SUMMARY OBJECTIVE: The aim of this study was to explore the correlation between skeletal muscle content and the presence and severity of metabolic dysfunction-associated fatty liver disease in patients with metabolic dysregulation in China. METHODS: A cross-sectional study was conducted among patients from the endocrinology outpatient department at Ningbo First Hospital, in Ningbo, China, in April 2021. Adult patients with metabolic dysregulation who accepted FibroScan ultrasound were included in the study. However, those without clinical data on skeletal muscle mass were excluded. FibroScan ultrasound was used to noninvasively evaluate metabolic dysfunction-associated fatty liver disease. The controlled attenuation parameter was used as an evaluation index for the severity of liver steatosis. Bioelectrical impedance analysis was used to measure the skeletal muscle index. RESULTS: A total of 153 eligible patients with complete data were included in the final analysis. As the grading of liver steatosis intensifies, skeletal muscle index decreases (men: Ptrend<0.001, women: Ptrend=0.001), while body mass index, blood pressure, blood lipid, uric acid, aminotransferase, and homeostatic model assessment of insulin resistance increase (Ptrend<0.01). After adjusting for confounding factors, a negative association between skeletal muscle index and the presence of metabolic dysfunction-associated fatty liver disease was observed in men (OR=0.691, p=0.027) and women (OR=0.614, p=0.022). According to the receiver operating characteristic curve, the best cutoff values of skeletal muscle index for predicting the metabolic dysfunction-associated fatty liver disease presence were 40.37% for men (sensitivity, 87.5%; specificity, 61.5%) and 33.95% for women (sensitivity, 78.6%; specificity, 63.8%). CONCLUSION: Skeletal muscle mass loss among patients with metabolic dysregulation was positively associated with metabolic dysfunction-associated fatty liver disease severity in both sexes. The skeletal muscle index cutoff value could be used to predict metabolic dysfunction-associated fatty liver disease. Abstract Kaempferol, an important flavonol, has numerous health-beneficial bioactivities and possesses a great potential for application in medicine, food, and cosmetics industries. To improve the production of kaempferol in an in vitro synthetic biosystem, we designed and constructed a panel of bifunctional enzymes by fusing the flavanone 3-hydroxylase (AtF3H) and the flavonol synthase (AtFLS1) of Arabidopsis thaliana with different orientation and different peptide linker type and length. By comparing the output of kaempferol, we obtained a highly active bifunctional enzyme AtF3H-(GGGGS)2-AtFLS1 with a Km value of 0.129 ± 0.016 mM. After optimization of a series of reaction parameters, kaempferol was produced at 100.54 ± 0.54 mg/L, the currently highest kaempferol output in an in vitro synthetic biosystem, and the substrate conversion rate was 68.26% ± 0.05%. In addition, we observed substrate inhibition for the AtFLS1, which eventually limited the production of kaempferol. This study provides a highly active biocatalyst for production of kaempferol and an insight into biosynthesis of other valuable molecules. Abstract Studies have reported that >91.9% of non-syndromic tooth agenesis cases are caused by seven pathogenic genes. Objective To report novel heterozygous PAX9 variants in a Chinese family with non-syndromic oligodontia and summarize the reported genotype-phenotype relationship of PAX9 variants. Methodology We recruited 28 patients with non-syndromic oligodontia who were admitted to the Hospital of Stomatology Hebei Medical University (China) from 2018 to 2021. Peripheral blood was collected from the probands and their core family members for whole-exome sequencing (WES) and variants were verified by Sanger sequencing. Bioinformatics tools were used to predict the pathogenicity of the variants. SWISS-MODEL homology modeling was used to analyze the three-dimensional structural changes of variant proteins. We also analyzed the genotype-phenotype relationships of PAX9 variants. Results We identified novel compound heterozygous PAX9 variants (reference sequence NM_001372076.1) in a Chinese family with non-syndromic oligodontia: a new missense variant c.1010C>A (p.T337K) in exon 4 and a new frameshift variant c.330_331insGT (p.D113Afs*9) in exon 2, which was identified as the pathogenic variant in this family. Abstract Pelvic inflammatory disease (PID) is an inflammation of the upper genital tract. PID is the leading cause of some severe sequelae in the absence of timely and accurate diagnosis and treatment. An appropriate animal model is needed to explore the underlying mechanism of PID sequelae. This study introduced an animal model of PID by vaginal injection of liquid Ureaplasma urealyticum combined with fatigue and hunger (UVF). This study was designed to test the feasibility of a rat model. A rat model was established using UVF irradiation. Levels of some inflammatory cytokines in the serum and the homogenates of the fallopian tubes were measured by ELISA, RT-PCR, and flow cytometry and compared with another rat model of Ureaplasma urealyticum liquids injected into the two uterus horns during laparotomy. Inflammatory alterations and adhesions were observed after hematoxylin and eosin (H&E) staining and detected using the Blauer scoring system. The results showed that the combined UVF and rat model caused apparent obstruction, edema, and adhesion in the fallopian tubes and connective tissues. The rat model showed upregulated CD4, CD8, and CD4/CD8 in peripheral blood mononuclear cells (PBMCs) and significantly increased levels of IL-4, IL-6, IL-10, and IL-17. UVF also enhanced the expression of tumor necrosis factor (TNF)-α, transforming growth factor (TGF)-β, vascular endothelial growth factor (VEGF) β, and matrix metalloproteinase (MMP)-2 (P<0.05). Although metastasis is the major cause of death in cervical cancer, the mechanism of metastasis is still unclear. The mRNA expression and protein level of latent transforming growth factor beta binding protein 1 (LTBP1) were detected in tumor tissues and paracancerous tissues from in-house samples. Cell proliferation, cell cycle, migration, and in vivo metastasis were determined after LTBP1 was knocked down. Then, 13 drugs were screened, and the changes in cell apoptosis and proliferation and tumor metastasis were detected after drug treatment in shRNA cells. In our in-house samples, LTBP1 was lowly expressed in cervical cancer tissues. After LTBP1 knockdown, cell proliferation was increased, and the ability of in vitro migration and in vivo metastasis was enhanced. Abstract To evaluate the liver stiffness measurement (LSM) of normal people before and after diet using real-time shear wave elastography (SWE), and to explore the effect of diet on liver elasticity. Fifty-four healthy volunteers were selected and 480 mL of a standard diet containing 391 kcal were administrated. The LSM before diet and at 30 minutes after diet was significantly higher than baseline LSM. The LSM at 60 minutes after diet was significantly higher than that before diet. The LSM at 120 minutes after diet was significantly less than that before diet. 75% of the volunteers reached the peak of liver stiffness at 30 minutes or 60 minutes after diet. The LSM increased by more than 20% in 31% subjects, increased by less than 20% in 48%, and remained or decreased in 21%. The blood flow volume of portal vein was 692.02 ± 130.69 mL/min before diet, and it reached the maximum value of 776.41 ± 151.45 mL/min at 30 min after diet. Diet could affect the Young’s modulus measurement of liver. In order to avoid the effect, it is suggested to use real-time shear wave elastic imaging to measure the Young's modulus of liver on an empty stomach.

A rapid and reproducible liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated to simultaneously determine sacubitril, valsartan and a metabolite of sacubitril (LBQ657) in human plasma using sacubitril-d4 and valsartan-d3 as the internal standards. Following protein precipitation, the analytes were operated on an Ultimate® XB-C18 column (2.1 × 50 mm, 3.5 μm, Welch) with a gradient elution with acetonitrile, and 5 mM ammonium acetate and 0.1% formic acid in water as the mobile phase. The detection was performed on a Triple Quad™ 4000 mass spectrometer coupled with an electrospray ionization source (ESI) under positive-ion multiple reaction monitoring mode. The linearities are 2.00-4000, 5.00-10000 and 5.00-10000 ng mL−1 for sacubitril, valsartan and LBQ657, respectively. The accuracy and precision of intra- and inter-day, dilution accuracy, recovery and stability of the method were all within the acceptable limits and no matrix effect or carryover was observed. The suitability of the method was successfully demonstrated in terms of the quantification of sacubitril, valsartan and LBQ657 in plasma samples collected from healthy Chinese volunteers in a clinical trial.

OBJECTIVES: We aimed to explore the effects of diet on the inflammatory response in middle-aged and elderly people with hypertension. METHODS: Thirty overweight or obese patients with stage one hypertension (age range, 45-75 years) were allocated to either the intervention or control group (n=15 per group; age- and sex-matched). Patients in the intervention group consumed a food powder supplement (100 g) instead of a regular meal. The control group maintained their normal dietary habits. This study lasted for six weeks. Blood pressure, inflammatory marker levels, and energy intake were measured before and after the study. RESULTS: After 6 weeks, the diet composition of the intervention group changed significantly (p<0.05). The intake of proteins, dietary fibre, monounsaturated fat, and polyunsaturated fat increased significantly (p<0.05), while the total energy intake trended towards an increase (p>0.05). In the control group, the total energy intake decreased significantly (p<0.05). The levels of nuclear factor-κB (NF-κB), soluble intercellular adhesion molecule-1 (sICAM-1) and high sensitivity C-reactive protein (hs-CRP) decreased, and adiponectin increased significantly in the intervention group (p<0.05); however, no significant changes were observed in the inflammatory marker levels of the control group. In the intervention group, systolic blood pressure decreased significantly (p<0.05), and diastolic blood pressure also exhibited a decreasing trend. No significant change in blood pressure was observed in the control group. CONCLUSION: The consumption of a food powder supplement can improve diet composition, decrease blood pressure and reduce inflammation in middle-aged and elderly overweight or obese hypertensive patients. The food powder supplement may also have an anti-atherosclerotic effect in hypertensive patients.

ABSTRACT We compared the morphology of the erythrocytes of five anurans, two toad species - Bufo gargarizans (Cantor, 1842) and Duttaphrynus melanostictus (Schneider, 1799) and three frog species - Fejervarya limnocharis (Gravenhorst, 1829), Microhyla ornata (Duméril & Bibron, 1841), and Rana zhenhaiensis (Ye, Fei & Matsui, 1995). We then reconstructed the ancestral state of erythrocyte size (ES) and nuclear size (NS) in amphibians based on a molecular tree. Nine morphological traits of erythrocytes were all significantly different among the five species. The results of principal component analysis showed that the first component (49.1% of variance explained) had a high positive loading for erythrocyte length, nuclear length, NS and ratio of erythrocyte length/erythrocyte width; the second axis (28.5% of variance explained) mainly represented erythrocyte width and ES. Phylogenetic generalized least squares analysis showed that the relationship between NS and ES was not affected by phylogenetic relationships although there was a significant linear relationship between these two variables. These results suggested that (1) the nine morphological traits of erythrocytes in the five anuran species were species-specific; (2) in amphibians, larger erythrocytes generally had larger nuclei.

In this study we attempted to confer E. coli strain capability of automatic excretion of the chemical by employing a native anaerobic promoter. Also, a novel DNA kit was designed for PCR preparation of linear DNA fragments to perform strain modification. The kit is primarily composed of two mother vectors, co-transformation of linear DNAs into E. coli can simultaneously introduce two butanol synthetic operons into the chromosome and create two in-frame gene deletions at targeted native loci. Results E. coli strain Bw2V carries plasmid pCNA-PHC and pENA-TA, both utilizes native anaerobic promoter Phya for the expression of butanol synthetic enzymes. When Bw2V was subjected in anaerobic fermentation using medium containing extra glucose, the accumulated n-butanol in the broth was up to 2.8 g/L in bioreactor; as the genetic element expressing the same pathway was introduced into the genome, the titer of butanol was 1.4 g/L. Conclusions The expression system using Phya is effective in applications that involve expression plasmids as also applicable in ectopic expression as single copy on the chromosome. Results imply that Phya can be subjected for broader application in bioproduction of more feedstock chemicals.

Background: Overweight and obesity are epidemic worldwide. Our previous study found that the prevalence of overweight and obesity in primary students is high. Objective: we sought to estimate whether the prevalence of overweight and obesity among university students is still high in China. Methods: A cross-sectional study was designed to collect the routine health screening data for university students in 2013. The height and weight of students were measured, and BMI was calculated with height and weight, we estimated the underweight and obesity prevalence of university students using by two references [Working Group on Obesity references in China (2004) and World Health Organization (WHO) criteria (2000)]. Results: Depending on the China references used, the overall prevalence of underweight, overweight (including obesity) and obesity in male university students were 14.2%, 14.7% and 4.2%, respectively; in female university students were 27.5%, 4.4% and 0.6%, respectively. Background: Previous study revealed that the link between dental caries and obesity has been controversial. The purpose of this research is to investigate the association between dental caries and obesity among primary school children in Wannan area, China. Methods: A cross-sectional study was designed to collect the routine health screening data for primary school children aged 5-14 years inWannan area,China, Overweight and obesity status were determined using the International Obesity Task Force standard (IOTF) BMI cut-off points. Caries status was recorded based on WHO recommendations. Results: Our results revealed that the overall caries prevalence of the subjects was 44.9%, Maximum number of caries affected children belonged to underweight and normal group, followed by overweight, and the least number was obesity. These differences were statistically significant (chi-square test, P < 0.001). Children with obesity were 1.908 times (OR =1.908; CI95%=1.750, 2.079) more likely have caries than children with underweight or health weight. Overweight children were 1.547 times (OR = 1.547; CI95% = 1.479, 1.618) more likely to have caries than children with underweight or health weight. After adjusted the gender and age, a statistically significant association was also observed between body mass index categories and caries. Conclusions: Obesity may have a significant effect on caries prevalence of primary school children in Wannan area, China. The importance of obesity should not only be emphasized with respect to general diseases but also with regard to carious lesions.

El propósito de esta investigación fue investigar la asociación entre la caries dental y la obesidad en niños de escuela primaria en el área de Wannan, China. Métodos: Se diseñó un estudio transversal para recoger los datos del chequeo rutinario de salud de los niños de escolarización primaria con edades de 5-14 en el área de Wannan, China. Se determinó el estado de sobrepeso y obesidad utilizando los puntos de corte estándar del IMC del Grupo de Trabajo Internacional en Obesidad (GTIO). El estado de las caries se registró de acuerdo con las recomendaciones de la OMS. Resultados: Nuestros resultados revelaron que la prevalencia global de caries en los sujetos fue del 44,9%. El máximo número de niños afectados por las caries estaba en los grupos normal y con peso bajo, seguido por el grupo con sobrepeso y por último en el grupo de obesidad. Estas diferencias fueron estadísticamente significativas (test Chi-cuadrado, P < 0,001). Los niños con sobrepeso tenían una probabilidad 1,547 veces superior (OR = 1,547; IC al 95% = 1,479, 1,618) de tener caries dental que los niños con un peso bajo o normal. Tras ajustar por sexo y edad, también se observó una asociación estadísticamente significativa entre el índice de masa corporal y la caries. Conclusiones: La obesidad puede tener un efecto significativo sobre la prevalencia de caries en niños en escolarización primaria en el área de Wannan, China. No sólo se debería poner el énfasis de la importancia de la obesidad en relación con las enfermedades generales sino también en relación con las lesiones cariales.

Moreover, it is observed that cracks are formed on the etched ribbon surface and plentiful Cu2O/CuO particles grow up from these crack walls if the dealloying time extends to long enough. Considering many fascinating properties of Cu2O/CuO particles and the amorphous alloy carrier, potential application fields of these amazing compounds will be developed in future.

Being a biocompatible, biodegradable and bioactive material, chitosan in the form of membranes offers a great potential as a substrate in mineralization processes in vitro. In the present work, chitosan with (without) bovine serum albumin used as templates induced the formation of calcium carbonate. The unusual morphology of calcium carbonate was get from the chitosan /bovine serum albumin system. The CaCO3 crystals obtained in system were characterized by scanning electron microscopy, Fourier transform infrared spectrography and powder X-ray diffractometry. The possible formation mechanism of CaCO3 was discussed.
