ABSTRACT Moringa oleifera Lam. is a tropical tree that belongs to the Moringaceae family and is popularly known worldwide for its multiple applications. This study aimed to evaluate the genetic variability of individuals from the Moringa Genebank of Embrapa Tabuleiros Costeiros, Sergipe state, Brazil. The Moringa Genebank comprises 25 accessions, represented by 177 genotypes, of which 18 were transferred in from an exchanged germplasm of the University of Florida, USA, and the others were from different states in Brazil. Leaves of each genotype were collected for DNA extraction and polymerase chain reaction (PCR) analysis using 20 Inter simple sequence repeats (ISSR) primers. A total of 141 bands were amplified, and 100 % were polymorphic. The average expected heterozygosity (He) and Shannon's index were 0.11 and 0.16, respectively. The highest genetic divergence was found between the M4 and M18 accessions, both from Florida, USA, whereas the closest pair of accessions was M23 and M24, both from Brazil. The cluster analysis obtained through the Structure software divided moringa genotypes into two groups. These results suggest low genetic diversity between the accessions in the Moringa Genebank. Therefore, the introduction of new accessions in the Moringa GeneBank is essential to increasing the genetic variability of the species to ensure its conservation and improvement. Resumo: Para espécies nativas do Brasil existe uma grande necessidade de informações científicas que permitam a produção e armazenamento eficiente de sementes. O presente trabalho avaliou as alterações fisiológicas e moleculares em sementes de Hancornia speciosa armazenadas em soluções conservativas. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema de parcelas subdivididas, com oito repetições. Nas parcelas, soluções conservativas (A, B, C e D) e nas subparcelas, períodos de armazenamento (50, 100, 150 e 200 dias). Foram analisadas as variáveis: teor de água, raios-X, germinação, comprimento da parte aérea, raiz e massa seca de plântulas, condutividade elétrica, concentração e qualidade do ácido ribonucleico, concentração de peroxidase e proteínas resistentes ao calor. O teor de água manteve-se acima de 50% e 88% das sementes estão cheias e bem formadas. Com o aumento dos períodos de armazenamento, ocorre redução na germinação, comprimento da parte aérea, raiz e massa seca de plântulas e aumento da condutividade elétrica. Proteínas resistentes ao calor não foram suficientes para proteger as sementes contra danos macromoleculares e as concentrações de RNA e peroxidase diminuíram ao longo do armazenamento. As sementes armazenadas nas soluções B e C são mais vigorosas, sendo tais soluções indicadas para conservação de H. speciosa.

Abstract: For native species in Brazil, there is a great need for scientific information that allows efficient production and storage of seeds. The present work evaluated the physiological and molecular changes in Hancornia speciosa seeds stored in conservative solutions. The experiment was carried out in a completely randomized design, in a split-plot scheme, with eight replications. In the plots, the conservative solutions (A, B, C, and D) were allocated, and the subplots the storage periods (50, 100, 150, and 200 days). The following variables were analyzed: water content, X-ray, germination, shoot length, root, dry mass of seedlings, electrical conductivity, concentration and quality of ribonucleic acid, the concentration of peroxidase, and heat-resistant proteins. The water content remained above 50%, and 88% of the seeds are full and well-formed. There is a reduction in the germination, shoot length, root, and dry mass of seedlings; and an increase of electrical conductivity with the increase of the storage time. Heat-resistant proteins were not sufficient to protect seeds against macromolecular damage, and RNA and peroxidase concentrations decreased with the increase of the storage time. The seeds stored in solutions B and C are more vigorous, being such solutions indicated for the conservation of H. speciosa.

A pesquisa, aqui apresentada, buscou verificar o grau de sucesso de usuários do Portal CAPES, na busca por um periódico específico e as razões para os casos de insucesso. A metodologia utilizada foi quantitativa, com coleta de alguns dados qualitativos. O estudo analisou, utilizando estatística descritiva, duas questões de um questionário sobre a satisfação dos usuários do Portal, as quais foram respondidas por 930 docentes de 17 universidades, das áreas de Ciências Biológicas, Ciências Humanas e Linguística, Letras e Artes. Os resultados mostram que 645 (70%) dos pesquisadores, quase sempre ou frequentemente, obtêm sucesso ao procurarem por um periódico específico no Portal. Os pesquisadores menos bem sucedidos são os da área de Linguística, Letras e Artes. Para 76% dos pesquisadores, a razão principal para a não obtenção de sucesso é a indisponibilidade do periódico no Portal. Respostas às questões abertas mostraram, também, que, mesmo estando o periódico no Portal, especialmente para literatura mais antiga, pode ocorrer a falta do fascículo específico buscado ou, ainda, estando o fascículo presente no Portal, o acesso ao texto completo do artigo procurado pode não ser permitido. Razões para insucesso na busca relacionadas a fatores, tais como usabilidade, dificuldades com idiomas, falta de treinamento e dificuldades de acesso, foram menos citadas pelos respondentes.

This paper evaluates the level of success of users of the CAPES Digital Library of Scientific Journals, when searching for a specific journal and the reasons for their failures. The methodology used was quantitative with some qualitative data collected. The study used descriptive statistics to analyze two questions of a survey concerning the Portal´s user satisfaction which were answered by 930 faculty members of 17 universities in Brazil. The areas of knowledge investigated were Biological Sciences, Humanities and Linguistics, Literature and the Arts. The results show that 645 (70%) of the researchers are almost always or frequently successful when searching for a specific journal in the Portal. Researchers in Linguistics, Literature and the Arts are the least successful. For 76% of the researchers, the main reason for not obtaining success is the absence of the journal in the collection. Responses to open questions reveal that, especially for older literature, the journal might be present in the collection but the specific issue needed may be absent or access to full text of the particular article sought may be restricted. Reasons for failure related to factors such as usability, difficulty with foreign languages and lack of training or difficulties for access were less cited by respondents.
