ABSTRACT This study aimed to identify candidate genes regulating ovarian development following ovarian tissue transplantation through transcriptome sequencing (RNA-seq) and weighted gene co-expression network analysis (WGCNA). Ovarian tissues were collected from 10 thirty-day-old donor chickens, and each ovary was divided equally into three parts of similar size, and transplanted in situ into two-day-old recipient chickens. Samples were collected on days 0 (untransplanted), 6, and 12 after transplantation. RNA was extracted from ovarian samples for construction of a transcriptome library, and then sequenced on the IlluminaHiSeqTM2000 sequencing platform. The sequencing results were evaluated to determine differentially expressed genes (DEGs) through GO function and the KEGG pathway analyses, Gene Set Enrichment Analysis (GSEA), WGCNA, and PPI networks. Some candidate genes were further validated through real-time fluorescence quantitative PCR (RT-qPCR). RNA-seq analysis identified 2242 up-regulated and 3095 down-regulated DEGs at 6 days after ovarian transplantation, and 2181 up-regulated and 2129 down-regulated DEGs were identified at 12 days after transplantation. Enrichment analysis showed that the genes were enriched in multiple inflammatory pathways, pathways involved in signal transduction and cellular processes. Through the WGCNA, five modules were constructed, from which 4 and 5 candidate genes were mined at 6 and 12 days after ovarian transplantation, respectively. Transcription factor prediction showed that GTF3A was the most important transcription factor. The results of RT-qPCR verification confirmed that the expression profile of 8 candidate genes was consistent with that of the sequencing results. In this work, we adopt the three-order perturbation formulae for g-factors (g‖, g⏊) of d1 ions in the octahedral environment to calculate the g-factors of W5+ ions in tungsten phosphate glasses containing lithium (P2O5-Li2W4O-Li2O). In the light of the high valence state of the studied W5+ centers and hence the strong covalency of the studied octahedral [WO6]7- cluster, we consider the contributions to g-factors from the ligand orbital and spin-orbit (SO) coupling interactions based on the cluster approach. The required tetragonal crystal-field parameters are calculated from the local structure of W5+ ions based on the superposition model. According to the theoretical calculations, we find that the octahedral [WO6]7- clusters possess the tetragonally compressed distortion with a shorter W-O bond length (≈ 1.54 Å) and a longer one (≈ 2.26 Å) along C4 axis and four normal W-O bond length (≈ 1.94 Å) in the perpendicular plane, which infers that the W5+ ions are in the form of tungstyl ions (i.e., WO3+). Based on the local structural data, the theoretical values of g‖ and g⏊ agree well with the experimental values.

RESUMO Infecção com o vírus Hepatite E (HEV) é uma importante questão de saúde pública global. Infecções HEV são reconhecidas como doença zoológica. Acredita-se que suínos são o principal reservatório de HEV. Recentemente iaques, vacas, e gado amarelo foram reportados como novos reservatórios do HEV. Porém, não se sabe se outras espécies de gado e búfalo são sensíveis a infecção HEV. Para investigar a prevalência de infecção HEV em búfalos, foram realizados prova de imunoabsorção enzimática e polimerização em cadeia inversa ancorada em transcrição. Apenas um búfalo foi positivo para o anticorpo anti-HEV IgM (1/106, 0,94%), e nenhum foi positivo para o anticorpo anti-HEV IgG. Para nossa surpresa cinco (5/106, 4,72%) e três amostras de leite (3/40, 7,50%) de búfalos foram positivos para HEV RNA. Todas as estirpes de HEV isoladas de búfalos pertencem ao genótipo 4. Resultados indicam que búfalos podem ser um reservatório de HEV.

ABSTRACT Hepatitis E virus (HEV) infection is an important global public health issue. HEV infections are recognized as a zoonotic disease. Swine are believed to be the main reservoir of HEV. Recently, yaks, cows, and yellow cattle have been reported as new reservoirs of HEV. However, whether other species of cattle and buffaloes are sensitive to HEV infection is unknown. To investigate the prevalence of HEV infection in buffaloes, enzyme-linked immunosorbent assay (ELISA) and reverse transcription-nested polymerase chain reaction (RT-nPCR) were performed. Only one buffalo was positive to anti-HEV IgM antibody (1/106, 0.94%), and none were positive for anti-HEV IgG antibody. To our surprise, five serum (5/106, 4.72%) and three milk samples (3/40, 7.50%) from buffaloes were positive to HEV RNA. All strains of HEV isolated from buffaloes belong to genotype 4. Results indicate that buffaloes may be a new reservoir of HEV.

ABSTRACT To investigate the hypothesis that APS can attenuate E. coli-induced microvascular dysfunction in chicken intestine, 60 0-day old male chickens were divided into three groups with 5 replications of 4 chicks. Chicken in the APS group were treated with 15 mg APS daily while the others were given 0 mg APS for 6 days. Then all 7-day old chicken were injected intraperitoneally by E. coli, except for the chicken in the control group. After 4 h, all chicken’s intestinal samples were collected to detect gene expressions involved in inflammatory factors and adhesion molecules. Results showed that APS attenuated the signs of edema and hemorrhage in 7-day old chicken intestinal mucosa which were induced by E. coli injection. Consistently, APS significantly reduced the increasing mRNA levels of inflammatory factors such as Tumor necrosis factor-a (TNF-a), interleukin (IL) -1β and inducible nitric oxide synthase (iNOS) (p<0.05), the same results were observed in vascular adhesion molecules such as E-selectin, intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1). Moreover, we observed that APS supplementation in water suppressed significantly (p<0.05) the decline of reparative factors such as epithelial growth factor (EGF) and basic fibroblast growth factor (bFGF) in E. coli injected group. Furthermore, supplementation with APS substantially blocked (p<0.05) the increase in Toll-like receptor-4 (TLR4) and Nuclear factor (NF)-κB mRNA abundance (p=0.087) induced by E. coli infection. This study indicated that microvascular injured chicken intestine induced by E. coli would be attenuated with feeding APS, and the mechanism of repairing were probably mediated through TLR4-NF-κB signal pathways.

ABSTRACT Heat stress induces oxidative stress, and reduces body antioxidant metabolite levels, which can affect poultry production performance. Dietary antioxidants protect birds against the adverse effects of heat stress. The effects of increasing concentrations of dietary curcumin on the antioxidant parameters of layers maintained under high-temperature conditions for nine weeks were evaluated. Roman laying hens (n = 336, 22 weeks old, 1420 g BW) were divided into three treatment groups. The first group served as a thermoneutral control (kept at 25 ± 1 °C). The second group was exposed to high temperatures (32 ± 1 °C, 6 h/d), given a basal diet. The third group was further divided into five treatment groups (100, 150, 200, 250, 300 mg/kg Curcumin) fed a basal diet (treatments H1, H2, H3, H4, H5) under high temperatures conditions (32 ± 1 °C, 6 hours/day). As a result of this study, total superoxide dismutase activity was significantly higher in H2 and H3 groups, and total antioxidant capacity was higher in H2, H3, and H5 groups. Catalase and glutathione peroxidase activity was significantly higher in the H3 group. Malondialdehyde concentration was lowered in curcumin supplemented hens compared with control groups hens. Laying hens in all curcumin treatment groups had slightly higher activities of CAT, SOD, GSH-Px, and T-AOC in the liver, heart, and lungs, compared with heat stressed control group. It was concluded that dietary curcumin given to laying hens under heat stress may enhance their antioxidant status, and alleviate the detrimental effects of stressful environmental conditions.

ABSTRACT The effects of oxidative stress induced by high temperature on the cell viability, proliferation, apoptosis and oxidative status of chicken embryonic fibroblasts (CEF) were analyzed. The viability, proliferation, apoptotic and anti-oxidative status were measured after incubating CEF at the temperatures of 37ºC (control) and 40-44ºC (experimental groups) for 6,12 and 24 hours. The results showed that at high temperature (42-43ºC), the viability of CEF cells decreased after 6, 12 and 24 h of incubation, but the difference was significant only at 43ºC. Cell proliferation was significantly reduced at 44oC/6h. The apoptotic rate of CEF cells was increased following heat treatments in a time-dependent manner. ROS formation increased with increasing temperature, but the difference was only significant at 44ºC/6,12h. Heat stress did not significantly affect the superoxide dismutase (SOD) activity. CAT activity was significantly decreased at 43ºC/24h and 44ºC/12 and 24h. Malondialdehyde (MDA) formation was significantly increased at 43ºC/12h and 44ºC/12 and 24h. In conclusion, heat stress induced the oxidative stress, decreasing the viability, proliferation and anti-oxidative response of CEF cells.

The majority of goats in Tanzania belong to the Small East African (SEA) breed, which exhibits large phenotypic variation. This study aimed to determine the genetic structure of, and relationships among four populations (Sukuma, Gogo, Sonjo, and Pare) of the SEA breed that have not been studied adequately. A total of 120 individuals (24 from each population) were analysed at eight microsatellite loci. In addition, 24 goats of the South African Boer breed were used as reference. Observed heterozygosity (Ho) ranged from 0.583 ± 0.04 for Sukuma to 0.659 ± 0.030 for Gogo, while expected heterozygosity (He) ranged from 0.632 ± 0.16 for Sukuma to 0.716 ± 0.16 for Boer. Five loci deviated from Hardy-Weinberg equilibrium (HWE) across populations. The mean number of alleles ranged from 4.75 ± 1.58 for Pare to 6.88 ± 3.00 for Sukuma. The mean inbreeding coefficient (F IS) ranged from 0.003 in Sonjo to 0.148 in Sukuma. The differentiation coefficient (F ST)was highest (0.085) between Boer and Sukuma and lowest (0.008) between Gogo and Sonjo. The largest genetic distance (0.456) was found between Sukuma and Boer, while the smallest (0.031) was between Gogo and Sonjo populations. Pare, Gogo, and Sonjo populations, formed one cluster, while Sukuma and Boer populations formed two separate clusters. From the findings, it can be concluded that the SEA goats in this study showed high in population genetic variation, which implies that there is good scope for their further improvement through selection within populations. The Sukuma population, which has fairly high inbreeding, is moderately differentiated from Pare, Sonjo, and Gogo goat populations, which showed a high level of admixture. Conservation and improvement strategies of the goats should be designed with first priority being on Sukuma goats.

The effect of particle size on the flotation kinetics of a low-rank coal was compared using air bubbles and oily bubbles. Five kinetic models were applied to the data from the tests to derive the relationships between the flotation rate constant, the ultimate recovery, and the particle size fraction. The results show that oily bubble flotation (OBF) resulted in a lower ash content and higher combustible recovery than air bubble flotation (ABF), especially for the -125 μm size fraction. It was found that the flotation of a low-rank coal in individual size fractions using air bubbles and oily bubbles can be best described by a first-order model with rectangular distribution of tendency to float and the classical first-order model respectively. OBF yielded higher ultimate combustible and ultimate ash recoveries. The flotation rate constants for OBF were higher in all the models except for the fully mixed reactor model. The ultimate recovery values and modified rate constants first increased and then decreased as the size fraction decreased. The trend of the flotation rate constants as a function of the size fraction was similar in all the models except for the fully mixed reactor model.

This study aimed to investigate the efficacy of minimally invasive tonsil surgery for the treatment of obstructive sleep apnea-hypopnea syndrome (OSAHS) in children. Tonsil ablation or turbinate reduction was performed on 49 pediatric patients with OSAHS by minimally invasive tonsil surgery. In order to evaluate the efficacy of surgery, a comparison was conducted between pre-operation and post-operation data in terms of the symptoms, signs and polysomnography test. Total effectiveness rate of the surgery was 83.7%. Subgroup analysis was also performed based on the severity of their conditions: mild, moderate, and severe groups had an effectiveness rate of 90.0, 88.9, and 66.7%, respectively (Hc=6.665, P<0.05). Postoperatively, the apnea-hypopnea index, the minimum oxygen saturation (SaO2), and corresponding symptoms improved compared to pre-operation conditions (P<0.05). Minimally invasive tonsil surgery was a safe and effective method for treating OSAHS in children.

Abstract: The local structure of the Cu2+ center in mixed alkali cadmium phosphate (10Li2O-10Na2O-20CdO-59.5P2O5) glass is theoretically investigated from the optical spectral data and high-order perturbation formulas of the electron paramagnetic resonance (EPR) parameters (g-factors g║,g┴ and the hyperfine structure constants A║,A┴ ) for a 3d9 ion in a tetragonally elongated octahedron. In these formulas, the ligand orbital and spin-orbit coupling contributions are taken into account based on the cluster approach in view of moderate covalency. The required crystal-field parameters are determined by the superposition model. According to the calculations, the impurity-ligand distances parallel and perpendicular to the C4 axis of [CuO6] 10- cluster are found to be, respectively, R ║ ≈2.051 Å and R ┴ ≈ 2.029 Å due to Jahn-Teller effect. Based on the above local structure data, the optical spectra are satisfactorily interpreted. The signs of A║ and A┴ for the Cu 2+ center are suggested in the discussion.

High plasma levels of homocysteine (Hcy) promote the progression of neurodegenerative diseases. However, the mechanism by which Hcy mediates neurotoxicity has not been elucidated. We observed that upon incubation with Hcy, the viability of a neuroblastoma cell line Neuro2a declined in a dose-dependent manner, and apoptosis was induced within 48 h. The median effective concentration (EC50) of Hcy was approximately 5 mM. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) nuclear translocation and acylation has been implicated in the regulation of apoptosis. We found that nuclear translocation and acetylation of GAPDH increased in the presence of 5 mM Hcy and that higher levels of acetyltransferase p300/CBP were detected in Neuro2a cells. These findings implicate the involvement of GAPDH in the mechanism whereby Hcy induces apoptosis in neurons. This study highlights a potentially important pathway in neurodegenerative disorders, and a novel target pathway for neuroprotective therapy.

Dada la creciente demanda en el fracturamiento de formación de baja permeabilidad, la interpretación matemática de datos de pozos se ha tornado importante para la generación de modelos matemáticos que estudien el comportamiento de la presión y se formulen metodologías para una caracterización más exacta de formaciones apretadas que contienen hidrocarburos. Por ende, en este artículo se presenta una metodología para interpretar pruebas de presión en yacimientos gasíferos bi-zonales drenados por un pozo vertical hidráulicamente fracturado (penetración parcial o complete) que utiliza el gráfico log-log de pseudopresión y derivada de pseudopresión. La metodología dada usa líneas y puntos característicos hallados en dicho gráfico de modo que se desarrollaron nuevas expresiones y correlaciones para longitud media de fractura, relación de penetración de la fractura, relaciones de movilidad y almacenaje, permeabilidad radial, tamaño de la zona interior, área de drenaje, permeabilidad vertical y factores de daño. Las expresiones nuevas se verificaron exitosamente mediante ejemplos sintéticos.

Due to the recent increase in the fracturing of low permeability formations, mathematical wellbore data interpretation has become important to generate mathematical models to study their pressure behavior and formulate interpretation methodologies for a more accurate characterization of tight hydrocarbon-bearing formations. This paper presents an analytical methodology of interpretation using the pseudopressure and pseudopressure derivative log-log plot for the characterization of hydraulically fractured (partially or fully penetrating) vertical wells completed in bi-zonal gas reservoirs. The methodology provided uses characteristic points and lines found on the pseudopressure and pseudopressure derivative plot so that new analytical expressions and correlations are developed to estimate half-fracture length, fracture penetration ratio, mobility and storativity ratios, radial permeability, size of the inner zone, well drainage area, vertical permeability and skin factor. The new expressions were successfully verified and tested with synthetic examples.

Understanding soil evaporation in reforestation processes in an area of extreme drought is important. We examined the effects of drip-irrigated, high-salinity groundwater on the formation of soil salt crusts and subsequent soil evaporation in the Taklimakan Desert Highway Shelterbelt. Soil evaporation was measured using micro-lysimeters (MLS, 20 cm in length x 10 cm in diameter), and chemical characteristics, including SOM, total salts, ion composition and pH, were analyzed. The results showed that the inhibition efficiency of salt crusts on soil evaporation decreased from the surface to the lower soil layers. Following irrigation, the inhibition efficiency was 33.0% in the shelterbelt, which was much higher than the 13.8% observed for the bare soil. Total salt content and ion contents (Cl¾, Na+, and SO4²>) were much higher for the soil salt crusts than for shifting sandy soils, while the contents of other ions (Mg²+, K+, Ca²+, HCO3>, and CO3²>) were very similar. The total salt content and ion composition of the salt crusts increased during the first 2 years of shelterbelt age, and decreased from 2 to 5 years. With the increasing age of the shelterbelts, the SOM of the salt crusts increased, whereas the total salts and pH decreased. We concluded that the formation of soil salt crusts results in reduced soil evaporation and the soil chemical condition functioned better for growth of the Taklimakan Desert Highway Shelterbelt.

Current therapy for pancreatic cancer is multimodal, involving surgery and chemotherapy. However, development of pancreatic cancer therapies requires a thorough evaluation of drug efficacy in vitro before animal testing and subsequent clinical trials. Compared to two-dimensional culture of cell monolayer, three-dimensional (3-D) models more closely mimic native tissues, since the tumor microenvironment established in 3-D models often plays a significant role in cancer progression and cellular responses to the drugs. Accumulating evidence has highlighted the benefits of 3-D in vitro models of various cancers. In the present study, we have developed a spheroid-based, 3-D culture of pancreatic cancer cell lines MIAPaCa-2 and PANC-1 for pancreatic drug testing, using the acid phosphatase assay. Drug efficacy testing showed that spheroids had much higher drug resistance than monolayers. This model, which is characteristically reproducible and easy and offers rapid handling, is the preferred choice for filling the gap between monolayer cell cultures and in vivo models in the process of drug development and testing for pancreatic cancer.

The separation performance of a raw coal sample from South Africa based on the dense gas-solid fluidized bed beneficiation technique was investigated. Analysis showed that the raw coal sample distributes mainly in the -50+6 mm size range with the -1.6 g/cm³ density fraction accounting for 78.20% of the cumulative mass distribution. A flow sheet of the sequential coal beneficiation technique was introduced. The experimental and simulation results of the hydromechanical and density distribution stability indicate that the dense gas-solid fluidized bed could provide a stable separation environment with uniform density fluctuations under suitable operating conditions. With the application of this technique on the raw coal sample, the separation results show that three products, consisting of 73.25% clean coal with an ash content of 9.59%, 11.31% middlings, and 15.44% gangue were effectively separated at two different densities of 1.82 g/cm³ and 1.57 g/cm³. This study provides an effective flow sheet for achieving the high-efficiency separation performance on raw coal from arid areas.
