Abstract This study aimed to assess the feasibility of comet and cytogenetic tests as tools for evaluating genomic instability in seeds of Oryza sativa L. (rice) and Phaseolus vulgaris (beans) L. from gene banks. Rice and beans were exposed to methyl methanesulfonate (MMS) as a reference DNA damaging agent. Seeds of two accessions of rice and beans were obtained from Embrapa Rice and Beans - Brazil. Seed groups were imbibed in three concentrations of MMS for three periods of time to carry out cytogenetic tests, and for one period for the comet test. At concentrations of 10 and 15 mg/L, MMS induced cytotoxic and/or mutagenic effects in the meristematic cells of roots from all the accessions of both species. In the comet test, MMS induced genotoxic effects at all the concentrations in the evaluated accessions of rice and beans, except in one accession of beans at the lowest concentration (5 mg/L). Both species showed sensitivity to MMS. The comet test can be proposed for the measurement of genomic instability in accessions of rice and beans in gene banks, as being more sensitive than the cytogenetic tests used.

This study aims to characterize the complex metal-clay formed by adsorption of lead. In this work, the bentonite clay named Fluidgel was calcined at 750 ºC and was used as adsorbent for lead removal. The characterization of this clay and complex metal-clay was carried out by X-ray diffraction (XRD), Fourier transformer infrared spectroscopy (FTIR), surface area (BET method), scanning electron microscopy (SEM) and chemical composition by energy dispersive X-ray (EDX). The removal of lead from aqueous solution was carried out in a fixed bed. Dynamic experiments were performed to evaluate the effect of flowrate on adsorption efficiency. The experiments were accomplished at room temperature, the clay adsorbent particle diameter was of 0.855 mm and the flow rate varied from 0.6 to 2 mL/min. The feed concentration of lead was about 0.24 mmol/L. Comparing the chemical compositions of the clays obtained by EDX before and after removal of the lead, it was concluded that the ion exchange process might be important to lead removal. From the semi-quantitative analysis of chemical composition in clays with and without adsorbed lead it was observed a reduction of the amount of Ca2+, K+ cations and the disappearance of Na+ cations, which was caused by cation exchange process.

Com o objetivo de obter informações básicas para o futuro zoneamento do Parque Nacional das Emas (PNE), foram realizadas amostragens ictiofaunísticas e de parâmetros limnólógicos durante o período de seca (setembro/1999) e de chuvas (dezembro/1999) em estações de coletas localizadas no interior do Parque e em áreas adjacentes (rios Araguaia e Sucuriú). Foram capturados 4.740 indivíduos, distribuídos em 22 espécies. Entre as espécies mais abundantes destacam-se Astyanax sp. 2 e Hasemania sp. no rio Araguaia e Astyanax scabripinnis cf. paranae e Hoplias aff. malabaricus no rio Sucuriú e PNE. A maior diversidade de espécies foi constatada para o rio Araguaia. Entretanto, para que medidas de manejo possam ser tomadas de forma criteriosa, estudos ecológicos mais detalhados das comunidades de peixes do cerrado devem ser conduzidos.

The relationship between habitats and the ichthyofauna composition in the Parque Nacional das Emas (PNE) and adjacent areas (the Araguaia and Sucuriú rivers) are provided and could be applied in determining the Park's future zoning. Samples of the ichthyofauna and limnological parameters were obtained during both dry (September 1999) and wet (December 1999) seasons. Ichthyofauna collections resulted in the capture of 4,740 specimens of 22 species. The most abundant species in the Araguaia River during the two sampling seasons were Astyanax sp. 2 and Hasemania sp. In the Sucuriú River and PNE, Astyanax scabripinnis cf. paranae and Hoplias aff. malabaricus were the most frequent species. The largest number of species and diversity index were recorded for the Araguaia River. However, sound management policies require more detailed studies on the fish communities of the Cerrado biome.
