Abstract INTRODUCTION: The aac(6’)-Ib-cr and bla KPC genes are spreading among Enterobacteriaceae species, including Providencia stuartii, in some countries of world. METHODS: These genes were investigated in 28 P. stuartii isolates from a public hospital in Recife, Pernambuco, Brazil, by PCR and sequencing. RESULTS: The aac(6’)-Ib-cr gene was detected in 16 resistant isolates, and the bla KPC gene was seen in 14. CONCLUSIONS: The presence of these genes in P. stuartii multi- and extensively drug-resistant isolates indicates that the resistance arsenal of this species is increasing, thus limiting the therapeutic options.

Abstract In this work, four isolates of endophytic fungi (Alternaria alternata, Colletotrichum gloesporioides, Glomerella cingulata and Nigrospora sphaerica), deposited in the culture collection ‘University Recife Mycologia’ (URM) at the Universidade Federal de Pernambuco, were characterized for the genes ITS 1 and 4 (region 5.8 S) and evaluated for taxol production.

RESUMO Na região Nordeste do Brasil, solos salinos são restritivos à produção de banana, tornandose necessário compreender os mecanismos de tolerância de sal. Dois genótipos de banana, Tap Maeo, tolerante, e Berlim, sensível, foram submetidos ao tratamento com 50 mol m-3 NaCl ou sem sal. Este estudo avaliou os efeitos do sal sobre os seguintes aspectos fisiológicos: área foliar, conteúdo e distribuição de Na+, integridade da membrana, atividade da AT Pase. Além disso, uma busca por genes diferencialmente expressos foi realizada usando a técnica Differential Display. O genótipo Tap Maeo apresentou a menor redução na área foliar, menor acúmulo de Na+ e malondialdeído (MDA) bem como maior atividade da H+AT Pase. Duas sequências diferencialmente expressas no genótipo tolerante (Musa 07, Musa 23) compartilham alto grau de identidade com as sequências de aminoácidos dos genes SOS1 e SOS2, respectivamente. O clone Musa 10 é muito semelhante à sequência de aminoácidos do gene da peroxidase do ascorbato, e o Musa 26 codifica a enzima aldeído betaína desidrogenase. Estes marcadores biológicos significativos indicam que a tolerância à salinidade em banana envolve pelo menos dois mecanismos simultâneos: a ativação da via SOS, aumentando a extrusão do Na+, e a ativação do sistema antioxidante, aumentando a síntese de APX e da enzima aldeído betaína desidrogenase.

ABSTRACT In the northeastern region of Brazil, saline soils are constraints to banana production, becoming necessary to understand the mechanisms of salt tolerance. Two bananas genotypes, Tap Maeo, tolerant, and Berlin, sensitive, were subjected to treatment with 50 mol m-3 NaCl or without salt. This study evaluated the effects of salt on the following physiological aspects: leaf area, content and distribution of Na+, membrane integrity, proton AT Pase activity. Besides, a search for differentially expressed genes was performed using the Differential Display technique. Tap Maeo genotype showed the smallest reduction in leaf area, smaller accumulation of Na+ and malondialdehyde (MDA), and higher activity of proton AT Pase activity. Two sequences differentially expressed in the tolerant genotype, (Musa 07, Musa 23), shared a high degree of identity with the amino acid sequences of the genes SOS1 and SOS2, respectively. The clone Musa 10 was highly similar to amino acid sequence of the ascorbate peroxidase gene, and Musa 26, encodes the enzyme betaine aldehyde dehydrogenase. These significant biological markers indicate that salinity tolerance in banana involves at least two simultaneous mechanisms: the activation of the SOS pathway, increasing the extrusion of Na+, and the activation of antioxidative system, increasing the synthesis of APX and betaine aldehyde dehydrogenase enzyme.

Foi determinada a ocorrência de espécies de fungos micorrízicos arbusculares (FMA) em áreas nativa (AN) e impactada (AI) pela mineração de gesso em Araripina, PE, Brasil, utilizando como culturas-armadilha: sorgo e amendoim. Os esporos de FMA foram caracterizados morfologicamente para identificação ao nível de espécie. DNA foi extraído de esporos para caracterização molecular a partir de sequencias da região ITS. No estudo molecular foram identificados três gêneros e quatro espécies (Funneliformis mosseae, Glomus sp., Glomus hoi, Claroideoglomus claroideum), que mostram capacidade para tolerar as condições de estresse da área minerada. Trabalhos em campo devem ser realizados para compreender melhor a dinâmica e funcionalidade dessas espécies de FMA que poderiam ser selecionadas para uso em programas de manejo, na recuperação de áreas similares.

The occurrence of arbuscular mycorrhizal fungi (AMF) in native (AN) and gypsum mining impacted (AI) areas in Araripina, Pernambuco State, Brazil, was determined using as trap cultures sorghum and peanut. The spores were morphologically characterized for identification at species level. DNA was extracted from spores and sequences of the ITS region used for molecular characterization. The molecular study confirmed the identification of three genera and four species (Funneliformis mosseae, Glomus sp., Glomus hoi, Claroideoglomus claroideum) that show ability to support stress conditions. Field work should be carried out to better understand the dynamics and functionality of these AMF species which could be selected for management programs in similar degraded areas.

A diversidade genética de catorze cultivares de cana-de-açúcar (Saccharum oficinarum) foi acessada por meio de marcadores moleculares ISSR. Objetivou-se caracterizar molecularmente as cultivares estudadas. Foram utilizados trinta e sete primers de ISSR, dos quais, oito foram eficientes na amplificação do DNA das amostras analisadas, sendo sete primers suficientes para distinguir todas as cultivares de cana-de-açúcar envolvidas nas análises. A faixa de amplicons variou de 300 a 2000 pb. As cultivares RB 92579 e RB 863129 apresentaram maiores coeficientes de similaridade (77%) enquanto as cultivares RB 961 e RB 931611 formaram o grupo com menor similaridade (22%). Os resultados indicam que os marcadores ISSR foram úteis na análise da diversidade genética e geração de padrões genéticos (fingerprint), em germoplasma de cana-de-açúcar. Marcadores ISSR cultivar-específico foram obtidos com o primer UBC 817 para as 14 cultivares testadas. Num próximo trabalho, mais primers ISSR serão utilizados para buscar mais polimorfismos dessas e de outras cultivares de cana-de-açúcar.

Genetic diversity of fourteen sugarcane cultivars was accessed by ISSR molecular markers. With the aim to characterizing and validating the efficiency of these markers in the fingerprint of studied cultivars, thirty seven ISSR primers were used, from which, eight were efficient for the DNA amplification. Seven primers were efficient to discriminate the fourteen studied sugarcane cultivars. The amplicons varied from 300 to 2000 bp. The cultivars RB 92579 and RB 863129 presented higher similarity coefficient (77%) while the cultivars RB 961 and RB 93611 formed the group with lower similarity (22%). The results suggested that ISSR markers were useful in the analysis of the genetic diversity and in the fingerprint in sugarcane germosplasm. In the next step more ISSR primers will be used in order to obtain more polymorphism from these varieties and to analyze more sugarcane cultivars.

O objetivo deste trabalho foi determinar o perfil de transcritos em plantas de tomate (Lycopersicon esculentum Mill.), durante a infecção com Fusarium oxysporum f. sp. lycopersici e após a aplicação foliar de ácido salicílico. A técnica de hibridização subtrativa por supressão (SSH) foi utilizada para gerar uma biblioteca de cDNA enriquecida por transcritos diferencialmente expressos. Foram identificados 307 clones, em duas bibliotecas subtrativas, que permitiram o isolamento de diversos genes de defesa com função em diferentes processos relacionados à resistência vegetal contra patógenos. Também foram isolados, nas duas bibliotecas, genes com função desconhecida, o que indica a possibilidade de identificação de novos genes que ainda não tenham sido relatados em estudos anteriores de resposta a estresses e defesa, em plantas. A técnica SSH é eficiente em identificar genes de resistência, ativados pelo ácido salicílico e pela infecção com Fusarium oxysporum f. sp. lycopersici. A aplicação dessa técnica não apenas possibilita isolar seqüências diferencialmente expressas, a baixo custo, como também permite a identificação de novas seqüências, em tomate, a partir de um número relativamente pequeno de seqüências.

The objective of this work was to determine the transcript profile of tomato plants (Lycopersicon esculentum Mill.), during Fusarium oxysporum f. sp. lycopersici infection and after foliar application of salicylic acid. The suppression subtractive hybridization (SSH) technique was used to generate a cDNA library enriched for transcripts differentially expressed. A total of 307 clones was identified in two subtractive libraries, which allowed the isolation of several defense-related genes that play roles in different mechanisms of plant resistance to phytopathogens. Genes with unknown roles were also isolated from the two libraries, which indicates the possibility of identifying new genes not yet reported in studies of stress/defense response. The SSH technique is effective for identification of resistance genes activated by salicylic acid and F. oxysporum f. sp. lycopersici infection. Not only the application of this technique enables a cost effective isolation of differentially expressed sequences, but also it allows the identification of novel sequences in tomato from a relative small number of sequences.
