ABSTRACT: In the state of Amazonas, the ‘Pêra’ orange scion is traditionally cultivated on ‘Rangpur’ lime tree. The restricted use of a single scion-rootstock combination has left local orchards vulnerable to adverse conditions and incidence of pests and diseases, which may negatively interfere with both productivity and fruit quality and the citrus activity sustainability in the state. The expansion of the number of scion-rootstock combinations becomes a viable alternative to overcome such problems. To meet this demand from citrus growers, 30 combinations with 10 scions and 3 rootstocks were evaluated in the sixth and seventh year after planting seedlings in the field in the municipality of Rio Preto da Eva, AM. In this experiment, productivity, juice yield, soluble solids, pH, titratable acidity, ratio and technological index were evaluated. ‘Pineapple’ scion with ‘Sunki Tropical’ and ‘BRS Bravo’ rootstocks can be productive alternatives in relation to traditional ‘Pêra’ orange scion under ‘Rangpur’ lime combination in the state of Amazonas. ‘Lima’ orange with ‘Sunki Tropical’ mandarin, ‘Rangpur lime SC’ and ‘BRS Bravo’ lime rootstocks can be alternatives for the fresh market in the state. Combinations that showed good fruit quality are ‘Pêra’, ‘Salustiana’, ‘Valência Tuxpan’ and ‘Westin’ scions with ‘BRS Bravo’ rootstock, and ‘Pêra D6’ scion with ‘Rangpur lime SC’ lemon rootstock. These combinations are alternatives for the diversification of orchards in the state of Amazonas. ABSTRACT This study aimed to assess an alkaline protease supplemented in diets with and without crude protein (CP) reduction on performance, apparent total tract digestibility (ATTD), blood parameters, and carcass and meat traits in growing-finishing pigs. Forty male pigs (26.2±1.2 kg) were randomly allocated into one of five treatments: negative control (NC, 2% and 1% reduction of CP in grower and finisher phases, respectively, no protease); NC150: NC + 150 mg protease kg−1 diet; NC300: NC + 300 mg protease kg−1 diet; PC: positive control (no CP reduction and protease); and PC300: PC + 300 mg protease kg−1 diet, with eight replicates of one pig/pen. Pigs fed NC showed greater average daily feed intake (ADFI) than pigs fed NC300 or PC and lower ADFI compared to pigs fed NC150. Pigs fed PC had lower ADFI than those fed PC300. Greater average daily gain and gain to feed ratio (G:F) were observed in pigs on NC compared with those on NC300 or NC150 and NC300, respectively. Pigs fed PC showed better G:F than pigs fed PC300. Lower coefficients of ATTD (CTTAD) of dry and organic matter (OM), digestible dry matter (DDM), digestible organic matter (DOM), and digestible protein were observed in growing II pigs fed NC compared with pigs fed NC150 or NC300. Pigs fed NC showed a lower DP compared with PC or NC150. Positive control group showed increased digestible protein compared with NC. Finishing II pigs fed NC showed lower DDM, DOM, CTTAD of OM, and gross energy than pigs fed NC150 or NC300. Pigs fed PC showed greater albumin concentration compared with pigs fed PC300 in finishing II. Pigs fed NC and PC300 showed greater luminosity in the l. thoracis muscle than pigs fed PC. A greater color score was evidenced in the l. thoracis in pigs fed PC compared with pigs fed PC300. The dietary supplementation of isolated alkaline protease and CP-reduced diets improves ATTD without supporting pig performance. ABSTRACT: Rabies is a viral encephalitis that affects mammals, including humans. Rapid and effective laboratory diagnosis of the rabies virus is critical for public health. This study evaluated the operational, technical, and financial viability of the RT-qPCR in replacement of inoculation in mice for diagnosing rabies in the laboratory routine. A total of 316 samples of mammalian brains were analyzed, 121 positives and 195 negatives, previously diagnosed by direct immunofluorescence (DIF) and mouse inoculation test (MIT). The samples were submitted to the duplex TaqMan RT-qPCR technique. The accuracy, sensitivity, and specificity of RT-qPCR were analyzed. We analyzed the costs for performing the RT-qPCR technique and compared it with the cost of MIT. The results showed 99.37% accuracy, 99.17% sensitivity, and 99.49% specificity by RT-qPCR when related to DIF and MIT results, which proved to be a robust and repeatable technique. The minimum time for a positive diagnosis was reduced in RT-qPCR (1 day) if compared to MIT (9.64 days), with a 17.7% reduction in the cost of the molecular technique. The present study demonstrated that the molecular biology technique is an efficient tool to diagnose rabies in the laboratory routine, being able to replace MIT.
Abstract Chromosomal patterns are valuable tools in evolutionary approaches. Despite the remarkable expansion of fish cytogenetic data, they are still highly deficient concerning deep oceanic species, including the Gempylidae snake mackerels. The snake mackerels are important commercial species composed by meso- and bento-pelagic predators with very limited information available about their lifestyle and genetics patterns. This study presents the first chromosomal data of two circumglobal species of this family, Ruvettus pretiosus and Promethichthys prometheus, from the São Pedro and São Paulo Archipelago. Conventional analyses, chromosomal staining with base-specific fluorochromes, and fluorescence in situ hybridization (FISH) for mapping of repetitive DNA classes were used. Both species have 2n = 48 chromosomes, but they highly differ regarding the karyotype formula (FN = 50 and FN = 84). The 18S rDNA/Ag-NOR and the 5S rDNA sites have a syntenic bi-telomeric array in R. pretiosus, but an independent distribution in P. prometheus. The transposable elements are dispersed, while the microsatellites are also clustered in the centromeric and terminal regions of some chromosomes. It is noteworthy that despite the 2n conservation, a marked macro and microstructural diversifications, mainly mediated by pericentric inversions, differentiates the karyotypes of the species, pointing to a particular chromosomal trajectory of the gempylids among marine fish. approaches meso bentopelagic bento pelagic family prometheus Archipelago analyses basespecific base specific fluorochromes FISH (FISH used n 4 chromosomes 5 84. 84 . 84) S rDNA/AgNOR rDNAAgNOR rDNA/Ag NOR Ag bitelomeric bi telomeric R P dispersed conservation diversifications inversions 8 AgNOR rDNAAg

Abstract Brazil harbors the highest richness of Convolvulaceae with 424 species recognized mainly distributed in the Amazon, Atlantic Forest, Caatinga and Cerrado phytogeographic domains. Seventeen of these species are representatives of Bonamia, with ten endemic to the country. The aim of the study was to map the distribution of this group to understand its richness, its sampling and detecting areas of endemism, valuable information for conservation. We collected data gathered from herbaria and from the online database. The data were refined (1) excluding of records not at the species level; (2) records with no identification of collection site or with only the identification of the state of collection. There was calculated the richness, the number of records and an estimate of richness per cell. We conducted a parsimony analysis of endemism for distribution analysis. Finally, the knowledge of richness for the species was analyzed. There were gathered 420 occurrence records, in 87 grid cells. Most grid cells observed in the study presented one species. Two endemic areas were found for the genus. The results contribute to the understanding of the distribution of the group in Brazil, highlighting shortfalls in collections. 42 Amazon Forest domains Bonamia country conservation database 1 (1 level 2 (2 cell Finally analyzed 8 genus collections 4 (

ATTRvPN PN . Abstract Background Hereditary transthyretin amyloidosis (ATTRv) is an inherited, progressive, and fatal disease still largely underdiagnosed. Mutations in the transthyretin (TTR) gene cause the TTR protein to destabilize, misfold, aggregate, and deposit in body tissues, which makes ATTRv a disease with heterogeneous clinical phenotype. Objective To describe the long-term efficacy and safety of inotersen therapy in patients with ATTRv peripheral neuropathy (ATTRv-PN). Methods Patients who completed the NEURO-TTR pivotal study and the NEURO-TTR OLE open-label extension study migrated to the present study and were followed-up for at least 18 more months to an average of 67 months and up to 76 months since day 1 of the inotersen therapy (D1–first dose of inotersen). Disease progression was evaluated by standard measures. Results Ten ATTRv-PN patients with Val30Met mutation were included. The mean disease duration on D1 was of 3 years, and the mean age of the patients was of 46.8 years. During an additional 18-month follow up, neurological function, based on the Neuropathy Impairment Score and the Polyneuropathy Disability Score, functionality aspects (Karnofsky Performance Status), and nutritional and cardiac aspects were maintained. No new safety signs have been noted. Conclusion The treatment with inotersen was effective and well tolerated for the average of 67 months and up to 76 months. Our results are consistent with those of larger phase-III trials. (ATTRv inherited progressive underdiagnosed (TTR destabilize misfold aggregate tissues phenotype longterm long term ATTRvPN. ABSTRACT Introduction: Internal carotid artery (ICA) stenosis causes about 15% of ischemic strokes. Duplex ultrasonography (DUS) is the first line of investigation of ICA stenosis, but its accuracy varies in the literature and it is usual to complement the study with another more accurate exam when faced with significant stenosis. There is a lack of studies that compare DUS with angiotomography (CTA) in the present literature. Methods: we performed an accuracy study, which compared DUS to CTA of patients in a tertiary hospital with a maximum interval of three months between tests. Patients were selected retrospectively, and two independent and certified vascular surgeons evaluated each image in a masked manner. When there was discordance, a third evaluator was summoned. We evaluated the diagnostic accuracy of ICA stenosis of 50-94% and 70-94%. Results: we included 45 patients and 84 arteries after inclusion and exclusion criteria applied. For the 50-94% stenosis range, DUS accuracy was 69%, sensitivity 89%, and specificity 63%. For the 70-94% stenosis range, DUS accuracy was 84%, sensitivity 61%, and specificity 93%. There was discordance between CTA evaluators with a change from clinical to surgical management in at least 37.5% of the conflicting reports. Abstract Groupers (Epinephelidae and Serranidae) have attracted special attention to fish farming, and their species offer good opportunities for successful hybridizations. Cytogenetic data allow a better understanding of the role of karyotypic diversification in the acquisition of post-zygotic reproductive isolation (RI). Thus, chromosomal analyses were performed on E. striatus (Caribbean Sea), E. coioides and E. tauvina (Indo-Pacific Region), using standard procedures and mapping of six repetitive DNA classes by the in situ hybridization. The three species have 2n=48 chromosomes. The karyotypes of E. coioides and E. striatus are composed only of acrocentric chromosomes (FN=48), while E. tauvina has 8 submetacentric chromosomes (FN=56). Heterochromatin has a preferential centromeric distribution, and the microsatellite repeats are dispersed throughout the chromosomes of all species. The 18S and 5S rDNA sites are unique but show a colocalization arrangement in E. tauvina and E. striatus. The chromosomal organization suggests that the three species still maintain a significant amount of syntenic regions. The range of the karyotype divergence and the RI levels showed low, but goes turn proportionally greater in relation to the divergence time between the parental species. The slow acquisition of postzygotic RI is consistent with the high karyotype homogeneity presented by Epinephelidae family. Serranidae farming hybridizations post zygotic RI. . ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others. publications problem uptodate up date classifications context exception (CTFB http//fauna.jbrj.gov.br/, httpfaunajbrjgovbr http //fauna.jbrj.gov.br/ , jbrj gov br (http://fauna.jbrj.gov.br/) 2015 Brazil 80 specialists 1 2024 133691 133 691 133,69 125138 125 138 125,13 82.3%, 823 82 3 (82.3% 102000 102 000 102,00 7.69%, 769 7 69 (7.69% 11000 11 11,00 . 3,567 3567 567 (3,56 2,292 2292 2 292 (2,29 1,833 1833 833 (1,83 1,447 1447 447 (1,44 1000 1,00 831 (83 628 (62 606 (60 520 (52 50 users science health biology law anthropology education others http//fauna.jbrj.gov.br/ faunajbrjgovbr //fauna.jbrj.gov.br (http://fauna.jbrj.gov.br/ 201 8 202 13369 13 133,6 12513 12 125,1 82.3% (82.3 10200 10 00 102,0 7.69% 76 6 (7.69 1100 11,0 3,56 356 56 (3,5 2,29 229 29 (2,2 1,83 183 83 (1,8 1,44 144 44 (1,4 100 1,0 (8 62 (6 60 52 (5 5 http//fauna.jbrj.gov.br (http://fauna.jbrj.gov.br 20 1336 133, 1251 125, 82.3 (82. 1020 0 102, 7.69 (7.6 110 11, 3,5 35 (3, 2,2 22 (2, 1,8 18 (1, 1,4 14 4 ( 82. ABSTRACT Background: Malaria is an acute febrile parasitic disease that significantly impacts global public health. In Brazil, the most studied endemic area for the disease is the Amazon region. This study aims to analyze temporal, spatial, and spatiotemporal patterns of malaria in the extra-Amazon region of Brazil over a 22-year period. Methods: We conducted a time-series study from 2001 to 2022, encompassing both autochthonous and imported cases. Time trend analysis was employed to assess fluctuations in incidence rates over the years. Spatial clusters of infection risk were identified using the Local Moran Index and Kulldorff's scan. Results: A total of 18,633 malaria cases were identified in the extra-Amazon region, including 1,980 autochthonous, 13,836 imported, and 2,817 of unknown origin. During the first period (2001-2011), 1,348 autochthonous and 9,124 imported cases were reported. In the second period (2012-2022), there were 632 autochthonous and 4,712 imported cases. The state of Espírito Santo exhibited a decreasing trend but maintained the highest incidence rates throughout the study. The number of municipalities at high risk for autochthonous cases declined, with Espírito Santo, Minas Gerais, and Piauí having the most municipalities with high rates. For imported cases, the federative units with the highest numbers in both periods were Ceará, Distrito Federal, Goiás, Minas Gerais, Piauí, and Paraná. Conclusions: The data reveal the areas most affected by malaria and thus of highest priority for implementing control strategies. ABSTRACT The umbu-cajazeira (Spondias sp.) is a fruit tree native to the semi-arid of Brazil, with significant environmental and agro-socioeconomic potential for the region. Determining leaflet area (LA) is essential for understanding the interactions between plant growth, physiology, nutrition, and environment. Thus, this research aims to construct allometric equations that non-destructively estimate the umbu-cajazeira (Spondias sp.) leaflet area, considering the leaflet dimensions (length and width). The leaflets were collected from mother plants of umbu-cajazeira (Spondias sp.) and individually measured for length (L), width (W), and LA using digitized images. These data were submitted for descriptive and regression analysis. The LA was estimated using, linear power, and exponential regression models based on the L and W of the leaflets. The best model and equation were chosen based on the following selection criteria: coefficient of determination, Willmott agreement index, root mean square error, and mean absolute error. The power model using LW presented the best equation ŷ = 0.76 * LW0.98 to estimate the leaflet area of umbu-cajazeira (Spondias sp.). This study provides a reliable, accurate, fast, and non-destructive approach for agronomic researchers and growers to determine the LA of the species. umbucajazeira umbu cajazeira Spondias sp. sp semiarid semi arid Brazil agrosocioeconomic agro socioeconomic region (LA growth physiology nutrition environment Thus nondestructively non destructively width. . width) L, , (L) W, (W) images analysis criteria determination index error 076 0 76 0.7 LW098 LW0 98 LW0.9 sp.. reliable accurate fast nondestructive destructive species (L (W 07 7 0. Abstract Objective: To analyze the performance of the cystic fibrosis (CF) newborn screening (NBS) program over its first five years in a Brazilian northeastern state. Methods: A population-based study using a screening algorithm based on immunoreactive trypsinogen (IRT)/IRT. Data were retrieved from the state referral screening center registry. The program performance was evaluated using descriptive indicators such as the results of an active search, coverage, newborn’s age at the time of blood sampling, the time between sample collection and its arrival at the laboratory, and the child’s age at diagnosis of disease. Results: The public CF screening program covered 82.6% of the 1,017,576 births that occurred, with an accumulated five-year incidence of 1:20,767 live births. The median (25th-75th) age at diagnosis was 3.5 (2.3–7.3) months. The sampling before 7 days of life for the first IRT (IRT1) increased between 2013 and 2017 from 42.2 to 48.3%. Around 5% of IRT1 samples and 30% of the second samples were collected after 30 days of life. In the first and second stages of screening, 23.6% and 19.9% of the infants, respectively, were lost to follow-up. In both stages of screening, the samples were retained at the health units for a median (25th–75th) of 9.0 (7.0–13.0) days. Conclusions: The coverage by the CF-NBS program was satisfactory as compared to other Brazilian state rates and the percentage of IRT1 samples collected within the first week of life increased progressively. However, time of samples retention at the health units, inappropriate sampling, inherent methodological problems, and loss of follow-up need to improve.
