ABSTRACT Objective: To evaluate the cumulative incidence, risk factors, and outcomes of COVID-19 in patients with Cushing's disease (CD). Subjects and methods: In all, 60 patients with CD following up in our outpatient clinic answered via phone interview a questionnaire about the occurrence of COVID-19 infection documented by RT-PCR (including the diagnosis date and clinical outcome) and vaccination status. Clinical and biochemical data on disease activity (hypercortisolism) and comorbidities (obesity, diabetes mellitus, and hypertension) were obtained from the patients' electronic medical records. Risk ratios (RRs) of risk factors were obtained using univariate and multivariate analyses. Results: The cumulative incidence of COVID-19 in patients with CD during the observation period was 31.7%, which was higher than that in the general reference population (9.5%). The cumulative incidence of COVID-19 was significantly higher in patients with hypercortisolism (57% versus 17% in those without hypercortisolism, p = 0.012) and obesity (54% versus 9% in those without obesity, p < 0.001) but not in patients with hypertension or diabetes mellitus. On multivariate analysis, hypercortisolism and obesity were each independent risk factors for COVID-19 (RR 2.18, 95% CI 1.06-4.46, p = 0.033 and RR 5.19, 95% CI 1.61-16.74, p = 0.006, respectively). Conclusion: The incidence of COVID-19 in patients with CD was associated with hypercortisolism, as expected, and obesity, a novel and unexpected finding. Thus, correction of hypercortisolism and obesity should be implemented in patients with CD during the current and future COVID-19 outbreaks. Resumo O aumento da modificação dos habitats e da perda de espécies demanda esforços consistentes para descrever e compreender os padrões de biodiversidade. O programa BIOTA/FAPESP foi criado nesse contexto e é uma iniciativa de sucesso para promover estudos em biodiversidade e conservação no Brasil. O BIOTA/Araçá é um projeto interdisciplinar que promoveu uma avaliação detalhada da biodiversidade da Baía do Araçá, um ecossistema costeiro localizado ao Norte do estado de São Paulo, Sudeste do Brasil. A baía engloba múltiplos habitats, tais como praias, manguezais, costões rochosos, e uma planície de maré, e também fornece importantes serviços ecossistêmicos. Infelizmente, a baía está sujeita à conflitos sócio-ambientais complexos que contrastam demandas econômicas, sociais e ambientais (i.e. a expansão das atividades do porto vizinho vs. a pesca artesanal de pequena escala e a proteção da biodiversidade). O presente estudo apresenta um levantamento das espécies bentônicas que ocorrem nos diferentes habitats da Baía do Araçá, incluindo dados obtidos durante o projeto BIOTA/Araçá e de investigações realizadas anteriormente na área. As espécies bentônicas desempenham um papel importante no ambiente marinho, e estudar a diversidade desses organismos que vivem associados ao fundo é indispensável para compreender o funcionamento do meio ambiente. A macrofauna, meiofauna, e microorganismos associados aos fundos consolidado e inconsolidado foram listados, e informações adicionais foram fornecidas para cada espécie, tais como a distribuição geográfica e nos habitats. O checklist inclui 826 espécies, quase 70% registradas durante o projeto BIOTA/Araçá. Os taxa mais especiosos foram os anelídeos (225 spp.), moluscos (194 spp.), e crustáceos (177 spp.). Abstract Increasing habitat modification and species loss demand consistent efforts to describe and understand biodiversity patterns. The BIOTA/FAPESP Program was created in this context and it has been a successful initiative to promote studies on biodiversity and conservation in Brazil. The BIOTA/Araçá is an interdisciplinary project that provided a detailed evaluation of the biodiversity of Araçá Bay, a coastal seascape located on the North coast of the state of São Paulo, Southeast Brazil. The bay encompasses multiple habitats, such as beaches, mangroves, rocky shores, and a tidal flat, and provides important ecosystem services. Unfortunately, the bay is the subject of complex social-environmental conflicts that oppose economic, social, and environmental demands (i.e., the expansion of neighboring harbor activities vs. small-scale artisanal fisheries and protection of biodiversity). The present study presents a survey of the benthic species occurring in the different habitats of Araçá Bay, including data obtained during the BIOTA/Araçá project and previous assessments of the area. The benthic species play an important role in marine environments and studying the diversity of these organisms that live associated with the bottom is indispensable for comprehending the environment’s functioning. The macrofauna, meiofauna, and microorganisms associated with soft and hard bottom were listed, and additional information, such as the habitat and geographical distribution, were provided for each species. The checklist includes 826 species, almost 70% recorded during the BIOTA/Araçá project. The most speciose taxa were the annelids (225 spp.), mollusks (194 spp.), and crustaceans (177 spp.). Seven benthic species are endemic to Araçá Bay, 14 are considered threatened, and seven are economically exploited. Furthermore, the bay is the type locality of many taxa, and 11 new benthic species were described based on specimens sampled during the project. Abstract This study evaluated the median lethal concentration of silver nanoparticles and their effects in fish tambaqui Colossoma macropomum. Therefore, an acute toxicity assay was carried out in completely randomized design evaluating six different concentrations of silver nanoparticles on blood parameters of tambaqui. The silver nanoparticles were produced by chemical reduction with polyvinyl alcohol (AgNP-PVA). The lethal concentration 50% (LC50) was estimated using probit regression. The blood was collected, analyzed and the data were submitted to T-test (dying x surviving fish) and Tukey test (surviving fish). An increase in glucose, hematocrit, total plasma protein, hemoglobin, erythrocytes, leukocytes, monocytes, and neutrophils as well as reduced MCV (mean corpuscular volume) in dying fish compared to surviving fish were observed. Survived fish exposed to 187.5 µg/L showed an increase in hematocrit, MCV, and MCH and a reduction in erythrocytes, total numbers of leukocyte, thrombocyte, lymphocyte, and neutrophil. The fish exposed to concentrations below 125 µg/L, had returned the blood parameter to baselines compared to control. The estimated LC50 was 165.09 µg/L and was classified as highly toxic for the fish tambaqui. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Objective: To compare plasma concentrations of cannabidiol (CBD) following oral administration of two formulations of the drug (powder and dissolved in oil), and to evaluate the effects of these distinct formulations on responses to emotional stimuli in healthy human volunteers. Methods: In a randomized, double-blind, placebo-controlled, parallel-group design, 45 healthy male volunteers were randomly assigned to three groups of 15 subjects that received either 150 mg of CBD powder; 150 mg of CBD dissolved in corn oil; or placebo. Blood samples were collected at different times after administration, and a facial emotion recognition task was completed after 150 min. Results: There were no significant differences across groups in the subjective and physiological measures, nor in the facial emotion recognition task. However, groups that received the drug showed statistically significant differences in baseline measures of plasma CBD, with a significantly greater difference in favor of the oil formulation. Conclusion: When administered as a single 150-mg dose, neither formulation of oral CBD altered responses to emotional stimuli in healthy subjects. The oil-based CBD formulation resulted in more rapid achievement of peak plasma level, with an approximate fourfold increase in oral bioavailability.

Resumo Uma nova espécie de Knodus das bacias dos rios Mearim e Munim, Nordeste do Brasil, é descrita com base em taxonomia integrativa, utilizando diferentes métodos moleculares de delimitação de espécies e abordagens independentes. A nova espécie possui a combinação de estados de caráter que geralmente é utilizada para diagnosticar o gênero. A nova espécie possui um padrão de coloração semelhante a K. victoriae, que também é morfologicamente semelhante a ela. A espécie aqui descrita difere de K. victoriae por possuir mais vértebras totais, mais raios ramificados na nadadeira anal e menos escamas circumpedunculares. Nós também fornecemos uma discussão detalhada das características morfológicas diagnósticas exibidas por espécies de Knodus de bacias hidrográficas adjacentes.
Abstract A new species of Knodus from the Mearim and Munim River basins, Northeastern Brazil, is herein described based on integrative taxonomy, by using different molecular based species delimitation methods and independent approaches. The new species possesses the combination of character states that usually diagnoses the genus. The new species possesses a similar colour pattern to K. victoriae, which is also morphologically similar to it. The species described herein differs from K. victoriae by possessing more total vertebrae, more branched anal-fin rays, and fewer circumpeduncular scales. We also provide a detailed discussion of the morphological diagnostic features exhibited by Knodus species from adjacent river basins.

RESUMO: O objetivo deste trabalho foi avaliar a série histórica (2010-2019) de condenações parciais e totais de carcaças de aves por síndrome ascítica em abatedouros registrados no SIF localizados nos principais estados produtores de carne de frango. Através de dados oficiais, foram calculados o índice de ocorrência de condenações (IOC) e o índice de sazonalidade ajustado (ISA). A taxa de condenação foi de 1.140 carcaças condenadas total ou parcialmente por síndrome ascítica a cada 1.000.000 de aves abatidas. O menor e maior IOCs foram encontrados em São Paulo (fevereiro/2010) e em Goiás (janeiro/2017), respectivamente. A ocorrência de condenações por esta síndrome teve comportamento cíclico ao longo da série histórica, apresentando picos de condenação em todos os anos avaliados, sendo os maiores ISAs encontrados nos meses de julho, agosto e setembro, com índices variando entre 1,24 e 1,54 nestes meses. Considerando o período de alojamento pré-abate, os maiores valores de ISAs coincidiram com o período mais frio do ano (Maio a Agosto) para todos os estados analisados. Os resultados mostram que síndrome ascítica é um problema crescente no Brasil, com maior ocorrência durante os meses frios do ano, impactando negativamente a saúde dos animais e a lucratividade dos produtores e indústrias da cadeia produtiva de frango de corte.
ABSTRACT: Over the last decade, Brazil registered a 10.56% increase in the number of poultry slaughtered in establishments registered under the “Serviço de Inspeção Federal” (SIF - Federal Inspection Service), as a result of technological advances in management, health and genetics applied to national aviculture. At slaughter, during post-mortem inspection, carcasses can be totally or partially condemned for various reasons, including ascitic syndrome. This syndrome has economic implications for the industry, in addition to being a problem for the health and welfare of poultry. The objective of this work was to evaluate the historical series (2010-2019) of partial and total condemnations of poultry carcasses due to ascitic syndrome in slaughterhouses registered under the SIF and located in the main poultry-producing states. Through official data, the condemnation occurrence index (COI) and the adjusted seasonal index (ASI) were calculated. The condemnation rate was 1,140 carcasses condemned, totally or partially, for ascitic syndrome for every 1,000,000 poultry slaughtered. The smallest and largest COIs were found in São Paulo (February 2010) and in Goiás (January 2017), respectively. The occurrence of condemnations for this syndrome was cyclical throughout the historical series, showing peaks of condemnation in all years evaluated, with the highest ASIs in July, August and September, and with rates varying between 1.24 and 1.54 in these months. Considering the period of pre-slaughter housing, the highest ASIs coincide with the coldest period of the year (May to August) for all analyzed states. The results show that ascitic syndrome is a growing problem in Brazil, with greater occurrence during the coldest months of the year, having a negative impact on animal health and the profitability of producers and industries in the poultry production chain.

ABSTRACT Objective To verify the presence of variants in HNF1B in a sample of the Brazilian population selected according to the presence of renal cysts associated with hyperglycemia. Subjects and methods We evaluated 28 unrelated patients with clinical suspicion of HNF1B mutation because of the concomitant presence of diabetes mellitus (DM) or prediabetes and renal cysts. Genotyping was accomplished using Sanger sequencing or multiplex ligation-dependent probe amplification (MLPA). In positive cases, available relatives were recruited. Results We found two patients with HNF1B mutations. The first presented the variant p.Pro328Leufs*48(c.983delC) and had DM, renal cysts, and hypomagnesemia. The second presented a heterozygous whole gene deletion in HNF1B, DM, renal cysts, body and tail pancreatic agenesis, and hypomagnesemia; this alteration was also found in his two siblings and his father. Conclusion The recruitment of suspected cases of HNF1B gene mutations in Brazilians due to hyperglycemia and renal cysts presents two positive cases. Our cases contribute to the annotation of clinical and biochemical phenotypes of this rare form of maturity-onset diabetes of the young (MODY).

Abstract: We have identified specimens of the leafhopper Gypona cerea DeLong & Freytag (Cicadellidae: Iassinae: Gyponini) collected from a crop of dwarf cashew (Anacardium occidentale Linnaeus 1753) located in Canto do Buriti, Piauí, Northeastern Brazil. The damage caused to the crop included inflorescences covered with sticky exudates, presence of sooty mold, and yellowing and drying of new shoots and inflorescences. Such documentation serves as a warning about possible outbreaks of infestation in the region and the likelihood of its expansion to other cashew producing areas.

Piperlongumine is a natural amide alkaloid isolated from several species of Piper and is described in the literature as selectively cytotoxic to several cancer cell lines. Inhibiting cell migration has gained considerable interest as an approach for discovering antimetastatic agents because this process is fundamental to metastasis. Piperlongumine, selected from cell-based assay screening of NuBBE Database, inhibited the migration of MDA-MB-231 breast cancer cells with an EC50 of 3.0 ± 1.0 µM by the Boyden chamber assay. A series of five analogous compounds based on the structure of piperlongumine were designed, synthesized and evaluated in cell migration and cytotoxicity assays. The analogue designed by molecular simplification ((E)-N-acryloyl-3-(3,4,5-trimethoxyphenyl)acrylamide) was the most active of the series, with an EC50 of 1.5 ± 1 µM. Additionally, this compound was selectively cytotoxic, with a selectivity index (SI) of 4.4.

RESUMO Este trabalho registra o primeiro relato de Megastigmustransvaalensis Hussey (Hymenoptera: Torymidae) em drupas de aroeira-vermelha brasileira, Schinusterebinthifolius Raddi (Anacardiaceae), em Sorocaba, estado de São Paulo, Brasil. Essa vespa é uma espécie invasora e foi encontrada danifi cando drupas de S.terebinthifolius em áreas urbanas (35,0±15,8%), florestas nativas (21,5±10,2%) e áreas de restauração (15,8±8,4%). A bioecologia e os danos causados por M.transvaalensis em drupas de S. terebinthifolius justifi cam mais estudos visando o manejo dessa vespa fi tófaga. Megastigmus transvaalensis tem um potencial de ser disseminado por todo Brasil e representa uma ameaça para a regeneração natural de S. terebinthifolius em florestas nativas e áreas de recomposição e restauração ecológica do país.
ABSTRACT This paper records the first report of Megastigmus transvaalensis Hussey (Hymenoptera: Torymidae) in Brazilian peppertree, Schinus terebinthifolius Raddi (Anacardiaceae) drupes in Sorocaba, state of São Paulo, Brazil. This wasp is an invasive species and was found damaging S. terebinthifolius drupes in urban areas (35.0±15.8%), natural forests (21.5±10.2%) and restoration areas (15.8±8.4%). The bio-ecology and damage caused by M. transvaalensis in the S. terebinthifolius drupes warrants further study focused upon the management of this phytophagous wasp. Megastigmus transvaalensis has a potential to be disseminated throughout Brazil and is posing a threat to the natural regeneration of S. terebinthifolius in the native forests and restoration areas and ecological regions of this country.

The adhesion of biofilm on dental prostheses is a prerequisite for the occurrence of oral diseases. Objective: To assess the antimicrobial activity and the mechanical properties of an acrylic resin embedded with nanostructured silver vanadate (β-AgVO3). Material and Methods: The minimum inhibitory concentration (MIC) of β-AgVO3 was studied in relation to the species Staphylococcus aureus ATCC 25923, Streptococcus mutans ATCC 25175, Pseudomonas aeruginosa ATCC 27853, and Candida albicans ATCC 10231. The halo zone of inhibition method was performed in triplicate to determine the inhibitory effect of the modified self-curing acrylic resin Dencor Lay - Clássico®. The surface hardness and compressive strength were examined. The specimens were prepared according to the percentage of β-AgVO3 (0%-control, 0.5%, 1%, 2.5%, 5%, and 10%), with a sample size of 9x2 mm for surface hardness and antimicrobial activity tests, and 8x4 mm for the compression test. The values of the microbiologic analysis were compared and evaluated using the Kruskal-Wallis test (α=0.05); the mechanical analysis used the Shapiro-Wilk's tests, Levene's test, ANOVA (one-way), and Tukey's test (α=0.05). Results: The addition of 10% β-AgVO3 promoted antimicrobial activity against all strains. The antimicrobial effect was observed at a minimum concentration of 1% for P. aeruginosa, 2.5% for S. aureus, 5% for C. albicans, and 10% for S. mutans. Surface hardness and compressive strength increased significantly with the addition of 0.5% β-AgVO3 (p<0.05). Higher rates of the nanomaterial did not alter the mechanical properties of the resin in comparison with the control group (p>0.05). Conclusions: The incorporation of β-AgVO3 has the potential to promote antimicrobial activity in the acrylic resin. At reduced rates, it improves the mechanical properties, and, at higher rates, it does not promote changes in the control.

A antracnose é a principal doença pós-colheita em frutos de goiabeira e visando reduzir o uso de agroquímicos tem-se investido em métodos alternativos de controle desta doença. O estudo teve como objetivo avaliar o rendimento da produção e a caracterização química de hidrolato e extratos provenientes de folhas e sementes de aroeira (Schinus terebinthifolius Raddi) em diferentes tempos de destilação. O trabalho inclui ainda a avaliação, in vitro, do potencial inibitório do fungo Colletotrichum gloeosporioides. O hidrolato e o extrato foram obtidos pelo método de hidrodestilação realizado em aparato de Clevenger e a determinação do perfil fotoquimico foi realizado por meio de cromatografia gasosa. Os ensaios com o fungo foram realizados a partir da adição dos subprodutos ao meio de cultura BDA em placas de Petri com 6 concentrações de extrato aquoso (5; 10; 15; 20; 25 e 30%) e 4 de hidrolato (10; 15; 20 e 25%) acondicionadas em câmara a 25 ºC. Os resultados mostraram que as folhas de S. terebinthifolius proporcionaram maior volume de hidrolato quando comparadas às de sementes, porém o rendimento de extrato aquoso foi semelhante para as duas estruturas vegetais estudadas. O tempo de hidrodestilação não exerceu influência no rendimento dos subprodutos. As análises cromatográficas não detectaram compostos presentes no hidrolato de aroeira. No ensaio in vitro de inibição do fungo, o extrato aquoso e hidrolato, em todas as concentrações testadas, não apresentaram potencial fungitóxico para o C. gloeosporioides, agente causador da antracnose em goiabas.
Anthracnose is the major disease seen post-harvest in the fruit of the guava, and with an aim to reducing the use of agrochemicals, there has been investment in alternative methods of controlling this disease. This study had as its objective to evaluate the yield and chemical characterisation of the hydrolate and leaf and seed extracts of the mastic tree (Schinus terebinthifolius Raddi) for different distillation times. The work also includes evaluating the in vitro inhibitory potential to the fungus Colletotrichum gloeosporioides. The hydrolate and the extract were obtained by hydrodistillation using a Clevenger apparatus, and the photochemical profile was determined by means of gas chromatography. Trials with the fungus were carried out by the addition of the by-products to a PDA culture medium in Petri dishes, with 6 concentrations of aqueous extract (5, 10, 15, 20, 25 and 30%) and 4 of hydrolate (10, 15, 20 and 25%), placed in a chamber at 25ºC. The results demonstrated that the leaves of S. terebinthifolius provide a greater volume of hydrolate when compared to that from the seeds, but the yield of aqueous extract was similar for the two plant structures under study. The period of hydrodistillation had no influence on the production of by-products. The chromatographic analyses did not detect compounds in the hydrolate of the mastic tree. For the in vitro trials of fungus inhibition, at all the concentrations under test neither the aqueous extract nor the hydrolate demonstrated antifungal potential for C. gloeosporioides, the causal agent of anthracnose in the guava.

Low-intensity resistance exercise does not affect cardiac autonomic modulation in patients with peripheral artery disease

OBJECTIVE: To analyze the effect of a single bout of resistance exercise on cardiac autonomic modulation in patients with peripheral artery disease. METHODS: Fifteen patients with peripheral artery disease (age: 58.3±4.0 years) underwent the following sessions in a random order: resistance exercise (three sets of 10 repetitions of the six resistance exercises with a workload of 5-7 in the OMNI-RES scale) and control (similar to the resistance session; however, the resistance exercises were performed with no load). The frequency domain (low frequency, high frequency and sympathovagal balance) and symbolic analysis (0V, 1V and 2V patterns) of heart rate variability were obtained before and until one hour after the interventions. RESULTS: After the resistance exercise and control sessions, similar increases were observed in the consecutive heartbeat intervals (control: 720.8±28.6 vs. 790.9±34.4 ms; resistance exercise: 712.9±30.1 vs. 756.8±37.9 ms; p<0.05) and in the pattern of the symbolic analysis with no variation (0V) (control: 25.1±3.5 vs. 33.4±4.1%; resistance exercise: 26.1±3.2 vs. 29.7±3.5%; p<0.05) until 50 min after both interventions. The pattern of two variations (2V) decreased similarly (control: 11.2±2.1 vs. 8.3±2.1%; resistance exercise: 9.5±1.7 vs. 7.8±1.7%; p<0.05). In contrast, the pattern of one variation (1V), the low and high frequency bands and sympathovagal balance did not change after the interventions (p>0.05). CONCLUSION: A single bout of resistance exercise did not alter cardiac autonomic modulation in patients with peripheral artery disease.
