The coronavirus disease (COVID-19) pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has had a profound impact on global health and socio-economic conditions. To date, various vaccines have been administered worldwide in an effort to curb the spread of the virus. Despite vaccination efforts, there have been complications. Existing antiviral drugs have shown limited effectiveness, prompting the use of computational methods to understand the dynamics of the virus and develop suitable treatments. The current study focuses on using biocompatible para-sulfonato-calix[4]arenes to dock against two key proteins of SARS-CoV-2, namely ribonucleic acid (RNA)-dependent RNA polymerase, and helicase. Docking results indicate a strong binding affinity of these compounds to the target proteins, with higher scores compared to commonly used medications. Molecular dynamics (MD) simulation validates the docking results, showing stable protein-ligand complexes over time. The compounds are also screened for absorption, distribution, metabolism, and excretion properties and toxicity, suggesting their potential as lead candidates for inhibiting the virus’s key proteins. However, further in vivo and in vitro studies are recommended to confirm these findings. Abstract The quality and quantity of the extracted DNA are two key aspects for a successful PCR (Polymerase Chain Reaction) amplification. Moreover, reduction in time and cost required for DNA extraction are also two considerable factors, in cases when large number of samples are to be analyzed within a limited time-frame and budget. Accordingly, the aim of this study was to compare and optimize performance of five different DNA extraction methods by boiling meat tissues from cattle, buffalo, sheep, goat, chicken, camel, horse and dog in PBS (Phosphate Buffer Saline), distilled water, alkaline lysis buffers 1, 2 or 3. The results indicated that the boiling of meat and its products in alkaline lysis buffers was a good method to extract crude DNA. The optimized crude DNA extraction protocol was coupled with PCR-RFLP (Restriction Fragment Length Polymorphism) analysis for meat species differentiation. This developed workflow was tested on fifty-three commercial beef and mutton samples, out of which three samples were found to be adulterated. In conclusion, the rapid crude DNA extraction protocol has led to the development of a direct PCR-RFLP workflow that is simple, time-saving and cost-effective for PCR-based identification of different meat species. Abstract Background: Plant-derived compounds are alternatives of synthetic insecticides in sustainable agriculture. Objective: This study investigated the phytotoxic effect of higher concentrations (2, 4, 8 and 16%) of four plants extracts (Azadirachta indica, Mentha arvensis, D. stramonium and Citrus limonium) on cotton plants. Methods: Each concentration was replicated four times to check the phytotoxic effect (CO2-in, CO2-out, H2O-in, H2O-out and photosynthesis absorption rate (PAR) in randomized complete block design. Data was recorded after 12, 24, 48 and 72 hours of spray with the help of Photosynthetic CL 340 meter. Results: The results showed that CO2-in was more affected by the D. stramonium (131.65±0.38) at 8% concentration. The overall progress showed that C. limonium was more affected the CO2-in of cotton crop. CO2-out was less affected by the C. limonium (117.83±1.46) at 4% concentration than M. arvensis (116.99±1.25) at 8% concentration and D. stramonium (115.77±0.74) at 16% concentration, but was more affected by the A. indica (118.15±0.71) at 4%. H2O-in was more affected by the C. limonium (0.39±0.05) than D. stramonium, A. indica and M. arvensis at 16% concentration. H2O-out of cotton was least affected by the D. stramonium (7.63±0.01) at 2% and more affected by the C. limonium (1.56±0.15) at 16% concentration. PAR was more affected by the A. indica (931.47±8.39) at 4% concentration and least affected by the M. arvensis (1499.7±9.94) at 8% concentration. Conclusions: Different dosages of various botanicals influenced the opening and closing of stomata and photosynthesis of cotton plants.

ABSTRACT BACKGROUND: The prevalence of Giardia lamblia in Pakistani children is currently unknown. The aim here was to evaluate the prevalence and risk factors of Giardia lamblia in children exhibiting diarrhea. DESIGN AND SETTING: Cross-sectional study at different district healthcare hospitals in Pakistan. METHODS: A total of 800 samples were collected from children aged 0-10 years. Information regarding personal data, demographic data and supposed risk factors was collected through a structured questionnaire. Giardia lamblia was detected through direct microscopy and antigens through the enzyme-linked immunosorbent assay (ELISA). RESULTS: The prevalence of Giardia lamblia was 2.75% through direct microscopy and inflated to 9.5% through ELISA. The demographic factors positively associated with occurrences of giardiasis were age (P = 0.035; odds ratio, OR = 1.96; 95% confidence interval, CI = 1.094-3.533), mother’s educational level (P = 0.031; OR = 2.67; 95% CI = 1.186-6.045) and father’s educational level (P = 0.004; OR = 3.56; 95% CI = 1.612-7.899). Similarly, among the supposed risk factors, rural residency (P = 0.032; OR = 1.76; 95% CI = 1.098- 2.851), absence of proper sewerage system (P = 0.000; OR = 6.60; 95% CI = 4.029-10.841) and unavailability of safe drinking water (P = 0.000; OR = 4.08; 95% CI = 2.207-7.547) were the factors strongly connected with giardiasis. Abdominal discomfort was a prominent clinical sign with 46% frequency. CONCLUSION: Various risk factors were associated with occurrences of Giardia, thus emphasizing the importance of parents’ education, safe drinking water and proper sewerage systems for Pakistani children’s health.

ABSTRACT: In this study an Iron oxide/carbon nanocomposite from maize straw was prepared and was characterized by XRD, SEM, EDX, FTIR, TG/DTA and Surface area analyzer. The adsorbent was fed to different groups of poultry birds along with aflatoxin B1. Different physiological and blood parameters were monitored in order to study the efficacy of the prepared adsorbent for binding of aflatoxin B1 in the gastrointestinal tract of chickens. It was found that adsorbent at dose of 0.3%/ kg feed was highly effective in detoxifying aflatoxin B1 in gastrointestinal tract of poultry birdswith no harmful effects. The high doses given to groups E and F; 0.4% and 0.5% respectively showed slight variation in tested parameters from group A. No negative symptoms associated with the use of activated carbon as previously reported were observed for the adsorbent under study.
