Bioelectrical impedance phase angle and nutritional status in children with intestinal failure on prolonged parenteral nutrition

Abstract Objective To compare the phase angle (PhA) through bioelectrical impedance (BIA) of children with intestinal failure (IF) using prolonged parenteral nutrition (PN) followed by an Intestinal Rehabilitation Program, with a control group. Methods Children under 10 years of age with IF using prolonged PN for >60 days (study group) were included. The control group consisted of healthy children without chronic pathologies, matched by sex and age. Anthropometric parameters evaluated were: weight, height, weight/age z-score (W/A), height/age z-score (H/A), BMI, BMI/A z-score, arm circumference, triceps skinfold, subscapular skinfold, mid-arm muscle circumference. BIA parameters were resistance (R), reactance (Xc), and phase angle (PhA). Results Twenty-eight children were included in the study group, median (IQR) age was 11 (8-27) months, 53.6 % were male. In the control group, 28 children were included, median (IQR) age was 12.5 (8-24.7) months, 50 % were male. Children from the study group had W/A z-scores and H/A z-scores significantly lower than controls. There was no significant difference between PhA in the study group and controls, [median (IQR) 4.3° (3.8;4.6) vs 4.0° (3.8;5.4) respectively, p = 0.980]. Prematurity was significantly higher in the study group than in the controls, but there was no significant correlation between gestational age at birth and PhA of the children from the study group. Conclusion Children with IF using prolonged PN showed lower W/A and H/A compared to the control group, but without significant difference between the PhA of children with IF compared to controls. RESUMO Foram avaliadas 60 adrenais de 30 cães, divididos em três grupos, de acordo com a idade: grupo 1 = jovens saudáveis (10 animais) - de um a três anos de vida; grupo 2 = adultos saudáveis (10 animais) - de quatro a nove anos de vida; e grupo 3 = senis saudáveis (10 animais) - a partir de 10 anos de vida. O delineamento experimental foi ao acaso, com significância de 5%. No comprimento, houve diferença significativa (P=0,017) entre os cães jovens e os idosos. A espessura do polo cranial apresentou diferença significativa (P=0,001) entre jovens e adultos quando comparados com o grupo de idosos. A espessura do polo caudal não foi significativa para adultos quando comparadas com idosos e jovens. Entre idosos e jovens, a diferença foi significativa (P=0,024). As médias das velocidades diferiram entre os grupos de animais jovens e adultos quanto à adrenal esquerda e a seu polo cranial. ABSTRACT Sixty adrenals of 30 dogs were evaluated, divided into three groups, according to age: Group 1 = healthy youngsters (10 animals) - from one to three years old; Group 2 = healthy adults (10 animals) - four to nine years old; and Group 3 = healthy senile animals (10 animals) - older than ten years old. The thickness of the cranial pole showed a significant difference (P=0.001) between young and adults when compared with the elderly group. The mean speeds differed between the groups of young and adult animals regarding the left adrenal and its cranial pole. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO O linfedema crônico progressivo (LCP) é uma enfermidade incapacitante de cavalos de tração que afeta o sistema linfático e se manifesta especialmente na região distal dos membros. A progressão da doença resulta em deformações na região distal dos membros, especialmente na quartela, com fibrose e lesões nodulares na pele. Foi atendido um garanhão da raça Friesian, de 15 anos de idade, com histórico de aumento de volume, nodulações e deformações na região distal dos membros torácicos, com início há aproximadamente quatro anos. Ao exame radiográfico simples, foram identificadas nodulações na pele e no subcutâneo e, ao exame venoso contrastado, identificou-se marcante dilatação II veia digital palmar comum em ambos os membros torácicos. Foi isolado Streptococcus equi e Proteus mirabilis ao cultivo tecidual e identificou-se sarna do tipo corióptica ao raspado de pele. No exame histopatológico, foram identificadas perivasculite e dilatação de vasos linfáticos. Os achados clínicos, histórico, de raça do animal e os exames complementares direcionaram o diagnóstico para o LCP. A pesquisa da literatura nacional não revelou descrições clínicas dessa enfermidade, mas, devido ao aumento do número de animais de tração no país nos últimos anos, a descrição dessa enfermidade se torna importante. O diagnóstico correto e precoce é necessário para retardar a progressão da doença. Nesse sentido, ressalta-se a relevância da divulgação nacional desses achados, a fim de contribuir para o diagnóstico precoce e permitir o retardo da evolução da doença.
ABSTRACT Chronic Progressive Lymphoedema (CPL) is a disabling disease of draft horses that affects lymphatic system function typically in the distal limbs. Deformities of distal limbs, especially of the pastern, with fibrosis and skin nodular lesions are consequences of disease progression. A 15-year-old Friesian stallion presented for evaluation with history of forelimb enlargement and nodule formations distal to the carpus for four years. Simple radiographs showed soft tissue nodular lesions and venous contrasted radiography showed intense enlargement of II common digital palmar vein of both forelimbs. Tissue culture tests revealed Streptococcus equi and Proteus mirabilis isolation and skin scraping test identified Chorioptes bovis mites. Histological examination revealed perivasculitis and lymph vessels distention. History, clinical and histological findings, and complementary exams suggested CPL diagnosis. We were unable to find previously published cases describing this disease in Brazil, where the increasing number of draft horses requires attention to this problem. The correct and early diagnosis substantially delays disease progression. Therefore, we highlight the need for nationwide propagation of these data to ensure better diagnosis and early treatment of future CPL cases.

Heart rate variability (HRV) is a relevant physiological variable for the estimation of cardiac autonomic function. Although the gold standard for HRV registration is the electrocardiogram (ECG), several applications (APPs) have been increasingly developed. The evaluation carried out by these devices must be compatible with ECG standards. The aim of this study was to compare the data obtained simultaneously with ECG and APP with chest heart rate transmitters. Fifty-six healthy individuals (28 men and 28 women) were evaluated at rest through a short simultaneous HRV measurement with both devices. Data from both acquisition systems were analyzed separately using their own analysis software and exported and analyzed using a validated software. Signal recordings were compatible between the two acquisition systems (Pearson r=0.99; P<0.0001). Although a high correlation was found for the HRV variables obtained in the time domain (Spearman r=0.99; P<0.0001), the correlation decreased in the frequency domain (Pearson r=0.85; P<0.0001) when two software programs were used. Comparison of the averages of spectral analysis parameters also showed differences when HRV data were analyzed separately in each device for low-frequency (LF) and high-frequency (HF) bands. Although the portability of these mobile devices allows for optimal HRV evaluation, the direct analysis obtained from these devices must be carefully evaluated with respect to frequency domain parameters.

RESUMO Objetivou-se avaliar um programa de controle da artrite encefalite caprina (AEC), por meio de testes diagnósticos sensíveis, separação de mãe e cria após o parto e medidas de manejo, com o intuito de formar rebanho livre do vírus. Utilizou-se um total de 47 cabritos da raça Saanen, mantidos isoladamente até o resultado dos primeiros testes de reação em cadeia de polimerase nested (PCR nested) e Western Blotting (WB), com base na coleta de sangue no momento do nascimento (M0). No PCR nested, quatro animais foram positivos, no M0, e foram eutanasiados. Posteriormente, os demais 43 cabritos foram submetidos à coleta de sangue aos 60 (M60) e 270 (M270) dias de vida para realização de novos testes de WB e PCR nested, que não detectaram animais positivos. Pode-se afirmar que a metodologia adotada neste estudo foi efetiva no controle da doença, nas fases de aleitamento e pós-aleitamento, e que a combinação do sistema de manejo, a fim de propiciar diminuição de risco de transmissão horizontal, com técnicas de diagnóstico mais apuradas, como o WB e a PCR nested, é relevante para elaboração de plano estratégico de controle da enfermidade.
ABSTRACT We aimed to evaluate a program to control Caprine Arthritis Encephalitis (CAE), using diagnostic tests, separation of the mother and postpartum and other management measures, in order to form a free flock of the virus. We used a total of 47 Saanengoats in isolation until the results of the first nested Polymerase Chain Reaction (nested PCR) and Western Blotting (WB) tests, based on blood collection at the time of birth (M0). In the nested PCR, 4 animals were positive, at M0, and were eliminated. Later, the other 43goats were submitted to blood collection at 60 (M60) and 270 (M270) days of life to perform new tests of WB and nested PCR, which did not detect positive animals. We can affirm that the methodology adopted in this study was effective in the control of the disease, in the phase of breastfeeding and post-breastfeeding, and that the combination of the management system, which allows a reduction of risk of horizontal transmission, with more accurate diagnostic techniques, such as WB and nested PCR, is relevant for the elaboration of a strategic plan for the disease control.

RESUMO Este artigo tem como objetivo relatar um caso de infecção por Mycobacterium bovis em um gato doméstico, com seis meses de idade, enfatizando seus aspectos clínicos e terapêuticos. O felino apresentava como histórico a ingestão de leite de vaca não fervido e posteriormente desenvolveu sinais de anorexia, tosse não produtiva e linfadenomegalia. Para elucidação do diagnóstico, foram realizados exames complementares de imagem, hematológicos, citológicos e reação em cadeia da polimerase (PCR). Diante da confirmação de infecção por Mycobacterium, o tratamento foi instituído por meio da associação de três antimicrobianos (enrofloxacino 5mg/kg, claritromicina 10mg/kg e rifampicina 15mg/kg), por via oral, a cada 12 horas, aos quais o animal respondeu positivamente, apresentando significativa diminuição dos sinais clínicos; entretanto, após 60 dias de tratamento, foi a óbito devido a um atropelamento. Conclui-se que a realização do tratamento em gatos com tuberculose pode ocasionar melhora dos sinais clínicos e que, embora de ocorrência rara no Brasil, essa enfermidade deve ser incluída como diagnóstico diferencial das enfermidades infecciosas nessa espécie.
ABSTRACT This article aims to report a case of Mycobacterium bovis infection in a six-month-old domestic cat, emphasizing its clinical and therapeutic aspects. The feline presented as history the intake of unboiled cow's milk and later developed signs of anorexia, non-productive cough and lymphadenomegaly. Additional imaging, hematological, cytological and polymerase chain reaction (PCR) tests were performed to elucidate the diagnosis. In the presence of Mycobacterium infection, treatment was instituted by the combination of three antimicrobials (Enrofloxacin 5mg/kg, Clarithromycin 10mg/kg, and Rifampicin 15mg/kg) orally every 12 hours, to which the animal responded positively, presenting significant decrease of the clinical signs, however after 60 days of treatment the cat died accidentally, hit by a car on the street. It is concluded that the treatment in cats affected by tuberculosis can cause improvement of clinical signs and that although rare in Brazil, this disease should be included as a differential diagnosis of infectious diseases in this species.

ABSTRACT Little is known about milk fatty acid (FA) profile of cows fed diets based on sugarcane (Saccharum officinarum L.) bagasse. This study aimed to evaluate FA profile and nutritional quality indexes of milk fat of cows fed sugarcane bagasse and concentrate in the Brazilian northeast semiarid region. Multiparous Girolando cows were allocated in four diets based on different levels of sugarcane bagasse (30%, 38%, 46%, and 54% total DM) added to concentrate. Fatty acids C18:1 trans-11 (lowest value for 46% inclusion) and conjugated linoleic acid (CLA) cis-9, trans-11 (lowest value for 46% inclusion) in milk fat presented a quadratic pattern (P < 0.05). This trend was also observed for C18:1 cis-9 (P < 0.05), but at a lower magnitude. There was a reduction in concentration of C18:1 trans-10 (0.320 to 0.148 g 100 g-1 total FA) and CLA trans-10, cis-12 (0.008 to 0.004 g 100 g-1 total FA) according to the levels of sugarcane bagasse increase. The greater inclusion of bagasse and consequent reduction in the proportion of concentrate, notably corn, cause an increase in concentration of medium-chain saturated FA and a linear reduction of C18:3 n-3 and C18:2 n-6, with a consequent increase in the indexes of atherogenicity (2.82 to 3.2) and thrombogenicity (3.39 to 3.84) and a decrease in ratio of hypo/ hyper-cholesterolemic (0.44 to 0.37) and ω-6:ω-3 (12.44 to 8.80) in milk fat. Results indicate that different levels of sugarcane bagasse change the FA content in cows’ milk when bagasse is an exclusive roughage source in the diet. The greater inclusion of sugarcane bagasse reduces the quality of milk fat of dairy cows. The lowest level of sugarcane bagasse inclusion tested (30%) promotes greater milk yield and healthier milk fat

Abstract: Lutjanids are marine fish and commonly inhabit coral reefs, which have migratory habits and feed in a general way, due to these characteristics, are susceptible to infections by parasites, such as cuculanid nematodes. This work aimed to report for the first time, the occurrence of Dichelyne (Dichelyne) bonacii in Brazil, in two new hosts, Lutjanus analis and Rhomboplites aurorubens, acquired from local artisanal fishermen at the Aracaju city public fishing terminal (10 ° 54’17 “S37 ° 2’56” W), northeast coast of Brazil. Adult nematodes were collected from the intestinal tract of the hosts, clarified in Aman’s Lactophenol and were briefly described here. Three adult males and five gravid females were found, indicating that the two species of fish acted as definitive hosts. Currently, this species of parasite has been reported only from fish collected in the coast of Mexico, which makes this work the first record of this parasite in Brazilian coastal waters. The results show that probably the range of hosts and the distribution of D. (Dichelyne) bonacii is not as restricted as data pointed in the literature, being a little wider, extending also to waters that bathe the Brazilian coast.

ABSTRACT Cationic derivatives of 5,10,15-tris[4-(pyridin-4-ylsulphanyl)-2,3,5,6-tetrafluorophenyl]-corrolategallium(III)pyridine and 5,10,15-tris[4-(pyridin-2-ylsulfanyl)-2,3,5,6-tetrafluorophenyl]-correlategallium(III)pyridine were synthesized and their photosensitizing properties against the naturally bioluminescent Gram-negative bacterium Allivibrio fischeri were evaluated. The cationic corrole derivatives exhibited antibacterial activity at micromolar concentrations against this Gram-negative bacterium strain.

RESUMO Hyptis pectinata, conhecida popularmente como Sambacaitá é uma erva medicinal nativa utilizada pela população local para o tratamento de enfermidades, cujas propriedades farmacológicas, antimicrobianas e medicinais foram relatadas em diversos estudos. O óleo essencial das folhas é rico em compostos terpênicos responsáveis pelas suas propriedades. Devido à existência de fatores que podem influenciar a composição química dos óleos essenciais, o objetivo deste trabalho foi avaliar a influência do armazenamento do óleo essencial Hyptis pectinata durante 360 dias em duas temperaturas diferentes. O óleo essencial foi destilado a partir de folhas secas de plantas de sete meses de idade, utilizando o método de hidrodestilação e a composição química foi determinada usando CG-EM e CG-DIC. Os efeitos do armazenamento no óleo essencial Hyptis pectinata foram testados ao longo de um ano (0, 15, 30, 60, 90, 120, 150, 180, 240, 300 e 360 dias), utilizando duas temperaturas: ambiente (±32ºC) e freezer (-20ºC). Os compostos predominantes no óleo essencial foram o β-cariofileno, óxido de cariofileno e germancreno D e houve variação na concentração dos constituintes químicos do óleo essencial, dependendo da temperatura de armazenamento, bem como ao longo dos 360 dias. O armazenamento do óleo essencial, à temperatura ambiente ao longo de um ano resultou em maiores concentrações de β-elemeno, α-copaeno, germacreno D, óxido de cariofileno e (E, E) -α-farneseno e menores teores de α-humuleno e β-cariofileno, quando comparado com os resultados obtidos a partir de armazenamento do óleo essencial no freezer. Estes resultados indicam que o óleo essencial deve ser estocado no freezer, o que propicia maior estabilidade na concentração dos constituintes químicos.
ABSTRACT Hyptis pectinata, popularly known as “sambacaitá”, is a native medicinal herb used by local people for treating disease. Its pharmacological, antimicrobial, and medicinal properties have been reported in several studies. The essential oil of the leaves is rich in terpene compounds responsible for its properties. Because there are factors that can influence the chemical composition of essential oils, the objective of this study was to evaluate the influence of storage on Hyptis pectinata essential oil for 360 days at two different temperatures. The essential oil was distilled from dried leaves of seven-month-old plants using the hydrodistillation method, and the chemical composition was determined using GC-MS and GC/FID. The effects of storage on the essential oil Hyptis pectinata were tested over the course of a year (0, 15, 30, 60, 90, 120, 150, 180, 240, 300, and 360 days), using two temperatures: room (±32ºC) and freezer (-20ºC). The predominant compounds in the essential oil are β–caryophyllene, caryophyllene oxide, and germancrene D. The concentration of the chemical constituents of the essential oil varied depending on the storage temperature, and over the 360 days. The storage of the essential oil at room temperature over one year resulted in higher concentrations of β-elemene, α-copaene, germacrene D, caryophyllene oxide, and (E,E)-α-farnesene and lower content of α-humulene and β-caryophyllene, compared to the results obtained from storing the essential oil in a freezer. These results indicate that the essential oil should be stored in a freezer, which provides greater stability to the concentration of the chemical constituents.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Genotoxic Maillard byproducts in current phytopharmaceutical preparations of Echinodorus grandiflorus

Extratos de Echinodorus grandiflorus, obtidos a partir de folhas secas por três técnicas diferentes, foram avaliados pelo ensaio de indução lisogênica bacteriana (Induteste), em relação às propriedades genotóxicas. As amostras foram previamente esterilizadas por vapor de água (autoclavação) ou luz ultravioleta. Somente os extratos preparados por infusão e esterilizados a vapor mostraram atividade genotóxica. A análise fitoquímica revelou a presença dos flavonóides isovitexina, isoorientina, swertisina e swertiajaponina. Isovitexina e isoorientina, isoladas de uma fração genotóxica, foram testadas separadamente e não apresentaram genotoxidade. O desenvolvimento de odor adocicado e escurecimento dos extratos submetidos ao calor motivaram a investigação quanto às substâncias precursoras da reação de Maillard no extrato. Ácidos aminados e açúcares redutores foram identificados e a presença de precursores da reação de Maillard no extrato foi confirmada por espectrofotometria no visível com a adição do inibidor bissulfito de sódio antes do aquecimento. Este é o primeiro relato de aparecimento de melanoidinas em uma técnica que pode ser usada para a preparação de fitoterápico. A atividade genotóxica de tais infusões preparadas ao calor implica em algum risco de desenvolvimento de doenças degenerativas com seu uso descontrolado e por longo prazo.
Extracts of Echinodorus grandiflorus obtained from dried leaves by three different techniques were evaluated by bacterial lysogenic induction assay (Inductest) in relation to their genotoxic properties. Before being added to test cultures, extracts were sterilized either by steam sterilization or ultraviolet light. Only the extracts prepared by infusion and steam sterilized have shown genotoxic activity. The phytochemical analysis revealed the presence of the flavonoids isovitexin, isoorientin, swertisin and swertiajaponin, isolated from a genotoxic fraction. They were assayed separately and tested negative in the Inductest protocol. The development of browning color and sweet smell in extracts submitted to heat, prompted further chemical analysis in search for Maillard's reaction precursors. Several aminoacids and reducing sugars were cast in the extract. The presence of characteristic Maillard's melanoidins products was determined by spectrophotometry in the visible region and the inhibition of this reaction was observed when its characteristic inhibitor, sodium bisulfite, was added prior to heating. Remarkably, this is the first paper reporting on the appearance of such compounds in a phytomedicine preparation under a current phytopharmaceutical procedure. The genotoxic activity of such heat-prepared infusions imply in some risk of developing degenerative diseases for patients in long-term, uncontrolled use of such phytomedicines.

A artrite-encefalite caprina (CAE) é diagnosticada rotineiramente pela técnica de imunodifusão em gel de agarose (IDGA), que é considerada pouco sensível. Objetivou-se com este estudo padronizar testes de Elisa-i e Western Blot (WB) para diagnóstico precoce de anticorpos em caprinos contra CAEV e comparar os resultados obtidos nesses testes com a prova de IDGA. Para a padronização dos testes Elisa-i e WB, utilizaram-se diferentes concentrações e diluições de antígeno, soros e conjugado. No Elisa-i, adotaram-se microplacas rígidas com 96 poços, sendo a combinação de concentração de 0,5µg/poço de antígeno e diluições de 1:100 de soro e 1:1500 de conjugado a que apresentou melhor resultado. No WB foram utilizadas membranas de nitrocelulose, definindo-se as diluições de 1:50 de soro e 1:15000 de conjugado. Para avaliar o desempenho das técnicas, 222 amostras de soro caprino foram testadas e os dados obtidos foram comparados com o IDGA. A sensibilidade e a especificidade do Elisa-i/IDGA, WB/IDGA e WB/Elisa-i foram de 70% e 91%, 100% e 72,6%, 84,6% e 76,5%, concomitantemente. O índice Kappa desses testes foi de 0,35, 0,2 e 0,36, respectivamente. As técnicas de Elisa-i e WB apresentaram-se mais sensíveis que a IDGA, podendo ser utilizadas como ferramentas para o diagnóstico precoce da CAE.
Caprine arthritis-encephalitis (CAE) is routinely diagnosed with the Agarose Gel Immunodiffusion (AGID) technique, which is considered to have low sensitivity. The objective of this study was to standardize testing i-Elisa and Western Blot for early detection of antibodies against CAEV in goats and compare the results obtained in these tests with proof of AGID. For standardization of i-Elisa and WB, different concentrations and dilutions of antigen, sera and conjugate were used. In the i-Elisa, rigid microplate with 96 wells was adopted, and the combination that showed the best result was a concentration of 0.5µg/ well of antigen and dilutions of the serum of 1:100 and conjugate of 1:1500. In the WB nitrocellulose membranes were used, and the dilutions of the serum were defined at 1:50 and conjugate at 1:15000. To evaluate the performance of the techniques, 222 goat serum samples were tested and the data were compared with the AGID. The sensitivity and specificity of Elisa-i/IDGA, WB/AGID and WB/Elisa-i were 70% and 91%, 100% and 72.6%, 84.6% and 76.5%, concomitantly. The Kappa index of these tests was 0.35, 0.2 and 0.36, respectively. The i-Elisa and WB techniques were more sensitive than the AGID and can be used as tools for early diagnosis of CAE.

Objetivou-se estudar a influência da artrite encefalite caprina no hemograma de cabras durante a lactação. Para tanto, 38 matrizes F1 Anglo-Nubiana x Saanen, com idade entre 14 e 38 meses e escore corporal entre dois e três, foram previamente testadas por IDGA e Western Blot para a divisão em grupo soropositivo (n = 19) e soronegativo (n = 19). Os grupos foram mantidos em piquetes distintos, sem contato físico, recebendo o mesmo manejo nutricional e sanitário. O sangue foi coletado por venipunção da jugular, mensalmente, desde o parto até o fim da lactação. Foram determinados os valores do eritrograma e do leucograma. Em dados momentos, os soropositivos apresentaram alterações morfológicas nas hemácias, com diminuição significativa do VCM, aumento significativo do HCM e CHCM. Não houve diferença significativa entre grupos na contagem de hemácias e volume globular. Os valores absolutos de monócitos e eosinófilos dos soropositivos, em algumas ocasiões, encontraram-se significativamente superiores aos dos soronegativos e acima da faixa de normalidade para a espécie caprina.
The objective was to study the influence of caprine arthritis encephalitis in the blood count of goats during lactation. To this end, 38 F1 Anglo-Nubian x Saanen reproductive female goats, aged 14 to 38 months and body condition score between 2 and 3, were first tested by AGID and Western Blot, and divided into either a seropositive group (n = 19) or a seronegative group (n = 19). The groups were kept in separate paddocks, without physical contact, receiving the same nutritional and health management. The blood of these animals was collected by jugular venipuncture, monthly, from the partum period until the end of lactation, and their erythrogram and leukogram values were determined. At given moments, the seropositive animals showed morphological changes in red blood cells, with significant decrease in MCV, coupled with a significant increase of MCH and MCHC. There was no significant difference between the groups in the values of red blood cells and packed cell volume. The absolute values of monocytes and eosinophils of the seropositive animals, at certain moments, were significantly higher than the seronegative ones and above the normal range for goats.
