Abstract The yerba-mate industry is one of the most important economic activities in Misiones, a province in the northeast of Argentina that is the world’s leading producer and exporter of this crop. White thread blight disease caused by Ceratobasidium niltonzousanum affects the cultivation reducing its quality and productivity. Due to the lack of a standardized visual method to quantify the severity of this disease in yerba mate, a diagrammatic scale was developed and validated. Yerba-mate branches were collected in a field in the north of Misiones province, and the actual severity was determined digitally. A six-level scale was developed using the DOSLOG software, based on the Weber-Fechner law. The validation was carried out by twenty raters. One evaluation without the diagrammatic scale and two evaluations with the scale were carried out in 14-day intervals. Accuracy, precision, and reproducibility of the scale were evaluated through linear regressions and correlation analysis, obtaining R2 values ranged between 0.70 and 0.94. Using the diagrammatic scale developed in this work, raters enhanced the accuracy and precision of the estimates, and the repeatability of the scale improved by 94.74%. The scale was appropriate to assess the damage of white thread blight in yerba mate.

Abstract Cassava cultivation in the Misiones province largely contributes to the national production. Part of it is used to obtain starch, which generates industrial effluents with a high load of organic matter and cyanogenic compounds. This constitutes a problem at the environmental level and bioremediation is presented as an alternative to reduce this waste, since it is based on the degradation of the pollutant using microorganisms present in the contaminated matrix. The objective of this work was to isolate microorganisms with starch degradation capacity and tolerance to cyanide from effluents of the starch industry. We isolated 93 bacterial strains and 15 were preselected by qualitative determination of amylolytic activity. Using the 16S rDNA gene for molecular identification, it was determined that some of these isolates belong to the genera Pseudomonas, Acinetobacter and Proteus. These strains were cultivated in minimal medium with different cyanide concentrations and it was possible to determine that one of them has amylolytic activity and tolerance to cyanide, both desirable characteristics for bioremediation of effluents from the cassava starch industry.

Resumen El cultivo de mandioca en la Provincia de Misiones aporta la mayor parte de la producción a nivel nacional. Parte de la misma es destinada a la obtención de fécula, actividad que genera efluentes industriales con una elevada carga de materia orgánica y compuestos cianogénicos. Esto constituye una problemática a nivel ambiental y la biorremediación se presenta como una alternativa para la reducción de estos residuos, ya que se basa en la degradación del contaminante utilizando los microorganismos presentes en la matriz contaminada. El objetivo de este trabajo fue aislar microorganismos con capacidad para degradar almidón y tolerantes a cianuro a partir de efluentes de una industria almidonera. Fueron aisladas93 cepas bacterianas y 15 de ellas fueron preseleccionadas mediante determinación cualitativa de actividad amilolítica. Utilizando el gen 16S rDNA para la identificación molecular, se determinó que algunos de estos aislamientos pertenecen a los géneros Pseudomonas, Acinetobactery Proteus. Esas cepas se cultivaron en medio mínimo con distintas concentraciones cianuro y se logró determinar que una de ellasposee tanto la actividad amilolítica como la tolerancia a cianuro, ambas características deseables para biorremediación de efluentes de la industria almidonera.

The cultivation of Ilex paraguariensis is important in northern Argentina, mainly in Misiones province. The presence of different symptoms of diseases caused by fungi has recently been observed. Neither the causative agents nor the epidemiological or economic damages have been identified yet. In this study, fungi were isolated from yerba mate leaves that showed visible symptoms of foliar lesions. Five isolated samples: Alternaría, Colletotrichum, Phoma and Pilidium; which were selected because of both, their micro and macro morphological characteristics. These fungi were identified by using an optical microscope together with morphological keys. Pathogenicity tests were carried out on yerba mate leaves and on one-year-old plants. The results showed that Colletotrichum sp. LBM 183 and Phoma sp. LBM 207 are pathogens typical of the yerba mate leaves capable of developing within the plant tissue, while Alternaría sp. LBM 186 and Pilidium sp. LBM 208 were able to sicken the plant by entering into the yerba mate root.

El cultivo de Ilexparaguariensis es importante en el norte de Argentina, principalmente en la provincia de Misiones. En los últimos años se ha observado la presencia de diferentes síntomas de enfermedades causadas por hongos de los cuales no se conoce agente causal, epidemiología y daño económico. En este estudio, se aislaron hongos de hojas de yerba mate que presentaban síntomas visibles de lesiones foliares. Cinco aislados (Alternaría, Colletotrichum, Phoma y Pilidium) fueron seleccionados por sus características micro y macromorfológicas. Estos hongos fueron identificados utilizando un microscopio óptico y claves morfológicas. Se realizaron ensayos de patogenicidad en hojas y en plantas de un año de yerba mate. Los resultados mostraron que Colletotrichum sp. LBM 183 y Phoma sp. LBM 207 son patógenos de hoja de yerba mate capaces de desarrollarse en el tejido. Mientras que Alternaría sp. LBM 186 y Pilidium sp. LBM 208 fueron capaces de enfermar la planta ingresando por la raíz de yerba mate.

The methodologies applied in agricultural and agroforestry systems management have great impacts on food production and biodiversity conservation. Diseases caused by phytopathogenic fungi are one of the major problems in the agriculture of Argentina. The need to find mechanisms that increase the productivity of agricultural and agroforestry systems has driven the search for less risky and more environmentally friendly control strategies of agricultural diseases. Trichoderma fungus is one of the most commonly used agents in biological control programs as a phytopathogenic fungi regulator. This review aims to compile all the available information regarding the state of art of Trichoderma in Argentina as a biological control agent, in order to reference different institutions of the country that focus their research on this fungus. Nearly 70 groups, representing universities and research institutes, have described approximately ten different species of Trichoderma with antagonistic capacity associated to 35 crops and 40 pathogens. The most used Trichoderma species as a biological control agent is Trichoderma harzianum, followed by Trichoderma koningii and Trichderma viride. Currently, there are around 14 bioproducts in the argentine market whose active principles are strains of Trichoderma. The growing initiatives of the national institutions on these studies are crucial for the development of innovative technologies to achieve a more productive and environmentally sustainable agriculture for the country.

Las metodologías que se aplican para manejar los sistemas agrícolas y agroforestales tienen efectos notables en la producción de alimentos y en la conservación de la biodiversidad. Las enfermedades que causan los hongos fitopatógenos son uno de los problemas críticos en la agricultura argentina. A partir de la necesidad de encontrar mecanismos que eleven la productividad del campo, en los últimos años se intensificó la búsqueda de estrategias de control de enfermedades agrícolas que sean alternativas al control químico, buscando, además, disminuir los riesgos ambientales y sanitarios derivados de éste. El hongo Trichoderma es uno de los agentes que más se usan en los programas de control biológico como reguladores de hongos fitopatógenos. Esta revisión tiene como objetivo realizar una compilación de toda la información disponible respecto al estado del arte de Trichoderma como controlador biológico en la Argentina, con el fin de referenciar diferentes instituciones del país que centralizan a este hongo en sus investigaciones. Cerca de 70 grupos de universidades e institutos de investigación describieron aproximadamente diez especies diferentes de Trichoderma con capacidad antagonista, se estudiaron alrededor de 35 cultivos y 40 patógenos. La especie de Trichoderma más utilizada como agente de control biológico es Trichoderma harzianum, seguida por Trichoderma koningii y Trichoderma viride. Actualmente, en el mercado argentino existen cerca de 14 bioproductos cuyo principio activo son cepas de Trichoderma. Las crecientes iniciativas de las instituciones nacionales sobre estos estudios son básicas para el desarrollo de tecnologías innovadoras y para lograr una agricultura más productiva y ambientalmente sostenible en el país.

Abstract Yerba mate (Ilex paraguariensis St. Hil.) is a species native to the subtropical regions of South America. Despite being an important crop for the region, there are few studies on the use of microorganisms to improve the growth of seedlings in the nursery stage. The objective of this study was to isolate spore-forming endophytic bacteria with plant growth promoting properties associated with yerba mate seedlings and determine their phytobeneficial effect under controlled laboratory conditions. Isolates were selected based on their sporulation capacity and evaluated for in vitro plant growth promoting properties (nitrogen fixation, phosphate solubilization, production of siderophores and synthesis of indolic compounds). Yerba mate seedlings were inoculated with the most promising isolates, which were identified via analyses of the sequence of their 16S rDNA gene as Bacillus circulans (12RS3) and Bacillus altitudinis (19RS3, T5S-T4). After 120 days plants showed higher root dry weight when inoculated with isolate 19RS3 and higher shoot dry weight with 19RS3 and T5S-T4. In conclusion, further studies to determine the ability of these isolates to adapt to the climatic conditions and to survive amidst the native soil microflora in yerba mate cultivated native soils, will be crucial for developing such strains as biofertilizer.

White rot fungi, such as Trametes villosa BAFC 2755, are the only organisms capable of efficiently degrading lignin due to their oxidative enzymatic system. One of the most important enzymes in this process is lacease. This is a phenol oxidase, which has applications in several biotechnological processes including pulp delignification, detoxification of recalcitrant chemicals, degradation of polycyclic aromatic compounds, bioremediation of contaminated soils and waters. The objective of this work was to evalúate the effect of pH, temperature and incubation time on the growth and oxidative potential of T. villosa BAFC 2755, for that were carried out a quantitative of tests in culture médium ME and activity in lacasa were determinated, to complete the analysis a gene fragment of this enzyme were characterize. The results obtained showed that the conditions that improved biomass production were not those that supported the greatest activity lacease. Maximun activity levéis (200 U/L), were recorded after 14 days, at pH 5.5 and 25° C. Lacase's isoenzyme profile was affected by pH, temperature and culture time, with three isoenzymes of 64 kDa, 123 kDa and 132 kDa. In addition, a nucleotide sequence of 779 bp was obtained from a gene coding for lacase, whose protein sequence showed 90% identity with sequences belonging to the lacase gene of other species of Trametes sp.

Los hongos de pudrición blanca, como Trametes villosa BAFC 2755, son los únicos organismos capaces de degradar eficientemente la lignina debido a que poseen un sistema enzimático oxidativo. Una de las enzimas más importantes en este proceso es la lacasa. Esta es una fenoloxidasa, que posee aplicaciones en varios procesos biotecnológicos incluyendo la deslignificación de la pulpa de papel, detoxificación de químicos recalcitrantes, degradación de compuestos policíclicos aromáticos, biorremediación de suelos y aguas contaminadas. El presente trabajo tuvo como objetivo evaluar el efecto del pH, la temperatura y el tiempo de incubación sobre el crecimiento y el potencial oxidativo de T. villosa BAFC 2755. Para ello se llevaron a cabo ensayos en medio de cultivo ME y se determinó la actividad lacasa. Para complementar el análisis de caracterizó una región del gen que codifica para una lacasa. Los resultados obtenidos mostraron que las condiciones que mejoraron la producción de biomasa no fueron las que soportaron la mayor actividad lacasa. Los máximos niveles de actividad se registraron a 25° C al día 14 para pH 5,5 siendo de 200 U/L. El perfil de isoenzimas de lacasa fue afectado por el pH, la temperatura y el tiempo de cultivo, encontrándose tres isoenzimas de 64 kDa, 123 kDa y 132 kDa. Además, se obtuvo una secuencia nucleotídica de 779 pb de un gen que codifica para lacasa, cuya secuencia proteica mostró identidad de 90% con secuencias pertenecientes al gen lacasa de otras especies de Trametes sp.

The aim of this research was to isolate and characterize Tríchoderma native strains from Misiones (Argentina) exploring their antagonistic capacities to phytopatogens fungi and their mass multiplication using different agricultural wastes. Fifteen native strains of Tríchoderma spp. were isolated from soil samples. These isolates were characterized via morphological observations and molecular phylogenetic analysis based on DNA sequences of the rDNA internal transcribed spacer region. The Tríchoderma native strains were identified as T. koningiopsis, T. harzianum, T. pleuroticola and T. brevicompactum. All strains showed antagonistic activities in vitro against Alternaría sp., Fusaríum sp. and Botrytis sp. T. koningiopsis LBM 090, LBM 091, LBM 092, and LBM 098 strains, T. pleuroticola LBM 097 and T. harzianum LBM 096 presented radial mycelial growth inhibition higher than 50% and antagonism index between 3 and 4 against the phytopathogens assayed. Among the different substrate sources evaluated, rice husk and rice polishing were the most suitable combination for mass multiplication of T. harzianum LBM 096.

El objetivo de esta investigación fue aislar y caracterizar cepas de Tríchoderma nativas de Misiones (Argentina) explorando sus capacidades antagónicas y su multiplicación masiva utilizando diferentes residuos agroindustriales. Quince cepas nativas de Tríchoderma spp. fueron aisladas de muestras de suelo. Estos aislamientos se caracterizaron mediante observaciones morfológicas y moleculares basados en secuencias de ADN de la región espaciadora transcrita interna del ADNr. Las cepas de Tríchoderma spp. fueron identificadas como T. koningiopsis, T. harzianum, T. pleuroticola y T. brevicompactum. Estas cepas mostraron actividades antagónicas in vitro contra Alternaría sp., Fusaríum sp. y Botrytis sp.. T. koningiopsis LBM 090, LBM 091, LBM 092 y LBM 098, T. pleuroticola LBM 097 y T. harzianum LBM 096 presentaron una inhibición del crecimiento micelial mayor del 50% y un índice de antagonismo entre 3 y 4 contra los fitopatógenos ensayados. La cáscara de arroz y el pulido del arroz fueron las combinaciones más adecuadas para la multiplicación de T. harzianum LBM 096.

The aim was to evaluate the diagnosis value of serological tests in a pediatric population with suspected celiac disease. In the serum of patients with classical symptoms, non-classical symptoms, or with an increased risk for celiac disease, antibodies anti-tissue transglutaminase IgA (a-tTG-IgA) and IgG anti-deamidated gliadin peptide (a-DGP-IgG) were measured. Endoscopic biopsy was performed on 97 patients: 62 of them had Marsh 3 lesion (celiac disease confirmed), 7 had Marsh 2 lesion and the other 28, Marsh 0-1 lesion. A 100% of positive predictive value for a tTG-IgA ≥ 100 U ml, and 95% for a-DGP-IgG ≥ 100 U/ml were obtained. When both (tTG-IgA and a-DGP-IgG) were ≤ 10 U/ml, the negative predictive value was 100%. Values of a-tTG-IgA ≥ 100 U/ml allowed the diagnosis of celiac disease in the studied population. When a-tTG-IgA and a-DGP-IgG, were both ≤ 10 U/ml it ruled out celiac disease.

Nuestro objetivo fue evaluar el valor diagnostico de pruebas serologicas de enfermedad celiaca en poblaciones pediatricas con sospecha clinica de celiaquia. En el suero de los pacientes con sintomas clasicos, no clasicos o pertenecientes a grupos de riesgo de celiaquia se dosaron anticuerpos anti-transglutaminasa tisular de tipo IgA (a-tTG-IgA) y anticuerpos contra los peptidos deaminados de gliadina de tipo IgG (a-DGP-IgG). Fueron biopsiados 97 pacientes: 62 presentaron histologia Marsh 3 (enfermedad celiaca confirmada), 7 histologia Marsh 2 y 28 histologia Marsh 0-1. Se obtuvo valor predictivo positivo 100% para valores de a-tTG-IgA ≥ 100 U/ml y 96% para a-DGP-IgG ≥ 100 U/ml. Cuando ambos anticuerpos fueron ≤ 10 U/ml se obtuvo valor predictivo negativo 100%. En nuestro estudio, los anticuerpos a-tTG-IgA permitieron el diagnostico de celiaquia cuando su valor fue ≥ 100 U/ ml. Cuando a-tTG-IgA y a-DGP-IgG fueron ambos ≤ 10 U/ml se pudo descartar enfermedad celiaca.

The white rot fungi secrete oxidative enzymes, such as laccase, with a capacity to play a part in environmentally friendly processes such as bioremediation. The aim of this study was to develop a culture medium supplemented with aromatic compounds and metal ions in order to increase laccase activity produced by Trametes sp. strain A. By means of a Plackett-Burman design, it was shown that Mn2 + was the only variable that had a significant positive effect on the enzyme activity. The highest laccase activity (272.46 U / L) was obtained at a concentration of 1.5 mM Mn2 + and maximum biomass (0.1272 g) at a concentration of 4 mM Mn2 +. It also was possible to establish a negative correlation between enzyme activity and growth of fungus. An isoenzyme of constitutive expression (60 kDa) and another inducible (100 kDa) were found in the presence of 2; 2.5; 3; 3.5 and 4 mM of Mn2+.

Los hongos de pudricion blanca secretan enzimas oxidativas, como la lacasa, con capacidad de intervenir en procesos amigables con el medioambiente, tales como la biorremediacion. El objetivo del presente trabajo fue formular un medio de cultivo suplementado con compuestos aromaticos e iones metalicos con el fin de aumentar los valores de actividad lacasa expresados por Trametes sp. cepa A. Utilizando un diseno de Plackett-Burman se logro establecer que el Mn2+ fue la unica variable que tuvo un efecto significativo positivo sobre la actividad. La mayor actividad lacasa (272,46 U/L) se obtuvo a una concentracion de 1,5 mM de Mn2+ y la maxima biomasa (0,1272 g) a una concentracion de 4 mM de Mn2+. Asimismo fue posible establecer una correlacion negativa entre la actividad enzimatica y el crecimiento del hongo. Se encontro una isoenzima de 60 kDa de expresion constitutiva y otra de 100 kDa inducible en presencia de 2; 2,5; 3; 3,5 y 4 mM de Mn2+.

This study presents a germination chamber built for a water hydroponic system provided with a fuzzy logic based controller. The system was built based on an Arduino electronics platform where the fuzzy control program and telemetry systems lie. Also, all data from sensors are stored for future studies. The telemetry system sends the data obtained by the embedded system to two computers, one conected via a USB (Universal Serial Bus) and the other conected via BT (Bluetooth). In both cases data are stored in virtual disks. The results show that the germination chamber built has better performance than the conventional floating root system.

Este estudio presenta una cámara de germinación construida para un sistema hidropónico de agua provisto de un controlador basado en lógica difusa. El sistema fue construido sobre la base de una plataforma electrónica Arduino, donde residen los sistemas de programas de control y telemetría difusos. Además, todos los datos de los sensores se almacenan para futuros estudios. El sistema de telemetría envía los datos obtenidos por el sistema a dos ordenadores, uno a través de un USB (Universal Serial Bas) y el otro a través de BT (Bluetooth). Todos los datos se almacenan en discos virtuales. Los resultados muestran que la cámara de germinación construida tiene mejor rendimiento que el sistema de raíz flotante convencional.

Some Trichoderma strains produce large amounts of hydrolytic enzymes, such as lendoglucanases,which have many industrial and biotechnological applications. The aim of the present work was to select the culture conditions favouring the secretion of cellulolytic enzymes of Trichoderma native strains from the Province of Misiones. Several experiments were done with carboxymethylcellulose 0.5% (w/v) as carbon source and Mandel's medium containing a complex nitrogen source. Inoculation method, buffer composition and incubation time were selected as independent variables. This work allowed us to establish that the inoculation as spore suspension, and the buffered medium with sodium succinate buffer pH 4.8 represent the most suitable growing conditions in a liquid medium for endoglucanases secretion by native strains of T. harzianum and T. koningiopsis.

Algunas especies de Trichoderma pueden producir grandes cantidades de enzimas hidrolíticas, tales como celulasas, las cuales poseen múltiples aplicaciones industriales y biotecnológicas. El objetivo del presente trabajo fue seleccionar las condiciones de cultivo en medio líquido que favorezcan la secreción de endoglucanasas por cepas del género Trichoderma nativas de la provincia de Misiones. Se llevaron a cabo experimentos utilizando carboximetilcelulosa 0,5% (p/v) como fuente de carbono y medio Mandels como complejo nitrogenado completo y se ensayaron diferentes condiciones de cultivo utilizando como variables el tiempo de incubación, la forma de inóculo y el sistema de control del pH del medio de cultivo. El presente trabajo permitió establecer que la inoculación en forma de suspensión de esporas y el tamponado del medio con buffer succinato de sodio pH 4,8 representan las condiciones de cultivo en medio líquido más adecuadas para la secreción de endoglucanasas por cepas nativas de T. harzianum y T. koningiopsis.

Cellulose is the main component of the cell wall with great biotechnological importance regarding the analysis of new cellulase producers microorganisms. The objective of this study was the design and testing of a submerged production system of endo-β-1.4-glucanase using pine chips as substrate; pH and T optimum action of the enzyme extracts by two white rot fungi Irpex sp. and Phlebia sp. were checked at laboratory scale. The production system proposed in this study showed a logarithmic behaviour with respect to enzyme production with maximum activity of 1 U / Lh for Irpex sp.strain and 1.37 U / Lh for Phlebia sp. strain. Enzymatic activity remained constant over time through washing and adding fresh medium, simulating a fed-batch production system. Extracts of both strains showed better performance at 50°C, with higher activity at pH 3 for extracts of Irpex sp. strain and pH 4.5 for Phlebia sp. strain. This system may be optimal to start scaling up studies.

La celulosa es el componente principal de la pared celular y es de gran importancia biotecnológica analizar nuevos microorganismos que produzcan celulasas. El objetivo de este trabajo fue diseñar y ensayar a nivel laboratorio un sistema de producción sumergido que utiliza chips de pino como sustrato para la producción de endo-1,4-β- glucanasa secretada por dos hongos de pudrición blanca, Irpex sp. y Phlebia sp. y verificar el pH y T óptimos de acción de los extractos enzimáticos. El sistema de producción propuesto en este trabajo mostró un comportamiento logarítmico respecto de la producción enzimática con un máximo de actividad de 1 U/L.h para Irpex sp. y 1,37 U/L.h para Phlebia sp. La actividad se mantuvo constante con el tiempo a través de lavados y agregado de medio fresco, simulando un sistema de producción fed-batch. Los extractos de ambas cepas mostraron una mejor performance a 50ºC, con una mayor actividad a pH 3 para los extractos de la cepa de Irpex sp. y a pH 4,5 para la cepa de Phlebia sp. Este sistema podría ser óptimo para iniciar los experimentos de escalado.

Polymorphisms of the adiponectin gene are associated with alterations in expression with subsequent decrease in plasma levels of adiponectin. The latter is associated with insulin resistance, obesity, and hypertension, all risk factors for cardiovascular disease and cancer. The objective of the present work was to standardize an amplification reaction, PCR, followed by detection by SSCP-HD (strand conformation polymorphism-heteroduplex simple) for the analysis of single nucleotide polymorphisms (SNPs) present in exon 3 of the gene ADIPOQ. DNA extractions were performed by the modified salting-out method, the primers were analyzed using bioinformatics programs, and the fragments were analyzed by the technique of Polymorphism, Single-Stranded- Heteroduplex (SSCP-HD). With the conditions used, the amplification of exon 3 of ADIPOQ was possible, identifying patterns of runs using the technique of SSCP-HD which will facilitate further studies of single nucleotide polymorphisms in the gene selected.

Los polimorfismos del gen de la adiponectina están relacionados con alteraciones en su expresión con la subsecuente disminución en los niveles plasmáticos de adiponectina, esto último se encuentra asociado a la insulino-resistencia, obesidad e hipertensión arterial, todos factores de riesgo para enfermedades cardiovasculares y oncológicas. El objetivo del trabajo fue estandarizar una reacción de amplificación, PCR, seguida de detección mediante SSCP-HD (polimorfismo de conformación de cadena simple-heterodúplex) para el análisis de los polimorfismos de simple nucleótido (SNPs) presentes en el exón 3 del gen ADIPOQ. Las extracciones de ADN se realizaron por el método Salting-Out modificado, los cebadores fueron analizados mediante programas bioinformáticos, y los fragmentos obtenidos fueron analizados mediante la técnica de Polimorfismo en la Cadena Simple-Heterodúplex (SSCP-HD). Con las condiciones utilizadas se logró la amplificación del exón 3 de ADIPOQ, identificándose patrones de corridas mediante la técnica de SSCP-HD que facilitarán el estudio posterior de los polimorfismos de simple nucleótido en el gen seleccionado.

β-thalassemias derive from a decrease in the synthesis of β-globin chains, being caused by different mutations in the β-globin gene. The molecular detection of these mutations allows the β-thalassemic genotype determination and therefore, a precise diagnosis and the underlying treatment. The aim of this study was to study the two most common finding mutations in the Argentinean population, CD39 and IVS1-110 mutations in the β-globin gene, by the ARMS-PCR technique. In 50 patients analyzed, three were positive for the CD39 mutation, corresponding to 6 % of the cases, while no patients with the IVS1-110 mutation were detected. This study provides new data on the frequency of genotypes (β/βCD39 and p / βIVS1-110) responsible for β-thalassemia heterozygote in our region.

Las β-talasemias obedecen a una disminución en la síntesis de cadenas de β-globina, y son causadas por diferentes mutaciones en el gen de β-globina. La detección molecular de estas mutaciones, permite determinar el genotipo de β-talasemia y así establecer con precisión el diagnóstico y el tratamiento a seguir. El objetivo del presente trabajo fue estudiar las dos mutaciones de hallazgo más frecuente en la población argentina, CD39 e IVS1-110 del gen de β-globina, mediante la técnica de ARMS-PCR. Del total de pacientes analizados (50), 3 resultaron positivos para la mutación CD39, correspondiente a un 6 % de los casos, mientras que no fueron detectados pacientes portadores de la mutación IVS1-110. Este estudio aporta nuevos datos sobre la frecuencia de los genotipos (β/βCD39 y β/βIVS1-110) responsables de β-talasemia heterocigota en nuestra región.

Molecular Epidemiology includes genotypic studies of different biological processes, including pathological diseases such as cancer. Therefore, the paraffin-embedded clinical samples analysis is very important for diagnosis, prevention and therapy. Previously, many problems associated with amplification of DNA obtained from paraffin-embedded tissue samples were reported. This paper compares the capability of nine methodologies for DNA extraction from paraffin-embedded clinical samples, amplifying it by PCR to analyze under-250-bp fragments corresponding to folate pathway enzymes polymorphisms. Modification of "salting-out" method with concentrations of 0.5 M NaCl and 2 % CTAB detergent and Proteinase K was the most suitable for amplifiable DNA obtention of selected amplicons. All methods showed better amplification with longer cycles and lower annealing temperatures as compared to less fragmented DNA samples.

La epidemiología molecular estudia variaciones genotípicas relacionándolas con diversos procesos biológicos, entre ellos los patológicos como el cáncer. Por ello el análisis de muestras clínicas parafinadas es de gran importancia para el diagnóstico, la prevención y la terapia. Se ha informado con anterioridad muchos problemas relacionados con la amplificación de DNA obtenido a partir tejido embebido en parafina (TEP). En este trabajo se comparó la capacidad de 9 metodologías para obtener DNA a partir de TEP y amplificarlo por PCR para analizar fragmentos de menos de 250 pb correspondientes con polimorfismos presentes en enzimas pertenecientes a la vía del folato. El método modificado de Salting-out con NaCl de 0,5 M, CTAB 2 % como detergente y Proteinasa K fue el más adecuado para obtener DNA amplificable considerando los amplicones elegidos. Todos los métodos mostraron una mejor amplificación con ciclos más prolongados y temperaturas de annealing menores comparado con DNA más íntegro.
