RESUMO: O objetivo deste estudo foi revisar os fatores tecnológicos e toxicológicos relacionados à presença de compostos carbonílicos encontrados em vinhos, incluindo acetaldeído, formaldeído, acroleína, carbamato de etila (CE) e furfural. O acetaldeído e o formaldeído podem ser formados através da oxidação do etanol e do metanol, respectivamente. A acroleína pode surgir como um produto de degradação térmica de glicerol, aminoácidos, carboidratos e triglicerídeos ou pela atividade metabólica de microorganismos. Além disso, a acroleína e o furfural são formados durante a combustão da madeira. Portanto, esses aldeídos podem estar presentes nas matérias-primas devido à contaminação ambiental. O furfural é também um produto da reação de Maillard formado a partir de açúcares e aminoácidos, enquanto o carbamato de etila ocorre através da reação entre uréia e etanol. Estes compostos podem reagir com SO2 e compostos fenólicos para formar adutos não voláteis, que modulam positivamente a estabilidade da cor, adstringência e aroma no vinho. No entanto, quando ingeridos através do vinho, os compostos carbonílicos que são eletrofílicos podem formar adutos com alvos nucleofílicos, como o DNA, resultando em genotoxicidade ao longo do trato gastrintestinal. Além disso, os compostos carbonílicos também induzem o aumento de espécies reativas de oxigênio e podem desencadear a apoptose, além de adenoma e carcinoma hepatocelular como consequência da hepatotoxicidade crônica. Doenças neurodegenerativas podem estar relacionadas à exposição aos compostos carbonílicos. Com isso, estratégias para reduzir os níveis desses compostos devem ser estudadas para obter o máximo das propriedades funcionais benéficas do consumo de vinho.

ABSTRACT: The objective of this study was to review technological and toxicological factors related to presence of carbonyl compounds found in wines, including acetaldehyde, formaldehyde, acrolein, ethyl carbamate (EC) and furfural. Acetaldehyde and formaldehyde may be formed through the ethanol and methanol oxidation, respectively. Acrolein may arise as a thermal degradation product of glycerol, amino acids, carbohydrates and triglycerides or by metabolic activity of microorganisms. In addition, acrolein and furfural are formed during wood combustion; therefore, these aldehydes may be present in raw materials due to the environmental contamination. Furfural is also a product of the Maillard reaction formed from sugars and amino acids, while ethyl carbamate occurs through the reaction between urea and ethanol. These compounds may react with SO2 and phenolic compounds to form non-volatile adducts, which positively modulates color stability, astringency and aroma in wine. However, when ingested through wine, electrophilic carbonyl compounds may form adducts with nucleophilic targets, such as DNA, resulting in genotoxicity along the gastrointestinal tract. Furthermore, carbonyl compounds induce the increase of reactive oxygen species and can trigger apoptosis, in addition to hepatocellular adenoma and carcinoma as a consequence of chronic hepatotoxicity. Neurodegenerative diseases may be related to the exposure to carbonyl compounds. Therefore, strategies to reduce the levels of these compounds should be studied in order to get the most out of the beneficial functional properties of wine consumption.

RESUMO: Os objetivos deste estudo foram verificar a ocorrência de álcool furfurílico (FA) e compostos carbonílicos (acetaldeído, acroleína, carbamato de etila (CE), formaldeído e furfural) em espumantes e avaliar, pela primeira vez, se o consumo das amostras em estudo poderia representar risco para a saúde do consumidor. Esses compostos são eletrofílicos e, portanto, podem se ligar covalentemente ao DNA, o que pode resultar em mutagenicidade. CE e formaldeído foram encontrados em baixos níveis (<1μg/L) em todas as amostras. Acetaldeído, furfural e acroleína também foram encontrados em baixos níveis (<1,5; 1,4 e 1,0μg L-1, respectivamente) em 57, 71 e 76% das amostras. Nas demais amostras, os níveis de acetaldeído, furfural e acroleína variaram de 5,2 a 54,8, 10,5 a 41,0 e 20,3 a 36,7μg L-1, respectivamente. O álcool furfurílico também foi encontrado em todas as amostras em níveis de 10,4 a 33,5μg L-1. A acroleína foi o único composto encontrado em níveis suficientes para representar risco à saúde, que ocorreu em 24% das amostras. Uma avaliação focada na origem da acroleína merece atenção, investigando a influência da concentração dos precursores e o papel da fermentação na formação do aldeído, além da avaliação da possível contaminação ambiental das uvas durante o cultivo.

ABSTRACT: The goals of this study were to verify the occurrence of furfuryl alcohol (FA) and carbonyl compounds (acetaldehyde, acrolein, ethyl carbamate (EC), formaldehyde and furfural) in sparkling wines and to evaluate, for the first time, whether the consumption of the samples under study could represent risk to consumers health. These compounds are electrophilic; and therefore, may covalently bind to DNA, which may result in mutagenicity. EC and formaldehyde were present at low levels (<1μg L-1) in all samples. Acetaldehyde, furfural and acrolein were also found in low levels (<1.5, 1.4 and 1.0μg L-1, respectively) in 57, 71 and 76% of samples. In the other samples, levels of acetaldehyde, furfural and acrolein ranged from 5.2 to 54.8, 10.5 to 41.0 and 20.3 to 36.7μg L-1, respectively. Furfuryl alcohol was also reported in all samples in levels from 10.4 to 33.5μg L-1. Acrolein was the only compound reported at levels sufficient to represent risk to health, which occurred in 24% of the samples. A study focused on the origin of acrolein deserves attention, investigating the influence of the concentration of precursors and the role of fermentation in the formation of this aldehyde, besides the evaluation of possible environmental contamination of grapes during cultivation.

Abstract Aspergillus westerdijkiae is one of the most important spoilage and toxigenic fungi contaminating coffee beans and may produce ochratoxin A (OTA), a mycotoxin that characterize a health risk to the coffee consumers. Biological control strategies can be used for prevention of fungal invasion and decrease mycotoxin exposure. The aims of this study were to evaluate the in vitro effect of three Bacillus sp. biocontrol candidates on A. westerdijkiae mycelial growth, spore counts and OTA production. A green-coffee based medium was inoculated with A. westerdijkiae and Bacillus spp. (B. safensis RF69, B. amyloliquefaciens RP103 and B. subtilis RP242) and after incubation, the fungal growth, sporulation and mycotoxin production was evaluated. Mycelial growth rate was reduced in a range between 76-95% and conidial production was also significantly decreased. All isolates were capable of reducing OTA production in a range between 62-96%. The results showed that the biocontrol candidates may be an effective control method for A. westerdijkiae and OTA in coffee.

RESUMO: O suco de uva foi incluído nas refeições de escolas públicas do estado do Rio Grande do Sul e Santa Catarina, Brasil. O suco destinado para as escolas é produzido por pequenas vinícolas através do método de arraste a vapor. Este método apresenta uma particularidade em relação aos demais métodos de produção do suco, pois as uvas não são maceradas para a extração do mosto. Cabe salientar que a maceração das uvas é o principal ponto crítico relacionado à contaminação dos sucos com ocratoxina A (OTA), que é uma micotoxina que apresenta efeitos nefrotóxicos e imunossupressores. O objetivo deste estudo foi verificar a ocorrência de OTA em sucos produzidos pelo método de arraste de vapor, pois o suco de uva pode ser uma fonte de exposição à OTA para crianças. Nenhuma amostra apresentou OTA em níveis maiores que o limite de detecção (0,05μg L-1) do método usado para determinar esta toxina. Este resultado evidenciou a qualidade adequada do suco produzido por este método de extração. Além disso, dados sobre a ocorrência de OTA são importantes para subsidiar estudos relacionados à exposição a compostos tóxicos através da dieta.

ABSTRACT: The grape juice was included in meals of public schools in the states of Rio Grande do Sul and Santa Catarina, Brazil. The juices intended for schools have been produced by small wineries through the steam distillation method. This method presents a particularity in relation to other juice production methods, since the grapes are not macerated to extract the juice. In literature is well established that maceration of grapes is the main critical point associated with the ochratoxin A (OTA) contamination in juices. OTA is a mycotoxin with nephrotoxic and immunosuppressives effects. The aim of this study was to verify the OTA occurrence in juices produced by steam distillation method, as grape juice may be a source of OTA exposure for children. No sample showed OTA levels higher than the detection limit (0.05μg L-1) of method used to determine this toxin. This study demonstrated the adequate quality of juice produced by this extraction method. Furthermore, data about OTA occurrence are important to support risk assessment studies related to the exposure to toxic compounds through diet.

A ocratoxina A (OTA) é uma micotoxina que possui propriedades nefrotóxicas, genotóxicas, teratogênicas e carcinogênicas. A presença dessa toxina em vinhos e sucos ocorre devido à contaminação das uvas por fungos toxigênicos. Estudos têm mostrado a presença desse metabólito secundário tóxico nestas bebidas, o que pode significar perdas econômicas ao setor vitícola, bem como problemas à saúde dos consumidores. Na Europa, vários estudos têm sido realizados com o objetivo de mapear as áreas em que o desenvolvimento de fungos ocratoxigênicos é mais favorável. Entretanto, no Brasil estes estudos ainda são incipientes. O Comitê de Especialistas em Aditivos Alimentares da Organização Mundial da Saúde (JECFA) estabeleceu a ingestão tolerável segura de 112ng de OTA por quilo de peso corpóreo por semana. Para verificar se população está exposta a OTA em níveis que representam risco para à saúde, é necessário comparar o parâmetro de ingestão segura, definido pelo JECFA, com os níveis de exposição a esta toxina. O monitoramento periódico dos níveis de OTA em alimentos e bebidas é justificado por alguns motivos que incluem: (i) os efeitos tóxicos dessa toxina, (ii) a publicação recente da legislação brasileira que estabelece limite máximo para OTA, (iii) a introdução do suco de uva na merenda escolar; e (iv) a recomendação da ingestão regular de vinho/suco de uva devido as suas propriedades funcionais.

Ochratoxin A (OTA) is a mycotoxin with nephrotoxic, genotoxic, teratogenic and carcinogenic properties. The presence of this toxin in wines and juices occurs due to the development of toxigenic fungi in grapes. Studies have shown the presence of this toxic secondary metabolite in these beverages may results in economic losses to the winery as well as health problems for consumers. In Europe, several studies have been done in order to map the areas where the development of ochratoxigenic fungi is more favorable. However, in Brazil these studies are still incipient. The Joint Expert Committee on Food Additives of the World Health Organization (JECFA) established the safe tolerable intake of 112ng OTA per kg of body weight per week. To verify whether the population is exposed to OTA levels that pose a risk to health is necessary to compare the parameter of safe ingestion defined by JECFA with the levels of exposure to this toxin. Periodic monitoring of the OTA levels in food and beverage has been justified by some reasons including: (i) the toxic effects of this toxin, (ii) the recent publication of the Brazilian legislation establishing maximum limit for OTA, (iii) the introduction of grape juice in school meals and (iv) the recommendation of regular wine intake because of their functional properties.

A part of apples destined to juice production is generally of poor quality. Apples from cold storage or recently harvest (ground harvested or low quality apples) are stored under ambient conditions until they are processed. Since Penicillium expansum and P. griseofulvum are the principal fungal species isolated from stored apples in Brazil, the objective of this study was to investigate the ability of these strains to produce patulin in apples and report the consequences of this type of storage in loss of quality. The toxin was quantified using thin layer chromatography and charge-coupled device camera (TLC-CCD). The rate and quantities that P. expansum and P. griseofulvum can grow and produce patulin are highly dependent on the fungal strain and time. Lesion diameter resulted to be independent of the strain considered. The maximum period of time which apples were kept at cold storage (4 ºC) without patulin accumulation was 27 days. When these apples were kept at 25 ºC during 3 days, both factors lesion diameter and patulin production increased significantly. These results confirm that time in which apples are taken out from cold storage room before juice production is critical in order to prevent patulin accumulation.

A cromatografia gasosa bidimensional abrangente (GC×GC) é uma técnica analítica relativamente nova, que tem sido amplamente empregada para análise de vários tipos de amostras complexas. A GC×GC proporciona maior capacidade de pico, sensibilidade e resolução, quando comparada à cromatografia gasosa monodimensional (1D-GC). Além disso, a estruturação dos cromatogramas bidimensionais auxilia na identificação de classes de compostos. No que diz respeito à análise de alimentos e bebidas, a eficiência superior da GC×GC resulta em benefícios únicos quando comparada às técnicas analíticas disponíveis para os mais variados objetivos de análise: separação de grupos de compostos aroma-ativos, verificação de perfil cromatográfico, análise de compostos alvo ou identificação de componentes desconhecidos. Quando compostos aroma-ativos estão presentes em nível de traços, a GC×GC também pode ser empregada como uma técnica de alto potencial devido a sua sensibilidade superior. Tendo em vista que a fração volátil de muitos alimentos e bebidas pode determinar a aceitação ou rejeição destes produtos por parte do consumidor, a caracterização da mesma pode ser feita com base na composição destes compostos voláteis. Neste trabalho, as aplicações da GC×GC para análise de voláteis em alimentos e bebidas são apresentadas, desde a fase inicial de sua implementação até 2010. Alguns tópicos relacionados aos princípios da GC×GC foram também discutidos.

Comprehensive two-dimensional gas chromatography (GC×GC) is a relatively new analytical technique, which has been widely used for the analysis of a variety of complex samples. GC×GC offers increased peak capacity, higher sensitivity, and improved resolution when compared to conventional one-dimensional gas chromatography (1D-GC). In addition, it generates structured two-dimensional chromatograms, which aids in the identification of compound classes. The enhanced separation provided by GC×GC has been a distinctive benefit when it is compared to other available analytical techniques that deal with the analysis of food and beverages, irrespective of the goal of the analysis, i.e. aroma group-type separation and fingerprinting, target-volatile compound analysis or identification of unknowns. When trace amounts of aroma active compounds are present, GC×GC may also be used as a very powerful tool for their analysis due to its higher sensitivity. As volatile fractions may determine the acceptance or rejection of many foods and beverages by the consumer, the characterization of these products may be done having volatile composition as a basis. Applications of GC×GC for the analysis of volatiles of foods and beverages were revised since the emergence of this technique until 2010. Some topics related to principles of GC×GC were also discussed.

A photometric procedure was developed for determination of aflatoxin B1 in peanuts by TLC-CCD technique. The quantification and detection limit were 1.2 μg kg-1 and 0.4 ng per spot, respectively, with mean recovery of 98%. The CCD camera is sufficiently sensitive to detect small changes in spots fluorescence intensity and the results for performance confirmed the efficiency of the method. Another important property of CCD detector is its linearity for a wide range of luminous stimulus determined by analysis of five-point calibration curves using the intensity of AFB1 fluorescence versus AFB1 concentration (0.8 to 4.8 ng per spot). The method was applied to the analysis of thirty nine peanut samples and aflatoxin B1 levels ranged from 16 to 115 μg kg-1. The TLC-CCD and the photometric procedure developed in this study demonstrated to be a simple and efficient tool for quantitative analyses of AFB1 in peanut samples.

A colonização dos Aspergillus da secção Nigri nas uvas durante o cultivo é a principal fonte de ocratoxina A (OTA) nos vinhos. A. carbonarius e A. niger são os principais produtores desta micotoxina em uvas e são fungos oportunistas que, se desenvolvem, principalmente, nas bagas danificadas durante seu amadurecimento. A produção de OTA em uvas é influenciada pelas condições climáticas e áreas geográficas, bem como pela variedade de uva, pelo sistema de cultivo e pelos danos causados nas uvas por insetos, infecção fúngica ou excesso de irrigação e chuva. As medidas para o controle de fungos toxigênicos devem considerar esses pontos críticos de controle. A OTA presente nas uvas é transferida para o vinho durante o processo de vinificação, sendo que um aumento na concentração de OTA ocorre após a maceração das uvas. Durante o envelhecimento do vinho, observa-se que a toxina permanece estável, pois a mesma concentração de OTA é encontrada no vinho após um ano de armazenamento. Boas práticas de produção, como, por exemplo, a seleção e separação dos cachos de uva com desenvolvimento fúngico visível auxilia, consideravelmente, na redução dos níveis de contaminação por fungos produtores de OTA, bem como dos níveis dessa micotoxina nos vinhos.

The infection of grapes by 'black aspergilli' in the field is the main source of ochratoxin A (OTA) in the wine. Aspergillus carbonarius and A. niger fungi are the main producers of this mycotoxin in grapes. They are opportunistic fungi that develop mainly on damaged berries at ripening. The production of OTA in grapes is influenced by climatic conditions, geographical location, grape varieties, crop system, berries damage caused by insets, fungal infection or excessive irrigation and rainfall. Control measures for toxigenic mycoflora in the vineyards must consider these critical control points. OTA in grapes is transferred to wine during vinification and an increase of OTA concentration in must was observed during maceration. The toxin remains stable during wine aging because the same OTA concentration is found in wine after 1 year. Good Agriculture Practices including balanced soil tillage, irrigation, nitrogen fertilization and pruning associated with Good Manufacturing Practices, such as separation of rot bunches helps considerably to reduce OTA-producing fungi and levels of mycotoxin in wine.

A patulina é uma micotoxina produzida por várias espécies de Penicillium, Aspergillus e Byssochlamys. Em experimentos com animais, ela demonstrou ter atividade mutagênica, carcinogênica e teratogênica. Tem sido freqüentemente encontrada em maçãs e derivados. A patulina é facilmente transferida da maçã para o suco durante o processamento devido a sua alta solubilidade em água. Essa micotoxina é muito estável ao aquecimento em meio ácido, como no suco de maçã. Assim, a presença de patulina em suco de maçã é um indicador da qualidade das maçãs utilizadas no processamento. Muitos métodos têm sido desenvolvidos para a determinação da patulina, principalmente baseados na extração líquido-líquido com acetato de etila e determinação por CLAE. É importante evidenciar a necessidade de legislação que regulamente limites dessa micotoxina em alimentos no Brasil. Esta revisão bibliográfica tem como objetivos descrever as principais características da patulina, a ocorrência, os aspectos toxicológicose os métodos desenvolvidos para sua detecção e controle durante os estágios da produção da maçã e suco.

Patulin is a mycotoxin produced by several Penicillium, Aspergillus and Byssochlamys species. Patulin is a highly toxic compound which has shown to be mutagenic, carcinogenic and teratogenic in experiments with animals. It has often been found in apples and apple products. Patulin is easily transfered into apple juice during processing due to its high solubility in water. This mycotoxin is very stable to heat in acidic medium as in apple juice. Thus, patulin content of apple juice is an indicator of the quality of the apples used to juice production. Many methods have been developed for the patulin determination mainly based on liquid-liquid extraction with ethyl acetate and use of HPLC for detection. It is important to show the need of legislation that imposes patulin limits in foods in Brazil. The objectives of this review are to describe the main patulin characteristics, occurrence, toxicological aspects, methods developed for patulin detection and control during the stages of apple and juice production.

O objetivo deste trabalho foi determinar a composição físico-química de méis de Apis mellifera L. produzidos em dois anos consecutivos na região noroeste do Estado do Rio Grande do Sul, Brasil. As características físico-químicas de 36 amostras de mel foram comparadas utilizando o teste T de Student e a adequação aos padrões da legislação brasileira da qualidade do mel foi verificada. Os resultados das análises dos méis variaram para as características analisadas conforme segue: pH (3,3-4,4), umidade (14,7-19,8%), acidez total (16,9-49,2meq kg-1), hidroximetilfurfural (0,15-48,3mg kg-1), açúcares redutores (60,1-75,9%), açúcares não-redutores (1,35-5,99%), cinzas (0,05-0,47%) e sólidos insolúveis (0,016-0,27g kg-1). A análise estatística das amostras de mel produzidas em dois anos consecutivos mostrou que houve diferença significativa na umidade, na acidez e na hidroximetilfurfural. Os méis produzidos nesta região apresentam boa qualidade e características físico-químicas compatíveis aos padrões da legislação brasileira.

The objective of this research was to determine the physicochemical composition of Apis mellifera L. honeys produced in two consecutive years in the northwest region of Rio Grande do Sul State, Brazil. The physicochemical characteristics of 36 honey samples were compared using Student’s T-test and their adequacy to standards established by the Brazilian legislation for honey quality was checked. The results varied as follows: pH (3.3-4.4), moisture (14.7-19.8%), acidity (16.9-49.2meq kg-1), hydroxymethylfurfural content (0.15-48.3mg kg-1), reducing sugar (60.1-75.9%), non-reducing sugar (1.35-5.99%), ashes (0.05-0.47%) and solids non-soluble in water (0.016-0.27g kg-1). The statistical analyses of honey samples produced in two consecutive years showed that the moisture, acidity and hydroxymethylfurfural content varied significantly. The honey samples produced in this region were in accordance with the Brazilian legislation for honey quality.
