This work includes the synthesis of phenanthrenequinone guanylhydrazone and phenanthro[9,10-e][1,2,4]triazin-3-amine to be tested as intercalate with DNA for treatment of cancer. The other synthesized product, 2-[(4-chlorophenylamino)methylene]malononitrile, was designed for future determination of its activity against leishmaniasis. A common problem about some articles on the literature is that some previously published compounds display error of their molecular structures. In this article it is shown the application of several procedures of nuclear magnetic resonance (NMR) to determine the complete molecular structure and the non questionable chemical shift assignment of the synthesized compounds, and also their analysis by molecular modeling to confirm the NMR results. To determine the capacity of pharmacological compounds to interact with biological targets is determined by docking. This work is to motivate the application of NMR and molecular modeling on organic synthesis, being a process that is very important for the study of the prepared compounds as interactions with biological targets by NMR.

The isoniazid-derived hydrazones 2-pyridinecarbaldehyde- (HPCIH), 2-acetylpyridine- (HAPIH), 2-benzoylpyridine- (HBPIH), 2-pyridineformamide- (HPAmIH) and 2-pyrazineformamide- (HPzAmIH) isonicotinoyl hydrazones, as well as the pyrazinamide-derived thiosemicarbazones 2-pyrazineformamide- (HPzAm4DH), N(4)-methyl-2-pyrazineformamide- (HPzAm4M), N(4)-ethyl-2-pyrazineformamide- (HPzAm4E) and N(4)-phenyl-2-pyrazineformamide- (HPzAm4Ph) thiosemicarbazones, were assayed for their action against intracellular amastigote form of Leishmania (Viannia) braziliensis strains and the glioblastoma multiforme (SF-295), colon adenocarcinoma (HCT-116), ovarian cancer (OVCAR-8) and acute myeloid leukemia (HL-60) human tumor cell lines. All compounds exhibited leishmanicidal effects, with concentrations at which 50% of parasites were inhibited (IC50) in the 10.70-18.84 µM range. Moreover, the compounds were up to 30-fold less toxic to macrophages than to the parasites. Pyrazinamide-derived thiosemicarbazones proved to have poor activity against the tumor cell lines at 5 µg mL-1, whereas, in general the isoniazid-derived hydrazones presented good activity, being HAPIH and HBPIH the most potent compounds (IC50 = 0.42-1.5 µM).

We report an alternative method for the synthesis of 2-(1,3-dithian-2-ylidene)-acetonitrile using 3-(4-chlorophenyl)-3-oxopropanenitrile and carbon disulfide as starting materials. The methanolysis of the intermediate 3-(4-chlorophenyl)-2-(1,3-dithian-2-ylidene)-3-oxopropanenitrile occurs via three possible intermediates, leading to the formation of the product at a 75% overall yield. Molecular modeling simulation of the reaction pathway using B3LYP 6-311G++(2df,2p) justified the proposed reaction mechanism.

2,2'-arilmetileno bis(3-hidróxi-5,5'-dimetil-2-ciclo-hexano-1-onas) com grupos para e orto no anel benzeno foram preparados e estudados por ressonância magnética nuclear (NMR) e modelagem molecular para determinar modificações conformacionais. Os resultados teóricos e experimentais mostraram que a interconversão conformacional nessas moléculas acontece por rotação do anel benzeno e leve movimentação dos anéis de dimedona, levando a variação do comprimento das ligações de hidrogênio intramoleculares. A presença de um na posição orto do anel benzeno modifica a interconversão conformacional, levando ao desaparecimento de uma ligação de hidrogênio intramolecular e sobreposição de diversos sinais de RMN. A correlação dos valores de σp com deslocamentos químicos, ângulos e cargas atômicas confirma que as propriedades eletrônicas dos grupos R-para estão envolvidas nas mudanças conformacionais e variação dos deslocamentos químicos. Esses resultados serão aplicados para o estudo da interação desses compostos com biomoléculas e no seu uso como materiais de partida no planejamento e síntese de novos agentes bioativos.

2,2'-arylmethylene bis(3-hydroxy-5,5-dimethyl-2-cyclohexene-1-ones) with para and ortho-R groups on the benzene ring were prepared and studied by nuclear magnetic resonance (NMR) and molecular modeling to determine their conformational exchanges. Experimental and calculated results indicated conformational interconversions in these compounds by rotation of benzene ring and slow movement of dimedone rings, leading to intramolecular hydrogen bond length variation. The presence of one R group at the ortho position on the benzene ring modifies conformational exchange, leading to disappearance of one intramolecular hydrogen bond and superposition of diverse NMR signals. The correlation of σp values with chemical shifts, angles and atomic charges confirms that para-R groups electronic properties are involved in conformational exchange and chemical shift variance. These results will be used to study the interaction of these compounds with bio-molecules and their use as starting materials for design and synthesis of new bioactive agents.

A elucidação das rotas de reativação da acetilcolinesterase humana (HuAChE) inibida por organofosforados é de crucial importância para o desenvolvimento de antídotos eficientes contra a intoxicação causada por agentes de guerra química. Com o objetivo de contribuir para uma melhor compreensão do mecanismo de reativação, foram aplicadas neste trabalho simulações de dinâmica molecular (DM) clássica para estudar as interações entre a pralidoxima e aminoácidos do sítio ativo da HuAChE inibida pelo agente neurotóxico tabun. Além disso, métodos híbridos de mecânica quântica/mecânica molecular (QM/MM) foram usados para propor um mecanismo de reativação para a enzima inibida. Os resultados mostraram que a DM clássica manteve a pralidoxima no interior do sítio ativo da enzima, em uma região favorável à ocorrência de uma possível reação de desfosforilação, que foi confirmada por métodos de QM/MM, levando à proposta de um mecanismo de reativação, energeticamente favorável.

The elucidation of the reactivation routes of human acetylcholinesterase (HuAChE) inhibited by organophosphorous compounds is of crucial importance to the development of efficient antidotes against poisoning by chemical warfare agents. In order to contribute to a better understanding of the reactivation mechanism, we applied, in this work, classical molecular dynamics (MD) simulations to study the interactions between pralidoxime and the active site's amino acids of HuAChE inhibited by the neurotoxic agent tabun. Further, quantum mechanical/molecular mechanical (QM/MM) hybrid methods were used to propose a reactivation mechanism for the inhibited enzyme. The results showed that the classic MD kept pralidoxime inside the enzyme's active site, in a favorable region to the occurrence of possible reactions of dephosphorilation, which were confirmed by QM/MM methods, and lead to the proposition of an energetically favorable mechanism of reactivation.

Neste trabalho, após validação por comparação com resultados obtidos por DFT, os métodos semi-empíricos PM6 e RM1 foram utilizados para análise conformacional de três oximas usadas em defesa química. Os resultados sugerem conformações de menor energia destes compostos que poderão ser utilizadas em futuras parametrizações para estudos por modelagem molecular.

In this work, after validation by comparison with results obtained by DFT, the semi-empirical methods RM1 and PM6 were employed to perform conformational analysis of three oximes employed in chemical defense. The results suggested low energy conformations for those compounds that could be useful in further parameterizations for molecular modeling studies.

Os agentes de guerra química constituem uma das maiores ameaças do mundo moderno. Dentre estes, destacam-se os agentes neurotóxicos, em virtude de sua alta letalidade e periculosidade. Eles são compostos organofosforados que atuam pela inibição da enzima acetilcolinesterase, a qual é fundamental no processo de transmissão de impulsos nervosos. Existem várias formas de tratamento para a intoxicação por organofosforados, mas nenhuma delas é eficaz contra todos os agentes conhecidos ou contra todos os seus efeitos. Esta revisão tem como foco o uso de compostos organofosforados como agentes neurotóxicos de guerra química. Após uma breve introdução histórica, será feita uma discussão sobre as principais características estruturais e biológicas da acetilcolinesterase, seguida por uma revisão das propriedades dos compostos organofosforados e da sua aplicação como agentes de guerra química. Por fim, serão discutidas as formas de tratamento contra estes agentes, com ênfase nas oximas usadas para reativar a acetilcolinesterase inibida.

Chemical warfare agents constitute one of the greatest threats in the modern world. Among them, the neurotoxic agents are of special interest due to their high lethality and danger. Neurotoxic agents are organophosphorus compounds that act by inhibiting the enzyme acetylcholinesterase, which is fundamental for the control of transmission of nervous impulses. There are several ways of treating intoxication by organophosphorus compounds, but none of them is efficient against all the known neurotoxic agents or against all of their effects. This review focus on the use of organophosphorus compounds as neurotoxic chemical warfare agents. After a brief historical introduction, it will be done a discussion about the structural and biological characteristics of acetylcholinesterase, followed by a review of the properties of organophosphorus compounds and their application as chemical warfare agents. Finally, the ways of treatment against intoxication with these agents will be discussed, with emphasis on the oximes used for reactivating the inhibited acetylcholinesterase.

Neste trabalho é proposto o primeiro modelo por homologia para a nucleosídeo hidrolase de Leishmania donovani construído a partir das estruturas das nucleosídeo hidrolases de Crithidia fasciculata e de Leishmania major. Usando as informações de interação entre o inibidor p-aminofeniliminoribitol e a nucleosídeo hidrolase de Crithidia fasciculata foram planejados dois novos potenciais inibidores, os quais apresentam novas interações com alguns resíduos da bolsa hidrofóbica do sítio ativo do modelo. Simulações por dinâmica molecular dos protótipos ancorados nos sítios ativos do modelo e das enzimas usadas como moldes, mostraram que, diferente do p-aminofeniliminoribitol, eles permaneceram ancorados nos sítios ativos das três enzimas ao longo de toda a dinâmica, interagindo fortemente com os aminoácidos da bolsa hidrofóbica.

In this work we propose the first homology model for nucleoside hydrolase from Leishmania donovani, built based on the crystallographic structures of Crithidia fasciculata and Leishmania major nucleoside hydrolases. We used the interaction information from the crystallographic model of the enzyme of C. fasciculata in complex with the inhibitor p-aminophenyliminoribitol, to design two new potential inhibitors, which present new interactions with some residues of the hydrophobic pocket of the model active site. Molecular dynamics simulations of the prototypes inside the active sites of the model and the template enzymes showed that, differently from p-aminophenyliminoribitol, they remained tightly bound inside the active sites, interacting strongly with the amino acids from the hydrophobic pocket.

Malaria is still a very serious worldwide public health problem affecting between 300 and 500 million people and causing from 1.0 to 2.5 million deaths annually. The major problems with this disease are the lack of efficient vaccines and the rapid emergence of chemotherapy resistant P. falciparum strains. In this work we present a synopsis and discussion on malaria to provide the readers with some information on the historical aspects and relevance of this disease as well as the chemotherapy in use today.

A Serina Hidroximetiltransferase de Plasmodium falciparum nunca foi vista como alvo para a quimioterapia antimalarial, possivelmente devido a sua grande similaridade seqüencial com a enzima humana. Esta similaridade sugere que este parasita pode ser incapaz de mutar esta enzima para desenvolver resistência à quimioterapia. Neste trabalho, diferenças observadas no comportamento dinâmico dos sítios ativos da estrutura cristalográfica da enzima humana e um modelo, por homologia, da enzima do parasita, ambos complexados com glicina, como N-glicina-[3-hidróxi-2-metil-5-fosfonooximetil-piridin-4-il-metano], e ácido 5-formil-6-hidrofólico, foram utilizadas para o planejamento de inibidores seletivos desta enzima como novos potenciais antimalariais. Estes potenciais inibidores são derivados do ácido 5-formil-6-hidrofólico com diferentes cargas e comprimentos de cauda. Simulações por dinâmica molecular e estimativas das energias de interação dos compostos nos sítios ativos de ambas as enzimas mostraram que os compostos com carga líquida negativa e caudas curtas ou caudas longas e anfóteras, seriam potencialmente mais seletivos em relação à enzima do parasita.

Plasmodium falciparum Serine Hydroxymethyltransferase has never been used as target for antimalarial chemotherapy, possibly because its great sequence similarity with the human enzyme. This similarity suggests implies that P. falciparum may not be able to mutate this enzyme to develop resistance for chemotherapy. In this work, we have used differences on the dynamic behavior of the active sites of the crystallographic structure of the human enzyme and a homology model of parasite's enzyme, both in complex with glycine, as N-glycine-[3-hydroxy-2-methyl-5-phosphonooxymethyl-pyridin-4-yl-methane] and 5-formyl-6-hydrofolic acid, to design prototypes for selective inhibitors of this enzyme as new potential antimalarials. Those potential inhibitors are 5-formyl-6-hydrofolic acid derivatives with different charges and tail lengths. Molecular dynamics simulations and interaction energy estimates of the compounds within the active sites of both enzymes showed that compounds with a negative net charge and either shorter tails or longer amphoteric tails, would be more selective towards pfSHMT.

Reativadores eficientes de Aceticolinesterase são fundamentais para o desenvolvimento de antídotos contra o envenenamento por pesticidas neurotóxicos e agentes de guerra química. Todavia, o mecanismo da reação de reativação e as características estruturais dos reativadores conhecidos são pouco compreendidos. Com o objetivo de estudar o comportamento dinâmico e o efeito da carga líquida do antídoto na reativação desta enzima, foi conduzido um estudo por dinâmica molecular da acetilcolinesterase humana inibida por tabun em complexo com o antídoto pralidoxima e com seu análogo deazapralidoxima nas formas neutra e aniônica. Os resultados mostraram que a carga positiva da pralidoxima é importrante para sua admissão e permanência dentro do sítio ativo. Além disso, os análogos, diferente da pralidoxima, quando colocados dentro do sítio ativo, se distanciam do resíduo serina fosforilado da enzima e são repelidos pelo potencial eletrostático na entrada do canal que conduz ao sítio ativo.

Efficient acetylcholinesterase reactivators are fundamental for the development of antidotes against poisoning by neurotoxic pesticides and chemical warfare agents. However, the mechanism of the reactivation reaction and the structural characteristics of the known reactivators are poorly understood. In order to study the dynamic behavior and the effect of the antidote net charge in the reactivation of this enzyme, we carried out a molecular dynamics study of human acetylcholinesterase inhibited by tabun in complex with the antidote pralidoxime and with its deaza analogues in the neutral and anionic forms. Results show that the positive charge of pralidoxime is important for its admission and permanence inside the active site. Also, the analogues, unlike pralidoxime, when forced inside the active site, move away from the phosphorilated serine residue of the enzyme and are repelled by the electrostatic potential at the entrance of the channel that conducts to the active site.

É proposto um modelo teórico para a pfDHFR-TS que inclui os 55 aminoácidos que não foram contemplados no modelo cristalográfico. O cálculo do potencial eletrostático sobre a superfície do modelo, revelou uma região contínua de potencial positivo conectando os dois sítios ativos, sugerindo um mecanismo otimizado de transporte de dihidrofolato.

We propose a theoretical model for pfDHFR-TS, which includes the 55 aminoacid residues ignored in the crystallographic model. The electrostatic potential calculation on the model surface revealed a continuous positive potential region between the two active sites, suggesting an optimized mechanism for dihydrofolate transport.

Cerca de 40% da população mundial está exposta à malária, o que resulta na morte de mais de dois milhões de pessoas por ano na África, América Latina, Ásia Meridional e Oceania. A forma mais grave de malária em humanos é causada pelo protozoário Plasmodium falciparum. A quimioterapia ainda é uma das principais estratégias de controle deste parasita. Muitos dos antimalariais atuam inibindo a enzima di-hidrofolato redutase (DHFR), o que resulta na morte do protozoário. Contudo, o desenvolvimento de resistência tem reduzido a eficiência dos antifolatos como antimalariais. Este fenômeno foi relacionado à ocorrência de mutações na di-hidrofolato redutase do parasita. Este artigo faz uma revisão rapida de algumas características da malária falciparum, seguida de uma revisão extensiva da di-hidrofolato redutase do P. falciparum e das mutações relacionadas à resistência a antifolatos.

About 40% of the world population is exposed to malaria, which results in the death of over 2 million people per year in Africa, Latin America, Southern Asia and Oceania. The most severe type of malaria in humans is caused by the protozoan parasite Plasmodium falciparum. Chemotherapy is still one of the main control strategies for this parasite. Many of the antimalarials act by inhibiting the enzyme dihydrofolate reductase (DHFR), resulting in protozoan death. However, the development of drug resistance is reducing the efficiency of antifolates as antimalarials. This phenomenon has been linked to the occurrence of mutations in the dihydrofolate reductase of the parasite. This article includes a preliminary review of some of the features of falciparum malaria, followed by a more extensive review of the dihydrofolate reductase of P. falciparum and the mutations related to antifolate resistance.

A reação de aminas com ditiocicloexilidenos é uma reação geral para a síntese de diaminometilenobarbituratos. Quando diaminas do tipo H2N-(CH2)n-NH 2 são utilizadas como nucleófilos os produtos majoritários de reação são compostos bicíclicos, cujos rendimentos aumentam conforme n diminui, n = 2, 3, 4. Quando n = 6, obtém-se altos rendimentos (80-90%) de novos compostos macrocíclicos de 18 e 27 membros. As atribuições dos deslocamentos químicos de ¹H e 13C RMN de todos os produtos são discutidas.

The reaction of amines with dithiocyclohexylidenes is a general reaction for the synthesis of diaminomethylidene barbiturates. When diamines of the type H2N-(CH2)n-NH 2 are used as nucleophiles bicyclic compounds are the major reaction products, with yields increasing as n decreases, n = 2, 3, 4. When n = 6, macrocyclic compounds, novel 18 and 27 member rings, are obtained in very good yields. The ¹H and 13C NMR assignment for all products is discussed.
