Cell-free antigens (CFAg) derived from Paracoccidioides brasiliensis have typically been used in immunodiffusion reactions for serodiagnosis or therapeutic follow-up of paracoccidioidomycosis patients. Thus, we investigated the usefulness of CFAg obtained from cultures at different ages, to evaluate cellular immunity by the footpad test, in experimental murine paracoccidioidomycosis. Male mice infected with P. brasiliensis 265 strain were challenged in the footpad with CFAg obtained from four- (4d CFAg) or 11-day-old cultures (11d CFAg). The increase in footpad swelling provoked by 4d CFAg and 11d CFAg was similar and showed significant difference in relation to control groups. However, the infiltrate pattern was strikingly different: 4d CFAg induced a predominant mononuclear infiltrate whereas 11d CFAg provoked a predominant polymophonuclear infiltrate. These different inflammatory patterns were associated with distinct electrophoretic characteristics. By comparison with 11d CFAg, 4d CFAg showed more numerous and intense bands, including a strong one of 43 kDa (gp43). These results suggest that CFAg derived from Pb 265 isolate can be used as a reagent to evaluate cellular immunity; however, the culture's age is critical because only young cultures are able to induce a typical mononuclear infiltrate. The efficacy of this new paracoccidioidin to assay the cellular immunity in infections caused by other P. brasiliensis isolates is under investigation.

Leishmaniasis, a zoonosis of worldwide distribution, presents a significant impact on immunosupressed patients. This study aimed to evaluate Leishmania chagasi infection in BALB/c mice immunosuppressed with dexamethasone. Spleen cells stimulated or not with L. chagasi were cultured for cytokine quantification (IFN-γ, IL-2, IL-4 and IL-10) by sandwich ELISA. Parasite loads in the spleen and liver were determined by means of culture microtitration. Immunosuppressed groups showed statistically lower spleen weight and CD4-cell percentage in blood on the day of infection and produced Th1 and Th2 cytokines on other days of the study. The other infected groups, weather immunosupressed or not, also produced Th1 and Th2 cytokines. Parasite loads in the spleen and liver were not statistically different among the groups. It was concluded that L. chagasi infection was not affected by dexamethasone-induced immunosuppression, probably due the reversible effect of the treatment.

The immune response in leishmaniasis may result in a polarization of the T lymphocyte subpopulation, altering cell phenotype and resulting in immune protection or disease exacerbation. Leishmania may persist in the body either during asymptomatic infections or after treatment, which represents high risk under immunosuppression. The objective of this study was to evaluate the effect of infection with immunosuppression by dexamethasone associated with pentoxifylline on animal weight, spleen weight, spleen and hepatic parasitic load and immunopathology, as well as the IFN-γ and IL-10 production in spleen cell culture of Balb/c mice infected with Leishmania chagasi. The infection did not cause body weight gain in animals, but both the weight and size of the spleen were increased. The immunosuppression using dexamethasone associated with pentoxifylline affected body weight gain and spleen weight and size in both infected and non-infected animals. The immunosuppression did not significantly alter the course of the splenic or hepatic parasite burden. Dexamethasone and pentoxifylline significantly affected cytokine production, but did not influence the Th1/Th2 ratio in infected animals.

Os mecanismos utilizados pelo Paracoccidioides brasiliensis para sobreviver em células fagocitárias ainda não estão elucidados. O metabolismo celular férrico é muito importante para o crescimento de inúmeros patógenos intracelulares cuja capacidade de se multiplicarem em fagócitos mononucleares é dependente da disponibilidade intracelular do íon ferro. Assim, o objetivo deste trabalho foi investigar o papel do ferro intracelular sobre a capacidade do P. brasiliensis sobreviver em monócitos humanos. O tratamento de monócitos com deferoxamina, uma droga quelante, diminuiu a sobrevivência de leveduras do fungo de forma dose-dependente. O efeito inibidor da deferoxamina sobre a sobrevivência do P. brasiliensis foi revertido por transferrina saturada com ferro (holotransferrina) mas não por transferrina insaturada (apotransferrina). Estes resultados sugerem que a sobrevivência do P. brasiliensis em monócitos humanos é dependente do íon ferro.

The mechanisms used by Paracoccidioides brasiliensis to survive into phagocytic cells are not clear. Cellular iron metabolism is of critical importance to the growth of several intracellular pathogens whose capacity to multiply in mononuclear phagocytes is dependent on the availability of intracellular iron. Thus, the objective of this paper was to investigate the role of intracellular iron in regulating the capacity of P. brasiliensis yeast cells to survive within human monocytes. Treatment of monocytes with deferoxamine, an iron chelator, suppressed the survival of yeasts in a concentration-dependent manner. The effect of deferoxamine was reversed by iron-saturated transferrin (holotransferrin) but not by nonsaturated transferrin (apotransferrin). These results strongly suggest that P. brasiliensis survival in human monocytes is iron dependent.

O conhecimento das formas químicas em que se encontra o cobre em solos, e suas relações com os teores disponíveis, são importantes para a previsão do seu comportamento no sistema solo-planta. Nesse sentido, este trabalho estuda a influência da calagem sobre os teores disponíveis e sobre o fracionamento de Cu em amostras de seis Latossolos. Essas amostras, submetidas ou não a calagem, receberam o elemento nas doses de 0,0, 20,0 e 40,0 mg dm-3 e foram incubadas por 30 dias. Terminada a incubação, procedeu-se a extração de Cu utilizando-se os extratores Mehlich-1, Mehlich-3, DTPA e EDTA. As amostras foram submetidas a um fracionamento que separou o Cu nas frações trocável, matéria orgânica, óxido de manganês, óxido de ferro amorfo, óxido de ferro cristalino e residual, além dos teores totais. Com sua aplicação ao solo o Cu foi retido, principalmente, na fração matéria orgânica, com pequena porcentagem retida na fração trocável. A calagem provocou redução nas frações trocável e matéria orgânica e aumento nas frações óxido de ferro, óxido de manganês e residual. Para os solos sem calagem, a fração matéria orgânica foi a que mais contribuiu para os teores determinados pelos extratores, com exceção do EDTA. Nos solos com calagem, o Cu ligado à matéria orgânica foi melhor correlacionado com os teores obtidos pelo DTPA e Mehlich-3.

The knowledge of the chemical forms of copper in soils and the relationships of these forms with soil copper availability are important for predicting the copper behavior in the soil-plant system. The present work studies the influence of liming on the available contents of copper as well as on the forms of copper fractions in six types of Oxisols. Soil samples, with and without liming, received copper at rates of 0.0, 20.0 and 40.0 mg dm-3 and remained incubated for 30 days. Then, available copper was extracted with Mehlich-1, Mehlich-3, DTPA and EDTA solutions, and analyzed by atomic absorption spectrophotometry. Additionally, soil samples were extracted in a sequential procedure to determine Cu in fractions of soil, as follows: exchangeable-Cu fraction, organic matter-Cu fraction, Mn oxide-Cu fraction, amorphous Fe oxide-Cu fraction, crystalline Fe oxide-Cu fraction, residual-Cu fraction, and the total Cu content in the soil. Soil samples to which Cu was added presented higher Cu retention in the organic matter fraction with a small percentage retained in the exchangeable-Cu fraction. Liming resulted in a decrease of Cu in the exchangeable and organic matter fractions and an increase in the Fe and Mn oxide fractions and in the residual fraction. Without liming, the organic matter fraction presented the highest contribution to Cu content found in the soil extracts obtained with all extractors, except EDTA. For treatments with liming, Cu contents in the organic matter fraction were better correlated to Cu contents in extracts obtained with DTPA and Mehlich-3.
