ABSTRACT This study aimed to determine the consumption of Planococcus citri (Risso, 1813) (Hemiptera: Pseudococcidae) nymphs and the predator:prey ratio required for high predatory efficacy and survival of Chrysoperla externa (Hagen, 1861) (Neuroptera: Chrysopidae) larvae under different densities of prey, reared in rose bushes. Consumption by first-, second-, and third-instar larvae of C. externa was assessed. The predatory efficiency and survival of the predator were calculated for larvae in the second instar under densities of 20, 40, 80, 120, and 160 nymphs of P. citri and predator:prey ratios of 1:5, 1:10, and 1:20. The assessments were performed after 24 and 48 hours of exposure between species. There was increasing consumption throughout the larval development of C. externa. The predator:prey ratios 1:5 and 1:10 were the most efficient for P. citri population reduction, but the 1:5 ratio led to lower survival of larvae than the 1:10 and 1:20 ratios. There was a positive correlation between C. externa larval density and cannibalism (ρ = 0.45) and a negative correlation between predator density and P. citri population density (ρ = -0.81). Third-instar larvae consume more P. citri nymphs than second-instar larvae. Second-instar larvae of this lacewing released at a 1:20 ratio caused total mortality of mealybug nymphs within 48 hours, regardless the density of P. citri, with high larvae survival of C. externa. Differentials in the epidemiological profile of canine visceral leishmaniasis in the semi-arid region of Paraíba, Brazil

RESUMO: O objetivo desta pesquisa foi estimar a prevalência da leishmaniose visceral canina (LVC) e identificar as diferenças nos fatores relacionados à sua ocorrência nas zonas urbana e rural do município de Santa Luzia, localizado no semiárido paraibano. Nos anos de 2015 e 2016, coletaram-se 779 amostras de sangue de cães. A prevalência foi determinada através de três técnicas sorológicas, Kit ELISA-S7®, teste rápido DPP® e Kit EIE-LVC®, considerando positivas as amostras que reagiram em, pelo menos, dois ensaios. Os fatores relacionados foram determinados por meio das análises estatísticas uni e multivariada das respostas dos tutores ao questionário epidemiológico. A prevalência de anticorpos anti-Leishmania infantum encontrada no município estudado foi de 15,00% (117/779), sendo maior na zona urbana (15,20%) do que na rural (13,60%). O bairro que apresentou maior prevalência foi o Frei Damião com 26,40% (33/125), sendo considerado um hotspot (OR 1,245; p=0,007). Outros fatores relacionados encontrados foram a criação semidomiciliar (OR 1,798; p=0,025), na zorna urbana, e cão de caça (OR 18,505; p=0,016), contato com bovinos (OR 17,298; p=0,022) e ambiente onde o cão é criado (OR 4,802; p=0,024), na zona rural. Verifica-se a elevada prevalência da leishmaniose visceral canina e a sua ampla distribuição no município de Santa Luzia. Diferenças epidemiológicas entre as zonas urbana e rural puderam ser observadas, demonstrando a necessidade de medidas de controle mais adequadas para cada localidade e comprovando o processo de urbanização.
ABSTRACT: The objective of this study was to estimate the prevalence of canine visceral leishmaniasis (CVL) and to identify the differences in associated factors to its occurrence in urban area and countrysides in the municipality of Santa Luzia located in the semi-arid region of Paraíba. In the years 2015 and 2016, 779 blood samples from dogs were collected. The prevalence was determined by three serological techniques, ELISA-S7® Kit, DPP® Rapid Test and EIE-LVC® Kit, considering positive the samples that reacted in at least two assays. Associated factors were determined by univariate and multivariate analyzes of the guardians’ responses to the epidemiological questionnaire. The prevalence of anti-Leishmania infantum antibodies in the studied municipality was 15.00% (117/779), being higher in the urban area (15.20%) than in the countryside (13.60%). The neighborhood with the highest prevalence was Frei Damião with 26.40% (33/125), being considered a hotspot (OR 1.245, p=0.007). Other associated factors were the semi-domiciliary breeding (OR 1.798, p=0.025), in the urban area, and hunting dog (OR 18.505, p=0.016), contact with cattle (OR 17.298, p=0.022) and environment where the dog is raised (OR 4.802, p=0.024) in the countryside. In the municipality of Santa Luzia, the prevalence of canine visceral leishmaniasis is high and the disease is widely distributed. Epidemiological differences between urban area and the countryside could be observed demonstrating the need for more adequate control measures for each locality and proving the urbanization process.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Abstract This study aimed to evaluate decomposition and release of C and of N from the litter of forest Caatinga species in the humification phase, as well as to estimate the residence time of C and N. Litter decomposition of the species was evaluated in 2014/2015 in the municipality of Floresta, Pernambuco, Brazil, using a method of the litter bags at 270; 300; 330; and 360 day. Litter from species of the Fabaceae family presented different decomposition groups, suggesting that decomposition was more influenced by the C/N ratio than N concentration only. The N release was 6.25% greater than the release of C. C will remain in the litter for more time, while N will be almost totally released in approximately 1.14 years. The species litter decomposition were shared, suggesting that the nutrients cycling is fundamental in the preservation of the Caatinga, mainly due to the longer residence time of C.

Resumo A fenologia é muito importante para o entendimento dos ciclos das plantas, a repetição de eventos biológicos e as causas desses ciclos em relação a fatores abióticos e bióticos. Diante disto, objetivou-se com este trabalho avaliar a época de ocorrência e intensidade das fases fenológicas vegetativas e reprodutivas e suas correlações com fatores externos e a influência da radiação fotossinteticamente ativa na biometria de frutos de Campomanesia adamantium (Cambess.) O. Berg. O estudo foi conduzido em fazenda localizada no município de Rio Verde-GO, Brasil. A caracterização da fenologia foi feita em dez indivíduos adultos. As observações fenologia das fases fenológicas vegetativas foram feitas mensalmente e os eventos fenológicos, reprodutivos, semanalmente. Foram utilizados dois métodos para o registro dos eventos fenológicos: o percentual de intensidade de Fournier e o índice de atividade. Foram feitas também avaliações do número total e diâmetros transversal e longitudinal dos frutos de dois ramos por planta com maior e menor incidência de radiação. Verificou-se atividade de folhas jovens e adultas em Campomanesia adamantium em quase todo o período de estudo, exceto nos meses de maio e agosto, para folhas jovens. A presença de brotos também foi bastante intensa durante o período. Observou-se um ciclo de fenofases de reprodução rápido e intenso entre os meses de outubro e janeiro para C. adamantium. A incidência da radiação interferiu na dimensão e produção dos frutos. Frutos desenvolvidos em ramos de sombra tiveram número reduzido e valores biométricos inferiores em relação àqueles que cresceram em ramos de sol.
Abstract Phenology is very important to the understanding of plant cycles, the repetition of biological events, and the causes of these cycles in relation abiotic and biotic factors. The aim of this to evaluate the time of occurrence and intensity of the vegetative and reproductive phenological phases, their correlations with external factors, and the influence of photosynthetically active radiation in the fruit Campomanesia adamantium (Cambess.) O. Berg. The study was conducted on a farm in the municipality of Rio Verde, Brazil. The phenological characterisation was performed using ten adult plants. Phenological observations of vegetative and reproductive phenological phases were made monthly and weekly, respectively. The total number and the transverse and longitudinal diameters of the fruit of two branches per plant, with higher and lower incidence of radiation, were also evaluated. The young and adult leaves in Campomanesia adamantium were active for almost the entire study period, except the young leaves were inactive in the months of May and August. The presence of sprouts was also very high during the study period. There was a phenophase cycle of rapid and intense play between October and January for C. adamantium. The incidence of radiation interfered with the size and production of the fruit. Fruits that developed in the shadow of branches were fewer and their biometric values lower than those who developed in sunlight.

Abstract Candida infection is an important cause of morbidity and mortality in immunocompromised patients. The increase in its incidence has been associated with resistance to antimicrobial therapy and biofilm formation. The aim of this study was to evaluate the efficacy of tea tree oil (TTO) and its main component – terpinen-4-ol – against resistant Candida albicans strains (genotypes A and B) identified by molecular typing and against C. albicans ATCC 90028 and SC 5314 reference strains in planktonic and biofilm cultures. The minimum inhibitory concentration, minimum fungicidal concentration, and rate of biofilm development were used to evaluate antifungal activity. Results were obtained from analysis of the biofilm using the cell proliferation assay 2,3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) and confocal laser scanning microscopy (CLSM). Terpinen-4-ol and TTO inhibited C. albicans growth. CLSM confirmed that 17.92 mg/mL of TTO and 8.86 mg/mL of terpinen-4-ol applied for 60 s (rinse simulation) interfered with biofilm formation. Hence, this in vitro study revealed that natural substances such as TTO and terpinen-4-ol present promising results for the treatment of oral candidiasis.

Electrochemical methods are considered useful tools for simulations of biological redox reactions. The activities of quinones depend on their bioreduction. Biologically active pterocarpanquinones LQB-149 (nitroderivative), 150 and 151 (bromo and chloroderivatives, respectively) were electrochemically investigated by cyclic voltammetry, differential pulse voltammetry, and in situ UV-Vis spectroelectrochemistry, in aprotic media (N,N-dimethylformamide (DMF) + tetra-N-butylammonium (TBAPF6)). The data obtained regarding their reduction mechanisms, positive reactivity with oxygen and analysis of the electrogenerated intermediates were useful in explaining their biological outcomes. The appearance of bands at 397 and 480 nm, for the halogenated compounds, suggests the generation of transient quinonemethides (QM), electrophilic intermediates related to their activity. As an additional proof for the intermediacy of QM, in the redox processes, chemical reduction of LQB-150, in the presence of hexanethiol was performed and led to a thioalkylated quinone. For the nitroderivative, a broad band appeared at 432 nm, corresponding to the generation of the nitroradical anion, giving rise to a dianion diradical, after reduction at the second wave potential. Computational data correlate well with electrochemical experiments. Homogeneous electron transfer to oxygen, yielding reactive oxygen species, the generation of electrophilic species and the radical reactivity, explain partially the mechanism of biological action.

RESUMO: A conservação do haylage (alimento pré-seco) pode ser desafiadora, considerando o aumento do risco de crescimento de fungos, com consequente produção de micotoxinas. Entretanto, quando a qualidade da higiene e armazenamento é assegurada, o haylage aumenta a palatabilidade da forragem e fornece suplemento de matéria seca suficiente ao longo do ano. Devido à falta de informação relativa aos efeitos dessa alimentação nos parâmetros sanguíneos de equinos alimentados exclusivamente com essa dieta, o objetivo do presente estudo é avaliar o perfil bioquímico sanguíneo dos equinos após administração da haylage em comparação com feno. Doze matrizes Quarto de Milha foram distribuídas em dois grupos, cada um recebendo feno ou haylage de Tifton 85 (Cynodon spp.) por um período de 28 dias. O perfil bioquímico foi realizado em cinco tempos (T) diferentes (T0, antes do início do experimento e cronologicamente, a cada sete dias após o fornecimento das dietas - T1, T2, T3 e T4) para análise de proteína total (PT) e seu perfil fracionado, das enzimas alanina aminotransferase, aspartato aminotransferase, γ-glutamil-transferase, dos produtos de catabolismo creatinina e ureia e, dos íons cálcio e fósforo. Micotoxinas no haylage foram investigadas e mantiveram-se abaixo dos limites determinados pela legislação brasileira. O perfil bioquímico revelou, somente, elevação da PT em T4 no grupo que recebeu haylage, o que pode estar relacionado à sua maior digestibilidade proteica. Nenhuma diferença foi observada nos outros parâmetros estudados em ambos os grupos experimentais. Conclui-se que Haylage é comprovadamente seguro, quando bem preparado para equinos, sem causar efeitos na saúde geral, conforme demonstrado pelos exames bioquímicos no presente estudo.
ABSTRACT: The conservation of haylage (a pre-dried feed) can be challenging, since there is an increased risk of mould growth, which can contaminate this foodstuff with mycotoxins. However, when the hygienic quality is secured, haylage enhances grass palatability and provide enough supply of dry matter throughout the year. Due to the lack of information regarding its effect on blood parameters in horses fed exclusively with this foodstuff, the aim of this study is to provide information regarding its use in comparison to hay and ensure that it does not affect horses’ biochemical profile. Twelve Quarter Horse broodmares were distributed into two groups, each fed with Tifton-85 (Cynodon spp.) hay or haylage for a period of 28 days, and the biochemical profile was done in five different times (T0 before the experiment started and, chronologically, seven days apart - T1, T2, T3 and T4), It was analyzed total protein (TP) and its fractioning; enzymes alanine aminotransferase, aspartate aminotransferase and γ-glutamyl-transferase; endogenous catabolism products urea and creatinine; and ions calcium and phosphorus. Mycotoxins in haylage were also investigated and remained below the legislation thresholds. Only TP was higher in the last sampling (T4) of the haylage group, which may be related to the foodstuff’s higher protein digestibility. No differences were observed between serum enzymes, urea, creatinine and Ca/P from both experimental groups. Haylage has proven to be safe, when well prepared for horses, without causing impairing side effects, as shown by the normal serum biochemistry parameters presented in this study.

RESUMO: Existe vasta literatura sobre a desnervação intrínseca de segmentos do tubo digestório através da aplicação de CB na serosa da víscera. Entretanto, essa técnica tem a desvantagem de causar peritonite, formação de bridas e alta mortalidade, não sendo factível para eventuais utilizações em humanos. O objetivo do presente estudo foi avaliar a viabilidade do Cloreto de benzalcônio (CB) induzir desnervação química intrínseca, por meio de aplicações intramurais em íleo de ratos wistar, além de aprofundar o conhecimento sobre a evolução da lesão neuronal causada neste processo. Foram utilizados 40 ratos, distribuídos em dois grupos (controle- GC e benzalcônio- GB) de 20 animais cada, subdivididos em quatro subgrupos de acordo com o tempo de avaliação pós-operatória de 24, 48 horas, 30 e 90 dias. Os animais foram submetidos à microinjeções intramurais de solução salina estéril 0,9% (GC) ou de cloreto de benzalcônio (GB) em porção ileal, e posterior análise histopatológica e imuno-histoquímica, para avaliação da lesão neuronal. Houve diminuição significativa (p<0,05) na contagem neuronal mientérica ao longo do tempo nos grupos GB2, GB3 e GB4. A imunomarcação específica positiva para H2AX e Caspase-3 confirmou os resultados obtidos na avaliação histopatológica, denotando início da lesão celular irreversível em 24 horas, evoluindo para apoptose neuronal em 48 horas após a aplicação do CB 0,3%. Nas condições em que este trabalho foi conduzido, é possível concluir que a aplicação de CB 0,3% por meio de microinjeções intramurais na parede ileal é capaz de induzir desnervação química intrínseca da porção ileal de ratos wistar e que o principal mecanismo de morte neuronal é a indução de apoptose.
ABSTRACT: Extensive literature is available about the intrinsic denervation of segments of the digestive tube through the application of CB in the serosa of the viscera. However, this technique has some disadvantages like causing peritonitis, flanges and high mortality, limiting its use in humans. The aim of the present study was to evaluate the feasibility of benzalkonium chloride (CB) to induce intrinsic chemical denervation, through applications of CB in the intramural ileum of wistar rats, as well as deepen the knowledge about the evolution of neuronal injury caused in the process. We used 40 rats, divided into two groups (control-GC and benzalkonium-GB) of 20 animals each, divided into four sub-groups according to the time of postoperative assessment of 24, 48 hours, 30 and 90 days. The animals were submitted to intramural microinjections of sterile saline solution 0.9% (GC) or benzalkonium chloride (GB) in ileal portion, and subsequent histopathological analysis and immunohistochemistry for evaluation of neuronal injury. A significant decrease (p<0.05) was found of the neuronal myenteric count over time in groups, GB3, GB4 and GB2. The specific positive immunolabeling for H2AX and Caspase-3 confirmed the results obtained in the histopathological evaluation, denoting the ignition of irreversible cell injury in 24 hours, evolving into neuronal apoptosis in 48 hours after application of the CB 0.3%. Under the conditions in which this work was conducted, it can be concluded that the application of CB 0.3% by means of microinjections intramural in the ileal wall is able to induce intrinsic chemical denervation of the diverticulum of wistar rats and that the main mechanism of neuronal death is induction of apoptosis.

Electrochemical methods are powerful in the characterization and design of redox-modulating agents. We, herein, report the electrochemical investigation, in aprotic medium, of eleven synthetic 3-thio-substituted-nor-beta-lapachones, along with the determination of cytotoxic activity and correspondent selectivity index, against several cancer cell lines and one normal cell. Four of the quinones are novel compounds. The redox behavior is representative of two independent systems: the easy reduction of the quinone moiety and, at far more negative potential, the reductive cleavage of the C-S-C bonding; and the anodic part controlled by the oxidation of the sulfur moiety. The compounds have shown relevant cytotoxic activity, with emphasis on 3-phenyl-thio-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione (compound 2), which mechanism of molecular action was shown to be related to reactive oxygen species (ROS) release. Despite the absence of a linear correlation, there is a trend: the majority of the thionaphthoquinones, with values of first wave reduction potential, less negative than -0.65 V, were active. The less electrophilic compound (3-(cyclohexylthio)-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione, cyclohexyl derivative) is also the less cytotoxic toward cancer cells. Agents containing chalcogens and quinones can be used to attack entities with a disturbed redox balance.

ABSTRACT Anthracnose is a crop disease usually caused by fungi in the genus Colletotrichum or Gloeosporium. These are considered one of the main pathogens, causing significant economic losses, such as in peppers and guarana. The current forms of control include the use of resistant cultivars, sanitary pruning and fungicides. However, even with the use of some methods of controlling these cultures, the crops are not free of anthracnose. Additionally, excessive application of fungicides increases the resistance of pathogens to agrochemicals and cause harm to human health and the environment. In order to find natural antifungal agents against guarana anthracnose, endophytic fungi were isolated from Amazon guarana. The compounds piliformic acid and cytochalasin D were isolated by chromatographic techniques from two Xylaria spp., guided by assays with Colletotrichum gloeosporioides. The isolated compounds were identified by spectrometric techniques, as NMR and mass spectrometry. This is the first report that piliformic acid and cytochalasin D have antifungal activity against C. gloeosporioides with MIC 2.92 and 2.46 µmol mL-1 respectively. Captan and difenoconazole were included as positive controls (MIC 16.63 and 0.02 µmol mL-1, respectively). Thus, Xylaria species presented a biotechnological potential and production of different active compounds which might be promising against anthracnose disease.

Resumo: Feridas de grandes extensões, com perda da viabilidade tecidual e retardo na cicatrização por segunda intenção são casos que se faz necessário o emprego de técnicas cirúrgicas reconstrutivas. O plasma rico em plaquetas (PRP) é um produto com maior concentração plaquetária, adjuvante no processo cicatricial de cirurgias reconstrutivas, auxiliando nos processos de hemostasia e estimulação da angiogênese. Dessa forma, delineou-se um estudo a fim de avaliar a eficácia do uso do gel produzido a partir do plasma rico em plaquetas (PRP) em flapes de avanço de padrão axial toracodorsal em coelhos, para avaliar a possibilidade de favorecer a integração do retalho no leito receptor. Utilizaram-se 30 coelhos da raça Nova Zelândia branco, separados em dois grupos de 15 animais, compreendendo os grupos plasma rico em plaquetas (GPRP), na qual empregou-se o gel antes da síntese da ferida cirúrgica, e controle (GC), na qual utilizou-se apenas solução fisiológica. Para obtenção do PRP, coletou-se sangue dos animais, e determinou-se a contagem plaquetária antes da preparação do gel. No início e término do experimento os animais foram pesados para posterior análise de ganho peso médio. Após o procedimento cirúrgico iniciou-se as avaliações macroscópicas no 3º, 7º e 14º dia, e avaliou-se presença ou ausência de exsudato, integridade da pele, edema, rubor e necrose. Após esta etapa, coletou-se o material da ferida cirúrgica para confecção das lâminas histológicas e posterior avaliação microscópica. Avaliou-se a proliferação vascular, presença de células mononucleares e polimorfonucleares, proliferação fibroblástica, colagenização, reepitelização e hemorragia. Os dados obtidos foram submetidos à análise estatística (Teste t Student, t emparalhado, e Kruskall Walis, sendo p<0,05). O ganho de peso médio não foi significativo entre os grupos; a concentração plaquetária da amostra final do PRP foi significativamente maior quando comparada com a inicial; exsudato e necrose foram significativamente maior no grupo controle quando comparado ao grupo PRP; proliferação vascular e reepitelização foram significativamente maior no grupo PRP, enquanto que a presença de mononucleares, polimorfonucleares, proliferação de fibroblastos, colagenização e hemorragia não foram significativas entre os grupos. Obteve-se no terceiro dia, diferença significativamente maior quanto à variável exsudato no grupo controle quando comparado ao grupo PRP, no sétimo dia exsudato e rubor foram significativamente maior no grupo controle, e ao décimo quarto dia exsudato e integridade da pele foram significativamente maior no grupo controle. Os resultados obtidos neste estudo evidenciaram que a utilização do plasma rico em plaquetas na forma de gel em cirurgia reconstrutiva foi capaz de estimular a angiogênese na ferida favorecendo o processo de cicatrização.
Abstract: Wounds of large tracts with loss of tissue viability and delayed healing by secondary intention are cases where it is necessary the use of reconstructive surgical techniques. The platelet rich plasma (PRP) is a product with a higher platelet concentration, adjuvant in the healing process of reconstructive surgeries, assisting in the processes of hemostasis and stimulation of angiogenesis. Thus, a study was devised to assess the efficacy of the gel produced from the platelet rich plasma (PRP) in the forward axial flaps thoracodorsal pattern in rabbits to evaluate the ability to promote the integration of the flap the recipient bed. We used 30 white rabbits New Zealand, separated into two groups of 15 animals, comprising the platelet rich plasma groups (GPRP), in which we used the gel before the synthesis of the surgical wound , and control (CG), in which only saline was used. To obtain the PRP, the blood was collected from the animals, was determined and the platelet count before preparation of the gel. At the beginning and end of the experiment the animals were weighed for analysis of average weight gain. After surgery began macroscopic in the 3rd, 7th and 14th day reviews, and we assessed the presence or absence of exudate, skin integrity, edema, redness and necrosis. After this step, yielded the material for the manufacture of surgical wound, followed by microscopic histological slides. We evaluated vascular proliferation, presence of polymorphonuclear and mononuclear cells, fibroblast proliferation, collagen deposition, reepithelialization and hemorrhage. The data were subjected to statistical analysis (Student t test, t paired, and Kruskal Wallis test, and p<0.05). The average weight gain was not significant between the groups; platelet concentration in the final sample of PRP was significantly higher when compared with the original; exudate and necrosis were significantly higher in the control group compared to PRP group, vascular proliferation and reepithelialization were significantly greater in PRP group, whereas the presence of mononuclear cells, polymorphonuclear cells, proliferation of fibroblasts, collagen and hemorrhage were not significant between groups. Obtained on the third day, as the difference significantly greater exudate variable in the control group compared to the PRP group; on the seventh day exudate and flushing were significantly higher in the control group, and the fourteenth day exudate and integrity of the skin were significantly higher in the control group. The results of this study showed that the use of platelet rich plasma gel in the form of reconstructive surgery was able to stimulate angiogenesis in the wound favoring the healing process.

Resumo: O plasma rico em plaquetas (PRP) é um produto de fácil obtenção a baixo custo, destacando-se pelos seus fatores de crescimento na reparação tecidual. Para obtenção do PRP, a centrifugação do sangue total é realizada com tempos e forças gravitacionais específicas. Assim, o presente trabalho teve por objetivo estudar o método da dupla centrifugação para obtenção do PRP, a fim de avaliar a eficácia de aumento da concentração de plaquetas no produto final, a preparação de gel de PRP e otimizar o tempo de preparação da amostra final. Quinze coelhos Nova Zelândia Branco, fêmeas, foram submetidos à coleta de sangue para a preparação de PRP. As amostras foram separadas em dois tubos estéreis contendo citrato de sódio. Os tubos foram submetidos ao protoloco de dupla centrifugação, com a tampa fechada a 1600 revoluções por minuto (rpm) durante 10 minutos, resultando na separação dos glóbulos vermelhos, plaquetas e plasma contendo os leucócitos. Na sequência, foram destapados para pipetar o plasma e transferí-lo para outro tubo de estéril. O plasma foi novamente centrifugado a 2000pm durante 10 minutos, resultando em duas partes: a parte superior, que consistia em plasma pobre em plaquetas (PPP) e a parte inferior do botão de plaquetas. Parte PPP foi descartado de modo que apenas 1ml de PPP permaneceu no frasco juntamente com o botão de plaquetas. Este material foi agitado suavemente para promover a ressuspensão das plaquetas, o que resultou na produção de PRP. O protocolo de centrifugação dupla foi capaz de promover a concentração de plaquetas 3 vezes maior em relação à amostra de sangue inicial. O volume de gluconato de cálcio utilizado para a ativação das plaquetas foi de 0,3ml, e foi suficiente para coagular a amostra, e o tempo de coagulação variou de 8 a 20 minutos, com uma média de 17,6 minutos. O tempo da centrifugação do sangue até a obtenção do PRP gel levou apenas 40 minutos. Concluiu-se que o PRP foi obtido com sucesso pelo protocolo de centrifugação dupla, sendo capaz de aumentar a concentração de plaquetas na amostra em comparação com o sangue total, permitindo assim a utilização de PRP em procedimentos cirúrgicos. Além disso, o tempo de preparação foi adequado, permitindo a obtenção do PRP em apenas 40 minutos, e a utilização de gluconato de cálcio foi capaz de promover a ativação das plaquetas.
Abstract: Platelet-rich plasma (PRP) is a product easy and inxpesnsive, and stands out to for its growth factors in tissue repair. To obtain PRP, centrifugation of whole blood is made with specific time and gravitational forces. Thus, the present work aimed to study a method of double centrifugation to obtain PRP in order to evaluate the effective increase of platelet concentration in the final product, the preparation of PRP gel, and to optimize preparation time of the final sample. Fifteen female White New Zealand rabbits underwent blood sampling for the preparation of PRP. Samples were separated in two sterile tubes containing sodium citrate. Tubes were submitted to the double centrifugation protocol, with lid closed and 1600 revolutions per minute (rpm) for 10 minutes, resulting in the separation of red blood cells, plasma with platelets and leucocytes. After were opened and plasma was pipetted and transferred into another sterile tube. Plasma was centrifuged again at 2000rpm for 10 minutes; as a result it was split into two parts: on the top, consisting of platelet-poor plasma (PPP) and at the bottom of the platelet button. Part of the PPP was discarded so that only 1ml remained in the tube along with the platelet button. This material was gently agitated to promote platelets resuspension and activated when added 0.3ml of calcium gluconate, resulting in PRP gel. Double centrifugation protocol was able to make platelet concentration 3 times higher in relation to the initial blood sample. The volume of calcium gluconate used for platelet activation was 0.3ml, and was sufficient to coagulate the sample. Coagulation time ranged from 8 to 20 minutes, with an average of 17.6 minutes. Therefore, time of blood centrifugation until to obtain PRP gel took only 40 minutes. It was concluded that PRP was successfully obtained by double centrifugation protocol, which is able to increase the platelet concentration in the sample compared with whole blood, allowing its use in surgical procedures. Furthermore, the preparation time is appropriate to obtain PRP in just 40 minutes, and calcium gluconate is able to promote the activation of platelets.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A formação de espécies paramagnéticas a partir da oncocalixona A em meio aprótico foi confirmada pela realização de experimentos eletroquímicos in situ em conjunto com ressonância de spin eletrônico (E-ESR). A alta deslocalização do radical gerado no primeiro potencial de redução está claramente evidenciada pelo acoplamento hiperfino do H-9 com a maior constante de acoplamento, além dos acoplamentos nas posições H-3 (próximo à quinona) e H-7 (distante do núcleo quinônico). Em meio prótico, além dos experimentos de dependência com o pH, oncocalixona A mostrou-se DNA-reativa por meio de experimentos eletroquímicos com sensores de DNA. Sua reação com N-acetilcisteína, com caracterização estrutural dos produtos de adição, evidencia sua habilidade como aceptor de Michael. Ambos os aspectos são importantes em termos de atividades biológicas/farmacológicas e indicam os presentes modelos como ferramentas importantes na avaliação de compostos biologicamente ativos.
The formation of paramagnetic species from oncocalyxone A in aprotic medium was confirmed by performing in situ electrochemical-electron spin resonance (E-ESR) experiments. The high delocalization of the radical generated at the first reduction potential is clearly evidenced by the hyperfine coupling of H-9 with the larger coupling constant, besides the couplings at the H-3 (close to quinone) and H-7 (far from the quinone nucleus) positions. In protic medium, together with pH dependence experiments, oncocalyxone A showed to be DNA-reactive through experiments with DNA sensors. Its reaction with N-acetylcysteine, with structural characterization of the addition products, proved its ability as Michael acceptor. Both aspects are important in terms of biological/pharmacological activities and indicate the present models as important tools in the screening of biologically active compounds.
