Resumo Nosso objetivo foi analisar a associação combinada entre características dos pais e indicadores de adiposidade dos filhos com o risco metabólico em adolescentes. Foi realizado estudo transversal com 972 adolescentes e seus pais. Observamos que adolescentes com sobrepeso que possuem mãe com peso normal apresentaram menor risco metabólico em comparação com seus pares com mães que apresentam sobrepeso. Concluímos que o status de peso da mãe modera a relação entre indicadores de obesidade e risco metabólico dos adolescentes.

Resumo Introdução: A queda da 25-hidroxivitamina D [25 (OH) D] na doença renal crônica (DRC) limita a capacidade renal de sintetizar a vitamina. A deficiência de vitamina D, (25(OH)D<20 ng/mL), é prevalente em pacientes com DRC e associada ao estresse oxidativo (EO). Avaliamos possível associação entre a deficiência de vitamina D e EO em pacientes pré-dialíticos. Métodos: estudo transversal com 206 pacientes com DRC. Exames para 25(OH)D, 1,25(OH)2D, marcadores inflamatórios e EO foram adicionados àqueles de rotina, incluindo creatinina, albumina, cálcio, fósforo, fosfatase alcalina, iPTH, glicose, hemoglobina, ácido úrico, colesterol total , LDL, HDL e triglicerídeos. Resultados: 55 pacientes com DRC tinham deficiência de vitamina D e os 149 tinham níveis normais da vitamina. Houve uma associação significativa entre a vitamina D e a taxa estimada de filtração glomerular (TFGe). Os níveis de homocisteína foram melhor previstos pela TFGe, gênero e idade; proteína C reativa de alta sensibilidade (hsCRP) por estadiamento e IMC; os metabólitos de óxido nítrico (NOx) aumentaram na doença tardia; a leptina foi influenciada pelo IMC, e mais alta em mulheres, assim como os níveis de adiponectina. Conclusões: biomarcadores do EO não correlacionaram com a deficiência de vitamina D, mas houve aumento de NOx nos estágios 4-5 da DRC. Apesar dos grandes números de pacientes com DRC, de biomarcadores inflamatórios e EO usados neste estudo, não houve associação entre os níveis de vitamina D e a TFGe. Mais estudos são necessários para avaliar a influência do status da vitamina D no EO em pacientes com DRC em pré-diálise.

Abstract Introduction: The progressive decline in 25-hydroxyvitamin D [25(OH)D] in chronic kidney disease (CKD) limits the kidney ability of synthesizing the vitamin. Vitamin D deficiency as defined by KDIGO (25(OH)D <20 ng/mL) is prevalent in CKD patients and associated to oxidative stress (OS). We studied a possible association between vitamin D deficiency and OS in pre-dialysis patients. Methods: A cross-sectional study with 206 CKD patients was carried out. Laboratory tests for 25(OH)D, 1,25(OH)2D, inflammatory markers, and OS were added to routine tests including creatinine, albumin, calcium, phosphorus, alkaline phosphatase, iPTH, glucose, hemoglobin, uric acid, total cholesterol, LDL, HDL, and triglycerides. Results: Vitamin D deficiency was present in 55 CKD patients and normal vitamin D levels were seen in 149 patients. There was a significant association between vitamin D and estimated glomerular filtration rate (eGRF). Homocysteine levels were best predicted by eGRF, sex, and age; high sensitivity C-reactive protein (hsCRP) by staging and BMI; nitric oxide metabolites (NOx) were increased in late disease; leptin was influenced by BMI and higher in women than man; and adiponectin levels were higher in women. Conclusions: OS biomarkers were not correlated with vitamin D deficiency but increased NOx were seen in stages 4-5 CKD patients. Even though a relatively large number of CKD patients was included and a broad number of OS and inflammatory biomarkers were used in this studied we failed to find an association between vitamin D levels and eGRF. More studies are needed to evaluate the influence of vitamin D status in OS in pre-dialysis CKD patients.

Resumo Introdução: A anemia na DRC pode ser dividida em anemia sem deficiência funcional de ferro e com deficiência funcional de ferro (ADFF). Diante do aumento dos casos de hemossiderose em pacientes em hemodiálise, atribuídos à reposição excessiva de ferro endovenoso, maiores conhecimentos sobre os fatores envolvidos na gênese da ADFF são importantes. Objetivos: documentar a prevalência de ADFF em renais crônicos em hemodiálise. Caracterizar clínica e laboratorialmente os portadores de ADFF em HD e avaliar o estado nutricional, estresse oxidativo e inflamatório. Estudo transversal, amostra de conveniência, envolvendo 183 renais crônicos em hemodiálise no sul do Brasil. Após aplicação dos critérios de exclusão, os pacientes foram separados em dois grupos: portadores de anemia com e sem deficiência funcional de ferro. Foram submetidos a questionário socioepidemiológico, à análise antropométrica e análise laboratorial dos marcadores de anemia, estresse oxidativo, inflamatórios e nutricionais. Análise estatística: programa GraphPad InStat versão 3.1. Foram aplicados os testes: Kolmogorov-Smirnov, qui-quadrado, t de Student e Mann-Whitney. Nível de significância adotado de 5%. Resultados: não houve diferença significativa nos marcadores inflamatórios entre os dois grupos. Houve diferença significativa nos marcadores de anemia e nutrição, significativamente menores nos pacientes com ADFF. Pacientes com ADFF receberam doses mais elevadas de ferro parenteral (p < 0,05). Discussão: ADFF esteve associada a menores valores de marcadores nutricionais, mas não esteve associada a marcadores inflamatórios ou de estresse oxidativo aumentados, como relatado na literatura. Estudos adicionais sobre o tema são necessários.

Abstract Introduction: Anemic patients with chronic kidney disease (CKD) can be divided into anemic patients without or with functional iron deficiency (FID). The increase in the number of cases of hemosiderosis in patients on hemodialysis (HD) attributed to excessive intravenous iron replacement has called for the investigation of the factors involved in the genesis of FID. Objectives: This study aimed to describe the prevalence of FID in patients with CKD on HD, characterize the included individuals in terms of clinical and workup parameters, and assess their nutritional, oxidative stress, and inflammation statuses. This cross-sectional study assembled a convenience sample of 183 patients with CKD on HD treated in Southern Brazil. Patients meeting the inclusion and exclusion criteria were divided into two groups, one with anemic subjects with FID and one with anemic patients without FID. Participants answered a questionnaire probing into socio-epidemiological factors, underwent anthropometric measurements, and were tested for markers of anemia, oxidative stress, inflammation, and nutrition. Statistical analysis: The date sets were treated on software package GraphPad InStat version 3.1. Variables were tested with the Kolmogorov-Smirnov, chi-square, Student’s t, and Mann-Whitney tests. Statistical significance was attributed to differences with a p < 0.05. Results: Markers of inflammation were not statistically different between the two groups. Markers of anemia and nutrition were significantly lower in patients with FID. Patients with FID were prescribed higher doses of parenteral iron (p < 0,05). Discussion: FID was associated with lower nutritional marker levels, but not to increased levels of markers of inflammation or oxidative stress, as reported in the literature. Additional studies on the subject are needed.

RESUMO Objetivo: Analisar o efeito de oito semanas de suplementação de ácido linoleico conjugado sobre o desempenho físico, a gordura de tronco e abdominal em mulheres com excesso de peso submetidas a um programa de treinamento aeróbio. Métodos: Vinte e oito mulheres com excesso de peso (índice de massa corporal ³ 25 kg/m2) foram separadas aleatoriamente por meio de um delineamento duplo cego para receber suplementação de ácido linoleico ou placebo associado a um programa de exercícios aeróbios (frequência = três sessões semanais, duração=30 min/sessão, intensidade=80% da frequência cardíaca máxima). A suplementação de ácido linoleico (3,2 g) ou de placebo (4,0 g) foi consumida diariamente (quatro cápsulas), durante oito semanas. As variáveis velocidade máxima atingida e tempo de permanência até a exaustão foram determinadas em teste incremental em esteira. A gordura de tronco foi estimada por absortometria radiológica de dupla energia. A circunferência de cintura foi utilizada como indicador de gordura abdominal. Resultados: Efeito principal do tempo (p<0,05) revelou aumento da velocidade máxima atingida (suplementação de ácido linoleico=+6,3% versus placebo=+7,5%) e tempo de duração até a exaustão (suplementação de ácido linoleico=+7,1% versus placebo=+8,6%) em teste incremental em esteira, sem diferenças entre os grupos (p>0,05). De forma similar, uma redução significante (p<0,05) na gordura relativa de tronco (suplementação de ácido linoleico=-1,7% versus placebo=-1,5%) e na gordura abdominal (suplementação de ácido linoleico=-4,7% versus placebo=-4,0%) foi encontrada após oito semanas de intervenção, sem diferenças entre os grupos (p>0,05). Conclusão: Os resultados do presente estudo sugerem que a suplementação de ácido linoleico não maximiza o desempenho motor e a redução da gordura de tronco e abdominal induzida pelo treinamento aeróbio em mulheres com excesso de peso.

ABSTRACT Objective: To analyze the effect of eight weeks of conjugated linoleic acid supplementation on physical performance, and trunk and abdominal fat in overweight women submitted to an aerobic training program. Methods: Twenty-eight overweight women (body mass index ³25 kg/m2) were divided randomly and double-blindly to receive conjugated linoleic acid or placebo, both associated with an aerobic exercise program (frequency = three times a week, duration=30 min/session, intensity=80% of maximum heart rate). Conjugated linoleic acid (3.2 g) and placebo (4.0 g) supplements were consumed daily (four capsules) for eight weeks. Maximum speed and time to exhaustion were determined in incremental treadmill test. Trunk fat was estimated by dual-energy X-Ray absorptiometry. Waist circumference was used as indicator of abdominal fat. Results: Main effect of time (p<0.05) showed increased maximum speed (conjugated linoleic acid=+6.3% vs. placebo=+7.5%) and time to exhaustion (conjugated linoleic acid=+7.1% vs. placebo=+8.6%) in the incremental treadmill test, with no differences between the groups (p>0.05). Similarly, significant reductions (p<0.05) in trunk fat (conjugated linoleic acid=-1.7% vs. placebo=-1.5%) and abdominal fat (conjugated linoleic acid=-4.7% vs. placebo=-4.0%) were found after eight weeks of intervention, with no differences between the groups (p>0.05). Conclusion: The results of this study suggest that conjugated linoleic acid supplementation does not maximize motor performance, and loss of body and abdominal fat induced by aerobic training in overweight women.

ABSTRACT Objective Paraoxonase 1 (PON1) polymorphisms are associated with an increased susceptibility to cardiovascular disease. PON1 Q192R polymorphism (rs662) partially determine PON1 hydrolytic activity and protect against oxidation of LDL and HDL. This study aimed to delineate the association of PON1 status (functional 192 genotype and plasma activity levels) and atherogenicity in urbans residents aged 40 years or more. Materials and methods Anthropometric data, lipid profiles, the atherogenic index of the plasma (AIP) and Framingham score risk were measured. Three kinetic assays were conducted to assay PON1 status using phenylacetate and 4-(chloromethyl)phenyl acetate as substrates. Results Smoking per se did not significantly impact the AIP but the interaction PON1 genotype by smoking significantly increased the AIP. In subjects with the RR genotype smoking increased the AIP index from (estimated mean ± SEM) -0.038 ± 0.039 to 0.224 ± 0.094. The QR genotype increased the Framingham risk index by around 1.3 points. Smoking by RR genotype carriers significantly increased the Framingham risk score (17.23 ± 2.04) as compared to smoking (13.00 ± 1.06) and non-smoking (7.79 ± 0.70) by QQ+QR genotype carriers. The interaction RR genotype by smoking was a more important predictor (odds ratio = 7.90) of an increased Framingham risk score (> 20) than smoking per se (odds ratio = 2.73). The interaction smoking by RR genotype carriers significantly increased triglycerides and lowered HDL cholesterol. Conclusion Smoking per se has no (AIP) or a mild (Framingham risk score) effect on atherogenicity, while the interaction smoking by PON1 RR genotype has a clinically highly significant impact on atherogenicity.

Resumo Introdução: Pacientes com doença renal em estágio terminal têm níveis de inflamação e estresse oxidativo maiores do que a população geral. Muitos fatores contribuem para isso, e o hormônio paratireoidiano (PTH) é um deles. Objetivo: Estudo foi realizado para avaliar a relação entre os níveis de PTH e níveis de inflamação e estresse oxidativo em pacientes em hemodiálise. Métodos: estudo transversal com pacientes de duas unidades de hemodiálise de Londrina, Brasil. Pacientes com condições causadoras de inflamação e estresse oxidativo foram exclusos. Níveis plasmáticos de PTH e parâmetros bioquímicos de inflamação (interleucina 1 e 6, fator de necrose tumoral alfa) e estresse oxidativo (capacidade antioxidante plasmática total, dialdeído malônico, hidroperoxidação lipídica, produtos avançados da degradação proteica, quantificação de metabólitos de óxido nítrico e 8-isoprostano) foram dosados antes da sessão de hemodiálise. Realizou-se análise de correlação entre os níveis de PTH - tanto como variável continua como variável categórica em tercis - e os parâmetros de inflamação e estresse oxidativo. Resultados: Não houve correlação do PTH com nenhum dos parâmetros testados, nem como variável contínua, nem como categórica. Conclusão: Neste estudo descritivo, os resultados sugerem que a inflamação e o estresse oxidativo em pacientes em hemodiálise provavelmente tem origem em mecanismos que não incluem o hiperparatireoidismo secundário.

Abstract Introduction: Patients at end stage renal disease have higher levels of inflammation and oxidative stress than the general population. Many factors contribute to these issues, and the parathyroid hormone (PTH) is also implicated. Objective: The study was conducted in order to assess the relationship between PTH levels and inflammation and oxidative stress in hemodialysis patients. Methods: Cross-sectional study with patients of two hemodialysis facilities in Londrina, Brazil. Patients with other conditions known to generate oxidative stress and inflammation were excluded. Blood levels of PTH and biochemical parameters of inflammation (interleukins 1 and 6, tumor necrosis factor-alpha) and oxidative stress (total plasma antioxidant capacity, malonic dialdehyde, lipid hydroperoxidation, advanced oxidation protein products, quantification of nitric oxide metabolites, and 8-isoprostane) were measured before a dialysis session. Then, we made correlation analyses between PTH levels - either as the continuous variable or categorized into tertiles-, and inflammatory and oxidative stress biomarkers. Results: PTH did not show any correlation with the tested inflammation and oxidative stress parameters, nor as continuous variable neither as categorical variable. Conclusion: In this descriptive study, the results suggest that the inflammation and oxidative stress of hemodialysis patients probably arise from mechanisms other than secondary hyperparathyroidism.

Resumo O objetivo deste estudo foi investigar a efetividade do treinamento com pesos (TP), seguido por um período semelhante de destreinamento, sobre a modulação do estresse oxidativo (EO) em mulheres idosas obesas. Quatorze mulheres (idade: 68,7 ± 4,8 anos; massa corporal: 71,3 ± 14,8 kg; estatura: 156,3 ± 7,2 cm; gordura corporal: 44,3 ± 4,4%) obesas (gordura corporal relativa ≥ 30%) foram submetidas a 12 semanas de um programa de TP seguido por igual período de destreinamento. Os produtos de proteínas de oxidação avançada (AOPP) e a capacidade antioxidante total plasmática (TRAP) foram utilizados como indicadores de estresse oxidativo. A AOPP não foi alterada pelo TP ou pelo destreinamento (P = 0,31). Por outro lado, a TRAP foi aumentada com o TP (+15,1%; P < 0,001) e se manteve elevada mesmo após 12 semanas de destreinamento (+10,5%; P < 0,001). Os resultados sugerem que o EO pode ser melhorado pelo TP e o período de 12 semanas de destreinamento não parece ser suficiente para desfazer as adaptações induzidas pelo TP em mulheres idosas obesas.

Abstract The aim of this study was to investigate the effect of resistance training (RT) followed by a similar detraining period on the modulation of oxidative stress (OS) in obese older women. Fourteen obese women (age: 68.7 ± 4.8 years, body mass: 71.3 ± 14.8 kg, height: 156.3 ± 7.2 cm, body fat: 44.3 ± 4.4%) were submitted to 12 weeks of a RT program followed by a similar detraining period. Advanced oxidation protein products (AOPP) and total radical-trapping antioxidant potential (TRAP) were used as oxidative stress indicators. AOPP was not changed by RT or detraining (P = 0.31). Furthermore, TRAP was increased with RT (+ 15.1%; P < 0.001) and remained high even after 12 weeks of detraining (10.5%; P < 0.001). The results suggest that OS can be improved by RT and the 12-week detraining period does not seem to be enough to reverse adaptations induced by RT in obese older women.

A doença de Parkinson (DP) é caracterizada pela degeneração progressiva dos neurônios dopaminérgicos na substância negra pars compacta. Além disso, o estresse oxidativo, presente nesta doença, causa ou contribui para o processo neurodegenerativo. Atualmente, a DP tem apenas tratamento sintomático e ainda nada pode ser feito para interromper o processo degenerativo. Este estudo teve como objetivo avaliar, comparativamente, a capacidade antioxidante do pramipexol, selegilina e amantadina em diferentes testes in vitro e oferecer possíveis explicações sobre os mecanismos moleculares antioxidantes destes fármacos. Avaliou-se a atividade antioxidante dos fármacos através da capacidade em diminuir ou sequestrar espécies reativas de oxigênio no burst respiratório, da capacidade em doar hidrogênio e estabilizar o radical livre 2,2-difenil-1-picril-hidrazil (DPPH•), de remover o radical 2,2'-azino-di-(3-etilbenzotiazolina-6-sulfônico (ABTS+) e da verificação do poder redutor/antioxidante do ferro (FRAP). Este estudo demonstrou que tanto o pramipexol como a selegilina, mas não a amantadina, possuem efeitos antioxidantes in vitro por eliminar o ânion superóxido no burst respiratório, doar elétrons no método ABTS e apresentar poder redutor sobre o ferro (FRAP). Essa capacidade antioxidante pode estar relacionada com a estrutura química desses medicamentos, sugerindo possíveis mecanismos neuroprotetores destes fármacos além de seus mecanismos de ação já conhecidos.

Parkinson's disease (PD) is characterized by progressive degeneration of dopaminergic neurons in the substantia nigra pars compacta. Furthermore, oxidative stress plays a role in PD, causing or contributing to the neurodegenerative process. Currently PD has only symptomatic treatment and still nothing can be done to stop the degenerative process of the disease. This study aimed to comparatively evaluate the antioxidant capacity of pramipexole, selegeline and amantadine in different in vitrostudies and to offer possible explanations on the molecular antioxidant mechanisms of these drugs. In vitro, the antioxidant capacity of the drugs was assessed by the ability of antiparkinsonian drugs to decrease or scavenge ROS in the neutrophil respiratory burst, ability of antiparkinsonian drugs to donate hydrogen and stabilize the free radical 2,2-diphenyl-1-picryl-hydrazyl (DPPH•), to scavenge 2,2'-azino-di-(3-ethylbenzthiazoline-6-sulphonic acid (ABTS+) and evaluation of the ferric reducing antioxidant power (FRAP). This study demonstrated that both pramipexole and selegiline, but not amantadine, have antioxidant effects in vitro by scavenging superoxide anion on the respiratory burst, donating electron in the ABTS+ assay and presenting ferric reduction antioxidant power. This chemical structure-related antioxidant capacity suggests a possible neuroprotective mechanism of these drugs beyond their already recognized mechanism of action.

Contexto Os anti-inflamatórios não-esteróides são considerados uma das mais importantes causas de reativação da doença inflamatória intestinal. Em relação aos inibidores seletivos da ciclo-oxigenase 2, os resultados são controversos tanto em estudos envolvendo humanos como na colite experimental. Objetivos Comparar os efeitos dos anti-inflamatórios não-esteróides, seletivos e não seletivos da ciclo-oxigenase 2, na colite experimental e, contribuir para o entendimento do mecanismo no qual os anti-inflamatórios não-esteróides provocam a exacerbação da colite. Métodos Seis grupos de ratos foram estudados: sem colite, com colite e com colite e tratados com celecoxib, cetoprofeno, indometacina ou diclofenaco. Foram determinadas a taxa de sobrevida, as concentrações de hemoglobina e albumina plasmática, as concentrações teciduais na mucosa colônica de interleucina-1ß, interleucina-6, fator de necrose tumoral alfa, prostaglandina E2, catalase, superóxido dismutase, substâncias reativas ao ácido tiobarbitúrico e quimiluminescência estimulada por hidroperóxido de tert-butil, e as concentraçãos plasmática e tecidual de leucotrieno B4. Resultados O grupo tratado com diclofenaco ou indometacina apresentaram as menores taxas de sobrevida, concentrações de hemoglobina e albumina, e as maiores concentrações plasmática e tecidual de leucotrieno B4 e tecidual de superóxido dismutase do que os groupos tratados com celecoxib. O grupo tratado com cetoprofeno apresentou um comportamento intermediário entre diclofenaco/indometacina e celecoxib, em relação a taxa de sobrevida e albumina. Os grupos sem colite, colite e colite tratado com celecoxib apresentaram menores concentrações de leucotrieno B4 e superóxido dismutase do que os grupos tratados com inibidores não seletivos ciclo-oxigenase 2. Conclusões Diclofenaco e indometacina apresentaram os maiores graus de exacerbação da colite entre os anti-inflamatórios não-esteróides, o celecoxib não apresentou exacerbação e o cetoprofeno apresenteou um comportamento intermediario entre diclofenaco/indometacina e celecoxib. Estes resultados sugerem que o leucotrieno B4 e superóxido dismutase estão envolvidos na exacerbação da colite experimental pelo anti-inflamatórios não-esteróides não seletivos ciclo-oxigenase 2.

Context Nonsteroidal anti-inflammatory drugs are considered one of the most important causes of reactivation of inflammatory bowel disease. With regard to selective cyclo-oxygenase 2 inhibitors, the results are controversial in experimental colitis as well as in human studies. Objectives The aim this study is to compare nonsteroidal anti-inflammatory drugs effects, selective and non selective cyclo-oxygenase 2 inhibitors, in experimental colitis and contribute to the understanding of the mechanisms which nonsteroidal anti-inflammatory drugs provoke colitis exacerbation. Methods Six groups of rats: without colitis, with colitis, and colitis treated with celecoxib, ketoprofen, indometacin or diclofenac. Survival rates, hemoglobin, plasmatic albumin, colonic tissue of interleukin-1ß, interleukin-6, tumor necrosis factor alpha, prostaglandin E2, catalase, superoxide dismutase, thiobarbituric acid-reactive substances, chemiluminescence induced by tert-butil hydroperoxides, and tissue and plasmatic leukotriene B4 were determined. Results The groups treated with diclofenac or indometacin presented lower survival rates, hemoglobin and albumin, higher tissue and plasmatic leukotriene B4 and tissue superoxide dismutase than the group treated with celecoxib. Ketoprofen presented an intermediary behavior between diclofenac/indometacin and celecoxib, concerning to survival rate and albumin. The groups without colitis, with colitis and with colitis treated with celecoxib showed leukotriene B4 and superoxide dismutase lower levels than the groups treated with nonselective cyclo-oxygenase 2 inhibitors. Conclusions Diclofenac and indometacin presented the highest degree of induced colitis exacerbation with nonsteroidal anti-inflammatory drugs, celecoxib did not show colitis exacerbation, and ketoprofen presented an intermediary behavior between diclofenac/indometacin and celecoxib. These results suggest that leukotriene B4 and superoxide dismutase can be involved in the exacerbation of experimental colitis by nonselective nonsteroidal anti-inflammatory drugs.

Passiflora incarnata é comercializada em muitos países como fitoterápico. Embora a bula da maioria dos fitoterápicos recomende que eles sejam usados sob supervisão médica, estudos sobre a toxicidade reprodutiva e do desenvolvimento desses produtos são raros e não obrigatórios para fins regulatórios. Neste estudo, realizamos uma avaliação da toxicidade reprodutiva da P. incarnata, administrada a ratas Wistar (30 ou 300 mg/kg, gavagem) durante a gestação e a lactação. Além disso, considerando as propriedades antioxidantes que têm sido atribuídas aos flavonoides presentes no gênero Passiflora, também avaliou-se o equilíbrio antioxidante/pró-oxidante no plasma destas fêmeas e conduziu-se um teste in vitro para avaliar o potencial antioxidante. O tratamento com P. incarnata não influenciou o peso corporal das fêmeas, bem como indicadores de toxicidade reprodutiva (perdas pós-implantação, número de filhotes vivos e peso da ninhada) e os parâmetros de função hepática (albumina, AST, ALT, GGT). A propriedade antioxidante da P. incarnata foi evidenciada tanto in vivo (aumento do potencial antioxidante total plasmático) quanto in vitro (diminuição do burst respiratório em neutrófilos). Os resultados deste estudo indicam que, nas condições experimentais avaliadas, o tratamento com P. incarnata durante a gestação e lactação apresentou efeito antioxidante, na ausência de toxicidade reprodutiva materna.

Passiflora incarnata is marketed in many countries as a phytomedicine. Even though the directions of most marketed phytomedicines recommend them to be used under medical supervision, reproductive and developmental studies are sparse and not mandatory for regulatory purposes. In this study, a reproductive toxicity evaluation of P. incarnata was conducted in Wistar rats gavaged (30 or 300 mg/kg) during pregnancy and lactation. Moreover, considering that antioxidant properties have been attributed to flavonoids present in the genus Passiflora, it was also evaluated the antioxidant/pro-oxidant balance in the plasma of these dams and the antioxidant potential in an in vitro test. P. incarnata treatment did not influence dams´ body weight as well as reproductive (gestation length, post-implantation loss, litter size, litter weight) and hepatic (albumin, AST, ALT, GGT) parameters. The antioxidant property of P. incarnata was evidenced both in vivo (increase in the total antioxidant plasmatic potential) and in vitro (decrease in neutrophil-induced respiratory burst). The results from the present study indicate that under the experimental conditions evaluated, P. incarnata treatment during gestation and lactation presented antioxidant activity in the absence of maternal reproductive toxicity.

OBJETIVO: Avaliar os indicadores de estresse oxidativo e marcadores inflamatórios em obesos antes e depois da gastroplastia vertical com bandagem em Y-de-Roux. MÉTODOS: Vinte obesos classe III foram submetidos à gastroplastia vertical com bandagem em Y-de-Roux, sendo sete homens e 13 mulheres, com idade média de 39 anos e 20 indivíduos não obesos, nove homens e 11 mulheres, média de idade de 38 anos. Foram determinados os níveis de malondialdeído no plasma, índice de estresse, capacidade antioxidante total, catalase e glutationa reduzida e oxidada, e marcadores inflamatórios (proteína C reativa e á-1 glicoproteína ácida). No grupo obeso, estes parâmetros foram determinados antes e 2, 6 e 12 meses após a gastroplastia vertical com bandagem em Y-de-Roux. RESULTADOS: No pré-operatório, o grupo obeso apresentou níveis elevados de marcadores inflamatórios, de estresse oxidativo (níveis de malondialdeído e índice de estresse) e menores níveis de indicadores de defesa antioxidante em relação ao grupo controle. O emagrecimento foi acompanhado de redução progressiva do níveis de malondialdeído e do índice de estresse. Foi observado aumento da concentração de glutationa reduzida e da capacidade antioxidante total e redução dos níveis de marcadores inflamatórios. CONCLUSÃO: A redução do peso melhora o estado inflamatório e os níveis de estresse oxidativo.

OBJECTIVE: To evaluate the oxidative stress and inflammatory markers in obese patients before and after Roux-en-Y vertical banded gastroplasty. METHODS: We studied 20 class III obese patients undergoing Roux-en-Y vertical banded gastroplasty, seven men and 13 women, mean age 39 years, and 20 non-obese subjects, nine males and 11 females, mean age 38 years. We determined the plasma levels of malondialdehyde, stress an index, total antioxidant capacity, catalase, reduced and oxidized glutathione and inflammatory markers (C reactive protein and á-1 acid glycoprotein). In the obese group, these parameters were determined before and 2, 6 and 12 months after gastroplasty. RESULTS: Preoperatively, the obese group showed elevated levels of inflammatory markers of oxidative stress (malondialdehyde levels and stress index) and lower levels of indicators of antioxidant defense compared to the control group. Weight loss was accompanied by gradual reduction in the levels of malondialdehyde and stress index. We found an increased concentration of reduced glutathione and total antioxidant status and reduced levels of inflammatory markers. CONCLUSION: Weight loss improves the inflammatory state and oxidative stress levels.

O objetivo do estudo foi analisar o impacto da prática habitual de atividade física, mediante estimativas do dispêndio energético diário (DEdiário), no perfil lipídico-lipoprotéico plasmático em amostra representativa de adolescentes. A amostra foi constituída por 452 sujeitos (246 moças e 206 rapazes) com idade entre 15 e 18 anos. As concentrações de lipídeos-lipoproteínas plasmáticas foram estabelecidas por procedimentos laboratoriais. As informações acerca da prática habitual de atividade física foram obtidas por intermédio de instrumento retrospectivo de auto-recordação. Estabeleceram-se estimativas quanto ao DE Diário com base no custo calórico associado ao tipo e à duração das atividades registradas pelos adolescentes. A amostra foi estratificada em três grupos de prática habitual de atividade física: os menos ativos, < 25º percentil do DE Diário; os moderadamente ativos, 25-75º percentil do DEdiário; e os mais ativos, > 75º percentil do DEdiário, e também pontos de corte para um perfil lipídico-lipoprotéico de risco aterogênico. Os resultados da análise de co-variância, controlando a participação do índice de massa corporal, mostraram que as concentrações de HDL-C foram significativamente diferentes de acordo com os grupos crescentes de DEdiário em moças (p = 0,009) e em rapazes (p = 0,010), porém, o comportamento de variação dos valores observados foi diferente nos dois sexos. A análise de regressão logística indicou que o risco relativo para menores valores de HDL-C foi duas vezes maior entre os adolescentes menos ativos em comparação com os mais ativos. Em ambos os sexos, as concentrações de colesterol total, LDL-C e triglicerídeos não apresentaram diferenças significativas entre os grupos de DEdiário. Em conclusão, a prática habitual de atividade física mais intensa e o maior DEdiário estão associados com mais elevadas concentrações de HDL-C em adolescentes de ambos os sexos, independentemente das variações do peso corporal. As associações entre DEdiário e os outros componentes plasmáticos de lipídeos-lipoproteínas tratados no presente estudo foram fracas e inconsistentes.

The aim of the present study was to analyze the impact of regular practice of physical activity facing estimates of daily energy expenditure (EEdaily) in the plasmatic lipid-lipoprotein profile in a representative sample of adolescents. The sample consisted of 452 subjects (246 females and 206 males), age between 15 and 18 years. The plasmatic lipid-lipoprotein concentrations were established through laboratory procedures. The data concerning the regular practice of physical activity were obtained though a retrospective instrument of self-recording. Estimates concerning the EEdaily were established based on the caloric cost associated with the type and the duration of the activities registered by the adolescents. The sample was divided in three groups of regular practice of physical activity: the least active, < 25º percentile of the EEdaily; the moderately active, 25-75º percentile of the EEdaily; and the most active, > 75º percentile of the EEdaily. Cutting points for an atherogenic lipid-lipoprotein risk were also established. The results of the analysis of covariance, having the participation of the body mass index controlled, showed that the HDL-C concentrations were significantly different according to the growing groups of EEdaily in females (p = 0.009) and in males (p = 0.010). However, the variation behavior of the values observed was different in the two sexes. The analysis of logistic regression showed that the relative risk for lower values of HDL-C was two times higher among the least active adolescents comparing with the most active ones. In both sexes the concentrations of total cholesterol, LDL-C and triglycerides did not present significant differences among the groups of EEdaily. In conclusion, more intense regular practice of physical activity as well as higher EEdaily are associated with higher concentrations of HDL-C in adolescents of both sexes, regardless of the variations of body weight. The associations between EEdaily and the other plasmatic components of lipid-lipoproteins treated in the present study were weak and inconsistent.

OBJETIVO: Analisar o impacto quanto ao uso de tabaco no perfil lipídico-lipoprotéico plasmático em amostra representativa de adolescentes. MÉTODOS: A amostra foi constituída por 452 sujeitos (246 moças e 206 rapazes) com idades entre 15 e 18 anos. Os participantes completaram questionário estruturado auto-administrado com relação ao uso de tabaco. As concentrações de lipídeos-lipoproteínas plasmáticas foram estabelecidas mediante procedimentos laboratoriais. Os procedimentos da análise de covariância, controlando a participação da ingestão de gordura saturada e de colesterol dietético, foram empregados para identificar as diferenças entre os valores médios. As estimativas de odds ratio foram utilizadas para estabelecer o risco relativo dos adolescentes fumantes apresentarem perfil lipídico-lipoprotéico de risco aterogênico. RESULTADOS: A proporção de fumantes foi de 20,9% entre os rapazes e 15,4% entre as moças. O consumo médio de cigarros por dia foi de 9,2 ± 4,7 nos rapazes e 5,6 ± 3,1 nas moças. Quando comparados com não fumantes, rapazes e moças fumantes apresentaram níveis séricos de colesterol total, LDL-colesterol, triglicerídeos e apolipoproteína B100 significativamente mais elevados, e níveis séricos de HDL-colesterol significativamente menores. Adolescentes fumantes tenderam a demonstrar risco de níveis de lipídeos-lipoproteinas plasmáticas alterados duas vezes maior que não fumantes. CONCLUSÃO: Intervenções direcionadas à adoção de um estilo de vida saudável, incluindo abstenção do uso de tabaco, deverão iniciar-se em idades precoces na tentativa de prevenir ou retardar o desenvolvimento de lesões ateroscleróticas e minimizar o aparecimento de coronariopatias prematuras na idade adulta.

OBJECTIVE: To analyze the impact of tobacco use on plasma lipid lipoprotein profile in representative sample of adolescents. METHODS: A sample of 452 subjects (246 girls and 206 boys) 15 to 18 years old were included in the study. Each participant completed a structured and self-administered questionnaire concerning tobacco use. Plasma lipid-lipoprotein concentrations were measured by standard procedures. Differences between mean values were evaluated by analysis of covariance, controlling for saturated fat and cholesterol intake. Odds ratio was used to estimate the relative risk of the smokers being classified with an undesirable level of a plasma lipidlipoprotein parameter. RESULTS: The proportion of smokers was 20.9% for boys and 15.4% for girls. The average consumption of cigarettes per day was 9.2 ± 4.7 for boys and 5.6 ± 3.1 for girls. When compared with non-smokers, boy and girl smokers showed a significantly higher serum levels of total cholesterol, LDL-cholesterol, triglycerides and apolipoprotein B100, and significantly lower serum levels of HDL-cholesterol. Adolescent smokers tended to show a two-fold higher risk of altered lipid-lipoprotein levels than non-smokers. CONCLUSION: The present data could imply that intervention promoting a healthy lifestyle, including non smoking, should start at an early age to prevent or delay development of atherosclerotic lesions and ultimately to minimize the appearance of premature coronary heart disease in adults.

O objetivo do presente estudo foi investigar associações entre a distribuição centrípeta da gordura corporal e lipídios-lipoproteínas séricos, pressão arterial e índice Homa-IR de resistência insulínica, mediante ajuste estatístico de indicadores quanto ao sobrepeso e à aptidão cardiorrespiratória. Foram analisados 89 voluntários (44 homens e 45 mulheres). A distribuição centrípeta da gordura foi analisada através de circunferência da cintura (CC) e o sobrepeso pelo índice de massa corporal (IMC). A aptidão cardiorrespiratória foi acompanhada pelo VO2máx estimado por teste de caminhada. Após ajuste pelos valores de IMC verificamos coeficientes de correlação significativos entre CC e níveis de pressão arterial e ApoB em homens, e entre CC e índice Homa-IR e triglicerídios em mulheres. Após ajuste pelos valores de VO2máx observamos correlações significativas entre CC e ApoB e índice Homa-IR em homens, e entre CC e índice Homa-IR em mulheres. Conclui-se que, dependendo do sexo, a quantidade e a distribuição da gordura corporal podem apresentar ações distintas na resistência insulínica e nas disfunções associadas. A aptidão cardiorrespiratória per se parece não contribuir na minimização da associação entre a distribuição centrípeta da gordura e o índice Homa-IR; porém, apresenta considerável impacto na associação entre a distribuição centrípeta da gordura e o metabolismo lipídico e os níveis de pressão arterial, sobretudo nos homens.

The purpose of this paper was to investigate associations between the centripetal distribution of the body fat and serum lipid-lipoproteins, blood pressure and the index Homa-IR of insulin resistance, adjusting for indicators of overweight and cardiorespiratory fitness. Eighty-nine voluntaries were analyzed (44 men and 45 women). The centripetal distribution of the body fat was analyzed through waist circumference (CC) and the overweight by the body mass index (BMI). The cardiorespiratory fitness was followed by the estimate VO2max by test of walking. After adjusted for BMI values were found significant coefficient of correlation between CC and levels of blood pressure and ApoB in men, and between CC and index Homa-IR and triglycerides in women. After adjusted for VO2max values were verified significant correlations between CC and ApoB and index Homa-IR in men, and between CC and index Homa-IR in women. In conclusion, depending on the sex, the quantity and distribution of the body fat can present different actions in the insulin resistance and associated dysfunctions. The cardiorespiratory fitness per se seems not to contribute on the minimization of the association between the centripetal distribution of the body fat and the index Homa-IR; but presents a considerable impact on the association between the centripetal distribution of the body fat and the lipid metabolism and the levels of blood pressure, mainly in men.
