Abstract Introduction: HIV pre-exposure prophylaxis (PrEP) has been shown to reduce the incidence of HIV, but its adoption in Colombia is still beginning. To advance the implementation of PrEP as a public health intervention, it is necessary to know the interest of populations at risk of HIV and the factors that would facilitate its use. Objective: Identify factors related to interest and intention to take PrEP in gay/bisexual men. Methodology: Analytical cross-sectional study in a non-probabilistic sample in main cities of Colombia. Sociodemographic characteristics, knowledge, motivations, behavioral skills, and psychosocial factors were explored; logistic models were used to estimate the relationship of these factors with interest and intention to use PrEP. Results and conclusions: 552 gay/bisexual men participated, 301 (54%) recruited online and 251 in person. Only 40% of the sample knew of the existence of PrEP. 57.4% (95% CI: 54.9%-62.4%) reported interest and 52.9% (95% CI: 48.5%-57.3%) intention to use PrEP. Interest and intention to use PrEP were related to positive attitudes and norms toward PrEP, as well as greater skills related to PrEP services. Stigma toward PrEP and depressive symptoms were related to lower interest and intention. Our results suggest the need to promote information about PrEP, especially in populations with more social disadvantages and who experience more stigma, as well as to develop community interventions that support motivations and skills to use PrEP.

ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others. publications problem uptodate up date classifications context exception (CTFB http//fauna.jbrj.gov.br/, httpfaunajbrjgovbr http //fauna.jbrj.gov.br/ , jbrj gov br (http://fauna.jbrj.gov.br/) 2015 Brazil 80 specialists 1 2024 133691 133 691 133,69 125138 125 138 125,13 82.3%, 823 82 3 (82.3% 102000 102 000 102,00 7.69%, 769 7 69 (7.69% 11000 11 11,00 . 3,567 3567 567 (3,56 2,292 2292 2 292 (2,29 1,833 1833 833 (1,83 1,447 1447 447 (1,44 1000 1,00 831 (83 628 (62 606 (60 520 (52 50 users science health biology law anthropology education others http//fauna.jbrj.gov.br/ faunajbrjgovbr //fauna.jbrj.gov.br (http://fauna.jbrj.gov.br/ 201 8 202 13369 13 133,6 12513 12 125,1 82.3% (82.3 10200 10 00 102,0 7.69% 76 6 (7.69 1100 11,0 3,56 356 56 (3,5 2,29 229 29 (2,2 1,83 183 83 (1,8 1,44 144 44 (1,4 100 1,0 (8 62 (6 60 52 (5 5 http//fauna.jbrj.gov.br (http://fauna.jbrj.gov.br 20 1336 133, 1251 125, 82.3 (82. 1020 0 102, 7.69 (7.6 110 11, 3,5 35 (3, 2,2 22 (2, 1,8 18 (1, 1,4 14 4 ( 82.
Abstract Globally, HIV is one of the major public health challenges of our time. In Latin America, the prevalence of HIV for men who have sex with men (MSM) and transgender women (TGW) is 30 times higher than for the general population. Colombia has the second highest rate of people with HIV in the region. PrEP therapy has been successfully implemented in other Latin American contexts. However, for an intervention such as PrEP to be effective and feasible in the Colombian context, it is necessary to explore how the social determinants of health (SDH) could potentially affect its implementation and uptake. To our knowledge, no other study in the country has explored this issue. Through a secondary analysis of qualitative data, we fill this gap of knowledge and generate evidence about the health inequities that could potentially limit the adoption of PrEP to prevent HIV in MSM and MTG populations. Objectives: To explore how the SDH affect the intention of health providers to implement PrEP as an HIV prevention strategy in Colombia. Methods: The study analyzed secondary data from interviews conducted with health providers in Colombia. A critical hermeneutic approach was used, and an SDH conceptual framework was followed for the thematic analysis. Results: The findings showed that stigma, cultural behaviours, armed conflict, lack of access to health services, and health literacy affect the adoption of PrEP in the country. Conclusions: For the successful implementation of PrEP in the country, it is necessary to consider the Colombian social and political context, especially in areas affected by violence and concentrated historical inequities.

Asbtract Introduction This study aimed to determine whether cytokine receptor-like factor 2 (CRLF2) antigen expression evaluated using multiparametric flow cytometry (MFC) could predict the genotype of CRLF2 and Janus kinase 2 (JAK2) status for application in the diagnosis of pediatric B-cell precursor acute lymphoblastic leukemia (BCP-ALL). Methods A total of 321 BCP-ALL bone marrow samples were collected, 291 at diagnosis and 13 at first relapse, while 17 samples were excluded due to low cellular viability. The CRLF2 antigen expression was evaluated using flow cytometry (percentage of positivity and median fluorescence intensity [MFI]). The CRLF2 transcript levels were assessed via quantitative reverse transcription polymerase chain reaction using SYBR Green. The CRLF2 rearrangements (CRLF2-r) were identified using the CRLF2 break-apart probe via fluorescence in situ hybridization. Sanger sequencing was performed to identify the JAK2 exon 16 mutations. Results We observed that 60 of the 291 cases (20.6%) presented CRLF2 antigen positivity, whereas the CRLF2 transcript overexpression was found in 19 of 113 cases (16.8%). The JAK2 mutation was found in four out of 116 cases (3.4%), all of which had CRLF2 ≥10% of positive cells and intermediate or high MFI (p < 0.0001). In addition, in the 13 cases with the CRLF2-r, a positive correlation was found with the CRLF2 antigen intermediate (61.5%) MFI (p= 0.017). Finally, the CRLF2-positive antigen was identified in the BCP-ALL subclones. Conclusion The identification of the CRLF2 antigen using the MFC, based on the percentage of positivity and MFI values, is a useful tool for predicting JAK2 mutations and CRLF2-r. receptorlike receptor like CRLF (CRLF2 MFC (MFC JAK (JAK2 Bcell B cell BCPALL. Moore (casi amenazada, NT), Miconia neomicrantha Judd & Skean y Guarea kunthiana A. Juss. (Preocupación Menor, LC).

Objective The global burden of diabetes mellitus will impact strongly American countries in the coming decades. Type 2 diabetes mellitus (T2DM) is a multifactorial disease and the basis for its genetic susceptibility remains not fully understood. Different population studies have demonstrated that variants of the TCF7L2 gene are strongly associated with an increased risk of T2DM. Moreover, institutions or countries with limited budget to conduct genetic research need cost effective methods for detecting DNA variants. Subjects and methods We standardized a rapid and simple allele-specific PCR method for genotyping the rs12255372 single nucleotide polymorphism (SNP) in a pilot study exploring the association of three TCF7L2 polymorphisms (rs7903146, rs12255372 and DG10S478) with T2DM in 70 patients and 73 controls from Venezuela. Results The performance of the designed allele-specific PCR reaction for rs12255372 genotyping was reliable and accurate. Patients carrying the TCF7L2 rs7903146 T allele (CT + TT genotypes) and heterozygous CT genotype had a significantly higher risk for T2DM (OR = 2.9 and 2.3, respectively). Although rs12255372 and DG10S478 risk alleles predominated in T2DM group no statistical significance was found. Conclusions We developed a novel allele-specific PCR method for easier and rapid detection of rs12255372 polymorphism without the use of expensive instrumentation and reagents. Our study in a relatively small sample of the Venezuelan population replicated the association of the rs7903146 SNP with T2DM. Further studies with larger sample size and more biochemical data should be conducted to explore the genetic basis of T2DM susceptibility in Venezuela.

Codon 72 polymorphism of the tumor suppressor gene TP53 has been associated with a higher risk in the development of several types of cancer. The polymorphism results in a variant protein with either an arginine (CGC) or a proline residue (CCC). The aim of this study was to analyze the association of the TP53 codon 72 polymorphism with the risk of developing gastric cancer in a high-risk population from the central-western region of Venezuela. DNA was extracted from paraffin-embedded gastric adenocarcinoma biopsies (n = 65) and endoscopic biopsies from chronic gastritis patients (n = 87). TP53 codon 72 polymorphism was determined by PCR-RFLP from all samples. Patients with gastric cancer had a significantly higher frequency (P = 0.037) of the Arg allele than those with chronic gastritis. A logistic regression analysis suggested that Arg carrier individuals had a 4.6-fold higher risk (95% CI 1.0-21.3) of developing gastric cancer. An increment of the Arg/Arg genotype was observed in poor-differentiated gastric adenocarcinoma (OR: 3.1; 95% CI 1.0-9.2), and of the Arg/Pro genotype in well/ moderate-differentiated adenocarcinoma samples (OR: 3.5; 95% CI 1.1-11.0), when comparing within the gastric cancer samples; and the last group also when contrasting it with chronic gastritis patients (OR: 2.4; 95% CI 1.1-5.2). The results of this study suggest that the carriage of the Arg allele could be associated with the development of gastric cancer in this Venezuelan population.

Telomeres are nucleoprotein complexes that protect the ends of eucaryotic chromosomes from degradation and recombination. In humans, telomeres measure 10-12 kbp in normal somatic cells, but they scarcely reach 1-2 kbp in tumor cells of rapid growth, due to the incomplete replication of these structures in each mitotic division. Telomeres shorten with age, which can be associated to genomic instability and to an increment of the risk of suffering from cancer. The standard method to measure the telomere length is the analysis of telomeric restriction fragments by Southern blot. However, a precise determination is not possible when the DNA is broken into small fragments or if it is scarce. In this work, a slot blot assay was adapted to quantify the relative content, instead of the length, of telomeric DNA from paraffin-embedded archival specimens of colon adenocarcinoma. The telomeric DNA content could be reproducibly measured with hardly 75 ng of highly degraded genomic DNA by chemiluminescent detection for hybridization.

No hubo muertes, y cuatro pacientes (7,5%) tuvieron secuelas.
Introduction: Colombia is a country with two whole IgG antivenom producers, but the expectancy of all the market are not fulfilled by different technical reasons. Objectives: To evaluate the efficacy and safety of a F(ab´)2 polyvalent antivenom produced in Mexico and of a new dosage regimen for Bothrops bites in Colombia. Methods: A clinical trial, including serum venom and antivenom measurements (ELISA), was performed during 9 months in 53 patients. Results: forty four patients were bitten by Bothrops asper in Antioquia and Chocó and 9 by B. atrox in Amazonas; on admission, all of them had nonclottable blood, 30 (56.6%) presented local and 24 (45.3%) systemic bleeding. The final envenoming grade was mild in 13 (24.5%), moderate in 30 (56.6%) and severe in 10 patients (18.9%). At the antivenom doses used in this study (5 vials for mild / moderate and 10 for severe envenoming), Antivipmyn Tri ® was 100% efficient to decrease significantly serum venom concentrations within the first treatment hour, and to stop local and systemic bleeding within 6-12 hours, 96.2% efficient to restore blood coagulation within 24 hours and 100% within 48 hours. Two patients (3.8%) had recurrence of coagulopathy without bleeding, and there were 12 recurrences of antigenaemia without clinical relevance. Ten (18.9%) patients suffered early mild adverse reactions to fabotherapy. There were no deaths and four patients (7.5%) presented sequelae. Conclusion: at the doses used in this study, Antivipmyn Tri® was efficient and safe for the treatment of Bothrops bites in Colombia.

There is an increasing need for investigations of the Brazilian native tree species, mainly the physiological quality of their seeds. To evaluate the desiccation tolerance and storability of Eugenia brasiliensis seeds, mature fruits collected at Mogi-Guaçu, SP, Brazil and their seeds were removed by washing and were dried at 36ºC until their water content was reduced from 48.9% (fresh seeds, wet basis) to 23.6% (final drying), totaling five drying levels. Samples of each drying level were stored at 7ºC in plastic bags for 270 days. Results showed that water content lower than 43.1% decreased both germinability[germination potential] and storability. Seeds with 48.9% water content showed 60% germination after 180 days and 19% after 270 days of storage inside plastic bags in a cold chamber. We concluded that E. brasiliensis seeds can be stored for 180 days at 7ºC and are intolerant to drying at 36ºC.

The physiological quality of seeds of native species is important to produce healthy saplings and therefore guarantee the success of programs to recover disturbed vegetation. This reinforces the necessity for investigating the physiological quality of those seeds. To evaluate the effects of different drying rates on the germination, moisture content and storability of Eugenia involucrata diaspores, mature fruits collected at Mogi Guaçu, SP, Brazil had their epi- and mesocarps removed by washing and were dried at 30, 40 or 50ºC until their water content was reduced from 57% (fresh diaspores) to 13% (final drying), totaling six drying levels. In a second experiment, diaspores had their moisture content reduced from 57% to 49%, at 30ºC, totaling six drying levels (0h, 1h, 2h, 3h, 4h and 5h), and were kept for 180 days in plastic bags under cold storage. The drying rate had no effect on tolerance to desiccation by E. involucrata diaspores; water contents lower than 51% decreased both germinability and storability. Diaspores can be stored for up to 180 days as long as their water content is reduced to 53% and they are kept inside plastic bags under cold storage.
