Orbitides are a class of small naturally occurring cyclic peptides with structural and functional diversities. Their chemical properties make this class feasible to be obtained by solid phase peptide synthesis (SPPS). Therefore, this synthetic accessibility enables useful application and facilitates the identification of analogues, bioactivity studies, and thus, enables them to be applied to obtain peptide libraries. The aim of this work was to investigate the effects of orbitides and their linear synthetic analogues on the migration of neonatal human foreskin fibroblasts. The screening of linear peptide analogues, originally designed from natural orbitides isolated from Jatropha species, demonstrated that some molecules (linear pohlianin B and linear ribifolin) have the potential to induce fibroblast migration and collagen deposition and may thus contribute to accelerating the processes of wound healing and tissue repair. These results also demonstrate the significance of using peptides as an important tool for the discovery of simple and novel drug scaffolds.

Actinoporinas constituem uma família de proteínas formadoras de poros com atividade hemolítica. A base estrutural para tal atividade parece depender do seu enovelamento correto, especialmente em torno de algumas regiões funcionais, incluindo um sítio de ligação a fosfocolina, uma região rica em resíduos de triptofano e o amino-terminal. Adicionalmente, diferentes condições de solvatação são reconhecidamente capazes de influenciar a formação de poros por actinoporinas, como observado para Sticolisina II (StnII) e Equinatoxina (EqtxII). Neste contexto, o presente trabalho visa caracterizar a influência de diferentes soluções no comportamento conformacional dessas proteínas, por meio de simulações de dinâmica molecular. Os dados obtidos fornecem insights estruturais sobre a dinâmica das actinoporinas em solução, caracterizando seu comportamento conformacional em nível atômico, de acordo com dados experimentais previamente obtidos para as atividades hemolíticas de StnII e EqtxII.

Actinoporins are a family of pore-forming proteins with hemolytic activity. The structural basis for such activity appears to depend on their correct folding. Such folding encompasses a phosphocholine binding site, a tryptophan-rich region and the activity-related N-terminus segment. Additionally, different solution conditions are known to be able to influence the pore formation by actinoporins, as for Sticholysin II (StnII) and Equinatoxin II (EqtxII). In this context, the current work intends to characterize the influence of distinct solution conditions in the conformational behavior of these proteins through molecular dynamics (MD) simulations. The obtained data offer structural insights into actinoporins dynamics in solution, characterizing its conformational behavior at the atomic level, in accordance with previous experimental data on StnII and EqtxII hemolytic activities.

O clássico e importante método do ácido pícrico, usado nas áreas de bioquímica e química de polímeros para a quantificação de grupos aminos, foi escolhido neste trabalho como modelo para investigar a importância do grau de solvatação de grãos poliméricos durante o protocolo analítico. Verificou-se que este método, proposto há cerca de três décadas atrás, falha na quantificação de grupos amino de peptidil-resinas contendo seqüência agregante e polar. Isto ocorre devido à solvatação insuficiente dos grãos quando apenas o CH2Cl2 é utilizado na etapa de ligação do ânion picrato e na subsequente etapa de lavagem. Demonstrou-se que a utilização nestas etapas, de soluções de CH2Cl2/DMF (dimetilformamida) e de CH2Cl2/EtOH permite uma determinação correta dos grupos amínicos de peptídil-resinas. Além da importância em si para o método da síntese de peptídeos em fase sólida, estes resultados representam também a primeira comprovação experimental da correlação quantitativa existente entre o grau de solvatação e a eficiência de um método analítico efetuado em grãos de resinas.

The classic and important picric acid method used in polymers biochemical and chemical fields of polymers for amine group quantification was chosen in this work as a model for evaluating the influence of the resin bead solvation during an analytical procedure. It was observed that this method, proposed almost three decades ago, failed to quantify amine groups of peptidyl-resin containing aggregating and polar sequence. This was due to inefficient solvation of resin beads when only CH2Cl2 was used for picrate anion binding and subsequent washing steps. It was demonstrated that the use of CH2Cl2/DMF (dimethylformamide) and CH2Cl2/EtOH solutions during these steps allows correct determination of peptidyl-resin amine groups. Besides the importance for the solid phase peptide synthesis methodology itself, these findings also represent the first quantitative demonstration of the relationship between solvation degree and the efficiency of a polymer-supported analytical method.

O método clássico de hidrólise para a quantificação de grupamentos aminoacílicos e peptídicos ligados a resinas em HCl 12 N : ácido propiônico foi reavaliada, estudando-se a influência da natureza da resina e do grupamento ligado à mesma. A estabilidade frente à hidrólise ácida foi dependente do amino acido C-terminal e a ordem de estabilidade ácida foi Phe > Val > Gly. Por outro lado, os dipeptídeos Ala-Gly, Ala-Val e Ala-Phe apresentaram maiores velocidades de hidrólise da resina, se comparadas com a dos correspondentes grupos aminoacílicos. Dentre as resinas testadas, a ordem de estabilidade foi: benzidrilamino-resina>p-metilbenzidrilamino-resina ≅4-(oximetil)-fenilacetamidometil-resina > copolímero de estireno clorometilado, contendo 1% de divinilbenzeno. Importante para a metodologia de síntese de peptídeos, os resultados demonstraram que tempos de hidrólise mais longos do que os propostos há anos na literatura (1 h a 130 °C e 15 min a 160 °C para os peptídeos ligados ao copolímero de estireno clorometilado, contendo 1% de divinilbenzeno), são necessários para uma clivagem quantitativa de alguns grupos ligados à resina. A ampla faixa de tempos de hidrólise observada variou de menos de 1 h até cerca de 100 h.

The classic hydrolysis procedure for quantification of resin-bound aminoacyl and peptidyl groups with 12 N HCl: propionic acid was reevaluated by studying the influence of the nature of the resin and the resin-bound group. Their stability during acid hydrolysis was dependent on the C-terminal amino acid, and the order of acid stability was Phe > Val > Gly. Otherwise, the dipeptides Ala-Gly, Ala-Val, and Ala-Phe displayed enhanced rates of hydrolysis of the resin if compared with their parent aminoacyl groups. Amongthe resins assayed, the order of acid stability was: benzhydrylamine-resin>p-methylbenzhydrylamine-resin ≅4-(oxymethyl)-phenylacetamidomethyl-resin > chloromethyl-copolymer of styrene-1%-divinylbenzene. Important for peptide synthesis method, the findings demonstrate that longer hydrolysis times than previously recommended in the literature (1 h at 130 °C and 15 min at 160 °C for peptides attached to the chloromethyl-copolymer of styrene-1%-divinylbenzene) are necessary for the quantitative acid-catalyzed cleavage of some resin-bound groups. The observed broad range of hydrolysis time varied from less than 1 h to about 100 h.
