predominated. Influences of local hydrodynamics (e.g., SPs sizes) and reduction in tourism during Covid-19 epidemic (e.g.,less SPs in summer) are discussed. RESUMO: Originária da América Tropical, a goiabeira (Psidium guajava L.) tem grande relevância para o Brasil. Contudo, o surgimento do patossistema designado como declínio da goiabeira, problema fitossanitário provocado pelo parasitismo das raízes pelo nematoide-das-galhas (Meloidogyne enterolobii Yang & Eisenback) em associação com fungos oportunistas, dizimou muitos pomares em todas as regiões do Brasil e em outros países. No presente estudo, propagou-se vegetativamente, por miniestaquia, acessos de goiabeiras seminíferas mantendo seus genótipos preservados e reavaliando a resistência por meio dos clones de forma a comprovar ou não as reações das plantas hospedeiras. Os resultados apontam para alta virulência do parasita, bem como alta hospedabilidade da espécie P. guajava, além da existência de grande variação da reação entre plantas do mesmo genótipo e entre genótipos distintos, o que indica que a estratégia de preservação do germoplasma e a reavaliação da reação em clones pode ser importante na busca e seleção de germoplasma com algum grau de resistência a M. enterolobii. A progênie da cv. Paluma P02R5R2 obteve a menor média de Fator de Reprodução do parasita (FR = 22,11) entre os genótipos avaliados, sendo classificada como moderadamente resistente e preservada para estudos posteriores. ABSTRACT: Native to the tropical Americas, guava (Psidium guajava L.) is an important crop in Brazil. In the present study, seminiferous guava accessions were vegetatively propagated by minigrafting and their genotypes preserved for resistance reassessment in clones to confirm or not host plant reactions. The results indicated a highly virulent parasite, high host suitability of the P. guajava species, and widely varying reactions among plants of the same genotype and between different genotypes, demonstrating that the strategy of preserving the germplasm and reassessing reactions in clones may be important in identifying and selecting germplasms with a degree of resistance to M. enterolobii. The progeny of cv. Paluma P02R5R2 obtained the lowest average parasite reproduction factor (RF = 22.11) among the genotypes evaluated and was; therefore; classified as moderately resistant and preserved for future research.
