RESUMO Foram avaliadas 60 adrenais de 30 cães, divididos em três grupos, de acordo com a idade: grupo 1 = jovens saudáveis (10 animais) - de um a três anos de vida; grupo 2 = adultos saudáveis (10 animais) - de quatro a nove anos de vida; e grupo 3 = senis saudáveis (10 animais) - a partir de 10 anos de vida. O delineamento experimental foi ao acaso, com significância de 5%. No comprimento, houve diferença significativa (P=0,017) entre os cães jovens e os idosos. A espessura do polo cranial apresentou diferença significativa (P=0,001) entre jovens e adultos quando comparados com o grupo de idosos. A espessura do polo caudal não foi significativa para adultos quando comparadas com idosos e jovens. Entre idosos e jovens, a diferença foi significativa (P=0,024). As médias das velocidades diferiram entre os grupos de animais jovens e adultos quanto à adrenal esquerda e a seu polo cranial. ABSTRACT Sixty adrenals of 30 dogs were evaluated, divided into three groups, according to age: Group 1 = healthy youngsters (10 animals) - from one to three years old; Group 2 = healthy adults (10 animals) - four to nine years old; and Group 3 = healthy senile animals (10 animals) - older than ten years old. The experimental design was randomized, with a significance of 5%. There was a significant difference (P=0.017) in length between young and elderly dogs. The thickness of the cranial pole showed a significant difference (P=0.001) between young and adults when compared with the elderly group. The thickness of the caudal pole was not significant for adults when compared to elderly and young people. Between elderly and young, the difference was significant (P=0.024). The mean speeds differed between the groups of young and adult animals regarding the left adrenal and its cranial pole. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Islet transplantation represents a therapeutic option for type 1 diabetes (T1D). Long-term viability of transplanted islets requires improvement. Mesenchymal stromal cells (MSCs) have been proposed as adjuvants for islet transplantation facilitating grafting and functionality. Stem cell aggregation provides physiological interactions between cells and enhances the in situ concentration of modulators of inflammation and immunity. We established a hanging-drop culture of adult human skin fibroblast-like cells as spheroids, and skin spheroid-derived cells (SphCs) were characterized. We assessed the potential of SphCs in improving islet functionality by cotransplantation with a marginal mass of allogeneic islets in an experimental diabetic mouse model and characterized the secretome of SphCs by mass spectrometry-based proteomics. SphCs were characterized as multipotent progenitors and their coculture with anti-CD3 stimulated mouse splenocytes decreased CD4+ T cell proliferation with skewed cytokine secretion through an increase in the Th2/Th1 ratio profile. SphCs-conditioned media attenuated apoptosis of islets induced by cytokine challenge in vitro and importantly, intratesticular SphCs administration did not show tumorigenicity in immune-deficient mice. Moreover, SphCs improved glycemic control when cotransplanted with a marginal mass of allogeneic islets in a diabetic mouse model without pharmacological immunosuppression. SphCs' protein secretome differed from its paired skin fibroblast-like counterpart in containing 70% of up- and downregulated proteins and biological processes that overall positively influenced islets such as cytoprotection, cellular stress, metabolism, and survival. In summary, SphCs improved the performance of transplanted allogeneic islets in an experimental T1D model, without pharmacological immunosuppression. Future research is warranted to identify SphCs-secreted factors responsible for islets' endurance. T1D. TD . The scientific publication landscape is changing quickly, with an enormous increase in options and models. Articles can be published in a complex variety of journals that differ in their presentation format (online-only or in-print), editorial organizations that maintain them (commercial and/or society-based), editorial handling (academic or professional editors), editorial board composition (academic or professional), payment options to cover editorial costs (open access or pay-to-read), indexation, visibility, branding, and other aspects. Additionally, online submissions of non-revised versions of manuscripts prior to seeking publication in a peer-reviewed journal (a practice known as pre-printing) are a growing trend in biological sciences. In this changing landscape, researchers in biochemistry and molecular biology must re-think their priorities in terms of scientific output dissemination. The evaluation processes and institutional funding for scientific publications should also be revised accordingly. This article presents the results of discussions within the Department of Biochemistry, University of São Paulo, on this subject.

ABSTRACT In the present work, different Brazilian biomes aiming to identify and select cyclodextrin glycosyltransferase-producer bacteria are explored. This enzyme is responsible for converting starch to cyclodextrin, which are interesting molecules to carry other substances of economic interest applied by textile, pharmaceutical, food, and other industries. Based on the enzymatic index, 12 bacteria were selected and evaluated, considering their capacity to produce the enzyme in culture media containing different starch sources. It was observed that the highest yields were presented by the bacteria when grown in cornstarch. These bacteria were also characterized by sequencing of the 16S rRNA region and were classified as Bacillus, Paenibacillus, Gracilibacillus and Solibacillus.

RESUMO A tartaruga-verde, Chelonia mydas, apresenta distribuição cosmopolita. No Brasil, ocorre na costa, porém desova em ilhas oceânicas. A helmintofauna de tartarugas-verdes é diversificada, podendo-se dizer que tem a maior diversidade comparada com outras espécies de tartarugas. Objetivou-se avaliar aspectos ecológicos da comunidade de helmintos gastrointestinais e relacionar com a condição corporal de tartarugas-verdes recolhidas no litoral do Espírito Santo. Foram utilizados 36 exemplares juvenis da espécie C. mydas. O trato gastrointestinal foi separado e dividido em porções: esôfago/estômago, intestino delgado e intestino grosso. Cada porção foi inspecionada à procura de parasitos, e os exemplares encontrados foram separados para posterior identificação. Das 36 tartarugas avaliadas, 34 estavam parasitadas por helmintos (94,44%), com um total de 10.734 helmintos. Foram encontradas 18 espécies de trematodas pertencentes a quatro famílias. A riqueza de espécies encontrada foi de 4,29±2,19 (1-10) e a intensidade média de infecção foi de 315,64±281,83 (2-994) helmintos. Os parasitos mais prevalentes foram Cricocephalus albus, Metacetabulum invaginatum e Neoctangium travassosi, ambos com 61,11% (22/36), Pronocephalus obliquus com 33,33% (12/36), e Glyphicephalus lobatus com 30,55% (11/36). O helminto mais abundante foi M. invaginatum com 70,63 helmintos/animal, seguido de C. albus com 58,77 helmintos/animal e N. travassosi com 41,75 helmintos/animal.
ABSTRACT The green turtle, Chelonia mydas has worldwide distribution. In Brazil, it is found on the coast, but spawning occurs on oceanic islands. The helminth fauna of green turtles is diverse and has the greatest diversity when compared with other species of turtles. This study aims to evaluate ecological aspects of gastrointestinal helminth community and connect to the body condition of green turtles collected on the coast of Espírito Santo. A total of 36 juvenile specimens of the species C. mydas were used. The gastrointestinal tract was removed and divided into portions: esophagus/stomach, small intestine and large intestine. Each portion was inspected looking for parasites and the specimens found were separated for later identification. Of the 36 turtles evaluated, 34 were parasitized by helminths (94.44%), with a total of 10,734 helminths. Results include findings of 18 species of trematodes belonging to four families. The species richness was 4,29 ± 2,19 (1-10) and the mean intensity of infection was 315,64 ± 281,83 (2-994) helminths. The prevalent parasites were Cricocephalus albus, Metacetabulum invaginatum and Neoctangium travassosi, both with 61,11% (22/36), Pronocephalus obliquus with 33,33% (12/36), and Glyphicephalus lobatus with 30,55% (11/36). The abundant helminth was Metacetabulum invaginatum with helminths 70,63/animal, followed by C. albus with helminths 58,77/animal and N. travassosi with helminths 41,75/animal.

Inflammatory cytokines and plasma redox status responses in hypertensive subjects after heat exposure

Hypertension is characterized by a pro-inflammatory status, including redox imbalance and increased levels of pro-inflammatory cytokines, which may be exacerbated after heat exposure. However, the effects of heat exposure, specifically in individuals with inflammatory chronic diseases such as hypertension, are complex and not well understood. This study compared the effects of heat exposure on plasma cytokine levels and redox status parameters in 8 hypertensive (H) and 8 normotensive (N) subjects (age: 46.5±1.3 and 45.6±1.4 years old, body mass index: 25.8±0.8 and 25.6±0.6 kg/m2, mean arterial pressure: 98.0±2.8 and 86.0±2.3 mmHg, respectively). They remained at rest in a sitting position for 10 min in a thermoneutral environment (22°C) followed by 30 min in a heated environmental chamber (38°C and 60% relative humidity). Blood samples were collected before and after heat exposure. Plasma cytokine levels were measured using sandwich ELISA kits. Plasma redox status was determined by thiobarbituric acid reactive substances (TBARS) levels and ferric reducing ability of plasma (FRAP). Hypertensive subjects showed higher plasma levels of IL-10 at baseline (P<0.05), although levels of this cytokine were similar between groups after heat exposure. Moreover, after heat exposure, hypertensive individuals showed higher plasma levels of soluble TNF receptor (sTNFR1) and lower TBARS (P<0.01) and FRAP (P<0.05) levels. Controlled hypertensive subjects, who use angiotensin-converting-enzyme inhibitor (ACE inhibitors), present an anti-inflammatory status and balanced redox status. Nevertheless, exposure to a heat stress condition seems to cause an imbalance in the redox status and an unregulated inflammatory response.

Eficiência de óleos essenciais na qualidade sanitária e fisiológica em sementesde feijão-fava (Phaseolus lunatus L.)

RESUMO A espécie Phaseolus lunatus L., conhecida popularmente como feijão-fava constitui uma das alternativas de renda e alimentação para a população da região Nordeste do Brasil. Um dos problemas enfrentados são as doenças que provocam grandes perdas na produção de sementes. O presente trabalho objetivou avaliar a eficiência dos óleos essenciais de copaíba, cravo-da-índia e manjericão na redução da incidência de fungos associados às sementes de feijão-fava, e sua interferência na qualidade fisiológica. Os tratamentos foram constituídos por óleos essenciais de copaíba, cravo-da-índia e manjericão nas concentrações de 0; 1; 1,5 e 2 mL. L-1 e fungicida (Captan®). Foram utilizadas 200 sementes por tratamento. Para análise da qualidade sanitária das sementes foi adotado o método de incubação em placas de Petri contendo dupla camada de papel-filtro umedecida com ADE, sob temperatura de 20 ±2 ºC, por 7 dias. A qualidade fisiológica das sementes foi determinada com base nos testes de germinação, emergência e vigor das sementes. Para o teste germinação foi empregado método de rolo de papel germitest e na emergência osemeio foi realizado em bandejas plásticas contendo areia lavada esterilizada, em condição de casa de vegetação, e após 9 dias da instalação foi determinado o vigor com base nos resultados de índice de velocidade de germinação e emergência; comprimento da parte aérea, raiz primária e plântula; e teor de massa seca da partes aérea, raízes e plântulas de feijão-fava. O delineamento experimental foi interiamente casualizados, em esquama de fatorial simples (3x4)+1 (óleos essenciais x concentrações + fungicida), com quatro repetições de 50 sementes. Os óleos essenciais de copaíba e manjericão reduziram consideravelmente o percentual de incidência dos fungos associados as sementes de feijão-fava. O óleo essencial de cravo-da-índia na concetração de 2 mL. L-1, reduziu a qualidade fisiológica das sementes de feijão-fava.
ABSTRACT The Phaseolus lunatus L. species, popularly known as lima bean, is one of the alternative sources of income and food for the population of Northeast Brazil. One of the problems is the diseases that cause great losses in seed production. This study evaluated the effectiveness of essential oils of copaiba, clove, and basil in reducing the incidence of fungi associated with lima bean seeds, as well as the effect on their physiological quality. The treatments consisted of essential oils of copaiba, clove, and basil at concentrations of 0; 1; 1.5; and 2 mL. L-1 and fungicide (Captan®). A total of 200 seeds were used per treatment. To analyze the sanitary quality of the seeds, the method used was incubation in petri dishes containing a double layer of filter paper moistened with distilled sterilized water at a temperature of 20 ± 2°C, for seven days. The physiological quality of the seeds was determined by testing their germination, emergence, and vigor. For the germination test, the method used was a roll of Germitest paper. For the emergence, the seeds were sown in plastic trays containing washed sterilized sand in greenhouse conditions. After nine days, the vigor was determined based on the results of germination and emergence speed rates; length of the aerial part, primary root, and seedling; and dry matter content of the aerial part, roots, and seedlings of the lima beans. The experimental design was completely randomized in a simple factorial arrangement (3x4)+1 (essential oils x concentrations + fungicide) with four replications of 50 seeds. The essential oils of copaiba and basil considerably reduced the percentage of incidence of the fungi associated with lima bean seeds. The essential clove oil at 2 mL. L-1 reduced the physiological quality of the lima bean seeds.

Resumo: O cerrado é a mais diversificada savana tropical do mundo e constitui grande importância ecológica. Ainda assim, são poucos os estudos realizados no Brasil a respeito das doenças que acometem os canídeos desse bioma, o que causa prejuízos à população de animais selvagens e à saúde pública. Essa pesquisa foi realizada com o objetivo de avaliar a ocorrência de anticorpos contra Leptospira spp. em três espécies de canídeos selvagens do cerrado brasileiro utilizando o teste de soroaglutinação microscópica (SAM). Foram testadas amostras de soro sanguíneo de 19 cachorros-do-mato (Cerdocyon thous), 14 lobos guarás (Chrysocyon brachyurus) e sete raposas-do-campo (Lycalopex vetulus) de vida livre das regiões de cerrado de Araguari e Uberlândia, MG, e Cumari, GO. Dessas 40 amostras, 14 (35%) foram soropositivas. Os sorovares detectados com maior frequência foram Copenhageni e Hardjo, porém evidenciou-se também reação aos sorovares Autumnalis, Grippotyphosa, Hbedomadis, Wolffi e Icterohaemorrhagiae. Esse trabalho foi o primeiro a relatar a presença de anticorpos anti-Leptospira spp. em L. vetulus e verificou-se que as três espécies de canídeos utilizadas nessa pesquisa podem atuar como possíveis hospedeiros de diversos sorovares de leptospiras no ambiente de cerrado.
Abstract: The Brazilian savanna, also known as Cerrado, is one of the world's richest and most ecologically invaluable tropical savanna regions. There are few studies in Brazil about the diseases that affect the wild canids of this biome, which may be harmful to wildlife populations and public health. The aim of this study was to evaluate the occurrence of antibodies against Leptospira spp. in three Cerrado wild canids species using the microscopic agglutination test (MAT). Serum samples were tested from 19 crab-eating foxes (Cerdocyon thous), 14 maned wolves (Chrysocyon brachyurus), and seven hoary foxes (Lycalopex vetulus), all free-ranging animals found in the municipalities of Araguari and Uberlândia, Minas Gerais State, and Cumari, Goiás State, Brazil. Fourteen (35%) of these samples were seropositive. The most frequent serovars detected in the samples were Copenhageni and Hardjo, but reactions to the serovars Autumnalis, Grippotyphosa, Hebdomadis, Wolffi, and Icterohaemorrhagiae also occurred. Notwithstanding other reported results, this study is the first to report the presence of antibodies against Leptospira spp. in L. vetulus. The three species of wild canids examined may act as potential hosts for several serovars of leptospira in Brazil's savanna environment.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Leukograms of healthy Holstein calves within the first month of life

Com a finalidade de estabelecer os valores de referência e de avaliar a influência do fator etário sobre o leucograma de bezerras sadias, da raça Holandesa, utilizaram-se amostras de sangue de 300 animais, distribuídos com igual número (n=20) por 15 grupos experimentais, de acordo com a idade: do nascimento até 8 horas de vida; de 9 até 16 horas; de 17 até 24 horas; 2 dias de idade; 3 dias; 4 dias; 5 dias; 6 a 7 dias; 8 a 9 dias; 10 a 11 dias; 12 a 13 dias; 14 a 15 dias; de 16 a 20 dias; de 2l a 25 dias; e de 26 a 30 dias de vida. Puderam-se observar valores máximos nas primeiras oito horas de vida para leucócitos (9305,0/µL), neutrófilos segmentados (6551,2/µL) e neutrófilos totais (6678,3/µL); entre 9 a 16 horas pós-nascimento para os neutrófilos bastonetes (133,3/µL). Para os linfócitos totais (4992,1/µL) e os linfócitos típicos (4686,1/µL) os valores máximos ocorreram entre 21 e 25 dias, e para os linfócitos atípicos (388,5/µL), entre 26 e 30 dias, demonstrando-se uma inversão na relação neutrófilo: linfócito, com o decorrer dos dias de vida. Constatou-se a da influência do fator etário no leucograma dos bezerros avaliados, no entanto acredita-se que a liberação de corticoides endógenos no momento do parto/nascimento, contribuíram para esta variação.
To establish reference values and to assess the influence of age on the leukograms of healthy Holstein calves, blood samples were obtained from 300 animals. These samples were distributed equally (n=20) among 15 experimental groups according to age: birth to 8 hours, 9 to 16 hours, 17 to 24 hours, 2 days, 3 days, 4 days, 5 days, 6 to 7 days, 8 to 9 days, 10 to 11 days, 12 to 13 days, 14 to 15 days, 16 to 20 days, 21 to 25 days and 26 to 30 days of age. The maximum numbers of leukocytes (9,305.0/mL), segmented neutrophils (6,551.2/mL) and total neutrophils (6,678.3/mL) were noted within the first 8 hours of life, while band neutrophils peaked in number (133.3/mL) between 9 and 16 hours after birth. Meanwhile, the maximum total lymphocyte (4,992.1/µL) and typical lymphocyte (4,686.1/µL) counts occurred between 21 and 25 days, whereas atypical lymphocytes (388.5/µL) reached their maximum number between 26 and 30 days, demonstrating an inversion of the neutrophil:lymphocyte ratio over time. Thus, the influence of age on the leukocyte count of the evaluated calves was verified. The release of endogenous corticosteroids during labor or at birth may contribute to this variation in leukograms with age.

La concepción de los profesionales de salud sobre el parto humanizado en la adolescencia

The aim of this study is to know the concept of health professionals of an obstetric unit in the southern region of Brazil about humanized childbirth of adolescents. It is a descriptive and explanatory study with qualitative approach which is linked to multicentric research called "Childbirth Humanized Care in Adolescents" developed in the obstetric unit of a college hospital in the south of Brazil. The subjects of this study were ten health workers of the obstetric unit of the mentioned institution. The data was collected through semi-structured interviews, from November 01st, 2008 to October 31st, 2009. For this research cut it was used the question 17. Two themes were highlighted as result of analysis and organization of the findings in the subjects interviewed: ambience as care space and light technologies as tools for humanization of attention in parturient adolescent. The investigation has indicated that the humanizing attention to adolescents needs more than problem solving or use of hard technologies, it requires attitudes and behaviors of health workers who contribute for the strengthening of the link among professionals, users, family and community.
Este estudo tem como objetivo conhecer a concepção dos profissionais de saúde de um centro obstétrico do sul do Brasil sobre o parto humanizado de adolescentes. Trata-se de um estudo descritivo e exploratório com abordagem qualitativa, estando vinculado à pesquisa multicéntrica intitulada "Atenção Humanizada ao Parto de Adolescentes", desenvolvida no centro obstétrico de um hospital de ensino do sul do Brasil. Os sujeitos do estudo foram dez trabalhadores da saúde efetivos do centro obstétrico da referida instituição. Os dados foram coletados a partir de entrevistas semi-estruturadas, entre 01 de novembro de 2008 a 31 outubro de 2009. Para este recorte da pesquisa utilizou-se especificamente a questão 17. Como resultado da análise e organização dos achados nos depoimentos dos sujeitos foram destacadas duas temáticas: a ambiência como espaço de cuidado e as tecnologias leves como ferramentas para a humanização da atenção à parturiente adolescente. A investigação apontou que a atenção humanizada ao parto de adolescentes necessita mais do que resolução de problemas ou uso de tecnologias duras e leve-duras, exige atitudes e comportamentos dos trabalhadores da saúde que contribuam para o fortalecimento do vínculo entre profissionais, usuárias, família e comunidade.

Identification of Staphylococcus spp. isolated during the ripening process of a traditional minas cheese

Estudou-se a dinâmica das populações de Staphylococcus spp. durante a maturação do queijo Canastra, em três fazendas localizadas no estado de Minas Gerais. A presença dos genes que codificam para a produção das enzimas coagulase (coa), termonuclease (nuc) e produção de enterotoxinas (sea, seb, sec e sed), em linhagens de Staphylococcus isoladas durante os 60 dias de maturação do queijo foi analisada. Também foi investigada a presença de enterotoxina estafilocócica A, C e D nas amostras de queijo. As amostras de queijo com 0, 7, 15, 30 e 45 dias de maturação apresentaram contagens de Staphylococcus spp. entre 10³ e 10(8)ufc / g. Todos os isolados coagulase positivo nos testes fisiológicos apresentaram o gene coa. Não foi observada associação entre os resultados obtidos com os testes bioquímicos e aqueles obtidos com a PCR usando iniciadores gene-específicos para linhagens coagulase negativa. Os genes da coagulase e termonuclease ocorreram simultaneamente em 41,3% dos Staphylococcus spp. testados. Nenhum dos isolados de Staphylococcus apresentou os genes que codificam para a produção das enterotoxinas SEA, SEB, SEC ou SED. As enterotoxinas A, C ou D não foram detectadas em nenhuma das amostras de queijo analisadas.
The population dynamics of Staphylococcus spp. was studied during the ripening of Canastra Minas cheese at three farms located in the State of Minas Gerais, Brazil. The presence of coagulase (coa), thermonuclease (nuc), and enterotoxin (sea, seb, sec, and sed) genes was investigated in Staphylococcus strains isolated during the 60-day cheese-ripening period. The presence of the staphylococcal enterotoxins A, C, and D was also investigated in the cheese samples. Cheese samples that were matured for 0, 7, 15, 30, and 45 days presented staphylococci counts from 10³ to 10(8)cfu/g. All isolates considered coagulase-positive by physiological tests had the coa gene. However, no association was observed between the results obtained with biochemical tests and those obtained by PCR using gene-specific primers for coagulase-negative strains. Coagulase and thermonuclease genes occurred simultaneously in 41.3% of Staphylococcus spp. tested. None of the investigated Staphylococcus strains expressed enterotoxins SEA, SEB, SEC, and SED. Enterotoxins A, C, and D were not detected in any of the cheese samples.

The essential oil from the leaves of Myrcia ovata Cambess., commonly used in Brazil for the treatment of gastric illnesses, was screened for antimicrobial activity and action in the formation of microbial biofilms by Enterococcus faecalis. The oil was obtained by hydrodistillation using a clevenger-type system. Its chemical composition was analyzed using GC and GC-MS. Both MIC and MBC of the essential oil were determined by broth microdilution techniques and agar dilution method. The essential oil showed antimicrobial activity against E. faecalis, Escherichia coli, Pseudomonas aeruginosa, Salmonella choleraesuis, Staphylococcus aureus, Streptococcus pneumoniae and Candida parapsilosis. The results showed that the essential oil of M. ovata Cambess. was effective against the formation of biofilm by E. faecalis when compared with the control. Four volatile compounds, representing 92.1 % of the oil, were identified and geranial was the major component (50.4 %). Efeitos dos glicosaminoglicanos e sulfato de condroitina A sobre a cartilagem articular normal e com doença articular degenerativa em cães

Avaliaram-se os efeitos dos precursores dos glicosaminoglicanos (GAG) e do sulfato de condroitina A (SC) sobre a histomorfometria da cartilagem articular normal ou de cartilagem de cães com doença articular degenerativa (DAD) experimental. Os grupos experimentais constituíram-se de animais com articulação direita normal, que não foi submetida a procedimento cirúrgico, e com articulação esquerda osteoartrótica e que foi submetida à intervenção cirúrgica. Os grupos foram subdivididos em animais com articulação não tratada e tratada, portanto: normais (N) (n=5), NGAG (n=5) e NSC (n=4); e osteoartróticos (O) (n=5), OGAG (n=5) e OSC (n=4). Secções de cartilagens do fêmur, da tíbia e da patela foram utilizadas neste estudo. Nos normais (N, NGAG e NSC), não se encontraram lesões que caracterizassem a DAD, embora tenha havido diminuição na celularidade nos de NGAG e NSC, em relação a N. Foram observadas alterações em graus variáveis entre os grupos osteoartróticos. Houve redução acentuada dos condrócitos no grupo O em comparação aos normais enquanto os grupos osteoartróticos tratados apresentaram celularidade semelhante aos normais tratados. Estes resultados foram confirmados pela análise do índice de proporção (IP), que se mostrou elevado em O, indicando menor síntese de proteoglicanos. Não houve diferença significativa entre os IPs dos grupos osteoartróticos tratados (OGAG, OSC) apesar do comportamento distinto do OSC ao assemelhar-se aos grupos N e NSC. Estes resultados sugeriram que o SC agiu na cartilagem osteoartrótica de maneira mais eficaz, reduzindo a perda de proteoglicanos e estimulando a viabilidade celular e a atividade metabólica.
The effects of precursors of glycosaminoglycans (GAG) and chondroitin sulphate A (CS) on the histomorphometry of normal articular cartilage and with experimental degenerative joint disease (DJD) in dogs were evaluated. The groups were constituted as follows: normal joints were not undergone to the surgical procedure and left osteoarthritics that suffered surgical intervention. These were then distributed into joints that did not receive drug treatment and those that received: normal (N) (n = 5); NGAG (n = 5), and NCS (n = 4); and osteoarthritics: O (n = 5); OGAG (n = 5); and OCS (n = 4). Cartilage sections of the femur, tibia, and patella were used in this study. In normal groups (N, NGAG, and NCS) no lesions were found that could characterize DJD, although the cellularity was shown to be slightly diminished in NGAG and NCS, in relation to N. Characteristic DJD disorders were identified in variable degrees among osteoarthritic groups and were more severe in O. There was marked reduction of the number of chondrocytes in group O, as compared to the normal groups, while treated osteoathritic groups showed similar cellularity to that of normal groups. These results were supported by the analysis of the index of proportion (IP), which was high in O, indicating less synthesis of proteoglycans. These results suggest that chondroitin A sulphate acted on osteoarthritic cartilage in a more efficient way than glycosaminoglycan precursors, reducing the loss of proteoglycans caused by the degenerative process, stimulating cellular viability and metabolic activity.
