A Vancomicina (VCM) é indicada no combate à infecções por Gram-positivas, porém não é considerada um agente de primeira escolha por causa dos seus efeitos adversos. Acredita-se que o estresse oxidativo é o principal mecanismo responsável pela lesão endotelial e consequente toxicidade da VCM, que varia desde flebites à nefrotoxicidade. Ademais, as recomendações de doses, diluições, taxas e tipos de infusão ainda são controversos. O objetivo deste trabalho foi determinar o efeito de diferentes diluições de VCM na lesão endotelial, hepática e renal utilizando parâmetros bioquímicos e análise histopatológica. Ratos Wistar foram divididos aleatoriamente em seis grupos e submetidos à canulação da veia femoral para a administração de medicamentos. Os grupos controle receberam 0,9 ml de salina e os outros receberam VCM (10mg/Kg/dia) nas diluições de 5.0 e 10.0 mg/ml durante 3 e 7 dias. Homocisteína, PCR-us, AST, ALT, GGT, ureia, creatinina, licopeno, alfa-tocoferol, beta-caroteno e retinol foram analisados. Fragmentos de rins, fígado e veia femoral canulada foram coletados. Este estudo mostrou alterações na ALT, caracterizando hepatotoxicidade. Entretanto, as diluições dos medicamentos não foram capazes de evidenciar alterações nos outros parâmetros bioquímicos. Por outro lado, alterações histopatológicas foram observadas nos rins e endotélio. Mais estudos são necessários para a caracterização do efeito da VCM quanto à toxicidade nos rins e endotélio e ainda na busca de marcadores bioquímicos capazes de evidenciar tais alterações morfológicas.

It is believed that oxidative stress is the primary mechanism of endothelial injury and the consequent VCM toxicity, which varies from phlebitis to nephrotoxicity. Moreover, dose recommendations, dilution, rates and types of infusion are still controversial. The aim of this study was to determine the effect of different VCM dilutions in endothelial, liver and kidney injuries by biochemical parameters and histopathological analysis. Wistar rats were randomly divided into six groups and subjected to femoral vein cannulation for drug administration. Control groups received 0.9 ml of saline and the others received VCM (10mg/Kg/day) at dilutions of 5.0 and 10.0 mg/mL for 3 and 7 days. Homocysteine, hs-CRP, AST, ALT, GGT, urea, creatinine, lycopene, alpha-tocopherol, beta-carotene and retinol were analyzed. Kidney, liver and cannulated femoral vein fragments were collected.This study showed alterations in ALT which featured hepatotoxicity. However, drug dilutions were not able to show changes in other biochemical parameters. In contrast, kidney and endothelium pathological changes were observed. More studies are needed to characterize VCM induced kidney and endothelium toxicity and biochemical markers able to show such morphological modifications.

RESUMO OBJETIVOS: Os efeitos do teste ortostático sobre o sistema cardiovascular já foram investigados, mas não há dados sobre o comportamento dos índices geométricos de variabilidade da frequência cardíaca. Aqui, foram avaliados os efeitos do teste ortostático ativo nos índices geométricos de variabilidade da frequência cardíaca em mulheres. MÉTODO: O estudo foi realizado em 12 mulheres saudáveis com idades entre 18 e 30 anos. Para a análise da variabilidade da frequência cardíaca, este parâmetro foi registrado batimento a batimento. Os intervalos RR foram transformados em figuras geométricas e, a partir destas, foi calculado o índice triangular, a interpolação triangular dos intervalos RR, o SD1, SD2 e a relacão SD1/SD2, que foram obtidos a partir da plotagem de Poincaré. A análise visual do gráfico foi também realizada. Para o teste, os participantes levantaram-se rapidamente de uma posicão sentada (em 3 segundos ou menos) e permaneceram imóveis, de pé, por 15 minutos. Amostras de variacão de frequência cardíaca foram coletadas em quatro momentos: repouso, 0-5 minutos, 5-10 minutos e 10-15 minutos a pé. RESULTADOS: O índice triangular, a interpolacão triangular dos intervalos RR, o SD1 e SD2 índices apresentaram-se reduzidos (p < 0,05) no intervalo 10-15 minutos depois de os voluntários se levantarem. A relacão SD1/SD2 apresentou-se reduzida durante o intervalo 0-15 minutos depois dos participantes passarem à posicão ortostática. CONCLUSÃO: Os índices geométricos de variabilidade da freqüência cardíaca diminuiram em resposta ao teste ortostático ativo em mulheres saudáveis.

OBJECTIVES: The effects of the orthostatic test on the cardiovascular system have been investigated, but there are no data on the behavior of the geometric indices of heart rate variability. We evaluated the effects of the active orthostatic test on the geometric indices of heart rate variability in women. METHOD: The study was performed on 12 healthy women between ages 18 and 30. For the analysis of heart rate variability, heart rate was recorded beat-to-beat. The RR intervals were transformed into geometric figures, and from these we calculated the triangular index, the triangular interpolation of RR intervals, the SD1, SD2 indexes and the SD1/SD2 ratio, which were obtained from the Poincare´ plot. Visual analysis of the plot was also performed. For the test, participants quickly stood up from a seated position in 3 seconds or less, and remained standing still for 15 minutes. Heart rate variability samples were collected at four moments: rest, 0-5 minutes, 5- 10 minutes and 10-15 minutes at standing position. RESULTS: The triangular index, the triangular interpolation of RR intervals, the SD1 and SD2 indices were reduced (p < 0.05) at 10-15 minutes after the volunteers stood up from seated position. The SD1/SD2 ratio was decreased at 0-15 minutes after the subjects changed from seated to orthostatic position. CONCLUSION: The geometric indices of heart rate variability decreased in response to the active orthostatic test in healthy women.

No adulto, a angiogênese é restrita a poucas condições fisiológicas. Ao contrário, a angiogênese descontrolada frequentemente está associada a patologias dependentes de angiogênese. Vários modelos animais foram descritos para ajudar na análise quantitativa da angiogênese in vivo e caracterizar moléculas pró e antiangiogênicas. Porém, ainda é necessário estabelecer um método quantitativo, reproduzível e específico para estudar os fatores pró e antiangiogênicos. Os objetivos deste trabalho foram padronizar um método para estudar angiogênese e investigar os efeitos da talidomida na angiogênese. Esponjas de 0,5 x 0,5 x 0,5 cm foram implantadas no dorso de camundongos controles e tratados (talidomida 200 mg/K/dia, por gavagem). Após sete dias, as esponjas foram retiradas. Avaliou-se a quantidade de hemoglobina na esponja e na circulação. A razão obtida entre esses valores foi testada por meio do teste não paramétrico Mann-Whitney. Os resultados mostraram que a angiogênese induzida pela esponja e quantificada por meio da razão hemoglobina esponja/circulação é um modelo útil para estudos in vivode angiogênese. Além disso, observou-se que a angiogênese induzida pela esponja diminui na presença de talidomida, corroborando para a validação do método padronizado.

In the adult organism, angiogenesis is restricted to a few physiological conditions. On the other hand, uncontrolled angiogenesis have often been associated to angiogenesis-dependent pathologies. A variety of animal models have been described to provide more quantitative analysis ofin vivo angiogenesis and to characterize pro- and antiangiogenic molecules. However, it is still necessary to establish a quantitative, reproducible and specific method for studies of angiogenesis factors and inhibitors. This work aimed to standardize a method for the study of angiogenesis and to investigate the effects of thalidomide on angiogenesis. Sponges of 0.5 x 0.5 x 0.5 cm were implanted in the back of mice groups, control and experimental (thalidomide 200 mg/K/day by gavage). After seven days, the sponges were removed. The dosage of hemoglobin in sponge and in circulation was performed and the ratio between the values was tested using nonparametric Mann-Whitney test. Results have shown that sponge-induced angiogenesis quantitated by ratio between hemoglobin content in serum and in sponge is a helpful model for in vivo studies on angiogenesis. Moreover, it was observed that sponge-induced angiogenesis can be suppressed by thalidomide, corroborating to the validity of the standardized method.
