This work has focused on the synthesis of three nanosystems composed of superparamagnetic iron oxide nanoparticles (SPIONs) coated either with a carboxylate platinum(IV) complex (PD = cis,cis,trans-[Pt(NH3)2Cl2(HOOCCH2CH2COO)(OH)]) or with platinum(II) complex functionalized dextrans (DexPt1 = [Pt(Dex-NH2)Cl3] and DexPt2 = [Pt(Dex-NH2)(NH3)2(H2O)]). All nanosystems have shown superparamagnetic behavior. Powder X-ray diffraction (XRD) has confirmed that the SPIONs were iron oxide phase and transmission electron microscopy (TEM) has shown average size of 6 nm (M6). Characterization of the nanosystems by inductively coupled plasma atomic emission spectroscopy (ICP AES) has revealed the presence of platinum on their surface (M6@PD, 0.54 mmol g-1 of Fe and M6@CA@DexPt1-2, 0.32-1.20 mmol g-1 of Fe); infrared spectroscopy (IR) and thermogravimetric and differential thermal analyses (TG-DTA) have confirmed the presence of dextran. Furthermore, the colloidal properties of these nanosystems (M6@PD and M6@CA@DexPt1-2) have been evaluated in water and in PBS buffer. Although M6@PD has shown good colloidal dispersion in water in the pH range of 2.0-8.0, the system underwent rapid agglomeration in PBS buffer. The M6@CA@DexPt1-2 nanosystems have exhibited improved colloidal behavior both in water and in PBS, where hydrodynamic sizes were kept below 100 nm over a large pH range (2.0-12.0). Furthermore, the latter systems have displayed isoelectric points below pH 5.0 and low surface charges at pH 7.0 (ζ-potential = -10 mV) and therefore PBS did not affect their colloidal stability.

Abstract Resistance to benznidazole in certain strains of Trypanosoma cruzi may be caused by the increased production of enzymes that act on the oxidative metabolism, such as mitochondrial tryparedoxin peroxidase which catalyses the reduction of peroxides. This work presents cytotoxicity assays performed with ferrocenyl diamine hydrochlorides in six different strains of T. cruzi epimastigote forms (Y, Bolivia, SI1, SI8, QMII, and SIGR3). The last four strains have been recently isolated from triatominae and mammalian host (domestic cat). The expression of mitochondrial tryparedoxin peroxidase was analyzed by the Western blotting technique using polyclonal antibody anti mitochondrial tryparedoxin peroxidase obtained from a rabbit immunized with the mitochondrial tryparedoxin peroxidase recombinant protein. All the tested ferrocenyl diamine hydrochlorides were more cytotoxic than benznidazole. The expression of the 25.5 kDa polypeptide of mitochondrial tryparedoxin peroxidase did not increase in strains that were more resistant to the ferrocenyl compounds (SI8 and SIGR3). In addition, a 58 kDa polypeptide was also recognized in all strains. Ferrocenyl diamine hydrochlorides showed trypanocidal activity and the expression of 25.5 kDa mitochondrial tryparedoxin peroxidase is not necessarily increased in some T. cruzi strains. Most likely, other mechanisms, in addition to the over expression of this antioxidative enzyme, should be involved in the escape of parasites from cytotoxic oxidant agents.

Quatro novos conjugados do tipo 6-aminocumarina-naftoquinona foram sintetizados e suas propriedades fotofísicas e eletroquímicas, investigadas. O composto 2-cloro-3-(2-oxo-2H-cromen-6-ilamino)-1,4-naftoquinona 1 não apresentou fluorescência apreciável, em comparação com a 6-aminocumarina, 6-AC. Visando entender as razões da extinção da fluorescência neste composto, duas estratégias foram imaginadas. Primeiramente, o composto 1 foi metilado no nitrogênio para remover a interação eletrostática intramolecular N-H...O=C que mantém as duas unidades fixas. Entretanto, as propriedades de emissão do produto 2 não se mostraram significantemente diferentes das do precursor 1. Como os cálculos usando a teoria do funcional da densidade dependente do tempo (TD-DFT) dos compostos 1 e 2 indicaram que a supressão da fluorescência relaciona-se ao caráter aceptor no anel naftoquinônico, a segunda estratégia envolveu a substituição do átomo de cloro na posição 2 do núcleo naftoquinônico por diferentes grupos doadores de elétrons (compostos 3-5). Novamente não houve mudanças apreciáveis nas propriedades de emissão. Para explicar estes resultados foram feitos cálculos TD-DFT dos estados fundamental (S0) e excitado (S1) de todas as moléculas em solução, os quais indicaram que o grupo fluorescente (6-AC) doa elétrons para o LUMO da naftoquinona, resultando em uma transferência de elétron fotoinduzida oxidativa (oxidative-PET).

Four novel 6-aminocoumarin-naphthoquinone conjugates were synthesized and their photophysical and electrochemical properties, investigated. 2-Chloro-3-(2-oxo-2H-chromen-6-ylamino)-1,4-naphthoquinone 1 did not present appreciable fluorescence in solution in comparison with 6-aminocoumarin, 6-AC. In order to understand the reasons for the fluorescence quenching in this compound, two strategies were attempted. Firstly, compound 1 was N-methylated to remove the intramolecular N-H...O=C electrostatic interaction that maintained the two units fixed, but the emission properties of the product 2 were not significantly different from those of 1. Time-dependent density functional theory (TD-DFT) calculations of compounds 1 and 2 indicate that the fluorescence quenching is related to the electron acceptor character of the naphthoquinone ring. The second strategy, therefore, involved the substitution of the chlorine atom in position 2 of the naphthoquinone nucleus for different electron donor groups (compounds 3-5), but again the emission properties did not change significantly. To explain these experimental findings, TD-DFT calculations of the ground (S0) and excited (S1) states of all molecules in solution were carried out. The results suggest that the energy states in these conjugates are such that the fluorescent group (6-AC) donates electrons to the naphthoquinone LUMO resulting in an oxidative photoinduced electron transfer (oxidative-PET).

Três novos complexos de platina(IV), cis,cis,trans-[Pt(HL1-3)Cl2(OH)2] 1b-3b (HL = 2-hidroxi3-[(R¹-amino)(piridin-2-il)metil]-1,4-naftoquinona, R¹ = n-butil, HL1; n-heptil, HL2 and n-decil, HL3) foram obtidos através da oxidação dos respectivos precursores cis-[Pt(HL1-3)Cl2] 1a-3a. Estudos de voltametria cíclica de 1b-3b em MeCN mostraram o processo redox quase reversível do íon naftoquinonato (NQO-, i.e. , L-) e o processo irreversível atribuído à redução do par Pt4+/Pt2+, em potenciais aproximadamente 400 mV menos negativos que no precursor da cisplatina cis,cis,trans-[Pt(NH3)2Cl2(OH)2]. Propõe-se que este processo Pt4+/Pt2+ seja favorecido, em 1b-3b, por uma interação de hidrogênio entre o grupo 2-hidroxil da naftoquinona e um ligante hidroxil axial. Estudos de citotoxicidade contra quatro linhagens de células tumorais humanas mostraram que, em geral, os derivados de platina(IV) e platina(II) exibem o mesmo perfil citotóxico, além de serem mais ativos que a cisplatina. Valores de CI50 in vitro mais baixos foram observados para 2b-3b, cujos ligantes possuem os maiores grupos R¹ (HL2-HL3), sendo, portanto, mais lipofílicos. Além disto, os valores semelhantes de CI50 dos complexos análogos de platina(II) e platina(IV) devem-se à rápida redução de 1b-3b in vitro para gerar 1a-3a.

Three novel platinum(IV) complexes cis,cis,trans-[Pt(HL1-3)Cl2(OH)2] 1b-3b (HL = 2-hydroxy-3-[(R¹-amino)(pyridin-2-yl)methyl]-1,4-naphthoquinone, R¹ = n-butyl, HL1; n-heptyl, HL2 and n-decyl, HL3) have been obtained from the oxidation of the respective precursors cis-[Pt(HL1-3)Cl2] 1a-3a. Cyclic voltammetry studies of 1b-3b in MeCN showed the quasi-reversible naphthoquinonate (NQO-, i.e. , L-) redox process and irreversible process attributed to the reduction of the Pt4+/Pt2+ pair, at potentials about 400 mV less negative than for the cisplatin precursor cis,cis,trans-[Pt(NH3)2Cl2(OH)2]. Hydrogen bond interaction between the naphthoquinone 2-hydroxyl group and an axially coordinated hydroxide ligand in 1b-3b has been proposed to favor the Pt4+/Pt2+ reduction. The cytotoxicity studies against four human cancer cell lines have shown that in general the platinum(IV) and platinum(II)derivatives exhibit the same cytotoxic profile and are all more active than cisplatin. The lowest in vitro IC50 values have been observed for 2b-3b, which bear ligands with the largest R¹ groups (HL2-HL3) being the most lipophilic. Furthermore similar IC50 values for platinum(II) and platinum(IV) complexes of the same ligands have been associated with rapid in vitro reduction of the latter complexes to afford 1a-3a.

A reação de tert-butil 2-(3-cloro-1,4-dioxo-1,4-di-hidronaftalen-2-ilamino)etilcarbamato com CF3COOH/CH2Cl2 fornece 5-cloro-3,4-di-hidrobenzo[f]quinoxalin-6(2H)-ona. Este composto sofre desidrogenação promovida por ácido na presença de água para dar a 5-clorobenzo[f]quinoxalin-6-ol, inédita. A estrutura molecular no estado sólido, determinada por um estudo de difração de raios X (XRD), e os dados em solução confirmam que a benzoquinoxalina existe na forma do tautômero enol-imina, tanto no estado sólido, quanto em solução, diferentemente de 5-cloro-3,4-di-hidrobenzo[f]quinoxalin-6(2H)-ona que exibe o arranjo ceto-amino. Cálculos de teoria do funcional da densidade (DFT) confirmam a preferência da benzoquinoxalina e dos compostos análogos contendo grupos H ou CH3 no lugar do Cl pela forma enol-imina. Sugere-se que a preferência da benzoquinoxalina pela estrutura enol-imina se deva ao maior caráter aromático desta estrutura em comparação com a forma ceto-amina. Os cálculos DFT dos dois tautômeros das benzo[a]fenazin-5(7H)-onas análogas às benzo[f]quinoxalin-6(4H)-onas indicaram que as estabilidades relativas são dominadas por efeitos de solvatação, no primeiro caso, e pelo grau de aromaticidade no segundo.

The reaction of tert-butyl 2-(3-chloro-1,4-dioxo-1,4-dihydronaphthalen-2-ylamino) ethylcarbamate with CF3COOH/CH2Cl2 yields 5-chloro-3,4-dihydrobenzo[f]quinoxalin6(2H)-one which undergoes acid-promoted dehydrogenation in the presence of water to give novel 5-chlorobenzo[f]quinoxalin-6-ol. The molecular structure of 5-chlorobenzo[f]quinoxalin6-ol in the solid state, determined by an X-ray diffraction (XRD) study, and the solution data confirm that it exists as the enol-imine tautomer, both in the solid state and in solution, differently from 5-chloro-3,4-dihydrobenzo[f]quinoxalin-6(2H)-one, which exhibits the keto-amine arrangement. Density functional theory (DFT) calculations confirmed the preference of 5-chlorobenzo[f]quinoxalin-6-ol and of the derivatives containing H and CH3 groups in place of the Cl atom for the enol-imine tautomer. It is suggested that the enol-imine structure is preferred for 5-chlorobenzo[f]quinoxalin-6-ol as a consequence of the higher aromatic character of this structure in comparison with the keto-amine form. DFT calculations carried out on the two tautomers of the benzo[a]phenazin-5(7H)-ones analogous to the benzo[f]quinoxalin-6(4H)-ones showed that the relative stabilities are dominated by solvation effects in the first case and the degree of aromaticity, in the latter.

Nanopartículas de sílica (NPs) modificadas covalentemente com uma aminonaftoquinona foram sintetizadas e caracterizadas. A aminopropilsilicagelnaftoquinona (APSGNQ) foi obtida por substituição nucleofílica do grupo metóxi da 2-metóxi-1,4-naftoquinona por NPs modificadas com o grupo aminopropil (APSG). Os espectros de ressonância magnética nuclear de 13C e 29Si no estado sólido confirmaram que a naftoquinona encontra-se ligada covalentemente à aminopropilsilicagel. Como o composto APSGNQ é solúvel em solventes orgânicos comuns, foi possível quantificar o ancoramento da naftoquinona na superfície das NPs modificadas por espectroscopia no ultravioleta-visível (0,56 mmol de naftoquinona ancorada por grama de APSGNQ), por comparação com o espectro do composto análogo 2-aminobutil-1,4-naftoquinona (ABNQ). Os dados das análises elementares indicaram que aproximadamente 8% da propilamina presente na superfície do composto APSGNQ não reagiu com a metóxinaftoquinona. Essas NPs de sílica multifuncionais têm potencial para aplicações médicas.

The synthesis and characterization of silica nanoparticles (NPs) covalently modified with an aminonaphthoquinone are reported. The aminopropylsilicagelnaphthoquinone (APSGNQ) was obtained by nucleophilic substitution of 2-methoxy-1,4-naphthoquinone with aminopropylsilicalgel (APSG) NPs. Solid state 13C and 29Si nuclear magnetic resonance spectra confirmed that the naphthoquinone is covalently bonded to APSG. Due to the solubility of APSGNQ in common organic solvents, solution ultraviolet-visible spectroscopy was used to determine the amount of naphthoquinone on the NPs surface (0.56 mmol of incorporated naphthoquinone per gram of APSGNQ) by comparison with the spectrum of 2-aminobutyl-1,4-naphthoquinone (ABNQ). Elemental analysis indicated that about 8% of the surface propylamine remained unreacted in APSGNQ. These multifunctional silica NPs have potential in medical applications.

Novas 2-(R-fenil)amino-3-(2-metilpropenil)-[1,4]-naftoquinonas (R = H, 4-OMe, 4-Ferrocenil, 4-Me, 3-Me, 4-I, 3-I, 4-CN, 3-CN, 4-NO2 e 3-NO2) derivadas do nor-lapachol [2-hidroxi-3-(2-metilpropenil)-1,4-naftoquinona] foram obtidas em bons rendimentos. A estrutura dos compostos foi proposta com base em estudos de difração de raios-X (R = OMe, 2b), dados de RMN de ¹H e 13C e cálculos teóricos utilizando o funcional B3LYP e a base 6-311+G(2d,p). Os potenciais de meia-onda das aminonaftoquinonas e o deslocamento químico do hidrogênio da cadeia 3-propenil dos compostos 2a-k mostraram boa correlação com as constantes de Hammett dos substituintes presentes no anel fenileno. A avaliação da citotoxicidade evidenciou atividade antitumoral promissora para o substrato metóxi-nor-lapachol 1 e o derivado 4-ferrocenil 2c.

Novel 2-(R-phenyl)amino-3-(2-methyl-propenyl)-[1,4]-naphthoquinones (R = H, 4-OMe, 4-Ferrocenyl, 4-Me, 3-Me, 4-I, 3-I, 4-CN, 3-CN, 4-NO2 and 3-NO2) derived from nor-lapachol [2-hydroxy-3-(2-methylpropenyl)-1,4-naphthoquinone] were obtained in good yields. Their structures were proposed on the basis of a single crystal X-ray diffraction study (R = OMe, 2b), ¹H and 13C NMR studies and calculations using the B3LYP functional and the 6-311+G(2d,p) basis set. The half-wave potentials of the aminonaphthoquinones and ¹H NMR chemical shifts of the 3-propenyl hydrogen in 2a-k show good correlation with the substituent Hammett constants on the phenylamino ring. The antitumor assays showed promising activity for substrate methoxy-nor-lapachol 1 and the 4-ferrocenyl derivative 2c.

Cálculos teóricos utilizando os funcionais B3LYP e PBE1PBE e as bases 6-31G(d) e 6-311+G(2d,p) para o sistema 3-(2-fenil-hidrazona)-naftaleno-1,2,4-triona, em solução (dmso) e em fase gasosa, evidenciaram, em ambos os casos, a maior estabilidade da forma ceto-hidrazona (rotâmeros Ia e Ib) comparada às formas enol-azo (rotâmeros IIa/IIb, por volta de 14 kcal mol-1) e III (aproximadamente 6 kcal mol-1). A natureza do substituinte no grupo fenil não influenciou a estabilidade relativa dos tautômeros. Estes resultados foram confirmados por dados espectroscópicos dos derivados HL1-HL13, sintetizados a partir da 2-hidroxi-1,4-naftoquinona e arilaminas (R = 4-OMe, 4-N2-C6H5, 4-Cl, 4-I, 3-I, 2-I, 4-COOH, 3-COOH, 4-CN, 3-CN, 4-NO2, 3-NO2, 2-NO2). A avaliação da atividade anticâncer in vitro (contra linhagens de células cancerosas SF-295, HCT-8, MDAMB-435 e HL-60) e bactericida (Bacillus cereus, Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia e Pseudomonas aeruginosa) dos compostos HL1-HL13 e dos seus respectivos complexos de cobre(II), [Cu(L1-13)2], foi avaliada. Em geral a atividade bactericida foi baixa, exceto para o derivado HL5 (R = 3-I), mais ativo do que o controle; entretanto, seu complexo não foi ativo. Por outro lado, a complexação levou, em geral, ao aumento da atividade antitumoral dos pré-ligantes. O complexo [Cu(L13)2] (R = 3-NO2) apresentou moderada citotoxicidade contra leucemia humana (HL-60).

DFT calculations using the B3LYP and PBE1PBE functionals with the standard 6-31G(d) and 6-311+G(2d,p) basis sets were carried out for the 3-(2-phenylhydrazone)-naphthalene-1,2,4-trione system in solution (dmso) and in the gas phase, and showed the keto-hydrazone forms (rotamers Ia and Ib) to be more stable than the enol-azo forms (rotamers IIa and IIb, by about 14 kcal mol-1) and III (by approximately 6 kcal mol-1), independently of the nature of the substituent in the phenylene ring. These results were confirmed by spectroscopic data on the derivatives HL1-HL13, obtained from 2-hydroxy-1,4-naphthoquinone and arylamines (R = 4-OMe, 4-N2-C6H5, 4-Cl, 4-I, 3-I, 2-I, 4-COOH, 3-COOH, 4-CN, 3-CN, 4-NO2, 3-NO2, 2-NO2). The in vitro antitumor (against SF-295, HCT-8, MDAMB-435 and HL-60 cancer cell lines) and antibacterial activities (Bacillus cereus, Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia and Pseudomonas aeruginosa) of compounds HL1-HL13 and of their respective copper(II) complexes, [Cu(L1-13)2], were tested. In general, these compounds exhibited low antibacterial activity, except for HL5 (R = 3-I), more active than the control; however, the corresponding complex was inactive. In contrast, increased cytotoxicity was observed upon complexation. Complex [Cu(L13)2] (R = 3-NO2) presented moderate cytotoxicity against human leukemia (HL-60).

Uma série de novas Bases de Mannich (HL1-HL13) derivadas da 2-hidroxi-1,4-naftoquinona (lausona), benzaldeídos substituídos [C6H2R¹R²R³C(O)H] e várias aminas primárias (NH2R4, R4 = n-butil, benzil, alil, 2-furfuril) e seus complexos de Cu2+, [Cu(L1)2]-[Cu(L13)2], foram sintetizados e caracterizados por métodos analíticos e espectroscópicos. As estruturas dos complexos 1(R¹ = R² = R³ = H; R4 = Bu), 2(R¹ = R³ = H; R² = NO2; R4= Bu) e 7(R¹ = OH; R² = R³ = H; R4= Bu) foram determinadas por estudos de difração de raios-X de monocristal. Todos os compostos cristalizam em grupos espaciais centrossimétricos, com um cobre no centro de inversão. Dois L- coordenam-se através dos átomos de oxigênio do naftalen-2-olato e do nitrogênio da amina secundária, formando anéis quelatos de seis membros ao redor do átomo de cobre em um ambiente trans-N2O2. A atividade antimicrobial de todos os compostos foi testada em sete diferentes linhagens de bactérias: Bacillus cereus, Bacillus subtilis, Escherichia coli, Enterococcus faecalis, Klebsiella pneumoniae, Pseudomonas aeruginosae Staphylococcus aureus. Em geral, as bases de Mannich foram mais ativas que os complexos, sendo HL11(R¹ = OH; R² =H; R³ = Me; R4= Bn) e HL13(R¹ = OH; R² = H; R³ = Br; R4= Bn) os inibidores mais potentes. O MIC para o composto mais ativo HL11contra S. Colifoi 20 µmol L-1 (8 µg mL-1), melhor que o cloranfenicol (90 µmol L-1) e bem abaixo da maioria dos valores descritos para outras naftoquinonas.

A series of novel Mannich bases (HL1-HL13) derived from 2-hydroxy-1,4-naphthoquinone (lawsone), substituted benzaldehydes [C6H2R¹R²R³C(O)H] and various primary amines (NH2R4, R4 = n-butyl, benzyl, allyl, 2-furfuryl), and their Cu2+ complexes, [Cu(L1)2]-[Cu(L13)2], have been synthesized and fully characterized by analytical and spectroscopic methods. The structures of complexes 1(R¹ = R² = R³ = H; R4 = Bu), 2(R¹ = R³ = H; R² = NO2; R4= Bu) and 7 (R¹ = OH; R² = R³ = H; R4= Bu) were determined by single crystal X-ray diffraction studies. All complexes crystallize in centrosymmetric space groups, with a copper atom in the inversion centre. Two L- coordinate through the naphthalen-2-olate oxygen and secondary amine-N atoms, forming six-membered chelate rings around the copper atom in a trans-N2O2 environment; spectroscopic data confirm that the other complexes exhibit similar molecular arrangement. The antimicrobial activity of all compounds has been tested on seven different strains of bacteria: Bacillus cereus, Bacillus subtilis, Escherichia coli, Enterococcus faecalis, Klebsiella pneumoniae, Pseudomonas aeruginosaand Staphylococcus aureus. In general, Mannich bases were more active than complexes, HL11(R¹ = OH; R² =H; R³ = Me; R4= Bn) and HL13(R¹ = OH; R² = H; R³ = Br; R4= Bn) being the most potent inhibitors. The MIC for the most active compound HL11against S. Coliwas 20 µmol L-1 (8 µg mL-1), better than Chloramphenicol (90 µmol L-1) and well below most values reported for other naphthoquinones.

A toxicidade da lausona (2-hidroxi-1,4)-naftoquinona e de diversos derivados foi avaliada frente à Artemia salina e ao molusco Biomphalaria glabrata, o principal vetor de transmissão da esquistossomose no Brasil. Entre os dezessete compostos testados, nove apresentaram um perfil de toxicidade menor que 100 µg/mL, sendo potenciais agentes moluscicidas de acordo com as designações da Organização Mundial da Saúde. No presente estudo, os compostos contendo substituintes apolares exibiram as maiores atividades. Estes compostos também se mostraram significantemente tóxicos frente à A. salina.

The toxic profile of lawsone (2-hydroxy-[1,4]naphthoquinone) and a series of [1,4]naphthoquinone derivatives was evaluated against the brine shrimp Artemia salina and against the mollusk Biomphalaria glabrata, the main transmitting vector of schistosomiasis in Brazil. Of the seventeen compounds tested nine fell below the threshold of 100 µg/mL set for potential molluscicidal activity by the World Health Organization. As a general rule derivatives with non-polar substituents presented the highest molluscicidal activities. These substances showed significant toxicity in A. salina lethality bioassay.

O lapachol, α e β-lapachona são naftoquinonas obtidas de espécies de Tabebuia, apresentam propriedades antiinflamatória, antibacteriana, anticâncer e tripanossomicida. O objetivo deste trabalho foi investigar um possível efeito espasmolítico destas naftoquinonas em íleo de cobaia, uma vez que, outras naftoquinonas inibem a atividade contrátil de músculos lisos. O lapachol, α e β-lapachona inibiram as contrações fásicas induzidas tanto por carbacol (CI50 = 1,5 ± 0,2 x 10-4; 7,3 ± 0,9 x 10-5 e 3,2 ± 0,5 x 10-5 M, respectivamente) quanto por histamina (CI50 = 3,6± 0,5; 3,6 ± 0,7 e 3,3 ± 0,6 x 10-5 M, respectivamente). Estes compostos também relaxaram o íleo pré-contraído com KCl (CE50 = 1,2 ± 0,4; 4,3 ± 0,8 e 2,7 ± 0,2 x 10-5 M, respectivamente); carbacol (CE50 = 2,6 ± 0,7; 3,5 ± 0,5 e 2,2 ± 0,7 x 10-5 M, respectivamente) ou histamina (CE50 = 3,0 ± 0,8; 1,1 ± 0,3 e 3,3 ± 0,6 x 10-5 M, respectivamente) de maneira dependente de concentração. Este efeito é provavelmente devido à inibição do influxo de Ca2+ através dos canais de Ca2+ dependentes de voltagem (CaV). β-lapachona antagonizou (pD'2 = 5,73 ± 0,12; "slope" = 1,51 ± 0,05) as contrações induzidas por CaCl2 em meio despolarizante nominalmente sem Ca2+. O achado de que a β-lapachona inibiu as contrações tônicas induzidas por S-(-)-Bay K8644 (CE50 = 1,4 ± 0,1 x 10-5 M) é sugestivo que o CaV envolvido é o do tipo L. Em conclusão, lapachol, α e β-lapachona apresentam atividade espasmolítica não seletiva em íleo de cobaia, e β-lapachona exerce este efeito pelo bloqueio dos canais CaV tipo L.

Lapachol, α and β-lapachone are naphthoquinones extracted from species of Tabebuia that have shown antiinflammatory, antibacterial, anticancer and trypanosomicidal properties. The aim of this work was to investigate the spasmolytic effect of these naphthoquinones on the guinea-pig ileum, since other naphthoquinones are known to depress the contractile activity of smooth muscles. Lapachol, α and β-lapachone inhibited the phasic contractions induced by both carbachol (IC50 = 1.5 ± 0.2 x 10-4; 7.3 ± 0.9 x 10-5 and 3.2 ± 0.5 x 10-5 M, respectively) and histamine (IC50 = 3.6± 0.5; 3.6 ± 0.7 and 3.3 ± 0.6 x 10-5 M, respectively). These compounds also relaxed the ileum pre-contracted with KCl (EC50 = 1.2 ± 0.4; 4.3 ± 0.8 and 2.7 ± 0.2 x 10-5 M, respectively); carbachol (EC50 = 2.6 ± 0.7; 3.5 ± 0.5 and 2.2 ± 0.7 x 10-5 M, respectively) or histamine (EC50 = 3.0 ± 0.8; 1.1 ± 0.3 and 3.3 ± 0.6 x 10-5 M, respectively) in a concentration-dependent manner. This effect is probably due to inhibition of calcium influx through voltage-gated calcium channels (Ca v). β-lapachone antagonized (pD'2 = 5.73 ± 0.12; slope = 1.51 ± 0.05) CaCl2-induced contractions in depolarizing medium nominally without Ca2+. The finding that β-lapachone inhibited the tonic contractions induced by S-(-)-Bay K8644 (EC50 = 1.4 ± 0.1 x 10-5 M) is suggestive that the L-type CaV is involved. In conclusion, lapachol, α and β-lapachone showed non-selective spasmolytic activity in guinea-pig ileum, and β-lapachone exerts this effect by to blockade of L-type CaV channels.

Compostos oxindol-ferrocenos com potencial atividade anticâncer foram preparados a partir de reações de oxindóis substituídos e carboxaldeído-ferroceno na presença de KOH como catalisador. Os produtos foram caracterizados por dados espectroscópicos incluindo RMN de ¹H e 13C, infravermelho e espectrometria de massas. As configurações E e Z foram estabelecidas por experimentos de nOe ou NOESY.

A series of ferrocenyl oxindoles with potential anticancer activity were prepared from the reactions of substituted oxindoles and ferrocenylcarboxyaldehyde in the presence of KOH as catalyst. The products were characterized by spectral data including ¹H and 13C NMR, IR and mass spectrometry. The E and Z configurations were established by nOe or NOESY experiments.
