Abstract Objective To assess a panel of cytokines and placental insufficiency with the risk of preterm delivery (PTD). Methods Nested case-control study into the BRISA birth cohort. Eighty-two mother-infant-placenta pairs were selected at 20+0 to 25+6 weeks. Circulating biomarker levels were performed using Luminex flowmetric xMAP technology. Cytokines classified as Th1, Th2 or Th17 and other biomarkers were selected. The ratio between birth weight and placental weight (BW/PW) was used as a proxy for placental efficiency. Spearman correlation, univariate analyses and logistic regression models were calculated. Sensitivity, specificity, positive and negative likelihood ratios were calculated using the Receiver Operating Characteristic curve. Results Mean gestational age was 250 days, 14,6% were small for gestational age, 4,8% large for gestational age and 13,4% stunted. Placental efficiency was higher for term newborns (p<0,001), and 18/22 (81%) preterm biomarker values were higher than the control group. Th1 cytokines were highly correlated, while the weakest correlation was observed in other biomarkers. Less education was associated with a higher risk of PTD (p = 0.046), while there was no appreciable difference in the risk of PTD for placental insufficiency. Biomarkers showed negligible adjusted OR of PTD (0.90 to 1.02). IL-6, IL-8, IL-1β, TNFβ, IL-4, IL-13, GCSF, MIP1A, VEGF, EGF, and FGF2 presented a higher sensitivity ranging from 75.56% to 91.11%. Conclusion IL-8, IL-12p40, IL-4, IL-13, GCSF, MIP1B, and GMSF in asymptomatic pregnant women were associated with PTD. This finding suggests an activation of maternal inflammatory response. . Resumo O objetivo deste estudo foi identificar espécies de Cryptosporidium em bovinos e ovinos do Paraná, região sul do Brasil. Amostras de fezes de 458 bovinos e 101 ovinos foram individualmente submetidas à análise molecular por PCR e nested PCR, utilizando-se iniciadores específicos para sequências da unidade ribossomal 18S (rRNA). As amostras positivas foram analisadas pelo polimorfismo de comprimento de fragmento de restrição (RFLP), seguido de sequenciamento genético para confirmação da espécie. A ocorrência de Cryptosporidium foi de 11,27% (63/559). Observou-se maior ocorrência em cordeiros (20,33%). Das 63 amostras positivas, foi possível identificar as espécies em 58 amostras por RFLP e sequenciamento genético. Foram identificadas cinco espécies de Cryptosporidium: Cryptosporidium andersoni, Cryptosporidium bovis, Cryptosporidium ryanae, Cryptosporidium xiaoi e Cryptosporidium parvum. A espécie mais predominantemente encontrada foi C. andersoni (41,38%) e a menos foi C. parvum (10,34%). Abstract The aim of this study was to identify Cryptosporidium species found in cattle and sheep in Paraná, southern region of Brazil. Individual fecal samples from 458 bovines and 101 sheep were submitted for molecular analysis by PCR and nested PCR using specific primers for sequences of the 18S ribosomal unit (rRNA). Positive samples were analyzed using restriction fragment length polymorphism (RFLP), followed by genetic sequencing for species confirmation. The occurrence of Cryptosporidium was 11.27% (63/559). The highest occurrence was detected in lambs (12/59, 20.33%). From the 63 positive samples, it was possible to identify the species in 58 of them by RFLP and genetic sequencing. Five species of Cryptosporidium were identified: Cryptosporidium andersoni, Cryptosporidium bovis, Cryptosporidium ryanae, Cryptosporidium xiaoi, and Cryptosporidium parvum. The most prevalent species was C. andersoni (41.38%) and the least predominant was C. parvum (10.34%). The most abundant species of Cryptosporidium in dairy calves were C. andersoni (11/25) and C. ryanae (6/25). Of the 17 positive sheep, nine (52.94%) were infected with C. andersoni. This finding is the first report on the occurrence of C. andersoni in naturally infected sheep in Brazil and the first observation of a high absolute occurrence of this Cryptosporidium species in sheep. Abstract Introduction Chronic intestinal constipation (CIC) presents an incidence of 2.6 to 30.7% in the overall population and due to the social reality imposed by the coronavirus pandemic, some behavior changes in the Brazilian population occurred that might or not be associated with alterations of CIC prevalence. Objective To assess CIC incidence in medical students before and during the COVID-19 pandemic in Brazil in a private higher educational institution in the city of São Paulo, state of São Paulo. Methods Clinic data were collected through Google Forms software from the same students seeking to analyze the variables before (year of 2019) and during the coronavirus pandemic. The data were: age, sex, body mass index, constipation referred in a subjective way and confirmed through the ROME III criteria, feces consistency and anxiety and/or depression during the pandemic. Results A total of 126 medical students from a private higher education institution from São Paulo, SP were included. The average age was 22.9 years old, 70.6% were female and the average BMI was 23.3 kg/m2. Regarding the ROME III criteria, 32.5% presented > 2 in 2019 and 42.1% during the pandemic. Concerning the feces consistency, 31.75 and 35.71% presented dry Bristol 1 feces or in both periods, respectively. Abstract We aimed to compare the visual search strategies of young soccer players with different positional roles. Data was collected with the Mobile Eye Tracking — XG to verify gaze behaviour through visual focus. At the laboratory, an 11-a-side match play video-based soccer simulation from a third-person perspective was presented to the participants. Players were grouped according to their positional roles: defenders (n= 6), midfielders (n= 6) and forwards (n= 5). Visual search strategies were classified according to five fixation locations: “player in possession”, “ball”, “teammates”, “opponents” and “space”. The number of fixations and the fixation duration in each location was assessed. One-way ANOVA was performed to compare mean values. Midfielders and forwards showed significant differences in the number of fixations (15.50± 5.85 vs. 27.80± 7.36; p = 0.023) and fixation duration (8.00± 3.11 secs vs. 20.10± 8.47 secs; p = 0.016) for the category “ball”, as well as in the number of fixations (22.00± 5.62 vs. 34.60± 1.81; p = 0.044) for “teammates”. No other differences were observed. We concluded that the positional role was not relevant to the visual search strategy adopted by young soccer players. This finding is important for coaches and researchers to understand the cognitive and perceptual skills of young soccer players. It can also help coaches design specific training with appropriate stimuli for the players’ development.

Participaram 160 pacientes com sintomas e/ou diagnóstico de Covid-19 que procuram assistência à saúde no estado. Os dados foram coletados por meio de um questionário eletrônico autoaplicável no período de julho a outubro de 2020, sendo a amostra caracterizada com auxílio do software SPSS versão 26; e os dados referentes à perspectiva dos pacientes sobre a assistência à saúde, submetidos à análise temática de conteúdo. Dentre os resultados, emergiram três categorias: Satisfação do paciente sobre a assistência à saúde; Perspectivas do paciente diante das condutas e práticas clínicas no atendimento à saúde; Sentimentos expressados pelo paciente ante o atendimento à saúde, sintomas e diagnóstico para a Covid-19. Destacou-se a ausência de apoio e suporte emocional para o paciente, além de déficits dos recursos humanos, materiais e estrutura, interferindo no gerenciamento do cuidado de saúde. Considera- se que compreender a perspectiva dos pacientes atendidos nos serviços de saúde poderá contribuir para a melhoria da qualidade do atendimento nos serviços de saúde.

It was highlighted the absence of emotional support to the patient, besides human resources, structure and material deficits, which interfere in health care management. It must be taken into consideration that understanding the patient’s perspective in health services can contribute to the quality improvement of health care.

Resumo O objetivo deste estudo foi avaliar as características genotípicas de Toxoplasma gondii isolados de galinhas caipiras da Região Metropolitana de Goiânia, Goiás, Região Centro Oeste do Brasil. A soroprevalência foi de 96% dos animais, determinada por hemaglutinação indireta. As amostras de cérebro e coração foram processadas através da digestão péptica para o bioensaio em camundongos. Os tecidos foram homogeneizados, e as amostras resultantes foram analisadas por reação em cadeia da polimerase (PCR), que possibilitou a detecção do DNA do parasito em 64% deles. Por meio do bioensaio em camundongos, foi possível detectar 15 isolados, 8 deles apresentando taquizoítos na lavagem peritoneal e 7 apresentando cistos cerebrais. A determinação dos genótipos de T. gondii foi realizada por PCR-RFLP com os seguintes marcadores: SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, alt. SAG2, Apico e CS3. Foi possível definir 3 genótipos, incluindo o ToxoDB # 65 e dois deles ainda não foram descritos na literatura. Portanto, conclui-se que os isolados obtidos na região metropolitana de Goiânia apresentaram diversidade genética relativamente baixa.

Abstract The aim of this study was to evaluate the genotypic characteristics of Toxoplasma gondii isolated from free-range chickens in the metropolitan area of Goiânia, Goiás, in Brazil’s central-west region. The seroprevalence rate was found to be 96%, according to an indirect hemagglutination assay. Brain and heart samples were processed by peptic digestion for a mice bioassay. The tissues were homogenized and the resulting samples were subjected to polymerase chain reaction (PCR), which revealed that 64% of them contained the parasite's DNA. The mice bioassay revealed 15 isolates, 8 of them tachyzoites isolates from the peritoneal lavage and 7 from brain cysts. T. gondii genotypes were determined through PCR-RFLP, using the following markers: SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, alt. SAG2, Apico and CS3. Three genotypes were identified, inclued ToxoDB #65, and the other two are not yet described in the literature. Hence, we conclude that the isolates obtained from the metropolitan area of Goiânia showed relatively low genetic diversity.

Resumo O gado é uma fonte importante de espécies zoonóticas de Cryptosporidium para o homem. O objetivo deste estudo foi investigar a presença de Cryptosporidium, identificar a espécie e determinar os fatores de risco relacionados ao meio ambiente, aos animais e ao manejo em bezerros leiteiros em oito estados brasileiros. Um total de 408 amostras fecais de bezerros, com idade entre 1 e 60 dias, foram analisadas. Um questionário epidemiológico foi preenchido. A triagem das amostras foi realizada pela técnica de Ziehl-Neelsen, e as amostras positivas foram submetidas à “nested” PCR. As espécies de Cryptosporidium foram identificadas pela técnica de PCR-RFLP, utilizando-se as enzimas SSPI, ASEI e MBOII. A técnica de Ziehl-Neelsen mostrou que 89,7% (35/39) das fazendas e 52,9% (216/408) das amostras foram positivas. Por meio de nested PCR, esses protozoários foram detectados em 54,6% das amostras. As 56 amostras submetidas à PCR-RFLP apresentaram Cryptosporidium parvum. Houve maior prevalência do parasita em animais de 7 a 28 dias (62,6%). Diarreia, idade entre sete e 28 dias, e fonte de água mineral foram fatores associados ao risco de infecção. O sistema de manejo do tipo “casinha” para bezerros foi associado à redução da infecção. Esses achados demonstram o alto nível de Cryptosporidium spp. em circulação nos rebanhos bovinos e o predomínio da espécie C. parvum.

Abstract Cattle are an important source of zoonotic species of Cryptosporidium for humans. The aim of this study was to investigate the presence of Cryptosporidium, identify the species and determine the risk factors relating to environment, animals and management among dairy calves in eight Brazilian states. A total of 408 fecal samples from calves aged 1-60 days were analyzed. An epidemiological questionnaire was completed. Sample screening was performed using Ziehl-Neelsen technique and the positive samples were subjected to nested PCR. Cryptosporidium species were identified by means of the PCR-RFLP technique, using SSPI, ASEI and MBOII enzymes. The Ziehl-Neelsen technique showed that 89.7% (35/39) of the farms and 52.9% (216/408) of the samples were positive. Through nested PCR, these protozoa were detected in 54.6% of the samples. The 56 samples subjected to PCR-RFLP presented Cryptosporidium parvum. There was higher prevalence of the parasite in animals aged 7 to 28 days (62.6%). Diarrhea, ages between seven and 28 days and a spring water source were factors associated with the risk of infection. The calf hutch-type management system was associated with reduced infection. These findings demonstrate the high level of Cryptosporidium spp. circulation in cattle herds and the predominance of the species C. parvum.

Resumo Métodos moleculares como a Copro-PCR se destacam no diagnóstico de T. gondii por serem altamente sensíveis e específicos, podendo distinguir T. gondii de outros coccídeos morfologicamente semelhantes. O objetivo foi a detecção da coproprevalência de Toxoplasma gondii por reação em cadeia da polimerase, em 149 amostras fecais de gatos errantes e domiciliados, utilizando-se três marcadores distintos (B5-B6, 18S e 529pb RE). Oocistos de T. gondii/H. hammondi foram detectados em 15,4% pelos exames parasitológicos de fezes (EPF), e 4% desses oocistos foram identificados positivamente como T. gondii pela Copro-PCR. A presença de material genético de T. gondii foi detectada em 16,1%, mas 12% das amostras positivas pelo Copro-PCR foram negativas no EPF. As amostras com resultados discordantes foram submetidas a uma nova Copro-PCR com os marcadores 18S e 529 e, das 17 amostras, 9 continham material genético de T. gondii. A comparação do EPF com os métodos moleculares mostrou que esse último foi mais sensível, pois detectou 22,1%, enquanto o EPF detectou 15,4%. Isso demonstra a alta sensibilidade e especificidade da Copro-PCR, principalmente com a associação de marcadores (k = 0,809); mas também confirma a importância do uso de técnicas moleculares em laboratórios, uma vez que a Copro-PCR foi capaz de detectar amostras consideradas negativas pelo EPF.

Abstract Molecular methods such as Copro-PCR stand out in the diagnosis of T. gondii, because they are highly sensitive and specific, and can distinguish T. gondii from other morphologically similar coccids. The purpose was the detection of Toxoplasma gondii copro-prevalence by polymerase chain reaction in 149 fecal samples from stray and domiciled cats, using three distinct markers (B5-B6, 18S and 529bp RE). Oocysts of T. gondii/H. hammondi were detected in 15.4% by parasitology fecal tests (PFT), and 4% of these oocysts were positively identified as T. gondii by Copro-PCR. The presence of T. gondii genetic material was detected in 16.1%, but 12% of the samples that tested positive by Copro-PCR were negative in PFT. Samples with discordant results were subjected to a new Copro-PCR with 18S marker and a 529, and of the 17 samples, 9 contained T. gondii genetic material. A comparison of the PFT and the molecular methods showed the latter was more sensitive, since it detected 22.1% while the PFT detected 15.4%. Demonstrating the high sensitivity and specificity of the Copro-PCR, particularly with the association of primers (k=0.809), but also confirms the importance of using molecular techniques in laboratories, since Copro-PCR was able to detect samples considered negative by PFT.

RESUMO No futebol, a fadiga é tradicionalmente investigada sob uma perspectiva neuromuscular e metabólica. Entretanto, já que o futebol é uma das modalidades esportivas com maior demanda cognitiva, acredita-se que a exigência de elevados níveis atencionais e as frequentes tomadas de decisões sejam fatores que influenciem o desempenho dos jogadores de futebol. Sendo assim, o objetivo do presente estudo foi verificar, através de uma revisão sistemática, os efeitos da fadiga mental sobre o desempenho físico, técnico, tático e cognitivo dos jogadores de futebol. Foram realizadas buscas nas bases de dados eletrônicas Pubmed, Web of Science e Scopus até 30 de abril de 2018. Incluiram-se artigos que utilizaram um protocolo de fadiga mental através das tarefas cognitivas realizadas previamente a uma tarefa física ou cognitiva relacionada ao futebol. Somente foram selecionados estudos que apresentaram um desenho experimental com a condição controle (sem fadiga mental) e experimental (com fadiga mental). No total, foram selecionados seis estudos que atenderam aos critérios estabelecidos, um estudo por busca reversa e outro por indicação dos autores. Os resultados indicaram menores distâncias percorridas em testes físicos, enquanto que os efeitos da fadiga mental sobre o desempenho físico em jogos reduzidos não foram evidentes. Em testes técnicos, houve maior número de penalizações em passes e menor precisão e velocidade da bola nos chutes na condição de fadiga mental. Em relação às variáveis táticas, foram encontrados efeitos prejudiciais da fadiga mental sobre a sincronização entre os jogadores das equipes e o desempenho tático individual em ações defensivas. Em testes cognitivos, também foram verificados efeitos negativos sobre o tempo e a precisão para a tomada de decisão dos jogadores em testes de vídeo. A partir dos resultados dos estudos analisados, torna-se possível inferir que a fadiga mental é um fator que influencia negativamente o desempenho dos jogadores de futebol. ABSTRACT Fatigue in soccer players is traditionally investigated based on neuromuscular and metabolic factors. However, given that soccer is one of the sports that has the highest cognitive demand, it is believed that players' performance might also be influenced by the high levels of attention, and frequent decision-making required in soccer. This systematic review aimed to verify the effects of mental fatigue on physical, technical, tactical and cognitive performance of soccer players. We searched in the electronic databases Pubmed, Web of Science and Scopus, for articles published up to 30 April 2018. We included articles that used a protocol of mental fatigue through cognitive tasks performed prior to a physical or cognitive task related to soccer. Only studies that presented an experimental design with the control condition (without mental fatigue) and the experimental condition (with mental fatigue) were selected. A total of six articles met the inclusion criteria, one study by backward reference search and other through the authors' indication. The results showed smaller distances covered in physical tests, while the effects of mental fatigue on physical performance in small-sided games were not clear. In technical tests, there were more penalties in passes and less accuracy and speed when kicking the ball when players were in conditions of mental fatigue. Regarding the tactical variables, it was found that mental fatigue had a detrimental effect on the synchronization between team players and on individual tactical performance in defensive actions. In cognitive tests, based on video recordings of game play, negative effects on the players' speed and accuracy of decision-making were observed. According to the results of the literature search, it can be inferred that mental fatigue is a factor that has a negative influence on soccer performance. Level of evidence II; Systematic review.

RESUMEN En el fútbol, la fatiga es tradicionalmente investigada desde una perspectiva neuromuscular y metabólica. Entretanto, ya que el f'útbol es una de las modalidades deportivas con mayor demanda cognitiva, se cree que la exigencia de elevados niveles de atención y las frecuentes tomas de decisión sean factores que influencien el desempeño de los jugadores de fútbol. Siendo así, el objetivo del presente estudio fue verificar, a través de una revisión sistemática, los efectos de la fatiga mental sobre el desempeño físico, técnico, táctico y cognitivo de los jugadores de fútbol. Fueron realizadas búsquedas en las bases de datos electrónicas Pubmed, Web of Science y Scopus hasta 30 de abril de 2018. Se incluyeron artículos que utilizaron un protocolo de fatiga mental a través de las tareas cognitivas realizadas previamente a una tarea física o cognitiva relacionada al fútbol. Solamente fueron seleccionados estudios que presentaron un diseño experimental con la condición control (sin fatiga mental) y experimental (con fatiga mental). En total, fueron seleccionados seis estudios que atendieron los criterios establecidos, un estudio por búsqueda reversa y otro por indicación de los autores. Los resultados indicaron menores distancias recorridas en tests físicos, mientras que los efectos de la fatiga mental sobre el desempeño físico en partidos reducidos no fueron evidentes. En tests técnicos, hubo mayor número de penalizaciones en pases y menor precisión y velocidad de la pelota en los tiros en la condición de fatiga mental. Con relación a las variables tácticas, fueron encontrados efectos perjudiciales de la fatiga mental sobre la sincronización entre los jugadores de los equipos y el desempeño táctico individual en acciones defensivas. En tests cognitivos, también fueron verificados efectos negativos sobre el tiempo y la precisión para la toma de decisión de los jugadores en tests de vídeo. A partir de los resultados de los estudios analizados, se hace posible inferir que la fatiga mental es un factor que influencia negativamente el desempeño de los jugadores de fútbol. Resumo O objetivo deste estudo foi relatar um surto de toxoplasmose humana em uma instituição de pesquisa em Londrina, Paraná, no período de dezembro de 2015 a fevereiro de 2016. Amostras de sangue de 26 indivíduos sintomáticos foram coletadas e o imunoensaio de quimioluminescência de micropartículas foi realizado para detectar IgM, IgG e teste de avidez de IgG específica em laboratório oficial. Um total de 20 pessoas com sintomas e sorologia compatíveis com toxoplasmose aguda (IgM positiva e IgG com baixa avidez) foi selecionado como casos, enquanto 45 funcionários assintomáticos que trabalhavam nas mesmas equipes e durante os mesmos turnos foram utilizados como controles. Todos os participantes da investigação responderam a um questionário epidemiológico. Foram coletadas três amostras de água e uma de lodo das cisternas de abastecimento da instituição, 10 de solo, 11 de vegetais, três amostras de fezes de gato e um cadáver de filhote felino doméstico para detecção de T. gondii pela reação em cadeia da polimerase (PCR). Após análise dos dados epidemiológicos, o consumo de hortaliças no restaurante da instituição foi a única variável associada à ocorrência da doença. Em resultados laboratoriais, todas as amostras apresentaram resultados negativos a PCR. O rápido reconhecimento do surto, notificação e investigação prematura poderia ter quebrado a cadeia de transmissão, evitando assim o surgimento de novos casos. Além disso, a adoção de boas práticas de manipulação de alimentos poderia ter impedido a ocorrência do surto.

Abstract The objective of this study was to report an outbreak of human toxoplasmosis at a research institution in Londrina, Paraná, from December 2015 to February 2016. Blood samples from 26 symptomatic individuals were collected and the microparticle chemiluminescence immunoassay was performed to detect IgM, IgG and specific IgG avidity test in the official laboratory. A total of 20 people with symptoms and serology compatible with acute toxoplasmosis (IgM positive and IgG with low avidity) were selected as cases, while 45 asymptomatic employees working in the same teams and during the same shifts were selected as controls. All the participants of the investigation answered an epidemiological questionnaire. Three samples of water and one sludge from the institution's supply cisterns, 10 soil samples, 11 plant samples, three cat fecal samples and one domestic feline cadaver were collected for analysis of the polymerase chain reaction (PCR) for T. gondii. After analyzing the epidemiological data, the consumption of vegetables in the restaurant of the institution was the only variable associated with the occurrence of the disease. In laboratory results, all the samples showed negative results to PCR. The rapid recognition of the outbreak, early notification and investigation could have broken the chain of transmission early, thus preventing the emergence of new cases. In addition, the adoption of good food handling practices could have prevented the occurrence of the outbreak.

Resumo Cryptosporidium é um protozoário com uma grande variedade de hospedeiros, incluindo os seres humanos. No entanto, poucas espécies têm sido descritas em aves (Cryptosporidium meleagridis, C. baileyi, C. galli e C. avium). O objetivo do presente estudo foi investigar a ocorrência de Cryptosporidium spp. em fezes de pombas-de-bando (Zenaida auriculata), e realizar a caracterização molecular dos isolados. Um total de 196 animais de ambos os sexos foram capturados, eutanasiados e o conteúdo intestinal recolhido para extração de DNA. Após a extração, realizou-se uma nested-PCR (nPCR), que amplifica um fragmento do gene 18S rRNA do Cryptosporidium spp.. Os fragmentos obtidos foram purificados e encaminhados para sequenciamento. Os resultados da n-PCR revelaram 30 animais (15.3%) positivos para Cryptosporidium spp.. Quanto ao sexo dos animais não foram observadas diferenças estatísticas significativas (p > 0.05). Somente 15 de 30 amostras positivas foram sequenciadas com sucesso e as espécies determinadas, das quais, 13 (86.7%) e 2 (13.3%) foram C. meleagridis e C. galli, respectivamente. Esse é o primeiro estudo com caracterização molecular de Cryptosporidium de fezes de pombas-de-bando (Z. auriculata), e propõe serem esses animais potenciais fonte de infecção de C. meleagridis para humanos.

Abstract Cryptosporidium is a protozoan parasite with a wide range of hosts, including humans. However, only a few Cryptosporidium species have been described in birds (C. meleagridis, C. baileyi, C. galli and C. avium). The aim of this study was to investigate the occurrence of Cryptosporidium spp. in feces of eared doves (Zenaida auriculata), followed by molecular characterization of the parasite. A total of 196 animals of both sexes were trap-captured; the animals were culled and the intestinal contents were collected for DNA extraction. After extraction, a nested-PCR (nPCR), which amplifies a fragment of the 18S rRNA gene of Cryptosporidium spp., was performed. The amplicons obtained were purified and sequenced. PCR analysis revealed that 30 animals (15.3%) were positive for Cryptosporidium spp. There was no significant sex-dependent enrichment of Cryptosporidium occurrence (p > 0.05). Only 15 out of the 30 positive samples were successfully sequenced and their species determined, of which, 13 (86.7%) and 2 (13.3%) were C. meleagridis and C. galli, respectively. Herein, we present for the first time a molecular characterization of Cryptosporidium from feces of eared doves (Z. auriculata) and propose that these birds are a potential source of C. meleagridis infection in humans.

Resumo Cryptosporidium e Giardia são protozoários causadores de diarreia em animais e humanos. A caracterização molecular destes protozoários em esgoto pode prover dados ainda desconhecidos da ocorrência de espécies. O objetivo do presente estudo foi monitorar a ocorrência de Giardia e espécies de Cryptosporidium em esgoto bruto e tratado em uma estação de tratamento de esgoto (ETE) de Londrina, Paraná. Amostras de esgoto bruto e tratado foram coletadas no período de um ano, com periodicidade quinzenal. A ocorrência destes protozoários foi caracterizada por meio de concentração das amostras e posterior extração de DNA seguida de nested-PCR para amplificação de fragmentos dos genes 18S rRNA de Giardia e 18S rRNA de Cryptosporidium. A caracterização das espécies de Cryptosporidium foi realizada por meio de análise por polimorfismo de comprimento do fragmento de restrição (RFLP) dos produtos obtidos. Foram coletadas no total 25 amostras de cada, esgoto bruto e esgoto tratado. Para Giardia, todas as amostras de esgoto bruto e 76% das de esgoto tratado foram positivas. Cryptosporidium esteve presente em 84% das amostras de esgoto bruto e em 8% do tratado. No esgoto tratado foi encontrado apenas C. muris, já nas amostras de esgoto bruto foram encontradas cinco espécies: C. muris, C. hominis, C. baileyi, C. suis e C. parvum em 100%, 19%, 9%, 9% e 4%, respectivamente. A presença de espécies mistas foi observada em 19,04% das amostras. A presença de Giardia e Cryptosporidium em esgoto tratado pode pôr em risco a saúde humana. A discriminação de espécies de Cryptosporidium revelou a presença de espécies zoonóticas incomuns como C. suis e C. muris.

Abstract Cryptosporidium and Giardia are protozoan parasites that cause diarrhea in humans and animals. Molecular characterization of these pathogens in sewage may provide insight on their occurrence and prevalence in Brazil. This study aimed to investigate the presence of Giardia and Cryptosporidium in raw and treated sewage from Londrina, Paraná, Brazil. Samples were collected every two weeks during a year. Samples were concentrated, then DNA was extracted and subjected to a nested PCR targeting the Giardia 18S rRNA gene and the Cryptosporidium 18S rRNA gene. Species of Cryptosporidium were characterized by restriction fragment length polymorphism (RFLP). All raw sewage and 76% of the treated sewage were positive for Giardia; 84% of raw sewage samples and 8% of treated sewage were positive for Cryptosporidium. C. muris, C. hominis, C. baileyi, C. parvum and C. suis were detected in 100%, 19%, 9%, 9% and 4% of raw sewage, respectively. C. muris was the only species found in treated sewage. Multiple species of Cryptosporidium were present in 19.04% of the raw sewage. Treated sewage water can pose a threat to human health. The speciation of Cryptosporidium revealed the presence of non-common zoonotic species as C. suis and C. muris.

RESUMO: A presença de anticorpos anti-Neospora caninum em bovinos de corte, abatidos na região norte do estado do Paraná, Brasil, foi avaliada. Foram coletadas 401 amostras de sangue, sendo 281 amostras no município de Rolândia e 120 no municipio de Borrazopolis, entre os meses de abril de 2015 e novembro de 2016. Do total de amostras, 289 eram de fêmeas e 112 amostras de machos, na faixa etária de um ano e meio até oito anos de idade. Foi realizada a reação da imunofluorescência indireta (RIFI) utilizando ponte de corte de 1:100. Em seguida, foram tabulados as variáveis para análise estatística (testes exato de Fisher e do qui-quadrado, p≤0,05). A análise mostrou que das 401 amostras, 37 foram positivas para N. caninum, indicando uma prevalência de 9,2 % e os títulos observados foram 1:100 (16), 1:200 (14) e 1:400 (7). As variáveis sexo, idade e local não diferiram estatisticamente (p>0,05). Nossos resultados demonstram uma soro-ocorrência de N. caninum em bovinos abatidos na região norte do Paraná.

ABSTRACT: The presence of anti-Neospora caninum antibodies in beef cattle slaughtered in the northern region of the state of Paraná, Brazil, was evaluated. A total of 401 blood samples were collected; 281 samples from the municipality of Rolândia and 120 from the municipality of Borrazópolis, between April 2015 and November 2016. Of the total samples, 289 were from females and 112 from males, aged one and a half to eight years. Indirect fluorescent antibody test (IFAT) was performed, using a cut-off of 1:100. Variables were tabulated for statistical analyses (Fisher’s exact test and chi-square tests, p≤0.05). The analysis showed that of the 401 samples, 37 were positive for N. caninum, indicating a prevalence of 9.2 %, and observed titers were 1:100 (16), 1:200 (14), and 1:400 (7). The variables sex, age, and location did not differ statistically (p>0.05). Our results showed a sero-occurrence of N. caninum in cattle slaughtered in the northern region of the state of Paraná.

ABSTRACT The aim of this study was to perform a systematic review of scientific papers that used spatial analysis tools in cases of leishmaniasis, in Brazil. The search for articles was carried out in PubMed, SciELO, Scopus and Web of Science databases. The keywords used in the identification of the articles were Thematic map AND Leishmaniasis, Spatial analysis AND Leishmaniasis, and Geoprocessing AND Leishmaniasis, in English language. A total of 360 articles were found, and 11 of them were analyzed after screening by title and abstract as well as reading of the full articles. The States studied were Sao Paulo, Acre, Maranhao, Piaui, Minas Gerais, Parana and Tocantins. Cutaneous leishmaniasis occurred predominantly in rural areas, with clusters in forest reserve regions or modified forest areas. Conversely, visceral leishmaniasis mainly occurred in peripheral and central urban areas associated with poorer environments and urban infrastructure, including worse sanitation. We conclude that the spatial distribution of leishmaniasis is closely related to the living environment of the risk population. The analyzed articles associated geospatial data with some risk factors for the disease, pointing out the locations where most cases occur, creating a relevant source to define control strategies.
