Abstract This study investigated the association between the IFITM3 rs12252 polymorphism and the severity and mortality of COVID-19 in hospitalized Brazilian patients. A total of 102 COVID-19 patients were included, and the outcomes of interest were defined as death and the need for mechanical ventilation. Genotypes were assessed using Taqman probes. No significant associations were found between the rs12252 polymorphism and COVID-19 outcomes in the original sample, both for death and the need for mechanical ventilation. A meta-analysis, incorporating previous studies that used death as a severity indicator, revealed no association in the allelic and C-recessive models. However, due to the rarity of the T allele and its absence in the sample, further replication studies in larger and more diverse populations are needed to clarify the role of rs12252 in COVID-19 prognosis. RESUMO Objetivou-se com este trabalho avaliar o efeito do primeiro desbaste sobre a morfometria de um plantio de Tectona grandis L.f. (teca). O delineamento experimental foi o de blocos ao acaso, com quatro tratamentos e três repetições sendo: T1 = testemunha (sem desbaste), T2, T3 e T4 = remoção da área basal de 20%, 30% e 40% respectivamente. As variáveis morfométricas avaliadas foram: proporção de copa (PC), índice de abrangência (IA), grau de esbeltez (GE), índice de saliência (IS), formal de copa (FC) e comprimento de copa (CC). A aplicação de desbastes em teca influenciou significativamente em todas as variáveis morfométricas avaliadas, com exceção de proporção de copa (PC) e índice de saliência (IS). As intensidades de desbastes (20%, 30% e 40%) não apresentaram diferença significativa para as variáveis: altura comercial do fuste (HF), comprimento de copa (CC), diâmetro de copa (DC), proporção de copa (PC) e forma de copa (FC) da Tectona grandis até o quarto ano pós-desbaste.

ABSTRACT The aim of this study was to evaluate the effect of first thinning on the morphometry of trees in a forest plantation of Tectona grandis Lf (teak). The experimental design used was randomized block with four treatments and three replications were: T1 = control treatment (no thinning), T2, T3 end T4 with the removal of basal area the 20, 30 and 40% respectively. The evaluated morphometric variables were: crown proportion (PC), coverage index (AI), slenderness degree (GE), salience index (IS), crown form (FC) and crown length (CC). The application of thinning in teak, significantly influenced in all morphometric variables analyzed, except for the proportion of room (PC) and the projection index (SI). The thinning intensities (20%, 30% and 40%) presented no significant differences for variables: commercial bole height (HF), crown length (CL), crown diameter (DC), crown proportion (PC) and crown form (FC) of teak to the fourth post-thinning year.

The selection of new microorganisms able to produce antimicrobial compounds is hoped for to reduce their production costs and the side effects caused by synthetic drugs. Clavulanic acid is a β-lactam antibiotic produced by submerged culture, which is widely used in medicine as a powerful inhibitor of β-lactamases, enzymes produced by bacteria resistant to antibiotics such penicillin and cephalosporin. The purpose of this work was to select the best clavulanic acid producer among strains of Streptomyces belonging to the Microorganism Collection of the Department of Antibiotics of the Federal University of Pernambuco (DAUFPE). Initially, the strains were studied for their capacity to inhibit the action of β-lactamases produced by Klebsiella aerogenes ATCC 15380. From these results, five strains were selected to investigate the batch kinetics of growth and clavulanic acid production in submerged culture carried out in flasks. The results were compared with the ones obtained by Streptomyces clavuligerus ATCC 27064 selected as a control strain. The best clavulanic acid producer was Streptomyces DAUFPE 3060, molecularly identified as Streptomyces variabilis, which increased the clavulanic acid production by 28% compared to the control strain. This work contributes to the enlargement of knowledge on new Streptomyces wild strains able to produce clavulanic acid by submerged culture.

No presente estudo, avaliaram-se os efeitos da alimentação de diferentes concentrados sobre o comportamento de ovinos. A hipótese é a de que a polpa cítrica estimularia a ruminação e reduziria o risco de ocorrência de urolitíase, podendo substituir outros concentrados. Dez ovinos adultos, mestiços da raça Santa Inês, foram distribuídos em um quadrado latino com cinco tratamentos, sendo quatro destes contendo dietas com 30% de feno de capim coast-cross e 70% dos seguintes concentrados: polpa cítrica peletizada, polpa cítrica farelada, fubá de milho e farelo de trigo, e uma dieta controle com 80% de feno e 20% de ração comercial peletizada. Após 21 dias de adaptação, a cada dieta os animais foram colocados em monitoramento visual, com registro de atividade a cada três minutos, durante 24 horas consecutivas, por meio do qual foi identificado o tempo gasto na ruminação, na ingestão de alimentos e em repouso. A posição dos animais (em pé ou deitados) também foi observada. O período diurno foi considerado entre seis e 18 horas. Para comparação entre os tratamentos, os dados foram avaliados por meio de ANOVA e do teste de Tukey. Para a avaliação circadiana e entre as posições, foi utilizado o teste t de Student. Dietas com polpa cítrica promoveram tempo de ruminação semelhante aos do grupo de controle (601, 590 e 669 min, respectivamente), mas superior ao grupo alimentado com fubá de milho (421min). A produção de saliva estimada no grupo controle (26L/d) foi maior do que nos demais grupos, e os grupos com polpa cítrica tiveram maior produção de saliva do que o grupo com fubá de milho (21L/d). Ovinos em dietas ricas em concentrados descansam mais durante a noite. Em todas as dietas, cerca de 90% da ruminação ocorreu com os animais deitados. A polpa cítrica peletizada e a farelada, e em menor grau o farelo de trigo, promoveram adequadamente a ruminação. Este concentrado pode ser utilizado como medida preventiva visando diminuir a incidência de urolitíase em rebanhos ovinos.

In this study we examined the effects of different feed concentrates on sheep behaviour. Our hypothesis was that citric pulp would stimulate rumination and be capable of replacing other concentrates traditionally used for feeding in confinement, to reduce the risk of urolithiasis. Ten adult Santa Inês sheep were distributed in a Latin square with five different diets, one control diet with 80% hay and 20% commercial feed and four diets containing 30% coast-cross hay and 70% of the following concentrates: pelleted citrus pulp, citrus pulp meal, cornmeal or wheat bran. After 21d of adaptation to each one of the five diets, the sheep were visually monitored for 24 h at 3 min intervals to record the time spent ruminating, time spent eating and time spent resting; the animals' positions (standing or lying down) were also noted. Daytime was considered to be from 06:00h to 18:00h. The data were evaluated using ANOVA, with Tukey post-hoc test or throughout Two-sample T test for circadian and position assessment. Citrus pulp diets resulted in time spent ruminating similar to the control diet (601, 590 and 669 min, respectively), but greater (P<0.05) than the cornmeal group (421min), which showed that citrus pulp generated effective rumination. The estimated saliva production in the control diet (26L) was greater than in the other groups, and was greater in the citrus pulp groups (24L/d) than cornmeal (21L/d). Feeding with cornmeal led to shorter time spent eating and time spent ruminating than all other diets. The sheep had higher time spent resting at night when fed concentrates (P<0.05). For all diets, about 90% of the time spent ruminating occurred with the animals lying down. Pelleted citrus pulp, citrus pulp meal and to a lesser degree wheat bran, led to adequate time spent ruminating. The use of citrus pulp can act as a preventive management measure to reduce the incidence of urolithiasis in sheep flocks.

Azospirillum brasilense is a diazotroph that associates with important agricultural crops and thus has potential to be a nitrogen biofertilizer. The A. brasilense transcription regulator NifA, which seems to be constitutively expressed, activates the transcription of nitrogen fixation genes. It has been suggested that the nitrogen status-signaling protein GlnB regulates NifA activity by direct interaction with the NifA N-terminal GAF domain, preventing the inhibitory effect of this domain under conditions of nitrogen fixation. In the present study, we show that an N-terminal truncated form of NifA no longer required GlnB for activity and lost regulation by ammonium. On the other hand, in trans co-expression of the N-terminal GAF domain inhibited the N-truncated protein in response to fixed nitrogen levels. We also used pull-down assays to show in vitro interaction between the purified N-terminal GAF domain of NifA and the GlnB protein. The results showed that A. brasilense GlnB interacts directly with the NifA N-terminal domain and this interaction is dependent on the presence of ATP and 2-oxoglutarate.

Azospirillum brasilense is a diazotroph found in association with important agricultural crops. In this organism, the regulation of nitrogen fixation by ammonium ions involves several proteins including the uridylyltransferase/uridylyl-removing enzyme, GlnD, which reversibly uridylylates the two PII proteins, GlnB and GlnZ, in response to the concentration of ammonium ions. In the present study, the uridylylation/deuridylylation cycle of A. brasilense GlnB and GlnZ proteins by GlnD was reconstituted in vitro using the purified proteins. The uridylylation assay was analyzed using non-denaturing polyacrylamide gel electrophoresis and fluorescent protein detection. Our results show that the purified A. brasilense GlnB and GlnZ proteins were uridylylated by the purified A. brasilense GlnD protein in a process dependent on ATP and 2-oxoglutarate. The dependence on ATP for uridylylation was similar for both proteins. On the other hand, at micromolar concentration of 2-oxoglutarate (up to 100 µM), GlnB uridylylation was almost twice that of GlnZ, an effect that was not observed at higher concentrations of 2-oxoglutarate (up to 10 mM). Glutamine inhibited uridylylation and stimulated deuridylylation of both GlnB and GlnZ. However, glutamine seemed to inhibit GlnZ uridylylation more efficiently. Our results suggest that the differences in the uridylylation pattern of GlnB and GlnZ might be important for fine-tuning of the signaling pathway of cellular nitrogen status in A. brasilense.

Avaliou-se o processo de enriquecimento protéico da palma forrageira (Opuntia ficus-indica Mill) com levedura Sacharomyces cerevisiae em cultivo semi-sólido, visando melhorar o valor nutritivo da palma para ser utilizada na alimentação de ruminantes. A levedura foi utilizada nas concentrações de 1, 2 e 3% em base úmida no substrato formado pela palma forrageira, incubada em biorreatores durante 6, 12, 24 e 36 horas de fermentação. O delineamento experimental foi inteiramente ao acaso, em arranjo de parcelas subdivididas com quatro repetições. O conteúdo de proteína bruta quando se utilizou concentração de 3% de inóculo, no período de seis horas, aumentou de 4,4% na forma in natura para 10,4% após o processamento. Os teores protéicos na concentração de 1% do inóculo foram de 6,1, 8,1, 8,1 e 9,2%; na concentração de 2%, 9,6, 9,7, 9,8 e 9,8% e na concentração de 3%, 10,4, 10,4 7,9 e 7,9%, nos períodos de 6, 12, 24 e 36 horas de fermentação, respectivamente. Uma fonte alternativa para arraçoamento de ruminantes, pode ser obtida pela bioconversão da palma forrageira.

The process of protein enrichment of the forage palm (Opuntia ficus-indica Mill) using the Saccharomyces cerevisiae yeast in a semi-solid culture to improve the nutritional value of forage palm for ruminants feeding was evaluated. The yeast concentrations of 1, 2 and 3% (wet basis) in the forage palm substrate were used. The periods of incubation were of 6, 12, 24, and 36 hours. A complete randomized experimental design in a split plot arrangement with four replicates was used. The crude protein content increased from 4.4% (in natura) to 10.4% when 3% of inoculums were used and the processing period was of 6 hours. The observed protein contents for 1% of the inoculum, used for the fermentation periods of 6, 12, 24, and 36 hours were 6.1, 8.1, 8.1, and 9.2%, respectively. These values were 9.6, 9.7, 9.8, and 9.8% for 2% of the inoculum, and 10.4, 10.4, 7.9, and 7.9% for 3% of the inoculum, respectively. An alternative for ruminant feeding can be obtained by bioconversion of forage palm.

The pupunha (Guilielma speciosa) is the fruit of a palm tree typical of the Brazilian Northern region, whose stem is used as a source of heart of palm. The fruit, which is about 65% pulp, is a source of oil and carotenes. In the present work, an analysis of the kinetics of supercritical extraction of oil from the pupunha pulp is presented. Carbon dioxide was used as solvent. The extractions were carried out at 25 MPa and 323 K and 30 MPa and 318 K. The chemical composition of the extracts in terms of fatty acids was determined by gas chromatography. The amount of oleic acid, a saturated fatty acid, in the CO2 extracts was larger than that in the extract obtained with hexane. The overall extraction curves were modeled using the single-parameter model proposed in the literature to describe the desorption of toluene from activated coal.
