The n-alkylcyclohexanes are poorly evaluated in biodegradation studies of Brazilian petroleum samples. In this context, this work aimed to use the homologue series of alkylcyclohexanes to assess extend of biodegradation of 26 petroleum samples from five Brazilian sedimentary basins, analyzed by gas chromatography coupled to mass spectrometry. The n-alkylcyclohexanes compounds were detected in all samples, with a distribution similar to the n-alkane profile, in which their abundance decreasing with increasing of the biodegradation extent. Based on this, it was suggested the use of the new parameter H30/DC together with the H30/AC already proposed, calculated by the ratio between C30 hopane to the dodecyl-cyclohexane, to assess biodegradation level according Peter and Moldowan scale (PM scale). In addition, it was observed an increasing in the relative abundance of DBE 2 from class O2, analyzed by ESI(-) FT-ICR MS, mainly due to the formation of monocyclic acids with biodegradation, which correlates with the decrease of n-alkylcyclohexanes, a possible precursor of these acids.

resumo Valsartana (VAL) foi submetida à degradação forçada em meio ácido conforme procedimento descrito no ICH. Os produtos de degradação (PDs) foram monitorados ao longo do tempo de degradação pela técnica de Cromatografia Líquida (LC) utilizando uma coluna Hypersil ODS (C18) (250 x 4,6 mm d.i., 5 µm). A detecção foi feita com dois detectores: espectrofotométrico (PDA) e espectrometria de massas (MS) por corrente iônica total. Ambas as técnicas falharam na identificação dos PDs obtidos ao longo do monitoramento, mostrando insignificantes variações na área do pico e permanecendo com pureza de pico ao longo de toda a eluição. Somente depois da avaliação por íon extraído (XIC), foi possível observar o aumento do íon m/z 306 e m/z 352 exatamente no mesmo tempo de retenção do íon molecular (m/z 436). Estes resultados mostram um caso simples e didático em que somente o uso de um método seletivo de LC-MS pode ser utilizado para monitorar produtos de degradação. Neste trabalho, é apresentado um caso real em que a separação por LC deve ser acoplada a métodos seletivos obtidos por MS, especialmente no estudo de PDs para novos fármacos, formulações e no desenvolvimento de métodos.

abstract Valsartan was submitted to forced degradation under acid hydrolysis condition as prescribed by the ICH. Degraded sample aliquots were separated via HPLC using a Hypersil ODS (C18) column (250 x 4.6 mm i.d., 5 µm). Either photodiode array (PDA) detection or mass spectrometry (MS) full scan monitoring of HPLC runs were used. HPLC-PDA failed to indicate Valsartan degradation under forced acid degradation, showing an insignificant peak area variation and that Valsartan apparently remained pure. HPLC-MS using electrospray ionization (ESI) and total ionic current (TIC) monitoring did not reveal any peak variation either, but inspection of the ESI mass spectra showed the appearance of m/z 306 and m/z 352 ions for the same retention time as that of Valsartan (m/z 436). These ions were identified as being protonated molecules of two co-eluting degradation products formed by hydrolysis. These assignments were confirmed by ESI-MS/MS with direct infusion of the degraded samples. The results showed that the use of selective HPLC-MS is essential for monitoring Valsartan degradation. Efficient HPLC separation coupled to selective and structural diagnostic MS monitoring seems therefore mandatory for comprehensive drug degradation studies, particularly for new drugs and formulations, and for method development.

Two methods were used to make crude preparations of surface-active compounds (SACs) produced by Gordonia amicalis grown on the medium containing 1% diesel oil. Using a 2:1 (v/v) solution of chloroform:methanol for extraction, Type I SACs were isolated and shown to produce oil in water (O/W) emulsions. Type II SACs were isolated by precipitation with ammonium sulfate and produced predominantly water in oil emulsions (W/O). The crude Type I and II preparations were able to produce a significant reduction in the surface tension of water; however, the crude Type II preparation had 10-25 fold higher emulsification activity than the Type I preparation. Both SAC preparations were analyzed by the TLC and each produced two distinct bands with Rf 0.44 and 0.62 and Rf 0.52 and 0.62, respectively. The partially purified SACs were characterized by the ESI(+)-MS, FT-IR and NMR. In each one of these fractions, a mixture of 10 oligomers was found consisting of a series of compounds, with masses from 502 to 899, differing in molecular mass by a repeating unit of 44 Daltons. The mass spectra of these compounds did not appear to match other known biosurfactants and could represent a novel class of these compounds.

Arrabidaea chica (H&B) Verlot is a plant popularly known as Pariri and this species is a known source of anthocyanins, flavonoids and tannins. This report describes an approach involving enzymatic treatment prior to extraction procedures to enhance A chica crude extract anticancer activity. Anticancer activity in human cancer cell lines in vitro using a 48 h SRB cell viability assay was performed to determine growth inhibition and cytotoxic properties. The final extraction yield without enzyme treatment was higher (24.28%) compared to the enzyme-treated material (19.03%), with an enhanced aglycones anthocyanin ratio as determined by HPLC- DAD and LC-MS with direct infusion.

A caracterização de amostras de vinho, sem pré-tratamento ou separação cromatográfica, por espectrometria de massas com ionização por eletrospray e infusão direta, em um processo denominado de fingerprinting, foi aplicado a várias amostras de vinho produzidas com uvas das variedades Pinot noir, Merlot e Cabernet Sauvignon, provenientes do Estado do Rio Grande do Sul, Brasil. Os fingerprints por ESI-MS das amostras detectaram mudanças que ocorreram durante o processo de envelhecimento para as três variedades de uvas. Análise de componentes principais (PCA) dos fingerprints em modo negativo foi usada para agrupar as amostras, salientar as principais modificações em composição e indicar os íons marcadores para cada grupo.

The characterization of wine samples by direct insertion electrospray ionization mass spectrometry (ESI-MS), without pre-treatment or chromatographic separation, in a process denominated fingerprinting, has been applied to several samples of wine produced with grapes of the Pinot noir, Merlot and Cabernet Sauvignon varieties from the state o Rio Grande do Sul, in Brazil. The ESI-MS fingerprints of the samples detected changes which occurred during the aging process in the three grape varieties. Principal Component Analysis (PCA) of the negative ion mode fingerprints was used to group the samples, pinpoint the main changes in their composition, and indicate marker ions for each group of samples.

As folhas de varias espécies de Passiflora são utilizadas como ansioliticas e sedativas. Passiflora alata Curtis, Passifloraceae consta em três edições da farmacopéia brasileira, porem não há muitos estudos sobre sua composição química. No passado, enfatizava-se a ação conjunta de alcalóides e flavonóides. Em trabalho anterior, não foi detectada a presença de alcalóides harmanicos através de CLAE. Assim, decidiu-se investigar a produção dos mesmos através de cultivo celular, introduzindo seu precursor metabólico L-triptofano. Os explantes foliares apresentaram resultados satisfatorios para germinação apos assepsia, e a formação de calo foi iniciada em meio MS com quantidades adequadas de fitohormonios, previamente determinadas. Sessenta dias após a inoculação os calos foram repicados para meio semi-solido com e sem L-triptofano (50, 100, 200 mg/L), mantidos por 90 dias em condições padrão. Amostras foram coletadas com 6, 16, 26, 36, e 90 dias, realizada extração acido-base e o extrato analisado por CLAE. Os resultados mostraram a ausência de harmana, harmina, harmol, harmalol e harmalina. Dois picos presentes nas amostras com L-triptofano foram coletados e analisados através de espectrometria de massas, electrospray modo positiva [ESI(+)-MS] e analise em tandem ESI(+)-MS/MS. Os espectros correspondentes foram similares, mostrando a perda consecutiva de 68 Da, atribuídos ao pirazol. Este fato aponta para uma transformação não enzimática, não relacionada a uma biossintese previamente descrita para alcalóides β-carbolínicos.

Preliminary work on Passiflora alata leaves failed to detect harmane alkaloids using LC. The aim of this work was to investigate the production of harmane alkaloids through the cell culture of P. alata, inducing its precursor (L-tryptophan). The leaf explants presented satisfactory results after disinfection, and the callus formation was initiated in MS media with adequate quantities of phytohormones. Sixty days after inoculation, calli were inoculated in the optimized semi-solid MS media, with and without the addition of L-tryptophan (50, 100, 200 mg/L) and kept in standard conditions for 90 days. Calli were collected on days 6, 16, 26, 36, and 90, followed by acid-base extraction, and analysed by LC. The results showed an absence of harmane, harmin, harmol, harmalol, and harmaline. With L-tryptophan feeding, two peaks were detected, collected and analysed through positive mode electrospray [ESI(+)-MS] and sequential analysis in tandem ESI(+)-MS/MS. The spectra obtained were very similar, with a repetition of the more intense ions, and consecutive loss of 68 Da units, attributed to the heterocycle pyrazole. It appeared that this transformation was not related to any enzymatic pathway previously described for the plant from L-tryptophan, and the biosynthesis of β-carboline alkaloids in callus culture of P. alata were not observed in this work.

CONTEXTO E OBJETIVO: Polimorfismos em genes do metabolismo do folato podem modular o risco materno para síndrome de Down (SD). Este estudo avaliou a influência do polimorfismo de deleção de 19 pares de base (pb) no íntron 1 do gene dihidrofolato redutase (DHFR) no risco materno para SD e investigou a associação entre esse polimorfismo e variações nas concentrações de folato sérico, homocisteína (Hcy) e ácido metilmalônico (MMA) plasmáticos. TIPO DE ESTUDO E LOCAL: Estudo transversal analítico realizado na Faculdade de Medicina de São José do Rio Preto (Famerp). MÉTODOS: 105 mães de indivíduos com trissomia livre do cromossomo 21 e 184 mães controles foram avaliadas. A análise molecular do polimorfismo foi realizada pela reação em cadeia da polimerase (PCR) por diferença de tamanho dos fragmentos. O folato foi quantificado por quimioluminescência, e Hcy e MMA foram determinados por cromatografia líquida/espectrometria de massas sequencial. RESULTADOS: Não houve diferença entre os grupos em relação às frequências alélica e genotípica (P = 0,44; P = 0,69, respectivamente). As concentrações de folato, Hcy e MMA não mostraram diferença significativa entre os genótipos, entre grupos (P > 0,05). CONCLUSÕES: O polimorfismo de deleção de 19 pb do gene DHFR não é um fator de risco materno para SD e não está relacionado com variações nas concentrações de folato sérico, Hcy e MMA plasmáticos na população estudada.

CONTEXT AND OBJECTIVE: Polymorphisms in genes involved in folate metabolism may modulate the maternal risk of Down syndrome (DS). This study evaluated the influence of a 19-base pair (bp) deletion polymorphism in intron-1 of the dihydrofolate reductase (DHFR) gene on the maternal risk of DS, and investigated the association between this polymorphism and variations in the concentrations of serum folate and plasma homocysteine (Hcy) and plasma methylmalonic acid (MMA). DESIGN AND SETTING: Analytical cross-sectional study carried out at Faculdade de Medicina de São José do Rio Preto (Famerp). METHODS: 105 mothers of individuals with free trisomy of chromosome 21, and 184 control mothers were evaluated. Molecular analysis on the polymorphism was performed using the polymerase chain reaction (PCR) through differences in the sizes of fragments. Folate was quantified by means of chemiluminescence, and Hcy and MMA by means of liquid chromatography and sequential mass spectrometry. RESULTS: There was no difference between the groups in relation to allele and genotype frequencies (P = 0.44; P = 0.69, respectively). The folate, Hcy and MMA concentrations did not differ significantly between the groups, in relation to genotypes (P > 0.05). CONCLUSIONS: The 19-bp deletion polymorphism of DHFR gene was not a maternal risk factor for DS and was not related to variations in the concentrations of serum folate and plasma Hcy and MMA in the study population.

Devido à importância das etapas de fermentação e secagem das sementes de cacau para as características sensoriais dos produtos finais obtidos, bem como para os teores de compostos fenólicos presentes, objetivou-se avaliar a influência do tempo de fermentação e do tipo de secagem de sementes de cacaueiro nos teores de compostos fenólicos, bem como nas características físicas, físico-químicas e sensoriais dos produtos obtidos. Os tempos de fermentação avaliados foram 3 e 7 dias e a secagem foi realizada naturalmente (ao sol) e artificialmente (estufa com circulação de ar a 35 °C). Os teores de compostos fenólicos totais, flavan-3-óis e procianidinas foram quantificados durante a fermentação e ao término da secagem. Constatou-se que grande parte dos compostos fenólicos foi perdida durante a fermentação, sendo que as amêndoas fermentadas por 3 dias apresentaram teores maiores dos compostos fenólicos avaliados. Contudo, observou-se nessa condição uma significativa perda na qualidade sensorial. A secagem natural das amêndoas fermentadas permitiu maior retenção de polifenóis e também melhores resultados sensoriais em relação à secagem artificial. A avaliação sensorial realizada com uma bebida formulada com o liquor obtido dos diferentes tratamentos demonstrou que a amostra fermentada durante 7 dias apresentou as melhores notas sensoriais, sendo que o produto seco naturalmente foi o mais bem avaliado em relação aos demais.

Due to the importance of the fermentation and drying stages in the sensory characteristics and in the phenolic content of cocoa products, the present study was carried out to evaluate the influence of the fermentation period and the type of drying of cocoa seeds, on the phenolic compounds content, as well as on the physical, physicochemical, and sensorial characteristics of the products. The periods of fermentation were 3 and 7 days and the fermented beans were dried naturally (sun drying) and artificially (under air circulation at 35 °C). The total phenolic compounds, flavan-3-ols and procyanidins content were quantified during fermentation and after drying. It was observed that most of the phenolic compounds were lost during the fermentation, and the beans that fermented for 3 days had higher levels of phenolic compounds. However, a significant loss in the sensorial quality was observed. The natural drying provided higher retention of phenols and the best sensorial results in comparison with artificial drying. The sensory evaluation, performed with a drink formulated with the liquor obtained from different treatments, showed that the product that fermented for 7 days presented the best sensorial notes, and the product dried naturally was considered the best among the others.

FUNDAMENTO: Polimorfismos em genes relacionados ao desenvolvimento da aterosclerose, angiogênese e metabolismo da homocisteína (Hcy) podem ser fatores de risco para a doença arterial coronariana (DAC). OBJETIVO: Avaliar o efeito dos polimorfismos VEGF C-2578A e MTHFR C677T na DAC e a associação desses polimorfismos com a gravidade e a extensão das lesões ateroscleróticas e concentrações de Hcy. MÉTODOS: 244 indivíduos foram avaliados através de angiografia coronariana e incluídos no estudo (145 com DAC e 99 indivíduos-controle). Os polimorfismos VEGF C-2578A e MTHFR C677T foram investigados através das técnicas de PCR-SSCP e PCR-RFLP, respectivamente. Os níveis de homocisteína plasmática foram mensurados através de cromatografia líquida/espectrometria de massa seqüencial (CL/EMS). RESULTADOS: Não houve diferença significante em relação à distribuição de alelos e genótipos entre os grupos, para ambos os polimorfismos. A análise univariada mostrou uma freqüência maior do genótipo VEGF -2578AA no grupo com doença em três vasos (p=0,044). Além disso, o genótipo VEGF -2578CA foi observado mais freqüentemente entre indivíduos com <95% de estenose (p=0,010). Após ajuste para outros fatores de risco para DAC em um modelo multivariado, observou-se que o polimorfismo VEGF C-2578A não era um correlato independente da DAC (p=0,688). O polimorfismo MTHFR não mostrou qualquer relação com a extensão e/ou gravidade da DAC. O polimorfismo MTHFR C677T não mostrou uma associação direta com hiperhomocisteinemia ou aumento das concentrações médias de Hcy no plasma. CONCLUSÃO: Embora haja uma aparente associação entre o polimorfismo VEGF C-2578A e o desenvolvimento de aterosclerose coronariana, essa associação não é independente dos fatores de risco cardiovasculares convencionais.

BACKGROUND: Polymorphisms in genes involved in the atherosclerosis development, angiogenesis, and homocysteine (Hcy) metabolism could be risk factors for coronary artery disease (CAD). OBJECTIVE: To evaluate the effect of the VEGF C-2578A and MTHFR C677T polymorphisms on CAD, and the association of these polymorphisms with the severity and extension of atherosclerotic lesions and Hcy concentrations. METHODS: Two hundred and forty-four subjects were evaluated by coronary angiography and included in the study (145 with CAD and 99 controls). The VEGF C-2578A and MTHFR C677T polymorphisms were investigated by the PCR-SSCP and PCR-RFLP techniques, respectively. Plasma Hcy was quantified by liquid chromatography/sequential mass spectrometry (LC-MS/MS). RESULTS: There was no significant difference in allele and genotype distribution between the groups, for both polymorphisms. The univariate analysis showed a higher frequency of the VEGF -2578AA genotype in the group with three-vessel disease (p=0.044). In addition, the VEGF -2578CA genotype was observed more frequently among individuals with <95% stenosis (p=0.010). After adjustment for other risk factors for CAD in a multivariate model, the VEGF C-2578A polymorphism was not found to be an independent correlate of CAD (p=0.688). The MTHFR polymorphism did not show any association with the extension and/or severity of the CAD. The MTHFR C677T polymorphism showed no direct association with hyperhomocysteinemia or increased mean plasma concentrations of Hcy. CONCLUSION: Although there is an apparent association between VEGF C-2578A and the development of coronary atherosclerosis, this association is not independent of conventional cardiovascular risk factors.

FUNDAMENTO: Polimorfismos en genes relacionados al desarrollo de la aterosclerosis, la angiogénesis y el metabolismo de la homocisteína (Hcy) pueden ser factores de riesgo para la enfermedad arterial coronaria (EAC). OBJETIVO: Evaluar el efecto de los polimorfismos VEGF C-2578A y MTHFR C677T en la EAC y la asociación de esos polimorfismos con la severidad y la extensión de las lesiones ateroscleróticas y concentraciones de Hcy. MÉTODOS: Se evaluaron a 244 individuos por medio de angiografía coronaria y se les incluyeron en el estudio (145 con EAC y 99 individuos-control). Los polimorfismos VEGF C-2578A y MTHFR C677T se investigaron mediante las técnicas de PCR-SSCP y PCR-RFLP, respectivamente. Se midieron los niveles de homocisteína plasmática por medio de cromatografía líquida/espectrometría de masa secuencial (CL/EMS). RESULTADOS: No hubo diferencia significante en relación con la distribución de alelos y genotipos entre los grupos, para ambos polimorfismos. El análisis univariado reveló una frecuencia mayor del genotipo VEGF-2578AA en el grupo con enfermedad en tres vasos (P=0,044). Además de ello, se observó el genotipo VEGF-2578CA con más frecuencia entre individuos con <95% de estenosis (p=0,010). Tras ajuste para otros factores de riesgo para EAC en un modelo multivariado, se evidenció que el polimorfismo VEGF C-2578A no era un correlato independiente de la EAC (p=0,688). El polimorfismo MTHFR no mostró cualquier relación con la extensión y/o severidad de la EAC. El polimorfismo MTHFR C677T no evidenció una asociación directa con hiperhomocisteinemia o aumento de las concentraciones promedio de Hcy en el plasma. CONCLUSIÓN: Si bien existe una aparente asociación entre el polimorfismo VEGF C-2578A y el desarrollo de aterosclerosis coronaria, esa asociación no es independiente de los factores de riesgo cardiovasculares convencionales.

O óleo do café torrado Brasileiro obtido através de extração com CO2 supercrítico mostrou consideráveis propriedades aromáticas, constituído principalmente por cinco compostos aromáticos: 2-metilpirazina; 2-furfuril álcool, 2,5-dimetilpirazina; γ -butirolactona e 2-furfuril acetato. Análises sensoriais foram usadas para verificar a influência da mistura destas importantes classes de compostos aromáticos de café (pirazinas, furanos e lactonas) e do óleo aromático de café torrado no aroma e sabor de bebidas preparadas com café solúvel instantâneo liofilizado e seco por spray dryer. Na análise da aceitação do aroma, a amostra preparada com café liofilizado sem a mistura de compostos voláteis (amostra 4) não diferiu da amostra de café liofilizados com adição de óleo (amostra 5) nem tão pouco da amostra de café liofilizado com adição da mistura dos cinco voláteis (amostra 3), coincidentemente oriundas do mesmo processo de secagem. Entretanto a amostra 3 não diferiu das amostras preparadas com café seco por spray dryer sem (amostra 1) e com (amostra 2) a mistura de voláteis. Indicando que a adição desta mistura, com relação a este atributo não interfere na aceitação desta bebida. Na avaliação do sabor, as amostras preparadas com café solúvel liofilizado enriquecida com óleo aromático (5) e as amostras com (3) e sem (4) a mistura dos cinco voláteis não diferiram significativamente, entretanto a amostra (4) não diferiu das amostras (1) e (2). Para este atributo, a adição do óleo aromático de café torrado ou da mistura de voláteis nas amostras de café liofilizado fez com que tivessem uma melhor aceitação que aquelas secas por spray dryer (1) e (2). Desta forma, o enriquecimento das bebidas com o óleo aromático de café torrado, ou mesmo com a mistura dos cinco componentes não influenciou a aceitação do consumidor com relação ao aroma, mas exerce influência com relação ao sabor.

The oil obtained from Brazilian roasted coffee by supercritical CO2 extraction shows considerable aromatic properties, mainly composed by five aromatic compounds, 2-methylpyrazine; 2-furfurylalcohol, 2,5-dimethylpyrazine; γ-butyrolactone and 2-furfurylacetate. Sensory analyses were used to verify the influence of a mixture of these important classes of aromatic coffee compounds (pyrazines, furans and lactones) and of the roasted coffee aromatic oil on the coffee aroma and flavour of black instant freeze and spray-dried coffee beverages. In the acceptance evaluation of the aroma, the samples prepared with freeze-dried instant coffee without the mixture of volatile compounds (sample 4) were not significantly different from the freeze-dried instant coffee in which the aromatic coffee oil was added (sample 5) and from the sample prepared with freeze-dried coffee in which the mixture of the five volatile was added (sample 3), coincidentally from the same drying process. Therefore, sample (3) did not differ from samples prepared with spray dried instant coffee without (sample 1) and to which (sample 2) the mixture of volatile was added. Therefore, with respect to this attribute, the addition of this mixture did not interfere in this drink acceptance. Taking into consideration the flavor, samples prepared with freeze-dried instant coffee in which the aromatic coffee oil was added (5) and the samples with (3) and without (4) the mixture of the five volatile was added did not differ significantly, however sample (4) did not differ from samples (1) and (2). Regarding this attribute, the addition of the aromatic oil of roasted coffee or a mixture of volatile in samples of freeze-dried instant coffee had a better acceptance than those dried by spray dryer (1) and (2). Thus, the enrichment of drinks with the aromatic oil of roasted coffee, or even with the mixture of the five components did not influence the consumer acceptance with respect to the aroma, but exerts influence with respect to flavour.

Propolis is mostly used as hydroalcoholic extract. Recently there has been a growing number of patents dealing with new solvents for preparing propolis extracts. This study aimed to prepare edible oil propolis extracts and compare their chemical composition and biological activity with ethanolic propolis extracts. ESI-MS and spectrophotometric methods were used for qualitative and quantitative analyses, respectively. Antibacterial activity was evaluated by diffusion in agar. Cytotoxicity was tested by MTT assay using tumor cell lines. The oil is able to extract bioactive compounds from propolis. Further studies are needed to improve extraction efficiency and to characterize the active components.

Chemical investigation of the MeOH extract of the leaves of Machaonia brasiliensis (Rubiaceae) resulted in the isolation and identification of 3beta-O-beta-glucopyranosyl stigmasterol, 3beta-O-beta-glucopyranosyl sitosterol, secologanoside, 7-O-beta-glucopyranosyl quercetagetin, 4,5-O-dicaffeoylquinic acid and 5-O-caffeoylquinic acid. The structures of these compounds were established by spectroscopic analysis, including 2D NMR experiments. The chemotaxonomic relevance of the isolation of secologanoside is discussed.
