RESUMO: A determinação de eugenol em óleo essencial de cravo-da-índia foi realizada utilizando um dispositivo operado com smartphone para captura e processamento de imagens. Para tal desenvolvimento foi utilizada a reação colorimétrica com o reagente de Folin-Ciocalteu. Foram avaliadas diferentes condições de iluminação para a captura das imagens diretamente em frascos descartáveis de 2 mL. O aplicativo livre PhotoMetrix UVC foi usado para a calibração por regressão de mínimos quadrados parciais com valores adequados (R2 maior que 0.99). A exatidão do método proposto foi comparada com métodos tradicionais, tais como cromatografia gasosa (CG) e espectrofotometria, sem diferenças significativas (teste t de Student (P > 0,05) e com concordâncias entre 97% e 101%. O método com smartphone permitiu a avaliação de várias amostras em poucos minutos, com etapas de análise simples e com fácil interpretação dos resultados. A escala miniaturizada permitiu o uso de pequenas quantidades de reagentes e mínima geração de resíduos. Portanto, o método proposto pode ser facilmente operado em campo e permite avaliar a qualidade de óleo essencial de cravo- da- índia em locais de acesso restrito, sem a necessidade de estrutura laboratorial e analistas especializados. ABSTRACT: The determination of eugenol in clove essential oil was performed using a smartphone-operated device, which was used for image capture and processing. The colorimetric reaction with the Folin-Ciocalteu reagent was used, and the lighting conditions were evaluated to capture images directly in 2 mL disposable vessels. The free application PhotoMetrix UVC was used for partial least squares regression calibration with suitable values (R² greater than 0.99). The accuracy of the proposed method was compared with traditional methods, such as gas chromatography (GC) and spectrophotometry, and it can be observed that there were no significant differences (Student’s t-test (P > 0.05), with agreements from 97% to 101%. The smartphone method allowed the evaluation of several samples in a few minutes, with simple analysis steps and easy interpretation of the results. The miniaturized scale allowed the use of small amounts of reagents with minimal waste generation. Therefore, the proposed method can be easily operated in the field and allows the evaluation of the quality of clove essential oil in places of restricted access without the need for a laboratory structure and specialized analysts. ABSTRACT This work aimed to evaluate the morphological, physical and chemical quality of fruits of the mandarin Satsuma 'Okitsu', which belongs to the Group of Satsumas (Citrus unshiu Marcovitch), the mandarin 'Marisol', which belongs to the Group of Clementines (Citrus clementina Hort. ex Tanaka) and the tangor 'Ortanique' [C. sinensis (L.) Osbeck x C. reticulata Blanco]. The study was carried out at the Citrus Collection of the Polo of Technological Modernization from the Medio Alto Uruguay, located in Frederico Westphalen, State of Rio Grande do Sul, Southern Brazil. Analyses were performed to assess the size, density, percentage of juice of the fruits, maturation rates in the fruit juice (pH, total soluble solids, titulabre total acidity and relation between total soluble solids/ titulabre total acidity or ratio), and the ascorbic acid content (Vitamin C). Data were compared by Tukey test at p<0.05. The tangor 'Ortanique' showed total soluble solids, titulabre total acidity and ratio significantly higher than the mandarines. The mandarin 'Marisol' showed higher ascorbic acid content. The results were promising, because the mandarin 'Okitsu' and 'Marisol' and the tangor 'Ortanique' yielded in 2013 and 2014 showed that the fruit flavor characteristics were within the range founded to other hybrid and mandarins produced in other regions of Brazil. In this way they will be offered as alternatives to citrus growers in this region, further extending the harvest period of mandarins and seedless hybrids.

RESUMO O presente trabalho teve por objetivo avaliar a qualidade morfológica, física e química dos frutos das tangerineiras Satsuma 'Okitsu' pertencente ao Grupo das Satsumas (Citrus unshiu Marcovitch), e 'Marisol', pertencente ao Grupo das Clementinas (Citrus clementina Hort. ex Tanaka) e do tangoreiro 'Ortanique' [C. sinensis (L.) Osbeck X C. reticulata Blanco]. O trabalho foi realizado na Coleção de Citros do Polo de Modernização Tecnológica do Médio Alto Uruguai (PMTec) localizado no município de Frederico Westphalen, RS. Foram realizadas análises para avaliação do tamanho, densidade, porcentagem de suco dos frutos, índices de maturação no suco dos frutos (pH, sólidos solúveis totais, acidez total titulável e relação sólidos solúveis totais/acidez total titulável ou ratio), e os teores de ácido ascórbico (Vitamina C). Os dados foram comparados pelo teste de Tukey a p<0,05. O tangor 'Ortanique' apresentou teores de sólidos solúveis totais, acidez total titulável e ratio, significativamente superior às tangerinas. A tangerina 'Marisol' apresentou maior teor de ácido ascórbico. Os resultados mostraram-se promissores, pois, as tangerineiras 'Okitsu', e 'Marisol' e o tangoreiro 'Ortanique', nas safras 2013 e 2014, apresentaram características de sabor dos frutos dentro da faixa de resultados encontrados para outros híbridos e tangerinas produzidas em outras regiões do Brasil. Desta maneira poderão oferecer como alternativas aos citricultores desta região, ampliando ainda mais o período de colheita de tangerinas e híbridos sem sementes.

RESUMO: A extração de óleos essenciais é comumente realizada pelo método de hidrodestilação, o qual é descrito em compêndios oficiais para o controle de qualidade de alimentos e plantas medicinais. Apesar do largo uso deste método de extração, poucos estudos têm sido propostos para avaliar o efeito da mudança da atmosfera durante a extração na composição e atividade antioxidante de óleos essenciais. Portanto, um estudo sobre a influência da introdução de oxigênio na extração de óleos essenciais de manjericão, capim-limão e limão foi realizado. A quantidade total de compostos oxigenados (e.g., linalol, cânfora, α-terpineol, neral, geranial, eugenol e α-muurolol) aumentou em todos os óleos essenciais extraídos com oxigênio. A atividade antioxidante avaliada pelo método ORAC aumentou significativamente (P<0,0001) com a introdução de oxigênio, variando de 618 para 906, de 355 para 613 e de 72 para 262µmol Trolox g-1 óleo essencial de manjericão, capim-limão e limão, respectivamente. Portanto, a modificação simples proposta mostrou-se uma alternativa adequada para obtenção de óleos essenciais com maior atividade antioxidante.

ABSTRACT: Essential oil extraction is commonly carried out by using the hydrodistillation method, which is described in official compendia of food quality control and medicinal plants. Despite the widespread use of this method, few studies have evaluated the effect of the atmosphere change during extraction on the composition and antioxidant activity of essentials oils. Therefore, a study of oxygen introduction influence during the extraction of essential oils from basil, lemongrass and lemon by hydrodistillation was performed. Total amount of oxygenated compounds (e.g., linalool, camphor, α-terpineol, neral, geranial, eugenol and α-muurolol) increased for all essential oils extracted under oxygen flow. Antioxidant activity evaluated by using the ORAC method significantly increased (P<0.0001) with oxygen from 618 to 906, 355 to 613 and 72 to 262µmol Trolox g-1 oil for basil, lemongrass and lemon, respectively. Therefore, the simple modification proposed could be considered a suitable alternative to obtain essential oils with higher antioxidant activity.

Introduction Candida dubliniensis, a new species of Candida that has been recovered from several sites in healthy people, has been associated with recurrent episodes of oral candidiasis in AIDS and HIV-positive patients. This species is closely related to C. albicans. The enzymatic activity of C. dubliniensis in response to oxidative stress is of interest for the development of drugs to combat C. dubliniensis. Methods Fluconazole- and amphotericin B-resistant strains were generated as described by Fekete-Forgács et al. (2000). Superoxide dismutase (SOD) and catalase assays were performed as described by McCord and Fridovich (1969) and Aebi (1984), respectively. Results We demonstrated that superoxide dismutase (SOD) and catalase activities were significantly higher (p<0.05) in the fluconazole- and amphotericin B-resistant strains of C. dubliniensis and C. albicans than in the sensitive strains. The catalase and SOD activities were also significantly (p<0.01) higher in the sensitive and resistant C. albicans strains than in the respective C. dubliniensis strains. Conclusions These data suggest that C. albicans is better protected from oxidative stress than C. dubliniensis and that fluconazole, like amphotericin B, can induce oxidative stress in Candida; oxidative stress induces an adaptive response that results in a coordinated increase in catalase and SOD activities.

As plantas medicinais (i. e. Discaria americana) têm sido utilizadas pela população por séculos, entretanto, o conhecimento popular não é suficiente para validá-las como medicamentos seguros e/ou efetivos. Assim, o presente estudo teve por objetivo avaliar a toxicidade aguda e subaguda, bem como o efeito ansiolítico e antinociceptivo dos extratos da casca da raiz e das partes aéreas da D. americana em camundongos. A toxicidade aguda foi avaliada pela administração dos extratos, via intraperitoneal. Para o estudo da toxicidade subaguda os animais foram tratados oralmente com os extratos por 14 dias. O efeito ansiolítico dos extratos foi determinado através do modelo do labirinto em cruz elevado e o efeito antinociceptivo, mediante o teste da placa quente. O valor da DL50 para o extrato das partes aéreas da D. americana foi definido como > 500 mg/kg, enquanto que para o extrato da casca da raiz foi estabelecido em 400 mg/kg. O extrato das partes aéreas da D. americana apresentou atividade ansiolítica (250 mg/kg) e antinociceptiva (125, 200 e 250 mg/kg). O extrato da casca da raiz da D. americana não apresentou efeito ansiolítico nem antinociceptivo.

Medicinal plants (e.g. Discaria americana) have been used by populations for centuries. However, popular use is not enough to validate these plants as safe and effective medicinal products. The present study sought to evaluate the acute and subacute toxicity as well as the anxiolytic and antinociceptive effects of D. americana root bark and aerial parts extracts in mice. In acute toxicity studies, mice were treated with single intraperitoneal doses of the aforementioned extracts. Subacute toxicity studies were performed by oral administration of the extracts over 14 days. Anxiolytic studies consisted of the elevated plus maze method, and antinociceptive studies were based on the hot plate test. The LD50 value for D. americana aerial parts extract was established at >500 mg/kg, and for the root bark extract, 400 mg/kg. D. americana aerial parts extract produced anxiolytic (250 mg/kg) and antinociceptive effects (125, 200 and 250 mg/kg). Conversely, D. americana root bark extract showed neither anxiolytic nor antinociceptive effects in mice.

Este estudo teve como objetivo otimizar a extração de ecdisterona em raízes de ginseng brasileiro. Primeiramente, para se avaliar a eficiência do solvente extrator, amostras de raízes dois acessos (BRA e JB-UFSM) de P. glomerata foram extraídas em Soxhlet com metanol e clorofórmio, separadamente, durante 4 horas. No segundo ensaio, com o intuito de se escolher o método extrator, a extração foi conduzida em Soxhlet e em ultrassom, utilizando metanol como solvente. Em P. tuberosa, as amostras foram extraídas com metanol, e a extração foi conduzida em Soxhlet e em banho ultrasônico. O conteúdo de ecdisterona foi determinado em Cromatógrafo Líquido de Alta Eficiência (CLAE). Em ambas as espécies, um maior conteúdo de ecdisterona foi detectado nas amostras extraídas com metanol e em Soxhlet. A metodologia proposta mostrou-se eficaz para a quantificação da ecdisterona a partir das raízes de P. glomerata e P. tuberosa, podendo ser aplicada no controle de qualidade de drogas vegetais e/ou fitoterápicos.

This study aimed at optimizing the extraction method from ecdysterone of Brazilian ginseng. Root samples of two accessions (BRA and JB-UFSM) of P. glomerata were extracted in a Soxhlet with methanol or chloroform for 4h. In the second trial, the extration was conduced in a Soxhlet or ultrasonic using metanol as a solvent. In P. tuberosa, the roots samples were extracted with methanol in a Soxhlet or in ultrasonic. The ecdysterone content was determinated using high efficiency liquid chromatography methods. In both studied species, the highest ecdisterone content was detected from samples extracted in a Soxhlet and using methanol as a solvent. This extration method has been successfully applied for determination of ecdysterone content from roots of Brazilian ginseng, and could be useful for the quality control of drugs and pharmaceutical formulations.
