Feasibility of replacing the viral isolation technique in mice with RT-qPCR for the diagnosis of rabies RTqPCR RT qPCR

RESUMO: A raiva é uma encefalite viral que acomete os mamíferos, incluindo humanos. O diagnóstico laboratorial rápido e eficaz do vírus da raiva é importante para a saúde pública. Este estudo teve como objetivo avaliar a viabilidade operacional, técnica e financeira da RT-qPCR em substituição à inoculação em camundongos para o diagnóstico da raiva na rotina laboratorial. Foram analisadas 316 amostras de cérebros de mamíferos, 121 positivas e 195 negativas, previamente diagnosticadas por imunofluorescência direta (IFD) e isolamento viral em camundongos, por inoculação intracerebral em camundongos. As amostras foram submetidas à técnica duplex TaqMan RT-qPCR. A precisão, sensibilidade e especificidade da RT-qPCR foram analisadas. Os custos para realizar a técnica de RT-qPCR foram analisados e comparados com o custo do isolamento viral em camundongos. Os resultados demostraram 99,37% de acurácia, 99,17% de sensibilidade e 99,49% de especificidade pela RT-qPCR quando relacionada aos resultados de IFD e isolamento viral em camundongos, que se mostrou uma técnica robusta e com repetibilidade. O tempo mínimo para diagnóstico positivo foi reduzido na RT-qPCR (1 dia) quando comparado ao isolamento viral em camundongos (9,64 dias), e com redução de 17,7% no custo pela técnica molecular. O presente estudo demonstrou que a técnica de biologia molecular é uma ferramenta eficiente para o diagnóstico da raiva na rotina laboratorial, podendo substituir o isolamento viral em camundongos. ABSTRACT: Rabies is a viral encephalitis that affects mammals, including humans. Rapid and effective laboratory diagnosis of the rabies virus is critical for public health. This study evaluated the operational, technical, and financial viability of the RT-qPCR in replacement of inoculation in mice for diagnosing rabies in the laboratory routine. A total of 316 samples of mammalian brains were analyzed, 121 positives and 195 negatives, previously diagnosed by direct immunofluorescence (DIF) and mouse inoculation test (MIT). The samples were submitted to the duplex TaqMan RT-qPCR technique. The accuracy, sensitivity, and specificity of RT-qPCR were analyzed. We analyzed the costs for performing the RT-qPCR technique and compared it with the cost of MIT. The results showed 99.37% accuracy, 99.17% sensitivity, and 99.49% specificity by RT-qPCR when related to DIF and MIT results, which proved to be a robust and repeatable technique. The minimum time for a positive diagnosis was reduced in RT-qPCR (1 day) if compared to MIT (9.64 days), with a 17.7% reduction in the cost of the molecular technique. The present study demonstrated that the molecular biology technique is an efficient tool to diagnose rabies in the laboratory routine, being able to replace MIT. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract This study aimed to assess the combined antibacterial effect of lectins and antibiotics on S. aureus ATCC 25923, multidrug-resistant E. coli ATCC 11303 and S. aureus ATCC 700698. Using the checkerboard assay, we evaluated the antibacterial effects of eight lectins isolated from marine organisms combined with two common antibiotics, oxacillin and tetracycline, on three virulent bacterial strains. Initially, none of the tested lectins exhibited antibacterial effects when used individually. However, when combined with antibiotics, the lectins exhibited synergistic, additive, antagonistic, or no interaction. Overall, the tested lectins alone had no effect on the efficacy of oxacillin. On the other hand, different lectins in combination with tetracycline potentiated its antimicrobial effect. Lectins from red algae of the Bryothamnium genus, for example, exhibited the most significant synergistic effects, reducing the minimum inhibitory concentration (MIC) of tetracycline by up to 16 times. Lectins from the Hypnea genus also reduced the MIC of tetracycline. Our findings suggest that some lectins binding to complex carbohydrates containing fucosylated cores (α1-6) are excellent candidates to boost the efficacy of some antibiotics. RESUMO O uso da irrigação desempenha papel fundamental no avanço das fronteiras agrícolas e no aumento da produtividade nas áreas de cultivo, principalmente em ambientes semiáridos que apresentam alta variação espaço-temporal de chuvas. Essa prática é sugerida até mesmo para cultivos adaptados como palma forrageira (Opuntia sp.) e sorgo (Sorghum bicolor). Assim, objetivou-se avaliar a rentabilidade do uso da irrigação no sistema solteiro e sistema consorciado palma-sorgo no semiárido. O delineamento experimental foi em blocos casualizados em esquema fatorial 5 × 3: cinco lâminas de irrigação por gotejamento (0, 25, 50, 75 e 100% da evapotranspiração de referência - ET0) e três sistemas de cultivo (palma forrageira solteira, sorgo solteiro e consórcio palma-sorgo), com quatro repetições. O tratamento 0% ET0 correspondeu à condição de sequeiro com uma lâmina de 355 mm. Foi conduzido um ciclo de palma e dois de sorgo (planta e rebrota) compreendidos de novembro 2014 a novembro de 2015. Verificou-se que os maiores custos para implantação dos sistemas de cultivo em condições irrigadas se devem à aquisição do sistema de irrigação e insumos, representando 85,3% do custo operacional efetivo. O sistema sorgo solteiro e o sistema palma forrageira-sorgo proporcionaram as maiores produtividades de matéria fresca (62.013,05 e 60.075,36 kg ha-1), respectivamente. Com base em indicadores econômicos, a adoção de lâminas de irrigação (25, 50, 75 e 100% ET0) promoveu lucros para os sistemas (solteiro e consorciado) a partir do segundo ano de implantação.
ABSTRACT Irrigation plays a fundamental role in advancing agricultural frontiers and increasing crop productivity, especially in semi-arid environments, because they exhibit high spatiotemporal rainfall variation. This practice is suggested even for adapted crops, such as forage cactus (Opuntia sp.), sorghum (Sorghum bicolor), and single or intercropping systems. In this study, aimed to evaluate the profitability of using irrigation in a single and intercropping forage cactus-sorghum system in a semi-arid region. The experimental design was in randomized blocks in a 5 × 3 factorial arrangement: five drip irrigation depths (0, 25, 50, 75, and 100% of the reference evapotranspiration - ET0) and three cropping systems (single forage cactus, single sorghum, and intercropping forage cactus-sorghum), with four replicates. The 0% ET0 treatment corresponded to rainfed conditions at a depth of 355 mm. A cycle of forage cactus and two sorghum cultivars (plant and regrowth) was conducted between November 2014 and November 2015. It was verified that the highest costs for implantation of the crop systems in irrigated conditions are due to the acquisition of irrigation and input systems, representing 85.3% of the effective operational cost. The single sorghum and forage cactus-sorghum systems provided the highest fresh matter productivity (62,013.05 and 60,075.36 kg ha-1, respectively). Based on economic indicators, the adoption of irrigation depths (25, 50, 75, and 100% ET0) promoted profits for the systems (single and intercropping) from the second year of implementation.

ABSTRACT Variation in rainfall affects crops; therefore, agricultural practices become essential for forage production in semi-arid regions. This study aimed to evaluate the influence of different agricultural practices on phyllochron, structural characteristics of forage cactus and millet, and their relations with crop yield using the principal component analysis (PCA). The design was in randomized blocks, with six treatments: sole cropped cactus without straw mulching, sole cropped cactus with straw mulching, sole cropped millet without straw mulching, sole cropped millet with straw mulching, and cactus intercropping with millet, with and without straw mulching, each with four replicates. There were three cactus cycles and nine millet cycles (three cycles of cultivars BRS1501 and six of IPA Bulk-1-BF). Biometric parameters were evaluated monthly, while yield was determined after the crop harvest. Phyllochron was determined with the regression analysis. The PCA was applied to structural characteristics and yield. The systems adopted did not influence the structural characteristics of cactus and millet ( p > 0.05), except for cladode thickness, which increased with straw mulching. Phyllochron of the millet reduced when the crop was intercropped. The structural characteristics of cladode length, width, and thickness influenced crop yield, mainly in systems with straw mulching. Straw mulching and intercropping alter phyllochron of cactus and millet. Abstract In recent years, cold atmospheric plasma (CAP) is used for surface disinfection. However, little is known about its ability to improve biocompatibility of metallic surfaces when compared to thermal plasma methods. In this context, the study aimed to evaluate the response of human endothelial cells (Ea.hy926) on titanium surfaces treated by non-thermal plasma method and thermal plasma method under nitriding atmosphere. The wettability was characterized by the sessile drop method, the topography and roughness were evaluated by atomic force microscopy (AFM), and the microstructure by grazing angle X-ray diffraction (GIXRD). Endothelial cells were cultured and evaluated for morphology by scanning electron microscopy and viability by an MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay. CAP treatment reduced the contact angle of the Ti surface (13.43° ± 1.48; p<0.05), increasing hydrophilicity. Rz roughness was higher on the nitrided surface (220.44±20.30; p< 0.001) compared to the CAP treated surfaces (83.29 ± 11.61; p< 0.001) and polished (75.98 ±34.21a); p<0.001). RESUMO O Brasil se destaca no cenário internacional em relação a produção de celulose de fibra curta. Apesar da grande biodiversidade brasileira, essa produção está baseada em clones de espécies exóticas de Eucalyptus. Nesse contexto, pode haver grande potencial na avaliação de novas fontes de fibras da flora brasileira, incluindo a região amazônica. O presente estudo teve como objetivo avaliar o potencial tecnológico da madeira de Ochroma pyramidale (Malvaceae) para produção de celulose kraft. Indivíduos de quatro anos de idade foram colhidos em um plantio comercial para produção de serrados no Estado do Mato Grosso. Determinamos as propriedades químicas (teores de holocelulose, lignina Klason, lignina solúvel, extrativos e cinzas), físicas (massa específica e porosidade) e morfologia de fibras (comprimento e largura de fibras, diâmetro do lume, espessura e fração de parede, coeficiente de flexibilidade, índice de enfeltramento e de Runkel) da madeira. Na polpação kraft a carga alcalina aplicada variou entre 10 e 24%, com intervalo de 2%. A madeira de Ochroma pyramidale apresentou características favoráveis à produção de polpa celulósica, como dimensões apropriadas das fibras e baixos teores de lignina, extrativos e cinzas. Os teores de álcali ativo residual e pH do licor negro apresentaram relação crescente com a carga alcalina aplicada nos processos de polpação. O acréscimo da carga alcalina reduziu o rendimento em polpação, número kappa, teor de rejeitos e aumentou o teor de ácidos hexenurônicos.
ABSTRACT Brazil stands out in the international scenario in the production of short-fiber pulp. Despite the great Brazilian biodiversity, that production is based on exotic Eucalyptus clones. In this sense, there may be great potential in the assessment of new sources of fibers from the Brazilian flora, including the Amazon. The present study aimed to assess the technical potential of the wood of Ochroma pyramidale (Malvaceae) for the production of kraft pulp. Four-year-old trees were harvested from a commercial forest for lumber production in Mato Grosso state (Brazil). We determined the wood’s chemical compositions (holocellulose, Klason lignin, soluble lignin, extractives and ash contents), physical properties (density and porosity), and fiber morphology (fiber length, width and thickness, lumen diameter, wall fraction, coefficient of flexibility, and slenderness and Runkel ratios). The wood was subjected to pulping with an effective alkali charge ranging from 10 to 24%, with intervals of 2%. Ochroma pyramidale wood presented characteristics favorable to the production of cellulosic pulp, such as appropriate fiber dimensions and low lignin, extractives, and ash content. The amount of residual active alkali and pH of the black liquor were positively related to the increase of the alkali charge employed in the pulping process. The increase of alkali charge decreased the pulp yield, kappa number and waste content, and increased the hexenuronic acid concentration.

Resumo: O estudo da fadiga em ambiente subaquático é dificultado pois envolve diversas variáveis, muitas delas estão relacionadas à interação do material com o meio em que está submetido. Em testes de fadiga em ambiente aquoso foi constatado o aparecimento de bolhas que se desprendem da região da ponta da trinca durante o ensaio, porém, até o momento a literatura não havia relatado esse fenômeno. Buscando a repetibilidade, este trabalho apresenta parâmetros de ensaio que promovam o desprendimento de bolhas, facilitando seu estudo e compreensão. Corpos de prova do tipo C(T) fabricados em aço ASTM A36 foram ensaiados em ambiente subaquático sob uma variação de carga constante (ΔP) de 5kN e frequência de 30Hz. Os desprendimentos das bolhas durante os ensaios foram registrados com uma câmera de alta velocidade (captura de 1000fps). Os resultados indicam que as primeiras bolhas desprendidas durante o ensaio são menores e aumentam de volume com o aumento do comprimento da trinca, chegando a um diâmetro médio máximo e após isso ocorre diminuição e supressão do fenômeno ao se aproximar do final do ensaio. A ocorrência foi associada à fragilização por hidrogênio, com existência já comprovada nesse tipo de ensaio
Abstract: The study of fatigue in an underwater environment is difficult because it involves several variables, many of them are related to the interaction of the material with the environment in which it is submitted. In fatigue tests in an aqueous environment, the appearance of bubbles that detach from the crack tip region during the test was observed, however, until now the literature had not reported this phenomenon. Seeking repeatability, this work presents test parameters that promote the detachment of bubbles, facilitating their study and understanding. Type C(T) specimens made of ASTM A36 steel were tested in an underwater environment under a constant load variation (ΔP) of 5kN and frequency of 30Hz. The detachments of the bubbles during the tests were recorded with a high-speed camera (capture of 1000fps). The results indicate that the first bubbles released during the test are smaller and increase in volume with increasing crack length, reaching a maximum average diameter and after that, there is a decrease and suppression of the phenomenon when approaching the end of the test. The occurrence was associated with hydrogen embrittlement, with proven existence in this type of test

Abstract INTRODUCTION: We evaluated the epidemiological and clinical profile of reported cases of bee sting incidents in Santa Catarina, Brazil. METHODS: This retrospective cohort study included all reported cases of bee sting incidents among the population of Santa Catarina from 2007 to 2017. RESULTS: In total, 8,912 cases were reported, corresponding to an overall rate of 12.3/100,000 population. The mean age was 29,8 years with 60.2% men. The lethality rate was 0.2%. CONCLUSIONS: Santa Catarina has a high incidence rate of bee stings, which is higher than the national average. The data presented in this study may be underestimated.

RESUMO: Rochas sedimentares da Formação Sete Lagoas afloram na margem esquerda do Rio São Francisco, na região de Januária-Itacarambi, Minas Gerais, em seções relativamente contínuas, tanto lateralmente como no empilhamento vertical. Boas exposições associadas à preservação de estruturas/texturas primárias favorecem o estudo de fácies, e de estratigrafia sequencial. Este estudo estratigráfico foi realizado com a integração de dados de mapeamento geológico, petrografia macro e microscópica e de três perfis estratigráficos chaves, onde foram realizados levantamentos quimioestratigráficos (C e O) de alta resolução. Foram reconhecidas oito fácies e quatro associações de fácies distribuídas em uma sequência composta por um trato de sistema transgressivo na base, e um trato de sistema de mar alto na porção superior, separados por uma superfície de inundação máxima. O padrão de empilhamento do trato de mar alto apresenta uma progradação das fácies, do alto do embasamento, em direção a áreas onde o topo do embasamento é progressivamente mais profundo. Este arcabouço estratigráfico, associado com outras feições estratigráficas e isotópicas, indica que o alto de Januária atuou como um alto do embasamento durante o evento sedimentar da Formação Sete Lagoas. Estudos quimioestratigráficos e petrográficos permitiram caracterizar o carbonato basal da Formação Sete Lagoas como um carbonato de capa na base do trato transgressivo, onde ocorrem dolomitos e pseudomorfos de aragonita associados a valores de δ13C de até -5‰. Registros do fóssil Cloudina, recuperados no trato de mar alto, indicam uma idade associada ao Ediacarano Superior para a porção superior da sequência estratigráfica estudada.
ABSTRACT: Sedimentary rocks of the Sete Lagoas Formation, exposed in the left margin of the São Francisco river (Minas Gerais State, Brazil), were deposited on the Januária-Itacarambi basement high. They show both lateral and vertical rock stacking along continuous outcrops, allowing us to carry out detailed facies analysis and sequence stratigraphy studies. Our studies also integrate data from geological mapping, macro and microscopic petrography and high-resolution C and O isotope analysis. Eight facies and four facies associations make up a sequence composed by a transgressive tract in the base, and a high stand tract in the upper portion, separated by a maximum flooding surface. The high stand tract shows a progradation stacking from the basement high apex towards progressively deeper basement areas. This stratigraphic framework, associated with others stratigraphic and isotopic features, indicates that the now exposed Januária basement high also represents a paleo-high during the sedimentation event. Aragonite pseudomorphs and dolomites coupled with δ13C values of -5‰ characterize the basal carbonate of the transgressive tract as a cap carbonate. Records of the Cloudina fossil, recognized in the high stand tract, indicate a Late Ediacaran age for the upper portion of the studied stratigraphic sequence.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Effect of breed and corpus luteum on pregnancy rate of bovine embryo recipients

The objective of this study was to evaluate pregnancy rates of recipients of different breed groups (Nellore and crossbreed), as well as the effects of size and type of the corpus luteum (CL) on plasmatic concentrations of progesterone and pregnancy rates of embryo recipients. A total of 152 heifers were synchronized with progesterone implants and on the day of embryo transfer, previously obtained by superovulation and frozen in ethylene glycol, the diameter and type of the corpus luteum (cavitary and compact) was measured and blood was collected for progesterone measurement. The pregnancy rate was 44.1%, with a diameter of corpus luteum higher in recipients that became pregnant (2.03±0.41) compared with non-pregnant ones (1.86±0.34 cm). Plasmatic concentrations of progesterone did not differ between pregnant (1.50±1.05) and non-pregnant (1.31±0.91 ng/mL) animals. The type of corpus luteum did not influence the pregnancy rates. Only Angus and crossbred Marchigiana differ among themselves in pregnancy rates (33.3 and 59.2%, respectively). The pregnancy probability was affected only by CL diameter, but not by P4 plasmatic concentration. Selection of the corpus luteum size at the time of embryo transfer is an important factor to increase pregnancy rates in recipients, and compact and cavitary corpora lutea do not influence the pregnancy rates of bovine embryo recipients. Nellore recipients have pregnancy rates that are satisfactory and comparable to crossbred (Bos taurus × Bos indicus) recipients.
