ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract The present work aimed to evaluate the influence of different eucalyptus hybrids on the edaphic macrofauna community. It is hypothesized that the density and diversity of edaphic fauna are influenced by the quantity and quality of the litter deposited by the different hybrids on the soil surface. The study area is in the city of Vitória da Conquista (BA) and consists of seven eucalyptus hybrids, one from sexual reproduction (seminal) and six from asexual (clonal) reproduction. Sampling of the soil macrofauna was performed by the Tropical Soil Biology and Fertility method. All visible invertebrates were collected and identified at the level of large taxonomic groups. The relative density, group richness, and diversity index were calculated. Hybrids did not influence the total density and richness of the soil macrofauna. However, it was possible to observe changes in litter accumulation and composition, and as a result in the diversity indexes and in the presence or absence of macrofauna groups. Among the studied hybrids, those from Eucalyptus urophylla (seminal, 1335 and 1249) promote greater macrofauna diversity and favor the occurrence of detritivores organisms, suggesting the occurrence of better quality and more palatable litter for the edaphic fauna.

Abstract Understanding the effects of different production systems on arbuscular mycorrhizal fungi (AMF) can help to interpret interactions between their components and to define management strategies. As a result, our study was conducted on soils under three coffee production systems (one homogeneous and two heterogeneous) and in a native forest located in the Bahia state, Brazil. This study aimed to answer the following questions: 1) Does the organization and management of the coffee production system affect the occurrence and diversity of AMF?; and 2) Is the seasonality effect similar between systems? To do so, soil samples (0-10 cm depth) were collected at two times of the year (rainy and dry). Number of spores (NS) and average richness did not show differences between the systems, only between seasons. There was a reduction in NS in the dry season (1.4 and 2.7 spores g-1 soil) in relation to the rainy season (3.8 to 12.5 spores g-1 soil). The influence of coffee production systems was observed in the presence and absence of some AMF species. The AMF community was shown to be related to the plant species composition of the system, which was reflected in the dissimilarity of heterogeneous systems in relation to the coffee monoculture system.

ABSTRACT The COVID-19 pandemic has caused a decrease in tourism activity. This has caused huge losses to the tourism industry, so different approaches have been devised for economic reactivation. This article aims to provide a novel strategy for mass molecular monitoring of clinically healthy individuals, and also including potentially asymptomatic carriers of SARS-CoV-2, who traveled to a tourist resort The monitoring consisted of collecting individual saliva samples (n=120) and conforming groups of 10 samples, thus setting 12 individual pools, which were analyzed by qRT-PCR. Obtained data showed that all the samples analyzed were negative, thus confirming that no individuals were infected with SARS-CoV-2 before and during the travel. The proposed molecular protocol allowed the analysis of massive saliva samples and detect individual infected persons (negative or positive). protocol for evaluate massive event, thus promoting the economic activation of the tourism industry.
RESUMEN La pandemia ha causado una disminución en la actividad turística afectado económicamente esta industria, en este sentido, se han planteado diferentes propuestas para reactivar esta actividad. Este articulo tiene como objetivo ofrecer una metodología para el monitoreo masivo de sujetos clínicamente sanos, incluyendo también portadores asintomáticos de SARS-CoV-2, que viajaron a un complejo turístico. El protocolo de monitoreo consiste en colectar muestra individual de saliva (n=120) y formar pools de 10 personas. Se analizaron un total de 12 pools, mediante RT-PCR (“Real-Time Polimerase Chain Reaction”). Los resultados mostraron que todas las muestras analizadas fueron negativas, esto indica que ningún individuo estaba infectado por SARS-CoV-2 antes y después del viaje. El protocolo molecular propuesto permite evaluar masivamente gran cantidad de muestras de saliva y detectar individualmente infectados (negativos o positivos), esto con el fin de promover activación de la industria turística.

RESUMO A toxoplasmose é uma zoonose de ampla distribuição, que acomete animais homeotérmicos, incluindo as aves e o ser humano. Galinhas podem ser consideradas indicadoras de contaminação ambiental. No Brasil, Toxoplasma gondii não apresenta estrutura populacional clonal (I, II e III), ocorrendo alta frequência de recombinação, o que resulta na grande diversidade genética observada. O presente trabalho teve como objetivo caracterizar geneticamente T. gondii de galinhas (Gallus domesticus) naturalmente infectadas do estado de Santa Catarina. Soros de 133 aves criadas extensivamente foram analisados pela reação de imunofluorescência indireta (RIFI) para detecção de anticorpos IgG contra T. gondii. Para o isolamento do parasito (bioensaio em camundongos), foram utilizados coração e cérebro de 30 aves que apresentaram títulos maiores que 64 na RIFI. Os isolados obtidos foram submetidos à caracterização genotípica por meio da RFLP-PCR utilizando 12 marcadores genéticos (SAG1, 5'-3'SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, Apico e CS3). Os resultados obtidos foram classificados de acordo com os genótipos presentes no ToxoDB (http://toxodb.org/toxo/). Das 133 amostras de soros de galinha analisadas, 84 (63,16%) foram positivas, com títulos de anticorpos variando de 16 a 1024. No bioensaio, foram obtidos 11 isolados (Ck 3, Ck 32, Ck 35, Ck 56, Ck 63, Ck 89, Ck 102, Ck 103, Ck 125, Ck127 e Ck 128). A análise genotípica revelou a presença de seis genótipos, três dos quais classificados como #26, #53 e #120, e três, NEO1, NEO2 e NEO3, não descritos anteriormente. Nenhuma linhagem clonal I, II ou III foi encontrada. O presente trabalho confirma a ampla diversidade genética do parasito observada no Brasil.
ABSTRACT Toxoplasmosis is a widespread zoonosis that can infect warm-blooded animals including birds and humans, and chickens are considered to be indicators of environmental contamination. In Brazil, Toxoplasma gondii has a non-clonal population structure composed of three lineages (I, II, and III), presenting high recombination, and resulting in wide genetic diversity. This study aimed to genetically characterize T. gondii isolates from naturally infected chickens (Gallus domesticus) in Santa Catarina state, southern Brazil region. Sera from 133 free-range chickens were analyzed by an immunofluorescence assay (IFA) to detect IgG antibodies against T. gondii. Brain and heart from 30 positive animals, based on IFA (≥ 1:64), were used to isolate the parasite using a mouse bioassay. Strain genotyping was performed by PCR-RFLP using 12 genetic markers (SAG1, 5´-3´SAG2, alt. SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, Apico, and CS3). The results were classified according to the genotypes based on the ToxoDB (http://toxodb.org/toxo/). Of 133 chicken sera analyzed, 84 (63.16%) were positive, with antibody titers ranging from 16 to 1024. Eleven isolates were obtained from mouse bioassay (Ck3, Ck32, Ck35, Ck56, Ck63, Ck89, Ck102, Ck103, Ck125, Ck127, and Ck128). Genotyping revealed six genotypes; three were classified as #26, #53, and #120, and three (NEO1, NEO2, and NEO3) were had not been previously described. No clonal lineages of type I, II, or III were identified. The present study confirms the high genetic diversity of T. gondii in Brazil.

Com os objetivos de verificar a ocorrência de anticorpos contra Neospora caninum em bovinos de corte da microrregião de Guara-puava, Paraná, e de correlacioná-la com a idade, o sexo e a raça dos animais, amostras de sangue de 250 bovinos foram analisadas por meio da Reação de Imunofluorescência Indireta (> 1:200). A análise estatística foi realizada por meio dos testes exato de Fisher e de qui-quadrado (p<0,05) para correlacionar os resultados da sorologia com as variáveis analisadas. Das 250 amostras avaliadas, 33 (13,2%) foram positivas para N. caninum. Os títulos obtidos para N. caninum foram 1:200 (8), 1:400 (14) e 1:800 (11). Animais soropositivos estavam presentes em 40% (10/25) das propriedades avaliadas. Os resultados demonstram ampla distribuição do protozoário entre os rebanhos bovinos de corte na região de Guarapuava, PR. Animais sem raça definida apresentaram maiores índices de soropositivos para N. caninum (P = 0,002). A idade apresentou associação positiva com a ocorrência de anticorpos contra N. caninum (P = 0,02), indicando que a transmissão horizontal tem importante papel na epidemiologia do coccídio.
Occurrence of anti-Neospora caninum antibodies in blood samples of 250 bovine beef cattle of the microrregion of Guarapuava, Paraná State was verified by Indirect Fluorescent Antibody Test (IFAT) (> 1:200) and correlated to age, sex and breed of animals. The statistical analysis was carried out through Fisher's Exact and qui-square tests (p < 0.05) to associate the results of the serology with the analyzed variables. From 250 evaluated samples, 33 (13.2 %) were positive for N. caninum. The titles obtained for N. caninum were 1:200 (8), 1:400 (14) and 1:800 (11). Seropositives animals were present in 40% (10/25) of the evaluated properties. These results demonstrate wide distribution of the protozoa among the beef cattle in the region of Guarapuava, PR. Animals without defined breed showed higher rates of seropositives for N. caninum (P = 0.002). The age had positive association with the incidence of antibodies against N. caninum (P = 0.02), indicating that horizontal transmission plays an important role in the epidemiology of this coccidia.

Medicinal plants are an important source of treatment for many ailments, although little is known of the potential genotoxic effects of most species. In the present study, two species from diverse and medicinally important genera - Maytenus rigida Mart., Celastraceae, and Aristolochia birostris Ducht, Aristolochiaceae - were analyzed to identify potentially significant secondary metabolites and the possible effects of their aqueous and alcoholic extracts on cell division in the onion root stem (genotoxicity test). The phytochemical testing revealed the presence of a number of potentially important secondary compounds in both species, including phenols, flavonoids, triterpenoids, steroids, and saponins. In the genotoxicity tests, no chromosomal abnormalities of any kind were observed in either species. In the case of M. rigida, a significant increase in mitotic activity was observed at the highest concentration. No significant tendency was recorded in A. birostris, although a considerable increase in the prophase was observed at all concentrations of the alcoholic extract. The triterpenoid content of both species may be especially important from a medicinal viewpoint, although recent findings on the carcinogenic potential of Aristolochia extracts demands caution in the interpretation of the results, and the need for further research.

RESUMO O objetivo deste trabalho foi realizar um estudo citomorfológico em plantas de arroz submetidas ao estresse salino (170 mM NaCl). Os efeitos da salinidade nas estruturas celulares foram analisados por meio de microscopia óptica e eletrônica de transmissão. Um processo de diferenciação precoce pôde ser observado na planta estressada, porém, o efeito deletério mais drástico foi encontrado nas membranas tilacóides dos cloroplastos. Estes resultados permitiram correlacionar o acúmulo de íons sódio, descritos na literatura, com os efeitos citomorfológicos do estresse salino em tecidos maduros de arroz.
ABSTRACT The goal of this work was to perform a cytomorphologic study of rice plants upon salt stress (170 mM NaCl). The effects of salinity on cell structures were analyzed by optical and electron transmission microscopy. An early differentiation process could be observed in stressed plants, however the most striking deleterious effect was found in the thylacoid membranes of chloroplasts. These results indicate a correlation between the previously described accumulation of sodium and the cytomorphological changes in chloroplasts of mature tissues of rice plants.

Bimetallic Pd-Pt films have been prepared on amorphous planar substrates by MOCVD using metal-acetylacetonate precursors. The effects of the MOCVD experimental conditions on the surface morphology, crystallite size and chemical composition of the bimetallic films have been analyzed. X-ray diffraction results show that bimetallic films consist of Pd-Pt solid solutions of variable composition depending on the initial metallic content in the inlet gases. Thus Pd-rich films can be obtained as preferred for the catalysts industry. The surface morphology of the bimetallic films varied from smooth to rough surfaces with the increase in total pressure. The crystallite size in the bimetallic films varies from 40 to 100 nm. Therefore, the MOCVD technique could be considered to be a viable alternative route for the preparation of bimetallic Pd-Pt films, which are in demand in the technology of supported bimetallic catalysts.
Se prepararon películas bimetálicas Pd-Pt sobre substratos amorfos por medio de la técnica MOCVD usando acetilacetonatos-metálicos como precursores. Se estudiaron los efectos de las condiciones experimentales MOCVD sobre la morfología superficial, tamaño de cristal y composición química de las películas bimetálicas. Los resultados de difracción de rayos X muestran que las películas bimetálicas consisten de soluciones sólidas Pd-Pt de composición variable dependiendo del contenido metálico inicial en el gas de alimentación. De esta forma, se pueden obtener películas ricas en paladio, las cuales son requeridas en la industria catalítica. La morfología superficial de las películas bimetálicas varía de una superficie suave a una superficie rugosa con el incremento de la presión total. El tamaño de cristal en las películas bimetálicas varía de 40 a 100 nm. Así, la técnica MOCVD puede ser considerada una ruta alterna viable para la preparación de películas bimetálicas Pd-Pt, las cuales tienen demanda en el desarrollo tecnológico de catalizadores bimetálitos soportados.

Adhesion molecule profiles of B-cell non-Hodgkin's lymphomas in the leukemic phase

We evaluated the expression of 10 adhesion molecules on peripheral blood tumor cells of 17 patients with chronic lymphocytic leukemia, 17 with mantle-cell lymphoma, and 13 with nodal or splenic marginal B-cell lymphoma, all in the leukemic phase and before the beginning of any therapy. The diagnosis of B-cell non-Hodgkin's lymphomas was based on cytological, histological, immunophenotypic, and molecular biology methods. The mean fluorescence intensity of the adhesion molecules in tumor cells was measured by flow cytometry of CD19-positive cells and differed amongst the types of lymphomas. Comparison of chronic lymphocytic leukemia and mantle-cell lymphoma showed that the former presented a higher expression of CD11c and CD49c, and a lower expression of CD11b and CD49d adhesion molecules. Comparison of chronic lymphocytic leukemia and marginal B-cell lymphoma showed that the former presented a higher expression of CD49c and a lower expression of CD11a, CD11b, CD18, CD49d, CD29, and CD54. Finally, comparison of mantle-cell lymphoma and marginal B-cell lymphoma showed that marginal B-cell lymphoma had a higher expression of CD11a, CD11c, CD18, CD29, and CD54. Thus, the CD49c/CD49d pair consistently demonstrated a distinct pattern of expression in chronic lymphocytic leukemia compared with mantle-cell lymphoma and marginal B-cell lymphoma, which could be helpful for the differential diagnosis. Moreover, the distinct profiles of adhesion molecules in these diseases may be responsible for their different capacities to invade the blood stream.

Acute promyelocytic leukemia (APL) is characterized by the expansion of blasts that resemble morphologically promyelocytes and harbor a chromosomal translocation involving the retinoic acid receptor a (RARa) and the promyelocytic leukemia (PML) genes on chromosomes 17 and 15, respectively. The expression of the PML/RARa fusion gene is essential for APL genesis. In fact, transgenic mice (TM) expressing PML/RARa develop a form of leukemia that mimics the hematological findings of human APL. Leukemia is diagnosed after a long latency (approximately 12 months) during which no hematological abnormality is detected in peripheral blood (pre-leukemic phase). In humans, immunophenotypic analysis of APL blasts revealed distinct features; however, the precise immunophenotype of leukemic cells in the TM model has not been established. Our aim was to characterize the expression of myeloid antigens by leukemic cells from hCG-PML/RARa TM. In this study, TM (N = 12) developed leukemia at the mean age of 13.1 months. Morphological analysis of bone marrow revealed an increase of the percentage of immature myeloid cells in leukemic TM compared to pre-leukemic TM and wild-type controls (48.63 ± 16.68, 10.83 ± 8.11, 7.4 ± 5.46%, respectively; P < 0.05). Flow cytometry analysis of bone marrow and spleen from leukemic TM identified the asynchronous co-expression of CD34, CD117, and CD11b. This abnormal phenotype was rarely detected prior to the diagnosis of leukemia and was present at similar frequencies in hematologically normal TM and wild-type controls of different ages. The present results demonstrate that, similarly to human APL, leukemic cells from hCG-PML/RARa TM present a specific immunophenotype.

The multidrug resistance P-glycoprotein is a transmembrane efflux pump expressed by lymphocytes and is involved in their cytolytic activity. In the present study, we investigated the age-related changes of P-glycoprotein function in normal peripheral blood lymphocytes. Blood samples from 90 normal volunteers (age range, 0 to 86 years) were analyzed. P-glycoprotein function was assessed by the flow cytometric rhodamine 123 assay. P-glycoprotein function was highest in cord blood and progressively declined with age in peripheral blood T CD4+ and CD8+ cells. In contrast, P-glycoprotein function did not vary with age in CD19+ B or CD16+CD56+ natural killer cells. These data suggest that the decline in P-glycoprotein function in T CD4+ and CD8+ lymphocytes as a function of age may contribute to the decrease in T cell cytolytic activity with aging.

Inotropic effects of extracts of Psidium guajava L. (guava) leaves on the guinea pig atrium

Many pharmacological effects have been ascribed to extracts of Psidium guajava L. (guava) leaves. However, in spite of its widespread use in Brazilian folk medicine and a reasonable number of scientific reports about it, we could not find any study dealing with its action on the mammalian myocardium. In the present study, by measuring isometric force, we observed that the crude extract of P. guajava (water-alcohol extract obtained by macerating dry leaves) depresses the guinea pig atrial contractility in a concentration-dependent fashion (N = 8 hearts, 15 trials). The compound with cardiac activity was concentrated by extraction in a Soxhlet apparatus using 17 M glacial acetic acid after removing the less polar fractions (hexane, chloroform, acetone, ethanol and methanol), suggesting that this compound is a highly polar substance. In the isolated guinea pig left atrium the acetic acid fraction (10-800 mg/l) of P. guajava 1) reversibly decreased myocardial force in a concentration-dependent fashion (EC50 = 0.07g/l, N = 5 hearts, 9 trials, P<0.05), 2) increased the atrial relaxation time measured at 20% of the force amplitude up to 35% (91 ± 15 to 123 ± 30 ms, N = 3 hearts, 6 trials, P<0.05), 3) abolished the positive staircase effect (Bowditch phenomenon) in a concentration-dependent fashion suggesting a decrease of the cellular inward calcium current (N = 4 hearts, 8 trials, P<0.05), and 4) its inotropic effect was abolished by cholinergic receptor blockade with 1.5 mM atropine sulfate, indicating a cholinergic involvement in the mechanism of action of the extract (N = 7 hearts, 15 trials, P<0.05). The acetic acid extract was 20 times more potent than crude extract (EC50 = 1.4 g/l). The results showed that extracts from P. guajava leaves depress myocardial inotropism.

The distinction between normal and leukemic bone marrow (BM) B-precursors is essential for the diagnosis and treatment monitoring of acute lymphoblastic leukemia (ALL). In order to evaluate the potential use of quantitative fluorescence cytometry (QFC) for this distinction, we studied 21 normal individuals and 40 patients with CD10+ ALL. We characterized the age-related changes of the CD10, CD19, TdT, CD34 and CD79a densities in normal and leukemic BM. Compared to normal adults, the B-precursors from normal children expressed significantly lower values of CD34-specific antibody binding capacity (SABC) (median value of 86.6 vs 160.2 arbitrary units (a.u.) in children and adults, respectively). No significant age-related difference was observed in the expression of the other markers in the normal BM, or in any of the markers in the leukemic BM. Based on the literature, we set the cut-off value for the normal CD10 expression at 45 x 10³ a.u. for both age groups. For the remaining markers we established the cut-off values based on the minimum-maximum values in the normal BM in each age group. The expression of CD10 was higher than the cut-off in 30 ALL cases and in 18 of them there was a concomitant aberrant expression of other markers. In 9 of the 10 CD10+ ALL with normal CD10 SABC values, the expression of at least one other marker was aberrant. In conclusion, the distinction between normal and leukemic cells by QFC was possible in 38/40 CD10+ ALL cases.

Surto alimentar por Salmonella Enteritidis no Noroeste do Estado de São Paulo, Brasil

Em 1993 ocorreu um surto alimentar em escola, com 211 afetados. Os dados epidemiológicos levantados por entrevista de amostragem de afetados e não afetados mostraram que os sintomas predominantes foram diarréia, febre (77,7%), dor abdominal (67,7%), vômito (65,8%), calafrios (54,5%) e cefaléia (44,5%). A mediana de incubação foi de 17 horas, com limites entre 3 e 29 horas. A duração da doença foi de 3 a 4 dias. O alimento consumido foi um tipo de patê, mistura de molho de maionese preparada com ovos crus com batata cozida, passado em pão. A análise de material biológico (3 coproculturas) e de restos de alimentos revelou a presença do mesmo microrganismo, a Salmonella Enteritidis. No caso dos alimentos, o número encontrado desta bactéria por gramo de produto era compatível com a quantidade de células necessária para desencadear a doença (10(4)e 10(5)/g). O antibiograma de todas as cepas isoladas revelou o mesmo padrão de sensibilidade. As falhas no preparo do alimento relacionadas com o levantamento indicam a possibilidade de contaminação endógena dos ovos; contaminação cruzada - o surto afetou três períodos escolares, sendo que para cada um o alimento foi preparado em separado - e as condições de manutenção do alimento após preparo e até o consumo. A observação por uma semana seguida das 3 merendeiras envolvidas, através de coprocultura, não indicaram que as mesmas fossem portadoras assintomáticas desta bactéria ou que tivessem sido envolvidas no surto em questão.
A foodborne outbreak which affected 211 persons occurred, in a School, in 1993. The epidemiological data obtained by interviewing the affected and non affected persons sampled showed as predominant symptoms: diarrhoea, fever (77.7%), abdominal cramps (67.7%), vomiting (65.8%), hot-and-cold sensations (54.5%) and headache (44.5%). The median incubation period was of 17 hours, the limits being 3 and 29 hours. The disease period was of from 3 to 4 days. The food concerned was a kind of paté, a mayonnaise mixture prepared with fresh eggs with boiled potatoes that was consumed with bread. The biological material analysis - 3 coprocultures, and leftovers of the food revealed the presence of one and the same organism: Salmonella Enteritidis. In the food, the numbers of this bacterium per gram were sufficient to account for the manifestation of the disease (104 and 105/g). The antibiogram of all the isolates showed the same sensibility pattern. The preparation related to this outbreak suggests the endogenous contamination of the eggs; the cross contamination - the outbreak affected three school periods, as the food was prepared separately for each school period; and the conditions under which the food was kept during the time from preparation to consumption. The observation of the 3 food handlers, by successive coprocultures, for one week, indicates that they were not asymptomatic carriers nor were they affected as a result of this outbreak by the causal bacteria.
