RESUMO O uso de cultivares de polinização livre de cebola predomina na região Sul do Brasil, devido à maior adaptabilidade às condições climáticas locais, enquanto híbridos comerciais têm demonstrado baixa adaptabilidade. A macho-esterilidade citoplasmática (CMS) é utilizada para o desenvolvimento de cultivares híbridas de cebola. Foram utilizados dois marcadores moleculares, 5'cob e orfA501, para diferenciar o tipo de citoplasma, e o marcador AcSKP1 para identificar o gene nuclear Ms responsável pela manutenção ou restauração da macho-fertilidade em cebola. Um total de 1.126 plantas de cultivares de cebola mais comuns no Sul do Brasil, incluindo Bola Precoce®, Crioula®, Valessul®, Mega®, Joia® e Princesa do Sul®, foram analisadas utilizando-se os três marcadores. A ocorrência do citoplasma S mostrou-se extremamente rara nas cultivares analisadas, sendo detectado em somente 1,8 % das amostras, enquanto o citoplasma T foi o mais frequente, sendo detectado em 56,3 % das amostras. Das 1.126 plantas analisadas, apenas três possuíam o citoplasma tipo S e eram recessivas para o locus Ms (Smsms). Também foram identificadas 49 plantas com citoplasma N (de acordo com a classificação de Engelke) e recessivas para o gene nuclear (Nmsms), das quais 45 eram produtoras de pólen. ABSTRACT Open-pollinated onion cultivars predominate in the southern region of Brazil, due to their higher adaptability to local climatic conditions, unlike commercial hybrids, which have shown a lower adaptability. Cytoplasmic male sterility (CMS) systems are employed to develop hybrid onion cultivars. Two molecular markers, 5'cob and orfA501, were used to differentiate the cytoplasm type, and the AcSKP1 marker to identify the nuclear male fertility-restoring locus (Ms). A total of 1,126 plants from the most common onion cultivars grown in southern Brazil, including Bola Precoce®, Crioula®, Valessul®, Mega®, Joia® and Princesa do Sul®, were analyzed using all the three markers. An extremely rare occurrence of the S cytoplasm was observed among the cultivars, being detected in only 1.8 % of the samples, while the T cytoplasm was the most prevalent, accounting for 56.3 % of the samples. ABSTRACT: Artificial neural networks (ANN) are computational models inspired by the neural systems of living beings capable of learning from examples and using them to solve problems such as non-linear prediction, and pattern recognition, in addition to several other applications. In this study, ANN were used to predict the value of the area under the disease progress curve (AUDPC) for the tomato late blight pathosystem. The AUDPC is widely used by epidemiologic studies of polycyclic diseases, especially those regarding quantitative resistance of genotypes. However, a series of six evaluations over time is necessary to obtain the final area value for this pathosystem. This study aimed to investigate the utilization of ANN to construct an AUDPC in the tomato late blight pathosystem, using a reduced number of severity evaluations. For this, four independent experiments were performed giving a total of 1836 plants infected with Phytophthora infestans pathogen. They were assessed every three days, comprised six opportunities and AUDPC calculations were performed by the conventional method. After the ANN were created it was possible to predict the AUDPC with correlations of 0.97 and 0.84 when compared to conventional methods, using 50 % and 67 % of the genotype evaluations, respectively. When using the ANN created in an experiment to predict the AUDPC of the other experiments the average correlation was 0.94, with two evaluations, 0.96, with three evaluations, between the predicted values of the ANN and they were observed in six evaluations. We present in this study a new paradigm for the use of AUDPC information in tomato experiments faced with P. infestans. This new proposed paradigm might be adapted to different pathosystems.

RESUMO. Este trabalho objetivou a construção de um mapa genético e identificação de locos de características quantitativas (QTLs) que controlam características do sistema radicular do arroz. Foi avaliada uma população F2:3 composta de 150 famílias a partir do cruzamento entre as variedades IAC 165 x BRS Primavera. A genotipagem foi realizada na população F2 utilizando 3.742 marcadores SNP (Polimorfismo de um Único Nucleótido). A avaliação do sistema radicular nas populações F3 foi realizada através de fenotipagem em larga escala baseada na geração de imagem com scanner raiz CI-600 e na quantificação através do software WinRhizo(r). Foi realizado experimento em delineando blocos casualizados, em casa de vegetação com três repetições. Foram consideradas as variáveis comprimento, área de contato da raiz e volume nas profundidades de 5 a 25 cm e 25 a 45 cm. A análise dos marcadores SNP permitiu a construção de um mapa genético com comprimento total de 1424 cM. O grupo de ligação com maior área de cobertura foi o número 3 com 270 cM (100 SNPs), seguido pelo grupo de ligação 1, com 249 cM (170 SNP), e 2 com 163 cM (99 SNP). A análise genética permitiu a detecção de QTL para todas as características estudadas.

ABSTRACT. The aim of this work is the construction of a genetic map and identification of quantitative trait loci (QTLs) that control characteristics of the root system of rice. We evaluated a F2:3 population composed of 150 families from the cross between the varieties IAC 165 × BRS Primavera. Genotyping was performed in the F2 population using 3,742 SNP (Single Nucleotide Polymorphism) markers. The evaluation of the root system in the F3 population was performed through a large-scale phenotyping method based on image generation with a CI-600 root scanner and on quantification through the WinRhizo(r) software. The experiment was arranged in a randomized block design with three replications performed under greenhouse. The variables analyzed were root length, root surface area and root volume at the depths of 5 to 25 cm and 25 to 45 cm. The SNP markers analysis allowed the construction of the genetic map with a full length of 1424 cM. The linkage group with the largest coverage area was number 3 with 270 cM (100 SNPs), followed by linkage group 1 with 249 cM (170 SNPs) and linkage group 2 with 163 cM (99 SNPs). The genetic analysis allowed the detection of QTLs for all the characteristics.

Analysis using Artificial Neural Networks has been described as an approach in the decision-making process that, although incipient, has been reported as presenting high potential for use in animal and plant breeding. In this study, we introduce the procedure of using the expanded data set for training the network. Wealso proposed using statistical parameters to estimate the breeding value of genotypes in simulated scenarios, in addition to the mean phenotypic value in a feed-forward back propagation multilayer perceptron network. After evaluating artificial neural network configurations, our results showed its superiority to estimates based on linear models, as well as its applicability in the genetic value prediction process. The results further indicated the good generalization performance of the neural network model in several additional validation experiments.

O objetivo deste trabalho foi verificar a existência de loco letal, em uma população hibrída de eucalipto, e quantificar a distorção de segregação no grupo de ligação 3 do genoma de Eucalyptus. Uma população híbrida de E. grandis x E. urophylla, que segrega quanto à resistência à ferrugem, foi genotipada com 19 marcadores microssatélites do grupo de ligação 3 do genoma de Eucalyptus. Para quantificar a distorção de segregação, modelos de máxima verossimilhança (ML), específicos para população exogâmica, foram utilizados. Esses modelos consideram os genótipos dos marcadores observados e o loco letal como parâmetros. As soluções ML foram obtidas por meio do algoritmo de esperança e maximização. Um loco letal, no grupo de ligação 3, foi verificado e mapeado com alta confiabilidade entre os microssatélites EMBRA 189 e EMBRA 122. Este loco letal causa intensa seleção gamética no genitor masculino. Nessa população, sua posição é de 25 cM de distância do loco que controla a resistência à ferrugem.

The objective of this work was to verify the existence of a lethal locus in a eucalyptus hybrid population, and to quantify the segregation distortion in the linkage group 3 of the Eucalyptus genome. A E. grandis x E. urophylla hybrid population, which segregates for rust resistance, was genotyped with 19 microsatellite markers belonging to linkage group 3 of the Eucalyptus genome. To quantify the segregation distortion, maximum likelihood (ML) models, specific to outbreeding populations, were used. These models consider the observed marker genotypes and the lethal locus viability as parameters. The ML solutions were obtained using the expectation‑maximization algorithm. A lethal locus in the linkage group 3 was verified and mapped, with high confidence, between the microssatellites EMBRA 189 e EMBRA 122. This lethal locus causes an intense gametic selection from the male side. Its map position is 25 cM from the locus which controls the rust resistance in this population.

No Brasil, a ferrugem causada por Puccinia psidii Winter destaca-se como a mais importante doença do eucalipto. O uso de genótipos resistentes é o principal método de controle, o que torna a detecção de marcadores ligados a resistência à ferrugem essencial para a seleção de genótipos resistentes. Neste estudo, uma progênie F1 composta por 131 plantas derivadas de um cruzamento interespecífico de Eucalyptus.Um mapa integrado foi construído para o grupo de ligação três com base em marcadores microssatélites. Para detecção de QTL foram utilizadas duas metodologias baseadas em alelos idênticos por descendência (IBD): marca simples de Haseman and Elston e mapeamento por intervalo de Fulker and Cardon. Ambas as metodologias evidenciaram associação mais significativa para o marcador Embra 125. O QTL que explicou 42 % da variação fenotípica foi mapeado a 0,02 cM desse marcador pela metodologia de intervalo de Fulker and Cardon. O marcador Embra125 têm potencial utilização em seleção assistida, com conseqüente aumento na eficiência durante a seleção de genótipos resistentes.

In Brazil the rust caused by Puccinia psidii Winter stands out as the most important disease of eucalyptus. The use of resistant genotypes is the main control method, which makes the detection of markers linked to rust resistance essential to the selection of resistant genotypes. In this study, an F1 progeny of 131 plants from interspecific crossings of Eucalyptus was used to identify markers linked to resistance genes for this pathogen. An integrated map was constructed for linkage group three based on microsatellite markers. For QTL mapping two methodologies based on alleles identical-by-descent (IBD) were used: single marker analysis of Haseman and Elston and the interval mapping procedure of Fulker and Cardon. Both methods showed significant association for the Embra 125 marker.The QTL that explained 42 % of the phenotypic variation was mapped to 0.02 cM of this marker by the Fulker and Cardon. Marker Embra 125 has potential use in assisted selection, thus increasing the efficiency of the selection of resistant genotypes.

Características oligogênicas de distribuição discreta e expressão governada por poucos genes de maior efeito têm se mostrado importantes na condução dos programas de melhoramento, com destaque para a resposta de resistência das plantas às doenças. Métodos tradicionais de detecção de QTL's, que pressupõem normalidade e herança governada por múltiplos fatores, não deveriam ser utilizados para mapeamento dessas características de distribuição discreta e interação epistática predominante. O objetivo deste trabalho é avaliar os resultados de um método para mapeamento e detecção de locos controladores da expressão de características oligogênicas, OTL's (Oligogenic Trait Loci). Esse método, definido como MMCO (Método de Mapeamento de Características Oligogênicas), utiliza funções de verossimilhança para obtenção de estimativas de ligação fatorial entre locos marcadores e locos controladores de características oligogênicas. Os resultados indicam que o método foi adequado para detecção de OTL's em populações F2 relativamente pequenas, compostas por 200 indivíduos, e que a determinação a priori do padrão de herança é condição necessária para a utilização dessa estratégia, que se diferencia por atender as pressuposições de análise, não necessitar de informação prévia de ordenamento entre as marcas e por permitir a obtenção de estimativas a partir da informação contida em todas as classes genotípicas.

Oligogenic traits are distinguished by their heritage ruled by higher effect genes and by their importance for cultivated plants, with emphasis to the plant disease resistance inheritance. The qualitative nature and epistatic interaction of these traits results in a heritage pattern that should not be interpreted through traditional QTL detection strategies. The objective of this work was to propose a method for Oligogenic Traits Loci (OTL) detection. This method, defined as Oligogenic Trait Mapping Method (OTMM) uses maximum likehood probability functions to obtain adjusted "r" estimates that express the distance among the molecular markers and the OTL loci. The results show that the method was adequate for OTL detection even in relatively small F2 populations. The prior definition of the oligogenic heritage pattern is one the main requirements of this method that focus in the attainment of the analysis presumptions without previous markers order information.

O objetivo deste trabalho foi alterar o método centroide de avaliação da adaptabilidade e estabilidade fenotípica de genótipos, para deixá-lo com maior sentido biológico e melhorar aspectos quantitativos e qualitativos de sua análise. A alteração se deu pela adição de mais três ideótipos, definidos de acordo com valores médios dos genótipos nos ambientes. Foram utilizados dados provenientes de um experimento sobre produção de matéria seca de 92 genótipos de alfafa (Medicago sativa) realizado em blocos ao acaso, com duas repetições. Os genótipos foram submetidos a 20 cortes, no período de novembro de 2004 a junho de 2006. Cada corte foi considerado um ambiente. A inclusão dos ideótipos de maior sentido biológico (valores médios nos ambientes) resultou em uma dispersão gráfica em forma de uma seta voltada para a direita, na qual os genótipos mais produtivos ficaram próximos à ponta da seta. Com a alteração, apenas cinco genótipos foram classificados nas mesmas classes do método centroide original. A figura em forma de seta proporciona uma comparação direta dos genótipos, por meio da formação de um gradiente de produtividade. A alteração no método mantém a facilidade de interpretação dos resultados para a recomendação dos genótipos presente no método original e não permite duplicidade de interpretação dos resultados.

ABSTRACT The objective of this work was to modify the centroid method of evaluation of phenotypic adaptability and the phenotype stability of genotypes in order for the method to make greater biological sense and improve its quantitative and qualitative performance. The method was modified by means of the inclusion of three additional ideotypes defined in accordance with the genotypes' average yield in the environments tested. The alfalfa (Medicago sativa L.) forage yield of 92 genotypes was used. The trial had a randomized block design, with two replicates, and the data were used to test the method. The genotypes underwent 20 cuts, from November 2004 to June 2006. Each cut was considered an environment. The inclusion of ideotypes of greater biological average production in the environments produced an arrow-shaped graphical dispersion directed to the right in which the most productive genotypes were placed near the tip of the arrow. With the alteration only five genotypes were classified into the former classes of the original centroid method. The arrow-shaped figure allowed a direct comparison of genotypes throughout the productivity gradient. The alteration performed in the method preserved the easy interpretation of results for genotype recommendations of the original method, and does leaves no room for ambiguity in interpretation of the results.
