ABSTRACT New technologies developed to control weeds in crops have flourished in recent years. Therefore, the objective of this work was to study the competitive ability of maize hybrids 2B433 (Enlist®), Pioneer 30F53 (VYHR®), and 13K288 PWE (Enlist®) and a conventional (non-transgenic) variety with a distinct biotechnological background when competing against wild poinsettia (Euphorbia heterophylla) and Alexandergrass (Urochloa plantaginea). Crops and competitors were studied in distinct plant proportions: 20:0, 15:5, 10:10, 5:15 and 0:20 plants per pot or 100:0, 75:25, 50:50, 25:75, and 0:100% (crop:weed) in replacement series experiments. Fifty days after emergence, the leaf area and aboveground dry mass were measured. Concomitantly, the photosynthetic and carboxylation efficiency and CO2 concentration were assessed in the leaf mesophyll. Maize hybrid 2B433 (Enlist®) showed better physiological and morphological performance compared to the conventional variety, and hybrids 13K288 PWE (Enlist®) and 30F53 (VYHR®) showed better performance when competing with wild poinsettia or Alexandergrass. The increased competitive ability seems to be due to the greater leaf area and aboveground dry mass reported for 2B433 (Enlist®). Therefore, the competitive ability presented by the hybrids was distinct, and we found no evidence that the transgenic event present in these hybrids affected their competitive ability. years Therefore B 2B43 Enlist®, Enlist Enlist® , F 30F5 VYHR®, VYHR VYHR® K 13K28 (Enlist® nontransgenic non (non-transgenic Euphorbia heterophylla Urochloa plantaginea. plantaginea . plantaginea) proportions 200, 200 20 0, 0 20:0 155, 155 15 5, 5 15:5 1010, 1010 10 10, 10:10 515 5:1 020 0:2 1000, 1000 100 100:0 7525, 7525 75 25, 25 75:25 5050, 5050 50 50, 50:50 2575, 2575 75, 25:75 0100% 0100 100% 0:100 cropweed crop weed (crop:weed experiments emergence measured Concomitantly CO mesophyll (VYHR® Enlist®. 2B4 30F 13K2 (Enlist 2 20: 1 15: 101 10:1 51 5: 02 0: 100: 752 7 75:2 505 50:5 257 25:7 010 0:10 (VYHR 2B 13K 10: 75: 50: 25: 01 0:1

Ezetimibe has significant lipid-lowering activity but presents low aqueous solubility. Co-crystallization techniques can be used to overcome this limitation. However, it is not yet clear whether the solubility of the cocrystal can be maintained after oral administration. In the present study, an ezetimi-imidazole cocrystal was synthetized by the reaction crystallization method, a promising strategy in cocrystal engineering. After conducting characterization tests, we determined the solubility and thermodynamic stability of the cocrystal. Also, the solubility advantage for both water and biorelevant fasted state simulated intestinal fluid (FaSSIF) was calculated. Dissolution was determined under supersaturated conditions, and the hypolipidemic activity of the cocrystal was assessed in vivo. The cocrystal exhibited high solubility and thermodynamic stability in FaSSIF. The solubility advantage was calculated as 22.4 in water and 11.8 in FaSSIF. The dissolution of the cocrystal resulted in supersaturation levels (σmax) of 1.3 in water and 3.3 in FaSSIF and the area under the curve (AUC) increased from 478.49 (ezetimibe) to 2963.62 µg min mL-1(cocrystal) in FaSSIF. The atherogenic index of the animals that received the cocrystal was similar to that of the control group. These results demonstrate that the inherent advantages of the co-crystallization process were maintained, enabling us to elucidate the behavior of the cocrystal. lipidlowering lipid lowering Cocrystallization Co limitation However administration study ezetimiimidazole ezetimi imidazole method engineering tests Also (FaSSIF conditions vivo 224 22 4 22. 118 11 8 11. σmax (σmax 13 1 3 1. 33 3. (LC (TAVR desenvolvimento tempo 200 2023 NSC. NSC . Abstract Background Robust data on the learning curve (LC) of transcatheter aortic valve replacement (TAVR) are lacking in developing countries. Objective To assess TAVR’s LC in Brazil over time. Methods We analyzed data from the Brazilian TAVR registry from 2008 to 2023. Patients from each center were numbered chronologically in case sequence numbers (CSNs). LC was performed using restricted cubic splines adjusted for EuroSCORE-II and the use of new-generation prostheses. Also, in-hospital outcomes were compared between groups defined according to the level of experience based on the CSN: 1st to 40th (initial-experience), 41st to 80th (early-experience), 81st to 120th (intermediate-experience), and over 121st (high-experience). Additional analysis was performed grouping hospitals according to the number of cases treated before 2014 (>40 and ≤40 procedures). The level of significance adopted was <0.05. Results A total of 3,194 patients from 25 centers were included. Mean age and EuroSCORE II were 80.7±8.1 years and 7±7.1, respectively. LC analysis demonstrated a drop in adjusted in-hospital mortality after treating 40 patients. A leveling off of the curve was observed after case #118. In-hospital mortality across the groups was 8.6%, 7.7%, 5.9%, and 3.7% for initial-, early-, intermediate-, and high-experience, respectively (p<0.001). High experience independently predicted lower mortality (OR 0.57, p=0.013 vs. initial experience). Low-volume centers before 2014 showed no significant decrease in the likelihood of death with gained experience, whereas high-volume centers had a continuous improvement after case #10. Conclusion A TAVR LC phenomenon was observed for in-hospital mortality in Brazil. This effect was more pronounced in centers that treated their first 40 cases before 2014 than those that reached this milestone after 2014. (LC (TAVR countries TAVRs s time 200 2023 CSNs. CSNs . ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others. publications problem uptodate up date classifications context exception (CTFB http//fauna.jbrj.gov.br/, httpfaunajbrjgovbr http //fauna.jbrj.gov.br/ , jbrj gov br (http://fauna.jbrj.gov.br/) 2015 Brazil 80 specialists 1 2024 133691 133 691 133,69 125138 125 138 125,13 82.3%, 823 82 3 (82.3% 102000 102 000 102,00 7.69%, 769 7 69 (7.69% 11000 11 11,00 . 3,567 3567 567 (3,56 2,292 2292 2 292 (2,29 1,833 1833 833 (1,83 1,447 1447 447 (1,44 1000 1,00 831 (83 628 (62 606 (60 520 (52 50 users science health biology law anthropology education others http//fauna.jbrj.gov.br/ faunajbrjgovbr //fauna.jbrj.gov.br (http://fauna.jbrj.gov.br/ 201 8 202 13369 13 133,6 12513 12 125,1 82.3% (82.3 10200 10 00 102,0 7.69% 76 6 (7.69 1100 11,0 3,56 356 56 (3,5 2,29 229 29 (2,2 1,83 183 83 (1,8 1,44 144 44 (1,4 100 1,0 (8 62 (6 60 52 (5 5 http//fauna.jbrj.gov.br (http://fauna.jbrj.gov.br 20 1336 133, 1251 125, 82.3 (82. 1020 0 102, 7.69 (7.6 110 11, 3,5 35 (3, 2,2 22 (2, 1,8 18 (1, 1,4 14 4 ( 82. Abstract Background: Transplant Renal Artery Stenosis (TRAS) is a recognized vascular complication after kidney transplantation. The overall risk predictors of TRAS are poorly understood. Methods: Retrospective analysis of patients with suspected TRAS (Doppler ultrasound PSV > 200 cm/s) who underwent angiographic study in a single center between 2007 and 2014. All patients with stenosis > 50% were considered with TRAS. Stenosis restricted in the body of the artery was also analyzed in a subgroup. Results: 274 patients were submitted to a renal angiography and 166 confirmed TRAS. TRAS group featured an older population (46.3 ± 11.0 vs. 40.9 ±14.2 years; p = 0.001), more frequent hypertensive nephropathy (30.1% vs. 15.7%; p = 0.01), higher incidence of Delayed Graft Function (DGF) (52.0% vs. 25.6%; p < 0.001) and longer Cold Ischemia Time (CIT) (21.5 ± 10.6 vs. 15.7 ± 12.9h; p < 0.001). In multivariable analyses, DGF (OR = 3.31; 95% CI 1.78-6.30; p < 0.0001) was independent risk factors for TRAS. DM and CIT showed a tendency towards TRAS. The compound discriminatory capacity of the multivariable model (AUC = 0.775; 95% CI 0.718-0.831) is significantly higher than systolic blood pressure and creatinine alone (AUC = 0.62; 95% CI 0.558-0.661). In body artery stenosis subgroup, DGF (OR = 1.86; 95% CI 1.04-3.36; p = 0.03) and Diabetes Mellitus (DM) (OR = 2.44; 95% CI 1.31-4.60; p = 0.005) were independent risk factors for TRAS. Conclusion: In our transplant population, DGF increased more than 3-fold the risk of TRAS. In the subgroup analysis, both DGF and DM increases the risk of body artery stenosis. The addition of other factors to hypertension and renal dysfunction may increase diagnostic accuracy.

Pharmaceutical cocrystals are composed of a drug and a coformer and are currently gaining attention due to their possibility of improving drug solubility. Quantitative determination of cocrystals is usually carried out by liquid chromatography method, but an alternative for the same is derivative UV spectrophotometry. Cocrystals of lamotrigine (LAM) were formed with nicotinamide (NIC) in order to improve the solubility of LAM. The aim of this study was to develop and validate a simple, low-cost, environment friendly, first-order derivative spectrophotometric method for simultaneous determination of LAM and NIC in cocrystals. Determination of LAM and NIC was performed at 244.4 and 271.6 nm, respectively. The method was accurate with recovery values of 98.21-101.52%, and precise (relative standard deviation (RSD) < 1.88%). Robustness evaluated by Plackett-Burman design showed no significant influence of the factors (pH, scanning speed, and sonication time) on LAM and NIC assays. The developed method was compared with a high performance liquid chromatography (HPLC) method and applied to study the cocrystal stoichiometry and solubility. The results indicated a molar ratio of 1:1 and that the cocrystal is more soluble than the drug. This study demonstrated that the first-order derivative method is feasible for drug and coformer determination in cocrystals and is a suitable alternative to chromatographic methods.

ABSTRACT Wasps are a diverse group of insects that possess a sting apparatus associated with a venom gland, which is used for predation and colony defense. The biochemistry of Hymenoptera venom has been evaluated in relation to allergy and immunology, and proteomics has been shown to be a powerful tool for the identification of compounds with pharmacological potential. Data on wasps venom the of genus Apoica are scarce, so the objective of the present work was to identify the venom proteins of the eusocial wasp Apoica pallens, as a first step towards further investigation of applied uses of the venom and its protein constituents. The venom proteins were separated by two-dimensional gel electrophoresis, followed by MALDI-TOF/TOF mass spectrometry. A total of 259 spots were detected, with molecular weights from 4.9 to 141 kDa. Thirty of these proteins were identified and classified into eight functional categories: allergen, enzyme, metabolism, structural, environmental response, proteoglycan, active in DNA and RNA, and unknown function. Due to the few available proteomic data for wasp venom, many proteins could not be identified, which makes studies with proteomic analysis of Hymenoptera venom even more important.

ABSTRACT: The aim of this study was to measure the occurrence of brucellosis, leptospirosis and neosporosis in cows from 25 family farms milk, with semi-intensive grazing system, located in the Southwest region of Paraná, Brazil. Eighty-four cows with retained placenta, between July 2013 to July 2014, diagnosed by the presence of fetal membranes in the uterus up to 12 hours after partum, decrease of appetite and of milk production, as well increase of rectal temperature were included into the study. The animals were submitted to blood collection for evaluating the seropositivity for brucellosis, leptospirosis and neosporosis. The blood was collected at 15 to 45 days postpartum, avoiding false negative results due to immune deficiency observed in the transition period. After collection, the blood was centrifuged, the serum was packed in three aliquots, identified and subsequently frozen. Serology was performed to diagnose brucellosis (technique of buffered acidified antigen), leptospirosis (through the microscopic agglutination test - MAT) and neosporosis (indirect immunofluorescence test). Animals were considered positive when presenting titer >1:100 (leptospirosis) and 1:200 (neosporosis). No animal with placenta retention presented Brucella abortus bacteria; 39% (33/84) were reactive to one or more Leptospira serovars, 15% (13/84) were positive to Neospora caninum, and 4% (4/84) had both diagnosis, being reactive for leptospirosis and neosporosis. In conclusion, data from this experiment inspires greater attention to leptospirosis and neosporosis in dairy cattle presenting placenta retention in southwest Paraná.

Abstract This study described a broad clinical characterization of classical homocystinuria (HCU) in Brazil. This was a cross-sectional, observational study including clinical and biochemical data from 72 patients (60 families) from Brazil (South, n = 13; Southeast, n = 37; Northeast, n = 8; North, n = 1; and Midwest, n = 1). Parental consanguinity was reported in 42% of families. Ocular manifestations were the earliest detected symptom (53% of cases), the main reason for diagnostic suspicion (63% of cases), and the most prevalent manifestation at diagnosis (67% of cases). Pyridoxine responsiveness was observed in 14% of patients. Only 22% of nonresponsive patients on treatment had total homocysteine levels <100 mmol/L. Most commonly used treatment strategies were pyridoxine (93% of patients), folic acid (90%), betaine (74%), vitamin B12 (27%), and low-methionine diet + metabolic formula (17%). Most patients diagnosed with HCU in Brazil are late diagnosed, express a severe phenotype, and poor metabolic control. Milder forms of HCU are likely underrepresented due to underdiagnosis.

Abstract Objective: This study describes a quantitative and qualitative methodology to assess hedonic responses to sweet stimulus in healthy newborns. Methods: A descriptive, cross-sectional, observational study, with healthy newborns (up to 24 h of life), between 37 and 42 gestational weeks, vaginally born and breastfed previously to all tests. The evaluation of the newborns reactions was performed by hedonic facial expression analysis, characterized by facial expressions with rhythmic serial tongue protrusion after neutral or sweet solution intake. Initially, 1 mL of water solution was provided to the newborn, followed by a 1-minute recording. Afterwards, the same amount of 25% sucrose solution was provided, performing a second recording. The concordance between researchers was analyzed by the Bland-Altman statistical method. Results: A total of 100 newborns (n = 49 males, n = 51 females; mean lifetime = 15 h 12 min ± 6 h 29 min) were recorded for neutral and sucrose solution intake, totaling 197 videos (n = 3 missing in the water treatment). These videos were double-blind analyzed and the test revealed a 90% concordance between the two trained researchers, in relation to both solutions. The intraclass correlation coefficient was 0.99 for both solutions, with a significant increase in frequency of hedonic expressions evoked by sucrose solution intake. Conclusions: These results confirm that the proposed method has an efficient power to detect significant differences between neutral and sucrose stimuli. In conclusion, this evaluation method of hedonic facial reactions in newborns reflects the response to a specific taste.

ABSTRACT Laetia suaveolens (Poepp.) Benth., Salicaceae, popularly known as "casinga-cheirosa", "caferana", or "laranjinha", is native to Brazil but not endemic to this country. A crude organic extract was obtained from the leaves and stem and intraperitoneally administered in male Balb-c mice. Its behavioral effects were evaluated in the open field and elevated plus maze in a two-stage experiment that assessed ten different parameters related to behavior as locomotion, emotionality, and anxiety. In the first stage of the experiment, intraperitoneal the crude organic extract administration dose-dependently impaired locomotion and emotionality 30–120 min after administration. A significant decrease in defecation was observed, which was related to emotionality. No alterations in the elevated plus maze were found; thus, this apparatus was not used in the next stage of the experiment. In the second stage, the previously determined non-lethal dose of 0.1563 g/kg was intraperitoneally administered, which impaired locomotion and rearing frequency and increased immobility time. Necropsy revealed smooth intestine hemorrhage. Rutin, leucoside, nicotiflorin, guaijaverin, and astragalin were isolated from the crude organic extract. This is the first time that these compounds have been identified in L. suaveolens. In conclusion, the crude organic extract impaired locomotion and emotionality and caused hemorrhage in male Balb-c mice, indicating that its consumption can be harmful to humans and animals. The present results provide a basis for further studies on the pharmacology, toxicology, and natural product chemistry of the crude organic extract.

ABSTRACT Drosophilidae is one of the most representative families of insects that occurs in fungal fruiting bodies of Basidiomycetes; however, the diversity and community structure of mycophagous Drosophilidae in the Neotropical region is poorly known. The aims of the present study were to describe the diversity of mycophagous Drosophilidae and to investigate its colonization of fungal hosts in a forest of southern Brazil. From 120 fungal samples (patches of mushrooms) of 17 Basidiomycetes genera, flies were recorded emerging from 70 samples and collected in adult stages of 25 fungal samples, for a total of 4897 drosophilids belonging to 31 species and 5 genera. Drosophila Fallén was the most species-rich genus, whereas Hirtodrosophila Duda was the dominant genus. Studies performed in the Holarctic region indicate that mycophagous drosophilid have generalist habits; however, our results showed that most drosophilids use fewer than two fungal hosts, and most species of Hirtodrosophila and Leucophenga were restricted to abundant fungal species, suggesting a specialization for these resources. The most specialized fauna emerged from Auricularia, which was the most frequent fungal genus in our collection, and this result supports the assumption that specialization depends on the availability of fungal resources over time.

Abstract The coccinellids Eriopis connexa (Germar), Harmonia axyridis (Pallas) and Olla v-nigrum (Mulsant) are important natural biological control agents. The purpose of this paper was to study the biology and create a fertility life table of these three coccinellid species. For the biology study, 50 insects/species were used and kept in groups of 10 in glass vials (2300cm3). For the three species studied, the viability of the total cycle varied from 45 to 50%. O. v-nigrum was the species which presented the longest oviposition period. However, H. axiridis demonstrated the best reproductive performance and ability of population growth in each generation. In conclusion, the use of commercially obtained pollen and A. kuenhiella eggs enables the development of coccinellids E. connexa, H. axyridis and O. v-nigrum under laboratory conditions, since the insects completed their biological cycle and originated adults with good reproductive performance.
